Abstract
Description
類鼻疽菌とは、環境中のグラム陰性細菌であり、ヒト疾患である、類鼻疽の原因物質である。鼻疽菌(B.pseudomallei)とは、タイおよびオーストラリア北部などの熱帯地域に特有であり、水田などの湿潤条件と、特に関連する。ヒトの曝露は、皮膚を介する経路を含む経路を介して生じる、例えば、ひっかき傷または他の病変の結果として生じる可能性があり、接種物のサイズに応じて、急性熱病をもたらしうる。細菌はまた、吸気を介して、ヒトに感染する結果として、より急性の肺感染ももたらしうる。
アルファ−ウイルスである、VEEVは、ウマおよびヒトを含む動物において、疾患を引き起こすことが可能であり、蚊およびダニなどの媒介節足動物を介して伝染する。VEEVは、生物兵器物質としての歴史的使用と関連している。
鼻疽菌は、排出ポンプにより、抗生剤を、能動的に排出し、これにより、この細菌を、複数の抗生剤に対して耐性とし、in vivoにおいて、効果的に処置することを、極めて困難とすることが公知である。実際、データは、単一の抗生剤投与レジメンが、鼻疽菌に対して、部分的に効果的であるに過ぎないことを示唆する。類鼻疽に対する有効性を改善するための、1つの選択肢は、組合せ療法の適用、すなわち、急性期のために、β−ラクタムおよびセファロスポリンなど、複数の抗生剤を投与して、重篤な敗血症および死亡を防止し、口腔内根絶期のために、コトリモキサゾールなどの抗生剤と、静菌性の広域スペクトル抗生剤である、クロラムフェニコールおよびドキシサイクリンとの混合物を投与して、残存する細菌を死滅させることである。しかし、このような手法は、このような抗生剤の組合せから生じうる副作用の危険性がある。
現在のところ、VEEVに対する抗ウイルス剤は、認可されていない。ウイルスは、宿主細胞内で複製する、それらの必要のために、細菌の病原体と比較して、処置するのがより困難なことで有名である。一般的なウイルス標的はまた、多種多様なウイルス(例えば、DNAウイルス、RNAウイルス、エンベロープウイルス)のために、同定することも困難であるため、抗ウイルス剤は、特異的なウイルス科またはウイルス属だけに対して有用である傾向がある。宿主特異的応答を、ウイルス感染へとターゲティングすることにより、より広範な手法が達成されうる。しかし、VEEVのための療法を含む、このような療法は、いまだ最適化されていない。
当業者により理解される、「ベシクル」という用語は、医薬剤などの化合物または物質を封入するか、またはこれと会合することが可能な、ナノ粒子状構造またはマイクロ粒子状構造を指す場合がある。医薬剤と、ベシクルとの会合について記載するのに、「封入された」という用語が使用される一方で、医薬剤は、ベシクル表面に結合させられることの結果としてもまた、ベシクルと会合することが想定される。
方法ステップa)およびb)に関して、加熱される成分に有害でない加熱温度が使用されるべきことが理解されるものとする。ステップa)について、ベシクル成分は、約130℃の温度で加熱されうる。しかし、市販の自動式実験室装置(例えば、Precision Nanosystem製のNanoassemblr(登録商標)などのマイクロフルイディクスデバイス)の使用を包摂する方法は、低温、例えば、65℃を利用しうる。低温で行われるステップa)について、ベシクル成分は、有機溶媒、例えば、濃度を100%とするメタノールの存在下で加熱されうる。
好ましくは、ステップb)の方法に関して、加熱温度は50℃〜60℃、特に60℃である。
製剤混合物は、振とうおよび後続のペレット化により加工されうる。例えば、2500rpmで、2分間にわたる、ボルテクシングによる振とう工程が適用されうる。代替的に、プローブ型のホモジナイザーも使用されうる。その後における、製剤混合物の濃縮は、遠心分離により達成されうる。好ましくは、製剤混合物は、例えば、100,000×gで、1時間にわたる、超遠心分離によりペレット化されうる。代替的に、ペレット化は、スピンカラムを使用するサイズ除外濾過などのサイズ除外濾過を介しても達成されうる。スピンカラム法は、自動式実験室装置上で調製されるベシクルに、特に適する。例えば、Nanoassemblr(登録商標)上で調製されたベシクルは、VivaSpinカラムにより、封入されていない薬物から分離されうる。
「二相性医薬ベシクル製剤」という用語は、ある量の医薬剤は、ベシクル内に封入される(すなわち、それらに結合させられ、かつ/またはそれらの中に封入される)が、残りの医薬剤は、ベシクルが懸濁させられる水溶液中に遊離する(すなわち、ベシクルに結合させられず、かつ/またはこれらの中に封入されていない)形でデザインされた製剤を指す。二相性医薬ベシクル製剤は、典型的に、医薬剤が、溶液中に所定の濃度の、同じ医薬剤に結合させられた/この中に封入されたベシクルを再懸濁させることにより調製される。好ましい組合せは、結合/封入抗生剤の、遊離抗生剤に対する50/50比、または平衡製剤もしくは安定製剤を達成するのに必要な比である。
本発明の方法は、それに、えり抜きの医薬剤が結合させられ、かつ/またはこの中に封入される、生体適合性で生体分解性のベシクルの作製を提供し、前記医薬剤を、動物へと送達するためのプラットフォーム技術を提供する。ベシクルの物理的特性は、本発明者らにより、医薬剤の、細胞への取込みを増強する特性;組織ターゲティング薬物送達を達成するための修飾に適する特性、例えば、病原体が常在しうるので、全病原体の除去の一助となるニッチ;バイオアベイラビリティーの増大;合成するのに廉価である特性;および凍結乾燥に適する特性を含む、一連の利点を付与することが示されている。
本発明者らはまた、本発明の方法が、様々な異なる抗微生物剤を、ベシクル内に封入する(すなわち、ベシクルに結合し、かつ/またはこの中に封入する)能力を有することも示した。二相性製剤は、複数の温度および時点の、液体形態および凍結乾燥形態における安定性のために開発され、評価された。例えば、方法を使用する、抗生剤の製剤の最適化は、−20℃で保持された場合の安定性と、最小阻害濃度(MIC)および最小殺菌濃度(MBC)について、非製剤化抗生剤と異ならない値を伴う、in vitroにおける、完全な抗微生物性活性とを保持することが裏付けられている。全ての被験温度および3カ月間にわたり、凍結乾燥状態で保管された、凍結乾燥二相性製剤の安定性は、使用についての現実的な概念を強調する。適合的であり、かつ/または新規である治療用製剤の安定性は、とりわけ、低温保管が、たやすく利用可能ではない、低中所得国(LMIC)へと販売される場合、ますます大きな関心の対象である。
