2. Description of the Related Art Propagation of microalgae (Japanese Utility Model Application Laid-Open No. 5-95266)
Japanese Patent Application Laid-Open No. Hei 6-30666) and the technique of culturing Aosa (Japanese Patent Application Laid-Open No. 7-203789) have already been described. These are techniques for promoting vegetative propagation of microalgae or blue-green alga bodies, and have devised a method for actively promoting photosynthesis. The purpose of the present invention is to promote the formation of germ cells (sporulation) of the green algae Aosa, and is different from the above-described technique which aims to activate photosynthesis and vegetative growth of a blue alga body. As a method for cultivating Aosa, a method called the slice method has been conventionally used, and a method is known in which Aosa alga bodies are cut into several cm squares and cultured (algae research method, edited by Kazutoshi Nishizawa, Mitsuo Chihara,
Kyoritsu Shuppan Co., Ltd., pp. 99-104). However, this method is intended only for culturing (growing) Aosa and not for forming germ cells.
Further, a section of a blue-green alga body which is appropriately cut out because the size is not taken into consideration has the following three results. 1. The section grows in all directions. 2. Each cell of the section germinates. 3. Each cell matures and releases germ cells. As described above, germ cell formation is not always induced, and maturation occurs accidentally.

[0003] By the way, as a method for obtaining germ cells from the green alga Mothae: The larvae that have matured and form germ cells are searched for from among the green alga, Aocoptera that grows naturally, and germ cells are extracted. 2. Cultivate the cultured alga body until it matures. Cultured alga bodies tend to mature when a new medium is replaced, and extract germ cells generated at that time. 3. Germ cells may be formed in protoplasts when protoplasts are produced from the thallus (naked cells removed by removing the cell wall covering the cells by enzymatic treatment). The germ cells generated at this time are extracted. However, in any case, the necessary amount of germ cells cannot be extracted when needed.

DISCLOSURE OF THE INVENTION The present invention relates to a method for inducing the formation of germ cells in the order of the green alga Ausa which contains a large amount of carotene and nutrients in the course of the growth of the order Ala. It is intended to extract and harvest cells by forming a large number of cells at any time.

According to the present invention, the present inventors have found that maturation is induced by finely cutting the leaf-shaped body of the green alga, Moth during the research and observation of the ecology and growth process of the green alga, Moth.
That is, by cutting the foliates of the green algae Ausa from small pieces and culturing them in seawater, a phenomenon occurs in which a large amount of male and female gametes and germ cells as zoospores are released from the cut frond fragments, resulting in a large amount of reproduction. Cells could be extracted and harvested.

[0006] A large number of viruses and bacteria are attached to the frond of the wild green algae Aurasian under natural conditions, which may affect the maturation reaction. Therefore, it is desirable that the thallus of the green algae Auricularia used for inducing the formation of germ cells is a thallus grown by culture. Cutting from 0.6 to 15mm
It should be finely chopped to the size within and washed well. The shape of the cut may be circular, square, or triangular. In order to induce the formation of germ cells in the thallus of the green algae, the cut thallus pieces must be placed in a container in which seawater is circulating. When the density of the thallus pieces is high, the uptake of substances necessary for maturation and the supply of carbon dioxide and light in photosynthesis may be hindered. Since the maturation reaction is an enzymatic reaction, it greatly depends on the temperature. If the temperature is high and is 30 to 40 degrees Celsius or higher, the enzyme may be inactivated and not mature. Conversely, if the temperature is too low, such as 10 degrees Celsius or less, the maturation reaction is delayed. Mature fronds are clearly distinguishable from immature fronds by color. Mature leaf pieces exhibit a yellow or brownish green color. This is due to the increased carotene content, which is a measure of the start of germ cell release from the thallus pieces. The release of these germ cells is facilitated by some dry stimuli. That is, the mature frond fragments are once withdrawn from the seawater, dried for several tens of minutes to the extent that they do not dry out, and when they are returned to the seawater again, the germ cells are rapidly released.

[0007] The fact that the released germ cells of the green algae Aurosa have the property of collecting in response to light (phototacticity) is utilized. By irradiating the seawater with light, the germ cells released from the leaf-shaped pieces of the green algae are collected, and the seawater containing the germ cells is collected and filtered to collect a large amount of germ cells of the green algae Got a way to do it.

According to the present invention, the maturation of the thallus of the green algae Alausa is promoted, artificially releasing the germ cells of the green alga Aausa, and irradiating the released germ cells with artificial light. It is a method that can be collected and harvested, and enables the production of germ cells of the green algae Aosa from the required amount whenever necessary.

[0009] Germ cells are harvested by a method of inducing maturation using Anaoasa, a species of the green algae Moth, described in Examples. Using a cork borer, the frond of Anaanasa 1
FIG. 1 shows a state of punching out a disk having a diameter of 1.5 mm.
2 is a punched hole after Ana Aosa was punched with a cork borer. FIG. 2 shows a state in which the punched disc-shaped anaamosa leaf piece 3 is transferred to a beaker 4 containing sterilized seawater 5. At this time, the disc-shaped leaflet pieces 3 are thoroughly rinsed with the sterilized seawater 5, and the sterilized seawater 5 is replaced several times. Maturation is induced in the discoid leaflet pieces 3. Finally, the amount of seawater is 1.5 mm diameter disc-shaped frond pieces,
0.5 ml or more is sufficient. The beaker 4 containing the leaflet pieces 3 is placed in a culture chamber where the temperature and light can be adjusted. The temperature was maintained at 20 degrees Celsius, and the light used a white fluorescent lamp and 1000 lUX.
A light period and a dark period are created by the degree. The light period is about 6 hours. For example, if the leaf-shaped piece 3 is cut out at 12:00 in the daytime, the dark period is from 6:00 to 6:00 a.m. Under the conditions described above, the maturation of the leaflets 3 of Ana-aosa is completed at 12:00 noon in the morning on the second or third day after cutting. When left under light conditions, germ cells are released, and when light is applied from one direction to the beaker 4, germ cells are gathered due to phototacticity. The suspension was collected and recovered by a pipette or the like, and the suspension was filtered to collect germ cells. In the examples described above, completely the same results can be obtained in the case of human grasses and Aonori, which are a kind of anaosa.
