本発明は、歯面での歯垢の形成及び付着を抑制する効果に優れた口腔用組成物に関する。

ムタステインは、不溶性グルカン合成酵素の働きを阻害することで歯垢の形成を抑制することが知られている。この働きを利用してう蝕を予防するためにムタステインを歯磨き剤等に添加して用いることが検討されている。

ところがムタステインだけでは歯垢の形成を抑制するには十分ではなく、デキストラナーゼ等の歯垢溶解物質と併用したり、S.mutansに対して免疫活性を有する抗体と用いたりすることが試されている。

しかしこれらは歯垢の形成を完全になくすものではないから、残った歯垢が徐々に蓄積していき虫歯や歯肉炎を誘発することがあるので、う蝕を予防するという点からすれば十分ではなく、さらに検討、改良することが必要であった。

【発明が解決しようとする課題】
本発明者らは、上記のような状況を鑑みて、ムタステインの歯垢の形成を抑制する働きを高めることを目的に検討をすすめる中で、この働きに加えて形成された歯垢の性状を歯面から剥離しやすいように変性させることができればう蝕を予防するのに有効であるとの考えに達した。
そこで本発明は、これら2つの働きを有し、う蝕の予防に有効となる製剤を開発して提供することを目的としたものである。

【課題を解決するための手段】
本発明者らは、上記課題を満足するため鋭意検討した結果、以下の手段によって課題を解決することを見い出した。
(1)歯垢を歯面から剥離しやすい性状に変性するために、0.01〜1重量%のムタステインと、0.01〜0.05重量%のカチオン性殺菌剤とを配合したことを特徴とする、口腔内洗浄用組成物。

本発明において、歯垢を剥離しやすい性状とは次のような状況になることをいう。すなわち、歯面に固着した歯垢(不溶性グルカン)は通常、強固に固着し、プレートを用いた洗浄試験では超音波洗浄機を用いなければとれない状態になるが、ムタステインとカチオン性殺菌剤を配合した製剤で処理すると、簡単な水洗で歯垢(不溶性グルカン)が剥離する状況になることである。このような状況になると、軽いブラッシングで歯垢が簡単に除去でき、ブラッシングの効果を高めることができる。

【発明の実施の形態】
本発明の口腔用組成物は、ムタステインを歯垢形成抑制剤とする口腔用組成物に、歯垢を剥離しやすい性状に変性し歯垢付着を抑制可能にするためにカチオン性殺菌剤を配合してなるものであり、カチオン性殺菌剤はその変性のために有効量配合すればよく、好ましくはムタステイン0.01〜1重量%に対して、カチオン性殺菌剤0.01〜0.05重量%、特に好ましくはムタステイン0.01〜0.5重量%に対して、カチオン性殺菌剤0.01〜0.05重量%である。

カチオン性殺菌剤としては、塩化セチルピリジニウム、塩化ベンゼトニウム、塩化ベンザルコニム、塩化デカリニウム、塩酸クロルヘキシジン、塩酸アルキルジアミノエチルグリシン等が挙げられ、これらの1種又は2種以上が使用できる。これらの中でも塩化セチルピリジニウムが本発明の効果が高く好ましい。
そして好適には、ムタステイン0.01〜0.2重量%に対して、塩化セチルピリジニウム0.01〜0.05重量%とする態様が挙げられる。

本発明の口腔用組成物は、各種製剤の歯磨剤、洗口剤とすることができ、特に液体歯磨、洗口剤とするのがよい。

そして液体歯磨、洗口剤とするには、水(精製水、イオン水等)及びエタノール等を溶媒として常法によって調製すればよく、また緩衝剤を用いてpHを5〜9、好ましくは5.5〜8.5とすると使用感がよくなる。

なおエタノールとしては、通常のエタノールの他に、ブルシン、ゲラニオール、香料等の各種変性剤で変性されたエタノールベースの変性アルコールを用いてもよい。

また添加される各種成分の溶解、分散及び可溶化をスムーズにするために、非イオン性界面活性剤を用いるとよく、例えば、グリセリン脂肪酸エステル、ソルビタン脂肪酸エステル、蔗糖脂肪酸エステル、ポリオキシエチレンアルキルエーテル、ポリオキシエチレンアルキルフェニルエーテル、ポリオキシエチレンポリオキシプロピレングリコール、ポリエチレングリコール脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、ポリオキシエチレン硬化ヒマシ油等が例示できる。

以下に実施例において本発明を具体的に説明するが、これらの実施例に限定されるものではない。

実施例1不溶性グルカンについての検討(1)粗グルコシルトランスフェラーゼの調製 Streptococcus.sobrinus OMZ176株を250mlのBHI(ブレイン・ハート・インフュージョン)培地を用いて37℃条件下で18時間培養後、この培養液を遠心分離(6000G,20分,4℃)して上清を得た。この上清を濾紙で濾過し、4Nの水酸化ナトリウムでpH7に調整し、50%飽和硫安を用いて、4℃条件下で1晩塩析した。
そして得られた塩析液を遠心分離(12000G,60分,4℃)し、その沈澱の硫安分画を0.05Mのリン酸カルシウム緩衝液(pH6.5)に溶解し、同緩衝液に対して、4℃で一昼夜透析した。この透析液を遠心分離(10000G,15分,4℃)し、不溶性物質を除去した後、0.05Mのリン酸カリウム緩衝液(PH6.5)で15mlにフィルアップしたものを粗グルコシルトランスフェラーゼとした。