本発明のベシクルは、例えば、使用サイトカインが上昇した状態に対する治療剤としての、空のNISV、または修飾NISVを開示する、PCT/GB96/01861において記載されているNISVである。当業者により理解される通り、このようなベシクルは、界面活性剤すなわち、ミセルなどの凝集物を形成する、水不溶性(または油可溶性)成分および水溶性成分の結果として、溶液中にNISVを形成することが可能な化合物;界面活性剤と混合して、ベシクルの物理的特性が依存する二重層を形成することが可能な疎水性物質;ならびにベシクルに、負の電荷を帯びさせることから、ベシクルを安定化させ、効果的な分散をもたらす、電荷産生性の両親媒性物質を含む。
好ましくは、界面活性剤、疎水性物質および両親媒性物質が、それぞれ5:4:1のモル比で提供される。この比は、本発明の方法において特に良好に作用することが示されている。
ターゲティングされる細胞または組織が必要とされる適用では、ベシクル成分が、in vivoにおける受容体媒介性輸送機構により認識されるリガンドをさらに含むことが好ましい。
「リガンド」という用語は、タンパク質受容体、例えば、宿主細胞受容体などの生体物質に結合することが可能であり、かつ/またはこれらと複合体を形成しうる物質を指す。リガンドの、本発明の方法により、すなわち、医薬ベシクルの形成時における、ベシクル内の封入、および/またはこれへの結合による組込みを介して作製されたベシクルとの会合は、医薬ベシクルの、in vivoにおける、特異的障壁を越える輸送、または例えば、宿主受容体媒介性輸送機構の結果としての、in vivoにおける、特異的なニッチ(例えば、細胞型、組織型、または細胞内の環境)への侵入を容易としうる。
タンパク質に関する記載を通して使用される、「変異体」という用語は、野生型配列、またはそれらが由来する塩基配列と異なる、アミノ酸の配列を指す場合があるが、この場合、変異体は、野生型配列または塩基配列の、所望の特性を保持する。例えば、トランスフェリンに由来する変異体は、元のトランスフェリンタンパク質と比べて、所望されるレベルの、リガンド構造/機能を保持するので、構造的/機能的変異体であると考えられるであろう。当業者により理解される通り、アミノ酸置換は、「保存的」置換、すなわち、アミノ酸を、同様の特性を伴う、別のアミノ酸で置きかえる置換の場合もあり、「非保存的」置換、すなわち、異なる特性を伴う、別のアミノ酸で置きかえる置換の場合もある。当業者によりさらに理解される通り、適切な変異体は、野生型配列または塩基配列と比較して、好ましくは、少なくとも70%同一、例えば、少なくとも75%同一、少なくとも80%同一、少なくとも85%同一、少なくとも90%同一であり、好ましくは、少なくとも95%同一である。
好ましくは、医薬剤−担体混合物とベシクル成分とは、それぞれ3:1の比で提供される。本発明者らは、この比が、本発明、特にBBBを越えるように適合したベシクルに特に適することを示した。
本発明者らは、NISVおよびこれらの変異体(ビロソーム;ブレインソーム)の作製を自動化し、迅速にスケールアップする能力の、市販のNanoAssemblr(登録商標)装置における裏付けに成功したことを示した。gNISVおよびtNISVのいずれも、製剤の自動化および迅速なスケールアップを可能とするNanoAssemblr(登録商標)を使用して作出された。例えば、ある範囲の抗ウイルス剤(例えば、IFN−α2βなどのインターフェロン)をロードされたブレインソームを、迅速に作製する、NanoAssemblr(登録商標)の、さらなる有用性は、ナノベシクルを、多目的的で魅力的な薬物送達技術として際立たせる。加えて、Nanoassemblr(登録商標)による、ブレインソームおよびビロソームの作製を、作出されるバッチの、サイズおよびローディングの点における特徴付けを伴い、成功裏にスケールアップし、自動化するための作業も企図されている。
好ましくは、医薬剤は、抗菌剤および/または抗ウイルス剤である。
本発明者らは、二相性のNISV製剤およびビロソーム製剤が、MICアッセイおよびMBCアッセイにおいて、抗微生物性活性を維持することが示されることを示すことに成功した。本発明者らにより企図された各アッセイでは、ビロソーム製剤化抗生剤は、非製剤化抗生剤と同等の抗微生物性活性を及ぼしたことから、製剤化の工程が、被験抗生剤に有害な作用を及ぼさなかったことを指し示す。さらに、報告されたデータは、ビロソームによる抗生剤の送達は、有意な生存上の利点をもたらすだけでなく、また、抗生剤の反復投与および感染と関連する、体重の減少という、重篤な副作用も低減することを示す。
好ましくは、抗菌剤は、フルオロキノロンおよびテトラサイクリンを含む群から選択される。
NISV製剤技術およびビロソーム製剤技術は、少なくとも3つの抗生剤(ドキシサイクリン、レボフロキサシン、シプロフロキサシン)の、NISVおよびビロソーム内への封入の成功へと適合させられている。成分(モノパルミトイルグリセロール、リン酸ジセチル、およびコレステロール)の混合物が、迅速に加熱され、選び出された抗生剤が添加され、次いで、抗生剤−ベシクル製剤を形成するように加工される。ビロソーム製剤は、加熱段階後において、抗生剤の溶解と同時に、胆汁酸塩を添加することにより調製される。製剤が伴う抗生剤濃度は、1〜100mg/mlでありうる。
好ましくは、抗ウイルス剤は、IFN−γ、IFN−α、カルボジン、DZ−13(Xie et al. Regulatory roles of c-jun in H5N1 influenza virus replication and host inflammation. Biochimica et Biophysica Acta. 1842(2014), 2479-88)、および抗ウイルス抗体、またはこれらの任意の変異体もしくは断片を含む群から選択される。
本発明者らにより開発されたブレインソームは、大型の抗体(VEEV Hu1A3B−7を含む)および/またはサイトカイン(例えば、IFNα2β)を封入することが可能であると示されているので有利である。さらに、gNISVは、in vivoにおける、医薬剤(例えば、IgG Ab)の、脳への送達を、遊離抗体と比較して増強することが示されている。
本出願者らは、NISVおよびgNISVによる、Hu1A3B−7 VEEV mAbの送達が、遊離mAbと比較して有利であることを示した。いずれのプラットフォームも、肺内のウイルス負荷を低減し、生存を延長し、死亡率を低減する。gNISVプラットフォームは、脳内のウイルス負荷を、著明に低減する。
本発明はまた、第1の態様に従い調製される、医薬ベシクル製剤も提供する。
第2の態様に従い、本発明は、a)モノパルミトイルグリセロール、コレステロールおよびリン酸ジセチルを含むベシクル;b)医薬剤;ならびにc)薬学的に許容可能な担体を含む、医薬ベシクル製剤を提供する。
