Description
ヒトモノクローナル抗体4E10、2F5、2G12及び3D6の作製、産生、及び特徴決定は、以前に記載されている(スザ(D’Souza)ら、J Infect Dis 1997;175:1056−1062;クナート(Kunert)ら、Biotechnol Bioeng 2000;67:97−103)。簡単に説明すると、抗体産生ハイブリドーマ(hybridomas)が、組み合わせポリエチレングリコール/電気融合法により作製された。無症候性HIV−1陽性ドナーに由来するPBMCが、マウスヒトヘテロミエローマ(heteromyeloma)細胞系CB−F7と融合させられた。ハイブリドーマ上清が、HIV特異的抗体産生に関してスクリーニングされ、陽性クローンが、ELISA、ウエスタンブロット及び免疫蛍光アッセイによりさらに分析された。安全な大量製造を可能にするため、そして2F5及び4E10のアイソタイプをIgG3からIgG1へ変化させるため、抗体は、IgG1(κ)としてチャイニーズハムスター卵巣細胞(CHO)において組換え発現させられた。後述の実施例1から7に記載された当研究には、2F5抗体及び4E10抗体の組換えCHO IgG1バージョンが使用された。
シンシチウム阻害アッセイ用のAA−2細胞系は、NIHエイズ・リージェント・リファレンス・プログラム(AIDS Reagent Reference Program)から入手された(M.ハーシュフィールド(Hershfield)により提供された)。細胞は、10%熱不活化FCS及び4mM L−グルタミンが補足されたRPMI−1640(Biochrom、Berlin、Germany)を含有している細胞培養培地(CCM)において週1回2回継代された。
HIV−1初代単離物92BR021/030、92RW009/021、92TH14/21/24及び92UG001/029/037は、HIVの単離及び特徴決定のためのWHOネットワーク(WHO Network for HIV Isolation and Characterization)から得られ、MRCエイズ・リージェント・プロジェクト(AIDS Reagent Project)により提供された。

4E10エピトープのマッピングのため、アナスペック社(AnaSpec,Incorporated)により製造されたHTLV−IIIMNの34個のオーバーラップしているgp41ペプチド(aa501から856、表1)を、エイズ・リサーチ・アンド・リファレンス・リージェント・プログラム(AIDS Research and Reference Reagent Program)より得た(番号2015から2049)。これらのペプチドの大部分が20アミノ酸長であり、連続ペプチド間には10アミノ酸のオーバーラップがあった。ペプチドGGGLELDKWASLは研究室内(in−hause)で合成された。
共焦点顕微鏡検査については、初代HIV−1ウイルス(S2/02、S2/04、S2/08)を感染させたPBMCを、37℃で1時間、スライド(Biorad、Munchen、Germany)に付着させた。同一ドナーの未刺激の未感染PBMC及び分裂促進因子により刺激された未感染PBMCを、陰性対照とした。スライドをPBSで洗浄し、20分間PBS中の5%スキムミルクによりブロッキングした。抗体4E10、2F5、2G12及びHIVIGを、100μg/mLの濃度で適用し、+4℃で1時間インキュベートした。細胞をPBS/スキムミルクで洗浄し、FITC標識ポリクローナルヤギ抗ヒトIgG(g)(Sigma、St.Louis、MO)と共に1時間インキュベートした。PBSによる徹底的な洗浄の後、細胞を20分間パラホルムアルデヒド(3%)で固定し、続いてPBSで洗浄した。蛍光シグナルを、バイオラド(Biorad)MRC600共焦点顕微鏡で可視化した。
シンシチウム阻害は、以前に記載されたように(パーチャー(Purtscher)ら、AIDS Res Hum Retroviruses 1994;10:1651−1658)、読み取りとしてシンシチウム形成を用いて、指標細胞系としてAA−2細胞を使用して査定した。簡単に説明すると、CCM(5μg/mL;Sigma、St.Louis、MO)を含有しているポリブレン中の抗体の段階希釈物を、37℃で1時間102から103TCID50/mLのウイルスと共にプレインキュベートした後、AA−2細胞を添加した。細胞を5日間インキュベートした後、シンシチウム形成を評価した。実験は、1希釈段階につき4から8個のレプリケート(replocates)を用いて実施した。1ウェル当たり少なくとも1個のシンシチウムの存在を、HIV−1感染の指標と見なした。50%阻害濃度(IC50)は、リード(Reed)及びミュンチ(Muench)、AM J Hygiene 1938;27:493−497の方法に従い計算した。実験は、2F5、2G12及び4E10を用いて実施した。
