1.材料及び方法
1.1 P-PMOの合成及び調製

Fmoc-β-Ala-OH充填済みWang樹脂(0.19又は0.46 mモル/g、Merck Millipore)を使用し、標準のFmoc化学を適用し、製造者の推奨に従うことによって、Intavis Parallelペプチド・シンセサイザーを使用して、10μモルスケールで、又はCEM Liberty Blue(商標名)ペプチド・シンセサイザー(バッキンガム、英国)を使用して、100μモルスケールで、ペプチドを調製した。Intavis Parallelペプチド・シンセサイザーを使用するケースでは、PyBOP/NMMカップリング混合物とともに、ダブルカップリング工程を使用し、各工程後に、無水酢酸カップリングを行った。CEM Liberty Blueペプチド・シンセサイザーを使用する合成については、アルギニンを除くすべてのアミノ酸について、単一基準カップリングを行ない、ダブルカップリングによって実行した。カップリングは、アルギニン残基を除き、60Wマイクロ波電力において、75℃、5分間で1回行い、それぞれ、二度カップリングした。各脱保護反応を、35Wマイクロ波電力において、75℃で、2回、そのうち1回目は30秒間、ついで、3分間行った。合成が完了した時点で、樹脂をDMFにて洗浄し(3×50ml)、固体相結合ペプチドのN-末端を、DIPEAの存在下、室温において、無水酢酸によってアセチル化した。N-末端のアセチル化の後、ペプチド樹脂を、DMFにて(3×20ml)及びDCMにて(3×20ml)洗浄した。トリフルオロ酢酸(TFA):H2O:トリイソプロピルシラン(TIPS)からなる分裂用カクテル(95%:2.5%:2.5%;3〜10ml)による、室温における、3時間の処理によって、ペプチドを固体支持体から分裂させた。ペプチドの遊離後、過剰のTFAを、窒素でのスパージングによって除去した。粗製のペプチドを、冷たいジエチルエーテルの添加(合成のスケールに応じて15〜40ml)によって沈殿させ、3200 rpmで5分間、遠心分離した。粗製のペプチドペレットを、冷たいジエチルエーテルにて3回洗浄し(3×15ml)、445-LCスケールアップモジュール及び440-LCフラクションコレクターを装着したVarian 940-LC HPLCシステムを使用するRP-HPLCによって精製した。0.1%TFA/H2OにおけるCH3CNの直線勾配を流速15ml/分で使用して、RP-C18カラム(10×250 mm、Phenomenex Jupiter)上でのセミ分取HPLCによって精製した。検出を、220 nm及び260 nmにおいて行った。所望のペプチドを含むフラクションを合わせ、凍結乾燥して、ペプチドを白色の固体として得た(収率については、表2を参照)。
マウスジストトロフィンエクソン-23用の25-merPMOアンチセンス配列(GGCCAAACCTCGGCTTACCTGAAAT(配列番号30))を使用した。ペプチドを、そのC-末端カルボキシル基を介して、PMOの3’-末端に結合させた。これを、DIPEA2.5当量の存在下、NMPにおいて、PyBOP及びHOAtを、それぞれ、2.5及び2当量使用して行い、DMSOに溶解したPMOの2.5倍過剰量のペプチドを使用した。いくつかの実施例では、ペプチドのC-末端カルボキシル基の活性化のため、PyBOPの代わりにHBTU2当量を使用した。一般に、N-メチルピロリドン(NMP、80μl)におけるペプチド(2500 nモル)溶液に、PyBOP(0.3M NMP溶液19.2μl)、HOAt(0.3M NMP溶液16.7μl)、DIPEA(1.0μl)、及びPMO(10mM DMSO溶液100μl)を添加した。混合物を40℃に2.5時間放置し、0.1%TFA水溶液(300μl)を添加することによって、反応をクエンチした。この溶液を、逆Gilson HPLCシステムを使用するイオン交換クロマトグラフィーによって精製した。PMO-ペプチド複合体を、20%CH3CNを含有する硫酸ナトリウム緩衝液(25mM、pH7.0)の直線勾配を使用するイオン交換カラム(Resource S4ml、GE Healthcare)上で精製した。塩化ナトリウム溶液(1M)を使用して、流速4l/分で、複合体をカラムから溶出した。所望の化合物を含むフラクションを合わせ、凍結乾燥して、ペプチド-PMO誘導体を白色の固体として得た。過剰の塩のペプチド-PMO複合体からの除去を、Amicon(登録商標)ultra-15 3K延伸フィルター装置を使用する、イオン交換後に集めたフラクションの濾過を介して行った。複合体を凍結乾燥し、MALDI-TOFによって分析した。複合体を、殺菌した水に溶解し、使用前に、0.22μmの酢酸セルロース膜を通して濾過した。ペプチド−PMOの濃度を、0.1N HCl溶液中、265 nmにおいて、モル吸光によって測定した(収率については、表3参照)。
