Description
実施例
樹状細胞(DC)は、一次T細胞反応の強力な活性化因子である。DCの独特な初回抗原刺激能力は、成熟化刺激との遭遇で獲得される。グラム細菌への暴露によって誘導される成熟により差次的に発現された遺伝子を識別するため、11000個の遺伝子および発現配列タグ(EST)を提示するマイクロアレイを用い、DC遺伝子発現の速度論的研究を行った。約3000個の差次的発現した転写物が識別された。予想外なことに、IL-2産生を引き起こす機能的インターロイキン(IL)-2転写物は、細菌遭遇後の早い時点で一過性にアップレギュレートされた。対照的に、マクロファージは、細菌刺激によってIL-2を産生しなかった。我々は、IL-2が、DCによる独特なT細胞刺激能力を付与するもう1つの重要な分子であることを明らかにした。
DC、マクロファージおよび培地。10%熱不活化ウシ胎仔血清(GIBCO)、100 IUペニシリン、100μg/mlストレプトマイシン、2mM L-グルタミン(すべてSigmaより)、および50μM 2-メルカプトエタノールを含有するIMDM(Sigma、St. Louis、MO)(完全IMDM)中、R1培地からの30%上清(GM-CSFを形質移入されたNIH3T3線維芽細胞からの上清)と共にD1細胞を培養した。マクロファージおよびDCは、4匹の異なるマウスから集めた同じ骨髄細胞から取得し、2個の別々の培養液に分割した。30%R1培地における14日間の骨髄培養後にmBMDCが得られた。M-CSF存在下の14日間の骨髄培養後にマクロファージが得られた。野性型およびDCは、同じ同腹仔のマウスから得た。低いB7.2発現およびCD40の欠如によって判断し、mBMDCが未成熟であった場合にのみ細菌活性化に使用した。部分的な自然活性化を示す細胞は捨てた。

IL-2 ELISAは、DuoSetキット(R and D、Minneapolis、MN)を用い、製造業者の推奨事項に従って行った。
情報の希薄化および混入を避けるため、転写分析には均一な集団が必要である。柔軟性のため、mBMDCは極めて不均一であり、成熟DCおよび中間期DCを混入させずに均一な未成熟細胞を得ることは不可能である。我々は、in vivoにおけるDC成熟プロセスを模倣し、細菌、細菌細胞産物または炎症性サイトカインに反応して完全に成熟させることができる均一かつ未成熟な増殖因子依存性(顆粒球単球コロニー刺激因子)マウスDCの増殖を可能にするDC培養系について報告している。この培養系を用いて得られる十分に特徴が明らかにされたDC系であるD1を用いた研究は、新鮮な脾DCまたはBMDCに見られるのと同様な成熟を示す。
実験情報を失う(分散)ことなく我々の全データセットを近似的に可視化するため、我々はまず、複雑なデータの次元を軽減することが可能である主成分分析(PCA)法を適用した。したがって、我々は、対応するトランスクリプトームを最もよく説明する速度点(kinetic point)の特徴について全体的に説明することができた(表1)。
得られた9930個の遺伝子およびESTのコレクション中で差次的に発現した遺伝子を識別するため、我々はまず、自己組織化マップ(SOM)に基づき発現パターンの類似性に従って遺伝子をグループ化するクラスター化アルゴリズムを適用した。二組の平均差(AvgDiff、発現レベルを示すパラメータ)を利用してデータにフィルターをかけ、最高速度値と最低速度値の変化が3倍以下である遺伝子またはESTをすべて除外した。このフィルターにより、クラスター化すべき配列は2951に減少し、その約半数はESTであった(図2b)。
我々は、クラスター32が、T細胞増殖因子IL-2をコードする転写物を含むことを観察した。細菌遭遇は、活性化後の早い時点(4〜6時間)でD1細胞における一過性のIL-2 mRNAアップレギュレーションを誘導した(図4a)。したがって、我々は、この分子がDCによって産生される重要な共刺激分子に相当するという仮説を検証した。
