本発明は、極微少量の液体を正確に吐出するピペットの製造方法に関する。

【従来の技術】
従来のピペットには、例えば、ガラス製のピペット本体にゴム袋を取付けたものや、ピペット本体をインジェクション成形法を用いた合成樹脂製として、これにマイクロシリンジを装備させたものがある。

前者は、比較的多量の液体を分注するものであり、ゴム袋を圧縮復元することによりサンプル液を吸入排出する構造になっている。

後者は10~2l〜10~7lの微少量液体を分注するもので、マイクロシリンジを持つ本体と、テーパ状に形成された使い捨て式のチップ(ノズル)からなり、マイクロシリンジの吸引、加圧によって、液体の吸入排出を行うようになっており、先端部のチップが使い捨てであるから、サンプル液交換時にピペットを洗浄する必要がなく、また他のサンプル液との混濁を容易に防ぐことができる。

その他、電磁モータ、圧電ポンプ等を用いたオートピペッタが考案されている。また、最近では、細胞、血液、遺伝子等の解析技術の発達にともない、解析の迅速化、使用サンプル液の低減が要求されるようになり、ピペットにおいてもナノオーダ(実際には10~9l〜10~12l)の分注能力を持つピペットへのニーズが高まりつつある。しかし、従来のピペットでは、上述したように10~7lの分注が限界であり、その精度も十分とは言えない。

このような事情から、最近は、インクジェットプリンタのヘッドで用いられるインク噴出機構が注目されており、実験段階ではあるが、論文も発表されている。

このインクジェット方式は、円筒型圧電素子を応用したアクチュエータを使用するものと、積層型またはスティック型圧電素子を使用するものとがある。

このうち、前者は、アクチュエータ(円筒型圧電素子)の内側にノズル部に通じるチューブ(サンプル液供給管)を挿入した構造となっている。アクチュエータに電圧を印加すると、その円筒型圧電素子の内径が縮み、チューブ内のサンプル液に圧力が加えられ、ナノオーダのサンプル液がノズル部より吐出される。

後者は、チャンバの壁の1つが圧電素子を接着した膜板(可撓性を有する薄板)となっており、圧電素子に電圧を印加して薄板を撓ませることにより、チャンバに圧力を加えてサンプル液をノズル先端より吐出させる。両者ともサンプル液はリザーバから供給される。吐出されたサンプル液の速度は、数m/s〜数十m/sである。

【発明が解決しようとする課題】
インクジェット方式をピペットに採用した場合には、次のような改善すべき点があった。

(1)分注量を極微少量にする目的は、高コストの試薬の総使用量を低減することにあり、より微少なチャンバーとその製造方法が要求される。従来の製造方法では、コスト及び精度の点から作製は困難である。特に、インジェクション成形法ではノズル先端部の精度に限界がある。

(2)微小な領域に分注液供給路などの複雑な構造を形成するため、一体成形は困難である。また、構成部品の接合に接着剤等を用いると接着剤の余分な部分へのはみだし、すなわち、液や気泡のスムーズな流れの妨げとなるようなサンプル液流路等への接着剤のはみだしや、接着剤のサンプル液への汚染などのクリーン度の低下を招くおそれがある。

本発明は、以上のような問題点を解決し、10~6l以下の極微少量の液体を精度良く扱え、しかも試薬等の使用に耐えるクリーン度の高いピペットの製造方法を提供することを目的としている。

【課題を解決するための手段】
本発明は、上記目的を達成するために、基本的には、次のような課題解決手段を提案する。

先端部に毛管現象により液体を吸い上げ可能にしたノズルを有し、その上部にチャンバが配置され、該チャンバの上部に、電気信号により微小変位する駆動機構を介して撓み力が付与される膜板が配置され、この膜板の撓み動作によりチャンバ内に満たされた液体を吐出させる構成からなるピペットにおいて、チャンバ、リザーバ、ノズル形成の際にマスキング及びエッチング加工を行う工程と、該チャンバ、リザーバ、ノズル接合の際に陽極結合を行う工程を含むことを特徴とする。

また、前記ピペットの構成部品全てをマスキング及びエッチングで形成した後、陽極結合することにより、複数個のピペットを1ロットで作成することを特徴とする。

【作用】
上記の製造方法によれば、低コストで量産に適用し易く、特に集積化されたピペットの製造に最も有効である。更に、この製造方法では接着剤を用いないので、サンプル液流路等への接着剤のはみ出しがなく、クリーン度の高いピペットを製造できる。

