Abstract
【課題】 C-Met調節用の組成物及び肝細胞再生を促進するための用途を提供することを課題とする。【解決手段】 本発明は、C-Met調節用の組成物及び肝細胞再生を促進するための用途を開示する。前記C-Met調節用の組成物の主要成分は、Antrodin A及び/又はDMB(4,7-Dimethoxy-5-methyl-1,3-benzodioxole)を含有し、肝細胞内の細胞の再生と修復に関連する遺伝子発現(c-Met、Notch等)を効果的に調節することで、損傷した肝細胞の修復及び肝細胞の再生を促進する効果を奏する。【選択図】 なし
Description
本発明は、肝疾患の治療に関連する組成物に関し、特に、C-Met調節用の組成物及び肝細胞を促進するために用いられる用途に関する。
肝臓は、人体の代謝反応において中心的役割を果たし、大部分の化学物質が全て肝臓に入り代謝するため、肝臓に有害な物質又はその代謝物の影響を容易に与え、肝機能が損なわれ、有害な物質又はその代謝物以外に、肝臓もウイルスに感染されて肝炎、肝硬変或いは肝癌等を発症していた。
肝疾患の初期症状は明らかではなく、かつ肝臓には痛みなどを感じる神経がないため、大部分の患者は、肝機能が異常であるか、又はウイルスに感染されたことに気付かない。したがって、肝疾患を有する大部分の患者は、何らかの症状が現れたときには、かなり病状が進んでいるため臨床上の治療の難易度が増していた。肝炎の治療を例にすると、現在臨床上、病気の原因に応じた薬物療法又は対症療法があり、例えばウイルスによる肝炎の場合、抗ウイルス薬が投与され、脂肪肝による肝炎の場合、体重管理を介して肝炎の悪化を遅らせるが、肝炎が悪化し続けると、肝細胞を壊死させて肝機能に不可逆的な損傷を与え、現在既存の薬物を投与しても回復に難しい。
本発明の主要目的は、肝損傷関連疾患の治療効果を奏するため、肝細胞の修復と再生に関連する複数の遺伝子発現(c-Met、Notch等)を調節することによって、損傷した肝細胞を効果的に修復すると共に肝細胞の再生を促進するC-Met調節用の組成物及び肝細胞を促進するための用途を提供することである。
本発明の副次的な目的は、肝細胞の成長を促進する効果を奏するため、C-Met調節用の組成物及び肝細胞を促進するための用途を提供することである。
これ故に、上記目的を達成するため、本発明は、C-Met調節用の組成物を開示する。その主成分が有効量のDMB化合物(4,7-Dimethoxy-5-methyl-1,3-benzodioxole)及び担体を含有し、前記担体が薬学的に許容される担体又は食品産業上許容される担体であり得る。本発明により開示されるC-Met調節用の組成物を肝疾患に罹った患者に投与することにより、肝臓損傷の状況を効果的に改善できることで、肝疾患の治療効果を奏することができる。
本発明の一実施形態において、前記DMB化合物の濃度は、1.25~20ppmの範囲である。
本発明の別の実施形態において、前記C-Met調節用の組成物は、有効量のAntrodin Aをさらに含有し、かつその濃度が8.75ppm以上140ppm未満である。
より良好な肝臓修復効果を奏するため、DMBとAntrodin Aの濃度比は、1:2~1:10であり、DMBとAntrodin Aの濃度比が1:5~1:10であることが好ましい。
本発明の別の実施形態は、肝細胞再生を促進する組成物としてDMB化合物又は/及びAntrodin Aを調製しすることであり、肝細胞の修復及び再生に関連する複数の遺伝子発現(c-Met、Notch等)を調節し、細胞内のフリーラジカル又は酸化ストレスを低減することから損傷した肝細胞を修復し、正常肝細胞の活性を維持し、再生を促進することで、肝細胞の損傷に関連する疾患の治療効果を奏することができる。
前記DMBの濃度は、1.25~20ppmの範囲であり、前記Antrodin Aの濃度が8.75ppm以上140ppm未満であり、前記肝細胞の再生を促進する組成物にDMB及びAntrodin Aを同時に含有する場合、DMBとAntrodin Aの濃度比は1:2~1:10であり、DMBとAntrodin Aの濃度比は1:5~1:10であることが好ましい。
