【発明の属する技術分野】
本発明は、不斉エポキシ化反応用触媒等の不斉酸化反応用触媒として有用な、アミノ酸を主骨格とした新規な光学活性ヒドロキサム酸配位子に関する。

【従来の技術】
エナンチオ選択性合成に用いる新規で効果的なキラル触媒の開発は、合成有機化学において最近特に注目されている(Comprehensive Asymmetric Catalysis I-III, 1999)。ヒドロキサム酸は、金属イオンに対する優れたリガンドであり、金属イオン検出の指標として用いられている(Chem. Rev. 33, 209, 1943)。1977年には、Sharplessらによって、ヒドロキサム酸が酸化に対して非常に抵抗力が高く、モリブデニルイオン及びバナジウムイオンに対して優れた結合性を有すると考えられることが報告された。この報告によると、VO(acac)2及びキラルヒロドキサム酸存在下でアリルアルコールの不斉エポキシ化が行われている(J. Am. Chem. Soc. 99, 1990, 1977、Aldrichim. Acta 12, 63,1979、CHEMTECH 15, 692, 1985、Angew. Chem. Int. Ed. Engl. 34, 1059, 1995、)。しかし、この方法による動的リガンド交換方法や、リガンド減速によって、この種の触媒は実用化しうるレベルにまで達することがなかった(Chem. Ber./Recueil 130, 887, 1997、Synlett 899, 2000)。本発明者らは最近、キラルビナフチル修飾ヒドロキサム酸を用いたバナジウム触媒によるアリルアルコールの不斉エポキシ化を報告(Org. Chem. 64, 338, 1999)し、バナジウムの酸化開始時における状態、ヒドロキサム酸の配位能、並びに金属結合部位及び酸化体間におけるπ作用若しくは立体反発等の、速度及びエナンチオ選択性の増加にキラルバナジウム錯体の特性の幾つかが重要な役割を果たしていることを明らかにした。

他方、コンビナトリアル・ケミストリーおよびその関連技術、周辺技術は、実験実施を超高速化することで高機能物質・目的機能物質の探索や構造最適化を実現する新たな方法論として1990年代中期より急速な発展を見せている。その根幹は目的脳物質の「候補群」の構造や調整方法をいくつかのユニットの「組み合わせ(コンビネーション)」から構築するように予め設計・設定することで、多数候補化合物群を迅速調整し、それらの機能を効率よく評価することにある。これにより従来に比較し3〜4桁以上の試行錯誤的検討を実施しようというものである。このようなコンビナトリアル・ケミストリーを利用した均一系触媒開発も知られている(化学工業 2000年2月号59〜65頁)。

【発明が解決しようとする課題】
本発明の課題は、不斉エポキシ化反応用触媒等の不斉酸化反応用触媒として有用な新規な光学活性ヒドロキサム酸配位子、特にアミノ酸を主骨格とした新規な光学活性ヒドロキサム酸配位子を提供することにある。

【課題を解決するための手段】
香月−Sharpless酸化は光学活性化合物を合成する有力な方法の一つである。本発明者らは光学活性ヒドロキサム酸ライブラリーを調製し、それらバナジウム錯体による不斉エポキシ化反応を検討した本発明者らによる前記キラルビナフチル修飾ヒドロキサム酸を用いたバナジウム触媒によるアリルアルコールの不斉エポキシ化方法を改良するべく鋭意研究し、コンビナトリアル及び関連技術(Curr. Opin. Chem. BIol. 3, 313, 1999、Comprehensive Asymmetric catalysis I-III 3, 1389)を用い、効果的な不斉エポキシ化用バナジウム触媒の同定を試み、ヒドロキサム酸耐性(hydroxamic acid-bearing)ビナフチル基を、リガンドライブラリー合成に適した新規α−アミノ酸ベースヒドロキサム酸に再構築し、非常に単純な構造を有するキラルα−アミノ酸ベースヒドロキサム酸リガンドが、アリルアルコール不斉エポキシ化において効率的な触媒であることを見い出し、本発明を完成するに至った。

