核内受容体
核内受容体は、細胞環境の化学的変化を転写上の変化、したがって生物学的変化に変換する際に極めて重要な役割を演じる。この機能は、細胞だけではなく生物体全体の恒常性を維持する上で不可欠である。核内受容体は、種にわたってその数に多大な差異があり、例えば、ヒトは48種を有し、C.エレガンス(C. elegans)は270種を有するが、後生動物においてのみ見出される。
核内受容体は質量50~100kDaの間を有し、成熟ポリペプチドは5つのドメインに分類される:
A/B:受容体間で高度に変動する。リガンドの非存在下では弱い転写活性化因子として作用するが、リガンドが結合すると強力な活性化因子として作用する活性化機能1(AF-1)を含有する。これは、EドメインのAF-2との相互作用によるものである。
C:DNA応答エレメントに結合する2つのジンクフィンガーを含有する高度に保存されたドメイン。
D:LBDとDBDの間を接続し、相互作用を可能にするフレキシブルドメイン。細胞トラフィッキング及び細胞内分布において重要である。
E:構造は高度に保存されているが、配列は中程度のみの保存である。リガンド結合キャビティを含有し、受容体にリガンド特異性を付与する。DBDとともに二量体形成面に役に立ち、コアクチベーター及びリプレッサーとも結合する。その作用がリガンド結合に左右される活性化因子2(AF2)を含有する。
F:高可変性C末端ドメイン。
核内受容体は、その作用機構に基づいて4つのタイプに分類できる。下は、各タイプのまとめである。

構成的アンドロスタン受容体(CAR)は - 又は核内受容体サブファミリー1、グループI、メンバ3 - 肝臓細胞でほぼ排他的に発現する核内受容体スーパーファミリーのメンバである。ここでは、CARは別の核内受容体であるプレグナンX受容体(PXR)と協力して、内因性物質及び生体異物の化学物質のセンサーを演じる。物質を活性化することによって結合すると、これらの受容体は、シトクロムp450を含めて、多数の遺伝子の活性をモジュレートし、したがってそれら化合物の代謝と排泄を担っている。CAR及びPXRが体内の外来化学物質の解毒に主要な役割を果たすのは、この結合と遺伝子活性化能によるものである。
上に示したように、CARは生体外異物及び内因性物質の代謝の重要な調節因子として機能する。それは肝臓において24の予測転写物を有する。これらの転写物の一部は、この受容体の低レベルの構成的活性の原因であるが、一方、他の転写物は誘導性であることが示された。構成的活性は、ステロイド受容体コアクチベーター1(SRC1)等の転写コアクチベーターとの相互作用によってモジュレートされると考えられている。この活性は、アンドロスタン等のインバースアゴニストの結合によって抑制することができる。
不活性CARはリン酸化され、細胞の細胞質に存在する。ここで、不活性CARは、ヒートショックタンパク質90(hsp90)及び細胞質CAR保持タンパク質(CCRP)と複合体を形成するが、この会合とは、CARを細胞質に維持し、したがって不活性のまま維持するところのものである。この不活性CARは2つの様式で、1)マウスCARリガンドであるTCPOBOPよる等の直接的なリガンド結合、又は2)フェノバルビタールを介した間接的な活性化、で活性化することができる。両経路によって、多タンパク質複合体からのCARの解離がもたらされ、核へのその移行が可能になる。核内では、CARは単量体として作用することができるか、又はレチノイドX受容体(RXR)とヘテロダイマーを形成することができる。核CARは、フェノバルビタール応答エレメント(PBREM)でDNAと結合し、これを通して、核CARは、CAR調節遺伝子、例えばCYP2B、CYP2C及びCYP3Aサブファミリーを活性化する。
TCPOBOPは、マウスCARに直接結合し、核へのその移行を誘導すると考えられる。しかし、この化学物質はヒトのCARに結合せず、したがって、ヒトの研究の場合CITCOが等価の化合物である。
