Description
シリカをベースとするRP(逆相)充填材は、通常クロマトグラフィーカラム中で使用される。これらはシリカ表面上に未反応のシラノール基を含む。残存シラノール部位は極性の検体と、特にイオン交換、水素結合及び双極子−双極子機構を介して塩基性化合物と相互反応する可能性がある。これらの相互作用により、保持の増大から過剰のテーリング(tailing)に広がる問題及び試料の不可逆的な吸着を生じる可能性がある。これらの望ましくない作用を、超純粋シリカを使用し、かつ疎水性リガンド及びエンドキャッピング試薬による表面被覆を最大にすることによって、最小化することができる。しかしながら、RP HPLCによる親水性検体の分離を行うために、高度に水性の移動相を必要とすることがよくある。このような条件下で操作した場合、常用のRPC18カラムは保持の可逆的な損失を示す。保持の損失の割合及び程度はカラムの相違によって大きく変化し得る。この作用の一般的な説明は、疎水性のアルキル鎖は湿潤性ではなく、シリカ表面に折りたたまれて親水性媒体に露出しないように見える、というものである。この折りたたまれた又はもつれた状態においては、保持の損失が生じる溶質との疎水性相互反応に対するアルキル鎖の表面積は小さくなっている。Reid, T.S.; Henry, R.A., American Laboratory 24-28(7月, 1999)。
初期に報告された材料は、2段階表面修飾によって製造された。Feibush, B. EP-A-0386926 (1990)。第1段階で、シリカ支持体をアミノプロピルシランと結合させた。第2段階で、このアミノ基を酸クロリドと反応させてアミド結合を形成した。この最初の製造方法には、二つの連続する反応について表面の再現性が低いという問題点がある。官能化した表面には、誘導化したアミノ基と誘導化していないアミノ基の混合物があった。従って、得られた相は未反応の塩基性部位が存在することにより望ましくないイオン交換反応を示した。
その後、材料は1段階表面修飾によって製造され、この方法では極性の官能基がシラン分子中に組み込まれた。Neue, U.D.; Niederlaender, C.L.; Perterson, J.S., EP-B-0579102 (1993)。シランとの1回の表面反応によって、アニオン交換作用がない唯一つのリガンド構造を得る。

本明細書はまず、本発明に従う極性相を有する修飾したシリカ支持体を説明する。次いで極性相を記載し、修飾したシリカ支持体の種々の製造方法を記載し、それは極性相の製造方法も含む。
修飾したシリカ支持体は以下の式1を有する:

R1=H、アルキル(例えばC1〜C30)、置換アルキル(例えばCH2OH)、アリール(例えばフェニル)又は置換アリール(例えばCH2Ph)であり;
R2=アルキル(例えばC1〜C30)、置換アルキル(例えばCH2OH)、アリール(例えばフェニル)又は置換アリール(例えばCH2フェニル)であり;かつ
R3及びR4は独立してアルキル、アリール、ヒドロキシル、又はアルコキシル、又はケイ素に結合した酸素を含む基である。
式(1)について、好ましい態様では、R1及び/又はR2は炭素鎖の長さがC1〜C30であるアルキル基である。RPクロマトグラフィー充填材として使用する場合、R1及びR2のアルキル炭素鎖は、修飾したシリカ支持体に逆相特性を与えるのに十分な長さであることが好ましい。適切な炭化水素鎖の長さは好ましくはC4〜C18の範囲であり、より好ましくはC10〜C16である。さらに、修飾したシリカ支持体は、イオン交換特性、リガンド交換特性、又は親水性を含むことができる。
好ましい態様において、Xはスルホニルである。スルホン類は以下の利点を提供する:
(1)高温で広いpH範囲における化学的安定性及び
(2)特異的な選択性。
置換基の典型的な数値は以下のとおりである:m=1〜3;p=1,2;n=0;R3及びR4=CH3;R1=C1〜C18;R2=C1〜C18。
本発明の修飾したシリカ支持体は極性単位としてアミノアルコールを含む。本発明の極性シラン試薬は、アミノアルコールのアミノ基とヒドロキシ基の異なる反応可能性を利用して、一端において炭化水素鎖により、他端においてシラン官能基によりアミノアルコールを誘導化する。次いで、極性部分を含むシランリガンドによってシリカ表面を修飾する。
修飾したシリカ支持体に埋め込まれた極性相は種々の極性埋め込みRP材料を含み、それにはエタノールアミド及びエタノールスルホンアミドRP相が含まれる。以下に十分に記載するように、好ましい態様では、極性相をシリカ支持体に埋め込む方法を1段階で行うことができ、この場合遊離のアミノ基は結合する前に消費される。この方法はシリカ表面上の残存アミノ基を回避するという利点を有し、該アミノ基は主の疎水性鎖が表面に結合する前にアミノ基が結合する場合に生じる可能性がある。