初期の細菌で初回抗原刺激されたDCにより産生されるIL-2のT細胞活性化における役割は、一次混合リンパ球反応(MLR)アッセイにおいてアロ反応性CD4+およびCD8+T細胞を刺激するIL-2-/-および野性型DCの能力を分析することにより検討した。mBMDCは、野性型およびIL-2-'-マウス20から入手し、感染多重度10で細菌により活性化した(図1)。野性型およびIL-2-/-DCを細菌刺激によって同様に活性化し(図5a)、それらは生存度の差を全く示さなかった(図5b)。
PCA法を用いて全データセットを可視化した。このようなタイプの研究では、PCAは、細胞活性化のプロセスを記述し、結果的に細胞分化のプロセスと区別することを可能にする。活性化は可逆的現象であり、所与の刺激物への暴露によって細胞は過渡的な機能的および表現型的改変を受け、次いで元の状態に戻る。これに反して、分化事象は、新たな機能的状態に向かう進行を誘導する不可逆的プロセスである。遺伝子発現プロフィールは、細胞表現型および機能の主要な決定要因であるため、速度論的研究において細胞遺伝子発現パターンに応用されるPCAにより、刺激後の同一細胞内における様々な状態の類似性を可視化することが可能である。
本研究では、我々は、DCによるIL-2産生を誘導する様々な刺激物の能力および活性化後にIL-2を産生するランゲルハンス細胞などの組織常在性未成熟DCの能力を分析する。我々は、TLRと結合するが炎症性サイトカインとは結合しないことが知られている刺激物のみがDCによるIL-2分泌を誘導できること、およびIL-2産生はDC組織の由来とは無関係であることを示す。実際に、脾臓、骨髄あるいは皮膚(ランゲルハンス細胞)からの未成熟DCは、微生物刺激活性化後にIL-2を分泌することができる。興味深いことに、T細胞と相互作用する早期の活性化DCでは、IL-2は、T細胞接触部位に局在する。最後に、本研究で我々は、DCによるIL-2産生が細菌またはLPS注射後にin vivoでも起きることを示す。
抗体および試薬。LPS(大腸菌026:B6、10μg/mlで使用)およびザイモサン(10μg/mlで使用)は、Sigma Chem.(St. Louis、MO)から入手した。rTNF(San Francisco)およびrIL-1(Genzyme、Cambridge MA)は、それぞれ100 U/mlおよび10μg/mlで使用した。IFNは、Schering-Plough(Dardilli、Fr.)よりご提供いただき、1000 U/mlで使用した。CpG(TCCATGACGTTCCTGATGCT)(配列番号5)およびCpG対照(TCCATGAGCTTCCTGATGCT)(配列番号6)オリゴはLife Technologiesから購入し、1□Mの濃度で使用した。PE結合抗IL-2、PE結合ラットアイソタイプ対照、ビオチン化抗CD8αおよびFITC結合抗CD11cモノクローナル抗体は、Pharmingenから購入した。Quantum-red結合ストレプトアビジンは、Sigmaから入手した。抗I-Ad/I-Ed mAb(クローンM5/114、ラットIgG2b)は、Dr. A. Ager(NIMR、London、UK)よりご提供いただいた。FITC結合抗ラットは、Jackson ImmunoResearch(West Grove、PA)から購入した。抗CD40抗体[クローンFGK45(10)]は、20μg/mlの濃度で使用し、モノクローナル抗ラット抗体(10μg/ml、PharMingen)とクロスリンクさせた。
DCは、いったん活性化されると細胞内でシグナルを伝達し、炎症性サイトカイン、ケモカインおよび共刺激分子などの、侵入した病原体に対する防御に関与する遺伝子の誘導をもたらす様々な機能性TLRを発現する。様々なTLRの活性化がIL-2産生を誘導するかどうかを検討するため、グラム陰性菌の成分であるLPS、およびTLR4を介してシグナルを送るグラム陽性菌の成分であるリポテイコ酸(LTA)、TLR9によって認識される細菌DNAのメチル化されていないCpGモチーフを含むオリゴDNA、ザイモサン、酵母細胞壁粒子、およびTLR2によって認識されるグラム陽性菌のペプチグリカン(peptiglycan)(PGN)により、in vitroにおいてマウスDCを刺激した。
