Abstract
Description
本発明は、医学及び免疫学の分野に関する。特に、本発明は、ABCA4関連の状態の処置、予防及び/又は遅延において使用することができる新規アンチセンスオリゴヌクレオチドに関する。
ABCA4における常染色体劣性変異は、中心視力喪失によって特徴付けられ、全盲に至ることが多い進行性障害である、スターガルト病を引き起こす。スターガルト病の典型的な特質は、患者の眼底全体にわたり分布した多くの黄色の点(斑点)の存在である。ABCA4遺伝子は、50個のエクソンで構成され、2273個のアミノ酸からなるタンパク質をコードする。このタンパク質は、錐体及び杆体視細胞の外節において発現され、光伝達後の老廃物の除去において重要な役割を果たす。
第1の態様では、本発明は、配列番号4に示すヌクレオチド配列を有するポリヌクレオチドに結合する、及び/又は配列番号4に示すヌクレオチド配列を有するポリヌクレオチドと相補的である、スプライシングを制御する(redirecting)ためのアンチセンスオリゴヌクレオチドであって、好ましくは、配列番号5を有するポリヌクレオチドに結合するか、又は配列番号5を有するポリヌクレオチドと相補的である、アンチセンスオリゴヌクレオチド、より好ましくは、配列番号80を有するポリヌクレオチドに結合するか、又は配列番号80を有するポリヌクレオチドと相補的である、アンチセンスオリゴヌクレオチド、さらにより好ましくは、配列番号7、8、9;11、12、13;15、16、17;19、20及び21からなる群から選択されるポリヌクレオチドに結合するか、又は配列番号7、8、9;11、12、13;15、16、17;19、20及び21からなる群から選択されるポリヌクレオチドと相補的である、アンチセンスオリゴヌクレオチドを提供する。
定義により、アンチセンスオリゴヌクレオチド(AON)は、その標的に対して実質的に相補的(アンチセンス)であり、AONが、対応するプレmRNA分子に結合することを可能にし、これにより、理論に制約されることは望まないが、スプライシングに必須なタンパク質の結合を防止する。本発明者らが、ABCA4におけるいくつかの変異について以前に示した通り(国際公開第2018/109011号)、通常、このような結合の欠如は、標的化されたエクソンのスキッピングをもたらす。

先ず、材料と方法セクションにおいて、実験の詳細について記載する一方、結果は、さらに下の結果セクションにおいて記載及び説明する。
AON設計及び検査
AONを設計するために、より小型の257bpの目的の領域を選択した(配列番号5)。その後、約20ヌクレオチドの長さのオリゴヌクレオチドを設計した。全オリゴヌクレオチドを、in silico RNA構造予測に付した。予測される構造の差を示した及び/又は最良の到達性を有した8種のAONを設計し、異なるケミストリーにおいて注文した(表2及び図2を参照)。
c.768G>Tをホモ接合型で保有するSTGD1患者由来の皮膚生検を得て、線維芽細胞株を作製した。次に、線維芽細胞株に、シクロヘキシミドの存在下で8種の異なるAON(表2に記載)をトランスフェクトした。次に、トランスフェクトされた細胞をRT-PCR解析に付した。
製造業者のプロトコールに従ってヌクレオスピンRNAクリーンアップ(NucleoSpin RNA Clean-up)キット(カタログ番号740955-50;Macherey-Nagel、Duren、Germany)を使用することにより、全RNAを単離した。RNAを定量化し、製造業者の説明書に従ってアイスクリプト(iScript)cDNA合成キット(カタログ番号1708891;Bio-Rad、Hercules、CA)を使用することにより、1μg RNAからcDNAを合成した。最後に、次のABCA4プライマーを使用してネステッドPCRを行うことにより、AONの有効性を評価した:PCR1(エクソン4_Fw:5’-CACCCGGAGAGAATTGCAG-3’(配列番号38)及びエクソン8_Rv:5’-CCTGCATACTCGGCCGATG-3’(配列番号39));PCR2又はネステッドPCR(エクソン5_Fw:5’-GGAATACGAATAAGGGATATCTTG-3’(配列番号40)及びエクソン7_Rv:5’-CTTGAATTCTTGGTGACATATCAG-3’(配列番号41))。
