I.X細胞の検出
具体的な実施の形態では、単セルPBMC吊り下げを以下のように準備することができる:
1.円錐管にPBSを入れて、少なくとも1:1まで希釈した血液試料を採取する。
2.希釈した試料の下に、元の試料体積と同じ量のFicollを置く。
3.制動OFFで室温で400 x g で20分間セントリフュージします。
4.PBSとFicoll層の境目にあるPBMCを新鮮なチューブに収穫する。
5.チューブにPBSを充填して電池を洗浄する。
6.300-400 x gで、2-8℃で4-5分間、電池を中心に分岐させます。上澄液を捨てる。
7.セルペレットを適切な量のフローキトメトリー貯留緩衝液または選択の緩衝液に再汚染し、細胞数および生存性分析を実施する。
8.Step 4のように中心核分裂電池は、最終電池濃度が1×107電池/mlになるように、適切なフローキトメトリー貯留バッファまたは緩衝液を適当な量で再費やします。
別の実施形態では、X電池を検出し、以下のように表現することができる:
1.セルサスペンション(106 セル)の100-uLアリコットをチューブに配布する。

更なる実施形態において、本発明は、島に反応するT細胞を刺激し検出するためのx-mAbの使用方法を提供する。x-mAbは、24ウェルプレートでCD4 T細胞を刺激するために使用することができる。x‐mAbをPBMCsに加え、表面抗原CD69のアップレギュレーションとCFSE希釈法で測定した拡散を測定してT細胞活性化を検討した。
本発明はまた、x-mAbを有する個々のT1Dの危険を決定するためのHLA-DQのジェノタイピング法を提供する。特定の実施の形態では、x-mAbは、ベータ鎖の57の位置のベータ鎖中の単一のアミノ酸に傾向があるHLA-DQ8とは異なるHLA-アレルを持っている個々に存在するので、PCRプローブを用いて危険にさらされている。DQ7を保有する個体は、T1Dと関連せず、この位置にアスパラギン酸を有するが、素因となるDQ8対立遺伝子を保有する個体は、この位置に非アスパラギン酸を有する。したがって、T1Dのリスクのスクリーニングには、x-mAb陽性の個人における遺伝子型HLA分子が関与するであろう。

RT‐PCRについては、製造者の指示に従い、RNeasy blood mini kit (Quigen)を用いて新鮮なPBMCからのRNを抽出し、次に濃度と純度のためのNanoDrop(ND‐1000分光光光度計)測定を行った。キットプロトコルに従い、RevertAid First Strand cDNA Synthesis Kit (Thermofisher)を用いて、精製された約1mmugの純化したRN上で逆転転写(RT) PCRを行い、cDNAを調製した。簡単に述べると、RNAを5X反応混合物、ランダムヘキサマープライマー、およびRevertAid M-MuLV RT (200 U/μL)酵素混合物と共に、最終容量20μL中、25oCで10分間インキュベートし、続いて42oCで60分間インキュベートし、70o Cで5分間不活化した。正(GAPDH比プライマー)と負(RT分子のない混合反応)反応を用いて、cDNA合成工程の結果を検証した。

VNARは新しい抗原レセプター(IgNAR)の可変領域である。IgNARは共有結合した重鎖ホモ二量体としてサメの血清中に存在する。それは、一定の領域の数が異なる可変領域(VNAR)から成る可溶性および受容体結合型として存在する。VNAR は CDRl と CDR3 で構成され、CDR2 の代わりに HV2 と HV4 ドメインがあります (例: Barelle とPorter, Antibodies, 4:240-258 (2015)) を参照してください)。本開示は、x-Idを特に結合するVNARを包含する。
この開示の反物は、全抗性又は単鎖Fc (scFc)であり、また、当該技術分野で知られている任意の定常領域を含むことができる。軽鎖定常領域は、例えば、カッパまたはラムダタイプの軽鎖定常領域、例えば、人間のカッパまたは人間のラムダ軽鎖定常領域であることができる。鎖定常領域は、例えば、アルファ-、デルタ-、エプシロン-、ガンマ-、又はムー型鎖定常領域、例えば、人間のアルファ-、人間のデルタ-、人間のエプシロン-、人間のガンマ-、又は人間のムー型鎖定常領域であることができる。特定の例では、抗x-Id抗体は、IgA抗体、IgD抗体、IgE抗体、IgGl抗体、IgG2抗体、IgG3抗体、IgG4抗体、またはIgM抗体である。
ある態様において、本開示の抗x-Id抗体は、二重特異性抗体である。二特異的抗体は、少なくとも2つの異なるエピトープに対する結合特異性を有する抗体である。典型的な二種特有の抗生物質は、x-Idタンパク質の二つの異なったエピトープに結合することがある。他のこのような抗物質は、x-Id結合部位と他のタンパク質の結合部位を結合することができる。二種性抗毒素は、それらの全長または低分子型(例えば、F(ab')2種の二種性抗毒素、sc(Fv)2種の二種性抗毒素、二体性抗毒素)として調製することができる。
本明細書で開示される抗毒素又は抗原結合フラグメントは、高分子(例えば、ポリエチレングリコール(PEG)、PEG (PEI-PEG)に改造されたポリエチレンミン(PEI)、ポリグルタミン酸(PGA) (N-(2-Hydroxypropyl)メタクリラミド(HPMA)コポリマー)、ヒト上記アルバミン又はその断片、放射性物質(例えば、90Y、131I)、蛍光物質、発光物質、ハプテン、エンザイム、メタルチレート、及び薬物を含む様々な高分子に結合することができる。
ここに記載されている抗生物質のx-Id結合特性は、任意の標準的な方法、例えば、OCTET(R)、Surface Plasmon Resonance (SPR)、BIACORETM分析、Enzyme Linked Immunosorbent Assay (ELISA)、EIA (エンザイム・イムノアッセイ)、RIA (ラジオイムアッセイ)、およびFluorescence Resonance Energy Transfer (FRET)によって測定することができる。
一実施の形態では、抗x-Id抗原またはそれの抗原結合フラグメントが、例えばミューテージェネシスによって、修正された抗物質のプールを提供するように修正される。その後、改良された抗生物質を評価し、1つ以上の機能特性(例えば、拘束力の改善、安定性の改善、抗原性の低下、生体内の安定性の増加)を有する抗生物質を同定する。1つの実装では、ディスプレイライブラリ技術を用いて、改良された抗生物質のプールを選択または画面する。次に、たとえば、より厳密であるか、より競合的な結合および洗浄条件を用いることによって、第2の図書館からより高い親和性が確認される。他のスクリーニング技術も使用できます。親和性成熟を効果的にする方法には、ランダムミューテージソン(例えば、福田ら、Nucleic Acids Res, 34:el27 (2006);標的化ミューテージソン(例、Rajpalら、Proc. E.x-id アンチボディの作成方法