抗けいれん薬であるフェノバルビタール(PB)によるCARの間接的活性化は、間接的CAR活性化のモデルとして広く認められている。PBによって、ホスファターゼPP2Aの活性化を通してCARの脱リン酸化が起こる。PP2A活性化の正確な機構は不明であるが、PBによってAMP活性化プロテインキナーゼが活性化され、このプロテインキナーゼは、多タンパク質複合体によって次いで動員されるPP2Aを活性化することができる。
マウスCARホモログのDNA結合部位が、Honkakoskiら(MOLECULAR PHARMACOLOGY、53:597~601頁(1998))によって特定された。この研究で、著者らは、RXR及びCARヘテロダイマーが、フェノバルビタール誘導に応答してシトクロムP-450 Cyp2b10遺伝子のフェノバルビタール応答性エンハンサーモジュール(PBREM)の部位に結合することを見出した。哺乳動物細胞株においてRXR及びCARの発現はPBREMを活性化したが、このことにより、CAR-RXRヘテロダイマーがCyp2b10遺伝子のトランス作用因子であることが示される。このヘテロダイマーが2つの不完全な直列反復配列-4モチーフに結合すること、及びこのモチーフがヒトで保存されていること、も示された。PBREMエレメントを図6に示すが、ここで、NR1とNR2は、太字体である不完全な直列反復配列を有する核内受容体結合部位である。NFI結合部位が示されている。CAR-RXRヘテロダイマーはNR1部位に結合すると思われる。

- 細胞を25,000細胞/300μlの密度で48ウェルプレートに播種した
2日目
- トランスフェクション当日、トランスフェクションするDNA(CARプラスミド/ルシフェラーゼ又はEPOと作動可能に連結されたPB1-MinTK/内部対照としてpcDNA6プラスミド)を100ng/μlストック溶液に希釈した。
48ウェルトランスフェクションあたり:
- DNA 45ng(各プラスミド15ng、pcDNA6、CAR、及び試験プラスミド)をOptimem培地4.1μlと混合した。
- FusionHD 0.5μlをOptimem培地4μlと混合した。
- これらの2つの溶液を混合し、室温で15分間インキュベートした。
- 次いで、最終溶液をウェルに滴下添加した。
- トランスフェクションの3時間後、誘導剤TCPOBOPを指示濃度で適当なウェルに添加した。
3日目
- 誘導の24時間後、培地を細胞から除去した。
- 細胞をDPBS 300μlで1回洗浄した。
- passive lysis buffer 100μl及び揺動しながらの15分間インキュベーションを使用して細胞を溶解した。
- benchtop centrifugeで最高速度でプレートを1分間遠心分離することにより、細胞デブリをペレット化した。
- ルシフェラーゼの場合、試料10μlを白色96ウェルプレートに移し、LARII基質50μlを注入して発光を測定した。
- β-ガラクトシダーゼ活性を、溶解物25μlを使用して、製造元の取扱説明書(哺乳動物β-ガラクトシダーゼアッセイキット、75707/75710、Thermo Scientific社)に従って測定した。溶解物25μlをマイクロプレートウェルに移し、β-ガラクトシダーゼアッセイ試薬25μlと混合し、室温に平衡化させた。混合物を37℃で30分間インキュベートし、吸光度を405nmで測定した。
- タンパク質濃度を、溶解物25μlを使用して、製造元の取扱説明書(Pierce(商標)BCAタンパク質アッセイキット、23225/23227、Thermo Scientific社)に従って測定した。溶解物25μlをマイクロプレートウェルに移し、希釈標準溶液200μlと混合した。混合物を37℃で30分間インキュベートし、室温に冷却し、次いで吸光度をおよそ562nmで測定した。タンパク質濃度を、既知のタンパク質濃度を有する標準物質をアッセイすることから作成されたタンパク質標準曲線に従って計算した。
図1AのPB1-MinTK構築物からのルシフェラーゼ発現が示したところは、細胞をビヒクル(DMSO)で処理した場合、プロモーターからの測定可能なルシフェラーゼ活性がほんのわずかであったことである。この活性は、プロモーターからの漏出発現に相当する。50nM又は150nMのTCPOBOP添加によってプロモーターの強力な誘導がもたらされたが、この場合、6倍までの誘導が測定された。250nMのTCPOBOP添加では、このプロモーターからの応答を誘発しなかったが、このことにより、細胞に過重な負担をかけることを防止するために恒常性制御機構が活性化された可能性があること、が示唆される。したがって、PB1-MinTK構築物は、Huh7細胞においてTCPOBOPを添加することによって誘導される。誘導はTCPOBOPの濃度に応じて調節可能であるが、より高濃度では低下する。

