- 緑内障は、とりわけ米国において失明の主な原因である疾患の群である。緑内障は通常、眼球内の流体圧がゆっくりと上昇し、視神経を損傷する場合に起こる。
- 視神経炎は視神経の炎症である。その原因には、感染および多発性硬化症等の免疫関連病が含まれる。原因が不明なこともある。
- 視神経委縮は視神経への損傷である。その原因には眼球への血流の不足、疾患、傷害、または毒物への曝露もしくは薬物[エタムブトール、イソニアジド、ディジタリス、抗生剤(クロラムフェニコール、スルホンアミド)およびアミオダロン]による誘起が含まれる。

本発明は、哺乳動物対象における眼科障害の処置および/または防止のための薬剤を提供する。薬剤は、その薬剤がたとえば疾患または障害の症状の改善において有益である場合に、対象への投与において使用することができる。特に、本発明において有用な薬剤は、配列番号3または配列番号4のポリペプチドである。即ち、本発明は特に、治療における使用のための配列番号3または配列番号4のポリペプチドを提供する。治療は典型的には本明細書で以下に記載するようにヒトまたは動物の身体に前記ポリペプチドを投与することを含む。
好ましくは、処置および/または防止は、ポリペプチドが投与されているかまたは投与されていた対象に副作用または有害影響を惹起しない。即ち、好ましくは、本発明のポリペプチドの投与は哺乳動物対象においていかなる望ましくない影響の原因にもならない。
好ましくは、本発明による使用のためのポリペプチドは、内因性(たとえばヒト)NGFに関する特定の薬剤によって選択的に認識され得る。用語「選択的に認識される」および「検出可能な」は、本明細書では相互交換可能に使用され、一般に、生体試料中のタンパク質の、好ましくは分子的手段による特異的同定を指す。
本発明によれば、本発明によるポリペプチドは、眼科障害の処置および/または防止における使用のためのものである。そのような使用およびその全ての実施形態のため、配列番号4のポリペプチドが特に好ましい。
本発明によれば、配列番号3または配列番号4のポリペプチドは、そのような投与を必要とする対象に投与してよい。そのような投与を必要とする対象は、本明細書に記載した障害に罹患した対象、そのような障害に罹患するリスクのある対象、またはその他そのような障害に苦しむ対象であってよい。薬剤は治療有効量で対象に投与される。治療有効量は本明細書の開示に鑑みて臨床医によって決定することができる。
本発明による処置または防止は、本発明のポリペプチドの投与、好ましくは局所投与によって行なってよい。本発明によれば、対象の眼は本発明のポリペプチドの優先的な投与部位である。一般に、本明細書においてポリペプチドが対象の眼に投与されるという場合には、文脈によって他が指示されない限り、そのような投与は眼の表面上、または眼の中であってよい。
本発明は、対象への投与のための異種ポリペプチドを提供する。
一般に、本発明によるポリペプチドは、繰り返しまたは単回投与で投与される。
本明細書に記載した薬剤および組成物は、有効量で投与される。本発明によれば、「有効量」は、単独でまたはさらなる用量とともに、所望の反応または所望の効果を達成する量または用量である。特定の障害の処置の場合には、所望の反応は、好ましくは疾患の経過の阻害に関連する。これは、疾患の進行の遅延および好ましくは疾患の進行の妨害または逆行を含む。疾患または状態の処置における所望の反応は、前記疾患または前記状態の発症の遅延または発症の防止も含み得る。一部の実施形態では、所望の反応は、障害の症状の局所的および/または全身的に完全な治癒を含む。
その他のパラメーターに依存することになる。したがって、本明細書に記載した薬剤の投与用量は種々のそのようなパラメーターに依存し得る。患者の反応が当初の用量では不十分な場合には、より高い用量(または異なる、より局所的な投与経路によって達成された効果的に高い用量)を使用してよい。
