三次元(3D)プリントは、組織工学用のスキャホールドおよびデバイスを製造する一般的な技術となりつつある。これは、患者特異的な設計、高い構造的複雑さ、および低コストでの迅速なオンデマンド製造を提供する3Dプリントの能力による。バイオ製造業における3Dプリントの広範囲の普及を制限する大きなボトルネックの1つは、「バイオ素材インク」の多様性の欠如である。
本開示は、例えば約10 x Yインチ、5 x Yインチ、2 x Yインチ、1 x Yインチ、0.5 x Yインチ、0.25 x Yインチまたは0.1 x Yインチであり、Yが0.1~1インチ、0.5~5インチ、0.2~1インチ、1~5インチまたは1~10インチであり得る寸法を有するその部分を含む、哺乳類の器官、例えばブタまたはヒトの器官、例えば肝臓、膵臓、腎臓、肺、脾臓または心臓由来の脱細胞化細胞外マトリクスを、熱の適用なしで脱水する方法を提供し、例えば、脱水は、約75°F未満、約70°F未満、約68°F未満、約25℃未満、約22℃未満または約19℃未満の温度で行われる。1つの態様において、当該部分は、脱水前に圧縮され得る。例えば、哺乳類器官由来の脱細胞化細胞外マトリクスの部分の厚みは、少なくとも0.01%、0.5%、1%、5%、10%、20%、50%、90%またはそれ以上圧縮され得る。1つの態様において、脱水された部分は、脱細胞化器官部分から水分を除去する周囲温度での脱水および任意の圧縮後に、元の器官の組成の大部分を維持している。脱水物は、その後、ミーリング(milling)、例えば凍結ミーリングに供され、ミーリングまたはミーリングおよび例えばふるいもしくは他のサイズ分離デバイスもしくは方法を用いるサイジング(sizing)を通じて獲得され得る、約0.01 mm~約0.05 mm、約0.05 mm~約0.1 mm、0.1 mm~約5 mm、約0.25 mm~約0.5 mm、約0.4 mm~約4 mm、約1 mm~約2 mm、約0.5 mm~約1.5 mm、約1.5 mm~約3 mmまたは約3 mm~約4 mmを含む、約0.001~0.005 mmから最大約10 mmの範囲のサイズの粒子の集団が提供され得る。全器官の細胞外マトリクスの元の組成は、脱水およびミーリングされた細胞外マトリクス粒子において、治療を可能にするまたは器官の機能を補助する3D構造のプリントに利用する器官特異的なインクを調製するための構造を提供する。インクは、溶媒、例えば水性溶媒、例えば水もしくはリン酸緩衝生理食塩水(PBS)中で組み合わせる前に脱水された粒子をサイジングすることによりまたは脱水された粒子を化学的もしくは酵素的消化に供することにより調製され得る。インクのバイオプリントは、サーマルインクジェットバイオプリント、ピエゾインクジェットバイオプリント、空気圧押し出しバイオプリント、機械押し出しバイオプリントまたはレーザー支援バイオプリントを含むがこれらに限定されない任意の方法を通じて行われ得る。したがって、粒子は、エクスビボ細胞アッセイを含むがこれらに限定されない方法またはインビボ用の組織工学構築物、例えばインビボ再モデリングのために移植されるもしくは機能的なインプラントを提供するよう細胞と組み合わされる灌流脱細胞化ECMに基づくプリントされた構造物の調製において有用な組成物で使用され得る。
本開示は、脱細胞化全器官物、例えば、灌流脱細胞化全器官物の脱水により調製される脱細胞化粒子生産物を提供する。多くの脱水法は、液体の除去を達成するために高温を用いる。本発明の方法は、脱細胞化物の選択またはサイジング、任意の圧縮、その後の、定義された時間の、例えばラミナーフローフード内での、スペーシング(spacing)を用いる。1つの態様において、脱水は、生産物中の含水量を、10%未満、例えば約9%、8%、7%、または5%未満にする。1つの態様において、圧縮および脱水は、生産物中の含水量を8%未満、例えば約7%、6%、5%または4%未満にする。脱水プロセスの周囲温度は、あらゆる細胞外マトリクス成分に対して非破壊的であり、それによってより高い効能を有する可能性のある生産物を生成する。この物質は、その後、3Dプリントを含むがこれらに限定されない多くの態様において微粒子として使用され得る微粒子粉末に凍結ミーリングされ得る、またはプロテアーゼ消化を含むがこれに限定されない、温度、化学的架橋、UV架橋もしくは他の手段よる再編成を通じてプリントされ得る溶液への消化により、ゲル形成のための液体に溶解され得、3Dプリントのインクとして使用するための液体に溶解され得る。この粒子はまた、直接的な治療能、例えば、創傷への直接適用、空隙部の充填、筋内注射、トンネル状の創傷および瘻孔への適用を有し得る。