R3、R4及びR5は独立してアルキル、アリール又は反応性離脱基であり、かつR3、R4又はR5の少なくとも一つは離脱基である。
離脱基は当業者に公知の離脱基を含み、離脱してシリカ支持体上にヒドロキシル基を経由する共有結合を形成することができる基である。このような離脱基はヒドロキシル、アルコキシル、ハライド及びアミノ基又はこれらの基から誘導する基を含むことができる。

式中、X、R1、及びR2は上記の式(1)で定義されている。
1.アミノアルコールのアルキル化、例えばエタノールアミンのアルキルブロミドによるアルキル化。段階3aの第1の反応は、炭酸カリウムの存在下における優勢なN−アルキル化として記載されている。この代わりに、強塩基を使用してアミノアルコールの優勢なO−アルキル化によってR1を導入することもでき、該強塩基は反応しないアミノ基の存在下にヒドロキシル基を脱プロトン化する(示していない)。
2.不飽和部分の導入、例えばO−アルキル化によるアリル基の導入も、段階3aの第2の反応に示されている。O−アルキル化の代わりに、アミノ基を介して不飽和フラグメントを結合させることができる(示していない)。
3.アミノアルコールのモノ置換アミノ基をアシル化剤、スルホニル化剤、又はイソシアナートと反応させて、ヒドロシリル化(段階3c)のために段階3bで形成される前駆体を製造する。
4.段階3cで、白金触媒の存在下に、モノ−、ジ−又はトリアルコキシ、クロロ又はアミノシランによってヒドロシリル化を行って、段階3dのためのシラン修飾剤を生成する。
5.段階3dで、還流下にトルエンのような有機溶媒中で、該シラン修飾剤をシリカゲルに結合させる。
6.任意の段階3eにおいて、残存シラノール基がエンドキャッピングされる(例えば式2のような離脱基を有するアルキルシランとの反応により)。
N−置換エタノールスルホンアミジル及びエタノールアミジルアルキルエーテルを合成するために、第2のアミノアルコールを出発物質として使用することができる。この場合、反応系列の第1段階(3a)を回避し、R1及びR2置換基の鎖長を反応式(4)に応じて変化させることができ、これはリガンド構造に疎水性の鎖を導入する別の方法を示している。例えば、R1=CH3、かつR2=C18、C10又はC16置換基であることが適切である。