好ましくは、医薬ベシクル製剤は、in vivoにおける受容体媒介性輸送機構により認識されるリガンドをさらに含む。

抗生剤である、レボフロキサシン≧98.0%(HPLC)、ドキシサイクリンハイクレート≧98.0%(HPLC)、およびシプロフロキサシン(Sigma−Aldrich)は、乾燥粉末形態で得、全ての研究で使用した。ビロソーム製剤を作り、非製剤化薬物として投与するために、抗生剤をPBS中に所定の濃度で懸濁させた。
ヒト化抗VEEVモノクローナル抗体は、既に記載されている通りに作製した(S.A. Goodchild, L.M. O’Brien, J. Steven, M.R. Muller, O.J. Lanning, C.H, Logue, R.V. D’Elia, R.J. Phillpotts, S.D. Perkins. A humanised murine monoclonal antibody with broad serogroup specificity protects mice from challenge with Venezuelan equine encephalitis virus. Antiviral Res., 90(2011), 1-8)。略述すると、Hu1A3B−7を発現させるベクターをトランスフェクトしたCHO DG44細胞を、10%(容量/容量)FBS、1%抗真菌剤、25mgのゲンタマイシン、1mMのピルビン酸ナトリウム、2mMのグルタミン、1%(容量/容量)ペニシリン/ストレプトマイシン、1%(容量/容量)の非必須アミノ酸および1%(容量/容量)の必須アミノ酸(Gibco)、ならびに10mMのメトトレキサート(Sigma)を補充した、IMDM培地中で培養した。Prosep−A(Millipore)を使用する、プロテインAアフィニティークロマトグラフィーを使用して、ヒト化抗体を精製し、PBSに透析した。
類鼻疽菌の臨床分離株である、K96423を、in vitro研究およびin vivo研究のために使用した。細菌をエアロゾル抗原投与のために、回転プラットフォーム上、37℃のルリア培養液中で増殖させ、L−寒天プレート上で数え上げた。
野兎病菌株であるLVSは、DSTLの培養物コレクション内、−20℃で保管されたワクチンアンプルから導出した。細菌を、補充BCGA(blood cysteine glucose agar)上、37℃で一晩にわたり培養し、PBSに採取し、0.15〜0.20の間のOD600(1mL当たり1×109CFU)を得るように希釈した。抗原投与のための細菌数は、試料の系列希釈(1:10)において寒天上で決定した。
組織培養物:L929細胞系(マウス線維芽細胞)およびVero(サル腎臓)細胞系(European Collection of Animal Cell Cultures、UK)を、推奨される培養培地を使用する、標準的方法により繁殖させた。実験の目的で、細胞を、2%(容量/容量)のウシ胎仔血清、2mMのl−グルタミン、50U/mlのペニシリン、および50μg/mlのストレプトマイシン(L15MM)を補充した、ライボビッツL−15培地中で維持した。培地および補充物質は全て、Sigma−Aldrich(UK)により供給された。
ウイルスの滴定:ウイルスの力価は、Vero細胞内の、1.5%(質量/容量)のカルボキシメチルセルロースオーバーレイ下におけるプラーク形成により決定した。略述すると、細胞を、24ウェルプレートに播種し(ウェル1つ当たりの細胞1×105個)、一晩にわたりインキュベートした。培地を除去し、細胞に、L15MM中で希釈されたウイルス100μlをオーバーレイした。室温で30分間にわたるインキュベーションの後、3%(質量/容量)のカルボキシメチルセルロース中で希釈された、2倍濃度のL15MM 1mlを、各ウェルに、注意深く添加した。細胞を、72時間にわたりインキュベートし、10%(容量/容量)のホルマール生理食塩液で、一晩にわたり固定し、次いで、0.1%(質量/容量)のクリスタルバイオレットで染色した。プラークをカウントし、ウイルスの量を計算した。
略述すると、1−モノパルミトイルグリセロール、コレステロール、およびリン酸ジセチルを、5:4:1のモル比で組み合わせ、130℃に加熱した。NISVを調製するために、PBS中に30mg/mlの、関与性の抗生剤を添加した。ビロソームのために、0.025Mの炭酸緩衝液中に、デオキシコール酸ナトリウムを伴う、30mg/mlの関与性の抗生剤を添加した。ベシクル成分への添加の前に、抗生剤を60℃にあらかじめ加熱した。その後、調製物を2分間にわたり十分にボルテクシングした。抗生剤は、ベシクルの形成に応じて封入されるか、または5サイクルにわたる凍結−融解の後で封入された。封入されなかった抗生剤は、遠心分離を介して除去し、ペレット化させ、ベシクルを単相性調製物に適切な緩衝液中、または二相性調製物をもたらすように、ある量の適切な抗生剤を、ベシクル内に封入された抗生剤と同等な濃度(すなわち、50/50の封入調製物:遊離調製物)で含有する、適切な緩衝液中に再懸濁させた。
86.3mgのグルコサミンを93μlのトリエタノールアミンおよび15mlのジメチルスルホキシド中で溶解させた。次いで、この混合物を283mgのパルミチン酸N−ヒドロキシスクシンイミドに添加し、4mlのクロロホルム中で溶解させた。溶液を暗所内、室温で48時間にわたり攪拌した。ミックスを冷却し、氷冷水を添加することにより、パルミトイル化グルコサミンを析出させた。次いで、ミックスを真空フィルターに適用して、析出物を分離した。個別の水、クロロホルム、およびエタノールによる洗浄をパルミトイル化グルコサミンに適用してから、40℃で、48時間にわたり乾燥させた。
トランスフェリンを10倍モル過剰量のトラウト試薬と、25℃で1時間にわたり混合することにより、ヒトホロトランスフェリンをチオールで修飾した。過剰量のトラウト試薬は、分子量カットオフを5kDaとするVivaspin 6カラムを使用して除去した。