本発明の実施形態において、前記肝細胞の再生を促進する組成物は、食品、健康食品又は医薬組成物であり得る。
本発明で開示されるC-Met調節用の組成物の主成分は、Antrodin A及び/又はDMB(4,7-Dimethoxy-5-methyl-1,3-benzodioxole)を含有し、肝細胞内の細胞再生及び修復に関連する遺伝子発現(c-Met、Notch等)を効果的に調節することで、損傷した肝細胞の修復及び肝細胞再生の促進の効果を奏することができる。
ここで、c-Met及びNotchは、各々肝細胞の修復及び再生の制御経路に関連する遺伝子である。肝細胞が肝毒性薬物によって刺激されるとc-Metの発現量が減少し、c-MetのダウンレギュレーションによりリガンドHGFとの結合が減少され、肝細胞の修復及び再生のメッセージ伝達を下がる。Notchの発現量は、肝細胞の複製に関連し、肝細胞内のROSの含有量を低下させて、肝細胞保護効果を奏することができ、すなわち、肝細胞内のNotch発現量が増加すると、肝細胞増殖の促進及び肝損傷改善の効果を奏することができる。
本発明で開示されるAntrodin A及びDMBは、化学合成法で調製することができ、真菌の菌糸体から抽出や精製して得ることもでき、例えばベニクスノキタケ菌糸体粉末をHPLC分析したところ、図1に示すように、その中にAntrodin A(76~77分頃の保持時間にピークが出現する)及びDMB(32~33分頃の保持時間にピークが出現する)が含有することが分かる。ここで使用するカラム:C18カラム(Merck Purospher STAR,5μm,4.5×250mm)、流速:1.0mL/分、温度:30℃、注入量:10μL、検出時間:120分間、移動相:0.1%ギ酸(A)及び100%アセトニトリル(B)、波長:254nmである。
言及すべき点は、Antrodin A及びDMBはベニクスノキタケから抽出・精製されることができ、先行技術文献ではベニクスノキタケ抽出物ががん細胞に対して細胞傷害活性を持つが、本発明で開示されるC-Met調節用の組成物の主要効用は、細胞を保護及び修復し、細胞の再生を促進することであり、すなわち本発明で開示されるC-Met調節用の組成物は、細胞に対して毒性が生じず、さらに細胞殺傷能力がないことである。
本発明で開示されるc-Met及びNotchは、各々肝細胞の修復及び再生の制御経路に関連する遺伝子である。具体的には、肝細胞が肝毒性薬物によって刺激されるとc-Metの発現量が減少し、c-MetのダウンレギュレーションによりリガンドHGF之との結合を減少させて、肝細胞の修復及び再生のメッセージ伝達を下がる。Notchの発現量は、肝細胞の複製に関連し、肝細胞内のROSの含有量を低下させて、肝細胞保護効果を奏することができ、すなわち、肝細胞内のNotch発現量が増加すると、肝細胞増殖の促進及び肝損傷改善の効果を奏することができる。
以下、本発明の技術的特徴及び効果を説明及び検証するため、いくつかの実施例を挙げて図表を参照しつつ、さらに説明する。
以下の実施例内で使用される細胞は、本発明の属する技術の分野における通常の知識を有する者が容易に入手できるものであるため、別途特許の寄託は必要とされない。
以下の実施例内で使用されるAntrodin A及びDMBは、化学合成法で調製して得られたものであり、化学合成法が本発明の属する技術の分野における通常の知識であるため、ここでその説明を省略する。
(実施例1:細胞活性の分析(一))
FL83Bマウスの正常肝細胞を採取していくつかの群(FL83B 1×104cells/well)に分け、各群の細胞量をそれぞれ異なる条件で処理してから24時間培養した後、市販MTS細胞活性分析キット(CellTiter-Glo(R) (登録商標) One Solution Assay)で各群の細胞活性を分析した。各群の細胞の処理条件及び対応する生存率を次の表1~表3に示す。
FL83Bマウスの正常肝細胞を採取していくつかの群(FL83B 1×104cells/well)に分け、各群の細胞量をそれぞれ異なる条件で処理してから24時間培養した後、市販MTS細胞活性分析キット(CellTiter-Glo(R) (登録商標) One Solution Assay)で各群の細胞活性を分析した。各群の細胞の処理条件及び対応する生存率を次の表1~表3に示す。
