本発明の多標的融合タンパク質における「免疫グロブリンFcドメイン」は、天然に存在する免疫グロブリンFcドメインの全てのアミノ酸残基を含むか、または天然に存在する免疫グロブリンFcドメインの一部のアミノ酸残基を含む。免疫グロブリンFcドメインは、本発明の多標的融合タンパク質に有利な薬物動態特性を提供し、長い血清半減期を含むが、これに限定されない。さらに、免疫グロブリンFcドメインはまた、例えば、タンパク質Aによって本発明の多標的融合タンパク質のアフィニティークロマトグラフィー、精製を可能にする。
本発明の多標的融合タンパク質における「CD80の細胞外ドメイン(ECD)」は、天然に存在するCD80 ECDの全てのアミノ酸残基を含むか、または天然に存在するCD80 ECDの一部のアミノ酸残基を含む。ある実施形態において、前記CD80 ECDは、CD80 IgVを含み、好ましくはヒトCD80 IgVを含み、より好ましくは、表6における配列番号13または14に示すアミノ酸配列を有する。

本発明の多標的融合タンパク質は、例えば、固体ペプチド合成(例えば、Merrifield固相合成)または組換え生産によって得ることができる。組換え生産のために、前記多標的融合タンパク質の各サブユニットをコードするポリヌクレオチドを分離し、宿主細胞内でさらにクローニングおよび/または発現させるために1つまたは複数のベクターに挿入する。前記ポリヌクレオチドは、従来の方法を用いて容易に分離および配列決定され得る。一実施形態では、本発明の1つ以上のポリヌクレオチドを含むベクター、好ましくは発現ベクターが提供される。
PD-1は、2つのリガンド、それぞれPD-L1およびPD-L2を有する免疫抑制タンパク質である。PD-1とPD-L1との間の相互作用は、例えば、腫瘍浸潤リンパ球の減少及び/又は癌細胞の免疫回避をもたらすことが知られている。PD-1とPD-L1またはPD-L2との局所相互作用を阻害することにより免疫抑制を逆転させることができる。PD-1とPD-L2との相互作用も阻止される場合、この効果は相加的である(IwaiY.ら、Involvement of PD-L1 on tumor cells in the escape from host immune system and tumor immunotherapy by PD-L1 blockade、Proc.Nat’l.Acad.Sci.USA、2002、99:12293-7)。免疫応答調節における免疫チェックポイントPD-1の信号伝達の重要性を考慮して、本発明は、本発明の多標的融合タンパク質と抗PD-1抗体とを含む、併用療法のための医薬組成物を開発している。
本明細書に開示される多標的融合タンパク質および医薬組成物は、癌の治療的および予防的使用を有する。例えば、多標的融合タンパク質、及び抗PD-1抗体とのその組成物は、種々のがん性疾患を治療及び/又は予防するために、インビトロ若しくはエクスビボ培養細胞に、又は被験者、例えばヒト被験者に投与することができる。

(1)目的タンパク質の一過性発現
293F(Invitrogenから入手可能、カタログ番号11625-019)細胞を、無血清CD 293培養液(Invitrogenから入手可能、カタログ番号11913-019)中で懸濁培養した。トランスフェクション前の細胞培養物を遠心分離して細胞ペレットを得、細胞濃度が1×106細胞/mlとなるように新鮮な無血清CD293培養液に細胞を懸濁した。細胞懸濁液を振盪フラスコに入れた。実施例1で調製した目的遺伝子を含む組換え発現ベクタープラスミドの各DNA250ugとポリエチレンイミン(PEI)(Sigma、カタログ番号408727)500ugをそれぞれ、細胞懸濁液100mlを例に、無血清CD293培養液1mlに加えて混合し、室温で8分間静置した後、PEI/DNA懸濁液を、細胞懸濁液100mlを入れた振盪フラスコに滴下した。穏やかに混合し、5%のCO2、37℃の振盪に置いて培養した(120回転/分)。5日後に培養上清を採取した。
