(1)第1の態様に記載のβ-グルカンまたは第1の態様に記載のβ-1,3/1,6-グルカンの組成物、および
(2)薬学的に許容されるベクターを含む。
(1)脱脂段階:南極の褐藻を乾燥および粉砕し、有機溶媒に浸し、攪拌して、脱脂藻類粉末を得る。
(2)水抽出:前記脱脂藻類粉末を室温下で水で攪拌および抽出して水抽出液を得る。
(3)等級付け段階:段階(2)で得られた水抽出液を遠心分離し、遠心分離によって得られた上澄みに1~3mol/Lの塩化カルシウム水溶液を加え、攪拌した後遠心分離し、上澄みを取って、透析または限外ろ過脱塩を実行し、減圧濃縮および乾燥して、粗多糖類を得る。
(4)精製段階:段階(3)に記載の粗多糖類を蒸留水で溶解し、蒸留水および塩化ナトリウム水溶液を移動相として使用し、陰イオン交換樹脂で分離および精製し、水溶出成分を収集し、減圧濃縮し、凍結乾燥して、前記β-1,3/1,6-グルカンを得る。
特に定義しない限り、本明細書および特許請求の範囲で使用される以下の用語は、当業者によって一般的に理解される意味を有する。特に明記しない限り、本明細書の全体で引用されるすべての特許、特許出願、出版物は、参照により本明細書に組み込まれる。
調製例1
(1)脱脂段階:南極の褐藻を乾燥および粉砕し、有機溶媒に浸し、攪拌して、脱脂藻類粉末を得る。
(2)水抽出段階:前記脱脂藻類粉末を室温下で蒸留水で攪拌および抽出して水抽出液を得る。
(3)分級段階:段階(2)で得られた水抽出液を遠心分離し、遠心分離によって得られた上澄みに1~3mol/Lの塩化カルシウム水溶液を加え、攪拌した後遠心分離し、上澄みを取って、蒸留水で透析または限外ろ過して脱塩し、減圧濃縮および乾燥して粗多糖類を得る。
(4)精製:段階(3)に記載の粗多糖類を蒸留水で溶解し、蒸留水および塩化ナトリウム水溶液を移動相として使用し、強陰イオン交換樹脂で分離および精製し、水溶出成分を収集し、減圧濃縮し、凍結乾燥して、前記β-1,3/1,6-グルカンを得る。

(4)精製段階:段階(3)に記載の粗多糖類を蒸留水で溶解し、蒸留水および塩化ナトリウム水溶液を移動相として使用し、強陰イオン交換樹脂で分離および精製し、水溶出成分を収集し、減圧濃縮する。
(5)第2回目の精製:段階(4)に記載の水溶出成分を、蒸留水を移動相として使用し、弱陰イオン交換樹脂で分離および精製し、水溶出成分を収集する。
(6)脱色段階:段階(5)のに記載の水溶出成分を活性炭カラムで分離および精製し、蒸留水を移動相として使用し、水溶出成分を収集する。減圧濃縮し、凍結乾燥して、前記β-1,3/1,6-グルカンを得る。
本実施例におけるβ-グルカン組成物は、十八角形レーザー光散乱装置(MALLS)および示差検出器(RI)と併用して、高性能ゲル浸透クロマトグラフィー(HPGPC)を使用して分析する。
1)β-グルカン組成物中の側鎖オリゴ糖の調製:β-グルカン組成物を溶解し、約5%の濃度に調製し、10μlのエンド-1,3-β-グルカナーゼを加え、40℃下で8時間酵素加水分解する。10分間煮沸し、遠心分離機で10000rpmで10分間遠心分離し、上澄みを収集し、凍結乾燥して、β-グルカン組成物中の側鎖オリゴ糖を得る。
本発明のβ-グルカン組成物は、三重らせん構造を有する。コンゴーレッドは、三重らせん鎖コンフォメーションを有する多糖類と複合体を形成することができ、複合体の最大吸収波長は、コンゴーレッドと比較して赤方偏移が発生する。本発明のβ-グルカン組成物は、塩基性条件下でコンゴーレッドと複合体を形成して、その最大吸収波長の赤方偏移が15nmを超えるようにすることができる(図8)。