一実施形態では、配列番号3のポリペプチドおよび配列番号4のポリペプチドは生物源から得られる。配列番号3のポリペプチドおよび配列番号4のポリペプチドは組み換え発現によって得てもよい。その目的のため、それぞれのポリペプチドをコードするオープンリーディングフレームが、組み換えタンパク質の源、たとえば宿主細胞またはタンパク質発現のための無細胞系に導入される。まさに、ヒトNGFがインビボで少量しか産生されないこと、マウスのNGFが通常、種々のタンパク質の不均一な混合物として産生されること(WO2000/022119A1を参照)、および本発明のポリペプチドが天然でなく、したがってインビボでは全く産生されないことを考慮すれば、本発明のポリペプチドを産生する最も有意義な可能性は、最新技術における野生型NGFについての等価の示唆に従って組み換え発現によることである(WO2000/022119A1、WO2008/006893A1、Rattenholl et al., Eur. J. Biochem, 2001, vol. 268, p. 3296-3303、米国特許公開2018/0086805A1)。しかし、そのようなポリペプチドを哺乳動物への投与のために十分な純度グレードで得ることは変わらぬ課題であった。本明細書で詳細に開示したように、この課題は本発明によって克服された(実施例1および2も参照)。

本発明のポリペプチドはその天然の状態で通常随伴する成分を実質的にまたは本質的に含まない。本発明のポリペプチドは、投与される前に単離される。一実施形態では、「単離されたポリペプチド」は、天然に存在する状態でそれを取り囲む組織等の細胞および細胞外の環境、たとえばそれが発現された宿主細胞等の細胞から精製されたポリペプチドを指す。別の実施形態では、「単離されたポリペプチド」はそれぞれ、その天然の細胞環境から、およびポリペプチドが通常存在する環境の他の成分との会合からのポリペプチドのインビトロの単離および/または精製を指す。
以下、本発明のポリペプチドを含む組成物について記述する。そのような組成物はそのまま、ならびに本発明による眼科障害の防止および/または処置における使用の特定の文脈において、本発明の一部である。即ち、本発明はそのような組成物の医療用途と組成物そのものの両方を指向している。

他に特定しない限り、以下の実験例は具体的に、野生型ヒトNGFに対するP61S R100Eの置換によって特徴付けられる配列番号4のポリペプチド(「NGF P61S R100E」と命名。Malerba et al. PLOS One, 2015, vol. 10, e0136425、配列番号4)、ならびにそのプロ形その他に関する。配列番号4のポリペプチドは「NGFムテイン」と称してもよいが、本発明により、また実験例、特に実施例4で実証されるように、このタンパク質に特異的な治療好適性が野生型ヒトNGFとは明らかに異なることは留意すべきである。同様に、実施例2に記載したように、配列番号4のポリペプチドの精製は野生型NGFの公開された精製プロトコルとは異なり、本発明による前記ポリペプチドを調製するための特定のプロセスは、特にデスノナ変異体およびトリプシンが存在しない高純度を達成するために好適である。
関連するタンパク質のタンパク質パラメーターの理論値を、ExPASyのProPram-Toolによって計算した。これはhttp://web.expasy.org/protparamで入手可能である。これらを以下の表2に示す。
SDS-PAGEおよびウェスタンブロット
SDS-PAGEおよびウェスタンブロットは、標準的な手順を使用して実施した。SDS-PAGEについては、NuPAGE MES操作緩衝液(Thermo Fisher社、製品番号NP0002)中、一定電圧(175V)の還元条件下で、12%Bis-TRIS NuPAGEゲル(Thermo Fisher社、製品番号NP0342BOX)を操作した。ウェスタンブロット用の一次抗体は、Santa Cruz Biotechnology社から購入した(NGF(H-20)sc-548)。結果の例を、たとえば図9Aおよび図10に示す。
CEX-HPLCは、Dionex社のProPac SCX-10を使用して実施した。カラムは50mMのクエン酸緩衝液、pH5.5、1mL/分で操作した。溶出のため、1MのNaCl(B)を添加し、Bを0~100%として50分にわたって線形勾配を実施した。結果の例を図9Bに示す。
SE-HPLCは、GE Healthcare社のSuperdex 200 Increase 10/300GLを使用して実施した。カラムはPBS中で操作した。生成物は280nmで検出した。
エンドトキシン、DNA、および宿主細胞タンパク質(HCP)は、標準的なプロトコルに従って決定した。
[実施例1]
産生株
