Abstract
Description
(i)初日のバースト放出は、境界電荷修飾界面ゾーンの二重電荷層内のカーゴ分子のより高い分配により、総送達の20%未満に減じられ得る。
(ii)送達装置は、より低いイオン強度の界面二重層ゾーンの収縮により(例えば、15mMのイオン強度の細孔流体の層の厚さは、150mMのイオン強度の層の3倍の厚さである)、少なくとも110mMのイオン強度を有する水性流体(例えば、150mMのイオン強度の涙液膜)と接触すると、要求に応じて自動的にカーゴ放出を開始できる。
(iii)装置ごとの薬物装填の総容量は、装置の水性多孔性、総表面積、表面電荷密度、及び界面二重電荷層の厚さを調整することによって正確に規定でき(細孔流体のイオン強度の平方根に反比例する)、結果的に、カーゴ送達の動力学は送達期間全体で正確且つ一貫している。さらに、正確な装填容量と放出速度により、装置は、過装填になることなく、意図した送達時間にちょうどいい量を収容できるように準備することができる。
「カーゴ分子」は、眼疾患を治療する機能又は健康を改善する機能のために眼系に送達可能な医薬又は非医薬化合物である。例えば、カーゴ分子は、薬物、ビタミン、又は栄養素である可能性がある。
本発明のコンタクトレンズ装置の親水性ポリマードメイン及び疎水性ポリマードメインは、親水性成分及び疎水性成分から作られる。1つの実施形態によれば、コンタクトレンズコンポジットの1つの成分は、表1A、1Bに列挙された親水性成分のいずれかから選択され、装置の親水性を高め、他の成分は、表1A、1Bに列挙された疎水性成分のいずれかから選択される。ナノコンポジットコンタクトレンズは、これらのアクリルオリゴマー又はプレポリマーから、熱硬化(100℃~120℃)工程によって作成され得る。
1つの実施形態によれば、親水性ポリマードメインは、疎水性ドメインの平均サイズよりも10分の1に小さく作られ、結果的に、疎水性ドメインが親水性ドメインと水性細孔の連続混合物に完全に浸されるように、複合材料の水親和性、接触角、及び毛管浸透を強化するために望ましい形態である。
(a)高分子ドメインは、高い光透過率を達成するために、5~50ナノメートルのサイズ、直径、又は最短軸(楕円形のドメインの3つの主軸の中で)を有し、
(b)界面ドメイン-細孔表面積は、延長された送達に十分なカーゴ化合物を収容するために5~500m2/gであり、
(c)水性細孔容積は、装置が150mMの通常の高イオン強度の涙液と接触した場合、送達デバイスの長期保存及び界面カーゴ分子のオンデマンド放出のために調製された低イオン強度(例えば、10mM未満)の水性流体を収容するために全装置容積の30~80%である。
本発明のコンタクトレンズ装置は、以下の望ましい生物医学的機能を有する:(a)眼科ケアの送達を目的とした医薬品/非医薬品カーゴの運送業者、(b)長期間(1~30日)カーゴの送達速度と精度を制御する放出装置、(c)設計された放出期間(数週間から1か月)にわたって眼の涙液と接触させた後、持続可能な放出を開始しながら、装置の棚保管中にカーゴが早期に浸出するのを防ぐ組み込みのトリガー機構。
本発明の装置では、境界荷電二重層は、(i)ポリマーと細孔の界面に境界電荷修飾剤を注入し、ここで、境界電荷修飾剤は、荷電頭基と、化学結合又は強力な物理的凝集のいずれかによってポリマー-細孔境界でポリマードメインに固定化される疎水性テールを有する分子であり、続いて(ii)反対に荷電したカーゴ分子を界面ゾーンに装填して、結合(スターン)及び拡散(デバイ)対イオン層を形成する、一連の工程で形成される。境界電荷修飾剤のポリマーへの結合力は、物理的又は化学的に結合されているかどうかにかかわらず、装置の使用寿命を通じて浸出を防ぐのに十分なほど強くなければならない(つまり、装置の寿命全体にわたって、浸出損失の合計が数パーセント未満になるようにする)。
境界表面電荷を組み込むことで、このような変更されたDCLで荷電カーゴ分子の分配を制御できる。反対に帯電した境界面の界面ゾーンでの荷電カーゴ分子の平衡分布は、以下に示す電気化学ポテンシャル方程式に従って、境界面電荷によって発揮される静的クーロンポテンシャルに比例して指数関数的に増加する可能性があり、ここでmはモル濃度、γは活量係数、zは電荷数、Fはファラデー定数である:

境界帯電表面の静電クーロンポテンシャルは、表面電荷密度と表面形状によって決まる。電荷密度は、界面に装填される境界電荷修飾剤の濃度を変更することによって調整できる。界面ゾーンでカーゴの電気化学ポテンシャルを下げるために必要な計算されたクーロンポテンシャルに従って、望ましい境界電荷密度がわかったら、荷電したカーゴ分子は、DCLをカーゴ装填溶液に浸すことによって界面ゾーンに装填できる。これらの2つの工程は、化学的に吸着された対イオンのスターン層と拡散性デバイイオン層を含む界面電荷二重層につながる。どちらも、反対の電荷間のクーロンポテンシャルにより大量のカーゴ分子を含む。DCLの細孔内のカーゴ分子の平衡分布は、反対に帯電した修飾剤の境界へのクーロン引力により、細孔流体よりも界面ゾーンではるかに高くなる。通常、スターン層には、境界面の電荷に強く(化学的に)結合しているイオンが含まれている。スターン層のクーロン引力は、kTの順で熱エネルギーよりもはるかに高く、ここでkはボルツマン定数である。デバイ層のカーゴ分子イオンへの表面電荷の引力は、シュテルン層によって遮蔽されるため、より弱い形の物理吸着(エネルギー~kT)になる。
デバイ層の電荷分布とそのような層の結果として生じる遮蔽されたクーロンポテンシャルは、ポアソン方程式を解いて拡散層の厚さ(デバイ長)を取得することで容易に計算できる。これはカーゴ分子の分配係数と放出速度に直接影響する。温度Tにおけるクーロンポテンシャルφの下での電荷の平衡分布は、以下である:
境界荷電修飾DCLによる荷電カーゴ種の吸着の界面分配及び容量は、細孔流体中の界面濃度に対する高い比率の界面濃度を有するDCL装置を設計するために容易に変更でき、そして従って、細孔流体からのカーゴ種のバースト放出が大幅に減少する。主なオプションは、境界電荷の装填密度を制御して、カーゴの界面分配を総カーゴ装填量の80%より高くすることである。第二に、カーゴ装填溶液のイオン強度も制限され、界面ゾーンの拡大された二重層により多くのカーゴ分子を含むためのより厚いデバイ層につながる可能性がある。
本発明は、本発明のコンタクトレンズと、装置の水性細孔のイオン強度の10%以内のイオン強度を有する保存流体とを含む保存パッケージを提供する。
表面注入修飾剤(境界電荷修飾剤)の界面電荷密度は、分子固有の電荷解離/会合定数(例えば、酸のpKa又は塩基のpKb)、疎水性テールの鎖長、及び細孔流体のpH値と相関している。疎水性鎖の長さは、解離/会合定数に直接影響するが、より重要なのは、バルクポリマードメインと細孔流体の間の修飾剤の溶解度分配である。どちらの影響も明らかにカーゴの放出速度に影響を与え、後者は装置の長時間の使用で電荷修飾剤の望ましくない浸出につながる可能性がある。
液体媒体のpHが1単位上昇又は低下すると、脂肪酸の陰イオンへの解離は、pKaの平衡方程式に従って約10倍変化する。脂肪酸の解離速度のこのような変化は、界面の表面電荷密度に比例して影響し、その結果、薬物の電気化学的ポテンシャルとコンタクトレンズからの放出速度に影響を及ぼす。
この例は、膨潤工程による界面電荷修飾剤の組み込みを示している。
この実施例では、コンタクトレンズ装置はHEMA 1-day ACUVUE Moist(登録商標)コンタクトレンズであり、脂肪酸はオレイン酸である。メタノールは膨潤には適した溶媒であるが、少量の残留物が目に有害になる可能性があるため、処理には適していない。エタノール中の従来のヒドロゲルpHEMAレンズの膨張は無視できるため、代わりに、HEMA相を膨張させることができる水/エタノール溶媒混合物を開発した。
図3及び4は、麻酔薬、TruEye(登録商標)及びOasys(登録商標)からのBUPの放出速度を示す。TruEye(登録商標)コントロールレンズは、装填されたBUPの80%を約5時間で放出した。オレイン酸を充填したTruEye(登録商標)の場合、TruEye(登録商標)13.5%OAレンズは300時間で80%のBUPを放出した。80%BUP放出の放出期間に基づくと、TruEye(登録商標)にオレイン酸が存在すると、BUPの送達が60倍に延長される。28.6%OAのTruEye(登録商標)の場合、レンズは450時間後に装填されたBUPの約60%しか放出しない。