本発明は、xクローノタイプのCDR3アミノ酸の代替物である: CARQEDTAMVYYFDYW (SEQ ID NO:1)が、1型糖尿病及び他の自己感染症を治療するとともに、T1Dを開発するための危険にさらされている個人を特定することを意図している。ある実施形態では、CDR3置換物は、MHCクラスII分子またはTCRに対するx-クローン型または超可変領域(すなわち、CDR3)ベアリングx-クローン型もしくは関連配列を有する抗体の結合または相互作用を増加させることによって、x-クローン型の抗原活性を正に調節する置換誘導体を含むことができる。これらの誘導体は、変調器またはワクチンモダリティとして使用することができる。これらの置換には、例えば、x-クローノタイプの異なる配列位置におけるアラニンスキャン誘導体、ならびにx-クローノタイプの異なる配列位置における非保存的置換が含まれる。他の実施形態において、CDR3の置換物は、MHCクラスII分子またはTCRとの超可変領域(CDR3)におけるxクローノタイプを含むxクローノタイプまたはxクローノタイプの結合または相互作用を減少または廃止することにより、xクローノタイプの抗原活性を否定的に変調する代替誘導体を含む。これらの置換には、例えば、x-クローノタイプの異なる配列位置におけるアラニンスキャン誘導体、ならびにx-クローノタイプの異なる配列位置における非保存的置換が含まれる。本発明はまた、ここに記載されている抗生物質のアミノ酸置換を熟考する。

それをエンコードしている抗毒剤又は抗原結合フラグメント、又はそれと同一の核酸は、例えば、それを必要としている被検者、例えば、人間又は動物の被検者に、様々な方法で管理することができる。多くの用途のために、投与経路は、静脈内注射または非経口、注入(IV)、皮下注射(SC)、腹腔内(IP)、または筋肉内注射、腫瘍内(IT)のいずれかである。他の親権行政形態も利用できる。そのようなモードの実施例は:動脈内、くも膜下、嚢内、眼窩内、心臓内、皮内、経気管、皮下、関節内、関節下、くも膜下、脊髄内、及び硬膜外及び盗血内注射を含む。
H.治療用機器・キット
(実施例1)1型糖尿病患者由来のユニークな二重受容体発現リンパ球に存在する公衆BCRは強力なT細胞自己抗原を符号化する
人間の被検者
末梢血サンプルは、Johns Hopkins Institutional Review Boardにより承認されたプロトコルを用いてドナーから入手した。すべてのドナーは、文書によるインフォームド・コンセントを提供した。T1D被検者はいずれも米国糖尿病学会の分類基準を満たしており、ジョンズホプキンス総合糖尿病センターで募集した。T1Dのないドナーは健常対照(HC)に分類され、正常ボランティアから募集された。ドナーの臨床的特徴を表1Aに要約する(データは示していない)。調査はヘルシンキ原理の宣言に従って行われた。Ficoll‐paque密度遠心分離(GE Healthcare)グラデーションを用いて、末梢血単核細胞(PBMCs)を新たに分離した。レパートリーをハイスループットで分析した被検者の膵島自己抗体プロフィールおよびHLA遺伝子型(表1Bおよび1C、図示せず)を、デンバーのバーバラ・デービス中心自己抗体/HLAコアラボで、確立された方法を用いて実施した。
LSRII多色フローサイトメーター(BDバイオサイエンス)を用いて細胞型を分析した。簡単に言うと、確立された方法(Dai et al., 2015; Martina et al., 2015)を用いて、示されたフッ素結合抗物質(主要資源表(示されていない))の所定の最適な濃度で、単セル吊りが氷上で20分間表面汚れた。獲得した試料(5 x 105 ~ 1 x 106 のライブイベント)は、単一の色の汚染で適切に補償されました。FlowJoソフトウェア(TreeStar)を用いてデータ解析、ゲーティング、及びグラフィック表示を行った。FSC-Height vs FSC-Width and SSC-Height vs SSC-Width plotsを用いた分析からダブルセットを除外した。複数の特殊性のコントロールが用いられた。これらには、CD5、CD19、TCR、IgD、ダンプゲート、および同型制御用のヒトFcRブロッキング試薬(Miltenyi Biotec)、蛍光マイナスワン(FMO)が含まれた。また、適切な場合には、不適切な細胞タイプが内部生体情報制御として使用され、in vitro刺激の場合には、我々は、ネガティブコントロールとして、計量されていない文化を使用した。
新しく分離されたPBMCはFITC結合反TCRαβ、ポリエチレン結合反IgD、APC結合反CD5、およびBV421結合反CD19で染められ、INSPIREソフトウェアを用いて低流速/高感度のImage Streamフローサイトメーター(アムニス社)でX60倍率で分析された。試料ごとに10,000件のイベントが獲得された。補償マトリックスの作成、各蛍光色素に対する最適レーザ出力の設定およびカメラの飽和を避けるために、単一のカラーコントロールを使用した。スペクトルクロストークを修正するために、すべてのファイルに補償マトリクスを適用した。正のカットオフ値はTCRαβの明るいディテール類似度(BDS)背景と不適切な信号(例えば側散乱)に基づいて計算されました。画像解析は、セルバウンダリーを設定し、内部化イベントのゲートを設定するために明るいフィールド画像を使用して、IDEAS 6.2ソフトウェアパッケージで実行された。補償データファイルは、傾きRMSと単一イベントと一致し、破片や多層セルラー事象のないアスペクト比(ダブレット)に基づいて、フォーカスセルを選択するゲーティング戦略を用いて分析された。TセルとBセルシングレットは、この方策を用いて、視野内での良い品質、フォーカスシングレットの選択により、最終ゲートの精緻化を可能にした。Tcon及びBcon電池のゲート後、表面プロフィール(CD19+CD5+TCR+IgD+)に基づき、個々のIgD+DE電池を識別し、指摘マーカーについて分析した。明視野イメージは、LEDベースの明視野照明装置で収集した。各プロットを手動で調整し、取得開始時に発生したマシンノイズをゼロに設定しました。