〔実施例1〕
図1に本発明のピペットの側面からみた要部断面図を示す。

図1において、先端部に毛管機能を有するノズル1、サンプル液収容部となるチャンバ2、可撓性を有する薄板で形成された膜板4、通気管(通気路;連通路)3、液溜め部となるリザーバ5、圧電素子6等がある。圧電素子6は静電アクチュエータに代えてもよい。

ピペット本体となる一つの組立体は、三層の組立要素(板体)11、12、13を接合して構成され、下層の組立要素11にはテーパ状の細径ノズル1が形成され、中層の組立要素12にはノズル1より大きな内径を有するチャンバ2が形成されると共に通気管3及びリザーバ5の底部が形成され、上層の組立要素13には圧電素子6の一部を収容するスペースとリザーバ5の底部以外の部分とが形成されている。

これらの組立要素11、12、13を三層に積層、接合することで一つの組立体1が構成され、組立要素12、13間に膜板4となるべき薄板を介在させる。これにより、ノズル1の上部にチャンバ2が配置され、チャンバ2の上部に膜板4が配置され、膜板4の接する位置にチャンバ2内に生じる気泡を逃すための通気管3が配置される。また、チャンバ2の斜め上方には、リザーバ5が大気開放の状態で配置される。リザーバ5は、その底部がチャンバ2の上部と同レベル或いはそれより上の位置にあって、このリザーバ5底部とチャンバ2上部とが通気管3を介して連通して成る。チャンバ2とリザーバ5とは、隔壁13aを介して上下方向に斜め隣に位置するレイアウト構成となっている。これにより、ノズル1、チャンバ2、通気管3、リザーバ5は一つの組立体(ピペット本体)に集中的に且つ一連に配設される。

図2は製造工程のフローチャートである。

まず、組立要素11の作製を行う。組立要素11の材料としてはシリコンウエハが好ましい。この組立要素(以下シリコンウエハという)11にエッチングを行い、ノズル先端部を作製する。

図3に示すように、1μmの酸化層(SiO2層10)を(100)面に持つシリコンウエハ11にレジスト9を塗布し、露光及び現像を行う。次にフッ化水素とフッ化アンモニウムの混合液でSiO2層10を部分的にエッチングし、さらに、フッ化水素でシリコンウェハー11の異方性エッチングを行う。

本実施例で作製したノズル1の縦断面図を図4に示す。
異方性エッチングの傾斜は約54°であるので、シリコンウェハー11の厚さとマスクのサイズによりノズル先端の穴の大きさを次式で決定できる。

膜板4(ダイヤフラム)は、金属等を圧延加工で引き延ばすことにより得られるが、その材料の熱膨張係数は、感光性ガラス12と同等かもしくはより高いものを選択する必要がある。これは、膜板4とチャンバ2、リザーバ5を接合する際に行う陽極結合では材料を300℃〜600℃に加熱するが、接合後に、加圧壁である膜板4に適度な弛みをつけるため、また熱膨張係数の差による接合界面の破壊を避けるためである。

本実施例で作製した膜板4を図7に示す。
厚さは10μm、材質はチタンであり、前述したようにリザーバ5を横切る部分を開孔すべく連結穴17が設けてある。

チャンバ2が形成されている感光性ガラス12を陰極側とし、400℃に加熱した後、電圧1.5kVを電極15間に5分間印加した。この方法は接着剤を使用しないのでピペットのクリーン度を容易に保つことができ、また接着剤等のはみ出しがないため、接合面に位置する通気管を正確に、且つ容易に形成できる。

本実施例は実施例1の方法を用いて複数個のピペットを1ロットで作製するものである。

各材料を、複数個の部品を形成できるように作製したマスクを用いてエッチングし、位置合わせを行った後一度の陽極結合で各材料を接合する。この際、ピペットは接合された1つの部材に複数個並べた形で成形されている。これを冶具に固定し、図10のようにスライサで切断ラインに沿って切断することにより、複数個のピペットが得られる。また、スライサによる切断工程を省くことにより、集積型ピペットが容易に得られる。

【発明の効果】
以上説明したように、本発明による製造方法によれば量産性が高く安価で、かつクリーン度の高い良好なピペットを得ることができる。