反応式(5)において、Xはカルボニル、スルホニル、カルバモイル、又はオキシカルボニルであり;R1及びR2=アルキル基、置換アルキル基、アリール基又は置換アリール基である。
本発明がさらに詳細に以下の例に記載されているが、本発明はこれに限定されない。
新規なN−置換カルバモイル、スルホンアミジル又はアミジルアリルエーテル(式1)を製造する一般的な手順
エタノールアミン(400mmol)を炭酸カリウム(20mmol)とトルエン(1000ml)中で混合した。次いで、得られた混合物を70℃で撹拌した。30分撹拌後、アルキルハライド(40mmol)を上記混合物にゆっくり添加した。反応混合物を70℃で24時間撹拌した。次いで、減圧下に全ての揮発物を除去し、残渣をEt2O/H2Oで抽出した。次いで、有機相をNa2SO4で乾燥し、減圧下に濃縮して対応するN−置換アミノ−エタノール1(反応式(3a))を得た。合成した化合物の構造は1H NMR分光分析で確認した。
THF(400ml)中のN−置換アミノ−エタノール1(20mmol)の溶液をTHF(400ml)中のNaH(22mmol)の懸濁液へ室温でゆっくり加えた。次いで、混合物を50℃で4時間撹拌し、引き続きTHF(50ml)中のアリルブロミド(24mmol)をゆっくり加えた。50℃で24時間撹拌後、減圧下に全ての揮発物を除去した。残渣をCH2Cl2とH2Oの混合物に溶解させた。有機相を水でさらに洗浄し、引き続きNa2SO4で乾燥した。有機相から揮発物を除去して、アリルエーテル2(反応式3a)を得た。合成した化合物の構造を1H NMR及び質量分光分析で確認した。
置換アリルエーテル2(15mmol)(反応式3b)をCH2Cl2(300ml)中のEt3N(30mmol)と混合し、0℃で20分間保持した。次いで、CH2Cl2(50ml)中の酸クロリド、スルホニルクロリド又はイソシアナート(15mmol)の溶液をゆっくり加え、反応混合物を周囲温度で12時間撹拌した。反応混合物を水で洗浄し、Na2SO4で乾燥した。減圧下に全ての揮発物を除去し、対応するN−置換極性アリルエーテル3(反応式3b)を得た。合成した化合物の構造を1H NMR及び質量分光分析で確認した。
ヒドロシリル化の一般的な手順
N−置換極性アリルエーテル3(10mmol)をモノ−、ジ−又はトリエトキシシランの混合物に添加した。次いで、触媒、例えば最小量のエタノール中のヘキサクロロ白金酸を添加した。50℃で24時間撹拌後、過剰のシラン及び溶媒を減圧下に除去して対応するN−置換極性シラン(表1)を得た。合成した化合物の構造を1H NMR分光分析で確認した。
シラノール活性
図1は、開発したエタノールスルホンアミドC16カラムと従来のC18カラム及び市販されている極性−埋め込み相A及びBを対比させたピリジン試験の比較クロマトグラムを示す。エタノールスルホンアミドC16カラムは優れた効率とピークの形を示している。試験条件:30℃、CH3CN/H2O=50/50、1ml/分、注入容量5μl、226nm。
アミトリプチリン試験
図2は、エタノールスルホンアミドC16及びエタノールアミドC18相上のアミトリプチリンを含む5個の検体の試験クロマトグラムを示す。両相とも優れたピークの形を示す。試験混合物は、ウラシル、トルエン、エチルベンゼン、アミトリプチリン及びキニザリンを含む。試験条件:30℃、20mM KH2PO4 80%MeOH、pH=7.0、ml/分、注入容量5μl、254nm。
極性試験
図3は、新規に開発したエタノールスルホンアミドC16及びエタノールアミドC18相の極性試験の結果を示す。両相は、酸性検体(フェノールとp−ブチル安息香酸)及び塩基性検体(ピリジンとN,N−ジメチルアニリン)について優れたピークの形を示す。試験条件:30℃、50mM KH2PO4(pH=3.2)/CH3CN=40/60、1ml/分、注入容量5μl、254nm。
相崩壊試験
図4は、プローブとしてアデニンを使用し、従来のRP C18相と比較した開発したエタノールスルホンアミドC16カラムの相崩壊試験の結果を示す。100%水性媒体に24時間さらした後で、従来のC18カラムは保持の損失及びカラム効率の低下という問題点を示す。新たに開発したスルホンアミドC16相は、100%水性緩衝液に200時間連続して曝す純粋な水性条件に対して免疫性である。
安定性試験
図5は、新たに開発したエタノールスルホンアミドC16相を使用する極度に過酷な条件下における安定性試験の結果を示す。試験条件:50℃、1%TFA(pH=1)CH3CN/水、50/50、連続流、1ml/分、注入容量5μl、254nm、検体:トルエン。トルエンに対する両者のk'値及び効率数は40リットル又は溶離液の追い出しの間変化しない。
極性選択性試験
図6は、従来のRP C18カラムと本発明のエタノールスルホンアミドC16及びエタノールアミドC18極性−埋め込み相の極性選択性試験の結果を示す。試験混合物は、ウラシル、ジメチルフタレート、ブチルパラベン、トルエン及びフェナンスレンの混合物である。エタノールスルホンアミドC16相は従来のRP C18材料に類似する選択性を示す一方、エタノールアミド相はトルエンとブチルパラベン検体について異なる順序の溶離を示す。試験条件:30℃、70%CH3CN、1ml/分、注入容量5μl、254nm。
本発明に従う埋め込み相を有する修飾したシリカ支持体は、シリカ表面上の極性基の遮蔽作用により、塩基性検体との相互反応が減少したことを示す。従って、シラノール活性は極端に低い。従来技術の極性−埋め込み相及び従来のC18相と比較すると、緩衝剤を含まない溶離液中のピリジンのピークは、最も低いテーリング因子及び最も高い効率性を新たに開発したエタノールスルホンアミドC16相において有する(図1)。アミトリプチリンのピークの形は、pH7でエタノールスルホンアミドカラム及びエタノールアミドカラムの両者において優れている(図2)。これらの二つの支持体は酸に対しても優れたピーク特性を与える(図3)。
さらに、シリカ支持体の修飾した固定相は、100%水性媒体中で使用した場合に優れたクロマトグラフィー特性を有する。100%の水に連続して200時間曝した後で、エタノールスルホンアミドC16は再現性のある結果を示し、一方、従来のRP C18カラムは24時間以内に崩壊した(図4)。
本発明の合成方法により、アミノアルコールを種々の極性官能基を含む新規な表面修飾剤に転換することが容易になる。本方法が多様であることから、特定のクロマトグラフィーの適用に適する種々の選択性を有する複数群の新規な極性−埋め込み相を得ることができる。エタノールスルホンアミドC16極性−埋め込み相が従来のRP C18カラムと類似する選択性を示す一方、エタノールアミドC18相が異なる順序の溶離を有することが図4に示されているように、明らかにされた。

- Xがスルホニルである、請求項1に記載の修飾したシリカ支持体。
- 修飾したシリカがシリカゲルである、請求項1に記載の修飾したシリカ支持体。
- シリカ表面のヒドロキシル基の少なくともいくつかが非極性及び/又は極性基によってエンドキャップされている、請求項1に記載の修飾したシリカ支持体。