ARE肝細胞を、製造元のマニュアルに記載の通りに培養及びトランスフェクトした。コラーゲンコーティングの6ウェルプレート中、培養培地2mlに200,000個の細胞を播種した。十分な接着のために、細胞を37℃及び5%CO2で4時間インキュベートした。トランスフェクトするDNAを含有するトランスフェクション混合物(ルシフェラーゼ及びβ-ガラクトシダーゼ含有プロモーターと作動可能に連結されたPB1-MinTK/PB1-CMV/PB1-SV40)200μlを添加し、細胞をオービタルシェーカー上で100rpmで37℃及び5%CO2で3時間インキュベートした。トランスフェクションの3時間後、CITCOを適当なウェルに添加した。細胞を37℃及び5%CO2での静的条件下で一晩インキュベートし、朝に培地を新鮮なARE肝細胞維持培地と交換した。読取りは誘導24時間後とした。
図2AのPB1-MinTK、PB1-CMV-MP、及びPB1-SV40-MPの各構築物からのルシフェラーゼ発現が示すところは、ミニマルプロモーターそれぞれが1μM CITCOを添加すると、PBREMエレメントからの発現をサポートすること、である。誘導はSV40とMinTKでおよそ7倍、CMV-IEプロモーターで2倍であった。各構築物の発現レベルは、CMV-IEと比較して、それぞれPB1-MinTK、PB1-CMV、及びPB1-SV40で、20%、10%及び55%である。SV40は、最高の発現を駆動することができるが、バックグラウンドレベルがより高いという代償が伴う。CMVミニマルプロモーターはほとんど又はまったく発現を示さない。このデータから、元のMinTk構築物は、発現レベルと、誘導性と、制御の厳格さ(すなわち、バックグラウンド発現を最小限に抑える)との間で最良の妥協点を提供することができると思われる。
PB1-MinTK、PB1-1-MinTK、PB1-2-MinTK、及びPB1-3-MinTKは、MinTKミニマルプロモーターの組合せでそれぞれ1、2、3、及び4つのPBREMエレメントを含有する。図3Cで、CITCO 1μlで誘導した場合の、PB1-1-MinTK、PB1-2-MinTK、及びPB1-3-MinTKの各構築物からのルシフェラーゼ発現が示すところは、多量体が誘導され、多量体によって発現レベルが増加することである。しかし、この発現レベルの増加は、PBREMエレメント(PB1~2)の3コピーまでに限って観察される、なぜなら、もう1つのエレメントの付加は、誘導及び発現のレベルに悪影響を与えると思えるからである。各多量体は、それぞれCMV-IEの1.5、4.1、2.66まで誘導される。しかし、誘導のレベルは、ここでPB1として示されている最初のPB1-MinTK構築物に類似する。これは、プロモーターのバックグラウンド活性の増加によるものである。結果をCMV-IEに対する比として表す。結果は3回の生物学的繰返しの平均である。
PB1-SV40、PB1-1-SV40、PB1-2-SV40及びPB1-3-SV40は、SV40ミニマルプロモーターの組合せでそれぞれ1、2、3、及び4つのPBREMエレメントを含有する。図3Aで、CITCO 1μlで誘導した場合のPB1-1-SV40、PB1-2-SV40、及びPB1-3-SV40の各構築物からのルシフェラーゼ発現が示すところは、多量体が誘導され、多量体によって実際発現レベルが増加することである。しかし前のように、発現レベルのこの増加は、PBREMエレメントの3コピーまでに限って観察される、なぜなら、もう1つのエレメントの付加は、誘導及び発現のレベルに悪影響を与えるようであるからである。各多量体は、それぞれCMV-IEの2.6、3.6、及び2.57まで誘導される。誘導のレベルは、9倍までの誘導を伴って、MinTkミニマルプロモーターで見られる6倍の増加よりも高い。ここでも、発現のバックグラウンドレベルが増加するが、これは、下に記載のCMV-MPプロモーターで見られるよりもはるかに少ない。結果をCMV-IEに対する比として表す。結果は3回の生物学的繰返しの平均である。