プロNGFをコードする遺伝子を、pET11a発現プラスミドにクローニングした。遺伝子はヒト(H.sapiens)から誘導し、2つの点変異(即ちP61SおよびR100E)をオープンリーディングフレーム中に導入した。続いて化学的に有力なロゼッタ(DE3)細胞を発現プラスミドによって形質転換し、単一のコロニーを選択した(得られた株をE5901-STRAIN(=E.coli Rosetta(DE3)/pET11a-プロNGF P61S R100E NGF RCB C-151101)と命名した)。1.0mLのアリコートを-60℃未満で保存した。
発酵のための複合培地
発酵のために使用した複合培地は、49.3g/Lの酵母抽出物、0.61g/LnのMgSO4・7H2O、0.5g/LのNH4Cl、14.2g/LのK2HPO4・3H2O、および10g/Lのグルコースからなっていた。この発酵のために使用した供給液は、263g/Lの酵母抽出物および133g/Lのグルコースからなっていた。
LB寒天培地を新たに注いだ。培地は10g/Lのペプトン、5g/Lの酵母抽出物、5g/LのNaCl、および15g/Lの寒天を含んでいた。オートクレーブの後に、培地に100μg/mlのアンピシリンおよび30μg/mlのクロラムフェニコールを添加した。
他に述べなければ、発酵、この実施例1における発酵は、Sartorius社製のBiostat Bユニットによって制御される1Lの撹拌ガラスバイオリアクター中で実施した。典型的には、pO2を30%に制御し、培養温度を37℃に設定して、2Mのリン酸および25%の水酸化アンモニウムを使用してpHを7に制御した。他に述べなければ、F0=6g/L/時間およびμ=0.25/時間の指数供給によってバッチ相を追跡した。実際的な理由により、全ての指数供給は2つの直線供給で近似した。典型的には、産生物発現の誘起は1mMのIPTGの添加によって実行し、誘起後は10g/L/時間の定常供給を適用した。Thermo Scientific社のSorvall Evolution RCを使用する遠心分離によって細胞のバイオマスを収穫した。遠心分離器にはSLC-6000ローターが取り付けられており、培養液を8500rpm、4℃で30分、遠心分離した。
所与の時点で、培養試料を10のOD600まで希釈し、この希釈液の100μLのアリコートからバイオマスをペレットとした。ペレットを150μlの(非還元性)Laemmli緩衝液中に再懸濁し、試料を95℃で5分、煮沸した。それぞれの試料10μlをNovex社の10% Bis-Trisゲルで分析した。電気泳動分離を125V、90分で実施し、ゲルをクーマシーで染色した。脱染したゲルをスキャンし、配列番号4のポリペプチドの前駆体に対応するバンドの存在量をデンシトメトリーによって定量した。使用したバイオマスの変動をさらに補正するため、配列番号4のポリペプチドの前駆体に対応するバンドの強度を、ハウスキーピングタンパク質の強度に正規化した。
配列番号4のポリペプチドの前駆体の標品を、European Brain Research Institute(EBRI,Rome,Italy)から入手した。標品をLaemmli緩衝液で65μg/mLの濃度に希釈した。記述したタンパク質濃度はEBRIによって定義されたものである。260、520、780、1040、および1300ngの配列番号4のポリペプチドの前駆体の標品を用いて標準曲線を作成した。標準曲線と同じゲル上で試料を分析し、試料の希釈係数を考慮して、所与の時間における産生物の絶対的産生収率を計算した。
上記に基づいて、産生株(E5901-STRAIN、上記参照)は、1Lスケールの発酵のために成功裡に使用されたと結論される。様々な培地組成を、細菌の増殖および産生物の発現を促進する能力について評価したが、5g/Lの酵母抽出物を添加した最小培地MMIが、発現収率および得られる細胞密度に関して満足できることが証明された。産生物の形成に関しては、主要な培養を抗生剤ありまたはなしで(アンピシリンおよびクロラムフェニコール、データは示していない)実施した場合に、有意な差は観察されなかった。
[実施例2]
本実施例で使用した配列番号4の前駆体は、実施例1に記載したように封入体で得られた。