ベシクルは、NanoAssemblr(商標)benchtop(Precision Nanosytems)を、60℃の温度で保持された、300μmのSHM(staggered herringbone micromixer)チップと共に使用して調製した。ベシクル成分:パルミチン、コレステロール、およびリン酸ジセチルを、メタノール(menthol)中に、5.62mg/mlの総濃度、5:4:1の比で可溶化させ、60℃に加熱した。抗体(ウサギIgG、mAbである、Hu1A3B−7、またはヒトIgG−FITC)を、PBS中に、2mg/mlで可溶化させた。水流と、溶媒流とを、12ml/分の組合せ流量、3:1の比で混合した。薬物のカプセル化と、ベシクルの製剤化は、マイクロフルイディクスチップ内で、同時に生じる。メタノールおよび封入されなかった薬物は、分子量カットオフを300kDaとする、Vivaspin20(Sartorious)スピンカラムを使用して除去した。封入された抗体の濃度は、標準的なブラッドフォードアッセイを使用して決定した。gNISVは、6.4mgのパルミトイル化グルコサミンを、メタノール中に溶解させたベシクル成分に添加して、同様に調製した。tNISVは、PEG−マレイミドを、NISVミックス中に組み入れることにより合成した。メタノールおよび封入されなかったカーゴを除去した後、ベシクルを、チオール化トランスフェリンと共に、25℃で、1時間にわたりインキュベートした。次いで、過剰量のトランスフェリンは、分子量カットオフを100kDaとする、Vivaspin20カラムを使用して除去した。
スクロースをNISV調製物に100mMの濃度まで添加した。24時間後、Christ Epsilon 2−4LD Freeze Dryer内で凍結乾燥させる前に、調製物を−80℃で静置し、復水器温度を−70℃として、−40℃で48時間にわたり乾燥させた。
ビロソームの、生物物理的特徴付け
ビロソーム調製物の大型バッチを凍結乾燥および保管のために製剤化およびアリコート分割した。アリコートを要求される通りに回収し、逆浸透水中に再懸濁させるのに続き、十分に振とうした。ベシクルサイズおよびゼータポテンシャルは、Malvern Zeta−sizer(Zetasizer 30000HS、Malvern Instruments Ltd.、UK)を使用して決定した。遠心分離によりベシクルをペレット化させ、HPLCを使用して、封入された抗生剤含量を決定した。
HPLC解析は、50℃で維持されたC18カラム(150mm×4.6mm、5μm)を使用して、Agilent1290 Infinity Series HPLC上で実行した。抗生剤(ドキシサイクリン、レボフロキサシンおよびシプロフロキサシン)のための移動相は、H3PO4およびアセトニトリルまたはメタノールにより、pH2とした、0.02MのNa2HPO4からなった。
安定性研究
NISV調製物およびビロソーム調製物の各々の大型バッチを製剤化し、アリコートを−20℃、4℃、室温、および37℃で、液体製剤または凍結乾燥調製物として静置した。アリコートを多様な時点において回収し、凍結乾燥調製物を逆浸透水中に再懸濁させるのに続き、十分に振とうした。ベシクルサイズおよびゼータポテンシャルは、Malvern Zeta−sizer(Zetasizer 30000HS、Malvern Instruments Ltd.、UK)を使用して決定した。遠心分離により、ベシクルをペレット化させ、HPLCを使用して、封入された抗生剤含量を決定した。
PK/PD研究
NISV製剤化抗生剤およびビロソーム製剤化抗生剤(50〜100mg/kg)の凍結乾燥調製物を、逆浸透水中に再懸濁させ、腹腔内経路または強制経口投与により、動物に投与した。他のマウス群は、同一濃度の抗生剤を、溶液として施された。初期実験では、血液を、24時間後までの時点において、浅静脈切開から得た。後続の実験では、マウスを終末麻酔により屠殺し、血液を心臓および肺から回収し、肝臓および脾臓を摘出した。各組織を秤量し、ホモジナイゼーションの前に、4倍容量の生理食塩液を添加した。試料を除タンパク質処理し、0.2μmのPFTEフィルターに通した。
抗生剤製剤についてのMICは、Clinical Laboratory Standards Institute(CLSI)ガイドラインに従い、培養液微量希釈法を使用して、鼻疽菌株であるK96243、および野兎病菌であるSchuS4について決定した。アッセイは、96ウェルマイクロ滴定プレート内のアイソセンシテスト培養液(BSAC)もしくはミュラーヒントン培養液(CLSI)、または病原体の要求に適する培養液中の抗生剤濃度を64mg/L〜0.03mg/Lの範囲とし、細菌の最終濃度を1mL当たり約5×105CFUとして実施した。37℃で、24時間にわたるインキュベーション後、プレートの光学濃度(OD)を自動式プレートリーダー内、590nmの波長で読み取った。MICは、細菌の増殖のうちの>80%を阻害する濃度として決定した。
抗生剤製剤についてのMBCは、増殖を示さない、100μLのMIC希釈液アリコートをL−寒天プレートまたはBCGAプレートに三連で播種し、37℃で48時間にわたりインキュベートすることにより決定した。MBCは、元の接種物中の細菌のうちの99.9%を死滅させる抗生剤の最低濃度として記録した。
ハチノスツヅリガ感染
ハチノスツヅリガ(G.mellonella)の幼虫を暗所で保管した。全てのアッセイで体重0.2〜0.3gの幼虫を利用した。10mlのハミルトンシリンジを使用して、左最後部腹脚を介して、接種物(幼虫1匹当たり3×106CFUの野兎病菌LVS)のアリコート10mlを、各幼虫の血体腔に注射した。感染の後、幼虫をプラスチック製容器内でインキュベートした。10mlのハミルトンシリンジはまた、感染の24時間後において、抗微生物剤(レボフロキサシン15mg/kg)および対照(PBS)を注射するのにも使用した。幼虫は、接触に応答した動きを提示しなくなったら、死んだと考えた。
BALB/cマウスをマウス12匹ずつの群に割り付け、1〜7日目に経口経路(10mg/mlの濃度で、毎日0.1ml)を介してPBS、空のビロソーム、空のNISV、抗生剤、NISV−抗生剤、またはビロソーム−抗生剤で処置した。糞便試料を処置の3日前、最終投与の24時間後(8日目)、処置の停止の14日後(22日目)、および28日後(36日目)に回収した。動物の体重および状態を、毎日記録した。8日目に、各群内の半分のマウスを屠殺し、血清、小腸、および大腸を回収した。血清試料は、−80℃で凍結させて保管し、大腸および小腸は、内容物を除去し、渦巻き状に巻き、10%ホルマリン中で固定した。残りのマウスは、さらに28日間にわたり、秤量および観察してから、8日目に屠殺したマウスについて記載した通りに、屠殺し、血清および組織を採取した。
製造元の指示書に従い、PowerSoil(登録商標)DNA単離キット(Qiagen)を使用して、ゲノムDNAを糞便試料から単離した。推奨される、250mgの土壌に対する代替物として、細胞の溶解を経るように、約200mgの糞便試料をPowerBeadチューブに添加した。50uLのC6液を使用して、精製DNAをスピンフィルターから溶出させ、PCR増幅まで、−20℃で保管した。