1つの態様において、哺乳類器官または組織を脱細胞化する方法は、その器官または組織へのカニューレ挿入を含む。器官または組織の脈管、導管および/または空隙部は、当技術分野で公知の方法および材料を用いてカニューレ挿入され得る。器官または組織を脱細胞化する上での次の工程は、細胞破壊媒体を用いてカニューレ挿入された器官または組織を灌流することである。器官を通じた灌流は、多方向(例えば、順行および逆行)であり得る。
脱水の前に、脱細胞化物は、様々なバッチで処理され、プールするおよび後日まとめて処理する目的で保管され得る。1つの態様において、当該方法は、穏やかな攪拌下での約5分間の、pH中性滅菌剤、例えば、500 ppm過酢酸(PAA)浴(5~10L)中での圧縮された脱細胞化全器官物のインキュベートを含む。乳酸、アルコール、過酸化水素、次亜塩素酸塩、二酸化炭素、ジクロロイソシアヌル酸ナトリウム、クロラミン-T等を含むがこれらに限定されない他の滅菌剤も使用され得る。1つの態様において、この後に、穏やかな攪拌下での5分間の脱イオン水浴(5~10L)に供される。滅菌および洗浄の後、物質は、その物質が乾燥しないよう、脱イオン水または他の生理学的に適合する溶液を用いて無菌的に包装され得る。包装物は、その後、4℃で保管される。数ロットの物質を蓄積した後、その物質はプールされ、他のクラスの製品を製造するために使用され得る。
脱水およびサイジングにより生成された粒子は、組織工学用のデバイスおよびスキャホールドを製造するために、押し出しベースの3Dプリント法、例えば、熱溶解積層モデリング(FDM)およびダイレクトインクライティング(DIW)において使用され得る。これらの方法において、インクは、レイヤー毎に3Dオブジェクトを構築するコンピュータモデルにより決定された事前に規定されたパスにしたがい粘性液としてノズルを通じて射出される。溶媒流延ダイレクトライティングインクは、押し出し後に構造を維持するヒドロゲルを含む。
インクジェットプリントは、固化後に構造物を形成する目的で、非常に小さな容積(1~100ピコリットル)の個々の液滴をノズルからプリント表面に付着させることを可能にする。このプリントプロセスを加速させるよう、数百の個別のノズルを含む多ノズルインクジェットプリントヘッドが開発されている。インクジェットプリンターは、液滴の生成機構に基づき、連続式インクジェット(CIJ)プリントおよびドロップオンデマンド(DOD)インクジェットプリントの2つのグループに大別される。CIJプリントにおいては、連続する液滴(直径約100μm)の流れが生成され、未使用のインクが再利用される。DODインクジェットプリントにおいては、(直径25~50μmの範囲の)個々の液滴が必要なときに生成される。組織工学用途においては、DOD型プリンターが一般的に使用されている。インクジェットプリントの能力は、空間分解能、例えば、位置的に高精度(x-y軸において約10μm)なECM粒子を含むピコリットルの液滴の配置である。インクの最も重要な特性は、粘性および表面張力である。インクの粘性は、適切に、1 x 105~1 x 106-1の間の高せん断速度の下で通常10 cP(mPa s)を下回る程度に低くすべきである。表面張力は、ノズルから射出される液滴の形状および基材上での液滴の形状を決定する。インクの表面張力値は、一般に、28~350 mN m-1の範囲である。プリントされるオブジェクトの分解能および精度は、隣接する液滴間(癒着)および個々の液滴と基材の間(例えば、表面張力およびぬれ性)の相互作用により決定される。液体から固体への相転移(すなわち、溶媒蒸発、温度制御転移、または前駆体溶液のゲル化)は、プリントされるオブジェクトの最終的な形状およびサイズを制御する。