1-day ACUVUE Moist(登録商標)コンタクトレンズでは、2.9 wt.%の最大オレイン酸装填が観察された。1-day ACUVUE Moist(登録商標)などの従来のヒドロゲルコンタクトレンズは、疎水性領域で構成されておらず、主にpHEMA親水性ポリマーで構成されている。その結果、これらのレンズはシリコーンヒドロゲルコンタクトレンズよりも含水量が高く、疎水性シリコーンドメインがないため、酸素透過性とオレイン酸などの疎水性分子の吸収が低下する。
図7及び8は、ヒスタミンH1受容体の比較的選択的で非競合的なアンタゴニストであるKTFの放出速度に対するオレイン酸の効果を示す。TruEye(登録商標)対照レンズは、50時間以内に装填されたKTFの80%を放出した。オレイン酸を装填したTruEye(登録商標)レンズは、TruEye(登録商標)5.1%OA、TruEye(登録商標)13.5%OA、及びTruEye(登録商標)28.6%OAで、それぞれ100時間後に装填されたKTFの40%、30%、及び18%のみを放出した。
この例は、境界陽イオン電荷が変更されたコンタクトレンズによる陰イオン薬物送達の拡張を示している。
図11、12、13及び14は、異なるイオン強度(I=167、500、1665mM)を有するPBS溶液中で調査された、TruEye(登録商標)及びDailies Total 1(登録商標)コンタクトレンズからのKTF及びTHCLの放出速度を示す。
図15、16、17及び18は、PBS又は脱イオン水中のTruEye(登録商標)及びDailies Total 1(登録商標)コンタクトレンズからのKTF及びTHCLの放出速度を示す。対照レンズとオレイン酸を装填したレンズの両方を調べた。オレイン酸を装填したレンズの場合、15mg/mLのオレイン酸浸漬濃度を利用した。この濃度で、オレイン酸の重量%は、ACUVUE TruEye(登録商標)及びDailies Total 1(登録商標)でそれぞれ2.5及び5.2%(オレイン酸の重量/未修正の乾燥レンズの重量)であった。
図21及び22は、異なる炭素鎖長を有する脂肪酸を充填したTruEye(登録商標)及びDailies Total 1(登録商標)コンタクトレンズにおけるKTF及びTHCLの取り込みを示す。図23、24、25、及び26は、PBS中のTruEye(登録商標)及びDailies Total 1(登録商標)コンタクトレンズからのKTF及びTHCLの対応する放出速度プロファイルを示す。対照レンズと脂肪酸を含むレンズの両方を研究した。レンズへの装填には、25mMの固定脂肪酸濃度を使用した。使用した5つの脂肪酸は、オクタン酸(C=8)、カプリン酸(C=10)、ラウリン酸(C=12)、ミリスチン酸(C=14)、オレイン酸(C=18)である。
図27、28、29、及び30は、異なるpHレベル(pH=5.5、6.4、及び7.4)のPBS溶液中で調査された、TruEye(登録商標)及びDailies Total 1(登録商標)コンタクトレンズからのKTF及びTHCLの放出速度を示す。
Claims (19)
- ナノコンポジットに埋め込まれたカーゴ分子を含むコンタクトレンズ装置であって、前記ナノコンポジットが、親水性ポリマードメイン、疎水性ポリマードメイン、水性細孔、及び境界荷電二重層を含み、ここで
(a)界面ドメイン-細孔表面積が5~500m2/g装置であり、
(b)水性細孔容積が装置の総容積の30~80%であり、
(c)カーゴ分子の少なくとも80%が、(i)水性細孔及び(ii)疎水性ポリマードメイン又は親水性ポリマードメインのいずれかの界面で形成される境界荷電二重層に分配され、
(d)帯電境界二重層が、0.005~0.5クーロン/m2の表面電荷密度を有し、そして