当初は、本発明のポリペプチドの改善された精製のプロセスは、これに反する指示がない状態で、文献で以前に報告されたNGF精製の基礎的な拠り所に従うことが道理であった。しかし、大スケールでの効率的な生産のためには、後日のスケールアップに適した適応を考慮すべきであることも留意された。したがって、Rattenhollら(上記)のWO2013/092776A1およびその他の出版物に基づいて、配列番号4のポリペプチドが同様にトリプシンを使用する非特異的な消化およびそれに続く精製を採用してそのプロ形を介して少なくとも実験室スケールのプロセスで得られることは道理であった。それによって配列番号4の高純度の成熟ポリペプチドが得られることは道理であった。しかし、配列番号4の高純度の成熟ポリペプチドが得られたのは、本実施例で報告した特定の適合化および改変を通してのみである。したがって、配列番号4のポリペプチドのそれを必要とする対象への投与は、本明細書に記載したように、特に配列番号4のポリペプチドが高純度であることに鑑みて可能になる。
[実施例2A]
配列番号4のポリペプチドの前駆体を発現する大腸菌細胞(「バイオマス」)を、実施例1に記載したように産生し、リゾチームの添加および引き続く氷上での超音波処理によって細胞を溶解した。封入体(「IB」)を(1)宿主細胞から抽出して6%のTriton X100(1.5MのNaCl、60mMのEDTA中)によって洗浄し、(2)6MのグアニジンHCl(「gHCl」)、0.1MのTris-HCl(pH8.0)、1mMのEDTA、100mMの(新鮮な)DTTの中で可溶化した。IBは室温で2時間、可溶化した。その後、37%のHClの添加によってpHを3~4に低下させた。このようにして得られた配列番号4のポリペプチドの前駆体の可溶化された前駆体を含む溶液(「ソルビリゼート」)を、6MのgHCl(pH3~4)に対して透析した。
[実施例2B]
以下、本発明に到達する経過において実施例2Aと比較して試験し実施した、いくつかの改善について記載する。文脈によって他が指示されない限り、明示的に指示していない全ての詳細は実施例2Aについて上述した通りである。
少量のIB(振盪フラスコ中の培養から例示的に受容した)は可溶化緩衝液(6MのgHCl、0.1MのTris-HCL、pH8、0.1MのEDTA、100mMの(新鮮な)DTT)に容易に溶解することが以前に報告されているが、高い細胞密度の発酵から得られたIBは完全には溶解することができなかった。これは本明細書において前記可溶化緩衝液に2Mの尿素を添加することによって解決することができ、これは可溶化収率を顕著に改善することが分かった(データは示していない)。疑いを避けるため、2Mの尿素は6MのgHClおよびその他の成分に加えて存在していた。
当初はRattenhollら(2001, Eur. J. Biochem, vol. 268, p. 3296-3303; Rattenholl, 2001, Dissertation zur Erlangung des akademischen Grades doctor rerum naturalium (Dr. rer. nat.), Martin-Luther-Universitat Halle-Wittenberg (Germany))に基づいて、しかし重要なことに後日のスケール変更(スケールアップ)も考慮して、折り畳み反応液1リッターあたり200~500mgの配列番号4のポリペプチドの前駆体、好ましくは折り畳み反応液1リッターあたり200~300mgの配列番号4のポリペプチドの前駆体で折り畳みを実施することを決定した。これにより、配列番号4のポリペプチドの可溶化された前駆体の比較的良好な収率がもたらされる。特に、再折り畳み反応の体積と比較してそのような増大したNGFの量によって、また再折り畳み反応がグルタチオンおよびアルギニン等の比較的高価な成分を含むという考慮の下に、再折り畳み反応の体積あたり比較的より多くの配列番号4のポリペプチドの前駆体を折り畳むことができることを考慮することが重要であり、それにより再折り畳みが生産スケールでも経済的に実行可能になる。
再折り畳みの後、配列番号4のポリペプチドの前駆体の精製を、かなり高いプロNGFの等電点を利用し、精製のためにカチオン交換固定相(即ちSPセファロース)を採用するアプローチによって行なった。この種類のクロマトグラフィーを運転するため、技術的な理由により再折り畳み緩衝液を低電導度の緩衝液に交換しなければならない。これを行なっている間に、顕著な量の配列番号4のポリペプチドの前駆体が沈殿した(データは示していない)。この観察は、緩衝液中におけるアルギニン濃度の低下に帰せられる。