レーザー支援3Dバイオプリント(LAB)は、例えば本明細書に記載されるECM粒子を含むバイオインクを含む「リボン」を通じてレーザーパルスを送る非接触でノズルを使わないプリントプロセスである。リボンは、エネルギーを吸収しその後にエネルギーをリボンに伝達することができるチタンまたは金の層によって支持される。バイオインクおよび任意で細胞は、リボンの底部で懸濁され、レーザーパルスによって蒸散されたときに、高圧のバブルを生成し、これが最終的に、リボンのちょうど向こう側に位置する受取側基材に個々の液滴を推進する。この工程は、3D構造物を機能的に作製するよう繰り返し行われる。LABは、プリント後の表現型および細胞生存性の高い保持力が実証されている。
光造形(SLA)バイオプリント法は、UV光またはレーザーが光重合可能な液体ポリマーの経路にパターン状に送られ、それによってそのポリマーを架橋し硬化した層にするプロセスである光重合を利用する。各層が重合されると、プリントプラットフォームは、ポリマー溶液中にさらに降下し、それによって3D構造物を形成するための複数回のサイクルが可能となる。
ヒドロゲルは、多量の水分を保持する能力を有する三次元ポリマーネットワークであり、調整可能な機構、分解性および官能化性を有する微小環境を提供する。ヒドロゲルインクは、それらが細胞および/または生化学分子、例えばECM成分を含む場合、バイオインクと称され得る。インクジェット、光支援および押し出しベースの3Dプリントシステムが、ヒドロゲルプリントの最も一般的な方法である。ヒドロゲルインクを設計する古典的アプローチは、プリント後直ちにネットワークを形成するポリマー溶液を調製することである。このネットワークは、外部刺激、例えば温度、光またはイオン濃度に応じて物理的または化学的に架橋され得る。物理架橋ヒドロゲルの主な利点は、その物質の毒性を低下させる化学的薬剤の非存在である。他方、化学架橋ヒドロゲルは、共有結合の形成を通じて調製され、得られるヒドロゲルは、機械的な力に対してより高い耐性を有するが、それは通常、物理的に架橋されたネットワークよりも大きな容積変化を引き起こす。

乾燥およびブレンドした灌流脱細胞化肝臓サンプルを、ラミナーフローフードにおいてサイジングした。脱細胞化肝臓ECM(MMまたはMD)の異なるサイズの細片、例えば0.5インチ x 0.5インチまたは1インチ x 1インチの細片(約15グラム)を、ポリカーボネート製グラインド用バイアルに入れた。バイアルの内容物を、SPEX Cryogrinderにおけるグラインドに供した。2つの異なるプロトコルを使用した。1つは、4グラインドフェーズ、10 CPS、5分予備冷却、2分冷却を含むものとした。他方のプロトコルは、2グラインドフェーズ、10 CPS、5分予備冷却、2分冷却を含むものとした。両方の生成物を、粒子サイズ分析のために2 mmふるいにかけた。4グラインドフェーズの生成物は、144 mgの>2.0 mm(1.1%)および13,051 mgの<2.0 mm(98.9%)を有した。2グラインドフェーズの生成物は、233 mgの>2.0 mm(2.2%)および10,228 mgの<2.0 mm(97.8%)を有した。各生成物(約1500 mg)を、非滅菌5 mL琥珀ガラスバイアルで包装し、ブチルストッパおよびアルミニウムシールにより封止した。含水量分析を行った(70℃)。表1は、各プロトコルおよび元ソース(MMまたはMD、両方とも肝臓由来灌流脱細胞化ECM)についての2つの異なるサンプルのデータを示している。
図1のデータは、実施例1の7つのサンプルタイプの圧縮および脱水が、ECMから高分子量コラーゲン(例えば、コラーゲン > 150 kDa)を除去しないことを示している。図2は、実施例1の7つのサンプルタイプについての粒子サイズ分析を示している。