食作用プロセス自体がIL-2産生に関してDCを活性化するかどうかを検討するため、対照として不活性ラテックスビーズまたはグラム陰性菌の大腸菌DH5αと共にDCをインキュベートし、培養上清中のIL-2を測定した。我々の結果から、不活性ラテックスビーズ単独では脾DCまたは骨髄DCのいずれにおいてもIL-2産生を誘導することができないことが明らかとなり、このプロセスには微生物刺激が必要であるというこれまでの知見を支持している。
DCによるIL-2分泌は、抗CD40抗体によるCD40の活性化などのT細胞ヘルプを模倣する刺激によっても誘導されると思われる。CD40の活性化は、D1細胞によってIL-2産生を誘導する際には微生物刺激と同程度に効率的な刺激である。対照的に、BMDCRAG2-/-培養液では、CD40刺激後の早い時点で少量のIL-2が上清中に検出されると思われる。このことは、細胞表面において新鮮な未成熟BMDCRAG2-/-が発現するCD40がD1系よりも少ない(細胞蛍光測定分析によりほとんど検出不可能)という事実に起因すると思われる。DCをまずLPSにより12時間処理し、次いでCD40活性化を行った場合には、IL-2産生の第2の遅延相が誘導されると思われ、微生物刺激による活性化後の早い時期には、DCが他の刺激に反応しないことを示している。
非リンパ組織に由来するDCが刺激によってIL-2を産生するか否かを調べるため、マウス表皮シートからトリプシン処理によって調製したランゲルハンス細胞をLPSで刺激し、6時間後にフローサイトメトリーによりIL-2産生を分析した。我々のデータは、極めて限られた数の非刺激ランゲルハンス細胞(MHCクラスII陽性細胞)がIL-2陽性であることを示していた。この数は、LPS刺激後に大きく増加した。IL-2産生の特異性を検証するため、LPS刺激の前にサイクロスポリンA(CsA)で上皮細胞を処理した。CsAは、LPS遭遇後のDCによるIL-2産生を大きく減少させる。CsA処理後、IL-2産生ランゲルハンス細胞の数は、未処理細胞に比べて減少した。
次いで、我々は、IL-2を産生する能力がin vivoにおけるDCに共通の一般的なDC特性であるのか、in vitroにおいて分化した細胞だけの特徴であるのかを検討した。この目的のため、GM-CSFまたはFLT3L形質導入腫瘍をマウスに移植し、それぞれCD8α-CD11c+またはCD8α+CD11c+脾DC集団を増殖させ(21)、LPSまたは大腸菌DH5αを腹膜内注射してDCを初回抗原刺激した。LPSまたは細菌処理マウスの脾臓におけるIL-2発現CD11c+DCの存在を細胞蛍光測定分析によって明らかにした。
ナイーブT細胞の初回抗原刺激は、長時間のT細胞受容体刺激を必要とし、この刺激は、DCとT細胞との接触領域で起こる特殊化した分子組織である免疫学的シナプスの形成によって行われる。高レベルの共刺激分子およびMHC-ペプチド複合体のDC発現ならびにサイトカインの分泌は、APCとしてのDCの高い効率の源である。IL-2は、独特なナイーブT細胞刺激能力をDCに付与するもう1つの分子であることから、早期の活性化DCが産生するIL-2が相互作用するDCとT細胞の接触領域で動員されるかどうかが検討されてきた。
DCに独特な特徴は、NK、BおよびT細胞反応を活性化する能力である。DCによるNKおよびB細胞活性化を担う機序はほとんど不明であるが、DCによるT細胞の活性化は、プロセシング機序の効率、高レベルの共刺激分子およびペプチド-MHC複合体の発現ならびに極性サイトカインの産生などの多くの要素に左右される。最近、最近刺激後の早い時期にDCがIL-2を産生することを我々が立証し、DCのT細胞との、おそらくNKおよびB細胞との相互作用を理解する新たな可能性が開けた。したがって、in vitroにおいてDCによるIL-2分泌を誘導する刺激物およびin vivoにおけるDCのIL-2産生能を明確にすることが重要であった。
