I.定義
i.ALDH2を標的とするオリゴヌクレオチド
本明細書では、複数の異なる種(ヒト、カニクイザル、及びマウス)のmRNAを含むALDH2のmRNAを調べることにより、インビトロ及びインビボでの試験で特定された、CNS内での強力なオリゴヌクレオチドが提供される。本明細書に記載されるように、かかるオリゴヌクレオチドを使用して、遺伝子活性(例えば、中枢神経系における)、この場合、ALDH2の活性を低下させることにより、神経疾患(例えば、神経変性疾患、認知障害、または不安障害)を有する対象において治療効果を得ることができる。本発明の方法及びオリゴヌクレオチドの標的とすることができる他の遺伝子としては、脊髄小脳失調症1型(アタキシン-1、及び/またはアタキシン-3)、β-アミロイド前駆体タンパク質遺伝子(APPまたはBACE1)またはその変異体、ジストニア(DYT1)、筋萎縮性側索硬化症「ALS」またはルーゲーリック病(SOD1)を引き起こす遺伝子として特定されているもの、及び中枢神経系の腫瘍につながる様々な遺伝子が挙げられる。例えば、本明細書では、付録Aに示される表にも示されるような(例えば、配列番号585に記載される配列を含むセンス鎖と配列番号595に記載される配列を含むアンチセンス鎖)、配列番号581~590、608、及び609のいずれか1つに記載される配列を含む、またはそれからなるセンス鎖と、配列番号591~600から選択される相補的配列を含む、またはそれからなるアンチセンス鎖とを有する強力なRNAiオリゴヌクレオチドが提供される。
RNAi、miRNAなどを含む、本開示の方法においてALDH2を標的化するのに有用なオリゴヌクレオチドの様々な構造が存在する。本明細書または他の場所に記載の構造のいずれかを、本明細書に記載される配列(例えば、配列番号601~607に示されるものなどのALDH2のホットポット配列)を組み込む、または標的化するためのフレームワークとして使用することができる。ALDH2発現を標的化する(例えば、RNAi経路を介して)ための二本鎖オリゴヌクレオチドは、一般に、互いに二本鎖を形成するセンス鎖とアンチセンス鎖を有する。いくつかの実施形態では、センス鎖とアンチセンス鎖とは共有結合していない。しかしながら、いくつかの実施形態では、センス鎖とアンチセンス鎖とは共有結合している。

いくつかの実施形態では、本明細書で提供されるオリゴヌクレオチドは、センス鎖及びアンチセンス鎖を含み、センス鎖もしくはアンチセンス鎖のいずれか、またはセンス鎖及びアンチセンス鎖の両方に3’オーバーハングが存在する。いくつかの実施形態では、本明細書で提供されるオリゴヌクレオチドの一方の5’末端は、他方の5’末端と比較して熱力学的に不安定である。いくつかの実施形態では、センス鎖の3’末端に平滑末端を、アンチセンス鎖の3’末端にオーバーハングを有する非対称なオリゴヌクレオチドが提供される。いくつかの実施形態では、アンチセンス鎖上の3’オーバーハングは、ヌクレオチド1~8個の長さ(例えば、ヌクレオチド1個、2個、3個、4個、5個、6個、7個または8個の長さ)である。
いくつかの実施形態では、オリゴヌクレオチドは、センス鎖とアンチセンス鎖との間の1つ以上(例えば、1個、2個、3個、4個、5個)のミスマッチを有する。センス鎖とアンチセンス鎖との間に複数のミスマッチが存在する場合、それらは連続して配置されるか(例えば、2個、3個、またはそれ以上が連続して)、または相補性の領域全体にわたって散在してもよい。いくつかの実施形態では、センス鎖の3’末端が1つ以上のミスマッチを含む。一実施形態では、2個のミスマッチがセンス鎖の3’末端に組み込まれる。いくつかの実施形態では、オリゴヌクレオチドのセンス鎖の3’末端のセグメントの塩基のミスマッチまたは不安定化は、おそらくはダイサーによるプロセシングを促進することにより、RNAiにおける合成二重鎖の効力を改善した。