[式(I)中、R1は、フェニル基、インドリル基、フタルイミド基又はメチルチオ基を置換基として有していてもよいC1〜6のアルキル基を示し、R2及びR3は、隣接する窒素原子と共に環状イミド基を形成する基であり、該基としては、フタロイル基、4−メチルフタロイル基、テトラフルオロフタロイル基、テトラクロロフタロイル基、2,3−ジメチルマロニル基、2,3−ジフェニルマロニル基、2,3−ナフタレンジカルボニル基、1,8−ナフタレンジカルボニル基、4−ブロモ−1,8−ナフタレンジカルボニル基又は2,3−(9H、10H−アントラ−9,10−ニル)マロニル基を示し、R4は、アリール基がC1〜6のアルキル基若しくはC1〜6のアルコキシ基で置換されていてもよいフェニル基若しくはナフチル基であるジアリールメチル基、フルオレニル基又は2,2’−エチレンジフェニルメチル基を示す。]で表される光学活性ヒドロキサム酸(請求項1)や、置換基を有していてもよいC1〜6のアルキル基が、メチル基、エチル基、イソプロピル基、イソブチル基、 sec −ブチル基、 tert −ブチル基、ベンジル基、3−インドリルメチル基、4−フタルイミドブチル基、2−メチルチオエチル基であることを特徴とする請求項1記載の光学活性ヒドロキサム酸(請求項2)や、R2及びR3が、隣接する窒素原子と共に環状イミド基を形成する基が、2,3−ジフェニルマロニル基、又は1,8−ナフタレンジカルボニル基であることを特徴とする請求項1又は2記載の光学活性ヒドロキサム酸(請求項3)や、ジアリールメチル基が、ジフェニルメチル基、ビス(o−トリル)メチル基、ビス(o−イソプロピルフェニル)メチル基、ビス(3,5−ジ− ( t−ブチル ) フェニル)メチル基、ジ(ナフト−1−イル)メチル基又はビス(2−メトキシナフト−1−イル)メチル基であることを特徴とする請求項1〜3のいずれか記載の光学活性ヒドロキサム酸(請求項4)や、下記式(II)

【発明の実施の形態】
本発明の一般式(I)で表される光学活性ヒドロキサム酸における、R1は官能基を含まない置換基を示し、R2及びR3は単独又は協働して窒素上の保護基を示し、R4は官能基を含まない置換基を示す。ここで、官能基を含まない置換基とは、不斉エポキシ化反応を阻害する官能基、例えばスルフヒドリル基(−SH)アミノ基(−NH2)、カルボキシル基(−COOH)等の官能基を含まない置換基をいい、かかる官能基を含まない置換基としては、水素原子、置換基を有してもよいアルキル基、置換基を有してもよいアルコキシ基、置換基を有してもよいアリール基、置換基を有してもよいアラルキル基、置換基を有してもよいイミダゾールやインドールなどの複素環を有する基等を挙げることができ、またこれら化合物における置換基としては、アルキル基、ハロアルキル基、ヒドロキシル基、アルコキシ基、カルボキシル基、アルコキシカルボニル基、アシル基、ニトロ基、シアノ基、アミノ基、アリール基、ハロゲン原子、アジド基、カルボニル基、ニトリル基、等を例示することができる。

上記一般式(I)におけるR1としては、次式に示されるアラニン(1a)、2−アミノ酪酸(1b)、バリン(1c)、イソロイシン(1d)、ロイシン(1e)、tert-ロイシン(1f)、フェニルアラニン(1g)、トリプトファン(1h)、リジン(1i)、メチオニン(1j)等の各種アミノ酸の側鎖であるメチル基、エチル基、nor-プロピル基、iso-プロピル基、nor-ブチル基、sec-ブチル基、tert-ブチル基等の置換基を有してもよいのアルキル基の他、メトキシ基、エトキシ基、プロポキシ基等のアルコキシ基や、ベンジル基、フェネチル基等の置換基を有してもよいアラルキル基を具体的に例示することができるが、これらの中でも、不斉エポキシ化反応における高いエナンチオ選択性を付与しうるt−ブチル基、ベンジル基等が好ましい。