マウスのH2k筋芽細胞を、ゼラチン(0.01%)被覆フラスコ内において、20%熱不活化ウシ胎児血清(FBS Gold;PAA laboratories)、2%ニワトリ胚抽出物(Seralab)、1%ペニシリン−ストレプトマイシン−ネオマイシン抗生物質混合物(PSN;Gibco)、及び3pg/μl γ-インターフェロン(Roche)を補足したダルベッコイーグル改変培地(DMEM;PAA laboratories)中、33℃において、10%CO2の下で培養した。細胞を、ゼラチン(0.01%)被覆24ウエルプレートに、密度2×105細胞/mlで播州し、33℃において、10%CO2の下で、2日間放置した。筋管に分化させるために、細胞を、さらに、5%ウマ血清(Sigma)及び1%PSNを補足したDMEMにおいて、37℃で、5%CO2の下、2日間生育した。
細胞を、血清フリーのOpti-MEMにおいて調合した、上述のように調製したペプチド−PMOとともにインキュベートし、350μlを各ウエルに正副1組として添加し、37℃で、4時間インキュベートした。ついで、遺伝子導入培地を、5%ウマ血清及び1%PSNを補足したDMEMと交換し、細胞を、37℃で、さらに20時間インキュベートした。細胞をPBSにて洗浄し、TRI RNA(Sigma)単離剤0.5mlを各ウエルに添加した。細胞を−80℃において、1時間凍結した。
1.5 RNAの採取及びネステッドRT-PCR分析
エタノールによる追加の沈殿とともに、TRI試薬を使用して、総細胞内RNAを抽出した。Nanodrop(登録商標)ND-1000(Thermo Scientific)を使用して、精製したRNAを定量した。RNA(400 ng)を、OneStep RT-PCR Kit(Roche、インディアナポリス、米国)を使用するRT-PCR用のテンプレートとして使用した。プライマー配列については表5を参照する。初期逆転写に関するサイクル条件は、1サイクルについて、50℃で30分間及び94℃で7分間、続いて、94℃で20秒間、55℃で40秒間及び68℃で80秒間の30サイクルであった。RT-PCR生成物1μlを、第2のPCR工程用のテンプレートとして使用した。94℃で30秒間、55℃で1分間及び72℃で1分間での25サイクルにおいて、SuperTAQ0.5Uを使用して、増幅を行った。1.5%アガロースゲルを使用する電気泳動によって、生成物を分離した。アガロースゲルの画像を、Molecular Imager ChemiDoc(商標名) XRS+イメージングシステム(BioRad、英国)上で得て、Image Lab(V4.1)を使用して、画像を分析した。エクソンスキッピングのアッセイデータを分析し及びブロットするために、Microsoft Excelを使用し、最後3回の独立した実験からのエクソン-23スキッピングの百分率として表した。
a)ペプチド変異体の合成
製造者の推奨に従いCEM Liberty Blue(商標名)マイクロウエーブ・ペプチド・シンセサイザー(バッキンガム、英国)及びFmoc化学を使用して、100μモルスケールで、ペプチドを合成した。使用した側鎖保護基は、トリフルオロ酢酸処理に対して不安定であり、DIPEAの存在下、PyBOP(5倍過剰)を使用して活性化された5倍過剰量のFmoc-保護アミノ酸(0.25 mモル)を使用して、ペプチドを合成した。N-Fmoc保護基を除去するために、ピペリジン(20%(v/v)DMF溶液)を使用した。アルギニン残基を除き、60Wマイクロ波電力において、78℃、5分間でカップリングを1回行い、2回ずつカップリングした。各脱保護反応を、35Wマイクロ波電力において、75℃で2回、その内1回は30秒間及びついで1回は3分間で行った。合成が完了した時点で、樹脂をDMFにて洗浄し(3×50ml)、固体相結合ペプチドのN-末端を、DIPEAの存在下、室温において、無水酢酸によってアセチル化した。N-末端のアセチル化の後、ペプチド樹脂をDMFにて(3×20ml)及びDCMにて(3×20ml)洗浄した。