hu−mAb 4E10(=4E10−IgG1)、2F5(2F5−IgG1)、2G12及びIgG1b12の中和活性を、以前に記載されたようにして(パーチャー(Purtscher)ら、AIDS Res Hum Retroviruses 1994;10:1651−1658)、PBMCに基づく中和アッセイにおいて決定した。抗体の段階希釈物を、37℃で1時間ウイルスと共にプレインキュベートした後、PBMCを添加し、さらに7日間インキュベートした。実験は1希釈段階につき4個のレプリケートを用いて実施した。アッセイ終了時に、高感度p24−ELISA(ステインドル(Steindl)ら、J Immunol Methods 1998;217:143−151)により、ウイルス増殖を測定した。対照培養物におけるp24抗原産生に対するmAb含有培養物におけるp24抗原産生の比率を、インプットp24を考慮に入れて計算し、50%の阻害を引き起こすmAb濃度(μg/mL)を線形回帰分析により決定した。各アッセイには、実際のTCID50を決定するためのウイルス力価決定を含めた。ウイルス力価が102から103TCID50の間にある場合にテストを妥当と見なし、最大の複製はインプットp24より少なくとも5倍高いp24濃度をもたらした。
組み合わせられたmAbの相乗中和効果の計算は、チョウ−タラレイ(Chou−Talalay)(チョウ(Chou)TC及びタラレイ(Talalay)、Adv Enzyme Regul 1984:22:27−55)の方法に従って決定した。簡単に説明すると、50%の阻害を達成するために必要な単一mAbの量を、mAbの組み合わせを使用した場合に必要とされる濃度と比較した。組み合わせ指数(CI)を、方程式CI=(D1)/(Dx)1+(D2)/(Dx)2に基づき計算した。(Dx)1及び(Dx)2は、50%の中和を達成するために必要とされた単独のmAb1及びmAb2の濃度であり、(D1)及び(D2)は組み合わせて使用した場合のmAb1又はmAb2の用量である。CI<1は相乗作用を示し、CI=1は相加効果(additive)を示し、CI>1は拮抗(antagonism)を示す。
4E10の結合領域を決定するため、ELISAにより、三十四(34)個のオーバーラップしているgp41MNペプチドを、4E10との反応性に関してテストした。最小2F5 gp41−エピトープを含むペプチドGGGLELDKWASLを陰性対照として使用し、gp41MN及びgp160IIIBを陽性対照とした。
4E10結合特徴の免疫蛍光分析
以前に報告された(ブチャチャー(Buchacher)ら、前記参照)、4E10の、感染PBMCの表面上に存在するHIV−1抗原との相互作用、及び可能性のあるMHCクラスIIとの交差反応性を研究するため、本発明者らは、4E10の、未刺激HIV−1陰性PBMC、PHA−Pにより刺激されたHIV−1陰性PBMC、及びHIV−1感染PBMCとの結合研究を実施した。共焦点顕微鏡検査及びフローサイトメトリーを使用した間接免疫蛍光により、4E10の結合を、2F5、2G12及びHIVIGの結合と比較した。
T細胞系馴化(TCLA)HIV−1株によるシンシチウム阻害アッセイ
AA−2細胞におけるTCLA単離物HTLVIIIB、HTLVIIIRF、HTLVIIIMN及びcl82によるシンシチウム形成を阻害するヒトモノクローナル抗体4E10(=4E10−IgG1)の能力を、mAb 2F5及び2G12の対応する能力と比較した。MAb 2G12は、V1ループ、V2ループ及びV3ループ又はCD4結合部位に無関係なgp120上のコンフォメーション感受性グリコシル化依存性のエピトープを認識する(トルコラ(Trkola)ら、J Virol 1996;70:1100−1108)。4E10及び2F5は、4個のウイルス全てのシンシチウム形成を阻害したが、2G12はHTLV−IIIMNを中和しなかった(表2)。
ウイルスとmAbとのプレインキュベーションのシンシチウム阻害に対する影響
標的細胞へ添加する前にウイルスを抗体と共にプレインキュベートした場合の影響を決定するため、別法としてプレインキュベーション工程なしのアッセイを実施した。ウイルス(HTLVIIIRF)及びmAb(4E10、2F5、2G12)を、同時にAA−2細胞に添加した。4E10(−IgG1)及び2F5(−IgG1)については、プレインキュベーションを行った場合と行わなかった場合とで結果は異ならなかったが、2G12については、1時間のプレインキュベーション工程の後に300%という劇的な増加が観察された。
初代HIV−1単離物の中和
次に、PBMCに基づく中和アッセイにおいて、異なるクレードの初代単離物のパネルに対するmAb 4E10(−IgG1)の中和活性を、2F5、2G12、IgG1b12の対応する活性と比較した。
ハイブリドーマmAb 4E10−lgG3及びCHO組換えmAb 4E10−IgG1の中和活性の比較
PBMCに基づく中和アッセイを、初代HIV−1単離物を用いて実施した(実施例5を参照のこと)。
mAb 4E10及び2F5の組み合わせ適用