(e)カーゴ分子を含む水性細孔が、0.1~100mMのイオン強度、及び200~300mMの浸透圧を有する、装置。 - 境界荷電二重層の個々は、(i)境界電荷修飾剤の頭基の電荷、及び(ii)荷電分子又は荷電頭基に対する対イオンの電荷から形成され、ここで、境界電荷修飾剤は、荷電頭基及び疎水性尾部を有する分子であり、そして境界荷電二重層に固定化される、請求項1に記載の装置。
- 前記親水性ポリマーが、logP<1を有するモノマーから形成され、前記モノマーは、ヒドロキシ基、アルキルグリコール、アミン、ラクタム、カルボキシル基、及びスルホン基から成る群から選択される1つ又は複数の親水性基を含む、請求項1又は2に記載の装置。
- 前記モノマーが、2-ヒドロキシエチルメタクリレート(HEMA)、N,N-ジメチルアクリルアミド(DMA)、N-ビニル-2-ピロリドン(NVP)、4,4-ジメチル-2-ビニル-2-オキサゾリン-5-オン、メタクリル酸、N-(ヒドロキシメチル)アクリルアミド、N-[3-(ジメチルアミノ)プロピル]メタクリルアミド、又はエチレングリコールジメタクリレートである、請求項3に記載の装置。
- 前記疎水性ドメインが、logP>3を有するモノマー又はオリゴマーから形成された疎水性ポリマーを含み、ここで前記モノマーは、アルキル基、芳香族基、エステル基、ペルフルオロアルキル基から成る群から選択される1つ又は複数の疎水性基を含む、請求項1~4のいずれか1項に記載の装置。
- 前記モノマーが、3-[トリス(トリメチルシロキシ)シリル]プロピルメタクリレート(TRIS)、3-メタクリルオキシ-2-ヒドロキシプロポキシ(プロピルビス(トリメチルシリルオキシ)メチルシラン(SIGMA)、ジメチルシロキサン、モノメチルシロキサン、フルオロシロキサン、又はメチルメタクリレート(MMA)である、請求項5に記載の装置。
- 前記境界荷電修飾剤はブレンステッド・ローリー酸である、請求項1~6のいずれか1項に記載の装置。
- 前記境界荷電修飾剤が、8~24の炭素鎖長及び1つの酸頭部を有する脂肪酸であり、そして前記カーゴ分子が正に帯電される、請求項7に記載の装置。
- 前記境界荷電修飾剤が、オレイン酸、リノール酸、α-リノレン酸、ミリスチン酸、又はステアリン酸である、請求項8に記載の装置。
- 前記境界荷電修飾剤が、ブレンステッド・ローリー塩基である、請求項1~6のいずれか1項に記載の装置。
- 前記境界荷電修飾剤が、アルキルアミン、エタノールアミン、又は8~24の炭素鎖長及びアルキルアミン、エタノールアミン、又は第四級アンモニウム頭基を有するカチオン性第四級アンモニウム塩であり、そして前記カーゴ分子は負に帯電している、請求項10に記載の装置。
- 境界荷電修飾剤が、スフィンゴシン、スフィンガニン、フィトスフィンゴシン、又は1,2-ジオレオイル-3-トリメチルアンモニウムプロパンである、請求項11に記載の装置。
- 前記水性細孔が、イオン強度及び浸透圧を達成するために、親水性及び非イオン性である1つ又は複数の化合物を含む、請求項1~11のいずれか1項に記載の装置。
- 前記化合物が、グリセロール、ポリエチレングリコール、ポリビニルピロリドン、グリコシド、及びカルボキシメチルセルロースから成る群から選択される、請求項13に記載の装置。
- 前記ナノコンポジットが透明なナノコンポジットである、請求項1~14のいずれか1項に記載の装置。
- 前記親水性ポリマー及び疎水性ドメインのそれぞれが、5~50ナノメートルの楕円形のドメインの3つの主軸のうちの直径又は最短軸を有する、請求項1~14のいずれか1項に記載の装置。
- 請求項1~16のいずれか一項に記載の装置及び、前記装置の水性細孔のイオン強度の≦10%のイオン強度を有する貯蔵流体を含む貯蔵パッケージ。
- 装置からカーゴ分子を放出する方法であって、以下の工程:
請求項17に記載の保管パッケージから装置を取り出す工程、
少なくとも110mMのイオン強度を含む水性流体中に装置を配置する工程、及び
水性細孔から水性流体へのイオン強度の増加によって引き起こされる水性流体へ前記カーゴを放出する工程を含む方法。 - 前記水性流体が涙である、請求項18に記載の方法。
