本発明は、ラフィノース液の精製方法に関するものであり、更に詳細には、本発明は、ラフィノースを製造する際に、ラフィノース液、特にケストース類(フルクトシルスクロース)を比較的多く含むラフィノース液の精製処理に関するものである。

【従来の技術】
ラフィノースは、甜菜糖蜜中に含まれており、従来は甜菜糖製造における不純物とされていた。しかしながら、近年になってラフィノースには整腸作用等のすぐれた生理作用があることが発見され、ラフィノースの有用性が見直されてきている。

ガラクトース、グルコース、フルクトースからなる三糖類であるラフィノースは甜菜糖蜜から結晶させて製造している(共立出版「化学大辞典9」第547頁)。具体的には、従来、製糖工程汁よりラフィノースを分離するには陽イオン交換樹脂クロマトグラフィーを採用し、得られたラフィノース画分を濃縮および結晶化することによってラフィノースを製造している。さらに、回収率を向上させるため、結晶化時の分離シラップを、クロマトグラフィー原料および結晶化原料に混合させ工程中を循環させている。

【発明が解決しようとする課題】
甜菜製糖工程汁より三糖類であるラフィノースを分離精製することは、前記したように、甜菜製糖における不純物(副生物)の有効利用の面から有意義なことであるが、従来、甜菜製糖工程汁からラフィノースの分離に採用している陽イオン交換樹脂クロマトグラフィーでは、三糖類であるケストース類(フルクトシルスクロース)とラフィノースの分離が不良であり、ラフィノース製造工程において回収率を向上させるために結晶化時のシラップをクロマトグラフィー原料および結晶化原料に混合させ工程中を循環させた場合、ケストース類が工程中に蓄積しラフィノース純度が下がって、それ以上のラフィノースの回収が困難になっていた。

現に、Bx50、50℃のシラップを25℃に冷却して結晶化した場合、シラップがラフィノース100%の場合にはラフィノースの結晶化率は82%に達するのに対して、ラフィノース60%及びケストース40%のシラップの場合には、ラフィノースの結晶化率は64%にまで低下してしまう。
このように、ケストース類の共存下においてラフィノースの効率的結晶化を行うことはきわめて困難であって、ケストース類の存在はラフィノースの工業的製造における大きな障害となっている。

ラフィノースを甜菜製糖工程汁から製造する場合、甜菜製糖工程汁にはケストース類が含まれているため、効率的にラフィノースを製造することができず、この点の解決策が強く求められていた。

【発明が解決しようとする課題】
本発明は、このような当業界の要望に応え、ラフィノースの効率的製造方法を新たに開発する目的でなされたものである。
そこで本発明者らは、各方面から研究した結果、Streptomyces rochei E87由来のイヌリン分解酵素がケストース類を分解する作用を有することをはじめて見出した。そして更に研究した結果、該酵素はケストース類(フルクトシルスクロース)のみを選択的に分解すること、そしてラフィノースの共存下においてラフィノースには全く作用しないことをはじめて発見した。

すなわち、本発明者らは、該酵素がラフィノースの存在下ケストース類のみを選択的に分解し、ラフィノースはそのまま保持されるというきわめて有用な新知見を得、この新知見に基づき、甜菜由来のラフィノース液に該酵素を作用させたところ、ケストース類の選択的分解を確認し、工業的にラフィノースの製造が可能であると確信するに至り、本発明を完成させたのである。

すなわち本発明は、イヌリン分解酵素を用いることにより、ラフィノース液に含まれるケストース類のみを選択的に分解し、もってラフィノースを精製、分離する点を基本的技術思想とするものである。
以下、本発明について詳述する。

【発明の実施の形態】
本発明を実施するには、イヌリン分解酵素を使用するが、本酵素の由来に格別の限定はなく、動物、植物、及び/又は微生物等すべての由来の酵素が使用可能である。
微生物由来の酵素としては、ストレプトマイセス属菌、例えば、ストレプトマイセス・ロケイ(Streptomyces rochei)もしくはストレプトマイセス sp.(Kawamura et al. 1989)由来、又は、オーレオバクテリウム属菌、例えばオーレオバクテリウム(Aureobacterium)sp.MCI−2494(Ohba et al. 1991)由来のイヌリン分解酵素が例示される。

これらの酵素の内、Streptomyces rochei E87由来のイヌリン分解酵素は、イヌリンのエキソ型加水分解酵素であって、イヌリンをエキソの形態で攻撃し、主生成物としてイヌロテトラオースを生成し、それからイヌロトリオースとフルクトースに分解する作用を有し、イヌロトリオースの高純度生産に用いられる酵素である(Tomita et al. : Letters in Applied Microbiology, 1995, 21, 330−333)。
イヌロトリオースの高純度生産に用いられる上記したイヌリン分解酵素は、本発明においてケストースを特異的に分解してラフィノースを精製する酵素として有利に使用できることがはじめて確認され、本酵素を生産する微生物であるStreptomyces rochei E87を工業技術院生命工学工業技術研究所にFERM P−15735として寄託した。