FACS分類単セル(分類方策については図12Aおよび12Bを参照)は、以下の修正を加えてSmart-seq2プロトコル(Picelliら、2014)で処理された。RNAClean XPビード(Beckman Coulter)を用いた逆転写の前にRNA精製を行った。cDNAは21 PCRサイクルで増幅され、その後AMPure XPビードでDNAクリーンアップされた。ライブラリーは、カスタムバーコードアダプターを使用して、Nextera XT Library Prep kit (Illumina) を使用して作成しました。バーコード化されたユニークなライブラリーは、NextSeq 500 シーケンサ(Illumina)で一緒に配列されました。
Anti-CD3/CD28ビーズ(106ビード/ウェル)の有無にかかわらず、新しく分離したPBMCを24ウェル組織培養プレートの井戸(1ml完全栽培メディアに106電池)の上に置き、37oCおよび5% CO2でインキュベートした。また、Anti-CD3(10mmug/ml)とAnti-CD28(10mmug/ml)をコーティングしたプレートを使用した(Yoneshiro et al., 2017)。7日間の培養後、生存細胞を収穫し、トリパンブルーを用いて数え、BD LSRIIフローサイトメーターを用いて、指摘分子の表現を分析した。絶対細胞数は、示されたサブセットの周波数に生存細胞数を掛け合わせて決定した。
新しく分離されたPBMCは、汚れに影響を密度血清を除去するために、温(37o C)1x PBSで2回洗浄され、電池は1.5~2.0×106電池/mlで温(37C o)1x PBSで再消費された。細胞には37℃で1~2分間、連続渦巻きにより1mm CFSE (eBioscience)とラベル付けした。標識反応は冷えた完全な文化媒体を加えることで消した。CFSE標識細胞は、1x PBSで洗浄され、完全なメディアで再消費され、24ウェル組織培養プレート(1.5~2.0×106セル/ウェル、1ミリリットルの完全なカルチャーメディア)にめっきされた。HLA‐DQ8分子の機能性を評価するために、24ウェルプレート(10μM)のウェルに、HC#1由来のIgD+DE(hIdと呼ばれる)由来の標識ペプチド(x‐Id、TP‐Id、mimotope、天然インシュリンおよびCDR3ペプチド)を担持したDQ8分子を固定化し、PBMCの中からCFSE標識CD4 T細胞を刺激するそれらの能力を調べた。並行実験で、マウス防HLA‐DQ(SPV‐L3; Abcam)および防HLA‐DR(L243; Abcam)の存在下(20 uM)で培養を活性化し、MHC制限を評価した。同様に、CFSE標識細胞もまた、可溶抗原として上述のペプチド(10mm)の有無で刺激された。別の実験では、mAb固有の拡散反応を評価するために、2.5および5ugの純化されたAbmRおよびmAbN (方法で後述)濃度を24ウェルプレートの井戸に固定し、CFSEラベルのPBMCを刺激するために用いた。刺激なしおよびCD3-28刺激ありのCFSE標識細胞を、それぞれ比陰性および陽性対照としてとった。7日間のインキュベーションの後、CFSElow CD4 T細胞の周波数を決定することによって評価された、図のレジェンドと拡散に示されるように、文化は染められた。
Golgi-Plug (Saxena et al., 2017; Xiao et al., 2011)の存在の下で、フォルボル12-ミリスチン酸13-酢酸(PMA) (50ng/ml)およびイオノマイシン(500ng/ml)を用いて、5% CO2で37oCで4時間刺激した。細胞内サイトカイン分析はメーカーの指示を用いて行った。簡単に言うと、表面に汚れた試料を固定し、mAbsを用いて指示された細胞内サイトキンに対して30分間浸透させ、洗浄し、獲得し、上述の方策を用いて分析した。
RPMI‐1640媒体の新しく分離されたPBMC(1X106)を刺激の30分前にCO2インキュベーターで37oCで休息させた。BCR信号化は、記載された時点のCO2インキュベーターの37oCで、BCRと10mmug/mlの山羊F(ab-2(Jackson ImmunoResearch)を交差させることによって引き起こされた(Wangら、2014)。逆時点で各標本にAnti-IgMを加え、全標本を一斉に固定することにより、時間経過分析を達成した。リン酸化の基礎レベルを決定するために、非刺激PBMCの平行培養物を時間ゼロで固定した。リン化CD79a (pIgα)を検出するために、電池を固定(1.5%パラホルムアルデヒド、5分、室温)、浸透(90%メタノール、10分、4°C)し、pCD79A (Igα、Tyr82)に特有のウサギに染め、続いてPE結合ヤギ反ウサギIgG (Jackson ImmunoResearch Laboratories)した。
分離方策および分離細胞集団の純度は、図12Aおよび12Bに示されている。各ドナーから2種類の分類を行い(HC#2を除く)、IGHVに使用された1種類とTRB分析に使用された2種類の分類を異なる時点で実施した。膵島自己抗体およびHLA遺伝子型を含むドナーの特徴を以下に示す(表1、図には示していない)。自己Bcon電池を、IGVH解析のための制御およびTRB解析のためのTcon電池として使用した。簡単に言うと、分離したばかりのPBMCは、CD19、CD5、IgD、およびTCRαβのために30分間、氷上で汚れ、徹底的に洗い、事前緩衝液(BD生命科学)で吊るした。プロピジウムヨウ素(PI)は、生存できない電池を除外するために、選別の直前に加えられた。整理した電池を氷上の50% FBSで採集した。IgD+DEgD電池はCD19、CD5、IgD、TCRαβ(800-1000電池/ソート)、IgD+DE電池はCD19+CD5+IgD+TCR電池(100-200電池/ソート)として識別されました。Bcon電池はCD19+CD5-TCR-、Tcon電池はCD19-CD5+TCRi+電池と同定されました。総DNAは、製造者の指示に従い、QIAMP DNAミニキット(Qiagen)を用いて選別電池から直接抽出した。本文に記載されているように、IgD+およびIgD-DE細胞から選別されたDNA、Bcon細胞およびTcon細胞をBCRまたはTCRBV配列決定に使用した。