本実施例において、本発明のβ-1,3/1,6-グルカン糖の、末梢血自然免疫細胞の二つの主要な集団、即ち、単球(CD11b+Ly6Chi、図10Aおよび10B)および顆粒球(CD11b+Ly6G+、図10Aおよび10C)に対する結合親和性および選択性を検出する。図10Dに示されるように、β-1,3/1,6-グルカン-FITCは、単球(12.8%)によく結合し、ごく少数の顆粒球(1.5%)に結合する。
血液単球は、腫瘍微小環境に動員されてマクロファージに分化し、免疫阻害および腫瘍促進微小環境を生成する。免疫阻害マクロファージを前炎症性状態にリモデリングし、腫瘍微小環境を操作し、インビボでの腫瘍の増殖を阻碍する。ヒト結腸直腸がん細胞DLD1異種移植マウスモデルにおける本発明のβ-1,3/1,6-グルカンの抗腫瘍効果を検出する。
本実施例において、対照グループおよび本発明のβ-1,3/1,6-グルカン処理グループのマウスの腹腔マクロファージを誘発し、9つの異なる結腸直腸がん細胞株と共培養し、マクロファージの食作用を検出する。
循環血液系における免疫細胞組成物を検出する。B細胞(CD19+)および樹状細胞(CD11c+)の割合が増加する一方で、骨髄細胞(CD11b+)の割合は、わずかに減少することが分かる(図14A)。β-1,3/1,6-グルカン治療は、骨髄性亜集団、前炎症性単球由来マクロファージ(CD11b+Ly6Chi)の形成を誘発する(図14B)。
免疫担当マウスの体内における本発明のβ-1,3/1,6-グルカンの抗腫瘍効果を検証するために、AOM/DSS誘発性C57BL/6Jマウス結腸直腸がんモデルにおいて、β-1,3/1,6-グルカンの抗腫瘍効果を検出する。腫瘍進行の後期において、β-1,3/1,6-グルカングループは、1および3mg/kgの用量下でvehicleグループよりも体重が高く、これらの治療されたマウスがより良い体調にあったことを示す(図15A)。3mg/kgのβ-1,3/1,6-グルカン治療によって結腸の長さがわずかに増加されたことを除いて(図15B)、結腸の長さに有意差は見られない。1mg/kgの低用量のβ-1,3/1,6-グルカンによって治療された後でも、マウスあたりに誘発された結腸直腸腫瘍の数は減少する(図15C)。図15Dに示されるように、β-1,3/1,6-グルカンによって治療された後、3mg/kgの用量下で、腫瘍直径が2mm~4mmおよび4mm以上である腫瘍の数が減少する。AOM/DSSを使用した後、本発明の1mg/kgのβ-1,3/1,6-グルカンは、vehicleよりも直径が4mmより大きい腫瘍の数をより少なく誘発する(図15E)。
AOM/DSS誘発および本発明のβ-1,3/1,6-グルカン治療後に、骨髄細胞(CD11b+)を含む様々な免疫細胞の変化を観察する。AOM/DSSは、循環血液系における骨髄細胞(CD11b+)の割合を増加させ、本発明のβ-1,3/1,6-グルカン治療によってこの傾向を逆転する(図16A)。同時に、β-1,3/1,6-グルカンは、インビボでAOM/DSS投与によって引き起こされた血液(図16A)および脾臓(図16B)でダウンレギュレートされたB細胞を増加させ、血液(図16A)およびリンパ節(図16C)におけるCD4T細胞の減少を逆転させる。従って、β-1,3/1,6-グルカン治療は、AOM/DSS投与によって引き起こされた免疫細胞組成の変化を逆転させる。さらに、本発明のΒ-1,3/1,6-グルカンは、炎症性因子IFNγ(図16D)、TNF-α等、およびケモカインCXCL1の分泌のアップレギュレーションを誘発する(図16E、図16F、図16G、図16H、および図16I)。