本研究では、我々は、DCがマウスCMV(MCMV)感染を許すことを立証する。より重要なことに、DCの感染は、T細胞反応の活性化に必要とされる危険なシグナルのその後の伝達を予防する。本研究は、DC機能のCMV媒介性損傷が、この病原体による感染に伴い、鍵になる因子としてIL-2を巻き込む免疫抑制の誘導に極めて重要であるという初めての証拠を提供する。
動物。6週齢の近交系C57BL/6(H2b、I-Ab)およびBALB/c(H2d、I-Ad)マウスをAnimal Resources Center(Perth、Western Australia)から入手し、University of Western AustraliaのAnimal Services Facilityにおいて特定の無菌条件で飼育した。すべての動物実験は、University of Western AustraliaのAnimal Ethics and Experimentation Committeeの承認を受け、National Health and Medical Research Council of Australiaのガイドラインに従って行った。
一段階増殖曲線
感染試験では、MEFを増殖させて集密とし、D1細胞は、継代日に感染させた。精製CD11c+細胞(DC)は、細胞分取直後に感染させた。MEFの感染では、MOI>3で集密単層にウイルスを加え、37℃で1時間吸収させた。1時間後に残留種菌を除去し、室温で1分間クエン酸緩衝液(40mMクエン酸、10mM KCl、135mM NaCl pH3.0)で細胞を処理し、結合ウイルスを可逆的に除去した。次いで、細胞を十分に洗浄し、新鮮なMEM/2%NCSを加え、様々な時間細胞をインキュベートした後、-70℃で冷凍した。
多段階増殖曲線は前述のように測定したが、ただしMOIを0.02まで減らした。成熟D1細胞の感染では、感染の2日前にLPS(Sigmaからの大腸菌026:B6)(10μg/ml)をD1細胞に添加した。
免疫反応の開始および調節においてDCが果たす中心的役割を考えて、我々は、感染に対するそれらの感受性および生産的MCMV複製を維持するそれらの能力を調べた。D1培養系を用いる我々の初期分析により、MCMV複製に対するDC成熟状態の関連性を評価することができた。高い感染多重度(MOI)(>3pfu/細胞)における未成熟D1細胞、または標準的マウス胚性線維芽細胞(MEF)培養液の感染はウイルス産生をもたらした。しかしながら、D1細胞におけるウイルス産生は、MEFにおけるよりも少なかった(図7a)。MEFの感染後、感染から18時間後にウイルス子孫が放出される(図7a)。対照的に、D1細胞では、感染後27時間まで感染ウイルスは検出されず、ウイルスの力価は、感染後4日間にわたり増加し、最終的にMEFによって産生されるレベルを上回った。
In vitroにおいてDCに感染させ得ることを立証したことから、in vivoにおける感染との関連でDCがどのような役割を果たしているかを判断することが重要となった。MCMVがin vivoにおいてDCに感染するか否かを明らかにするため、我々は、LacZ遺伝子産物を発現する組み換えウイルスを利用した。LacZの発現は、M33 ORFのイントロン中のLacZカセットの安定な挿入によって生じる。この挿入は、感染との関連でLacZの安定な発現をもたらし、この発現がウイルス複製に対する有害な影響を及ぼさないことはすでに分かっている。LacZ遺伝子産物のβ-ガラクトシダーゼの基質としてFDGを用いると、蛍光分析によってMCMV感染細胞を検出することが可能であった(図7b)。CD11c、MHC-IIおよびFDGによる標識化後に、感染の過程における感染DCの割合を測定した。脾DCは感染から24時間後にMCMV陽性(10.10±4.21%)となり、2日目までに大部分が感染した(74.61±6.22%)。
未成熟抗原プロセシング細胞から成熟抗原提示細胞へのDCの成熟は、飲食作用の能力の低下を伴う。フルオレシン(Fluorescin)結合デキストラン(FITC-DX)の取り込みを測定することにより、MCMV感染D1細胞の飲食作用能力を未成熟またはLPS刺激D1細胞と比較した。感染から2日後、未成熟D1細胞に比べ、MCMV感染細胞では50%の取り込み低下が観察されたが(図8a)、完全な阻害は、感染から4日後に観察された(図8a)。