IGHVおよびTRBVクローン型の分析は、調査レベル分解能(Adaptive Biotechnologies)で免疫SEQプラットホームを用いて、各選別細胞型由来のゲノムDNAについて実施した。High-through sequencingと高度なバイオインフォマティクスパイプラインを組み合わせたCDR3地域分析(Carlson et al., 2013; DeWitt et al., 2016; Robins et al., 2009)を用いて、multiplex PCRを行った。試料は試料ごとに40ngから100ngのゲノムDNA増幅した。T1D#1からIgD-を、T1D#2からIgD+セルを配列エンスしようとしたところ、失敗しました。TCRβおよびIGH 配列は、Adaptive Biotechnologies でご利用いただけます。個々のサンプルからのRaw ImmunoSeqデータをImmunoSeqアナライザー2.0ソフトウェア(Adaptive Biotech)で処理した。処理されたデータの測定メトリックは、tsvファイル形式でエクスポートされ、Rプラットホームを使用して分析されました。vGeneのアイデンティティが不明またはフレーム外のクローン化は下流分析から除外した。各細胞種のvGeneについては、「試料中に存在する細胞ゲノムの推定数」のメトリック値を合計し、それに対応するパーセンテージを算出した上で、別個の細胞クローンの数を算出した。割合量化は、vGene (VHとVβの使用法が、異なるセルタイプと試料を通して公平かつ一貫して比較されるようにすること、そして、DEサブセットセルの異なる割合と非常に小さな割合の配列から生じるいかなる影響も最小限に抑えること)のための均一な基準を提供する。パーセンテージをバープロットで可視化し、異なるセル型間でのvGeneの使用を直線的に比較した。異なるセルサブセットにおけるvGeneの有無は、vGeneの用法に基づいて決定した。異なるセルサブセット間のユニークで共有されたvGenesを特定し、R Limmaパッケージの機能を用いてVenn図に表示した。vGene突然変異はIMGTデータベースとのアラインメントに基づいて同定され、その上で生殖系列との差異がマークされ、計数され、記録される。生きたImmunoSeqデータtsvスプレッドシートの「vAlignution Count」の欄に、vGene突然変異値がさらに合計され、Rを用いたボックスプロットと散布図の組合せで表示された。選択されたクローンのアミノ酸配列を用いたストライング検索を行い、個々の被検者におけるそれらの存在を決定した。インバリアントクローン型のアミノ酸配列の文字列検索、「CARQEDTAMVYYFDYW」(SEQ ID NO:1)もまた、ナイーブ細胞および記憶部B con細胞の3700万個の固有BCR配列の公衆ImmunoSEQデータベース3に対するR script、およびnPOD適応免疫レパートリーから調べたIBCのデータベース3に対してR scriptを用いて実施された。
x‐Idクローノタイプが末梢血中で検出できるかどうかを判定するために、PBMCの分析のために2つのPCRプローブを設計し使用した。最初のプローブでは、VH04-b-specific sense primer (5GCTGCTGGATGTGATGGTAGTA-3) (SEQ ID NO:17) とアンチセンスプライマー (CCCAGTAGTAAACCATA) (SEQ ID NO:18) がCDR3領域全体を補完するものであった(図3J)。第2のプローブでは、VH04-b特異的プライマーを、JH04に及ぶリバースプライマー(3TCCCTGGCCAGTAGTCAAAGTAGTAGTA-5)(配列番号:19)と対合させ、N2領域で終了させた(図14参照)。簡単に述べると、RNeasy血液ミニキット(Quigen)を用いて新鮮なPBMCからRNAを抽出し、NanoDrop (ND-1000分光光度計)により分析して純度を評価し、濃度を測定した。RevertAid First Strand cDNA Synthesis Kit (Thermofisher)を用いて精製RNA約1μgを逆転写(RT) PCRし、キットプロトコルに従ってcDNAを調製した: RNAを5X反応混合物、ランダムヘキサマープライマー、およびRevertAid M-MuLV RT (200 U/μL)酵素混合物と共に、最終容量20μLで25℃で10分間インキュベートし、続いて42℃で60分間、そして70℃で5分間不活化した。正(GAPDH特異的プライマー)および陰性(RT酵素なしの反応混合物)対照反応を用いて、cDNA合成の特異性を検証した。2X QIAGEN HotStarTaqマスターミックスを用いて調製された全体積の25マイクロL cDNAを以下の条件下で実施した:初期変度は95°Cで3分、95oCで30s、54oCで30s、40サイクルで72oCで1分、続いてサーモサイクラー(BioRad T100)を用いて10分間72oCで最後の延長ステップ。PCR産物アガロースゲル上で200bpのバンドサイズとして可視化され、バンドは、Johns Hopkins Medical Institute GRCF配列決定コアでPCR精製キット(Quigen)とサンガー配列決定を用いて、切断され、浄化された。Immunogenetics IMGT/V-QUESTソフトウェアを用いて、配列を分析した。
新しいペプチド系は、HLA-DQ8(PDB ID 1JK8)(シャープ、2012)に結合したインスリンB鎖エピトープの結晶構造から構築された。インスリンのエピトープ配列は、VMDからのミューテータープラグインを使用して新しいペプチドエピトープ配列にミュートされ、新しいペプチドエピトープが望ましい登録されていることを確認した。HC#1のCDR3エピトープも、新しいペプチドシステムと同じ手順でインスリン-boundエピトープ構造(PDB ID 1JK8)から作られました。スーパーアゴニストシステムは、HLA‐DQ8(PDB ID 5UJT) (Wangら、2018)に結合したインスリンミモトープの結晶構造から構築された。このシステムのために、スーパーアゴニストに一致するようにエピトープを変異させることに加えて、両方のHLA鎖を変異させ、インスリン結晶構造(PDB ID 1JK8)中のHLAの配列に一致させた。より具体的には、ディスタル残渣の他に、HLAの残渣72Cを1JK8 HLA配列に一致するようにIsoleucineにミュートした。各システムをTIP3P水箱に溶かし、Na+とCl-を用いて100mM濃度で充電、中和、電化した。
結合親和性自由エネルギー動乱(FEP)法を用いて計算した。製造MDシミュレーションの最後の構造をFEP演算処理のために選んだ。束縛(HLA +エピトープ)と自由状態(エピトープのみ)の自由エネルギー動乱算出を算出し、各算出に対して6個のレプリカを用いた。エピトープ間の広範な配列差のために、エピトープをエピトープの長さであるポリグリシンの中性中間配列に変異させた。2 つのトポロジーは、VMD からのMutator プラグインを使用して実装されました。各システムは、エピトープからポリグリシンへ、最大0.04の増加を用いて、端部に向かってより小さい増加で、各システムの少なくとも34のFEP窓を合計し、ゆっくりと変形した。各FEPウィンドウは1nsで実行され、800ns以上のシミュレーション(6つのレプリカ×34の窓×2つの状態(複雑+自由)×2つのエピソード)が得られた。静電は=0.1からオンになった。各窓の収束は、レプリカ間の値を比較することによって評価された。CHARMM36プロテインフォースフィールドとTIP3P水モデルを用いたNAMD2.11は、MD作業に一致するFEP算出に使用された。FEPの軌跡を観察することにより、ポリグリシンはシフトレジではなく、エピトープのスタートレジストリを維持する。自由エネルギー誤差バーは標準誤差である。
