以下のようにして、RGMBを指標として美白用素材のスクリーニングを行った。

(材料)
使用細胞:ヒトiPS細胞由来感覚神経前駆体細胞(AXOL Bioscience社)
使用培地:播種培地、維持培地(AXOL Bioscience社)
RGMBの定量方法:ELISA(ポリクローナル ウサギ抗ヒトRGMB抗体(LifeSpan Biosciences社製)を使用)
試験対象であるエキスは、下記表に示す。

Cells used: Human iPS cell-derived sensory neuron precursor cells (AXOL Bioscience)
Medium used: seeding medium, maintenance medium (AXOL Bioscience)
RGMB quantification method: ELISA (using polyclonal rabbit anti-human RGMB antibody (manufactured by LifeSpan Biosciences))
The extracts to be tested are shown in the table below.
(方法)
(1) ヒトiPS細胞由来感覚神経前駆体細胞を1x105 cells/ウェルずつ48ウェルプレートに播種し、メーカーのプロトコールに従ってDay24まで培養し、感覚神経細胞へ誘導
(2) Day25:培養上清を半量回収し、エキス含有培地(エキス添加群)または溶媒含有培地(溶媒対照群)を半量添加、3日間培養
(3) Day28:培養上清を半量回収、細胞毒性がないことを目視にて確認
(4) Day25およびDay28の培養上清中のRGMB量をELISAキットにて定量
(5) 感覚神経細胞の3日間のRGMB分泌量を解析し、エキスの影響を評価
(Method)
(1) Human iPS cell-derived sensory neuron progenitor cells were seeded in 48-well plates at 1x10 5 cells/well, cultured until Day 24 according to the manufacturer's protocol, and induced into sensory neurons.
(2) Day 25: Collect half of the culture supernatant, add half the extract-containing medium (extract-added group) or solvent-containing medium (solvent control group), and culture for 3 days.
(3) Day28: Collect half of the culture supernatant and visually confirm that there is no cytotoxicity
(4) Quantify the amount of RGMB in the culture supernatant on Day 25 and Day 28 using an ELISA kit
(5) Analyze the amount of RGMB secreted by sensory neurons for 3 days and evaluate the effect of the extract
(結果)
図2および3に示すように、感覚神経細胞のRGMB分泌量を抑制する物質として、スギナエキス、ヒメフウロエキス、イチョウエキス、イザヨイバラエキス、オウバクエキスおよびボタンエキスを選択することができた。
また、図4に示すように、感覚神経細胞のRGMB分泌量を抑制する物質として、ホーリーバジル抽出物、およびインディアンデーツを選択することができた。
(result)
As shown in FIGS. 2 and 3, we were able to select Equisetum extract, Himefuuro extract, Ginkgo biloba extract, Rose rose extract, Phellodendron bark extract and Botan extract as substances that suppress the amount of RGMB secreted by sensory nerve cells.
Moreover, as shown in FIG. 4, it was possible to select holy basil extract and Indian dates as substances that suppress the amount of RGMB secreted by sensory nerve cells.