16Sのユニバーサル真正細菌プライマーである、515F GTGCCAGCMGCCGCGGTAAおよび806R GGACTACHVGGGTWTCTAATを利用して、bTEFAP(登録商標)DNA解析サービスを介する方法により、各試料の微生物環境を、HiSeq 2500上で査定した。各試料は、HotStarTaq Plus Master Mix Kit(Qiagen、Valencia、CA)を、以下の条件:94℃で3分間に続き、94℃で30秒間;53℃で40秒間;および72℃で1分間の28サイクル下で使用する、単一ステップの、30サイクルのPCRを受け;この後、72℃で5分間にわたる、最終的な伸長ステップを実施した。PCRの後、異なる試料からの、全ての単位複製配列産物を、等濃度で混合し、Agencourt Ampure beads(Agencourt Bioscience Corporation、MA、USA)を使用して精製した。製造元のプロトコールに従い、Illumina HiSeq化学反応を利用して、試料をシーケンシングした。
骨髄細胞は、8週齢の雄BALB/cマウスの大腿骨から得、GM−CSFエンリッチ完全DMEM(DMEM10%ウシ胎仔血清、100U/mlのペニシリン、100μg/mlストレプトマイシン、および2mMのL−グルタミン)を使用して、樹状細胞に分化させた。これは、規定的に、80%のCD11c+細胞を結果としてもたらす。次いで、BMDCを、24ウェル組織培養プレート上、完全RPMI(RPMI1640、10%のウシ胎仔血清、100U/mlのペニシリン、100μg/mlのストレプトマイシン、および2mMのL−グルタミン)中に、ウェル1つ当たりの細胞1×106個で播種し、LPS(3μg/ml)で刺激するか、または対照として、非刺激のまま放置した。同時に、細胞を、溶融法により作製されたNISV、もしくは超音波処理法により作製されたNISVで処置するか、非処置のまま放置した。24時間後に、細胞を採取し、フローサイトメトリーによる解析の前に、抗CD40−APC、抗CD80−PerCP、および抗CD86−PE−Cy7の1/100希釈液で染色した。補償電圧および光電子倍増管電圧は、単一染色対照を使用して設定した。FMO(fluorescence minus one)対照は、適正なゲーティング戦略を同定するために使用し、陰性対照を作製するために使用した。
6〜8週齢の雌BALB/cマウス(Charles River、UK)を、クラスIII高格納精度の堅固なアイソレーターに移し、そこで、不断の給餌および給水を施し、少なくとも5日間にわたり馴致させた。マウスを、処置群(群1つ当たり15匹)に割り付け、5つのケージで飼育した。コンピュータ制御型送達プラットフォーム(Biaera Technologies)を使用して、マウスに、鼻腔限定曝露システムにより、エアロゾル経路を介して、50〜100CFU(10MLD)の、鼻疽菌であるK96243、または野兎病菌であるSchuS4を抗原投与した。抗生剤投与を、感染の6時間後に開始する、最適用量未満の治療研究デザインを使用し、毎日1回7日間だけにわたり投与して、治療有効性について調べた。抗生剤であるレボフロキサシンおよびドキシサイクリンを、50mg/kgで、毎日、経口経路により送達し、処置を、7日間にわたり継続した。処置群は、ビロソーム送達型抗生剤または非製剤化抗生剤を含んだ。処置群1つ当たりのマウス5匹ずつの亜群を、感染後3日目に殺処分して、肺、脾臓、および肝臓における細菌負荷を決定した。Biaearaエアロゾルデバイスを使用して、マウスに、エアロゾルによるVEEV Trinidad Donkey Strainを抗原投与した。マウスを、毎日2回ずつチェックし、臨床症状について評定し、マウスを、毎日秤量した。所定の客観的臨床徴候のセットに基づき、人道的限界に達したマウスは、速やかに殺処分した。異なる時点において、一部のマウスについては、生存時間を記録し、他のマウスは、組織を解析するために殺処分した。全ての手順および飼育は、UK Animal(Scientific Procedures)Act(1986)に準拠した。
マウス組織(脳、肺、肝臓、および脾臓)内に存在するVEEV株であるTrDの量は、Vero細胞内の滴定により決定した。組織を除去し、70μmのナイロン製の細胞ストレイナー(BD Falcon)に通すことにより、2mlのL15MM中でホモジナイズした。次いで、細胞懸濁液を、L15MM中で、系列希釈(1:10)した。希釈されたホモジネート(100μl)を、単層コンフルエントのVero細胞を含有する、24ウェルプレートのウェルに添加した。細胞を、72時間にわたりインキュベートし、この時間の後、10%(容量/容量)のホルマール生理食塩液を添加することにより、単層を固定し、0.1%(重量/容量)のクリスタルバイオレットで染色した。
サイトカイン解析のために、細胞懸濁液の200mlアリコートを、2000rpmで、5分間にわたり遠心分離した。サイトカイン解析のために、上清を除去し、−80℃で保管した。サイトカインのレベルは、製造元の指示書に従い使用される、23プレックスマウスLuminexアレイ(Bio−Rad)を介して測定した。加えて、洗浄ステップのために、磁気プレート洗浄機(Bio−Rad)を使用し、最終的に、PBS中に4%のパラホルムアルデヒドを、4℃で、少なくとも24時間にわたり使用して、試料を固定した。
ホルマリン固定パラフィン包埋(FFPE)組織試料を使用して、組織中のウサギIgGを位置特定するプロトコールを標準化した。キシレンならびにエタノールおよび水の段階系列を使用し、最後に、TBSで処理して、組織切片を、脱パラフィン化および再水和させた。プロテイナーゼKによる酵素的消化(DAKO)、ならびにpH6.0およびpH9.0の緩衝液(DAKO)による高温抗原賦活化(HTAR)を含む、いくつかの方法を使用して、4μmの組織切片内の抗原を賦活化させた。プロテイナーゼK前処理により、最良の結果が得られた。酵素的消化を停止させた(TBS中で3回の洗浄)のに続き、ポリクローナルヤギ抗ウサギIgG Alexa568標識抗体を伴うインキュベーション(室温および1:100の希釈率で30分間にわたる)を行った。TBSにより3回にわたる洗浄の後、Vectashield antifade hardset封入剤(Vector laboratories)を使用して、スライドに封入し、Nikon Ni−SE顕微鏡内の蛍光下で研究した。
同様に、同じ抗原賦活化法(プロテイナーゼK)を使用して、光学顕微鏡による、ウサギIgGの免疫組織学的検出のためにも、組織切片を使用した。脱ろう/再水和プロトコールは、メタノール中の過酸化水素溶液を使用して、内因性ペルオキシダーゼをクエンチングするステップを含んだ。ビオチニル化ヤギ抗ウサギIgGポリクローナル抗体(Pierce)(室温で、1:200に希釈して、30分間にわたる)に続き、アビジン−ビオチンペルオキシダーゼ複合体反応(Pierce)を行った。NOVARED(Vector)を使用して、反応を生じさせた。