いくつかの実施形態では、本明細書に記載されるALDH2発現を低減するためのオリゴヌクレオチドは、一本鎖である。そのような構造には、これに限定されるものではないが、一本鎖RNAiオリゴヌクレオチドが含まれ得る。最近の取り組みにより、一本鎖RNAiオリゴヌクレオチドの活性が実証されている(例えば、Matsui et al.,Molecular Therapy,2016,24(5):946-955)。ただし、いくつかの実施形態では、本明細書で提供されるオリゴヌクレオチドは、アンチセンスオリゴヌクレオチド(ASO)である。アンチセンスオリゴヌクレオチドは、5’~3’方向に書いた場合に特定の核酸の標的セグメントの逆方向の相補体を含み、細胞内でその標的RNAのRNaseHによって媒介される切断を誘導するように、または(例えば、ミクスマーとして)細胞内の標的mRNAの翻訳を阻害するように適当な修飾が行われた(例えば、ギャップマーとして)一本鎖オリゴヌクレオチドである。本開示で使用するためのアンチセンスオリゴヌクレオチドは、例えば、アンチセンスオリゴヌクレオチドの改変(例えば、長さ、核酸塩基の糖部分(ピリミジン、プリン)、及び核酸塩基の複素環部分の改変を含む)に関するその開示について参照によって本明細書に援用する米国特許第9,567,587号に示されるものを含む、当該技術分野では周知の任意の適切な方法で改変することができる。また、アンチセンス分子は、特定の標的遺伝子の発現を低下させるために数十年にわたって使用されてきた (例えば、Bennett et al.,Pharmacology of Antisense Drugs,Annual Review of Pharmacology and Toxicology,2017,57:81-105を参照)。
オリゴヌクレオチドは、特異性、安定性、送達、バイオアベイラビリティ、ヌクレアーゼ分解に対する耐性、免疫原性、塩基対合特性、RNA分布及び細胞取り込み、ならびに治療または研究用途に関連する他の特性を改善または制御するために様々な方法で改変することができる (例えば、Bramsen et al.,Nucleic Acids Res.,2009,37:2867-2881;Bramsen and Kjems,Frontiers in Genetics,2012,3:1-22)。したがって、いくつかの実施形態では、本開示のオリゴヌクレオチドは、1つ以上の適当な改変を含むことができる。いくつかの実施形態では、改変ヌクレオチドは、その塩基(または核酸塩基)、糖(例えば、リボース、デオキシリボース)、またはリン酸基に改変を有する。

いくつかの実施形態では、オリゴヌクレオチドは、修飾されたヌクレオシド間結合を含む。いくつかの実施形態では、リン酸の修飾または置換は、少なくとも1個(例えば、少なくとも1個、少なくとも2個、少なくとも3個、又は少なくとも5個)の修飾ヌクレオチド間結合を含むオリゴヌクレオチドを生じ得る。いくつかの実施形態では、本明細書に開示されるオリゴヌクレオチドのいずれか1つは、1~12個 (例えば、1~12個、1~10個、2~10個、2~8個、4~6個、3~10個、5~10個、1~5個、1~3個または1~2個)の修飾ヌクレオチド間結合を含む。いくつかの実施形態では、本明細書に開示されるオリゴヌクレオチドのいずれか1つは、1、2、3、4、5、6、7、8、9、10、11、または12個の修飾ヌクレオチド間結合を含む。

