【従来の技術】
前記一般式〔I〕で表される3,5共役エノール構造を持つステロイドの6位のフッ素化は、親電子型のフッ素化剤を用いて行われている。親電子型のフッ素化剤としては、FClO3(パークロリルフルオライド)(例えば、J. Org. Chem., 29, 2982‐2986(1964)、Oraganic Reactions, chapter 2, Modern Method to Prepare Monofluoro aliphatic compounds, pp.225‐236 等参照。)が用いられている。また、酢酸と酢酸ナトリウムの存在下低温でフッ素ガスを作用させる方法(特開昭59−139398号、特開昭60−6700号公報)などがあるが、収率、毒性、爆発性等の取り扱いの困難なガスを使用しなければならないといった欠点があった。本発明者らはこれらの欠点を解消するため、次式〔II〕で示すN−フルオロピリジニウム塩

しかし収率が低く、反応時間が長いという欠点があった。

【発明が解決しようとする課題】
本発明者らはこれらの欠点を克服すべく鋭意研究を重ねた結果、窒素−フッ素結合を有する特定の親電子型フッ素化剤を、一般式〔I〕で表される3,5共役エノール構造を持った特定のステロイドと反応させる際に、有機カルボン酸を添加すると、6位がフッ素化されたステロイドの収率が向上し、かつ必要とする反応時間が短縮できることを見いだし、本発明を完成するに至った。

(N−フルオロピリジニウム塩)
本発明で使用する窒素−フッ素結合を有する親電子型フッ素化剤のうちの1つである、一般式〔II〕のN−フルオロピリジニウム塩は、例えば特公平2−33707号公報に記載されているように、ピリジン化合物をブレンステッド酸またはルイス酸の存在下に、フッ素ガスと反応させることによって得られる。これを製造するための反応としては、例えば、次のようなものがある。

本発明において添加する有機カルボン酸としては、酢酸、蟻酸、トリフルオロ酢酸等が好ましい。有機カルボン酸の添加量は特に限定されるものではないが、収率、経済性等を考慮すると、ステロイドに対して0.2〜3.0倍モルが好ましく、0.3〜3.0倍モルがより好ましく、0.5〜2.0倍モルが最も好ましい。

(溶媒)
本発明に使用される溶媒は、とくに限定されるものではないが、ジクロロメタン、1,2−ジクロロエタン、1,1,3−トリクロロエタン、1,1,2,2−テトラクロロエタン、ヘキサフルオロベンゼン等のハロゲン系溶媒や、アセトニトリル等を用いるのが好ましい。