上記一般式(I)におけるR2及びR3としては、同一又は異なって、アルキル基、アリール基、シクロアルキル基、ヒドロキシル基、アルコキシ基、カルボキシル基、アルコキシカルボニル基、アシル基、イミン基、スルホンアミド基、カルバメート基等を示す他、R2及びR3は互いに結合して隣接する窒素原子と共に芳香族性若しくは非芳香族性の環状基を形成してもよい。これら環状基としては、他の縮合環又は置換基を有してもよい、隣接する窒素原子と共に環状イミド基を形成する基を好適に例示することができる。隣接する窒素原子と共に環状イミド基を形成する基としては、具体的に、次式に示されるフタロイル基、4−メチルフタロイル基、テトラフルオロフタロイル基、テトラクロロフタロイル基、2,3−ジメチルマロニル基、2,3−ジフェニルマロニル基、2,3−ナフタレンジカルボニル基、1,8−ナフタレンジカルボニル基、4−ブロモ−1,8−ナフタレンジカルボニル基、2,3−(9H、10H−アントラ−9,10−ニル)マロニル基等を例示することができ、これらの中でも不斉エポキシ化反応における高いエナンチオ選択性を付与しうる2,3−ジフェニルマロニル基、1,8−ナフタレンジカルボニル基等が好ましい。

上記一般式(I)におけるR4としては、メチル基、エチル基、nor-プロピル基、iso-プロピル基、nor-ブチル基、sec-ブチル基、tert-ブチル基等のアルキル基や、次式に示されるジフェニルメチル基、ビス(o−トリル)メチル基、ビス(o−イソプロピルフェニル)メチル基、ビス(3,5−ジ−(t−ブチル)フェニル)メチル基、ジ(ナフト−1−イル)メチル基、2,2′−エチレンジフェニルメチル基、ビス(2−メトキシナフト−1−イル)メチル基等の置換基を有してもよいアルキル基や、メトキシ基、エトキシ基、プロポキシ基等のC1〜6のアルコキシ基や、ベンジル基、フェネチル基等の置換基を有してもよいアラルキル基を例示することができるが、これらの中でも、不斉エポキシ化反応における高いエナンチオ選択性を付与しうる次式で示されるジアリールメチル基、特にオルト位に置換基を有するビス(o−トリル)メチル基、ビス(2−メトキシナフト−1−イル)メチル基、ビス(o−イソプロピルフェニル)メチル基や、ジ(ナフト−1−イル)メチル基等が好ましい。

本発明のアミノ酸を主骨格とするヒドロキサム酸は有用な不斉配位子であり、この不斉配位子はアミノ酸(光学活性化合物)、アミノ酸の窒素上の保護基(イミド型等の保護基)、そしてヒドロキシルアミン部分の微調整可能な三部分に分割され、それぞれを変化させることにより多数の類似した配位子を容易に合成することができるという、不斉配位子として重要な要素である柔軟性を兼ね備えている。例えば、コンビナトリアル手法を用いてバナジウム触媒を用いるアリルアルコールの不斉エポキシ化反応等に最適なヒドロキサム酸配位子を探索することができ、触媒活性が高く、高いエナンチオ選択性ででアリルアルコールを不斉エポキシ化する光学活性ヒドロキサム酸を同定することができる。

不斉配位子の効果をみるため、次式に示されたようなアリルアルコールのバナジウム触媒を用いる不斉エポキシ化反応に応用し、エナンチオ選択性の評価から最適な配位子を設計することができる。最適化するために、配位子のアミノ酸、アミノ酸の窒素上の保護基、及びヒドロキシルアミン三部分を種々変化させ、具体的には、アミノ酸、保護基、ヒドロキシルアミン部分と段階的に変化・検討していき、最終的に前記式(II)に示されるヒドロキサム酸配位子等の、高収率かつ高エナンチオ選択性を有するエポキシアルコールを得ることができる、最適化されたヒドロキサム酸配位子を得ることができる。

上記光学活性を有するヒドロキサム酸は、アミノ酸又はペプチドとカルボン酸無水物とを反応させ、生成したカルボン酸をカルボン酸クロライドに変換し、次いでこのカルボン酸クロライドとヒドロキシルアミンとを反応させることによって製造することができ、本発明にはかかる光学活性ヒドロキサム酸の製造方法により得られるヒドロキサム酸も包含される。前記アミノ酸としては、アラニン、2−アミノ酪酸、バリン、イソロイシン、ロイシン、tert-ロイシン、フェニルアラニン、トリプトファン、リジン、メチオニン、アルギニン等の各種アミノ酸を例示することができ、前記ペプチドとしては上記アミノ酸を構成アミノ酸とするジペプチド、トリペプチドを例示することができる。