下記表1の結果からも分かる通り、Antrodin Aの濃度が140ppmの時、肝細胞に対して毒性があり、肝細胞の生存率の低下が生じ、また濃度が8.75~70ppmの時、正常肝細胞の活性及び生存率を効果的に維持及び向上できた。表2の結果からも分かる通り、各濃度のDMBを添加すると、正常肝細胞に毒性が生じないだけでなく、肝細胞の成長も助ける。
表1と表3の結果を比較すると、Antrodin Aの添加濃度が同じ条件下で、DMBを併せて添加した正常肝細胞の生存率は、Antrodin Aのみが添加された細胞生存率より高いことが分かり、Antrodin Aの濃度が細胞毒性を生じない時、DMBを添加した場合細胞活性の向上を助け、Antrodin Aの濃度が細胞毒性を生じさせた時、DMBを添加した場合、肝細胞をAntrodin Aで生じた細胞毒性影響から保護し、細胞の活性及び生存率を効果的に向上できることを示している。
(実施例2:細胞活性の分析(二))
FL83Bマウスの正常肝細胞を採取していくつかの群(FL83B 1×104cells/well)に分け、各々先に10mMアセトアミノフェン(Acetaminophen)で24時間処理し、肝細胞が損傷したように見えるようにさせ、アセトアミノフェンを除去し、次に異なる濃度のAntrodin A及び/又はDMBで24時間処理した後、市販MTS細胞活性分析キット(CellTiter-Glo(R) (登録商標) One Solution Assay)で各群の細胞活性を分析し、各群の細胞の処理条件及び対応する生存率を下記表4~表6に示す。
FL83Bマウスの正常肝細胞を採取していくつかの群(FL83B 1×104cells/well)に分け、各々先に10mMアセトアミノフェン(Acetaminophen)で24時間処理し、肝細胞が損傷したように見えるようにさせ、アセトアミノフェンを除去し、次に異なる濃度のAntrodin A及び/又はDMBで24時間処理した後、市販MTS細胞活性分析キット(CellTiter-Glo(R) (登録商標) One Solution Assay)で各群の細胞活性を分析し、各群の細胞の処理条件及び対応する生存率を下記表4~表6に示す。
表4及び表5の結果から分かる通り、Antrodin A或いはDMBの添加は、損傷した肝細胞の修復を助け、かつ肝細胞の活性を増加させることができる。DMBの濃度がAntrodin Aより低い場合、損傷した肝細胞を修復する良い効果を有する。
次に、表6の結果から分かる通り、同時にAntrodin A及びDMBで損傷した肝細胞を処理すると、肝細胞への修復効果は、Antrodin Aのみで処理した場合よりも優れ、かつDMBとAntrodin Aの濃度比が1:2~1:10の場合において損傷した肝細胞の活性及び生存率の向上を助けることができ、ここで、DMBとAntrodin Aの濃度比が1:5~1:10の場合、損傷した肝細胞を修復する効果が高くなった。
(実施例3:遺伝子発現の分析)
Real-time qPCRキットで実施例2における異なる濃度のAntrodin A及びDMBで処理された細胞内の遺伝子発現を分析し、結果を下記表7~表9に示す。

表7~表9の結果から分かる通り、Antrodin A及びDMBは、いずれも損傷した肝細胞がc-Met及びNotchの発現量を増加させるのを助け、損傷した肝細胞の修復及び再生を加速する効果を奏し、かつAntrodin A及びDMBで細胞を同時に処理すると、損傷した肝細胞内のc-Met及びNotchの発現量はAntrodin Aのみで処理した場合よりも有意に優れ、DMBがc-Met及びNotchを調節する能力は高いことを示している。
実施例1~実施例3の結果をまとめると、本発明で開示されるC-Met調節用の組成物は、肝細胞に対して細胞毒性が生じないだけではなく、損傷した肝細胞の修復及び肝細胞再生促進の効果を奏することができるため、有効量のC-Met調節用の組成物を肝疾患の患者に投与した場合、肝臓の損傷を治療し、肝機能向上の効果を奏することができる。
