本発明は、極微量の液体試料中の成分を検出する場合に利用される電気泳動用チップに関する。

【従来の技術】
近年、Science, Vol.261, P.895-897 (1993)に記載されているように、ガラス(例えば、パイレックス(Corning Glass Works社(米国)の登録商標)ガラス)基板を材料とした電気泳動部材上に液体試料を導入するための流路と液体試料を分離するための流路を、半導体製造技術を基盤とするマイクロマシニング技術を用いて形成した電気泳動装置が開発されている。この電気泳動装置で用いる電気泳動部材を電気泳動チップと呼ぶ。電気泳動チップを用いた電気泳動装置は、従来のキャピラリー電気泳動装置と比較して、高速分析が可能、溶媒消費量が極めて少ない、必要とするサンプルが極微量、装置の小型化が可能などの利点を有する。

これらの特徴は、分析化学の分野において従来の分析装置では実現が困難であった、現場(オンサイト又はベッドサイド)分析を可能とするものとして、またDNA(デオキシリボ核酸)分析などの分野に対しては高速分析による多検体処理が可能なものとして、また創薬分野などのハイスループットスクリーニングに有利なものとして有望視されている。

通常、ガラス表面はマイナスに帯電しており、ガラス表面と泳動バッファ(導電性)の間には電気二重層が形成され、電気浸透流が起こる。この電気浸透流が分析の障害となる場合、これを抑制するために流路表面は化学的に修飾される。化学修飾は、例えば表面をシリル化した後にアクリルアミドコーティングを行う方法が採られる。これには放置時間を含めて3日程度が必要である。

この煩雑な操作を不要とするため、分析前に吸着防止用試薬を流すことによりコーティングを行う方法も用いられる(以下この方法をダイナミックコーティングと称する)。しかし、ダイナミックコーティング表面の耐久性が低く、使用回数に応じて分析再現性が低下してくるため、繰り返し使用する場合にはダイナミックコーティングを繰り返し行う必要がある。

【発明が解決しようとする課題】
このように内面をコーティングしたチップを作製する工程において、耐久性の高いコーティングを行うためには長時間を費やし、逆に簡単な方法でダイナミックコーティングを行えば、耐久性が低いという問題がある。 本発明は、このような課題を解決することができる分析再現性に優れた電気泳動チップと、簡単な工程で実現できる製造方法を提供することを目的とするものである。

【課題を解決するための手段】
本発明は、液体試料を導入するための試料導入口、導入された液体試料の流路、及び液体試料を排出する試料排出口が設けられたガラス製電気泳動チップであって、流路内面が炭素を主成分とする皮膜で被われていることを特徴とするものである。
その炭素を主成分とする皮膜の好ましい一例は炭素皮膜である。

そのガラス製電気泳動チップの内面に炭素を主成分とする皮膜を析出させる1つの好ましい方法は、炭素化合物を含むガスを、加熱した電気泳動チップに接触させてその流路内面を含むチップ表面上に熱CVD(chemical vapor deposition)法により皮膜を析出させる方法である。

炭素化合物を含む材料ガス、例えばCH3OH、CH3COCH3などの有機化合物ガス中に、500〜700℃に加熱されたチップを置くことにより、流路内面を含むチップ表面でガスの分解反応が生じ、流路内面を含むチップ表面に炭素を主成分とする皮膜、例えば非晶質グラファイト膜などが形成される。

【発明の実施の形態】
図1は本発明の電気泳動チップの一実施例である。(A)は平面図、(B)はそのA−A線位置での断面図、(C)はそのB−B線位置での断面図である。
本図において1,2はガラス基板、例えば合成石英基板である。ガラス基板1の片面には、数100μm以下の幅、深さを持ち液体試料用流路として用いる微小な流路溝8が形成されている。一方、ガラス基板2には試料導入又は排出のための穴3が形成されている。両基板1,2の接合すべき面を向かい合わせて密着させ、後で説明するフッ酸溶液による接合などの手段で気密に接合することで内部に液体試料用の流路溝8が形成されている。

流路溝8の内面には電気浸透流を抑えるために炭素を主成分とする皮膜、例えば炭素皮膜が形成されている。その炭素皮膜は穴3の内面にも形成されている。

このような構成の電気泳動チップにおいて、液体試料用の微小な流路溝8の一部を測定室8aとして使用すれば、十分に微小な体積の測定室8aを実現できる。測定室8aの部分には検出光4が照射され測定が行なわれる。

次に、上述した電気泳動チップを作製するプロセスについて図2により説明する(Trans. IEE Japan, Vol.119-E, No.10, Oct., 1999 「電気泳動用石英ガラス製マイクロチップの開発」参照)。

まず、図2(a)に示したように、合成石英製のガラス基板1を洗浄した後、薄膜形成装置(例えばスパッタ成膜装置)にてエッチング保護膜5、例えば膜厚3000Åのシリコン(Si)薄膜を形成する。さらにその上に、エッチング保護膜5をパターニングするためのフォトレジスト6、例えばAZ4620(商標:Clariant社製)を3000rpm、40秒間の条件にてスピンコートする。使用するフォトレジスト6の材質及び厚みは特に限定されるものではなく、後のエッチング工程における溶液に耐える材質及び厚みであればよい。また、エッチング保護膜5の材料及び厚みも特に限定されるものではなく、後のガラス基板1のエッチング工程における溶液に耐える材質及び厚みであればよい。