オリゴヌクレオチドの使用を促進するための様々な製剤が開発されている。例えば、オリゴヌクレオチドは、分解を最小限に抑え、送達及び/または取り込みを促進し、または製剤中のオリゴヌクレオチドに別の有益な特性を与える製剤を使用して対象または細胞環境に送達することができる。いくつかの実施形態では、本明細書では、ALDH2の発現を低下させるためのオリゴヌクレオチド(例えば、一本鎖または二本鎖オリゴヌクレオチド)を含む組成物が提供される。かかる組成物は、標的細胞の隣接環境にまたは全身的に対象に投与された場合に、オリゴヌクレオチドの十分な部分が細胞に入ってALDH2発現を低下させるように適当に配合することができる。各種の適当なオリゴヌクレオチド製剤のいずれかを使用して、本明細書に開示されるALDH2を低減させるためのオリゴヌクレオチドを送達することができる。いくつかの実施形態では、オリゴヌクレオチドは、リン酸緩衝生理食塩水などの緩衝液、リポソーム、ミセル構造、及びカプシド中に配合される。いくつかの実施形態では、裸のオリゴヌクレオチドまたはそのコンジュゲートが、水または水溶液(例えば、pH調整された水)中に配合される。いくつかの実施形態では、裸のオリゴヌクレオチドまたはそのコンジュゲートが、塩基性緩衝水溶液(例えば、PBS)中に配合される。
i.細胞内のALDH2発現の低下
いくつかの実施形態では、細胞内のALDH2の発現を低下させる目的で、本明細書に開示されるオリゴヌクレオチドのいずれか1つの有効量を細胞に送達するための方法が提供される。本明細書で提供される方法は、任意の適当な細胞タイプで有用である。いくつかの実施形態では、細胞は、ALDH2を発現するあらゆる細胞である(例えば、肝細胞、マクロファージ、単球由来細胞、前立腺癌細胞、中枢神経系の細胞(例えば、ニューロンまたはグリア細胞)、内分泌組織、骨髄、リンパ節、肺、胆嚢、肝臓、十二指腸、小腸、膵臓、腎臓、胃腸管、膀胱、脂肪及び軟組織及び皮膚)。いくつかの実施形態では、細胞は、対象から得られ、限られた数の継代を行っていてもよい初代細胞であり、それにより、細胞は天然の表現型特性を実質的に維持している。いくつかの実施形態では、オリゴヌクレオチドが送達される細胞は 、エクスビボまたはインビトロである (すなわち、培養中の細胞または細胞が存在する生物に送達することができる)。具体的な実施形態では、中枢神経系(CNS)内のALDH2のみの発現を低下させる目的で、本明細書に開示されるオリゴヌクレオチドのいずれか1つの有効量を細胞に送達するための方法が提供される。
別の態様において、本開示は、対象の神経学的疾患を治療するためにALDH2の発現を低下させるための方法に関する。いくつかの実施形態では、方法は、治療を要する対象の脳脊髄液に本明細書に開示されるオリゴヌクレオチドのいずれか1つの有効量を投与することを含み得る。かかる治療法を用いて、例えば、中枢神経系(例えば、体性感覚野、海馬、前頭葉、線条体、視床下部、小脳、及び脊髄全体)におけるALDH2の発現を低下させることができる。本開示は、神経学的疾患のリスクを有する(または感受性を有する)対象を治療する予防的及び治療的方法の両方を提供する。いくつかの実施形態では、本開示は、神経疾患を治療するためのオリゴヌクレオチドの方法または使用を提供する。いくつかの実施形態では、神経疾患は、神経変性疾患、認知障害、または不安障害である。CNSにおけるALDH2発現に関連する例示的な疾患としては、とりわけ、老人性痴呆、ジスキネジア、アルツハイマー病(AD)、及びパーキンソン病(PD)が挙げられる。
いくつかの態様において、本開示は、対象における標的遺伝子の発現を低下させるためにオリゴヌクレオチド誘導体(例えば、コンジュゲートA、B、C、D、E、F、またはG)を使用する方法を提供する。