液体窒素で冷凍保存されたマウス結腸がん細胞株HCT-116を蘇生させ、10%のウシ胎児血清を含む5A培地で培養し、培養フラスコ内で培養細胞を増殖させ、必要な量の細胞に達した後、細胞を消化して収集し、10%ウシ胎児血清を含む5A培地で25000万/mlの細胞懸濁液に希釈し、従来の消毒後、0.2mlの各マウスをマウスの右前肢腋窩に皮下接種し、腫瘍が1gの組織塊に増殖した場合、動物を犠牲にして腫瘍組織を剥がし、約2~3立方ミリメートルの腫瘍塊に切断し、カテーテル法によって継代ヌードマウスの皮下に移植し、2回連続継代する。表に示されるHT-29、SW-480、DLD-1およびRKO細胞株を同じ方法でモデリングおよび実験する。担癌ヌードマウスを約1gの腫瘍増殖で継代させた後、上記の方法を用いて腫瘍塊移植モデリングを実行し、移植後2日目にマウスをランダムにグループに分けて投与を開始する。

S180マウス線維肉腫細胞を使用し、10%FBSを含むDMEM高グルコース培地を使用して培養増殖する。細胞が対数増殖期に入ると、次の試験を実行することができる。対数増殖期の細胞を収集して遠心分離し、カウントする。細胞濃度を調整し、第1世代の繁殖マウスとして昆明マウスの腹腔に1匹あたり0.2mlを接種する。1週間飼育した後、第1世代の繁殖マウスの腹腔内の腹水を遠心分離してカウントし、細胞濃度を調整して、第2世代の繁殖マウスとして昆明マウスの腹腔に1匹あたり0.2mlを接種し、さらに1週間飼育した後、第2世代の繁殖マウスの腹腔内の腹水を遠心分離してカウントし、細胞濃度を調整して、昆明マウスの背中に1匹あたり0.2mlを皮下接種する。すべての接種プロセスは、超クリーンなワークベンチで無菌的に実行される。接種後、マウスの増殖状況を観察し、マウスの増殖状態に応じてスクリーニングし、村主―ニング後、ランダムにクループに分ける。実験結果は、次の表に示され、3mg/kgのβ-1,3/1,6-グルカンは、手術後の腫瘍の再増殖を有意に阻害する可能性がある。
マウス肺がんルイス細胞を使用し、10%FBSを含むDMEM高グルコース培地を使用して培養増殖する。細胞が対数増殖期に入ると、次の段階の試験を実行することができる。対数増殖期の細胞を収集して遠心分離し、カウントする。細胞濃度を調整し、繁殖マウスとしてマウス1匹あたり0.2mlをC57BL/6マウスの右側の腋窩に皮下接種する。腫瘍が1000mm3より大きく増殖すると、無菌的に取り出し、秤量し,質量対体積比1:4の塩化ナトリウム注射液を加えて希釈して粉砕し、従来の消毒後、0.2mlの各マウスをマウスの右前肢腋窩に皮下接種して継代する。腫瘍が1000mm3より大きく増殖すると、第2世代の腫瘍を取り出し、無菌的に取り出し、秤量し,質量対体積比1:4の塩化ナトリウム注射液を加えて希釈して粉砕し、従来の消毒後、0.2mlの各マウスをマウスの背中に皮下接種し、接種後、マウスの増殖状況を観察し、マウスの増殖状態に応じてスクリーニングし、スクリーニングした後ランダムにグループに分けて投与する。実験結果は、次に表に示されるようであり、本発明のβ-1,3/1,6-グルカンは、1mg/kg、3mg/kg、9mg/kgでの手術後の腫瘍の再増殖を有意に阻害することができる。