次いで、図2(b)に示したように、フォトマスク7を用いてフォトレジスト6を紫外光9で露光し、続いて現像してフォトレジスト6をパターニングするする。ここで、フォトレジスト6の露光は、一般に半導体製造に用いられているアライナを用いて行なうことができる。露光後のフォトレジスト6を現像する現像液は、用いるフォトレジストを現像するために使用されているものであれば、特に限定されるものではない。

続いて、図2(c)に示したように、SF6ガス中での高周波プラズマを用いたドライエッチングにより、フォトレジスト6のパターンをマスクとしてエッチング保護膜5をパターニングする。ここで、エッチングガスは特に限定されるものではなく、シリコンが問題なくエッチングされるガスであればよい。

さらに、図2(d)に示したように、パターニングされたエッチング保護膜5及びフォトレジスト6をマスクとして、石英ガラス基板1を例えば46%フッ酸水溶液にてエッチングして試料用流路溝8を形成する。ここで、石英ガラス基板1のエッチング液は特に限定されるものでなく、石英ガラスが問題なくエッチングされる溶液であればよい。

続いて、図2(e)に示したように、フォトレジスト6を完全に除去した後、エッチング保護膜5をエッチング除去する。

一方、他方のガラス基板2に対しては、図2(f)に示したように、例えばサンドブラスト等の加工により液体試料導入又は排出のための貫通穴3を形成しておく。

最後に、(a)〜(e)の工程により試料用の流路溝8を形成したガラス基板1と工程(f)により貫通穴3を形成したガラス基板2を重ね合わせ、例えば1%のフッ酸水溶液を界面に介在させ、必要に応じて1MPa程度の荷重を印加しつつ、室温で24時間放置することで、図2(g)に示されたようにガラス基板1と2を接着させて電気泳動チップを完成する。

この状態の流路内面表面には、通常、OH基が存在し、その量によって流路内壁−溶液間の電位差、ひいては電気浸透流が生じる。電気浸透流は分析に利用する場合もあるが、例えばDNA分析の場合、分離性能を上げるために、電気浸透流を抑制する必要がある。

そこで、この実施例では、図3に示されるように、電気泳動チップの流路溝8の内面に炭素を主成分とする皮膜として炭素皮膜10を形成する。炭素皮膜10は流路溝8の内面だけでなく、ガラス基板2の穴3の内面にも形成されている。

次に、電気泳動チップの流路溝8の内面に炭素を主成分とする皮膜を形成する方法を図4を参照して説明する。
図4は熱CVD装置12を示しており、横型の円筒状石英管14の周囲にヒータ16を備えた炉が配置されている。石英管14の一端から炭素化合物を含んだ材料ガスが供給され、他端から排出されるようになっている。石英管14内の温度は温度センサ(図示略)により検出され、その検出温度が所定の温度になるように炉のヒータ16の通電が制御される。

図2の工程で作製された電気泳動チップ20を図4に示す熱CVD装置12内に入れ、ヒータ16によって電気泳動チップ20を600〜700℃に加熱しながら材料ガスを導入する。材料ガスはCH3OHであり、その供給流量は0.1L/分である。電気泳動チップ20では流路内面を含むチップ表面でガスの分解反応が生じ、流路内面を含む電気泳動チップ20表面に炭素皮膜10が形成される。

図5に図1に示した本電気泳動チップを検出セルとして用いた光学測定装置の一例を示す。電気泳動チップは流路溝8の測定室8a部分の表面側に検出光を入射させるための入射窓、裏面側に検出光を出射させるための出射窓を形成し、その他の部分からは外乱光の侵入を防ぐために、入射窓と出射窓を除いてその表面と裏面が遮光膜で被われている。

図5において、21は重水素ランプ、タングステンランプ及び分光器が内蔵されて所定の波長の光を送り出す紫外可視光源、22はフォトダイオードアレイ検出器を使用した測光光学系を有する光検出器であり、いずれも紫外可視測定に一般的に用いられるものである。23はステージであり、検出計セルとしての電気泳動チップ20を位置決めできる凹部24が設けられている。そして、電気泳動チップ20をこの凹部24に挿入することにより、ステージ23に形成された入口流路25と電気泳動チップ20の試料導入口26とが密着でき、ステージ23に形成された出口流路27と電気泳動チップ20の試料排出口28とが密着できるようになっている。さらに、光源21からの光が電気泳動チップ20の入射窓から入射でき、出射窓からの光が光検出器22に入射して受光されるようになっている。これにより、ステージ23の凹部24にこの電気泳動チップ20をセットすれば、光学測定が可能になる。

上記の実施例では、炭素を主成分とする皮膜として炭素皮膜について述べたが、材料ガスの選択によっては、その他の炭素を主成分とする皮膜を形成することも可能である。この場合の皮膜は電気浸透流の抑制効果のある膜であれば何でも良い。

【発明の効果】
本発明が対象とする電気泳動チップは、フォトファブリケーション技術にて高精度に形成した幅、深さ共に微小で流路断面積が分離キャピラリーカラムとほぼ同程度な流路を測定室として使用できるため、分離能力を損なわない程度に微小な体積の測定室を実現できる。
そして、本発明によれば、その電気泳動チップの流路内面に炭素を主成分とする皮膜を形成したことにより、電気浸透流を抑制することができ、しかもこの炭素を主成分とする皮膜はダイナミックコーティングよりも耐久性が高く、化学修飾によるコーティングよりも短時間に形成することができる。
本発明の製造方法によれば、簡単な熱CVD法により耐久性のある電気浸透流抑制皮膜をもつ電気泳動チップを製造することができる。
