Abstract
Description

Claims (15)
- ポリヌクレオチド配列決定方法であって、以下:
(a)同じヌクレオチド配列を有する多様な鋳型ポリヌクレオチド鎖がフローセルの表面に結合する部位を含むフローセルに、第一の鎖伸長酵素およびブロック化、ラベル化ヌクレオチドの混合物を導入する工程であって、第一の鎖伸長酵素は、コピーポリヌクレオチド鎖が対応する鋳型鎖の配列に基づいて適切な一のブロック化、ラベル化ヌクレオチドをコピーポリヌクレオチド鎖中に組み込むように構成される、工程;
(b)組み込まれていないブロック化、ラベル化ヌクレオチドをフローセルから洗い流す工程;
(c)組み込まれていないブロック化、ラベル化ヌクレオチドのフローセルからの洗い流しの間またはその後ブロック化、アンラベル化ヌクレオチドの混合物を含む組成物をフローセルに導入する工程であって、ブロック化、アンラベル化ヌクレオチドの混合物のうちのあるブロック化、アンラベル化ヌクレオチドは、コピーポリヌクレオチド鎖が対応する鋳型鎖の配列に基づいてコピーポリヌクレオチド鎖中への組込みに利用可能であり、ただし、コピー鎖における以前に組み込まれたヌクレオチドは、もしあれば、ブロック化されない、工程;および
(d)コピー鎖中に組み込まれた一のブロック化、ラベル化ヌクレオチドの同一性を検出する工程であって、もしあれば、その一方でブロック化、アンラベル化ヌクレオチドの混合物はフローセルと共にインキュベーションされる、工程

- 第一の鎖伸長酵素およびブロック化、ラベル化ヌクレオチドの混合物は、組み込まれていないブロック化、ラベル化ヌクレオチドをフローセルから洗い流すことに先立ってフローセルと共に10秒またはそれよりも短い間インキュベーションされる、請求項1~9のいずれかに記載の方法。
- 第一の鎖伸長酵素およびブロック化、ラベル化ヌクレオチドの混合物は、組み込まれていないブロック化、ラベル化ヌクレオチドをフローセルから洗い流すことに先立ってフローセルと共に7.5秒またはそれよりも短い間インキュベーションされる、請求項1~9のいずれかに記載の方法。
- ブロック化、アンラベル化ヌクレオチドの混合物を含む組成物は、コピーポリヌクレオチド鎖が対応する鋳型鎖の配列に基づき、適切な一のブロック化、アンラベル化ヌクレオチドがコピーポリヌクレオチド鎖中に組み込まれるように構成される第二の鎖伸長酵素を含み、ただし、コピー鎖において以前に組み込まれたヌクレオチドは、もしあれば、ブロックされない、請求項1~12のいずれか一項に記載の方法。
- 第一の鎖伸長酵素および第二の鎖伸長酵素は、同じ酵素または異なる酵素である、請求項13に記載の方法。
- ブロック化、アンラベル化ヌクレオチドの混合物を含む組成物は、9~9.5のpHを有し、および緩衝剤、洗浄剤、硫酸マグネシウム、EDTA、および抗酸化剤を含む、請求項1~14のいずれか一項に記載の方法。