配列番号4のポリペプチドの製造のためにはプロテアーゼ(トリプシン)が必須であり、したがって理想的には選択された特定のトリプシンは以下の基準に合致すべきことは道理である。
1.組み換え源から誘導されること。動物を用いない原材料の証明が、後に要求されるプロセスのGMP適合性のために重要である。
2.トリプシンの副活性が低いこと。注目すべきことに、トリプシンは自己分解を受けることがある。このプロセスはいわゆるプソイドトリプシンをもたらすことがあり、これは幅広い基質スペクトルを有し、キモトリプシン様活性を保有している。自己分解を低減するためにCa2+(たとえば1mMのCaCl2)を添加してもよい。しかし今日では典型的には「改変されたトリプシン」があらゆるプロトコルに適用され、これは厳しい配列特異性を必要とする(たとえばペプチドフィンガープリンティングのため)。この改変されたトリプシンは、典型的にはトリプシンのリジン残基の露出したεアミノ基のアシル化によって得られる。
3.再現性のある産生プロセスを可能にするため、バッチ間の変動が少ないこと。あるいは、選択された酵素は、それぞれのバッチの比活性を記述する証明とともに送達される必要がある。したがって、酵素の必要量は質量よりもむしろ活性に基づいてよい。
意図したトリプシン処理のための最適な酵素/基質比の当初のスクリーニング探索の中で、捕捉カラムから得られた配列番号4のポリペプチドの前駆体(上記参照)を使用した。以前に確立されたプロセス(European Brain Research Institute(EBRI)、詳細は公開されていない。Rottenhollら(上記)に基づく)とは対照的に、トリプシン処理の前にさらなる疎水性相互作用クロマトグラフィーを使用しないことを決定した。トリプシン処理の前にそのような第2のカラム精製ステップを省略するという決定は、主として2つの思考の流れに基づいていた。一方では、捕捉カラムの後で得られる産生物はSDS-PAGEによれば既に実質的に純粋であった。他方では、トリプシン処理それ自体が、残存する宿主細胞タンパク質(HCP)の消化によって不純物プロファイルの改善を助ける可能性がある。
成熟ポリペプチドの仕上げのためにSPセファロース固定相(注:SPセファロースはカチオン交換固定相である)を採用した、以前に確立されたプロセス(European Brain Research Institute(EBRI)、詳細は公開されていない。Rottenhollら(上記)に基づく)とは対照的に、本明細書では以下の考慮に基づいてより好適な固定相を探索した。SPセファロースカラムに効率的に負荷するために、たとえば緩衝液の交換によって、配列番号4のポリペプチドの前駆体を含む溶液の電導度を低下させる必要がある。しかし、溶液のイオン強度の低下は標的分子の沈殿をもたらすことが知られており(たとえば実施例2A)、したがって低電導度の緩衝液への緩衝液変更は避けるべきである。さらに、カチオン交換固定相は既に配列番号4のポリペプチドの前駆体の捕捉に使用されており、残存する夾雑物のより良い分離を達成するためには直交選択性が好ましい。トリプシン処理反応の精製にSP固定相を使用することに反対する第3かつ最後の論拠は、配列番号4のポリペプチドのおそらく残存している前駆体はこのカラムに結合しており、結合選択性によってではなく、単に溶出の選択性によって、配列番号4の成熟ポリペプチドから分離できることである。
エンドトキシンおよびDNAをさらに枯渇させるため、追加のアニオン交換膜をプロセスに含めた。一般にかつ共通して知られているように、膜クロマトグラフィーは、分析または精製される成分(本例では配列番号4のポリペプチド)を含む溶液が、通常は荷電している膜を通過するまたは通り抜けることで特徴付けられる。その目的のため、本例ではSTIC膜(Sartorius,Goettingen,Germany)を、図1の指示した場所に組み込んだ。配列番号4のポリペプチドは膜に結合しないことが示され、したがって膜クロマトグラフィーが配列番号4のポリペプチドの精製のために好適であるという概念の証明が提供された。膜クロマトグラフィーを含む全体のプロセスにおける組み込みの説明のため、図1を参照されたい。
