細胞株
ヒト肝細胞癌(HCC)HepG2(ATCC(登録商標)HB-8065(商標))およびHep3B(ATCC(登録商標)HB-8064(商標))細胞は、ATCCから購入した。細胞を、5%CO2の雰囲気中37℃で、イーグル最小必須培地(EMEM)(Cellgro,USA)、10%FBS(HyClone,USA)、ストレプトマイシン(100μg/mL)およびペニシリン(100U/mL-1)(Cellgro)中で単層として培養した。HepG2細胞は、5μg/cm2のコラーゲン濃度でコ
ラーゲン(Gibco,USA)被覆したプレート上で増殖させた。
pMRNA-CTX-mRNA鋳型の構築
実験で使用した全てのmRNAのインビトロ合成用のプラスミドpMRNA-CTx-mRNA鋳型形成マトリックスは、市販のmRNAExpress(商標)mRNA合成キット(SBI,USA)に従って構築した。全てのプラスミドをE.coli(Invitrogen,USA)中で増殖させ、Qiagen MiniまたはMaxi Kit(Qiagen,USA)を使用して精製した。pDRAW32ソフトウェア(www.acaclone.com)を使用して、全てのプラスミドの制限地図を作成した。
図2に示すように、遺伝子の5’末端のATGコドンまたは3’末端の終止コドン(TAA、TAG、TGA)の直前で最適な翻訳のためにコザック配列と隣接した1つ以上の遺伝子の配列は、GeneArt社によって合成され(コドン最適化なし)、図2でpMA-T-CTx-Geneと示されるプラスミドDNA(DNAプラスミドまたはベクターと称される)として供給された。コード配列に隣接する独自の5’および3’UTR領域は、全ての合成配列に含まれていた(添付の配列には示されていない)。5’UTRは合成であり、コザック配列を含有し、3’UTRはマウスアルファグロビンUTRに基づき、また120塩基のポリAテールを含む。図2に示す合成ベクターをpMRNA-CTx-mRNAとして作製するために、単数または複数の遺伝子を含有するヌクレオチド断片を、制限エンドヌクレアーゼ、ここではEcoRIおよびNheIを使用してDNAプラスミドから切り出し、pMRNA鋳型プラスミドのEcoRI/NheI制限部位にサブクローニングした:この鋳型プラスミドは、T7 RNAポリメラーゼによって認識されるT7プロモーター、5’および3’UTR、ならびにポリA配列を含む。

miRNA結合部位配列を含むmRNA配列の生成(miR-122を使用した例が図3に示される)について、3つの変異体を作製する例示的な方法を以下に記載する。変異体1では、miRNA結合部位配列の2つのコピーが、終止コドンと3’UTRの+1位との間に含まれる。変異体2では、miRNA結合部位配列の2つのコピーが、3’UTRの開始点または5’末端に含まれ、変異体3では、miRNA結合部位配列の2つのコピーが、3’UTRの終了点または3’末端に含まれる。
図5に示すように、コザック配列および終止コドン(TAA、TAG、TGA)と上記の方法のように隣接し、さらに終止コドンの後にmiRNA結合部位配列の2つのコピーを含む1つ以上の遺伝子の配列は、GeneArt社によって合成され(コドン最適化なし)、プラスミド(ここでも同様にプロテインAを用いて示されている)DNA pMAT-CTx-Geneとして供給された:このプラスミドの骨格は上記と同じである。次いで、この配列を鋳型プラスミドにクローニングして、上記方法のいずれかにより合成ベクターを作製した。

miRNA修飾3’UTRを用いてまたは用いずにmRNAのIVTを行うために、市販のmRNAExpress(商標)mRNA Synthesis Kitを使用した。IVTベクターのDNA鋳型は、上記のプロトコルに記載される通りに構築した。インビトロmRNA合成の手順は、製造業者のプロトコルに従って行った。端的に述べると、特定の5’および3’プライマー(キットに付属)を用いたPCR反応を使用して、DNA配列にポリAテールを付加した。インビトロ転写中、DNA鋳型に合成されたmRNAを、アンチリバースキャップアナログ(ARCA)修飾ヌクレオチド(5-メチルシチジン-5’-三リン酸)でキャッピングした。キャップアナログ、プソイドウリジン-5’-三リン酸およびポリAテールをインビトロ転写mRNAに組み込み、宿主細胞の安定性
を高め、免疫応答を低下させた。
Combined Therapeutics社の製剤の送達および修飾のプラットフォーム(DMPCTx)は、イオン化可能な脂質様物質C12-200、リン脂質DOPE、コレステロール、および脂質固定ポリエチレングリコールC14-PEG-DSPE2000混合物の多成分ナノ粒子である。この特定のDMPCTxの組成、C12-200:mRNAの比重量比(10:1)、ならびに脂質様物質、リン脂質、コレステロール、およびPEGのモル[%]組成(表4)を最適化したところ、インビボで製剤の高い有効性が明らかになった(Kauffman K.J.,Nano Letter.2015,15,7300-7306)。これらの例示的な成分の化学構造を図8に示す。