上記の実施例2(1)で回収した培養上清中に存在する目的タンパク質を、pH7.4のPBS溶液で平衡化したHiTrap MabSelect SuRe 1mlカラム(GE Healthcare Life Sciences製品、カタログ番号:11-0034-93)を用いて精製した。手短に述べると、HiTrap MabSelect SuRe 1mlカラムを、pH7.4のPBS溶液を用いて0.5ml/分の流速で、10カラム床体積で平衡化した。上記実施例2(1)で回収した培養上清を、0.45μmろ過膜を用いてろ過した後、pH7.4のPBS溶液で平衡化したHiTrap MabSelect SuRe 1mlカラムにローディングし、上清をロードした後、該カラムをまずpH7.4のPBS溶液で流速0.5ml/分で5~10個のカラム床の体積を洗浄し、その後に100mMのクエン酸緩衝液(pH4.0)で流速0.5ml/分で溶出する。溶出ピークを収集し、目的タンパク質BY18.1、BY31.19、BY24.4、BY24.5、BY24.12がそれぞれ溶出ピークに存在する。
組換えヒトCD28(北京義翹神州生物技術有限公司製、カタログ番号:50103-M08H)、組換えヒトPD-L1(北京百普賽斯生物科技有限公司、カタログ番号:PD1-H5229)、組換えヒトCTLA-4(北京義翹神州生物技術有限公司製、カタログ番号:11159-H08H)をそれぞれ0.5μg/ml、0.25μg/ml、1.0μg/mlに希釈し、96ウェルELISAプレート(Corning社、品番:42592)でコーティングした。上記実施例2(2)で精製した融合タンパク質BY24.4、BY24.5、BY24.12、BY31.19を2000μg/mlに希釈し、次に3倍のシリーズ希釈を行い、合計16個の勾配を希釈し、各濃度勾配に対して複穴検出を行った。希釈試料50μLを、上記の組み換えヒトCD28、組み換えヒトCTLA-4、または組み換えヒトPD-L1でコーティングした96ウェルプレートにそれぞれ加え、37℃で2時間インキュベートした。3回洗浄後、西洋ワサビペルオキシダーゼ標識ヤギ抗ヒト二次抗体(北京中衫金橋公司製、品番ZDR-5301)を加え、37℃で1時間インキュベートした。3回洗浄後、3,3’,5,5’-テトラメチルベンジジン(TMB)基質発色液(北京康偉世紀生物科技有限公司、製品番号:CW0050)50μl/ウェルを加えた。10分後、2NのH2SO4を加えて発色を終了した。ELISAリーダーを用いて、450nmで各ウェルの吸光度OD値を決定した。
表面プラズモン共鳴測定は、BIAcore(登録商標)T100装置(GE Healthcare Biosciences AB、スウェーデン)で25℃で行った。
異なる健常ヒトに由来するCD4+Tリンパ球及び樹状細胞(DC)は、北京時合生科学技術株式会社から購入した。CD4+Tリンパ球及び樹状細胞(DC)を、それぞれ、96ウェル細胞培養プレートに1×105細胞/ウェル及び1×104細胞/ウェルで播種した。試験を、それぞれBY18.1群(1.0μg/mL)、BY24.4群(1.14μg/mL)、BY24.4(1.14μg/mL)+BY18.1(1.0μg/mL)、BY24.5(1.14μg/mL)+BY18.1(1.0μg/mL)、BY24.12(0.84μg/mL)、およびBY24.12(0.84μg/mL)+BY18.1(1.0μg/mL)の7群に分けた。各群の3つのウェルに対して、要求に従って、抗体BY18.1、融合タンパク質BY24.4、BY24.5、BY24.12、または抗体BY18.1と各融合タンパク質BY24.4、BY24.5、BY24.12との組合せを、示される最終濃度まで添加する。最終的に10%の牛胎児血清を含む1640培地を最終容量が200μlとなるように添加した。37℃、5%のCO2で培養する。
本実施例の主な目的は、本発明の多標的化融合タンパク質とPD-1抗体とのインビボ抗腫瘍における相乗効果を検討することであり、従って、PD-1抗体及び各融合タンパク質の用量は、いずれも低用量であり、高用量では腫瘍抑制効果が良好でPD-1抗体及び各融合タンパク質相互間の相乗効果は見られない可能性がある。
Claims (14)
- 血管内皮細胞の増殖をブロックし、T細胞を活性化する多標的融合タンパク質であって、血管内皮増殖阻害剤ドメイン(i)、免疫グロブリンFcドメイン(ii)、およびCD80細胞外ドメイン(ECD)(iii)を含む、多標的融合タンパク質。
- 前記(i)は、抗VEGF抗体及び/又は抗VEGFR抗体から誘導される抗原結合断片及び/又はVEGFR細胞外受容体機能領域を含み、