4E10及び2F5は近接したエピトープを認識するため、初代ウイルスの中和における2F5及び4E10の可能性のある阻害性相互作用を、2F5抗体又は4E10抗体のいずれかを含有している調製物を、2個の抗体の組み合わせ(1:1比率)を含有している調製物と比較することにより、5個の異なる初代ウイルス(S2/02、S2/03、S2/04、S2/06及びS2/08)に関して研究した。2F5(−IgG1)と4E10(−IgG1)との組み合わせは、全ての実験において、わずかに相乗的又は相加的な効果(CI≦1)をもたらし;いずれの実験においても、拮抗的な効果は検出されなかった。2G12及びIgG1b12も含めた、初代単離物P6/71を用いた付加的な実験の結果が、図4(A)、図4(B)及び図4(C)に示されている。この実験に関する組み合わせ指数(CI50)は、4E10と2F5との組み合わせで0.64、4E10と2G12との組み合わせで0.23、4E10とIgG1b12との組み合わせで0.87であり、2G12及びIgG1b12も、4E10と拮抗せず、相乗的ではないにしても相加的な効果を示すことを示している。
Claims (11)
- TCLA単離物HTVL IIIMNのgp41のaa672から677(NWFDIT)、NWFNIT,SWFGIT,TWFGIT,又は、NWFSITを有するアミノ酸配列と結合し、かつ、ECACC受託番号90091704のハイブリドーマから産生されるモノクローナル抗体2F5、及び/又は、ECACC受託番号93091517のハイブリドーマから産生されるモノクローナル抗体2G12による中和に対して非感受性のHIV−1変異体を中和する活性を有する、ECACC受託番号90091703のハイブリドーマから産生されるモノクローナル抗体4E10−IgG3以外のHIV−1中和モノクローナル抗体であって、
前記HIV中和モノクローナル抗体が、ECACC受託番号01110665のチャイニーズハムスター卵巣細胞(CHO)から産生されるモノクローナル抗体4E10−IgG1である
ことを特徴とするHIV−1中和モノクローナル抗体。 - 請求項1に記載の抗体を含有することを特徴とする、哺乳動物細胞のHIV−1感染の阻害又は防止に使用される医薬組成物。
- 前記医薬組成物が、請求項1に記載の抗体のほか、1つ以上の前記抗体とは別のHIV−1中和抗体を含有することを特徴とする請求項2に記載の医薬組成物。
- 前記医薬組成物が、ECACC受託番号90091704のハイブリドーマから産生されるモノクローナル抗体2F5、及び、ECACC受託番号93091517のハイブリドーマから産生されるモノクローナル抗体2G12のうち一方又は両方を含有することを特徴とする請求項2または3に記載の医薬組成物。
- HIV−1感染に対して使用される医薬組成物の製造用の、ECACC受託番号01110665のチャイニーズハムスター卵巣細胞(CHO)から産生されるモノクローナル抗体4E10−IgG1。
- 前記医薬組成物が、ECACC受託番号90091704のハイブリドーマから産生されるモノクローナル抗体2F5、及び、ECACC受託番号93091517のハイブリドーマから産生されるモノクローナル抗体2G12のうち一方又は両方による中和に対して非感受性のHIV−1株による哺乳動物細胞の感染の阻害又は防止に使用されるものであることを特徴とする請求項5に記載のECACC受託番号01110665のチャイニーズハムスター卵巣細胞(CHO)から産生されるモノクローナル抗体4E10−IgG1。
- モノクローナル抗体4E10−IgG1の4E10結合パラトープと反応性であるペプチドの誘発、または、スクリーニング用の、ECACC受託番号01110665のチャイニーズハムスター卵巣細胞(CHO)から産生されるモノクローナル抗体4E10−IgG1。
- 前記ペプチドが、抗イディオタイプ抗体であることを特徴とする請求項7に記載のECACC受託番号01110665のチャイニーズハムスター卵巣細胞(CHO)から産生されるモノクローナル抗体4E10−IgG1。
- 前記ペプチドが、TCLA単離物HTVL IIIMNのgp41のaa672から677(NWFDIT)、NWFNIT,SWFGIT,TWFGIT,又は、NWFSITを有するアミノ酸配列を含むHIV−1のgp41の断片を模倣するペプチドであることをと特徴とする請求項7または8に記載のECACC受託番号01110665のチャイニーズハムスター卵巣細胞(CHO)から産生されるモノクローナル抗体4E10−IgG1。
- 請求項1に記載のHIV−1中和モノクローナル抗体を含有することを特徴とする、HIV−1感染に対して哺乳動物レシピエントに能動免疫を与えるための医薬組成物。
- ECACC受託番号90091704のハイブリドーマから産生されるモノクローナル抗体2F5、及び、ECACC受託番号93091517のハイブリドーマから産生されるモノクローナル抗体2G12のうち一方又は両方である、請求項1に記 載のHIV−1中和モノクローナル抗体とは別のHIV−1中和モノクローナル抗体を含有することを特徴とする請求項10に記載の医薬組成物。