標的細胞に構築物mRNAを成功裏にトランスフェクトし、続いて異なる細胞型で差次的発現を駆動する本発明の可能性を調査するために、miRNA-122結合部位で修飾されたDMPCTx mRNAプラットフォームを肝細胞癌のモデルで使用した。
蛍光イメージングと定量化
ヒト肝細胞癌株HepG2およびHep3Bの単一トランスフェクションを以下のように行った:トランスフェクションの1日前に、HepG2およびHep3B細胞をそれぞれ2.7x105/ウェルおよび2x105/の密度で12ウェルプレートに別々に播種した(EMEM/10%FCS)。翌日、PBS単独のビヒクル対照、0.5μg/ウェルのmRNA-mCherry-DMPCTx、または0.5μg/ウェルのmRNA-mCherry-122-DMPCTx(配列番号3を含む)のいずれかを細胞にトランスフェクトした。トランスフェクションは、mRNA-DMPCTxをウェル内の培養培地に直接添加し、必要に応じて培養細胞を穏やかに混合することにより行った。

miRNA-122は、豊富な肝臓特異的miRNAであり、その発現はヒト原発性肝細胞癌(HCC)、ならびにHep3BおよびHepG2等のHCC由来細胞株では著しく減少する。この試験の目的は、miRNA-122を標的とする配列(例えば、図3の上の変異体1に示されるような配列2)の挿入によるmRNA配列の3’非翻訳領域(UTR)の修飾が、正常な肝細胞において翻訳抑制および/または脱アデニル化、それに続く外因性mRNAのデキャッピングをもたらし得るが、試験したHCC細胞株ではもたらされないことを実証することであった。

-122のレベル低下はマウスの肝細胞癌と関連している(Kutay et al,2006)。したがって、マウスの健康な肝細胞株Aml12を使用してmiRNA-122活性の内因性効果を調べた。
別の実験において、ウエスタンブロッティングを用いて、トランスフェクション後に最終的に示されるタンパク質発現レベルを以下のように決定した。
例示的な25kDaのヒトタンパク質(「プロテインA」と表示)の腫瘍特異的発現レベルを評価するために、肝癌細胞(HepG2およびHep3B)と健康な肝細胞(HMCPP5)の両方を12ウェルプレートに播種し、実施例1でmCherryトランスフェクションについて前述したように、ヒトプロテインA(25kDa)を発現するナノ製剤化mRNA(mRNA-A-DMPCTx)、または3’UTRに2つのmiRNA122結合配列(配列番号2)を含むヒトプロテインA(約25kDaのヒトタンパク質)を発現するmRNAである変異体1(mRNA-A-miRNA122-DMPCTx)を0.5μg/ウェルでトランスフェクトした。トランスフェクションの24時間後、全タンパク質抽出後に免疫ブロットを行った。
ギ二次抗体(1:10000、Abcam)とともに1時間室温でインキュベートした。タンパク質抗体複合体は、それぞれClarity(商標)Western ECL Substrate(Bio Rad)およびLI-COR(登録商標)システム(LI-COR)を使用して視覚化および想像した。

