Description
抽出溶剤にはアセトンあるいはヘキサン、アセトン−ヘキサン混合液、メタノールなどの有機溶剤を用いることが出来、活性マグネシアあるいは活性アルミナ等の吸着剤を用いてのカラム分離精製が可能である。展開溶剤にはアセトン、ヘキサン、メタノール等が用いることが出来る。
有色馬鈴薯「インカのめざめ」の塊茎を、スラーサー等を用いて2mm厚にスライスした直後に、該塊茎スライス片を直接2倍量程度の99.5容量%のエタノール溶液に投入し、約4℃下で12時間浸漬抽出した。
次に、該浸漬液を遠心分離することにより固形物を除去し、馬鈴薯カロチノイド系色素抽出液を得た。更に次いで、該馬鈴薯カロチノイド系色素抽出液を40℃以下の温度で減圧濃縮して、本発明のアポトーシス誘導物質を調製した。このようにして得られた本発明のアポトーシス誘導物質を実施例1とする。
ヒト胃癌細抱を10%牛胎児血清含有RPMI_1640培地で培養した。5×105/mlの密度になるようにヒト胃癌細胞を調整し、50%エタノール(対照群)、実施例1のアポトーシス誘導物質を50%エタノールに溶かした溶液(実験群)をそれぞれ添加し、37℃、95%−5%CO2の条件下で胃癌細胞を3日間培養した。3日間の培養後に胃癌細胞を遠心分離した後、上清を除去した後に残った細胞ペレットをPBS(−)で一回洗浄した。該細胞ペレットに細胞融解用バッファーを加え、細胞を融解させた。RNAse溶液を加え、50℃で2.5時間反応させてから、プロテアーゼK溶液を加え、50℃で2.5時間反応させてから、DNA断片を抽出した。DNA抽出液とゲルローディング液を混合して、2%アガロースゲル板のウェルに添加し、100Vで電気泳動を行った。ゲルを水に浸してから、UVトランスイルミネーターでエチジウムブロマイド蛍光を発するDNAを検出した。
発癌剤として、ベンゾ(α)ピレンのオリーブ溶液をマウスの胃へゾンデにて注入し、胃を前癌状態にした10匹のマウスを2群用意した。該前癌状態のマウスの1群に対して、有色馬鈴薯「インカのめざめ」の生塊茎を蒸かしたもの(以下、単に「インカのめざめ」という。)を餌として5ヶ月間与えた。実験開始から5ヶ月後、実験に供したマウスを屠殺し、マウス1個体当たりの胃の中に惹起された腫瘍の発生数と腫瘍量重を算出した。尚、上記前癌状態にした他のマウス群に対して、男爵いもを蒸かしたものを餌として5ヶ月間与えた。このマウス群を比較対照用とした。
バター6.0重量部、脱脂粉乳7.5重量部、水71.5重量部、実施例1のアポトーシス誘導物質のペースト5.0重量部、砂糖8.0重量部及び乳化安定剤0.7重量部の全成分を混合して80℃まで昇温し、ホモジナイザーで均一に乳化した後、翌日まで5℃でエージングする。次いで、0℃まで冷却した後、よくミキシングして癌予防効果が付与された乳飲料を得ることができる。

グミキャンディー還元麦芽糖水飴150重量部を加熱し、減圧下で水分約15w/w%に濃縮し、常法にしたがって、これにゼラチン13重量部を水18重量部に溶解したものと、実施例1のアポトーシス誘導物質のペースト2重量部、クエン酸2重量部および適量の香料を混合し、成形、包装して癌予防効果が付与されたグミキャンディーを得ることができる。
全卵15重量部、水85重量部、上白糖80重量部、食塩0.6重量部、実施例1のアポトーシス誘導物質のペースト3重量部を充分に混合する。次に、薄力粉100重量部、ベーキングパウダー3重量部、重曹1.5重量部、増粘剤1.5重量部を篩いに通し、混合する。最後に、ケーキ用ショートニング15重量部を加えて、泡立器で充分に攪拌混合する。この生地を焼成型に入れ、190℃で25分間焼成することにより、癌予防効果が付与されたパウンドケーキを得ることができる。
Claims (2)
- 肉部にカロチノイド系色素を含む馬鈴薯の品種であって、Solanum tuberosum ssp. tuberosam L.に比べ、生芋肉部の色調が黄色で測色計による色測においてL*、a*、b*表色系のb*が30以上であり、肉部の色調を形成する主成分であるカロチノイド系の黄色色素ゼアキサンチンの生芋肉部の濃度が3μg/g以上である馬鈴薯の品種から、50%以上のアルコールを抽出溶媒として用い、10℃以下で少なくとも15時間以上抽出して得られるゼアキサンチン及びルテインを主成分として構成されたカロチノイド系色素含有抽出物よりなることを特徴とする、
