【従来の技術】
モノクローナル抗体は極めて広範囲に用いられており、基礎研究から医薬品にまで応用されている。モノクローナル抗体は、抗原を免疫したマウスの脾細胞とミエローマ細胞とを融合してハイブリドーマ細胞を作製し、その培養上清から精製することにより、一般的に得られている。ハイブリドーマ細胞の樹立に際して、アフィニティの高い優れた抗体を産生する株を選択するため、各々のクローンにより産生されるモノクローナル抗体をアッセイすることによりスクリーニングする必要がある。

従来の方法としてはELISA法が多く使われてきた。この手法はすでに確立されているが、操作は煩雑であり、抗体の濃度を変えて、多くのサンプル評価するとなると、測定数が極めて多くなり、非常に手間がかかっていた。

一方、金属基板表面に固定した抗体を用い、ラベル不要な分析技術である表面プラズモン共鳴(SPR)法、エリプソメトリ法によって抗原抗体反応を測定する方法は開示されており既知である(例えば、特許文献1および2参照)。しかし、これらの開示は、個々の抗原抗体反応に関してSPR法が用いられることを示しているのみであり、抗原に対してアフィニティの高い抗体をどのようにして効率良く選び出すかは示されていない。すなわち、本発明にあるような、抗原が同一である抗体を複数種類用意し、その抗体を金属基板上に固定化したアレイを使って抗体のスクリーニングを行った例は開示されていない。本発明は金属基板上に作製した抗体のアレイを用いて、抗体をスクリーニングする方法を開示する。

【発明が解決しようとする課題】
本発明の課題は、抗体を金属基板上に固定化したアレイを用い、SPR法によってスクリーニングしてアフィニティの高い抗体を効率よく選び出すことにあり、この技術はモノクローナル抗体を産出するハイブリドーマ株を樹立する際などに利用することが出来る。

【課題を解決するための手段】
本発明者らは鋭意検討した結果、以下に示す手段により、上記課題を解決できることを見出した。
1.抗原となる分子が同一である複数の抗体のアミノ基を、金薄層に覆われた平面ガラス基板上の官能基と反応させて該抗体を表面に固定化し、残存する該官能基にアミノ基末端ポリエチレングリコールを反応させブロッキング反応を行ったアレイを用い、
抗原を1μg/mlから8μg/mlまで濃度(C)を段階的に倍増させていき、各々の濃度における平衡時のシグナル(Req)を表面プラズモン共鳴イメージング法によって測定し、
Req/CとCをプロット(スキャッチャ−ドプロット法)した直線の傾きから抗原抗体反応の結合平衡定数を算出し、
アフィニティの高い抗体を選び出すことを特徴とする抗体のスクリーニング方法。
2.抗体がモノクローナル抗体もしくは、複数のモノクローナル抗体の混合物であることを特徴とする1に記載の抗体のスクリーニング方法。
3.アミノ基末端ポリエチレングリコールが、分子量2,000のアミノ基末端ポリエチレングリコールであることを特徴とする1又は2に記載の抗体のスクリーニング方法。
4.官能基がスクシンイミド基であり、アミンカップリング法により、抗体を表面に固定化することを特徴とする1〜3のいずれかに記載の抗体のスクリーニング方法。
5.金薄層に覆われた平面ガラス基板上に以下(i)〜(iv)の方法によりスクシンイミド基を導入すること特徴とする4に記載の抗体のスクリーニング方法。
(i)金薄層に覆われた平面ガラス基板上全体に、末端にチオール基を有するポリエチレングリコール(PEGチオール)を結合させる工程
(ii)フォトマスクを載せ、超高圧水銀ランプで照射し、照射された部分のPEGチオールを除去する工程

【発明の実施の形態】
以下に本発明を詳細に説明する。本発明は、複数の抗体を同一金属基板上に固定化したアレイを用いて抗体のスクリーニングを行う方法を開示している。

本発明において、スクリーニングの対象となる抗体は複数であり、抗原となる分子は同一である。ただし、エピトープは単一とも同一とも限定されない。すなわち、抗体はモノクローナル抗体であってもポリクローナル抗体であってもよく、エピトープの異なるモノクローナル抗体の組み合わせであってもよい。組み合わせ/配合率が異なる場合も、アフィニティが異なるため、異なる抗体として扱う。複数の抗体は一つの金属基板に固定化され、アレイが形成される。