MCMVが生産的にDCに感染できることが明らかになったことから、これらの細胞の表現型に関してこのような感染の影響を判定することが重要となった。D1系を用いて初期分析を行った。D1細胞は、CD11c、CD11bおよびCD54ならびにMHCクラスIおよびII、CD40、CD80およびCD86を発現する。MCMVによる未成熟D1細胞の感染後(MOI>3)、2段階の表現型変化が観察された。感染後2日目には、MHCクラスIおよびII、CD40、CD54およびCD86のレベルは、LPS活性化後に観察されるレベルに類似したレベルまで増加した(図9a)。興味深いことに、CD80のレベルは未変化のままであった。感染から4日後に同じ抗原を分析した場合、試験したすべてのマーカーの発現に減少が観察され(図9b)、MHCクラスIIの場合に最も顕著な影響が起きた。
MCMV感染がDCの表現型活性化を妨害することを考えると、MCMVによる感染後のDCが他の刺激物による活性化に反応性のままであるか否かを判定することが重要となった。感染後2または4日目にLPSで48時間刺激されたMCMV感染D1は、活性化刺激物に反応せず、共刺激分子の発現に増加は観察されなかった(図12)。
MCMVによる感染後のDCで観察される表現型変化がそれらの機能に影響を与えるか否かを判定するため、我々は、サイトカイン産生との機能的関連が存在するか否かを調べた。D1細胞は、LPS処理などの活性化刺激物に反応してIL-12を分泌した(図13a)。しかしながら、MCMVに感染したD1細胞は、感染後2および4日目までIL-12を分泌する能力の低下を示した(図13a)。同じことは、2または4日間MCMVに感染させたマウスの脾臓から単離されたDCについてもあてはまった(図13b)。
MCMV感染DCのナイーブ同種T細胞の増殖を刺激する能力は、D1細胞(図14aおよびb)または精製DC(図14c)を用い、in vitroにおいて評価した。LPS活性化D1細胞は、処理から2日後と4日後の双方で同種T細胞の最も効率的な刺激物であり、同系対照に比べ、5〜10倍の増殖を誘導した(図14aおよびb)。
CMV感染の1つの目立った特徴は、感染プロセスの初期を特徴付ける一過性であるが重大な免疫不全である。一過性で数週間から数ヶ月続くが、CMV関連免疫不全は、宿主の生存に対して重要な意味を有している。移植手術を受ける患者では、このウイルス誘発性抑制は実に有益なことがあり、移植拒絶反応の低下と関わっていることが分かっている。しかしながら、二次的日和見感染の発生率が上昇するため、CMV誘発性免疫抑制は、好ましくない転帰につながることが多い。したがって、CMV誘発性免疫抑制をよりよく理解することは、改良されたウイルス療法の設計における重要な検討材料である。
未成熟および成熟樹状細胞(DC)の機能的および表現型的特徴を十分に明らかにしてきた。微生物またはリポ多糖(LPS)などの細菌産物と腫瘍壊死因子(TNF-α)を含む炎症性分子は共に、DCに先天性および適応性免疫反応を開始させかつ増幅させるDC成熟プログラムを活性化すると考えられている。しかしながら、DCの機能状態は様々な刺激物によって誘導され、免疫反応の結果に関係しているという証拠が増えつつある。したがって、我々は、LPS刺激あるいはTNF-α刺激後の成熟移行型DCおよび未成熟DCの転写プログラムを比較した。約6,500のマウス遺伝子およびESTを提示するGeneChip(登録商標)オリゴヌクレオチドマイクロアレイをこの分析に使用した。2種類の刺激物により遺伝子発現の極めて様々な変調が観察された。LPS処理細胞のみが、決定的な増殖停止ならびに免疫反応の適切な活性化および制御に一致する遺伝子の発現パターンを示した。
細胞および試薬。D1細胞は、マウス脾DCから入手し、前述の30%R1馴化培地を添加したIMDM中にin vitroで維持した。LPS(大腸菌血清型026:B6)はSigma Chemical Co.から購入し、10μg/mlで使用した。マウスTNF-α(Genetech Inc.、San Franscisco、CA)は、100U/mlで使用した。細胞は同時に増殖させ、採集した。
