Description
<薬用人参カルスの分化誘導>水洗いしたアメリカ薬用人参を70%エタノール水溶液に30秒間浸漬した後、2%次亜塩素酸ナトリウム水溶液に10分間浸漬して滅菌した。
培養条件Bにおける液体培地中のIAA濃度を表1に記載した濃度に変更した以外は実施例1と同様にして培養し、乾燥収量比、およびサポニン含量比を計算した。実施例2〜4における培養条件BのIAAの濃度は、それぞれ培養条件Aに対して1/4、1/3、1/2倍の濃度である。
培養条件Bにより培養する工程を設けず、培養条件Aでの継代培養を12ヶ月間行った後に、実施例1と同様にして培養し、乾燥収量比、およびサポニン含量比を計算した。
培養条件Bを、培養条件Aと同じ組成の液体培地を400mL使用する条件に変更した以外は実施例1と同様にして培養し、乾燥収量比およびサポニン含量比を計算した。
培養条件Bにおける液体培地中のIAA濃度を0.5ppmに変更した以外は実施例1と同様にして培養し、乾燥収量比およびサポニン含量比を計算した。培養条件BのIAAの濃度は、培養条件Aに対して1/8倍の濃度である。
薬用人参としておたね人参を使用した点、培養条件Aおよび培養条件Bの条件を表2に記載した培地組成および使用培地量とした点、そして、培養条件A、培養条件Bでの継代培養を、培養条件A、培養条件B、培養条件A、培養条件Bの順にそれぞれ2ヶ月間交互に培養を行って合計8ヶ月間の継代培養とした点以外は実施例1と同じ操作を行い、おたね人参カルス培養物の継代培養を行った。
培養条件Bにおけるカイネチン濃度を表2に記載した濃度に変更した以外は実施例5と同様にして培養し、乾燥収量比、およびサポニン含量比を計算した。実施例6〜8における培養条件Bのカイネチンの濃度は、それぞれ培養条件Aの2、3、5倍の濃度である。
培養条件Bによる培養工程を設けず、培養条件Aでの継代培養を8ヶ月間行った以外は実施例5と同様にして培養し、乾燥収量比、およびサポニン含量比を計算した。
培養条件Bを、培養条件Aと同じ組成の液体培地を450mLに変更した以外は実施例5と同様にして培養し、乾燥収量比、およびサポニン含量比を計算した。
培養条件Bにおける培地中のカイネチン濃度を2ppmとした以外は実施例5と同様にして培養し、乾燥収量比、およびサポニン含量比を計算した。この培養条件Bにおけるカイネチンの濃度は、培養条件Aに対して10倍である。
Claims (5)
- 培養条件Aでの液体培養および培養条件Bでの液体培養を交互に繰り返すことにより薬用人参カルスを継代培養する方法であり、培養条件Bが、培養条件Aに比べて使用培地量が110〜200容量%であるとともに、液体培地中のオーキシン類の濃度が1/7〜1/2倍であることを特徴とする薬用人参カルスの継代培養方法。
- 培養条件Aおよび培養条件Bのオーキシン類の濃度が0.1〜10ppmの範囲である請求項1記載の薬用人参カルスの継代培養方法。
- 培養条件Aおよび培養条件Bの液体培地がともにサイトカイニン類を含有しており、培養条件Bのサイトカイニン類の濃度が培養条件Aのサイトカイニン類の濃度に比べて2〜5倍である請求項1記載の薬用人参カルスの継代培養方法。
- 培養条件Aおよび培養条件Bのサイトカイニン類の濃度が0.01〜5ppmの範囲である請求項3記載の薬用人参カルスの継代培養方法。
- 薬用人参カルスがオタネ人参、チクセツ人参、アメリカ人参、三七人参、シベリア人参から選ばれる薬用人参のカルスである請求項1〜4のいずれか1項記載の薬用人参カルスの継代培養方法。
