多くの分泌タンパク質が翻訳後グリコシル化され、この過程により、糖部分(例えば、グリカン、糖類)がタンパク質の特定のアミノ酸に共有結合される。真核細胞においては、次の2種類のグリコシル化反応が起こる:(1)グリカンが認識配列Asn−X−Thr/Ser(ここで、「X」はプロリン以外の任意のアミノ酸である)のアスパラギンに連結されるN結合型グリコシル化、及び(2)グリカンがセリン又はスレオニンに連結されるO結合型グリコシル化。グリコシル化のタイプ(N結合型又はO結合型)にかかわらず、各部位(O又はN)と結合されるグリカン構造は広範囲にわたるため、タンパク質グリコフォームには微小不均一性がある。

本明細書に記載される方法によって生成される組み換えグリコシル化タンパク質及び抗体を含む組成物もまた、本明細書において提供される。例示的な実施形態では、抗体組成物はグリカン(例えば、ガラクトシル化グリカン、末端β−ガラクトース、G1、G1a、G1b及び/若しくはG2ガラクトシル化種、非フコシル化グリカン、コアフコース、又はこれらの組み合わせ)の量を調節する方法によって調製される。例示的な態様では、抗体は、IgG1抗体である。したがって、増加又は減少したADCC活性を有するグリコシル化非フコシル化IgG1抗体(抗HER2抗体、抗TNFα抗体又は抗CD20抗体(トラスツズマブ、インフリキシマブ又はリツキシマブを含む)など)を含む抗体組成物(ここで、グリコシル化非フコシル化IgG1抗体(抗HER2抗体、抗TNFα抗体又は抗CD20抗体(トラスツズマブ、インフリキシマブ又はリツキシマブを含む)など)は、IgG1抗体組成物上のグリカン(例えば、ガラクトシル化グリカン、末端β−ガラクトース、G1、G1a、G1b及び/若しくはG2ガラクトシル化種、非フコシル化グリカン、コアフコース、又はこれらの組み合わせ)の量を調節する(例えば、増加又は減少させる)ことによって、対照又は参照抗体組成物と比較して、特定のADCC活性、又は増加若しくは減少したADCC活性を有するように操作されている)が本明細書において提供される。
治療用モノクローナル抗体のトラスツズマブ(抗HER2)、リツキシマブ(抗CD20)及びインフリキシマブ(抗TNFα)をCHO細胞中に発現させ、標準的な製造プロセスにより高濃度溶液として製造した。この研究で用いたmAbは、CD20、EGFRファミリーメンバーであるHER2、及びTNFαなどの受容体を標的としている。
β−(1−4)−ガラクトシダーゼ(QA−Bio)及びβ−1,4−ガラクトシルトランスフェラーゼ(Roche)のインビトロでの脱ガラクトシル化及びガラクトシル化能を利用して、トラスツズマブ、リツキシマブ及びインフリキシマブ用のガラクトースリモデリングシリーズの試料を作製した。ガラクトースを除去するために、トラスツズマブ、リツキシマブ及びインフリキシマブを、最初に、50mMのリン酸ナトリウム(pH6.0)を含有する反応緩衝液の存在下、1/50の比のβ−(1−4)−ガラクトシダーゼ(QA−Bio)と、37℃で1〜2時間インキュベートした。次いで、プロテインAアフィニティークロマトグラフィー精製(ガラクトシダーゼ及び他の成分を除去するために使用される)を、Agilent1100シリーズHPLCシステムで、予め充填したプロテインAカラム(Poros PrA、Applied Biosystem)を用いて、3mL/分の流量で実施した。適切な量の各試料をカラムに注入した後、100%緩衝液A(20mMトリス−HCl/150mM NaCl、pH7.0)をカラムに1.4分間通し、続いて100%緩衝液B(0.1%酢酸)を2.9分間通し、その間に画分を集めた。溶出したmAbを含有する画分を、3kDaカットオフ膜を有するAmicon Ultra遠心フィルターを使用して所望の緩衝液系中に透析濾過した。
