即ち、本発明は、新規な微弱光測定システムを用いることにより生体内における動物の皮膚において紫外線照射下で産生される活性酸素を直接観測できる方法を提供するものであり、さらに、この方法を用いた活性酸素の発生機構の解明と新しい皮膚傷害防御薬の開発方法を提供するものである。

本発明の活性酸素と反応して発光する物質としては、前記した活性酸素の中の特定の活性酸素種と反応するものであってもよいが、いずれの活性酸素種と反応するものであってもよい。また、発光する光としてはCCDカメラなどの検出装置で測定することができる光又は波動であるならば特に制限はないが、可視光線又は蛍光による可視又は紫外線領域の光が好ましい。好ましいが、活性酸素と反応して発光する物質としては、例えば、ルミノール又はCLA(2−メチル−6−フェニル−3,7−ジヒドロイミダゾ−[1,2−a]ピラジン−3−オン)などが挙げられる。

本発明のスクリーニング方法により、簡便かつ正確に生体内で活性な抗酸化剤を見出すことができることになる。したがって、本発明は当該スクリーニング方法によって得られた抗酸化剤組成物を提供するものでもある。

次に本発明をより具体的に説明する。
本発明者らは、紫外線照射後の皮膚において発生する活性酸素種の検出と同定について検討した。
活性酸素と反応して発光する物質(以下、単に発光物質という。)としてCLAを用いた場合、照射終了30秒後から30分間にわたって非常に強い発光が観測された。この惹起された発光がSOD前処置によってほぼ完全に抑制され、β−カロテン(β-calotene )前処置では若干抑制されたことから、紫外線照射後の皮膚において発生する主な活性酸素種は・02 −であり、さらに一部102も発生していることが示された。

発光物質としてCLAを用いた場合、塗布5分後から15分間にわたって発光が観測された。ただし、正常皮膚からの発光量は紫外線照射後の発光量と比較するとかなり弱いものであった。この場合の発光はSOD前処置によってほぼ完全に抑制され、β−カロテンにはほとんど影響されなかったことから、正常の皮膚において発生している主な活性酸素種は・02 −であることが示された。