好ましくは、前記抗VEGF抗体及び/又は抗VEGFR抗体の抗原結合断片は、抗VEGF抗体及び/又は抗VEGFR抗体のFab、Fab’、F(ab’)2、Fv、単鎖Fvであり、より好ましくは、前記抗VEGF抗体及び/又は抗VEGFR抗体の抗原結合断片は、配列番号1/2、3/4及び5/6から選択される、ペアとなる重鎖可変領域配列/軽鎖可変領域配列に含まれる重鎖CDR及び軽鎖CDRの6個全て、又は前記6個全ての重鎖CDR及び軽鎖CDRの1個以上と1、2、3、4、又は5個のアミノ酸変化(例えば、アミノ酸置換又は欠失)を有する配列を含み、さらに好ましくは、前記抗VEGF抗体及び/又は抗VEGFR抗体の抗原結合断片は、配列番号1/2、3/4及び5/6から選択される、ペアとなる重鎖可変領域配列/軽鎖可変領域配列、又は前記ペアとなる重鎖可変領域配列/軽鎖可変領域配列と少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又はそれ以上の配列同一性を有する配列を含み、最も好ましくは、前記抗VEGF抗体及び/又は抗VEGFR抗体の抗原結合断片は、Bevacizumab、Ranibizumab、又はRamucirumabのFabであり、
好ましくは、VEGFR細胞外受容体機能領域は、VEGFR1の免疫グロブリン様ドメイン2およびVEGFR2の免疫グロブリン様ドメイン3を含み、または、前記VEGFR細胞外受容体機能領域は、VEGFR1の免疫グロブリン様ドメイン2、VEGFR2の免疫グロブリン様ドメイン3およびVEGFR2の免疫グロブリン様ドメイン4を含み、または、前記VEGFR細胞外受容体機能領域は、VEGFR1の免疫グロブリン様ドメイン2を含み、より好ましくは、前記VEGFR細胞外受容体機能領域は、配列番号7~9に示すアミノ酸配列から選択されるもの、または配列番号7~9に示すアミノ酸配列と少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%またはそれ以上の同一性を有する任意のアミノ酸配列を有する、請求項1に記載の多標的融合タンパク質。 - 前記(ii)は、ヒト免疫グロブリンFcドメインであり、好ましくは、前記(ii)は、ヒトIgG1、IgG2、IgG3、またはIgG4のFcドメインであり、より好ましくは、前記(ii)は、配列番号10、11または12に示すアミノ酸配列のFcドメインを含み、または配列番号10、11または12に示すアミノ酸配列と少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%またはそれ以上の配列同一性を有するFcドメインを含む、請求項1または2に記載の多標的融合タンパク質。
- 前記(iii)は、ヒトCD80 ECDを含み、好ましくは、前記(iii)は、ヒトCD80 IgVまたはヒトCD80 IgVIgCを含み、より好ましくは、前記(iii)は、配列番号13または14に示すアミノ酸配列、または配列番号13または14に示すアミノ酸配列と少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、またはそれ以上の同一性を有するアミノ酸配列を有する、請求項1~3のいずれか一項に記載の多標的融合タンパク質。
- 前記(i)、(ii)および/または(iii)の間のペプチドリンカーをさらに含み、好ましくは、前記ペプチドリンカーは、1つ以上のアミノ酸を含み、より好ましくは、少なくとも5つのアミノ酸を含み、最も好ましくは、配列番号20~46から選択されるペプチドリンカーを含む、請求項1~4のいずれか一項に記載の多標的融合タンパク質。
- 前記融合タンパク質は、N末端からC末端に向かって、(i)、(ii)、および(iii)の順、(iii)、(i)および(ii)の順、または(iii)、(ii)および(i)の順で有効に連結される、請求項1~5のいずれか一項に記載の多標的融合タンパク質。
- (a)完全長抗VEGF抗体、完全長抗VEGFR抗体、又は完全長抗VEGF及びVEGFR二重特異性抗体、および前記抗体の2本の重鎖のそれぞれのC末端において有効に連結されたCD80 ECD、
(b)完全長抗VEGF抗体、完全長抗VEGFR抗体、又は完全長抗VEGF及びVEGFR二重特異性抗体、前記抗体の2本の重鎖のそれぞれのN末端において有効に連結された1つのCD80 ECD、および前記抗体の2つの軽鎖のそれぞれのN末端において有効に連結された1つのCD80 ECD、