トリフルオロ酢酸(TFA):H2O:トリイソプロピルシラン(TIPS)からなる分裂用カクテル(95%:2.5%:2.5%;10ml)による、室温における、3時間の処理によって、ペプチドを固体支持体から分裂させた。過剰のTFAを、窒素でのスパージングによって除去した。分裂したペプチドを、氷冷したジエチルエーテルの添加によって沈殿させ、3000 rpmで5分間、遠心分離した。粗製のペプチドペレットを、冷たいジエチルエーテルにて3回洗浄し(3×40ml)、445-LCスケールアップモジュール及び440-LCフラクションコレクターを装着したVarian 940-LC HPLCシステムを使用するRP-HPLCによって精製した。0.1%TFA/H2OにおけるCH3CNの直線勾配を流速15ml/分で使用して、RP-C18カラム(10×250 mm、Phenomenex Jupiter)上でのセミ分取HPLCによって精製した。検出を、220 nm及び260 nmにおいて行った(収率については、表4を参照)。
マウスジストトロフィンエクソン-23用の25-merPMOアンチセンス配列(GGCCAAACCTCGGCTTACCTGAAAT(配列番号30))を使用した。ペプチドを、そのC-末端カルボキシル基を介して、PMOの3’-末端に結合させた。これを、DIPEA2.5当量の存在下、NMPにおいて、PyBOP及びHOAtを、それぞれ、2.5及び2倍当量を使用して行い、DMSOに溶解したPMOの2.5倍過剰量のペプチドを使用した。いくつかの実施例では、ペプチドのC-末端カルボキシル基の活性化のため、PyBOPの代わりにHBTU(2当量)を使用した。一般に、N-メチルピロリドン(NMP、100μl)におけるペプチド(10μモル)溶液に、HBTU(0.3M NMP溶液76.6μl)、HOAt(0.3M NMP溶液66.7μl)、DIPEA(4.0μl)、及びPMO(10mM DMSO溶液400μl)を添加した。混合物を40℃に2時間放置し、0.1%TFA水溶液(1ml)を添加することによって、反応をクエンチした。カチオン交換クロマトグラフィーカラム(Resource S 6mlカラム、GE Healthcare)において、25%アセトニトリルを含有する25mM硫酸ナトリウム緩衝液(pH7.0)を使用して、反応混合物を精製した。塩化ナトリウム溶液(1M)を使用して、流速6ml/分で、複合体をカラムから溶出した。Amicon(登録商標)ultra-15 3K遠心分離フィルター装置を使用して、過剰の塩のペプチド−PMO複合体からの除去を、イオン交換後に集めたフラクションの濾過を介して行った。複合体を凍結乾燥し、MALDI-TOFによって分析した。使用前に、複合体を、殺菌した水に溶解し、0.22μmの酢酸セルロース膜を通して濾過した。ペプチド-PMOの濃度を、0.1N HCl溶液中、265 nmにおいて、複合体のモル吸光によって測定した。全体収率はPMOに基づき29〜46%であった(表5)。
実験を、オックスフォード大学生物化学ユニットにおいて、ホーム・オフィス・プロジェクト・ライセンス認定の下、施設内レビューに従って行った。マウスを最少疾病施設において飼育した。環境的には、温度制御し、12時間毎の明−暗サイクルとした。全ての動物に、自由に、市販の齧歯動物用飼料及び水をとらせた。
実験を、年齢10〜12週の雌mdxマウスにおいて行った。P-PMO10mg/kgの単回の尾静脈注射の前に、mdxマウスを拘束した。注射から1週間後、動物を犠牲にし、TA、心臓及び横隔膜筋を取り出し、ドライアイス冷却イソペンタン内で瞬間凍結させ、−80℃で保存した。
シングル投与後のジストロフィンの修復の期間を査定するために、筋肉(TA及び横隔膜について)の1/3又は厚さ7μmの横断凍結切片(心臓について)90個を、緩衝液300μl(50mM Tris(pH8)、 150 mM NaCl、1%NP40、0.5%デオキシコール酸ナトリウム、10%SDS及びプロテアーゼ/ホスファターゼ阻害剤)中で溶解し、続いて、13000 rpm(Heraeus、#3325B)で10分間遠心分離した。上澄液を集め、100℃で3分間加熱した。タンパク質をBCAによって定量し、タンパク質/サンプル40μgを、既に記載されている(19)ように、NuPAGE3〜8%Tris-Acetateゲル中で分解した。タンパク質を、30Vで1時間、続いて、100 Vで1時間、孔サイズ0.45μmのPVDF膜に移し、既に記載されている(37)ように、モノクロナール抗-ジストロフィン抗体(1:200、NCL-DYS1、Novocastra)及び抗-ビンキュリン抗体(ローディングコントロール、1:100000、hVIN-1、Sigma)にて探索した。第2の抗体(IRDye 800CWヒツジ抗-マウス)を希釈度1:20000(LiCOR)で使用した。