スクリーニングの際の測定には、抗原を含んだ溶液が基板上に曝露され、抗原抗体反応によって、抗原がトラップされ、このトラップされた状態を観察する。
表面に抗原が吸着するのを観察する手段としてはSPR法が好ましい。SPRはラベルフリーの相互作用解析方法であり、抗原を蛍光物質や放射線同位体でラベルする必要がないため好ましい。また、リアルタイムでSPRシグナルを観察できるため、相互作用が飽和するまで抗原を含む溶液をセンサー表面に流し続けることができる。また、半定量的な検出方法であるため、シグナルの変化が吸着した量に対応するため、Kinetics解析が可能であり好ましい。
Kinetics解析によって抗原抗体反応の平衡定数が得られ、アフィニティの高い抗体を選ぶことが可能となる。

一度に行えるスクリーニングの件数は、SPRで観察できる数、すなわちアレイのスポット数が上限である。アレイのスポット数としては、好ましくは6個以上、より好ましくは12個以上、さらに好ましくは20個以上である。また、スポット数の上限としては観察が行える限り限定はされないが、概ね10000個である。
これらのアレイに複数種、好ましくは4種以上、さらに好ましくは6種以上のの抗体をスポットする。スポットは抗体1種に対して1箇所でも良いし、複数箇所にスポットしてデータのn数を増やすことも好ましい。

金属基板の金属としては金が好ましい。金はSPRによる観察が可能であることと、金−硫黄結合によって、表面に官能基を導入するのが容易であるからである。金属基板としては金薄層にコーティングされた平面ガラス基板であることが好ましい。ガラスはさまざまな屈折率を有するものが揃っており、切り出すことで容易にスライドを得ることができるため好ましい。また、平面基板である方が成形は容易かつアレイ作製が容易であり好ましい。

SPR法としては、広範囲にSPRシグナル変化を観察することのできるSPRイメージング法が好ましい。SPRイメージング法は偏光とした光束が金属薄層をコーティングしたガラス基板に照射され、その反射像を波長フィルターで通した上で、CCDカメラ等で撮影する方法である。

こうして抗原抗体反応が観察され、そのKinetics値はSPRシグナル変化から解析される。アフィニティの低い相互作用の場合、相互作用させた後に、緩衝液を流すことで解離が観察され、解離の速度から解離速度定数が算出できる。しかし、抗原抗体反応の場合、アフィニティが高く、解離を解析するのは困難である。そこで、平衡定数はスキャッチャードプロット法によって計算されるのが好ましい。この方法においては、濃度(C)を段階的に上昇させていき、その濃度におけるシグナルの平衡値Reqを求める。Req/CとReqとプロットし、その傾きから平衡定数が求まる。

抗体を固定化する手段としては、さまざまな手段が挙げられる。表面のカルボキシル基を水溶性カルボジイミドとN−ヒドロキシスクシンイミドによって活性化し、抗体中のアミノ基と反応させるアミノカップリング法が容易であり、好ましい。また、抗体をペプシン消化によってF(ab’)2とし、還元することでFab’として、チオールカップリングにより、表面に形成したマレイミド基、ジスルフィド基と反応させることができる。また、抗体に含まれる糖鎖を用いて、固定化する手段も考えられる。

以下に実施例を示して本発明を具体的に説明するが、本発明は実施例に限定されるものではない。

[実施例]
末端にチオール基を有するポリエチレングリコール(PEGチオール:日本油脂製SUNBRIGHT MESH−50H)を1mMの濃度で7mlのエタノール:水=6:1の混合溶液に溶解させた。PEGチオールの分子量は5,000であり、親水性が非常に高い。また、PEGの末端はメトキシ基であり、反応性をほとんど有さない。

18mm四方、1mm厚のLak10ガラススライドにクロムを3nm蒸着し、金を45nm蒸着した金蒸着スライドを、上記PEGチオール溶液に3時間浸漬させ、金基板全体にPEGチオールを結合させた。

スライドをミリQ水とエタノールで洗浄し、空気噴霧により乾燥させたのち、このスライドの上に図1に示すフォトマスクを載せ、500W超高圧水銀ランプ(ウシオ電機製)で2時間照射し、照射された部分のPEGチオールを除去した。フォトマスクのパターンは500μm×500μmの四角形が96個並んだものである。