PB1-CMV、PB1-1-CMV、PB1-2-CMV、及びPB1-3-CMVは、CMVミニマルプロモーターの組合せでそれぞれ1、2、3、及び4つのPBREMエレメントを含有する。図3Bで、CITCO 1μlで誘導した場合のPB1-1-CMV、PB1-2-CMV、及びPB1-3-CMV構築物からのルシフェラーゼ発現が示すところは、多量体が誘導され、多量体によって実際発現レベルが増加することである。しかし前のように、発現レベルのこの増加は、PBREMエレメントの3コピーまでに限って観察される、なぜなら、もう1つのエレメントの付加は、誘導及び発現のレベルに悪影響を与えるようであるからである。各多量体は、CMV-IEのそれぞれ1.9、2.67、及び2.67まで誘導される。誘導のレベルは、MinTk又はSV40のミニマルプロモーターのどちらででも観察されるよりも低く、最大5倍である。CMVミニマルプロモーターを使用すると、発現のバックグラウンドレベルが極めて高レベルに増加するように思われ、したがって、評価し得る最も劣る候補であるとすることができる。
先の適用から、構築物PB1(単一のマウスPBREMエレメントとMin-TKプロモーター)及びPB1-2(3×PBREMとmin-TKプロモーター)をマウスでのin vivo試験用に選択した。これは以下のように実行した。
1日目:
- 15cmプレートにHEK293-AAV細胞を播種する。トランスフェクションの当日、70~80%のコンフルエントである
- 各プレートの最終容量:15ml
2日目:トランスフェクションミックスを調製する:
- DNAミックス/プレート:pDG9(AAV9用パッケージングプラスド):10.5μg/pHGTI(Ad.ヘルパープラスミド):31.5μg/ベクタープラスミド:10.5μg/無血清DMEM/Optimem中で調製
- トランスファーミックス/プレート:PEI:125μl/無血清DMEM/Optimem中で調製
- トランスフェクションミックスにDNAミックスを添加する。混合し、室温で15~20分間放置する。
- トランスフェクションミックス3mlを各プレートに滴下添加し、穏やかに分配する。24時間インキュベートする。
3日目:
- 培地をP/Sと2%FCSを補充したDMEM 15mlに交換する。48時間放置する。
5日目:
- 上清を収集し、各チューブ25mlで50mlチューブにプールする。-20℃で保存する。
- 細胞を収集する:各プレートにPBS 5mlを添加する→50mlのチューブに掻爬し収集する。
- プレートを洗浄するために、さらにPBS1mlを添加し、回収する。
- 1500rpmで5分間回転する。
- 上清を除去し、ペレット化細胞を1ml/プレートのTD溶解バッファに再懸濁する→プールする
- -80℃で保存する。
細胞:
- ペレットを5回凍結融解する→37℃でおよそ20分間、次いでドライアイス(又は-80℃)でおよそ20分間。
- 20%デオキシコール酸25μl/ml細胞(又は10%デオキシコール酸50μl/ml)を添加する。
- ベンゾナーゼ8μl/ml細胞を添加する。
- 37℃で30分間インキュベートする。
- 4Krpmで30分間回転する。
- 0.45μMフィルターを使用して上清をろ過する。
- 4℃で最長24時間保存する。
上澄み:
- ベンゾナーゼ2.5μl/25ml上清を添加する。
- MgSO4 50μl/25ml上清を添加する。
- 37℃で30分間インキュベートする。
- 4Krpmで30分間回転させる。
- 0.45μMフィルターを使用して上清をろ過する。
- 4℃で最長24時間保存する。
HPLC精製
- 両系列を20%ETOHに入れる→テンプレート→システムウォッシュ
- 系列AをPBSに置き、系列Bをグリシンに置く→テンプレート→システムウォッシュ
- カラムを機械にいれる→マニュアルラン→流速:5ml/min→25mlの間又はUVラインがフラットになるまでラン。