癌は、人間の死の主要な原因の一つであり、癌症例の約1/4の病因および発病のメカニズムは、慢性炎症に関連する。炎症性腸疾患(inflammatory bowel disease、IBD)は、病院や発病メカニズムが不明な腸の炎症性疾患の一種であり、その病理学的特徴から、潰瘍性大腸炎(ulcerative colitis、UC)およびクローン病(Crohn’s disease、CD)と分けられ、年々増加し、かつ若くなりつつある。IBD患者の結腸直腸がん(colorectal cancer、CRC)の発生リスク率は増加し、8~10年後に毎年年0.5%~1%増加し、30年後にIBD患者の最大18%がCRCに発展する可能性がある。IBD関連CRCは、すべての結腸直腸がんの1%~2%しか占めていないが、それは、IBD患者が死亡する従来の原因である。アゾキシメタン(azoxymethane、AOM)/デキストラン硫酸ナトリウム(dextran sodium sulfate、DSS)によって誘発された大腸炎関連の癌(colitis associated cancer、CAC)動物モデルは、IBD誘発性CRCのプロセス全体をうまくシミュレートするため、新薬の有効性およびメカニズムを研究するために広く使用され、その病理学的変化および発癌を解明することは、結腸直腸がん治療の新しい候補標的を発見するのに役立つ。
試験方法は、次のとおりである。
免疫機能が正常なマウスでの動物実験(尾椎の静脈内投与)
1.実験方法:
腫瘍細胞:S-180、接種部位:肩甲骨、マウス種:KM(雄のマウス)、マウス数:各グループ13匹、腫瘍細胞接種の翌日から投与開始、投与頻度:毎日投与、投与サイクル:20日、投与方法:尾椎の静脈内注射。
空白対照グループ:モデルグループ(CNグループ)
陽性対照グループ:シスプラチン(2mg/kg)(PCまたはCPグループ)、椎茸多糖類LNT(1.5mg/kg)
実験グループ:BL:β-1,3/1,6-グルカン低用量(0.3mg/kg)、BM:β-1,3/1,6-グルカン中度用量(1.5mg/kg)、BH:β-1,3/1,6-グルカン高用量(7.5mg/kg)。
結果は、図18A~Bに示されるようである。陽性対照グループおよび実験グループのマウスは、接種後20日でもまだ良好な生理学的状態にあり、動物は、犠牲になる前に非常に活発で、行動が機敏である。
モデルグループと比較して、β-1,3/1,6-グルカンは、低用量、中度用量、高用量でS-180腫瘍の増殖を有意に阻害することができ、腫瘍阻害率は、60%~70%に達する。シスプラチングループのマウスの胸腺重量は、モデルグループよりも有意に低く、β-1,3/1,6-グルカン投与グループ(0.3、1.5、7.5mg/kg)の胸腺重量は、シスプラチングループよりも有意に高く、高用量BG136投与グループのマウスの胸腺重量は、モデルグループと同等である。これは、β-1,3/1,6-グルカンがマウスの免疫系を活性化することにより、抗腫瘍効果を発揮する可能性があることを示す。
1.実験方法:
腫瘍細胞:S-180、接種部位:腋窩、マウス種:昆明マウス(Kunming)、マウス数:各グループ13匹(雌マウス7匹および雄マウス6匹)、投与開始時間:接種翌日から投与開始、投与頻度:毎日投与、投与サイクル:10日、投与方法:経口投与、投与量(mg/kg):1 モデル(CN)、2 シスプラチン(CPまたはPC):1.5mg、3 β-1,3/1,6-グルカン:1mg(B1)、4 β-1,3/1,6-グルカン:5mg(B5)、5 β-1,3/1,6-グルカン:25mg(B25)、6 シスプラチン1.5mg+BG1mg(CPB1)、7 シスプラチン1.5mg+BG5mg(CPB5)、8 シスプラチン1.5mg+BG25mg(CPB25)。
実験結果は、図19AおよびBに示されるようである。当該実験結果から、β-1,3/1,6-グルカンの経口投与は、S-180腫瘍に対して有意な阻害効果があり、β-1,3/1,6-グルカンの経口投与量が1mg/kgである場合、全体の阻害率は、58%以上であり、雄マウスの腫瘍阻害率は、70.4%に達することが分かる。BG136を25mg/kgのシスプラチン(1.5mg/kg)薬物と併用すると、阻害率は、さらに改善され、腫瘍阻害率は、77.3%に達する。