スライドを水洗後、空気噴霧により乾燥させ、自動スポッターに装着した。13種類の抗ヤギIgGモノクローナル抗体を100μg/mlの濃度に調整し(PBS(−))、10μlを96穴プレートのA1〜12、B1のウェルに用意した。自動スポッターの内部湿度を81±2%、温度25℃に保ち、n=4でスポッティングを行った。湿度はアズワン社NT−1800デジタル温湿度計をチップ置場横に置いて測定した。

スポッティングのパターンを図2に示す。このように、96穴プレートに入れたサンプルの入れ方を反映した形でスポッティングを行った。B2〜12のウェルにはモノクローナル抗体サンプルは入っていないため、ブランクのスポットとなる。

スポッティングに用いたピンの先端の図を図3に示す。ピンの直径は487μmであり、200μmの幅の溝が入っている。従って、面積は0.09mm2である。また、ピンが取り付けられたヘッド部分の内部構造の概略図を図4に示す。ピンはガイドを通して内部のピン固定治具に取り付けられており、ピン固定治具は上下方向に移動可能となっている。ピン固定治具の上には緩衝材として軟質ウレタンフォームが設けられ、ピンが基板に押し付けられた時には、ピンが上方に移動し、緩衝材が圧縮され、衝撃を緩和すると共にピンに適度な応力を与える。

スポッティングは自動的に行われる。図5に使用した自動スポッターの内部の配置の概略を示す。ピンは96穴プレートに浸漬され、ウェル内のサンプルを保持し、スライド上へと移動し、スポッティングを行う。実施例の場合はn=4であるため、96穴プレートとスライドの間を3回往復する。4回目のスポッティングが終わった段階でピンは洗浄浴へと移動し、ミリQ水で2回洗浄される。洗浄後にドライヤー内にて上下運動が30mmの振り幅で3回行われ、ピンが乾燥される。次のウェルにてサンプルと採取し、同様の動作は付属したコンピュータで制御されて自動的に行われる。
湿度の調整は超音波により発生した霧をスポッター内に吹き込む方式とし、内部湿度はチップ置き場周辺での湿度を測定し、このデータを元に超音波のON/OFFを切り替えて調整した。

スポッティング終了後、スポッターからスライドを取り出し、直ちに加湿チャンバー内にスライドを移し、2時間保持してスライド上の官能基と抗体のアミノ基を反応させ、抗体を表面に固定化した。加湿チャンバーは、容器の底部に水を張り、中段にスライドを置ける構造となったものであり、密閉が可能であるため、長時間乾燥させることなく反応の進行を続けることが出来る。内部の湿度はほば100%である。
反応終了後のチップをミリQ水で数回洗浄した上で、空気噴霧により水を取り除き、分子量2,000のアミノ基末端ポリエチレングリコール水溶液を2mg/mlの濃度でpH8.5に調製し、300μlをスライド上に注ぎ、残存するスクシンイミド基にポリエチレングリコールを反応させ、ブロッキング反応を行った。表面固定化の反応スキームを図5に示す。

抗体を固定化したスライドを3回水洗した後、空気噴霧により乾燥させ、SPRイメージング装置(東洋紡績製)にセットした。緩衝液としてPBS(−)を250μl/mlの流速で30分通液し、100μl/mlの流速で30分通液して安定化させた。その際のSPRイメージング像を図7に示す。白くなっているスポット部分には抗体が固定化されており、その他の部分には固定化されていない。ピンが接触してサンプル液がスポットされた部分には全く傷は見当たらなかった。

次に抗原であるヤギIgGを1μg/mlから8μg/mlまで濃度(C)を段階的に倍増させて流し、各々の濃度における平衡時のシグナル(Req)を得た。この際のSPRシグナルの変化を図8に示す。

Req/CとCをプロットした(スキャッチャードプロット)結果を図9に示す。いずれもほぼ直線関係が得られている。図9の直線の傾きから結合平衡定数を算出し、表1に示す。ただし、シグナルが低いために相関係数が0.8未満となったデータは省いている。
このように金属基板上に抗体を固定化し、アフィニティの高い抗体を選び出すことができた。

【発明の効果】
本発明により、簡便に抗体をスクリーニングして、アフィニティの高い抗体を選択することが可能である。同一の抗原に対する複数の抗体は金属基板上に固定されてアレイが作製され、表面プラズモン共鳴法によってラベルフリーに抗原への結合挙動が観察され、平衡定数が容易に得られる。