最初に、トラスツズマブ及びリツキシマブ原薬(「DS」)を、Agilent 1100シリーズHPLCで、カスタマイズしたglycap−3Aカラム(低密度FcγIIIa受容体、3×150mm、Zepteon)を使用して、2つの画分(フロースルー及び溶出液)に分離した。移動相Aは、20mMトリス(pH7.5)、150mM NaClを含み、移動相Bは50mMクエン酸ナトリウム(pH4.2)であった。0.5mL/分の流量で勾配(0%Bで8分間保持、0%Bから18%Bへ22分間)を使用して、フコース濃縮(フロースルー)mAb及び非フコース/HM濃縮(溶出)mAbの両方を得た。非フコシル化種及びHM濃縮種の両方を含む溶出液画分を、Endo−H(QA−Bio、PN E−EH02)でさらに酵素処理して、高マンノース種を除去した。具体的には、50mMリン酸ナトリウムの反応緩衝液(pH5.5)中、37℃で、24時間、mAbをEndo−Hと共にインキュベートした。最終mAb濃度は4mg/mLである。
glycap−3Aカラムからフロースルー画分を集め、β(1,4)ガラクトシダーゼで処理して末端ガラクトースを除去することにより、様々なレベルのガラクトースを含むフコシル化トラスツズマブ及びリツキシマブを生成した。次いで、G0F濃縮mAbを、10mMのUDP−ガラクトース、100mMのMES(pH6.5)、20mMのMnCl2、及び0.02%のアジ化ナトリウムを含有する反応緩衝液中で、β−1,4−ガラクトシルトランスフェラーゼ(Roche)と共に37℃でインキュベートした。酵素とmAbの最終的な比は6/1(μL/mg)であり、mAb濃度は2mg/mLであった。異なる時点で反応混合物からアリコートを取り出し、続いて瞬間冷凍して反応を停止させることにより、異なるレベルのガラクトースを有するmAbを得た。プロテインAクロマトグラフィーを実施し、溶出液を、Amicon Ultra遠心フィルターを使用して所望の緩衝液系に透析濾過した。
所望のグリカン特性及び最小レベルの高分子量種を確実にするために、濃縮及びリモデリングした全ての試料を、親水性相互作用液体クロマトグラフィー(HILIC)及びサイズ排除クロマトグラフィー(SEC)で特性評価した。
ADCCに対する非フコシル化トラスツズマブの計算による全体的影響(これは、G0及びG1からの個々の影響、並びにDSロットに対するそれらの相対比に基づく)を確認するために、実験は全体的な非フコースの影響を直接測定するように設計された。
ADCCアッセイを、エフェクター細胞としてFcγIIIa(158V)発現NK92(M1)細胞を使用し、標的細胞としてトラスツズマブにHCC2218細胞、リツキシマブにWIL2−S細胞、インフリキシマブにCHO MT−3細胞を用いて行った。標的細胞を最初にカルセイン−AMで標識した後、トラスツズマブについては3.3〜2000ng/mL、リツキシマブについては0.0155〜100ng/mL、インフリキシマブについては0.01024ng/mL〜100000ng/mLの漸増濃度でインキュベートした。次いで、エフェクター細胞を、約1〜2時間の間、25:1のE:T比で、オプソニン化標的細胞に添加した。溶解した標的細胞から放出されたカルセインを、Envision(Perkin Elmer)蛍光プレートリーダーで反応上清の蛍光を測定することによって決定した。SoftMaxProソフトウェアを使用する拘束4パラメーターフィットを使用してデータを平均蛍光値にフィットさせ、EC50標準/EC50試料比によって計算し、参照標準に対するADCC活性の割合として報告した。各アッセイを3回行い、平均及び標準偏差を報告した。
リツキシマブに対するCD20抗原結合アッセイを、蛍光阻害を報告する競合的アッセイフォーマットを利用して、ヒトβ−リンパ芽球様細胞株であるWIL2−S細胞を用いて行った。試験試料は、WIL2−S細胞の細胞表面発現CD20への結合について、参照標準の固定濃度のAlexa−488標識形態と競合する。0.5mg/mLのBSAを含有するPBS中で、4.92〜3000ng/mLの最終濃度範囲まで8つの濃度にわたって段階希釈することによって、参照標準、アッセイ対照及び試験試料の用量応答曲線を作成した。Alexa−488標識競合体を最終的なウェル内濃度100ng/mLに希釈する。30,000細胞/ウェルに希釈した試料、競合体及びWIL2−S細胞を96ウェルプレートに2連で加え、プレートシーラーで密封する。次いで、プレートを室温で4.5〜6時間インキュベートした後、Acumen(登録商標)eX3イメージングサイトメーター(TTP Labtech)で蛍光シグナルを測定する。蛍光シグナルの用量依存的減少が、試験試料の濃度の上昇に伴い検出される。参照標準試料に対する試験試料の相対CD20結合を報告する。SoftMaxProによる4パラメーター曲線フィットを使用してデータを平均発光値にフィットさせ、IC50標準/IC50試料を計算し、相対結合活性パーセントとして報告した。各試料を3つの独立したアッセイで試験し、最終結果を3回の測定の平均として報告する。トラスツズマブ結合アッセイを、SKBR−3を標的細胞として使用し、用量応答曲線を(0.016〜10μg/mL)の範囲としたことを除いて、同様の方法で実施した。