P-PMO処理mdxマウスにおけるジストロフィンの修復レベルを、C57BL/10野生型コントロールマウス(100%と仮定する)との相対比として表示した。このため、P-PMO処置マウスと並行して、5連続C57BL/10タンパク質希釈を含めて、標準曲線を作成した。希釈シリーズは次の通りである:1レーンについて負荷した40μg総タンパク質の75%、40%、15%、5%又は0%は、C57BL/10タンパク質溶解物からのものであり、残りは、未処置mdxタンパク質溶解物からのものである。これらの基準物を定量し、処置したmdxサンプルと並行して、ウエスタンブロットにおいて使用した。全ての標準及び処置サンプルについて、ジストロフィン強度の定量をFluorescence Odysseyイメージングシステムによって行い、全てのサンプルにおいて、ビンキュリン蛍光強度に対する割合を算定することによって正規化した。標準の正規化値を、既知のジストロフィン濃度に対してブロットして、最良適合の数式を得て、P-PMO処置mdxマウスの各サンプルの正規化値を内挿するため、この式を使用した。
ペプチド−PMOにて処置した骨格筋及び心筋組織について、マウスDMD転写からのエクソン23の排除の定量化を行った。簡単には、Trizol系抽出法を使用して、ホモジナイズした組織からRNAを抽出し、ランダムプライマーを使用して、cDNAを合成した。プライマー/プローブをIntegrated DNA Technologiesによって合成し、スキッピングしていない生成物(mDMD23-24、表6参照)を表す、エクソン23-24に及ぶ領域を増幅する、又はエクソン22及び24の境界に及ぶプローブ(mDMD22-24)を使用して、特異的に、転写欠損エクソン23を増幅するようにデザインした。各転写のレベルを、公知の転写量を使用して調製した標準曲線への較正によって測定し、スキッピング率を、[スキップ]/[スキップ+アンスキップ]によって得た。
ここに示す結果は、ここで調製したペプチド−PMO複合体の、細胞内でのエクソンスキッピング活性における明らかな用量応答効果を証明している(図1)。また、これは、DPEP2シリーズのペプチド、すなわち、本発明のペプチドの全てが、細胞における十分な細胞浸透力を有しており、治療用と考えられることを強調している。
ここに示す結果は、さらに、適切な疾患のマウスモデルにおける、本発明のペプチド−PMO複合体のインビボ活性を強調している(図2)。全体として、結果は、本発明のペプチド複合体の活性は、前脛骨筋において最大であり、>横隔膜>心筋であることを示唆している。これらの結果は、本発明のDPEPsが良好なインビボエクソンスキッピング活性を有し、ジストロフィンのインビボ発現の増大を提供することを証明している。さらに、本発明のDPEPsは、同じ複合体において使用される際、少なくとも骨格筋においては、従来の細胞透過性ペプチド、例えば、「PIP」ペプチド及びR6Glyと比べて遜色がない。本発明のペプチド複合体の全てが、同じ複合体で使用される際の公知のR6Glyコンパレーターよりも高い活性を有することは注目に値する。
それ故、本発明のDPEP2ペプチドは、ヒトの神経筋系疾患の治療のための治療用複合体の効力を改善するための有望な細胞透過性ペプチドを提供する。
Claims (25)
- アミノ酸残基40個以下の全長を有するペプチドであって、1個以上のカチオン性ドメイン;及び各々、アミノ酸残基少なくとも3個を含んでなる2個以上の疎水性ドメインを含んでなり、疎水性ドメインの1個は、ペプチドのC-末端に位置し、疎水性ドメインの1個は、ペプチドのN-末端に位置することを特徴とするペプチド。
- 疎水性ドメインの1個は、ペプチドのC-末端を形成し、疎水性ドメインの1個は、ペプチドのN-末端を形成する請求項1に記載のペプチド。
- 各疎水性ドメインは、アミノ酸3〜6個の長さを有し、好ましくは、各疎水性ドメインは、アミノ酸5個の長さを有する請求項1又は2に記載のペプチド。