AON8は、イントロン6における代替のスプライス部位を遮断し、c.768G>T変異により弱められてはいるが依然として用いることができる、本来のスプライスドナー部位に戻るようにスプライス部位を制御することができた(図3A及び図3B)。結果として、ABCA4プレmRNAスプライシングを回復させることができる。
次に、本出願人らは、より多くのAON(配列とケミストリーの観点から異なる)を使用し、複数の細胞系(HEK293T細胞及び患者由来線維芽細胞)を用いた、実施例1の拡大について報告する。
AON設計
AONを設計するために、235bpの小型のABCA4領域を選択した;ABCA4 c.669~c.768+135。その後、総計26種のAON(それぞれ21ヌクレオチドの長さ)を設計し、AON10~AON35と表示した。AON10~AON34は、2’-O-メチル(2’-OMe)化学修飾により設計し、AON35は、2’O-メトキシエチル(2’-O-MOE)ケミストリーにより設計した。最初の結果の後に、2’-O-MOEケミストリーを有するさらに5種のAONを注文し、AON36~AON40と表示し、これらは、それぞれAON14、15、16、19及び23の配列に対応する。ケミストリー毎に、それぞれAON22及び35に相補的な配列を有するセンスオリゴヌクレオチドを注文した。全AON(及びSON)は、表3に収載されている。収載されているAONは全て、ホスホロチオエート骨格を有する。

全検査を2回繰り返して行った。HEK293T細胞を6ウェルプレートに播種し、野生型ABCA4エクソン6及びイントロン6(BA4_WT)を含有するミディジーン、並びにc.768G>T変異を含有する以外は同じミディジーン(BA4_MUT)を6μgトランスフェクトした。一晩インキュベーション後に、HEK293T細胞をトリプシンで消化し、12ウェルプレートに蒔いた。細胞は、付着するようになったら、ヒュージーン(FuGENE)(カタログ番号E2311;Promega)を使用して1μM濃度のAON又はSONをトランスフェクトし、48時間インキュベートした。トランスフェクトされた細胞は、RT-PCRによりRNAレベルで解析した。
製造業者のプロトコールに従ってヌクレオスピンRNAクリーンアップキット(カタログ番号740955250;Macherey-Nagel、Duren、Germany)を使用することにより、全RNAを単離した。RNAを定量化し、製造業者の説明書に従ってスーパースクリプト(SuperScript)VILO cDNA合成キット(カタログ番号11755050;Invitrogen)を使用することにより、1μg RNAからcDNAを合成し、その後、20ng/μlの作業濃度となるようにH2Oで希釈した。次のBA4ミディジーン特異的プライマーを使用して逆転写PCRを行うことにより、AONの有効性を評価した:ABCA4 ex6_Fw:5’-CTTCAGCCAGAGACGCGGGGC-3’(配列番号81)及びpCI-Neo-Rho ex5_Rev:5’-AGGTGTAGGGGATGGGAGAC-3’(配列番号82)。RHOエクソン5を標的とするプライマーを使用したRT-PCRは、ミディジーントランスフェクション有効性を評価するための対照として使用した(pCI-Neo-Rho ex5_Fw:5’-ATCTGCTGCGGCAAGAAC-3’(配列番号83)及びpCI-Neo-Rho ex5_Rev:5’-AGGTGTAGGGGATGGGAGAC-3’(配列番号84))。25μlの総反応物における10μMの各プライマー、2μMのdNTP、2.5mMのMgCl2、1Uのタック(Taq)ポリメラーゼ(Roche、Basel、Switzerland)及び40ngのcDNAにより、次のPCR条件を使用して、RT-PCR解析を行った:94℃で3分間、続いて、94℃で30秒間、58℃で30秒間及び72℃で2分間を35サイクルと、72℃で3分間の最終伸長。PCR産物は、2.5%アガロースゲルにおいて分離し、選択されたバンドは、サンガー配列決定により確認した。観察された非特異的バンドは、解析に考慮しなかった。フィジー(Fiji)ソフトウェアを使用して、ヘテロ二重鎖、変異体及び野生型バンドの半定量的解析を行った。ヘテロ二重鎖バンドは、グラフ表示のため、正しい転写物と異常な転写物の間で等しく分布した。