不死化DE電池を生成するために、説明した戦略(図6A)を用いて、FACSAria IIを用いて、新しく分離されたPBMCからIgD+ DE電池を分類した。選別された電池は、24時間早く照射された繊維電池でコーティングされた96ウェルマイクロプレート(ATCC(R)55-XTM)のウェルあたり10、25、50、または100セルで播種された。Caputoら(Caputo and Flytzanis, 1991)によって記載された方法に従って、培養物に2.5μg/mlのCpG ODN 2006(ODN7909)およびB95-8細胞(ATCC(R) VR-1492)由来のEBV上清ストックをパルスした。栽培は、5~7日毎に培地の半分を新しい培地に置き換えて維持した。不死細胞は、50~100個のDE細胞を播種した栽培で8日後に見られた。リンパ芽球様細胞株(以下、x-LCL)を選択し、その後の分析を行った。ある実験では、x-LCLから単セルを選別し、Ig重鎖とL鎖の表現を検討した。第二の設定では、希釈(0.3セル/ウェル)を制限してx1.1クローン化を生成するためにx-LCLを用いた(Hamadら、1994)。x1.1クローンの細胞を用いて、BCRおよびTCRの分析、ならびに自然抗体産生を行った。
著者らは、不死化DE細胞集団の単セルクローン性を保証するために2つのアプローチを使用し、RNAキャッチ緩衝液を含有する上述のようにFACSAria II (BD Biosciences, Bedford, MA)を使用して、96ウェルマイクロプレート上で成長したモノクローナルDE細胞を選別した(Smithら、2009)。
浄化と消化の後、単一細胞からの、増幅されたcDNAsは、ヒトIgGを含む式ベクトルにクローン化され、また、前述したように(Smith et al., 2009)、Ig醍醐味の定常領域(AbVec-hIgG1, AbVecIg)を含む式ベクトルにクローン化された。軽鎖Ig遺伝子を含む重およびAbVecIgG1を含むAbVec-hIgG1は、ポリプラスジェット-プライムトランスフェクション(who)および製造者の指示を用いて293Aセルラインに共移植された。移送された293A電池は、4~5日間、セラムフリー基底媒体で抗物質を秘密にし、mAbRは固定性タンパク質Aカラム(Pierce)を用いて精製した。反体式と純度はSDS-PAGEによって確認され、EZQタンパク質定量キット(Invitrogen)を用いて精製された抗たんぱく濃度を測定した。
TCRαおよびTCRα鎖の遺伝子を、Eugsterら(Eugsterら、2013)によって記載された方法の改良版を用いてクローン化した。簡単に言うと、合計RNは、RN抽出キット(バイオラボ)を用いて、x1.1クローンの細胞から分離された。cDNAを、OneStep RT-PCRキット(Qiagen)を用いて、αおよびα鎖のための特定のプリマーを用いて、ネストされたPCRにより、αおよびα鎖を別々に増幅するのに使用し、また、ベータ鎖のためのデグレートプライマーと混ぜ合わせた。増幅された製品を1.5%のアガロースゲルで可視化し、pGEM(R)-T Easyベクトル(Promega)にクローン化した。プラスミド抽出キット(Qiagen)とM1プライマー(Eugsterら、2013)を用いてサンガー配列を用いてDNAを抽出した。可変領域の完全な配列を用いて、IMGTV-QuestによるVDJの使用とCDR3を確認した。
x1.1クローンの細胞を完全なメディアで3~4日間拡大し、PBSで洗浄し、盆地メディア(スター法)で5日間栽培した。分泌されたmAbNは、SDS-PAGEを用いて上清中に検出された。秘密のmAbのアイソタイプは、Pro-DetectTM Rapid Antibody Isotyping Assay Human Kit (Thermo Fisher)を用いてIgMと決定した。抗体濃度は、EZQ Protein Quantitation Kit (Invitrogen)を用いて測定した。
我々は、確立された方法(Crawford et al., 2011)を用いて、(NIH Tetramer Core Facility)によって提供されたバイオティレートHLA-DQモノマーを用いて3つのDQ8量体を作った。量体の1つは、HLA-DQとx-Idペプチド(x-Idテット)、1つはインスリンミモトープ(mim-tet)、3つ目はクリップ (クリップ-tet)で構成されている。トロンビンを用いて切り口を除去し、空のモノマーに0.2mm/mlのx-Idまたはインスリン-mimotopeペプチドを装填した。積荷は、2.5mm/ml(0.25%) n-octyl-i-Dglucopyranosideおよび1-M Pefabloc SC (Sigma-Aldrich)の存在下で72時間、37℃で実施した。ペプタイドを積載したHLA‐DQ一量体は、それぞれ1:4のモラル比でポリエチレン結合ストレプタビジン(eBioscience)でテトラメリド化された。HLA-DQ/CLIPモノマーは、汚れの中では負の制御としてテトラメリド化された。量体錯体の成功形成は、穏やかなSDS‐PAGEによって確認された。量体染料は、FACSバッファーの室温で1時間、HLAクラスII量体と2マイクログラム/mlのPBMCsをインキュベートした。表面CD4、TCR、CD19の個体固有を用い、試料を採取した。以上のように分析した(Dai et al., 2015)。
図中には、実験例複製と試料の大きさの説明が記載されています。Prism 6(GraphPad Software)を用いて、結果の統計的意義を行った。適切な場合には、独立サンプルt-testまたはペアサンプル生徒t-testを用いて分析を行った。結果は、特に明記しない限り、平均@SEMで表した。p?0.05は統計的に有意とみなした。

結果