- FACSチューブを用意する(細胞用に10本、上清用により多く):Tris 30μl/チューブを添加する→ベクターを収集するために機械に置く。
- 廃液系列を別のチューブに入れて、カラムに再び通すことができるようにする。
- 試料をランする(流速は低い、システムがどのくらい速いか及び試料をどのくらい濃縮するかに依存する;細胞の場合は流速はより遅く、上清の場合はより高くなる)。
- 廃液をカラムに通してランさせた後、PBSで洗浄する→流速:UVラインがフラットになるまで5ml/min。
- 設定→フラクションサイズ:1ml;流速:1ml/min;濃度%B:100%→ラン。
- 収集を開始する。ピークを探す。ピークはベクターの精製を示す。これらのベクターを含有するチューブをマークする。
- 終了前にプログラムを保存する。
- PBSで洗浄する→75ml、5ml/min
- Na3PO4で洗浄(カラムを保存するため)→75ml、5ml/min
- カラムを取り外し、4℃で保存する。
- PBSで機械を洗浄する→テンプレート→システム洗浄
- 20%ETOHで機械を洗浄する→テンプレート→システム洗浄
- 両系列を20%ETOHに置き、シャットダウンする
- PBS 2Lを大きなバケツに添加し、透析カセット(Side-A-Lyzer; Thermo Scientific社)を置いてプライミングする。
- 注射器と針を使用してマークしたFACSチューブからベクターを収集し、透析カセットに添加する→膜から余分な空気を取り除き、カセットの上にゴムを注意深く配置し、PBS含有バケツにこれを浮かべる→ゆっくり回転するローター上で室温で一晩放置する。
翌日:
- PBS 5mlを遠心フィルター(Amico Ultra 15; MERCK社)上に添加することによってメンブレンをプライミングする→4Krpmで5分間回転させる。
- メンブレンの内側から余分なPBSを除去する。
- カセットからベクターを取り出し、メンブレンにロードする→4Krpmで5分間回転させる
- 内部にベクターを有するメンブレンを数回洗浄し、次いで、2ml遠心チューブフィルター0.22μM(Spin-X; COSTAR社)に回収する。
- 13Krpmで3分間回転させる。
- フィルター及び一定分量:1×100μl(注入用)を取り出す、残部は10~2μl一定分量。
- 80℃で保存する。
- ウイルスの定量化を、ルシフェラーゼ遺伝子に対するプライマーとプローブでqRT-PCRを使用して実行した。
指定のAAV血清型はAAV9としたが、これは、ほとんどの組織及び器官に対して指向性があり、したがって、本発明のプロモーターの特異性に関する理想を与えると思えるからである(AAV指向性の問題を回避する)。実験からのアウトプットは、注射後5日目の第1の読取りにより視覚的に測定されたルシフェラーゼ活性とした。次いで、マウスを毎週モニタリングし、35日後、CMV-IEを含むコントロールベクターAAV9が一定した着実な結果を示したら、誘導プロファイリングのベースラインを確立した。この時点で誘導物質を添加したが、測定値については誘導の前後に取った。より詳細については下を参照されたい。
マウス:
- 成体(8週齢)のCD1雄マウスに尾静脈を介してAAV9ベクターを注射した。
- 計5×1011ベクターゲノムコピー数/mlをマウス1匹あたりに投与した。
- 注射の5日後にマウスを画像化した。マウスはまず麻酔され、ルシフェリンの腹腔内注射を受けた(300μlのルシフェリンストック15mg/ml)。5分後、マウスをIVISマシンに配置し、画像を取得した。
画像化:
- 画像に使用した露光時間を1秒及び10秒とした。
- 画像を週に1回撮影した
- 更に、誘導剤又はリプレッサーを投与する前に、マウスを4日間にわたって毎日画像化した。
誘導:
誘導剤(フェノバルビタール)を5mg/mlの濃度とした。マウスは腹腔内に10μl、すなわちマウスあたり50マイクログラム(各マウスは体重概ね30g)を受けた。