線維芽細胞トランスフェクション
健康な個体及びABCA4 c.768G>Tがホモ接合型のSTGD1患者由来の皮膚生検を得て、2種の線維芽細胞の細胞株を作製した。線維芽細胞を6ウェルプレートに蒔き、シクロヘキシミドの存在下でヒュージーン(カタログ番号E2311;Promega)を使用して1μM濃度のAONをトランスフェクトした。トランスフェクトされた細胞は、RT-PCRによりRNAレベルで解析した。
製造業者のプロトコールに従ってヌクレオスピンRNAクリーンアップキット(カタログ番号740955250;Macherey-Nagel、Duren、Germany)を使用することにより、全RNAを単離した。RNAを定量化し、製造業者の説明書に従ってスーパースクリプトVILO cDNA合成キット(カタログ番号11755050;Invitrogen)を使用することにより、1μg RNAからcDNAを合成し、その後、20ng/μlの作業濃度となるようにH2Oで希釈した。次のABCA4プライマー:エクソン5_Fw:5’GGAATACGAATAAGGGATATCTTG-3’(配列番号85)及びエクソン7_Rev:5’-CTTGAATTCTTGGTGACATATCAG-3’(配列番号86)を使用した線維芽細胞対照細胞株におけるRT-PCR、並びに次のABCA4プライマー:PCR1エクソン4_Fw:5’-CACCCGGAGAGAATTGCAG-3’(配列番号87)及びエクソン8_Rev:5’-CCTGCATACTCGGCCGATG-3’(配列番号88)並びにPCR2エクソン5_Fw:5’GGAATACGAATAAGGGATATCTTG-3’(配列番号89)及びエクソン7_Rev:5’-CTTGAATTCTTGGTGACATATCAG-3’(配列番号90)を使用したABCA4 c.768G>Tを保有する線維芽細胞におけるネステッドPCRを行うことにより、AONの有効性を評価した。25μlの総反応物における10μMのエクソン5_Fw及びエクソン7_Rwプライマー、2μMのdNTP、2.5mMのMgCl2、1Uのタックポリメラーゼ(Roche、Basel、Switzerland)並びに80ngのcDNAにより、次のPCR条件を使用して、線維芽細胞対照細胞株におけるRT-PCRを行った:94℃で3分間、続いて、94℃で30秒間、58℃で30秒間及び72℃で2分間を35サイクルと、72℃で3分間の最終伸長。25μlの総反応物における10μMのエクソン4_Fwプライマー及びエクソン8_Rwプライマー、2μMのdNTP、2.5mMのMgCl2、1Uのタックポリメラーゼ(Roche、Basel、Switzerland)並びに80ngのcDNAにより、次のPCR条件を使用して、ABCA4 c.768G>T変異を保有する線維芽細胞におけるRT-PCRを行った:94℃で3分間、続いて、94℃で30秒間、58℃で30秒間及び72℃で2分間を40サイクルと、72℃で3分間の最終伸長。上に記載されている通りにネステッドPCRを行い、0.5μlのPCR1産物を、次の条件を使用したネステッドPCRのためのPCR鋳型として使用した:94℃で3分間、続いて、94℃で30秒間、58℃で30秒間及び72℃で2分間を35サイクルと、72℃で3分間の最終伸長。全PCR産物は、2.5%アガロースゲルにおいて分離し、選択されたバンドは、サンガー配列決定により確認した。フィジーソフトウェアを使用して、変異体及び野生型バンドの半定量的解析を行った。アクチン(ACTB)を増幅して、ローディング対照とした。プライマー:P49750_Fw_ACTBエクソン3:5’-ACTGGGACGACATGGAGAAG-3’(配列番号91)プライマー:P49751_Rev_ACTBエクソン4:5’-TCTCAGCTGTGGTGGTGAAG-3’(配列番号92)。
HEK293T細胞におけるAONレスキュー