β、IL-12p40およびIL-6を定量した。
DC初回抗原刺激に対するTNF-α対LPSの差次的影響を検討するため、我々は、数千個の遺伝子の発現を同時に分析することができるAffymetrix Gene-Chip技術を用いた。情報の希薄化および混入を避けるため、これらの分析には均一な細胞集団が必要である。骨髄由来マウスDCは極めて不安定であり、成熟DCおよび中間期DCを混入させずに均一な未成熟細胞を得ることは不可能である。新鮮なDC機能と密接に平行する細胞系が妥当な選択肢である。したがって、我々は、前述のマウスDC系D1を利用した。D1は、未成熟状態で培養液中に無期限に維持することができる脾性、骨髄性および増殖因子依存性のDC系である。この細胞系は、様々な刺激物を用いて完全な成熟にすることができる。特に、D1細胞は、表現型の特徴(クラスIIおよび共刺激分子のアップレギュレーション)および抗原提示、遊走の阻害、抗原取り込みの遮断、細胞骨格再配列のような機能的特徴によって評価されるように、LPSまたはTNF-α刺激から18時間後には成熟状態に到達する。
AvgDiff=(Σ(PM-MM))/(対のn)
20のAvgDiffは、検出遺伝子について蛍光強度の低値に近い。各プローブアレイの絶対(個別)分析を行い、分析した各標的cRNAにおける遺伝子発現レベルを測定した。次いで、各チップアレイについてシグナル強度の平均値を測定し、固定された任意の値(目標強度)にスケーリングすることにより、ハイブリダイズされたアレイ間でシグナル強度を正規化した。我々は、マウスGeneChipについてシグナル強度の平均値を考慮して算出された100の目標強度を用いた。GeneChipの性能および試料調製の変動を最小限に抑えるためにこの手順を開発した。
分化の終了は、増殖している細胞の増殖停止をもたらす。未成熟な脾臓のマウスDCにおける細胞周期制御は厳密に制御されていないという証拠があり、線維芽細胞および内皮細胞の放射線処理された間質細胞単層上にそれらをプレートした場合に、細胞数の僅かな増加が観察されることがある。成熟DCは増殖能力を完全に失い、活性化から8〜9日後にアポトーシスによって死滅する。
LPS刺激後に増殖停止したDCの生存には、成熟DCをリンパ組織中に移動させ、ナイーブCDB+およびCD4+T細胞を初回抗原刺激させることが必要と思われる。この仕事を行うため、DCは、MHCクラスIおよびクラスII分子上に負荷するべきペプチドを天然タンパク質抗原から産生する能力を拡大しなければならない。興味深いことに、抗原提示機能に関与する遺伝子は、LPSまたはTNF-α活性化D1細胞において特有の発現パターンを示した。
生理学的免疫反応は、十分に制御された炎症反応に由来している。炎症プロセスの活性化および制御に関与する遺伝子は、2種類の異なる刺激物の存在下で成熟したDCにおいて差次的に調節される(表2)。LPS成熟DCにおいて、それほどではないにせよTNF-α処理細胞において、補体分子C1qの抑制または強力なダウンレギュレーションが観察される(表2)。
Claims (21)
- in vitroでリンパ球を活性化するための方法であって、大腸菌、リポ多糖(LPS)、酵母、ザイモサン、メチル化されていないCpGモチーフを含むオリゴDNA、リポテイコ酸およびペプチドグリカン(PGN)より選択される樹状細胞刺激物の存在下で樹状細胞を培養および刺激して、IL-2の産生を誘導することと、IL-2および樹状細胞を含む得られた培養液とリンパ球を接触させ、それによってリンパ球を活性化することを含む方法。
- 前記リンパ球が免疫系のエフェクター細胞である、請求項1に記載の方法。
- 前記リンパ球がNK細胞、NKT細胞、B細胞、またはT細胞である、請求項1に記載の方法。
- 前記樹状細胞がリンパ球を含む対象にとって内因性である、請求項1に記載の方法。
- 前記樹状細胞がランゲルハンス細胞、CD8α陽性細胞、CD8α陰性細胞、またはCD11c陽性細胞である、請求項1に記載の方法。