実験の結果を図8で見られる。図8Aは、試験した各構築物からの代表的なマウスを示す。ここでは、PB1とPB1-2の両方の発現が肝臓に限定されているのに対し、一方、CMV-IEプロモーターはマウスのほぼ全組織で発現していることがわかる。更に、0時間では、PB1及びPB1-2のマウスはルシフェラーゼ遺伝子の発現をまったく示さず、これによって、厳格に制御された発現が示唆される。しかし、誘導剤フェノバルビタールを添加すると、PB1とPB1-2の両方からの発現が増加することがわかる。誘導の大きさと持続時間にはいくらかの変動がある。例えば、PB1は、9時間で見られた最大活性で概ね10倍を誘導したが、誘導は24時間で完了であり、これに対して、PB1-2は24時間での最大活性で概ね50倍の増加を有し、この場合、注入後48時間まで誘導は完了しない(図8B及び図8C)。これらのデータは、モデル細胞株で観察された発見を裏付けるものであり、in vivoで使用するための本発明の誘導性システムの可能性を更に実証するものである。本発明のシステムは、誘導剤がヒトの推奨投薬量よりも10分の1で投与された場合でも、バックグラウンドが低く且つ誘導性が良好である。
上で論議のように、核内受容体CARはヒトとマウスの両方のDNA配列に結合する。配列アライメントからわかるように、種間にはある程度の配列分散がある。51-bpのモジュラーPBREMエレメントはそれ自体、異なる3部分に分解することができる(下の表を参照されたい)。1)NR1領域(NR1エレメントを含有する)、誘導性活性のほとんどを担うと考えられている、2)NF1領域(NF1エレメントを含有する)、他の核内受容体と結合し、活性化CARの非存在でのバックグラウンドレベルの低下を担うことができる、及び3)NR2領域(NR2エレメントを含有する)、CARの誘導性活性にやはり関与してくる。

- 配列番号7、配列番号10、配列番号11、配列番号12、配列番号68、配列番号69、配列番号70又は配列番号71による配列、又はそれらのいずれか1つの機能的バリアントを含む、請求項1から17のいずれか一項に記載の遺伝子治療ベクター。
- 合成肝臓特異的発現カセットは、リボソーム結合部位、開始コドン、終止コドン、転写終止配列、転写後調節エレメントをコードする核酸及び/又はポリAエレメントのうちの1つ又は複数、好ましくはそれらのすべてを提供又はコードする配列を含む、請求項1から18のいずれか一項に記載の遺伝子治療ベクター。
- 遺伝子はタンパク質又はRNAをコードする、請求項1から19のいずれか一項に記載の遺伝子治療ベクター。
- 遺伝子は、治療用発現産物、好ましくは肝臓における異常な遺伝子発現に関連付けられる疾患又は状態の処置で使用するのに適した治療用タンパク質をコードする、請求項1から20のいずれか一項に記載の遺伝子治療ベクター。
- 遺伝子は、メガヌクレアーゼ、ジンクフィンガーヌクレアーゼ(ZFN)、転写活性化因子様エフェクターベースのヌクレアーゼ(TALEN)、又はクラスター化され規則的に間隔が空いた短い回文構造の反復(CRISPR-Cas)システム等の、部位特異的ヌクレアーゼをコードする、請求項1から21のいずれか一項に記載の遺伝子治療ベクター。
- レトロウイルス、レンチウイルス、アデノウイルス、又はアデノ随伴ウイルス(AAV)の各ベクター等の、ウイルスベクターである、請求項1から22のいずれか一項に記載の遺伝子治療ベクター。

- 遺伝子に作動可能に連結された合成肝臓特異的誘導性プロモーターであって、
CARとRXRのヘテロダイマーによって結合及び活性化されることが可能なCREを含む、合成肝臓特異的誘導性プロモーターが含まれる、
発現カセット。 - 遺伝子はレポーター遺伝子ではない、請求項25に記載の発現カセット。
- 遺伝子は治療用発現産物をコードする、請求項25又は26に記載の発現カセット。
- 配列番号7~18及び59~71のうちのいずれか1つによる配列、又はそれらのいずれか1つの機能的バリアントを含む合成肝臓特異的誘導性プロモーター。
- 請求項1から24のいずれか一項に記載の遺伝子治療ベクター、請求項25から27のいずれか一項に記載の発現カセット、又は請求項28に記載のプロモーター、を含む組換えビリオン。