デジタル画像解析のために、FFPE切片を、免疫組織化学で染色した。画像解析ソフトウェア(Nikon Br−NIS)を使用して、平均値強度を計算した。最大強度を100とする。
プログラムである、SPSS V21.0(IBM)またはGraphpad PRISM V6.0を使用して、様々な統計学的解析を実施した。Graphpad PRISM V6.0を使用して、グラフを構築した。ログランク検定を使用して、生存データを比較した。連続データは、条件が満たされる場合(ガウス分布、および不等分散のためのルビーン/バートレット検定について評価するQQプロット)は、パラメトリック解析(ANOVA、T検定、GLM)により解析し、これらの基準が満たされない場合は、ノンパラメトリック検定(クラスカル−ワリス、マン−ホイットニー、ムード)により解析した。場合によって、対数変換などの変換を使用して、パラメトリック基準に達することも可能であった。二値データについては、分割表を使用した。ファミリーワイズエラーのための多重検定補正は、解析との個別の比較時に実施した。これらは、ボンフェローニ補正およびダン補正を含んだ。
カプセル化の効率および安定性についての評価
カプセル化の効率は、遠心分離により、封入されなかった抗生剤を除去し、HPLCを介する、速やかな解析のために、ペレットを、適切な緩衝液中に再懸濁させることにより決定した。生成された値は、ベシクルに封入された抗生剤の量を表す。定量のために、HPLCを使用して、単相性製剤および二相性製剤の両方を、時間経過にわたる、抗生剤の、各製剤型中の貯留により決定される安定性について調べた。NISV試料およびビロソーム試料の、ゼータポテンシャルおよびサイズもまた測定した。

抗生剤の、NISVまたはビロソームによる製剤が、抗微生物機能に有害な影響を及ぼす可能性を除外するために、in vitroにおいて、製剤の、野兎病菌および鼻疽菌に対するMICおよびMBCについて調べた。これらのアッセイの各々では、NISV製剤化抗生剤およびビロソーム製剤化抗生剤は、非製剤化抗生剤と同等の抗微生物性活性を及ぼした(表3)ことから、製剤化の工程が、抗生剤カーゴに、有害な影響を及ぼさなかったことを指し示す。

レボフロキサシンを組み込むNISVおよびビロソームを、in vivoにおける、PK/PD解析のための、見本製剤として選択した。時間経過研究では、雌Balb/cマウス(群1つ当たりのn=5)に、それぞれ、腹腔内経路または経口投与経路により、NISVまたはビロソームを投与し、血液および組織を、24時間中に複数の間隔で回収した。血液試料および組織試料を、HPLCによる抗生剤濃度についての解析のために加工した。二相性のNISV内またはビロソーム内で製剤化され、凍結乾燥したレボフロキサシン、または遊離レボフロキサシンについての、Cmax(投与間隔中の最大薬物濃度)、Tmax(Cmが観察される時点)、および曲線下面積(AUC、すなわち、総薬物濃度−時間経過曲線下面積)の計量を、血液(パネル1)、肝臓(パネル2)、または脾臓(パネル3)について、表4に示す。ビロソーム内で経口投与されたレボフロキサシンは、肝臓貯留についてのAUC値を、2倍に増大させ、最高の効能を裏付けた。

Balb/cマウスに、14日間にわたり、毎日施される、シプロフロキサシン製剤、レボフロキサシン製剤、およびドキシサイクリン製剤の忍容性を査定する研究を実施した。120匹の雌Balb/cマウスを、マウス10匹ずつ、12の群に割り付け、1〜14日目において、方法で示した通りに処置した。動物の体重および状態を、毎日記録した。この研究中に、マウスは死ななかった。しかし、処置経路間および抗生剤製剤間で、体重の変化が見られた(図3に示される結果)。
生理食塩液中またはビロソーム内に製剤化された、異なる抗生剤による処置の、体重の増加、ミクロビオーム組成物、小腸組織学、および血清セロトニンレベルに対する効果を査定した。レボフロキサシン、ドキシサイクリン、またはシプロフロキサシンを、経口において、7日間にわたり施し、半分のマウスを、8日目に屠殺して、処置の直接的な影響を査定した。糞便ペレットを、8日目に屠殺したマウスから回収した。残りのマウスは、処置の停止後、28日間にわたりモニタリングし、22日目に、糞便ペレットを回収し、36日目に、終了時の試料を回収した。
体重の増加において見られる有意差を査定するために、各動物の、百分率による体重変化についての曲線下面積(AUC)を計算し、一元ANOVA検定を使用して、群を比較した。この解析では、データを、8日目の、投与期間の終了時まで査定し、屠殺された動物については、36日目に査定した。8日目までのデータについての解析では、ANOVA検定は、群間で有意差が見られることを指し示し(p=0.0001)、36日目までのデータでもまた、ANOVA検定は、有意差を指し示した(p=0.0020)。データに対して群間比較を実施して、単一変数の影響を査定した。この解析が、PBS中のシプロフロキサシンにより処置された群と、シプロフロキサシンビロソームにより処置された群との間で、統計学的有意差が見られることを指し示した(p=0.0152)ことは重要である。同様の正の傾向は、PBS中のレボフロキサシンを、レボフロキサシンビロソームと対比して比較する場合、およびPBSを、空のビロソームと対比して比較する場合にも見られた。
ビロソームが、抗生剤誘導性の体重減少を改善する能力について探索するために、マウスのミクロビオームを、遊離抗生剤またはビロソームカプセル化抗生剤による処置後36日間にわたりモニタリングした。マウスに抗生剤を投与し、ミクロビオームを、次世代シーケンシングにより特徴付けした。属データを、それらが属する、具体的な門に類別し、各処置群についての、百分率による存在度を計算した。データは、面グラフ/積み重ね棒グラフ上でグラフ化した。
結果は、ビロソームが、ミクロビオームに対して、有害作用を及ぼさなかったことを裏付ける(図8)。一般に、抗生剤処置の効果は、8日目に、グラム陰性菌である、バクテロイデス門(Bacterioidetes)を低減し、グラム陽性菌である、フィルミクテス門(Firmicutes)を増大させ、36日目までに、処置前の比への、漸進的な回復をもたらす。しかし、これらの変化は、ビロソーム処置群内で、少量のウェルコミクロビウム門(Verucomicrobia)種の出現を伴ったが、これは、ミクロビオーム内で、有益な効果を及ぼすと考えられる。