また、前記カルボン酸無水物としては、例えば、無水フタル酸、無水コハク酸、無水マレイン酸などの飽和又は不飽和脂肪族ジカルボン酸無水物、テトラヒドロ無水フタル酸、ヘキサヒドロ無水フタル酸(1,2−シクロヘキサンジカルボン酸無水物)、1,2,3,4−シクロヘキサンテトラカルボン酸1,2−無水物などの飽和又は不飽和非芳香族性環状多価カルボン酸無水物(脂環式多価カルボン酸無水物)、無水ヘット酸、無水ハイミック酸などの橋かけ環式多価カルボン酸無水物(脂環式多価カルボン酸無水物)、無水フタル酸、テトラブロモ無水フタル酸、テトラクロロ無水フタル酸、無水ニトロフタル酸、無水トリメット酸、メチルシクロヘキセントリカルボン酸無水物、無水ピロメリット酸、無水メリト酸、1,8;4,5−ナフタレンテトラカルボン酸二無水物などの芳香族多価カルボン酸無水物を例示することができ、フタル酸、4−メチルフタル酸、テトラフルオロフタル酸、テトラクロロフタル酸、2,3−ジメチルマロン酸、2,3−ジフェニルマロン酸、2,3−ナフタレンジカルボン酸、1,8−ナフタレンジカルボン酸、4−ブロモ−1,8−ナフタレンジカルボン酸、2,3−(9H、10H−アントラ−9,10−ニル)マロン酸を好適に例示することができる。

前記ヒドロキシルアミンとしては特に制限されるものではないが、アリールハライドと蟻酸エチル(HCOOEt)とを反応させて得られるジアリールアルコールに、ヒドロキシルアミン塩酸塩(NH2OH・HCl)を反応させることにより得ることができるジアリールメチルヒドロキシルアミンが好ましく、具体的には、ビス(0-トリル)メチルヒドロキシルアミン、ビス(2-イソプロピルフェニル)メチルヒドロキシルアミン、ビス(3,5-ジ-tert-ブチルフェニル)メチルヒドロキシルアミン、ジ(ナフサ-1-yl)メチルヒドロキシルアミン、ビス(2-メトキシナフサ-1-yl)メチルヒドロキシルアミン、2,2′-エチレンジフェニルヒドロキシルアミンを好適に例示することができる。

本発明の不斉酸化反応用触媒は、光学活性ヒドロキサム酸を有効成分とするものであればどのようなものでもよく、かかる不斉酸化反応としては、不斉エポキシ化反応等を挙げることができる。本発明の光学活性ヒドロキサム酸は、上記不斉エポキシ化反応の中でも、バナジウム触媒を用いるアリルアルコールの不斉エポキシ化反応に好適に用いることができる。

【実施例】
以下本発明を実施例に基づいて詳細に説明するが、本発明の技術的範囲はかかる実施例により何ら制限されるものではない。
実施例1[分析方法及び材料]
赤外線スペクトルは、Shimadzu FTIR-9100で測定した。1H NMRスペクトルは、Varian Gemini-300(300MHz) 分光計で記録した。1H NMRの化学的置換は、内標準(0ppmにおけるテトラメチルシラン)に比例するダウンフィールドppmで表わした。分解パターンは、一重項をs、二重項をd、三重項をt、四重項をq、多重項をm、ブロードピークをbrとして示した。13C NMRスペクトルは、Varian Gemini-300(75MHz)分光計で測定し、内標準(77.0ppmにおけるCDCl3)としての溶剤を使用したppmで表わした。51V NMRスペクトルは、Varian Gemini-300(79MHz)分光計で測定し、 外標準(0ppm)としてのVOCl3と比較したppmで表わした。気液クロマトグラフィ(GLC)分析は、フレームイオン化検出器及びβ-TA(0.25mm×25m)の毛管カラムを備えたShimadzu GC-17A機器で、キャリヤーガスとしての窒素を使用して行った。高度液体クロマトグラフィ(HPLC)分析は、Shimadzu LC-10AD機器とSPD-M10A UV検出器でキラルの静止カラム(Daicel,AS又はOD−H)を使用して行った。光学的旋光度は、JASCO DIP-100デジタル偏光計で測定した。実験は全てオーブンで乾燥させたガラス機器中でマグネチックスターラーを用いて行われた。反応生成物は、シリカゲルE.Merck9385又はシリカゲル60エキストラピュア上でフラッシュクロマトグラフィにより精製した。薄層クロマトグラフィ(TLC)分析をMerckプレコートTLC板(シリカゲル60GF254、0.25mm)にて行った。微量分析は、名古屋大学農学部にて実施した。