プロセスの堅牢性を精査するため、プロセスを5回実施し、得られた分画をその収率および純度に関して解析した。これらの操作を通して、プロセスの詳細の着実な最適化を追求し、最終の最適化されたプロセスの詳細(図1を参照)が確立されるまで、緩衝液の組成、勾配、その他を選定した。結果は、実験室スケールでほぼ50~100mgの配列番号4のポリペプチドが1つの一貫した生産運転から得られることを示している。注目すべきことに、得られた産生物は、SDSアクリルアミドゲル電気泳動およびそれに続くクーマシー染色または銀染色によって、比較的純粋であることが一貫して見出された(夾雑する宿主細胞タンパク質は5%未満、切断されたNGFは微量のみ。データは示していない)。
このプロセスのため、配列番号4のポリペプチドの再折り畳みされた前駆体を、SPセファロースFF(「FF」は速い流量、即ち比較的大きな粒子の固定相を意味する)を使用して捕捉し、続いてトリプシンで処理して成熟NGFを得た。この目的のため、再折り畳み反応のためのアルギニンの濃度を1M(先行技術で推奨された)から350mMに低下させた。
[実施例3]
実施例2に従って以前調製した配列番号4のポリペプチドを含む製剤を、Eng et al., 1997, Anal. Chem., vol. 69, p. 4184-4190による示唆を考慮して調製した。

- 第1の実施形態:上記の製剤を-70℃で凍結保存し(データは示していない)、投与前に解凍することができる。
- 第2の実施形態:上記の製剤を冷蔵庫中、好ましくは+2~+8℃の温度範囲で保存することができる(データは示していない)。
- 第3の実施形態:上記の製剤を乾燥または凍結乾燥に供し、次いでたとえば室温で保存することができる(データは示していない)。これは投与前に再構成することができる。
[実施例4]
本実施例の目的は、非ヒト動物における眼科障害の処置および/または防止のための配列番号4のポリペプチドの有効性を検討することである。
視神経挫滅(PNC)モデル
ラット(Sprague Dawley、雄、180~200g、Charles River社、Italy)を、温度および湿度を制御した動物設備中で飼育した(12時間の暗/明サイクル、飼料および水への自由なアクセス)。行動実験は、静かな温度制御(20~22℃)された部屋で午前9時から午後5時まで、薬物処理の状態について知らされていないオペレーターが実施した。
・媒体または配列番号4のポリペプチドによる処置:20、2、0.2μg/mlの溶液、体積2μlの硝子体内注射。傷害の後4日目に開始し、7日目まで続ける。
・媒体または配列番号4のポリペプチドによる処置:200μg/mlの溶液、体積25μlの点眼。傷害の後4日目に開始し、7日目まで続ける。1日1回、2回、または3回。
切除した網膜をホルマリン中で24時間、後固定し、パラフィン包埋した。組織学的検査のため、いくつかのホルマリン固定パラフィン包埋切片(5μm)をヘマトキシリンおよびエオシン(H&E)で染色した。他の切片は、キシレンと低下する量のエタノールとのシリーズ(100%、90%、および70%)中の逐次インキュベーションによって脱パラフィンおよび再水和した。切片を10mMのクエン酸緩衝液(pH6.0)中で5分間マイクロウェーブ処理することによって、抗原を回収した。切片を一次抗体(Brn3a、1:500およびRBPMS 1:500)と湿潤チャンバー中で1時間、インキュベートした。次いで切片を、前希釈したビオチン化抗マウス/ウサギIgG二次抗体またはビオチン化した抗モルモット二次抗体(1:200、#BA-7000、Vector Laboratories社、Burlingame,US)と室温で30分、およびアビジン-ビオチン複合体溶液(#PK-7200,Vectastain Elite ABC-HRP Kit,Vector Laboratories社、Burlingame,US)中、室温で30分、インキュベートした。次いで色素の発色反応のために切片をペルオキシダーゼ基質(#K3468,ImmPACT DAB,Vector Laboratories社、Burlingame,US)に4~6分間移し、蒸留水で濯いでマウンティングした。核はMayerのヘマトキシリンで対比染色した。組織を可視化し、光学顕微鏡Leica DM2500(Leica Microsystems社、Milan,Italy)を使用してデジタルイメージを捕捉した。
