Description
眼中グルコースセンサの構成
ポリエチレングリコールヒドロゲルの構造的に安定なポリマー(PEGH、Shearwater Polymers社)を使用して眼中グルコースセンサを構成した。PEGHを眼中レンズ(Alcon Laboratories、円周6mm、厚さ1mm)中に固定化した。Ballerstadt & Schultz, Anal. Chim. Acta 345, 203-12, 1997及びRussell & Pishko, Anal. Chem. 71, 3126-32, 1999によって記載されているように、化学的に固定化されたペンダントテトラメチルローダミンイソチオシアネートコンカナバリンA(TRITC−ConA, Sigma)を受容体部分としてPEGHに組み込み、フルオレセインイソチオシアネートデキストラン(FITC−デキストラン、Sigma)を競合物質部分としてUV光下での重合によって組み込んだ。FITC−デキストランはTRIT−ConAに結合したが、FITC蛍光は、蛍光共鳴エネルギー移動によって消光された。グルコース濃度の増大がFITC−デキストランを遊離させ、グルコース濃度に比例する蛍光を生じさせた。
眼中グルコースセンサの生体内埋め込み
例1に記載の眼中レンズグルコースセンサを、血中グルコース濃度112mg%の生きたニュージーランドラビットの前房に埋め込んだ。眼の中でインプラントは緑の蛍光(518nm)を示す明るい点として見えた。埋め込み後6ヶ月で、生体顕微鏡スリットランプを用いた入念な検査は、毒性、拒絶又はいかなる反応の徴候をも示さなかった。
図1に示すスキームにしたがって、フルオレセイン(517nm)とローダミン(570nm)との双方からの蛍光を計測するための装置を構成した。装置は、フルオレセイン用1個及びローダミン用1個の2個の励起光源を有するものであった。2個の光電子増倍管を使用して二つの波長(517nm及び570nm)における信号の感知を最大にした。ウサギ眼に埋め込んだカプセル封入眼中グルコースセンサからの蛍光の検出を試験した。加えて、溶液中でセンサ(ローダミン−コンカナバリンに結合したフルオレセイン−デキストラン)へのグルコース添加(0〜500mg/dl)に応答する蛍光の変化を装置によってモニタし、従来の実験用走査型蛍光光度計(SpexFluorolog)を使用して得られたデータと比較した。
Claims (7)
- 眼中及び/又は血中グルコースレベルを計測するためのキットであって、
(a)使用者の眼(1)に角膜の外側から光を照射して眼中グルコースセンサを励起するための照射手段(10)であって、前記眼中グルコースセンサが、眼性流体と接触しており、前記照射手段(10)による照射を受けると第一及び第二の波長帯域を有する全蛍光を発することができる照射手段(10)と、
(b)両帯域を有する前記全蛍光を、前記第一の波長帯域を有する第一の蛍光と、前記第二の波長帯域を有する第二の蛍光とに分割するための光路分割手段(11)であって、前記第一の蛍光が第一の光路に沿って移動し、前記第二の蛍光が第二の光路に沿って移動する光路分割手段(11)と、
(c)第一の蛍光の強度を第一の波長で検出するための、第一の光路中に位置する第一の検出手段(14)と、
(d)第二の蛍光の強度を第二の波長で検出するための、第二の光路中に位置する第二の検出手段(17)と、
(e)第一の蛍光と第二の蛍光との強度の比を計算し、計算した強度の比に基づいて、使用者の眼性流体中の眼中グルコース濃度を、所定の較正表又は較正曲線にしたがって決定するための計算手段と、
(f)血中グルコース濃度と眼中グルコース濃度との所定の相関関係を参照することにより、計算手段によって決定された眼中グルコース濃度を血中グルコース濃度に変換するための算術手段と、
(g)使用者の眼(1)に照射された光の位置を観察するための瞳孔整合ビューワと、
を含むキット。 - 算術手段によって決定された血中グルコース濃度を視覚的に示すための表示手段をさらに含む、請求項1記載のキット。
- 信号をポンプに伝送するための伝送手段をさらに含み、前記信号は、ポンプが所定量のインスリンを使用者の組織に注入するための指示を含む、請求項1又は請求項2記載のキット。
- 前記照射手段(10)が、異なる第三及び第四の波長の光を発し、ここで第三及び第四の波長帯域はそれぞれ異なり、第一及び第二の波長帯域と異なる、請求項1〜3のいずれか1項記載のキット。
- 請求項1〜4のいずれか1項記載のキットと、異なる既知のグルコース濃度の少なくとも3種の溶液とを含む、請求項1〜4のいずれか1項記載のキットを較正するためのキット。
- 前記既知のグルコース濃度が0〜500mg/lの範囲の濃度で均一に分布している、請求項5記載のキット。
- 光を眼(1)から瞳孔整合ビューワ(21)へ向けるための偏光子(3)をさらに含む、請求項1〜6のいずれか1項記載のキット。