トルエン及びジクロロメタン(CH2Cl2)は、4Åモレキュラーシーブ(MS)で保存した。N,Nジイソプロピルエチルアミン(i-Pr2NEt)は、KOHペレットで保存した。VO(O-i-Pr)3及び78%のtert-ブチルハイドロパーオキサイド(TBHP)は、High Purity Chemicals及びKatayama Chemicalsからそれぞれ購入した。α-アミノ酸、酸無水物、及び単純化学製品は、購入したものを使用した。アリルアルコール等は、以前分離・同定したものを用いた。これらは他でも引用している。ジフェニルメチルヒドロキシルアミン及び9-フルオレニルヒドロキシルアミンは、文献(J.Chem.Soc., Perkin Trans.I; 643, 1979)記載の方法に従って作製した。

実施例2(アミノ酸由来のヒドロキサム酸配位子の合成)
アミノ酸を主骨格とするヒドロキサム酸配位子を以下の反応式により合成した。
<Step1> 各種アミノ酸[1](3mmol)とカルボン酸無水物[3](3.3mmol)をDMF(1.5mL)に溶解させ、100℃で6時間から12時間攪拌し、反応溶液を室温まで冷却した後、酢酸エチルを使って分液ロートに移し、水で数回有機溶媒を洗浄した。有機層を硫酸ナトリウムで乾燥し、ロータリーエバポレーターを使って溶媒を留去し、ヘキサンを加えて固体を析出させることにより粗生成物[2]を得た。また、必要に応じてカラムクロマトグラフィー(酢酸エチル/ヘキサン=1:2)を用いて精製を行った。収率は27〜100%であった。

<Step2と3> カルボン酸[2](1.0mmol)をジクロロメタン(10mL)に溶解させ、オキサリルクロライド(1.5equiv)とDMF(触媒量、〜0.01mL)を加え、室温で2時間攪拌した後、溶媒をアスピレーターで留去し、真空ポンプを用いて十分溶媒を留去することによりカルボン酸クロライド[4]を粗生成物として得た。続いて、このカルボン酸クロライド[4]をジクロロメタン(10mL)に溶かし、そこへヒドロキシルアミン[5](0.9mmol)のジクロロメタン溶液(5mL)を室温で加え、1時間から2時間攪拌したところでエチルジイソプロピルアミン(0.9mmol)を加え、さらに30分間攪拌を行った。反応混合物を塩化アンモニウム飽和水溶液に流し込み有機層を分離し、さらに、水層から有機化合物をジクロロメタンで抽出し、前記有機層と抽出液との混合物を炭酸水素ナトリウム水溶液で洗浄し、硫酸ナトリウムを用いて有機層を乾燥させ、ロータリーエバポレーターを使って溶媒を留去し、カラムクロマトグラフィー(シリカゲル60エキストラピュア)を用いて精製し、ヒドロキサム酸[6]を収率〜50%で得た。次いで、適当な溶媒を用いて純ヒドロキサム酸を再結晶化した。

上記アミノ酸[1]としては、アラニン(1a)、2−アミノ酪酸(1b)、バリン(1c)、イソロイシン(1d)、ロイシン(1e)、tert-ロイシン(1f)、フェニルアラニン(1g)、トリプトファン(1h)、リジン(1i)、メチオニン(1j)を用い、カルボン酸無水物[3]としては、無水フタル酸(Pht)、無水4−メチルフタル酸(1k)、無水テトラフルオロフタル酸(1l)、無水テトラクロロフタル酸(1m)、無水2,3−ジメチルマロン酸(1n)、無水2,3−ジフェニルマロン酸(1o)、無水2,3−ナフタレンジカルボン酸(1p)、無水1,8−ナフタレンジカルボン酸(1q)、無水4−ブロモ−1,8−ナフタレンジカルボン酸(1r)、無水2,3−(9H、10H−アントラ−9,10−ニル)マロン酸(1s)を用いた。各種アミノ酸[1]と各種カルボン酸無水物[3]との反応生成物である各種N−ジアシルアミノ酸[3]の物性を以下に示す。