ONC後4日目に媒体で開始した硝子体内処置に供したラットでは、モデルの設定の間に生成したデータと一致して、網膜神経節細胞の数の約60%の減少が観察された。3つの異なる用量レベル(20、2、0.2μg/mlの溶液、注射体積2μl)で硝子体内投与した配列番号4のポリペプチドは、網膜神経節細胞の損失に対して用量依存性の保護効果を発揮し、これは20μg/mlの溶液の硝子体内注射を受けたラットで統計的有意に達した。この群では、ONCに供した眼における網膜神経節細胞の数は、健康な反対側の眼と同様であった(図3)。
[実施例5]
本実施例の目的は、非ヒト動物における眼科障害の処置および/または防止のための配列番号4のポリペプチドの有効性を検討することである。
1.本実施例は、眼窩下神経の圧縮(CION)のマウスモデルにおける点眼の後の、配列番号4のポリペプチドと、野生型ヒトNGF(配列番号2のポリペプチド)および野生型マウスNGF(mNGF)との発痛活性を比較することを目的としている。
2.NGFアナログ(配列番号4のポリペプチド)の効果を、配列番号2のポリペプチド、mNGF、および痛覚応答を惹起することが知られている刺激性参照化合物であるカプサイシンと比較して、点眼の後の急性痛覚応答を誘起する能力を測定することによって評価する。以下の用量を試験する。等張の食塩水(NaCl 0.9%)で希釈して0.5、1、5、および10μg/5μl/眼。カプサイシンは0.001~0.5nmol/5μl/眼で試験する。
3.次いで閾値以下の用量の様々な化合物を、眼窩下神経の圧縮(CION)モデルにおける点眼の後の痛覚応答を惹起する能力について試験する。
マウスNGF(マウスNGF、mNGF)
マウスNGFは高純度の天然マウスNGF2.5S(>95%)であり、Bocchini et al., 1969, Pro.c Natl. Acad. Sci. U S A, vol. 64, p. 787-794によって記載された方法に従ってマウスの顎下腺から抽出および精製によって得た。mNGFは、UniProt P01139のポリペプチド配列の残基129~241からなっている。
動物実験は、動物介護手順に関する欧州連合(EU)のガイドラインおよびEU指令2010/63/EUのイタリアの法律(DLgs26/2014)の適用に従って行なう。研究はUniversity of Florenceの研究許可#194/2015-PRの下で実施する。C57BL/6マウス(雄、25~30g、Envigo社、Milan,Italy)を痛覚試験に使用する。動物は、温度および湿度を制御した動物設備中で飼育する(12時間の暗/明サイクル、飼料および水への自由なアクセス)。行動実験は、静かな温度制御(20~22℃)された部屋で午前9時から午後5時まで、薬物処理の状態について知らされていないオペレーターが実施する。
CIONは報告されたように(Vos et al., 1994, J. Neurosci., vol. 14, p. 2708-2723; Luiz et al., 2010, Neuropeptides, vol. 44, p. 87-92)、C57BL/6マウスで実施する。簡単に述べると、ケタミン(90mg/kg)とキシラジン(3mg/kg)の混合物の腹腔内(i.p.)注射によってマウスを麻酔し、鼻の側面の左上唇の皮膚に切開を作成し、次いで眼窩下神経の吻端を露出した。次いで、緩く締める2本の結紮糸(#6/0絹縫合糸)を2mmの距離で眼窩下神経の周囲に設ける。シャム手順では、左の眼窩下神経を露出するが結紮しない。硫酸ネオマイシンおよびスルファチアゾール(粉末、それぞれ0.05gおよび9.95g、Boehringer Ingelheim Italia S.p.A,Italy)を創傷に適用し、切開創を縫合する。マウスを追跡し、適切に水分補給し、麻酔から完全に回復するまで制御された温度(37℃)で維持する。全ての実験は手術後10日目に実施する。実験の終了時に、CO2および10~50%のO2の吸入によってマウスを安楽死させる。
以前記載されたように(De Petrocellis et al., 2010, Pharmacol. Res., vol. 63, p. 294-299)、急性痛覚応答を誘起するために、試験化合物、即ち配列番号4のポリペプチド、配列番号2のポリペプチド、mNGF(全て0.5、1、5、および10μg/5μl/眼)およびカプサイシン(0.5nmol/5μl/眼)またはそれぞれの媒体(等張食塩水、0.9%のNaClおよび1%のジメチルスルホキシド、DMSO)の点眼(5μl)を使用する。マウスは個別にプレキシグラスチャンバーの中に収容し、刺激前20分、馴化する。薬物を眼に点滴した後の眼を拭う動作の回数を5分の期間について記録し、刺激の指標とみなす。