次に、DEのTCRβチェーン(TRBV)とIg重鎖(IGHV)レパートリーを分類し分析し、ゲノムDNAとハイスループットImmunoSEQ (Adaptive Biotech)を用いてそれらの従来のレパートリーと比較した。この分析は、DEのクローン性を決定し、また、あるプラズマ膜たんぱく質の別のものへの移行(すなわちトロゴサイトシス(LeMaoultら、2007)のようなタンパク質レベルでの予期せぬアーチファクトを排除するために重要であった。
xクローノタイプのDEと普及をさらに明らかにするために、IgD+とIgD-DEのレパートリーを取得・分析し、HC#1のBcon 細胞のレパートリーと比較することができた。HC#1のDEsのレパートリーは、Bcon 細胞(図3Gと表6は示されていない)と同様に多様であることがわかった。IgD+電池、IgD-電池、HC#1のBcon 電池(表6Aは示されていない)では、IGHV04-b遺伝子の使用はまれであった(0.015%以下)。さらに重要なことは、HC#1のIgD+、IgD-およびBcon 細胞のレパートリー(表6B、6Cおよび6Dは示されていない)から、xクローノタイプは存在しなかったことである。それにもかかわらず、T1D被検者と同様にHC#1におけるIGHV04‐b{IgD#細胞#1は、DH05‐18遺伝子ではなく、IGHJ04‐01*2遺伝子を用いた1つのクローンタイプから成り、それらのCDR3配列(CARQRFWSGPLFDYW) (配列番号21)はx‐クローンタイプのそれと部分的に一致した(太字)。しかし、IGHV04-b+IgD+DE電池は、IGHJ04-01*2を使用した5つのクローノタイプで構成されていたが、DH05-18ではなかった(図3H)。さらに、T1D患者と同様に、HC#1のDEクローノタイプは、ほとんど体性突然変わりはないが、生殖ラインの構成であった(図3I)。したがって、HC#1におけるDEのレパートリーは、3つのT1D被検者におけるものとは異なり、多様であり、x-クロノタイプを含まなかった。
本研究では、T1D被検者でクローン性に増殖し、TCRおよびBCRの発現によりエピトーム化されたB細胞およびT細胞の両方の系統マーカーを内包するまれなリンパ球(DE)について報告する。クローナルに拡張されたDEsは、糖尿病発生DQ8分子の最適なレジストリを持つ、Ig重鎖の抗原結合部位にある強力な自己抗原(x‐自己抗原)をエンコードする。x自己抗原ペプチドは、HC被検者ではなくT1D由来のCD4 T細胞を強固に刺激するDQ8分子と機能的複合体を形成する。さらに、xクローン型OOBmAbもCD4 T細胞を刺激する。競合結合抑制分析は、x-mAbとインスリンミモトープが重複するT細胞サブ集団を刺激することを示す(図7C)。まとめると、これらの知見は、T細胞とB細胞の区画化は絶対的ではなく、このパラダイムの違反因子が自己免疫の鍵となる要因となりうることを示している。

患者サンプル中のx-mAbの検出
本発明者は、血液液中のx-mAbの存在を測定するために、ELISAアッセイを開発した。特定の実施例において、この方法は、リスクのある個人を識別するために、ベアリング/ベアリングは新しい患者におけるT1Dの診断を確認するために、xイディオタイプを持っている個人のスクリーニングに使用することができる。特に、健康な個人からの血液が対照として用いられる。
議定書:
1.40Cの重炭酸緩衝液に様々な濃度の防xmAbを含んだ高い親和性96ウェルプレートを一晩でコーティングする。
2.皿を4回、0.05%のリン酸緩衝塩200ウルで洗います。
3.ブロック緩衝液(20% FCS)を用いて、370Cで2時間ブロックプレート。
4.1:50から2倍の連続希釈液をブロッキングバッファーで準備する。
5.200マイクロリットルのPBSTで皿を6回洗いなさい。
6.3つの複製井戸に希釈した血清を加える。
7.バックグラウンドシグナルの制御には、防x-mAbまたは血清を欠いた井戸を使用する。
8.一晩40℃で皿を育てる。
9.200マイクロリットルのPBSTで皿を6回洗いなさい。
10.1: 10,000に薄めた二次検出(ヤギ防IgG)を用意する。
11.指示されているように、別の井戸に二抗体反応を加えてください。
12.370Cで30分間、皿を育てる。
13.200マイクロリットルのPBSTで皿を6回洗う。
14.各ウェルにTMB溶液分間加え、硫酸で反応を止めます。

1型糖尿病の予測
ラベルのx-mAbは、T1Dが発達するリスクの高い個人(例えば、2つ以上のT1D抗毒薬を持つ人)のT1D比X細胞の存在は、誰がT1Dを発達させるだろうかと予測することを示している。
1型糖尿病の予防と治療
細胞に結合し、細胞を不活性化または破壊するX細胞の表面上のT1D疾患特定のアミノ酸配列(「x-mAbs」)に対するヒト化抗体は、その病原性作用を妨げる。X細胞をT1D自己反応性T細胞の集団に加えると、著しく活性化され、急速に分裂し、サイトカインを分泌する。x-mAbの無力化作用を実証するために実験が行われ、以下のような実験が行われている:
1.x-mAbおよびT1D比X-細胞をT1D自己反応性T細胞の集団に加え、サイトカイン放出および自己反応性T細胞の拡大がin vitroで遮断されることを実証する。
2.T1Dのヒト化マウスモデルにおいて、T1D比X細胞の添加は、膵島細胞のリンパ球浸潤(膵島炎)およびT1Dをもたらす。
3.T1Dのヒト化マウスモデルでは、T1D比X電池を投与されたマウスをx-mAbで処理すると、インスリン炎とT1Dの発生が阻止される。
4.T1Dを発症するリスクの高い患者(例えば、2以上のT1D抗体を有する患者)をx-mAbで治療すると、この疾患は防止される。
5.T1Dを発症するリスクが高い(例えば、T1D比X細胞および1以上のT1D抗体を有する)患者をx-mAbで治療することにより、この疾患は防止される。
Claims (55)
- 患者から得た生物学的サンプルからSEQ ID NO:1をエンコードするヌクレオチド配列を検出する方法。
- 発見ステップがポリメラーゼ連鎖反応(PCR)を含む記載の方法。
- PCRが、SEQ ID NOS:17-18を含むプライマーを用いた増幅を含む記載の方法。
- 記載の方法で、PCRがSEQ ID NOS:23及び19からなるプライマーを用いた増幅を含むもの。
- 記載の方法で、ヌクレオチド配列がSEQ ID NO:25を含む。
- 生物試料が周辺血液であることを特徴とする請求項1の方法。
- 発見ステップを更に配列決定を含む、請求項2-6のいずれかの方法。
- 記載の方法で、HLA-DQアレルをゲノタイピングする工程をさらに含む。
- 患者が1型糖尿病(T1D)のリスクがあるかどうかを判定するための方法であって、以下のステップを含む、方法:
(a)患者から得た生物学的サンプルからSEQ ID NO:1をエンコードする核酸配列の検出;