実施例3(ヒドロキシルアミンの合成;方法1)
ヒドロキサム酸の合成に用いるヒドロキシルアミンを以下の反応式により合成した。
<Step1> 三口フラスコにジムロート冷却管と滴下ロートと三方コックを取り付け、十分乾燥させ不活性ガス雰囲気下とし、削状マグネシウム(Mg)とマグネシウムが浸漬する程度のTHFを入れた。滴下ロートにアリールハライド[7]のTHF溶液を入れ、滴下ロートからこの溶液のだいたい1割程度の量の溶液を加えた。しばらく攪拌を行っていると反応容器が発熱してくることにより反応が開始したことを確認し、残りのアリールハライドのTHF溶液をゆっくりと滴下し、全て滴下し終わったら反応温度を60℃に昇温して数時間攪拌を続けて反応を完結させた。反応溶液を室温ぐらいまで冷却し、蟻酸エチル(HCOOEt)をゆっくりと添加した。再び反応温度を60℃まで昇温し6時間攪拌を行った後、室温に冷却して、反応混合物を塩化アンモニウム飽和水溶液に流し込み、ジエチルエーテルで3回抽出を行い、硫酸マグネシウムを用いて乾燥させる。溶液をロータリーエバポレーターで濃縮し、ヘキサンを加えることにより固体を析出させ、アルコール[8]の粗生成物を得た。収率は〜100%であった。

<Step2> ジクロロメタンに酢酸ナトリウム(AcONa)(656mg、2mmol)とジクロロ酢酸(CHCl2COOH)(2.0mL、24mmol)を加え、溶液になるまで室温で攪拌する。この溶液にヒドロキシルアミン塩酸塩(NH2OH・HCl)(556mg、8mmol)を加え、1時間室温で攪拌し、得られた白色懸濁液にアルコール[8]のジクロロメタン溶液を添加した。反応時間はアルコールの種類により異なり、反応が平衡に達するまでに早いものは2時間、長いものでは数日間要した。反応溶液に氷を加え続いて炭酸水素ナトリウム飽和水溶液を加えて反応溶液を中和した。これを分液ロートに流し込み、ジクロロメタンを使って3回抽出操作を行い、有機層を硫酸ナトリウムで乾燥し、ロータリーエバポレーターを使って溶液を濃縮し、ヘキサンを加えることにより結晶を析出させた。結晶がうまく析出しない場合は一度カラムクロマトグラフィーをかけ、精製することにより析出させた。ヒドロキシルアミン[5]が粗生成物として収率〜50%で得られた。

【発明の効果】

In the asymmetric epoxy reaction, epoxy alcohol (91%; 62% ee) is obtained when hydroxamic acid (1 g) is used as the hydroxamic acid, and epoxy alcohol when the peptide-based hydroxamic acid described in Example 5 is used. Example 7 (Asymmetric epoxidation reaction; reaction 2)
Using an optically active hydroxamic acid, an asymmetric epoxidation reaction of alcohol using a vanadium catalyst was performed according to the following reaction formula. 【The invention's effect】
The novel optically active hydroxamic acid ligand of the present invention is composed of three parts, an amino acid (optically active compound), a protecting group on the nitrogen of the amino acid (an imide-type protecting group), and a hydroxylamine moiety. It is possible to synthesize many similar ligands. There are many commercially available amino acids and protecting groups, and there is an advantage that they are easily available. Also, the route for synthesizing the ligand is very short and practical, with two routes. Further, when an asymmetric epoxidation reaction of allyl alcohol using a vanadium catalyst is performed using the optically active hydroxamic acid ligand of the present invention, it becomes possible to achieve a high selectivity of 95% ee at the maximum. The asymmetric epoxidation reaction using the optically active hydroxamic acid ligand of the invention is characterized by high catalytic activity and selectivity, use of a non-halogen solvent, and ease of operation. Moreover, high selectivity can be achieved by using 0.001 to 0.01 equivalent of vanadium concentration with respect to the substrate.