白血球和血小板に対する化学療法と併用したβ-1,3/1,6-グルカンの効果
3x105個のマウス黒色腫細胞株B16(PerkinElmerからの贈り物)の細胞懸濁液をC57BL/6Jマウス(雌、6~8週齢、済南朋悦実験動物会社)に皮下注射する(Overwijk & Restifo、2001)。腫瘍移植から約2日後、カルボプラチン(30mg/kg、週2回)の腹腔内注射およびBG136(4mg/kg)または椎茸多糖類LNT(2mg/kg)の尾静脈内注射を実行し、投与期間中に腫瘍体積を検出し、15日目に、動物を犠牲にした後、腫瘍質量を検出する。動物を犠牲にした後、心臓から採血し、EDTA-抗凝固チューブに入れ、混合した後、50μlの全血を採集し、血液分析器で免疫細胞の濃度を検出する。
免疫細胞は様々な種類の腫瘍細胞で重要な役割を果たすため、β-1,3/1,6-グルカンの抗腫瘍、白血球および血小板増加効果は、様々な腫瘍細胞(例えば、肺がん、腎臓がん、肝臓がん、乳がん等)で役割をはたし、かつ免疫細胞は、腫瘍の転移および発生においても調節的な役割を果たすことにより、β-1,3/1,6-グルカンは、腫瘍細胞の転移および発生を阻害することができる。
使用されたPD-1抗体:名称:インビボMAb抗マウスPD-1(CD279)、Bioxcellから購入
(i)結腸がんMC38マウス皮下移植腫瘍に対するPD-1抗体と併用したβ-1,3/1,6-グルカンの静脈内注射の効果
本実施例でマウス結腸がん細胞株MC38を選択し、PD-1抗体と併用したβ-1,3/1,6-グルカンがマウス移植腫瘍増殖に及ぼす影響を検出し、試験結果は、図20A~Cに示されるようである。
本実施例において、RT-PCR方法によって、マウス前立腺がん細胞MC38移植腫瘍における様々なサイトカインの発現を定量化する。結果は、図21A~Fに示されるようである。
マウス結腸がん細胞株MC38を選択し、マウス移植腫瘍の増殖に対するPD-1抗体と併用したβ-1,3/1,6-グルカンの影響を検出する。結果は、図22A~Cに示されるようである。

Glcβ1-6Glcβ1-6Glcβ1-6Glcβ-であることを特徴とする、請求項1に記載のβ-グルカン組成物。 - 前記β-グルカン組成物において、重合度5~10の重量含有量は、0~50.0%であり、重合度10~20の重量含有量は、0~80.0%であり、重合度20~25の重量含有量は、0~25.0%であり、重合度25~30の重量含有量は、0~45.0%であり、重合度30~40の重量含有量は、0~30.0%であり、重合度40~50の重量含有量は、0~15.0%であり、重合度50~60の重量含有量は、0.1%~55.0%であり、重合度60~70の重量含有量は、0.1%~20.0%であり、重合度70~80の重量含有量は、0.1%~15.0%であり、重合度>80の重量含有量は、0~40.0%であることを特徴とする、請求項1に記載のβ-グルカン組成物。
- 前記β-グルカン組成物において、重合度5~10の重量含有量は、0~45.0%であり、重合度10~20の重量含有量は、0~75.0%であり、重合度20~25の重量含有量は、0~20.0%であり、重合度25~30の重量含有量は、0~40.0%であり、重合度が30~40であるβ-グルカンの重量含有量は、0~25.0%であり、重合度40~50の重量含有量は、0~10.0%であり、重合度50~60の重量含有量は、0.1%~50.0%であり、重合度60~70の重量含有量は、0.1%~15.0%であり、重合度70~80の重量含有量は、0.1%~10.0%であり、重合度>80の重量含有量は、0~35.0%であることを特徴とする、請求項3に記載のβ-グルカン組成物。