(c)CD80 ECD、CD80 ECDのC末端において有効に連結された二量体形態の免疫グロブリンFcドメイン、及び前記二量体形態の免疫グロブリンFcドメインのC末端において有効に連結された、抗VEGF抗体及び/又は抗VEGFR抗体から誘導される抗原結合断片、
又は
(d)CD80 ECD、CD80 ECDのC末端において有効に連結された二量体形態の免疫グロブリンFcドメイン、および前記二量体形態の免疫グロブリンFcドメインのC末端において有効に連結されたVEGFR細胞外受容体機能領域を含み、
好ましくは、前記抗体は、IgGクラスの抗体、特にIgG1サブクラス、IgG2サブクラス、IgG4サブクラスの抗体であり、より特にIgG4サブクラスの抗体であり、さらに好ましくは、前記IgG4サブクラスの抗体は、Fcドメイン中のS228位にアミノ酸による置換を含み、より好ましくはアミノ酸によるS228P置換であり、さらに好ましくは、前記抗体の軽鎖型は、それぞれκ型またはλ型であり、好ましくはκ型であり、
好ましくは、前記全長抗VEGF抗体は、Bevacizumabであり、前記全長抗VEGFR抗体は、Ramucirumabである請求項6に記載の多標的融合タンパク質。 - 配列番号80の融合タンパク質の第1のサブユニットおよび配列番号82の融合タンパク質の第2のサブユニットを含む融合タンパク質(1)と、
配列番号84の融合タンパク質の第1のサブユニットおよび配列番号86の融合タンパク質の第2のサブユニットを含む融合タンパク質(2)と、
配列番号88の融合タンパク質サブユニットを含む融合タンパク質(3)とからなる群から選択される、請求項1~7のいずれか一項に記載の多標的融合タンパク質。 - 請求項1~8のいずれか一項に記載の多標的融合タンパク質をコードするポリヌクレオチド。
- 請求項9に記載のポリヌクレオチドを含むベクターであって、好ましくは発現ベクターであり、最も好ましくは二重発現カセットを有するグルタミンシンテターゼ発現ベクターである、ベクター。
- 請求項9に記載のポリヌクレオチドまたは請求項10に記載のベクターを含む宿主細胞であって、好ましくはCHO、HEK293またはNSO細胞である、宿主細胞。
- 請求項1~8のいずれか一項に記載の多標的融合タンパク質を生成するための方法であって、前記多標的融合タンパク質の発現に適した条件下で、請求項11に記載の宿主細胞を培養するステップ(i)と、前記融合タンパク質を回収するステップ(ii)とを含む、方法。
- 請求項1~8のいずれか一項に記載の多標的融合タンパク質と、抗PD-1抗体とを含み、好ましくは前記抗PD-1抗体が、配列番号47/48、49/50、51/52、53/54、55/56、57/58、59/60、61/62、63/64、65/66、67/68、69/70および71/72からなる群より選択される、ペアとなる重鎖可変領域配列/軽鎖可変領域配列に含まれる重鎖CDR及び軽鎖CDRの6個全て、又は前記6個全ての重鎖CDR及び軽鎖CDRの1個以上と1、2、3、4、又は5個のアミノ酸変化(例えば、アミノ酸置換又は欠失)を有する配列を含み、さらに好ましくは前記抗PD-1抗体が、配列番号47/48、49/50、51/52、53/54、55/56、57/58、59/60、61/62、63/64、65/66、67/68、69/70および71/72からなる群より選択される、ペアとなる重鎖可変領域配列/軽鎖可変領域配列、又は前記ペアとなる重鎖可変領域配列/軽鎖可変領域配列と少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%又はそれ以上の配列同一性を有する配列を含み、最も好ましくは前記抗PD-1抗体が、ニボルマブ、ピズマブ、およびツムマブ(Pembrolzumab)からなる群より選択される、医薬組成物。
- 個体における癌性疾患(例えば、固形腫瘍および軟組織腫瘍)の治療または予防のための医薬の製造に使用され、好ましくは、癌性疾患が、黒色腫、乳癌、結腸癌、食道癌、消化管間質性腫瘍(GIST)、腎臓癌(例えば、腎細胞癌)、肝臓癌、非小細胞肺癌(NSCLC)、卵巣癌、膵臓癌、前立腺癌、頭頸部腫瘍、胃癌、血液悪性疾患(例えば、リンパ腫)であり、特に、前記疾患は、結腸癌またはトリプルネガティブ乳癌であり、好ましくは、前記個体は哺乳動物であり、より好ましくはヒトである、請求項1~8のいずれか一項に記載の多標的融合タンパク質、請求項13に記載の医薬組成物の使用。