(b)HLA-DQ7アレルまたはHLA-DQ8アレルの存在を検出するためのHLA-DQのジェノタイプ。これは、SEQ ID NO:1およびHLA-DQ8を有する患者がT1Dの危険にさらされ、SEQ ID NO:1およびHLA-DQ7を持たない患者はT1Dの危険にさらされず、SEQ ID NO:1を持たない患者はT1Dの危険にさらされない。 - 以下のステップを含む、患者が自己免疫疾患の危険があるかどうかを判定するための方法:

- SEQ ID NO:1を具体的に結合する、それらの抗原結合フラグメント。
- B細胞レセプターがSEQ ID NO:1で構成される、(i)B細胞レセプター上で表現されるB細胞レセプターを具体的に結合する、又はその抗原結合フラグメント、又は(ii)SEQ ID NO:1で構成される自由浮遊する、その断片。
- SEQ ID NO:1を含む、特定的に結合する、その抗原結合フラグメント。
- 請求項12-14のいずれかの抗原結合フラグメント、すなわち、抗原または抗原結合フラグメントが、SEQ ID NO:1への抗原の結合を防止または減少させる。
- 抗原結合フラグメントが、scFv、sc(Fv)2、Fab、F(ab)2、およびダイアボディからなるグループから選択される、請求項12-15のいずれかの抗原結合フラグメントまたは抗原結合フラグメント。
- T1Dを有する被検者における第1種糖尿病(T1D)の治療又は予防の方法、又は、請求項12-16のいずれかの抗原結合フラグメントの治療効果のある量を患者に施術する工程を含むその危険性。
- 重鎖相補性決定領域(CDR)1、2および3を含む単離された抗体またはその抗体結合フラグメントであって、重鎖CDR1が、配列番号60に示されるアミノ酸配列、または配列番号60に示されるアミノ酸配列で2つ以下のアミノ酸位置での置換を有するアミノ酸配列を含み、重鎖CDR2が、配列番号62に示されるアミノ酸、または配列番号62に示されるアミノ酸で2つ以下のアミノ酸位置での置換を有するアミノ酸を含み、重鎖CDR3が、配列番号64に示されるアミノ酸配列、または配列番号64に示されるアミノ酸で2つ以下のアミノ酸位置での置換を有するアミノ酸配列を含む、単離された抗体またはその抗体結合フラグメント。
- 単離された140又は抗原結合フラグメントが更に軽鎖CDR1、2及び3を含み、軽鎖CDR1はSEQ ID NO:66に記載されたアミノ酸配列、又はSEQ ID NO:66に記載されたアミノ酸配列は2又はそれ以下のアミノ酸位置に置き換えられ、軽鎖CDR2はSEQ ID NO:68に記載されたアミノ酸配列、又はSEQ ID NO:68に記載されたアミノ酸配列は2又はそれ以下のアミノ酸位置に置き換えられ、軽鎖CDR3はSEQ ID NO:9に記載されたアミノ酸配列、又はSEQ ID NO:9に記載されたアミノ酸配列は2又はそれ以下のアミノ酸位置に置き換えられた。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプター、(2)SEQ ID NO:1で構成されるB細胞レセプター、(2)および(SEQ ID NO:1で構成される自由浮遊型の)を具体的に結合する、(1)B細胞レセプターの断片であるB細胞レセプター、または、(2)それの反対又は抗原結合フラグメントが、それぞれSEQ ID NO:60、62および64に規定されるアミノ酸配列で構成されるCDR1、CDR2およびCDR3から成る重鎖可変領域(VH)で構成される自由浮遊型の。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプターに、(2)SEQ ID NO:1で構成されるB細胞レセプターまたは(2)SEQ ID NO:1で構成されるフリーフローティング(SEQ ID NO:1で構成される)を具体的に結合する、(1)B細胞レセプターの断片又は抗原結合断片は、SEQ ID NO:66、68及び9に規定されるアミノ酸配列から成るCDR1、CDR2及びCDR3で構成されるライトチェーン可変領域(VL)で構成される。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプター上に表されるB細胞レセプター、又は(2)それが構成されるSEQ ID NO:1で構成される自由浮遊型のバラエティを具体的に結合する、単離された、又は抗原拘束する断片、又は(2)SEQ ID NO:1で構成される:
(a)SEQ ID NOS:60, 62, 64に記載されているアミノ酸配列からなるCDR1, CDR2, CDRからなるVH;
(b)SEQ ID NOS:66, 68, 9に記載されているアミノ酸配列からなるCDR1, CDR2, CDR3からなるVL。 - 請求項18-22のいずれかの単離された核酸分子で、当該請求項又は抗原結合フラグメントをエンコードしている。
- 請求項23の核酸分子からなるベクトル。
- 請求項24のベクトルからなる宿主細胞。
- 宿主細胞が原核細胞または真核細胞である、請求項25記載の宿主細胞。
- 宿主細胞による、自己又は抗原結合フラグメントの表現に適した条件下で請求項25の宿主細胞を(a)文化する段階、及び(b)それらの抗原結合フラグメントを回収する段階で構成される、抗原結合フラグメントを作る方法。
- 宿主細胞が原核または真核細胞であることを示す請求項27の方法。
- 請求項18-22のいずれか1つによれば、それらの抗原結合フラグメント又は抗原結合フラグメントを含む組成物と、適切な医薬品担体。
- 請求項29の組成物であって、組成物は、静脈、筋肉内、口下、皮下、腹内、筋内、または筋肉内の行政のために定式化される。
- 請求項18-22のいずれかの抗原結合フラグメントまたは抗原結合フラグメントの治療効果のある量を哺乳動物に管理する工程を含んでいる哺乳動物における糖尿病を治療する方法。
- 請求項18-22のいずれかのうちの1つで、哺乳動物に、抗原結合フラグメントまたは抗原結合フラグメントの治療効果的な量を与える工程を含んでいる哺乳動物における自己感染病を治療する方法。
- 抗原結合フラグメントがscFv、sc(Fv)2、Fab、F(ab)2、およびダイアボディからなるグループから選択される請求項22の抗原結合フラグメント又は抗原結合フラグメント。