c.768G>T変異(35-nt拡張)に関連するスプライス欠損をレスキューすることができる最も強力なAONを同定するために、「オリゴ-ウォーク」を行い、それによると、代替のスプライス部位(位置c.768+35における)前後の領域を標的とするAON配列は、次から次に1個又は2個のヌクレオチドのみ異なる(図4)。位置c.768にG(WT)又はT(MUT)のいずれかを有するABCA4のゲノム断片を有するミディジーンを、HEK293T細胞へとトランスフェクトし、その後、異なるAONなし又はありでコトランスフェクトした。最初のシリーズにおいて、AONの大部分は、ホスホロチオエート骨格及び2’-O-メチル修飾(2’-OMe)による以前に使用されたケミストリーを含有し、AON35のみが異なり、2’-OMeの代わりに2’-O-メトキシエチル(2’-O-MOE)を有し、AON22と同じ配列を有した。図5から観察できるように、全AONは、ある程度まで、MUTミディジーンをトランスフェクトした場合に観察されるスプライシング欠損を制御することができた。AON14、15、16、19、22及び23は、2’-OMeケミストリーを有する場合に最も強力であると思われた。興味深いことに、近年「発表された」2’-O-MOEケミストリーを有するAON35は、定量化及びサンガー配列決定によって確認される通り、正常なABCA4転写物に向けてスプライシングを制御することを最も可能とした。全実験を2回繰り返して行った。WTミディジーンを有するHEK293T細胞へのAON投与の結果は、いかなるスプライシング欠損も示さなかった(データ図示せず)。
線維芽細胞におけるAONレスキュー
c.768G>T変異が原因の異常ABCA4スプライシングを潜在的に制御することができるAONの数を拡大することにより、多数の興味深い知見が得られた。先ず、「オリゴ-ウォーク」と組み合わせたABCA4ミディジーン系を使用した場合、本出願人らは、ほぼ全てのAONが、ある程度まで、スプライシングを制御することができたが、AON間に有意差が観察されたことを発見した。AON14、15、16、19、22及び23は全て2’-OMeケミストリーを有し、最も効果的であった。興味深いことに、2’-O-MOEケミストリーを有したAON35は、全ての2’-OMeケミストリーよりも優れていた。全て2’-O-MOEケミストリーを有する多数の追加的なAON(AON36~AON40)に関して同様の観察が得られたが、この場合もやはり、AON配列間で異なる有効性を有した。線維芽細胞において、AONは、HEK293T細胞と比較したほど有効ではなく、例えば、HEK293Tミディジーン系においては問題ではない何かしらの、内在性環境におけるABCA4プレmRNAのフォールディングが原因で、これらの細胞によるAONの取込みの有効性が低いこと、及び/又はAONに対する標的転写物の到達性が低いことのいずれかを示唆する。

- 配列番号4に示すヌクレオチド配列を有するポリヌクレオチドに結合する、及び/又は配列番号4に示すヌクレオチド配列を有するポリヌクレオチドと相補的である、スプライシングを制御するためのアンチセンスオリゴヌクレオチドであって、好ましくは、配列番号5を有するポリヌクレオチドに結合するか、又は配列番号5を有するポリヌクレオチドと相補的である、アンチセンスオリゴヌクレオチド、より好ましくは、配列番号80を有するポリヌクレオチドに結合するか、又は配列番号80を有するポリヌクレオチドと相補的である、アンチセンスオリゴヌクレオチド、さらにより好ましくは、配列番号7、8、9;11、12、13;15、16、17;19、20及び21からなる群から選択されるポリヌクレオチドに結合するか、又は配列番号7、8、9;11、12、13;15、16、17;19、20及び21からなる群から選択されるポリヌクレオチドと相補的である、アンチセンスオリゴヌクレオチド。
- 前記アンチセンスオリゴヌクレオチド中のヌクレオチドが、RNA残基、DNA残基、又はヌクレオチドアナログ若しくは等価物であり得る、請求項1に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド。