(実施例4:安全性試験)
本実施例において慢性B型肝炎にかかっている計29名の被験者を2つの群に無作為に分け、ここで、対照群(n=14)は、C-Met調節用の組成物を投与したが、抗ウイルス薬entecavirを投与せず、治療群(n=15)が抗ウイルス薬entecavir及びC-Met調節用の組成物を投与した。
本実施例において慢性B型肝炎にかかっている計29名の被験者を2つの群に無作為に分け、ここで、対照群(n=14)は、C-Met調節用の組成物を投与したが、抗ウイルス薬entecavirを投与せず、治療群(n=15)が抗ウイルス薬entecavir及びC-Met調節用の組成物を投与した。
本実施例で投与されたC-Met調節用の組成物は、614.01ppmのDMB及び3937.39ppmのAntrodin Aを含有し、すなわち本実施例で投与されたC-Met調節用の組成物内のAntrodin AとDMBの比率が6.5:1程度である。薬投与期間は、48週間であり、投与治療開始後の12、24、36、48、60、72週目の治療効果及びウイルス学的再発(virologocal replapse)をそれぞれ検出及び記録し、結果を下記表10及び表11に示し、表内の空白はデータがないことを意味する。さらに、治療群において、治療中及び治療後のB型肝炎ウイルスのe抗原の有無の患者の治療効果及び再発状況を分析し、結果を表12に示し、表内の空白はデータがないことを意味する。表10~表12内のウイルス学的再発とは、ヌクレオチド抗B型肝炎ウイルス薬物の治療終了後少なくとも3ヶ月間隔で2回連続してB型肝炎-DNAが2000IU/mLを超えたと定義される。ウイルス学的再発とは、ヌクレオチド抗B型肝炎ウイルス薬物の治療終了後、ALTが正常値の2倍以上上昇し、かつB型肝炎-DNAが2000IU/mLを超えたと定義された。
また、治療群と対照群の血液及び尿を収集し、AST、ALT、ビリルビン(bilirubin)、アルカリホスファターゼ(ALK-P)、r-GT(Gamma-glutamyl transferase)、AFT(Alpha-Fetoprotein)、PT(Prothrombin Time)、APTT(Activated Partial Thromboplastin Time)、アルブミン(Albumin)、グロブリン(Globulin)、腎臓BUN、クレアチニン(Creatinine)、空腹時血糖値(GlucoseAC)、グリコシル化ヘモグロビン(HbA1c)、白血球、赤血球、ヘモグロビン、ヘマトクリット(HCT)、赤血球分布幅(RDW)、血小板、平均赤血球容積(MCV)、平均赤血球ヘモグロビン量(MCH)、平均赤血球ヘモグロビン濃度(MCHC)、平均血小板容積(MPV)、尿比重(specific gravity)、尿pH、尿路上皮細胞数を検出し、さらに2つの群の各検出指標を分析し、結果を表13~表41に示す。
上記結果は、本発明で開示されるC-Met調節用の組成物の長期投与が、ウイルス性肝炎患者の健康に悪影響を及ぼさないことを示している。言い換えれば、本発明で開示されるC-Met調節用の組成物は、確実に安全な肝臓保護成分であり、抗ウイルス薬の補助薬として使用できる。
Claims (6)
- 少なくとも1つの肝臓修復に関連する遺伝子の発現を促進する組成物の調製におけるDMB(4,7-Dimethoxy-5-methyl-1,3-benzodioxole)の使用であって、
前記肝臓修復に関連する遺伝子は、c-Met及び/又はNotchである、使用。 - 前記DMBの濃度は、1.25~20ppmの範囲である、請求項1に記載の使用。
- 有効量のAntrodin Aをさらに含有し、かつその濃度が8.75ppm以上140ppm未満である、請求項1に記載の使用。
- 前記DMBと前記Antrodin Aの濃度比は、1:2~1:10である、請求項3に記載の使用。
- 前記DMBと前記Antrodin Aの濃度比は、1:5~1:10である、請求項4に記載の使用。
- 肝臓修復に関連する遺伝子の発現を促進する前記組成物は、肝臓保護組成物である、請求項1に記載の使用。