- 分離された、又は抗原結合フラグメントは、(1)B細胞レセプターがSEQ ID NO:1を構成する、B細胞レセプターに表現されるB細胞レセプターを、(2)それの抗原結合フラグメントは、SEQ ID NO:74に記載されるアミノ酸配列から成るVHを構成するSEQ ID NO:1を構成する自由浮遊する。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプターに、(2)SEQ ID NO:1で構成されるB細胞レセプターを具体的に結合する単離された、又は(2)SEQ ID NO:1で構成される自由浮遊性の、(この場合、それらの抗原結合フラグメントは、SEQ ID NO:76に記載されるアミノ酸配列で構成されるVLで構成される)B細胞レセプターを具体的に結合する、単離された、又は抗原結合フラグメントである。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプター、(2)SEQ ID NO:1で構成されるB細胞レセプター、又は(2)SEQ ID NO:1で構成される自由浮遊性のビーズであり、このような、それらの、又は抗原結合フラグメントは、SEQ ID NO:74に記載されるアミノ酸配列で構成されるVH及びSEQ ID NO:76に記載されるアミノ酸配列で構成されるVLで構成される。
- 重鎖CDR1が、配列番号78に記載のアミノ酸配列、または配列番号78に記載のアミノ酸配列の2つ以下のアミノ酸位置での置換を有するアミノ酸配列、配列番号80に記載のアミノ酸、または配列番号80に記載のアミノ酸の2つ以下のアミノ酸位置での置換を有する重鎖CDR2、および配列番号82に記載のアミノ酸配列、または配列番号82に記載のアミノ酸の2つ以下のアミノ酸位置での置換を有する重鎖CDR3を含む、重鎖相補性決定領域(CDR)1、2、および3を含む、単離された抗体またはその抗体結合フラグメント。
- 請求項37の単離された抗菌作用又は抗原結合フラグメントはさらに軽鎖CDR1、2及び3を含み、軽鎖CDR1はSEQ ID NO:84に記載されたアミノ酸配列、又はSEQ ID NO:84に記載されたアミノ酸配列は2又はそれ以下のアミノ酸位置に置き換えられ、軽鎖CDR2はSEQ ID NO:86に記載されたアミノ酸配列、又はSEQ ID NO:86に記載された2又はそれ以下のアミノ酸位置に置き換えられたアミノ酸配列及びSEQ ID NO:88に記載された軽鎖CDR3は2又はそれ以下のアミノ酸位置に置き換えられ、又はSEQ ID NO:88に記載されたアミノ酸配列である。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプター、(2)SEQ ID NO:1で構成されるB細胞レセプター、又は(2)それを構成する自由浮遊性の(SEQ ID NO:1で構成される)それらの、又は抗原結合フラグメントは、それぞれ、SEQ ID NO:78、80及び82に規定されるアミノ酸配列から成るCDR1、CDR2及びCDR3で構成される重鎖可変領域(VH)で構成される。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプターに、(2)SEQ ID NO:1で構成されるB細胞レセプターまたは(2)SEQ ID NO:1で構成されるフリーフローティング(SEQ ID NO:1で構成される)を具体的に結合する、(1)B細胞レセプターの断片又は抗原結合断片は、それぞれSEQ ID NO:84、86および88に規定されるアミノ酸配列から成るCDR1、CDR2およびCDR3から成る軽鎖可変領域(VL)で構成される。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプター上に表されるB細胞レセプター、又は(2)それが構成されるSEQ ID NO:1で構成される自由浮遊型のバラエティを具体的に結合する、単離された、又は抗原拘束する断片、又は(2)SEQ ID NO:1で構成される:
(a)SEQ ID NOS:78, 80, 82に記載されているアミノ酸配列からなるCDR1, CDR2, CDRからなるVH;
(b)SEQ ID NOS:84, 86, 88に記載されているアミノ酸配列からなるCDR1, CDR2, CDR3からなるVL。 - 請求項37-41のいずれかのうちの、抗原結合フラグメント又はそれらをエンコードする孤立した核酸分子。
- 請求項42の核酸分子からなるベクトル。
- 請求項43のベクトルからなる宿主細胞。
- 宿主細胞が原核細胞または真核細胞である、請求項44記載の宿主細胞。
- 宿主細胞による、自己又は抗原結合フラグメントの表現に適した条件下で、(a)請求項44の宿主細胞を栽培する段階、及び(b)それらの抗原結合フラグメントを回収する段階で構成される、抗原結合フラグメントを作る方法。
- 宿主細胞が原核または真核細胞である記載の方法。
- 請求項37-41のいずれかによれば、それらの抗原結合フラグメントまたは抗原結合フラグメントと適当な医薬品担体とを含む組成物。
- 請求項48の組成物であって、組成物が静脈、筋肉内、口下、皮下、腹内、皮内又は筋肉内の行政のために定式化されるもの。
- 請求項37-41のいずれかの抗原結合フラグメントまたは抗原結合フラグメントの治療効果のある量を哺乳動物に管理する工程を含む哺乳動物における糖尿病を治療する方法。
- 請求項37-41のいずれかの抗原結合フラグメントまたは抗原結合フラグメントの治療効果のある量を哺乳動物に管理する工程を含む哺乳動物における自己感染病を治療する方法。
- 抗原結合フラグメントが、scFv、sc(Fv)2、Fab、F(ab)2及びダイアボディからなるグループから選択される請求項41の抗原結合フラグメント又は抗原結合フラグメント。
- B細胞レセプターがSEQ ID NO:1で構成される、(1)B細胞レセプターに表現されるB細胞レセプターを具体的に結合する、分離された、又は抗原結合フラグメント、又は(2)その抗原結合フラグメントが、SEQ ID NO:90に記載されたアミノ酸配列からなるVHを含むSEQ ID NO:1で構成される自由浮遊する、SEQ ID NO:1で構成される。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプターに、(2)SEQ ID NO:1で構成されるB細胞レセプターを具体的に結合する単離された、又は(2)SEQ ID NO:1で構成される自由浮遊性の、それらの、又は抗原結合フラグメントは、SEQ ID NO:92に記載されるアミノ酸配列で構成されるVLで構成される。
- (1)B細胞レセプターがSEQ ID NO:1で構成されるB細胞レセプター、(2)SEQ ID NO:1で構成されるB細胞レセプター、(2)SEQ ID NO:1で構成される自由浮遊性のバラエティッド、すなわち、そのバラエティック抗原結合フラグメントは、SEQ ID NO:90で規定されるアミノ酸配列で構成されるVHと、SEQ ID NO:92で規定されるアミノ酸配列で構成されるVLで構成される。