したがって、全体として、ミクロビオーム解析は、ビロソームプラットフォームが、in vivoにおいて、忍容良好であり、抗生剤による撹乱後の腸内において、バクテロイデス門の回復、および有益な細菌(ウェルコミクロビウム門)の増殖を促しうることを裏付けた。
骨髄細胞を、8週齢の雄BALB/cマウスの大腿骨から得、GM−CSFエンリッチ培地を使用して、骨髄由来樹状細胞(BMDC)に分化させた。これは、規定通りに、80%のCD11c+細胞を、結果としてもたらした。BMDCを、96ウェル組織培養マイクロ滴定プレート上に、ウェル1つ当たりの細胞5×105個で播種し、LPS(3ug/ml)で刺激するか、または対照として、非刺激のまま放置した。同時に、細胞を、溶融法により作製されたNISV、もしくは超音波処理法により作製されたNISVで処置するか、非処置のまま放置した。24時間後に、細胞を採取し、フローサイトメトリーによる解析の前に、CD40、CD80、またはCD86に対する抗体で染色した。いずれかのNISV製剤で処置された、LPS非刺激細胞内では、CD40、CD80、またはCD86のレベルは、影響を受けなかった(図9;*p<0.05、**p<0.01)。しかし、いずれかのNISV製剤で処置された、LPS刺激細胞内では、CD40およびCD80のレベルは、下方調節された(図9)。これに対し、いずれかのNISV製剤で処置された、LPS刺激細胞内では、CD86レベルが上昇したが、これは、骨髄性DCの成熟に典型的である(図9)。
抗生剤投与を、感染の6時間後に開始する、最適用量未満の治療研究デザインを使用して、類鼻疽のエアロゾルモデルにおいてレボフロキサシンおよびドキシサイクリンのビロソーム製剤の有効性について調べ、毎日1回7日間だけにわたり経口経路を介して投与して、治療有効性について調べた。処置群は、ビロソーム送達型抗生剤または非製剤化抗生剤を含み、対照群は、PBSまたは空のビロソームを施された。正確に同じデザイン、条件、および同一な処置群により、この研究を、2回にわたり反復し、解析のために、データを、層別化し、組み合わせて、ビロソームによるカプセル化についての、遊離レボフロキサシンと対比した、生存に対する有意な利点(p=0.014)(図10)、およびビロソームによるカプセル化についての、遊離ドキシサイクリンと対比した、生存に対する有意な利点(p<0.001)(図11)を示した。グラフ上に表されたデータは、群1つ当たりのマウスを10匹とする、カプランマイヤープロットである。いずれの図も、実験1(左上パネル)、実験2(右上パネル)、および組合せ(下パネル)の層別化データの結果を示す。PBS処置群および空のビロソーム処置群を、対照として組み入れた。
ビロソームによる抗生剤送達の有益な効果についての他の証拠は、ビロソーム封入レボフロキサシンを投与されたマウスが、一部の、鍵となる炎症性サイトカインのレベルを、非製剤化レボフロキサシンを施されるマウスと比較して、有意に(p<0.05:肺におけるIL−6、脾臓におけるIL−13、肝臓におけるエオタキシン、肺および肝臓におけるGM−CSF、肝臓におけるMIP1b)低減した、野兎病菌研究から導出された。バークホルデリア属(Burkholderia)感染マウスにおけるサイトカインレベルの解析も、同様の傾向を示した。
ある範囲の抗ウイルス剤の、改変NISV内の封入
脳炎ウイルス(例えば、VEEV)に効果的な治療を開発するために、NISVを、それらの血液脳関門(BBB)を越える通過を容易とするように改変した。この手法は、パルミトイル化グルコサミンでコーティングするか、またはNISV製剤内のコレステロール部分をトランスフェリンに共有結合的に連結して、それぞれ、gNISVおよびtNISV(「ブレインソーム」)を創出することにより達成した。in vivoにおける、Glut−1受容体(BBBの内皮組織内でよく見られる)への結合を目的として、パルミトイル化グルコサミンを、gNISVに組み込んだ。同様に、ヒトホロトランスフェリンのチオール化により、tNISVを調製して、NISV内に含まれる、通常のコレステロールのうちの一部(29%)を置換した、マレイミド修飾コレステロール成分への結合を可能とした。
ナノベシクルの作製(ブレインソームおよびビロソーム)は、選択されたコーティングを伴い、サイズ範囲を最適化したナノベシクルを調製するように、規定通りに自動化され、かつ、一貫した製造工程を達成することを目的とする、NanoAssemblr(登録商標)により、スケールアップに成功している。NanoAssemblr(登録商標)を使用して、NISV、ブレインソーム、およびビロソームの全てを作ることに成功している(表6)。ブレインソームをもたらすための、NISVの改変は、ゼータポテンシャルまたはベシクルサイズの特性に対して、非コーティングNISVと比較して、著明な影響を及ぼさなかった。
まず、染料は、通常の状況下では、BBBを越えることができないので、gNISVを、モデルカーゴとしてのヘキスト染料で染色した。染色されたgNISVを、Balb/cマウスに静脈内注射し、90分後に、脳組織を摘出した。予測される通り、ヘキスト染料は、単独では、BBBを越えず、脳内では検出されなかった。ヘキスト染料をカプセル化したgNISVが、BBBを越えて、細胞の核を染色したのに対し、ヘキスト染料をカプセル化する、非改変NISVでは、核染色は、ほとんど検出されなかった。これらのデータは、gNISVが、BBBを越え、それらのカーゴを放出することが可能であることを指し示す。
gNISVが、このモデルカーゴを送達することを裏付けたので、その後、最終的に、ヒト化抗VEEVモノクローナルIgGである、Hu1A3B−7をカプセル化する意図で、gNISVを使用して、ポリクローナルウサギ抗体(分子量:150kDa)をカプセル化した。
Hu1A3B−7の、gNISVおよびtNISVへのカプセル化を試みた。ウサギIgGの、gNISV内の静脈内送達の後における、マウス脳についての免疫組織化学解析は、非製剤化ウサギ抗体を注射されたマウスに由来する脳組織とは対照的に、これらのマウスに由来する脳血管系と関連するだけでなく、また、脳組織内に広がったウサギ抗体の存在も裏付けた。
エアロゾル化VEEVの感染後における、見本NISV製剤についての、詳細な評価