- in vitroで樹状細胞におけるIL-2産生を誘導する方法であって、前記樹状細胞のトール様受容体を活性化する薬剤と前記樹状細胞とを接触させることを含み、前記薬剤が、大腸菌、LPS、酵母、ザイモサン、メチル化されていないCpGモチーフを含むオリゴDNA、リポテイコ酸およびPGNより選択される方法。
- 前記トール様受容体(TLR)がTLR2、TLR4、またはTLR9である、請求項6に記載の方法。
- in vitroでの細胞ベースの治療法のための、IL-2を産生する樹状細胞を製造する方法であって、前記樹状細胞のトール様受容体を活性化する薬剤と前記樹状細胞とを接触させることを含み、該活性化が前記樹状細胞のIL-2産生を誘導するものであり、前記薬剤が、大腸菌、LPS、酵母、ザイモサン、メチル化されていないCpGモチーフを含むオリゴDNA、リポテイコ酸およびPGNより選択される方法。
- 樹状細胞成熟に影響を与える薬剤をスクリーニングする方法であって、被験薬剤の存在下および非存在下で、大腸菌、LPS、酵母、ザイモサン、メチル化されていないCpGモチーフを含むオリゴDNA、リポテイコ酸およびPGNより選択される微生物刺激および未成熟樹状細胞をインキュベートし、被験薬剤の存在下および非存在下で樹状細胞におけるIL-2発現を検出することを含み、ここで被験薬剤によって引き起こされるIL-2発現の量の増加または減少が樹状細胞成熟に影響を与える薬剤であることの指標である方法。
- 前記被験薬剤が、化合物、小分子、ポリヌクレオチド、およびポリペプチドからなる群から選択される、請求項9に記載の方法。
- 前記未成熟樹状細胞がD1細胞である、請求項9に記載の方法。
- 樹状細胞によるIL-2の発現の量に影響を与える薬剤の能力をテストするためのアッセイ系であって、被験薬剤、大腸菌、LPS、酵母、ザイモサン、メチル化されていないCpGモチーフを含むオリゴDNA、リポテイコ酸およびPGNより選択される微生物刺激、および未成熟樹状細胞が入った容器を含み、前記被験薬剤によって引き起こされるIL-2発現の量の増加または減少が樹状細胞成熟に影響を与える薬剤であることの指標である系。
- 前記被験薬剤が、化合物、小分子、ポリヌクレオチド、およびポリペプチドからなる群から選択される、請求項12に記載の系。
- 前記未成熟樹状細胞がD1細胞である、請求項12に記載の系。
- 樹状細胞のIL-2産生を引き起こす微生物刺激に対応する樹状細胞成熟用遺伝子発現プロフィールのライブラリーを作成する方法であって、
未成熟樹状細胞を前記微生物刺激と共にインキュベートすること、
前記微生物刺激により活性化された前記樹状細胞によるIL-2の産生を検出すること、
前記微生物刺激が遺伝子発現に変化を引き起こす樹状細胞中の遺伝子を識別すること、および
前記微生物刺激の遺伝子発現プロフィールであって、前記微生物刺激の存在下で樹状細胞における発現を変化させる1種または複数の遺伝子を含むプロフィールを作成すること
を含み、
前記微生物刺激が、大腸菌、LPS、酵母、ザイモサン、メチル化されていないCpGモチーフを含むオリゴDNA、リポテイコ酸およびPGNより選択される方法。 - 前記樹状細胞がD1細胞である、請求項15に記載の方法。
- 免疫抑制性ウイルス感染を治療するための治療剤候補をスクリーニングする方法であって、被験薬剤の存在下および非存在下において免疫抑制性ウイルス感染に関係する免疫抑制性ウイルスと樹状細胞とをインキュベートすること、前記樹状細胞のIL-2産生のレベルを測定することを含み、前記被験薬剤によって引き起こされる前記IL-2産生レベルの増加が、免疫抑制性ウイルス感染を治療するための治療剤候補であることの指標である方法。
- 前記被験薬剤が、化合物、小分子、ポリヌクレオチド、およびポリペプチドからなる群から選択される、請求項17に記載の方法。
- 前記樹状細胞が未成熟樹状細胞である、請求項17に記載の方法。
- 前記樹状細胞がD1細胞である、請求項17に記載の方法。
- 前記免疫抑制性ウイルスがHIVである、請求項17に記載の方法。
