本発明は、新規な光学活性二座ホスフィン配位子パラジウム錯体及びその用途に関する。

本発明の新規な光学活性二座ホスフィン配位子パラジウム錯体は、医薬品の製造中間体である光学活性β−ヒドロキシケトン類を製造する際に、触媒として用いることができる。

【従来の技術及び発明が解決しようとする課題】
近年、遷移金属錯体を触媒として用いる有機合成反応は精力的に開発されており、中でも光学活性二座ホスフィン配位子との組合せによる不斉合成は、望みとする光学活性体を効率的に得る方法として活発に研究がなされている。例えば、遷移金属としてパラジウムを用いる不斉触媒の開発はこれまでに幾つか報告されており、下記の(A)の構造を有するPd(BINAP)2や(B)(Ozawa,F.;Kubo,A.;Matsumoto,Y.;Hayashi,T.Organometallics,1993,12,4188)等が知られているが、光学活性二座ホスフィンを配位子として持ち、さらに水分子又は水酸基が配位したパラジウム錯体は知られていない。

光学活性β−ヒドロキシケトンを得る触媒的不斉アルドール反応は、光学活性ルイス酸触媒によるシリルエノールエーテル、ケテンシリルアセタール又はケテンシリルチオアセタールとアルデヒドとの縮合反応である。たとえば(a)Heathcock,C.H.In Asymmetric Synthesis;Morison,J.D.,Ed.;Academic press:New York,1984;Vol.3,Chapter2.(b)Heathcock,C.H.In Comprehensive Organic Synthesis;Perganon Press:Oxford,1991;Vol.2,Chapter1.5〜1.9などの文献に報告されている。しかし、これら不斉ルイス酸触媒によるアルドール反応では、極めて高い不斉収率が報告されているものの、ルイス酸の性質上、医薬品の製造において様々な酸素官能基等に親和性をもつことが予想され、このような反応を複雑な化合物に適応する場合には困難が予想される。

一方、様々な酸素官能基等に親和性をもたないと予想される遷移金属エノレートを経る触媒的不斉アルドール反応に関しては、Reetz,M.T.;Vougioukas,A.E.Tetrahedron Lett.,1987,28,793.Roos,G.H.P.;Haines,R.J.;Raab,C.E.Synth.Commun.1993,23,1251.などの文献に報告されているが、不斉収率は20%程度であり、新規な光学活性な配位子をもつ遷移金属錯体及び、該錯体を用いた効率の良い不斉合成法の開発が望まれていた。不斉アルドール反応はエリスロマイシンA、エリスロマイシンB等ポリオキソマクロライド抗生物質のアグリコンの不斉合成に応用できることから、アルドール反応の立体化学に関する研究が活発におこなわれてきた。又、β−ヒドロキシケトンを部分構造に有する化合物としてはジンゲロール類縁化合物が挙げられる。ジンゲロール類は強心作用(特開昭64−921号公報及び特開平6−40895号公報)、血小板凝集作用(特開昭63−72625号公報)、鎮痛作用(特開昭59−1684号公報)、抗炎症作用(特開平3−90021号公報)、抗寄生虫作用(特開平2−4711号公報)等が知られている化合物である。またHMG−CoA還元酵素阻害剤のメバロン酸部位にはβ−ヒドロキシエステルを有しており、効率の良い光学活性β−ヒドロキシケトンを得る方法は有用である。しかし、既知の錯体(A)や(B)を用いてもアルドール反応は進行せず、β−ヒドロキシケトンは得られない。

【課題を解決するための手段】
本発明者は、上記課題を解決すべく鋭意努力した結果、水分子又は水酸基が配位した新規な光学活性二座ホスフィン配位子パラジウム錯体を開発した。
該錯体はアルドール反応の触媒として極めて有用である。しかも反応中間体としてパラジウムエノレートを経るという、これまでに知られている不斉ルイス酸触媒による反応と異なる、全く新しいタイプの反応であり、高い化学収率及び光学収率を与える不斉触媒として有効であることを見出した。
また、本発明のパラジウム錯体合成に用いる光学活性二座ホスフィン配位子は、両エナンチオマーが入手容易であるため、ふたつの光学活性体を作り分けることが可能であり、自由に立体を制御できる新規な光学活性二座ホスフィン配位子パラジウム錯体および、該錯体による光学活性β−ヒドロキシケトン類を得る新規反応系を確立し、本発明を完成するに至った。