VEEV mAbである、Hu1A3B−7(S.A. Goodchild, L.M. O’Brien, J. Steven, M.R. Muller, O.J. Lanning, C.H, Logue, R.V. D’Elia, R.J. Phillpotts, S.D. Perkins. A humanised murine monoclonal antibody with broad serogroup specificity protects mice from challenge with Venezuelan equine encephalitis virus. Antiviral Res., 90(2011), 1-8;WO2011036435A1において、さらに開示されている)は、in vivoにおける感染研究時に使用される、えり抜きの抗ウイルス剤であった。VEEV感染についての、致死性のエアロゾルモデルを、約300PFUのVEEV Trinidad Donkey Strainに曝露される、BALB/cマウスに対して使用した。この抗原投与は、感染の約2日後に、死亡までの時間を、5〜6日間の間とする、感染の臨床徴候を引き起こす。
小スケールの予備研究を行って、IV経路を介して、治療を施すことの有用性について調べた(表7)。エアロゾル経路を介して、群(PBS対照群、空のベシクル対照群、VEEV Hu1A3B−7 mAb群、NISV Hu1A3B−7 mAb群、およびgNISV Hu1A3B−7 mAb群)1つ当たり5匹ずつのBALB/cマウスを、VEEVに曝露し、静脈内処置を、感染の24時間後に、1回施した。対照マウスが、疾患に斃れる前の、感染後5日目に、全てのマウスを殺処分した。ウイルス力価のために、脳組織および肺組織を摘出し、加工した。静脈内送達されたNISVプラットフォームは、肺内および脳内のウイルス負荷を、著明に減少させた(PBS対照と比較して、P=0.001であり、遊離mAbと比較して、P=0.007であり;データは示さない)。
実験1からの生存データと、実験2からの生存データとの組合せは、NISV Hu1A3B−7 VEEV mAb製剤およびgNISV Hu1A3B−7 VEEV mAb製剤のいずれの効能も、PBSおよび遊離Hu1A3B−7 VEEV mAより、有意に良好であったことを示す(それぞれ、p=0.0294およびp=0.0030;図16)。
Claims (29)
- 医薬ベシクル製剤を調製するための方法であって、
a)モノパルミトイルグリセロール、コレステロールおよびリン酸ジセチルを含むベシクル成分を50℃〜150℃の範囲の温度で加熱するステップ;
b)医薬剤を薬学的に許容可能な担体中に溶解させ、得られた医薬剤−担体混合物を30〜99℃の範囲で加熱するステップ;
c)医薬剤−担体混合物をベシクル成分に添加して、製剤混合物をもたらすステップ;ならびに
d)製剤混合物を加工して、医薬剤と担体とが複数のベシクル内に封入された医薬ベシクル製剤を形成するステップ
を含み、
医薬ベシクル製剤が薬学的に許容可能な担体中に溶解した、既知数量の医薬剤に復元されて、二相性医薬ベシクル製剤をもたらす
方法。 - モノパルミトイルグリセロール、コレステロールおよびリン酸ジセチルが、それぞれ5:4:1のモル比で提供される、請求項1に記載の方法。
- デオキシコール酸ナトリウムが、ベシクル成分中に、かつ/または医薬剤を薬学的に許容可能な担体中に溶解させる場合に提供される、請求項1〜2のいずれか1項に記載の方法。
- ベシクル成分が、in vivoにおける受容体媒介性輸送機構により認識されるリガンドをさらに含む、請求項1〜3のいずれか1項に記載の方法。
- リガンドが、グルコサミンおよび/もしくはトランスフェリン、またはこれらの任意の変異体もしくは断片である、請求項4に記載の方法。
- グルコサミンが、パルミトイル化グルコサミンである、請求項5に記載の方法。
- 共有結合が、チオール化リガンドと修飾コレステロールとの間に形成されるように、リガンドがチオール化され、コレステロール部分をコレステロール−PEG−マレイミドにより置きかえることにより、コレステロールが修飾される、請求項4〜請求項6に記載の方法。
- 医薬剤−担体混合物とベシクル成分とが、それぞれ3:1の比で提供される、請求項1〜7のいずれか1項に記載の方法。
- 医薬ベシクル製剤が、加工後において、少なくとも1つの凍結乾燥および融解のサイクルを経る、請求項1〜8のいずれか1項に記載の方法。
- 医薬剤が、抗菌剤および/または抗ウイルス剤である、請求項1〜9のいずれか1項に記載の方法。
- 抗菌剤が、フルオロキノロンおよびテトラサイクリンを含む群から選択される、請求項10に記載の方法。
- 抗菌剤が、レボフロキサシン、シプロフロキサシン、およびドキシサイクリンを含む群から選択される、請求項10〜請求項11に記載の方法。
- 抗菌剤が、30mg/mlの濃度である、請求項10〜請求項12に記載の方法。
- 抗ウイルス剤が、IFN−γ、IFN−α、カルボジン、DZ−13、および抗ウイルス抗体、またはこれらの任意の変異体もしくは断片を含む群から選択される、請求項10に記載の方法。
- 抗ウイルス抗体が、Hu1A3B−7 VEEV mAb、またはこの任意の変異体もしくは断片である、請求項14に記載の方法。
- a)モノパルミトイルグリセロール、コレステロールおよびリン酸ジセチルを含むベシクル;
b)医薬剤;ならびに
c)薬学的に許容可能な担体
を含む医薬ベシクル製剤。 - モノパルミトイルグリセロール、コレステロールおよびリン酸ジセチルが、それぞれ5:4:1のモル比で存在する、請求項16に記載の医薬ベシクル製剤。
- in vivoにおける受容体媒介性輸送機構により認識されるリガンドをさらに含む、請求項16〜請求項17に記載の医薬ベシクル製剤。
- リガンドが、グルコサミンおよび/もしくはトランスフェリン、またはこれらの任意の変異体もしくは断片である、請求項18に記載の医薬ベシクル製剤。
- ベシクルが、50〜4000nmのサイズである、請求項16〜請求項19に記載の医薬ベシクル製剤。
- 医薬剤が、抗菌剤および/または抗ウイルス剤である、請求項16〜請求項20に記載の医薬ベシクル製剤。
- 抗菌剤が、フルオロキノロンおよびテトラサイクリンを含む群から選択される、請求項21に記載の医薬ベシクル製剤。
- 抗菌剤が、レボフロキサシン、シプロフロキサシン、およびドキシサイクリンを含む群から選択される、請求項22に記載の医薬ベシクル製剤。
- 抗菌剤が、30mg/mlの濃度で存在する、請求項21〜請求項23に記載の医薬ベシクル製剤。
- 抗ウイルス剤が、IFN−γ、IFN−α、カルボジン、DZ−13、および抗ウイルス抗体、またはこれらの任意の変異体もしくは断片を含む群から選択される、請求項21に記載の医薬ベシクル製剤。

- 類鼻疽菌(Burkholderia pseudomallei)、野兎病菌(Francisella tularensis)、またはVEEVによる感染の防止または処置における使用のための、請求項16〜請求項26に記載の医薬ベシクル製剤、または請求項27に記載の医薬キット。