- 前記β-グルカン組成物は、重合度が20~80であるβ-グルカンで構成され、ここで、重合度20~25の重量含有量は、13.2%~19.8%であり、重合度25~30の重量含有量は、29.1~43.7%であり、重合度30~40の重量含有量は、18.2~27.4%であり、重合度40~50の重量含有量は、7.3~10.9%であり、重合度50~60の重量含有量は、4.5%~6.7%であり、重合度60~70の重量含有量は、3.5%~5.3%であり、重合度70~80の重量含有量は、4.2%~6.2%であることを特徴とする、請求項4に記載のβ-グルカン組成物。
- 前記β-グルカン組成物は、重合度が20~80であるβ-グルカンで構成され、ここで、重合度20~25の重量含有量は、16.5%であり、重合度25~30の重量含有量は、36.4%であり、重合度30~40の重量含有量は、22.8%であり、重合度40~50の重量含有量は、9.1%であり、重合度50~60の重量含有量は、5.6%であり、重合度60~70の重量含有量は、4.4%であり、重合度70~80の重量含有量は、5.2%であることを特徴とする、請求項4に記載のβ-グルカン組成物。
- 前記β-グルカン組成物は、重合度が10~80であるβ-グルカンで構成され、ここで、重合度10~20の重量含有量は、40.6%~60.8%であり、重合度20~25の重量含有量は、13.4%~20.0%であり、重合度25~30の重量含有量は、7.7%~11.5%であり、重合度30~40の重量含有量は、7.6%~11.4%であり、重合度40~50の重量含有量は、4.2%~6.2%であり、重合度50~60の重量含有量は、2.3%~3.5%であり、重合度60~70の重量含有量は、1.6%~2.4%であり、重合度70~80の重量含有量は、0.8%~1.2%であることを特徴とする、請求項4に記載のβ-グルカン組成物。
- 前記β-グルカン組成物は、重合度が10~80であるβ-グルカンで構成され、ここで、重合度10~20の重量含有量は、50.7%であり、重合度20~25の重量含有量は、16.7%であり、重合度25~30の重量含有量は、9.6%であり、重合度30~40の重量含有量は、9.5%であり、重合度40~50の重量含有量は、5.2%であり、重合度50~60の重量含有量は、2.9%であり、重合度60~70の重量含有量は、2.0%であり、重合度70~80の重量含有量は、1.0%であることを特徴とする、請求項4に記載のβ-グルカン組成物。
- 免疫関連疾患を改善または治療するための組成物の調製における、請求項1~8のいずれか1項に記載のβ-グルカンの使用。
- 前記免疫関連疾患は、腫瘍または炎症であることを特徴とする、免疫関連疾患を改善または治療するための組成物の調製における、請求項9に記載のβ-グルカンの使用。
- 前記腫瘍は、結腸直腸がん、肺がん、線維肉腫、黒色腫、腎臓がん、肝臓がん、乳がん、またはその組み合わせから選択されることを特徴とする、免疫関連疾患を改善または治療するための組成物の調製における、請求項10に記載のβ-グルカンの使用。
- 免疫関連疾患を改善または治療するための組成物であって、
前記組成物は、
(1)上記請求項中に記載のβ-グルカン、および
(2)薬学的に許容される担体を含むことを特徴とする、前記組成物。