実施例の最初の部分では、点眼(5μl/滴/眼)によって投与した種々の用量(0.001~0.5nmol)のカプサイシンの、5分の期間における眼拭いの回数として測定した急性痛覚応答を誘起する能力について試験した。カプサイシンは、カプサイシンの点眼(0.001~0.5nmol/5μl/眼)の後で測定した眼拭い応答の増加によって実証されるように、用量依存性の痛覚応答を誘起した(図5)。
[実施例6]
本実施例の目的は、ヒトにおける眼科障害の処置および/または防止のための配列番号4のポリペプチドの有効性をさらに支持することである。
Claims (20)
- 哺乳動物対象における眼科障害の処置および/または防止における使用のための、配列番号3のポリペプチドおよび配列番号4のポリペプチドの中から選択されるポリペプチド。
- 前記哺乳動物対象がヒトである、請求項1に記載の使用のためのポリペプチド。
- 配列番号4のポリペプチドである、請求項1または2に記載の使用のためのポリペプチド。
- 眼に投与するための、請求項1から3のいずれか一項に記載の使用のためのポリペプチド。
- 前記投与が眼への局所投与および硝子体内投与からなる群から選択され、局所投与が好ましい、請求項4に記載のポリペプチド。
- 繰り返して投与される、請求項1から5のいずれか一項に記載の使用のためのポリペプチド。
- 少なくとも1日3回、繰り返して投与される、請求項6に記載の使用のためのポリペプチド。
- 3~30日、好ましくは7~14日の期間、繰り返して投与される、請求項6または7に記載の使用のためのポリペプチド。
- 前記眼科障害が視神経の損傷および/または障害を含む、請求項1から8のいずれか一項に記載の使用のためのポリペプチド。
- 前記眼科障害が網膜神経節細胞の障害によって特徴付けられる、請求項9に記載の使用のためのポリペプチド。
- 視神経の損傷に続いて投与される、請求項1から10のいずれか一項に記載の使用のためのポリペプチド。
- 視神経の損傷の誘起の少なくとも4日後に投与される、請求項11に記載の使用のためのポリペプチド。
- 前記眼科障害が、緑内障、神経栄養性角膜炎、視神経炎、視神経委縮、視神経頭ドルーゼン、および視覚経路神経膠腫からなる群から選択される少なくとも1つを含む、請求項1から12のいずれか一項に記載の使用のためのポリペプチド。
- 用量/各用量が1眼あたり0.3~30μgのポリペプチド、より好ましくは1眼あたり1~10μgのポリペプチド、最も好ましくは1眼あたり5μgのポリペプチドの量を有する、請求項1から13のいずれか一項に記載の使用のためのポリペプチド。
- 前記処置および/または防止が、前記哺乳動物対象に痛覚過敏症を惹起しない、請求項1から14のいずれか一項に記載の使用のためのポリペプチド。
- 前記ポリペプチドが水性媒体中に含まれ、前記水性媒体が前記哺乳動物対象に投与される、請求項1から15のいずれか一項に記載の使用のためのポリペプチド。
- 以下の
a)0.2~20mg/mlの前記ポリペプチド(好ましくは2mg/ml)、
b)5~100mMの酢酸ナトリウム緩衝剤(好ましくは20mM)、
c)5~100mMのメチオニン(好ましくは20mM)、
d)5.0~6.0のpH(好ましくはpH5.5)
を含む組成物中に含まれる、請求項16に記載の使用のためのポリペプチド。 - 前記ポリペプチドの分解物を実質的に含まず、特に前記ポリペプチドのデスノナ変異体を実質的に含まない、請求項1から17のいずれか一項に記載の使用のためのポリペプチド。
- 前記ポリペプチドが組み換え発現および精製によって得られ、前記精製が混合モード固定相上での精製を含む、請求項1から18のいずれか一項に記載の使用のためのポリペプチ
- 配列番号3のポリペプチドおよび配列番号4のポリペプチドの中から選択されるポリペプチドを含む組成物であって、pH5.0~6.0のpH(好ましくはpH5.5)によって特徴付けられ、以下の
a)0.2~20mg/mlの前記ポリペプチド(好ましくは2mg/ml)、
b)5~100mMの酢酸ナトリウム緩衝剤(好ましくは20mM)、
c)5~100mMのメチオニン(好ましくは20mM)
を含む、組成物。