実施例1:中枢神経系(CNS)へのGalNAc結合ALDH2オリゴヌクレオチドの送達
中枢神経系(CNS)は保護された環境である。脳脊髄液(CSF)中の循環タンパク質含有量は血漿中の含有量の1%未満であり、CSFには内因的なヌクレアーゼ活性はほとんどない。CNSは、血液脳関門が免疫細胞の循環を妨げることから「免疫特権化」されている。CSF中に投与されたオリゴヌクレオチドは 、CSFのバルク流によって分布し、組織中で長い半減期を有する(脳室内(ICV)注入後、脳及び脊髄内で最大200日)。神経細胞はオリゴヌクレオチドを容易に取り込む。RNAiオリゴヌクレオチドのサイズ及び/または親油性は、CSFからのオリゴヌクレオチドの排除を減少させるように操作することができる。しかしながら、RNAiオリゴヌクレオチドは、血液脳関門を通過しないため、CNS内に直接投与する必要がある(例えば、髄腔内またはICV注射)。オリゴヌクレオチドは リンパ系を介してCSFから除去され、全身投与されたオリゴヌクレオチドと同じ懸案事項/制限の対象となる(例、腎毒性、血小板減少症)。本開示の一実施形態では、活性ガイド鎖は、CNSからの急速な排除から化合物を保護するために化学的に修飾されたより大きなオリゴヌクレオチド担体として調製される。オリゴヌクレオチド担体の化学的修飾としては、それぞれ、ガイド鎖がRISCに積み込まれ、標的mRNAを阻害するまでの間、分子全体を除去するCNSの能力を低下または遅延させることを目的とした、単純に大きな分子サイズ、親油性、二量体化、電荷または極性の改変、分子量の増加が含まれる。
GalNAc結合ALDH2オリゴヌクレオチド(S585-AS595-コンジュゲートA)を上記と同じアッセイを使用し、ただし、2つの異なる濃度(250μg及び500μg)で試験した。GalNAc結合ALDH2オリゴヌクレオチドを ICVによりマウスに投与し、投与後7日目または28日目に組織(線条体、皮質(体性感覚野及び前頭葉)、海馬、視床下部、小脳、脊髄)を採取した。各組織中に残存するALDH2のmRNAレベルを、RT-PCTを用いて評価した。各組織中のGalNAc結合ALDH2オリゴヌクレオチドの量を、SL-qPCTを使用して評価した。実験デザインを表2に示す。

単回ボーラスICV注射後の脳及び脊髄におけるGalNAc結合ALDH2オリゴヌクレオチド(S585-AS595-コンジュゲートA)の効果の持続時間の評価も行った。GalNAcコンジュゲートALDH2オリゴヌクレオチドを、250μg及び500μgの2つの用量レベルで右側脳室へのICV注入によってCD-1雌マウス(6~8週齢)に投与した。注入の7、28、及び56日後にマウスを屠殺し、組織(線条体、皮質(体性感覚野及び前頭葉)、海馬、視床下部、脊髄)を採取した。各組織中に残存するALDH2のmRNAレベルを、RT-PCTを用いて評価した。実験計画を下記表3に示す。
ニューロンへの送達にGalNAc結合が必要であるかどうかを判定し、中枢神経系(CNS)でALDH2阻害活性を有するGalNAc結合ALDH2オリゴヌクレオチドの構造変異体を特定するため、一群のALDH2 RNAiオリゴヌクレオチド誘導体を設計した(コンジュゲートA~G、図23)。すべての誘導体は、テトラループ構造の有無にかかわらず、センス鎖の5’末端に異なる構造を形成している。オリゴヌクレオチド誘導体のテトラループ部分における例示的な修飾ヌクレオチドを図22に示す。加えて、いずれのオリゴヌクレオチドも、2’-フルオロ及び2’-O-メチル修飾ヌクレオチド、ホスホロチオエート結合の組み合わせをさらに含み、及び/またはアンチセンス鎖の5’末端ヌクレオチドに位置するリン酸アナログを含んでいる。