mRNAの修飾は、mRNA翻訳を肝細胞癌Hep3Bおよび肝芽腫HepG2に著しく限定することができるが、正常なヒト肝細胞ではそうではない。
ここでは、本発明の方法によって可能になり、かつ上記の実施例に示される、提供されたmRNA構築物の差次的発現を、腫瘍溶解性ウイルス療法と組み合わせて使用することができることを記載する。特に、腫瘍溶解性ウイルスが病原性遺伝子を除去するように修飾され、健康な細胞におけるその複製能力を弱める場合、本発明を使用して、癌細胞等の罹患細胞でそれらの遺伝子またはその等価物の機能を回復することができる。この可能性を調査するために、US3を欠く腫瘍溶解性ウイルスHSV-1(R7041)(Leopardi et al,1997,PNAS 94;7891-7896を参照)と、US3をコードするmRNA構築物を提供し、miRNA-122結合部位で修飾されたDMPCTxプラットフォームとの組み合わせを、肝細胞癌モデルで使用した(配列番号4)。
細胞培養
ヒト肝細胞癌(HCC)HepG2およびHep3B細胞を、5%CO2の雰囲気中37℃で、イーグル最小必須培地(EMEM,Cellgro,USA)、10%FBS、ストレプトマイシン(100μg/mL)およびペニシリン(100U/mL-1)中で単層として培養した。HepG2細胞は、5μg/cm2のコラーゲン濃度でコラーゲン被覆したプレート上で増殖させた。
凍結したR7041ウイルスを37℃の水浴中で解凍し、バスソニケーター(Q500ソニケーター、Qsonica,USA)を使用して30秒間超音波処理し、次いで氷に移し、すぐに使用できる状態にした。
US3変異体R7041ウイルスは、健康な細胞に対して実質的に無病原性であると考えられ(Leopardi et al.1997)、免疫不全の無胸腺マウスにおいて良好な安全性さえ示している(Liu et al.2007,Clin Cancer
Res 2007;13(19))。肝細胞癌細胞に対するR7041ウイルスの有効性のベースラインを確立するために、モデル細胞株を腫瘍溶解性ウイルスのみで処理した。Hep3BおよびHepG2株からの細胞を、それぞれウェル当たり15,000および17,000で、96ウェルプレートに3通り播種した。
ヒト肝細胞癌に対するR7041ウイルスとmRNA-US3-DMPCTxの組み合わせ効果を評価する前に、0.04μg/mL mRNA-US3でのmRNA-US3-DMPCTxによるHep3BおよびHepG2細胞のトランスフェクションが、MTSアッセイにより測定された細胞生存率に対して著しい効果を与えないことを確認した。
A-US3-DMPCTx単独(y軸のバツ印)、様々な希釈率のR7041単独(灰色の三角形/ひし形)、および組み合わせ(黒丸)について生存率に対する影響が示されている。
インビボ手法に対する本発明の適用可能性を決定するために、同所性ヒト肝細胞癌のマウスモデルを使用した。miRNA-122結合部位によって駆動される差次的発現は、インビトロで健康なマウスAml12肝細胞に適用可能であることが上で示された(実施例1を参照)。
動物
6~8週齢の雌(CB17/Ics-PrkdcSCID/IcrIcoCrl)Fox Chase SCIDマウスは、Charles River,UKから購入した。全てのインビボ手順は、UKのCrownBioでSubcommittee on Research Animal Careによって承認された。
同所性HCCモデルを作製するために、ホタルルシフェラーゼを発現するヒトHep3B細胞株の生物発光変異体(Hep3B-cLuX)を使用した。細胞は、10%熱不活性化FBS、2mM L-グルタミン、1%NEAAを添加したEMEM培地(Sigma,UK)で培養した:2μg/mLのピューロマイシン(Sigma)で毎週細胞を処理した。
麻酔下で、20μLの1:1のPBS:Matrigel(商標)に懸濁したヒトHep3B-cLuX細胞(2×106)を、29G針を使用して肝臓の左葉上部に注射した。注射部位を吸収性ゼラチンスポンジ(AGS)で覆い、AGSを乱すことなく肝臓を腹腔内に戻し、皮膚を縫合して閉じた。生物発光イメージング(BLI)により、腫瘍増殖を週2回確認した。
mCherry配列を含むmRNA配列、およびmiRNA-122を含むmCherryの配列(配列番号3)を、「DMPCTxの合成とmRNAの製剤化」および表4で前述したように製剤化した。選択的腫瘍標的化と非罹患肝細胞の温存を評価するために、同所性肝癌を有するマウスの尾静脈に製剤化mRNAを注入した。端的に述べると、20μLの1:1のPBS:Matrigel(商標)に懸濁した2×106のヒトHep3
B-cLuX細胞を、上記のように肝臓の左葉上部に注射した。次いで、同じく上記のように、BLIイメージングによって腫瘍増殖をモニタリングした。8日後、腫瘍が確立されたとき(BLI≧6×106)、マウス当たり20μgの製剤化mRNA-mCherry-DMPCTx、mRNA-mCherry-122-DMPCTxまたはmRNA-A-122-DMPCTxを、尾静脈を介して注射し、血流を戻すことにより送達粒子を肝臓に取り込ませた。24時間後、最後のBLIを行い、マウスを安楽死させ、肝臓を切除し、局在化する肝臓病変でエクスビボBLIにより画像化した。
端的に述べると、エクスビボイメージングの後、腫瘍のある左肝葉を取り除き、2%PFAで固定し、4℃の30%ショ糖溶液(PBS中、pH7.4)に浸漬し、OCTに包埋し、ドライアイス浴で予冷したイソペンタン中で凍結させ、-80℃で保存した。5μm凍結切片(Leica CM300,USA)を、DAPI(VECTASHIELD、Vector Laboratories,USA)またはH&E(ヘマトキシリンエンドエオシン)染色による核対比染色に供した。腫瘍標的化は、蛍光顕微鏡および/またはソフトウェアを使用して、腫瘍および健康な肝臓におけるmCherry対mCherry-122の発現レベルを決定することにより評価した。腫瘍組織および健康な組織は、H&E染色によって決定した。
実施例4に記載のインビボマウスモデルを、US3 mRNA DMPCTx miRNA-122構築物を含む送達粒子組成物の投与に適用した。Hep3Bヒト癌を含有するマウスの肝臓におけるUS3の差次的発現を、抗US3ポリクローナル抗体を用いた免疫組織化学を使用して分析した。その結果を図18に示す。腫瘍細胞(濃い染色)と非罹患細胞(淡い染色)との間で、US3タンパク質レベルに目に見える違いがあることがわかる。病理学者によって独立して確認されたように、差次的発現は腫瘍の境界を描出する。したがって、本発明の組成物は、哺乳動物対象において潜在的な治療増強因子の差次的発現をインビボで成功裏に駆動できると結論付けることができる。
新鮮な凍結切片を5μm(ミクロン)で切断し、約1時間風乾した後、室温(RT)で15分間、4%パラホルムアルデヒドで固定した。水道水を流して切片を洗浄し、PBS-0.1%Tweenに移した。切片を2.5%正常馬血清(すぐに使用できる、ImmPRESS HRP抗ウサギIgGペルオキシダーゼポリマー検出キット、Vector
MP-7401)とともに20分間インキュベートした。スライドの水気を切り、1:400に希釈したUS3に対する一次抗体(Acris AP55266SU-N)とともにインキュベートした。抗体をPBS-0.1%Tweenで希釈し、一次抗体を省いた陰性対照を含め、スライドを抗体希釈液PBS-0.1%Tweenとともに1時間室温でインキュベートした。スライドをPBS-0.1%Tweenで洗浄し、ELGA社製の装置で精製した水で希釈した0.3%過酸化水素で内因性ペルオキシダーゼを10分間ブロックした。スライドをPBS-0.1%Tweenで洗浄し、ImmPress抗ウサギIgG試薬(すぐに使用できる、ImmPRESS HRP抗ウサギIgGペルオキシダーゼポリマー検出キット、Vector MP-7401)とともに30分間室温でインキュベートした。スライドをPBS-0.1%Tweenで洗浄し、色素原ImmPACT DAB(ImmPACT DABペルオキシダーゼ(HRP)基質、Vector SK-4105)とともに5分間インキュベートし、次いで、ELGA社製の装置で精製した水で洗浄し、Mayerのヘマトキシリンで必要に応じて対比染色した。ELGA社製の装置で精製した水でさらに短時間の洗浄を行い、5分間水道水を流して青色に染色した。スライドを脱水し、透明化し、マウントし(95%IMS、99%IMS×2、キシレン×2)、次いでカバースリップで覆った。