- 請求項1から24のいずれか一項に記載の遺伝子治療ベクター、請求項25から27のいずれか一項に記載の発現カセット、請求項28に記載のプロモーター、又は請求項29に記載のビリオンを含む、医薬組成物。
- 請求項1から24のいずれか一項に記載の遺伝子治療ベクター、請求項25から27のいずれか一項に記載の発現カセット、請求項28に記載のプロモーター、又は請求項29に記載のビリオン、を含む細胞。
- 肝臓細胞である、任意選択で細胞培養中の肝臓細胞又は対象の肝臓に存在する肝臓細胞である、請求項31に記載の細胞。
- 疾患、好ましくは肝臓における異常な遺伝子発現に関連付けられる疾患の処置に使用するための、請求項1から24のいずれか一項に記載の遺伝子治療ベクター、請求項25から27のいずれか一項に記載の発現カセット、請求項28に記載のプロモーター、請求項29に記載のビリオン、又は請求項30に記載の医薬組成物。
- - 遺伝子に作動可能に連結された合成肝臓特異的誘導性プロモーターであって、
CARとRXRのヘテロダイマーによって結合及び活性化されることが可能なシス調節エレメント(CRE)を含む、合成肝臓特異的誘導性プロモーター、

- 前記発現カセット内の誘導性プロモーターに作動可能に連結された遺伝子からの発現産物の発現を誘導することができる誘導剤を、前記細胞に投与する工程と、

細胞、好ましくは肝臓細胞において発現産物、好ましくは治療用発現産物を生成する方法。 - 遺伝子からの発現産物の発現に適した条件下で前記肝臓細胞を維持する工程を含む、請求項34に記載の方法。

- 細胞に投与される誘導剤の濃度を経時的に変化させる工程を含む、請求項34から38のいずれか一項に記載の方法。
- 合成肝臓特異的発現カセットを細胞に導入する工程を含む、請求項34から39のいずれか一項に記載の方法。
- 請求項1から24のいずれか一項に記載の遺伝子治療ベクターを肝臓細胞に導入する工程、その後に、
細胞に誘導剤を投与する工程、
を含む、対象の肝臓細胞において治療用導入遺伝子を発現させる方法。 - - 治療用産物をコードする核酸を含む、請求項1から24のいずれか一項に記載の遺伝子治療ベクター、請求項25から27のいずれか一項に記載の発現カセット、請求項29に記載のビリオン、又は請求項30に記載の医薬組成物を対象の肝臓に導入する工程と;
- 治療有効量の治療用産物が対象において発現されるように、対象に誘導剤を投与する工程と、
を含む、それを必要とする対象の、好ましくはヒトの遺伝子治療方法。 - 対象に提供される治療用遺伝子産物の投薬量をモジュレートするために、対象に投与される誘導剤の量を経時的に変化させる工程を含む、請求項42に記載の方法。
- - 対象において発現される治療用産物の量を決定する工程、又は治療用産物に対する対象の応答を評価する工程と:
a)対象においてより高い量の治療用産物が望ましい場合、対象に投与される誘導剤の量を増加する工程、又は
b)対象においてより低い量の治療用産物が望ましい場合、対象に投与される誘導剤の量を減らす工程と、
を含む、請求項42又は43に記載の方法。 - 対象における治療用産物の量を変更するために誘導剤を変える工程を含む、請求項42から44のいずれか一項に記載の方法。
- 本明細書に記載の任意の状態又は疾患の処置のための医薬組成物の製造のための、
請求項1から24のいずれか一項に記載の遺伝子治療ベクター、請求項25から27のいずれか一項に記載の発現カセット、請求項28に記載のプロモーター、請求項29に記載のビリオン、の使用。 - 更なる態様では、本発明は、以下の工程を含む、発現産物を生成するための方法を提供する:
a)遺伝子に作動可能に連結された合成肝臓特異的誘導性プロモーターであって、CARとRXRのヘテロダイマーによって結合及び活性化されることが可能なシス調節エレメント(CRE)を含む合成肝臓特異的誘導性プロモーター
が含まれる発現カセットを含む、真核細胞、好ましくは動物細胞、より好ましくは哺乳動物細胞、より好ましくは肝臓細胞の集団を用意する工程と;
b)前記細胞集団を培養する工程と;
c)前記発現カセット中の誘導性プロモーターに作動可能に連結された遺伝子からの発現産物の発現を誘導することができる誘導剤を、前記細胞集団に投与する工程と;
d)発現産物を回収する工程。