イヌリン分解酵素としては、精製酵素が使用できることはもちろんであるが、粗製酵素、酵素を含有した酵素含有物、そ(れら)の処理物も使用することができる。
酵素含有物は、本酵素を含有するものがすべて包含され、例えば本酵素生産菌菌体、同生産菌の培養液、同生産菌の培養物(菌体、培養液及び/又は培地成分等の混合物)も、酵素含有物に包含される。
また、酵素の処理物としては、精製酵素、粗製酵素、及び/又は酵素含有物を適宜処理したものをすべて指し、例えば、乾燥物〜ペースト化物〜濃縮物〜希釈物等が挙げられる。

本酵素をケストース類含有ラフィノース液と接触させれば、ケストース類のみが選択的に分解されて、クロマト処理によってラフィノースと分離され易い二糖類及び単糖類となる。
この場合、ラフィノースには本酵素は作用しないので、本酵素処理後においてもラフィノースは分解されることなくそのまま保持される。

したがって、本酵素で処理した後は、常法にしたがってラフィノースの精製処理を行えばよく、陽イオン交換樹脂クロマトグラフィー、高速液体クロマトグラフィー等のクロマト処理、結晶化処理その他常法にしたがってラフィノースを精製、分離することができる。
これらの処理は、バッチ式でもよいし反応カラムを用いて連続的に行ってもよい。酵素処理も同様であって、常法にしたがって固定化した又は固定化しない酵素を用いて、バッチ式ないし連続式に酵素処理することができる。
以下、本発明の実施例について述べる。

Streptomyces rochei E87(FERM P−15735)を、下記する(1)〜(3)の各培地を用いて、以下のように培養した。なお、培地は121℃、10分間オートクレーブした。

スラント培地に接種後、27℃、3日間培養。スラントから1白金耳を試験管の10mlGPYS培地に接種し、27℃、1日間振とう培養した。その培養液0.3mlを500ml坂口フラスコの30ml酵素生産培地に接種し、32℃、3日間振とう培養した。得られた培養液を遠心分離し上清を粗酵素液とした。

以下の組成の反応液を40℃で8時間反応させたのち、糖濃度を比較した。糖濃度は高速液体クロマトグラフィー(Shodex Ionpack KS801、80℃、水1ml/min、以後HPLC)で分析した。

ケストース類を含むラフィノース液を塩酸でpH6.2に調製し、実施例1の粗酵素液を等量加え、40℃で8時間攪拌し反応させた。結果は下記表2のとおりであった。

実施例1の粗酵素液を、セラミック担体を充填し40℃に保温したカラムにSV0.1で通液して、酵素を固定化した。ケストース類を含むラフィノース液を塩酸でpH6.2に調整して、この固定化カラムにSV0.1で通液した。結果は下記表3のとおりであった。

【発明の効果】
上記から明らかなように、本発明の酵素によるラフィノース液の精製処理法によって、クロマトグラフィーでラフィノースと分離が不良であったケストース類を分解し、ラフィノース純度を向上させるとともに、同収率を向上させた。その結果、食品分野におけるラフィノース需要の増加に対し、供給量を増やすことができる。

また、従来より甜菜糖の製造において不純物として扱われてきたラフィノースに対して、本発明はその有効利用の途を拓いたものであって、本発明はこの点においても卓越している。
本発明について、ケストース類を含有するラフィノース液であれば、甜菜製糖由来のラフィノース液に限定されることなく、すべてのラフィノース液に対して適用できることはいうまでもないことである。[0001]

Separation and purification of raffinose, a trisaccharide from sugar beet sugar process juice, is significant from the aspect of effective utilization of impurities (by-products) in sugar beet sugar as described above. In the cation exchange resin chromatography used for the separation of raffinose, the separation of the trisaccharide kestoses (fructosyl sucrose) and raffinose is poor, and during crystallization to improve the recovery rate in the raffinose production process. The present invention has been made for the purpose of newly developing an efficient method for producing raffinose in response to such a demand in the industry.
As a result of studying from various directions, the present inventors have found for the first time that an inulin-degrading enzyme derived from Streptomyces rochei E87 has an action of degrading kestoses. As a result of further studies, it was discovered for the first time that the enzyme selectively decomposes only kestoses (fructosyl sucrose) and does not act on raffinose in the presence of raffinose.