- 約8~約40ヌクレオチド、好ましくは、約10~約40ヌクレオチド、より好ましくは、約14~約30ヌクレオチド、より好ましくは、16、17、18、19、20、21、22、23又は24ヌクレオチド等の約16~約24ヌクレオチドの長さを有する、請求項1に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド。
- 配列番号34、51、52、53、54、55、56、57、58、59、60、61、73、75又は77を含むか、又は配列番号34、51、52、53、54、55、56、57、58、59、60、61、73、75又は77からなる、請求項1~3のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド。
- 2’-O-メチル修飾されたリボース(RNA)、2’-O-エチル修飾されたリボース、2’-O-メトキシエチル修飾されたリボース、2’-O-プロピル修飾されたリボース及び/又はこれらの修飾を有するハロゲン化された誘導体等の置換された誘導体等の、2’-Oアルキルホスホロチオエートアンチセンスオリゴヌクレオチドを含む、請求項1~3のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド。
- 配列番号34、51、52、53、54、55、56、57、58、59、60、61、73、75又は77を含むか、又は配列番号34、51、52、53、54、55、56、57、58、59、60、61、73、75又は77からなり、2’-O-メチル修飾されたリボース(RNA)又は2’-O-メトキシエチル修飾されたリボース及びホスホロチオエート骨格を含む、請求項1~5のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド。
- 当該分子の発現を促す条件下に置かれると、請求項1~4のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチドを発現する、ウイルスベクター。
- 請求項1~4のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド又は請求項7に記載のウイルスベクター及び薬学的に許容できる賦形剤を含む、医薬組成物。
- 硝子体内投与のためのものであり、0.05mg~5mgの範囲の総アンチセンスオリゴヌクレオチドの量で投与される、請求項8に記載の医薬組成物。
- 硝子体内投与のためのものであり、片眼当たり0.1~1mgの範囲の、スプライシングを制御するための総アンチセンスオリゴヌクレオチド、例えば片眼当たり約0.1、0.2、0.3、0.4、0.5、0.6、0.7、0.8、0.9又は1.0mgのスプライシングを制御するための総アンチセンスオリゴヌクレオチドの量で投与される、請求項8及び9に記載の医薬組成物。
- 医薬としての使用のための、好ましくはABCA4のスプライシングを調整することを必要とするABCA4関連疾患又は状態を処置するための医薬としての使用のための、請求項1~4のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド、請求項7に記載のベクター又は請求項8~10のいずれか一項に記載の医薬組成物。
- ABCA4のスプライシングを調整することを必要とするABCA4関連疾患又は状態を処置するための、請求項1~4のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド、請求項7に記載のベクター又は請求項8~10のいずれか一項に記載の医薬組成物の使用。
- 細胞におけるABCA4のスプライシングを調整するための方法であって、前記細胞を、請求項1~4のいずれか一項に記載のスプライシングを制御するためのアンチセンスオリゴヌクレオチド、請求項7に記載のベクター又は請求項8~10のいずれか一項に記載の医薬組成物と接触させるステップを含む方法。
- 前記ABCA4関連疾患又は状態が、スターガルト病である、請求項11に記載の使用のためのスプライシングを制御するためのアンチセンスオリゴヌクレオチド、請求項12に記載の使用又は請求項13に記載の方法。