前記miRNA結合部位配列は、前記非罹患細胞と前記罹患細胞との間でコード配列の差次的発現を提供し、

(i)TNFα、TNFβ、IFNα、IFNβ、IFNγ、IL1、IL2、IL3、IL4、IL5、IL6、IL7、IL8、IL9、IL10、IL11、IL12、CCL2、CCL3、CCL4、CCL5 CXCL9、およびCXCL10のうちの1つ以上から選択される免疫応答および炎症に関与するサイトカイン、
(ii)GM-CSF、TLR7およびTLR9のうちの1つ以上から選択される樹状細胞活性化剤、
(iii)CD40、CD40L、CD160、2B4、Tim-3、GP-2、B7H3およびB7H4のうちの1つ以上から選択されるそれらの細胞受容体およびそれらのリガンドを標的とする分子、
(iv)TGF-β阻害剤、
(v)T細胞膜タンパク質3阻害剤、
(vi)プログラム死1(PD1)、プログラム死リガンド1(PDL1)、プログラム死リガンド2(PDL2)、細胞傷害性Tリンパ球抗原4(CTLA4)、およびリンパ球活性化遺伝子3(LAG3)の阻害剤、ならびに

- 前記腫瘍溶解性ウイルスは、水疱性口内炎ウイルス、マラバウイルス、ポリオウイルス、レオウイルス、麻疹ウイルス、ニューカッスル病ウイルス、コクサッキーウイルスA21、パルボウイルス、単純ヘルペスウイルス1型、およびアデノウイルスのうちの1つ以上を含む群から選択される、請求項50~56のいずれか一項に記載の方法。
- 前記腫瘍溶解性ウイルスは、単純ヘルペスウイルスである、請求項50~57のいずれか一項に記載の方法。
- 前記コード配列は、US3をコードする、請求項58に記載の方法。
- 前記mRNA配列は、配列番号4を含む、請求項59に記載の方法。
- 前記コード配列は、ICP6をコードする、請求項58~60のいずれか一項に記載の方法。
- 前記mRNA配列は、配列番号5を含む、請求項61に記載の方法。
- 前記罹患細胞は、新生物細胞を含む、請求項50~62のいずれか一項に記載の方法。
- 前記標的器官は、肝臓、脳、肺、乳房、および膵臓からなる群から選択される、請求項50~63のいずれか一項に記載の方法。