IgG分子におけるADCC機能の高精度制御のための非フコシル化−ガラクトシル化及び非フコシル化−非ガラクトシル化グリカン基の組合せの使用
抗体のADCC活性を大きく増強する際のグリカン非フコシル化の役割は、この略20年にわたって知られている(Mizushima et al.,2011)が、ガラクトシル化又は高マンノースなどの他のグリカン構造の成分の寄与は、十分には理解されていなかった。例えば、ガラクトシル化のADCCへの影響に関する公表されたデータには議論の余地があった。グリカン構造の全体的な複雑さ及び不均一性には意味があり、一般的なモノクローナル抗体は多くは、20を超える個々のグリカン種を示す。グリカン−ADCC関係及びグリカン組成物の全体的な複雑さに関する理解が限られているため、治療用抗体に対するADCC機能の正確な制御は依然として困難であった。種々のグリカン群におけるグリカン構造の例を図2に示す。
ADCC活性に対する末端ガラクトースの影響
FcγIIIa受容体結合及びその後のADCC活性に対する末端ガラクトースの効果を調べるための有効な方法は、脱ガラクトシル化及び/又は完全ガラクトシル化タンパク質を生成し、次いで、ADCC活性に対するガラクトースの影響を試験することであった。しかしながら、大部分の治療用IgG1は不均一なグリカン種の集団を含み、製剤内にはガラクトシル化されていない種、一部ガラクトシル化されている種、及び完全にガラクトシル化されている種の全てが存在する。さらに、完全にガラクトシル化された種(G2Fなど)は、一般的に他の2つのタイプの種と比較してわずかな割合でしか存在しない。例えば、Raju,T.S.,et al.,Glycoengineering of therapeutic glycoproteins:in vitro galactosylation and sialylation of glycoproteins with terminal N−acetylglucosamine and galactose residues.Biochemistry,2001.40(30):p.8868−76を参照されたい。したがって、IgG1のADCC活性に対するガラクトシル化の影響を評価するために、関連する全てのグリカン形態の役割を解読する機会が存在する。Fc N結合型グリカン種の一般的な形態(0、1又は2個のガラクトース残基を有するものを含む)を図2に示す。
非フコシル化mAbのADCC活性に対する末端ガラクトースの影響
非フコシル化mAb種に対するガラクトースの影響を評価するために、末端ガラクトースを有する非フコシル化種及び末端ガラクトースを有さない非フコシル化種の両方を生成した。実験計画では2つの因子を考慮した。第1に、代表的なmAb産生プロセスからのロットを出発物質と使用することによって、関連するグリカン種の影響に焦点を当てた。ここでは、非フコシル化G0及びG1グリカンが優勢な非フコシル化種であった。ガラクトースを含まない非フコシル化トラスツズマブ(G0)及びガラクトースを含む非フコシル化トラスツズマブ(G1)の両方は濃縮したが、他のグリカン属性(マンノースなど)は一定に保った。第2に、濃縮非フコシル化G0mAb及びG1mAbは、ADCC応答が非フコシル化種(ADCC活性に大きな影響を有することが知られている)の高含量のためにアッセイの作業範囲外であるため、直接測定することができなかった。したがって、最小のADCC活性を有すると予想されるG0Fグリカンを濃縮したmAbを、次に、非フコシル化G0及びG1種を濃縮した試料と種々の比でブレンドして、ブレンドした物質のADCC活性を各mAbのADCCアッセイの作業範囲に収まるようにした。
フコシル化mAbのADCC活性に対する末端ガラクトースの影響
フコシル化トラスツズマブ及びリツキシマブのADCC活性に対する末端ガラクトースの影響の評価も実施した。まず、FcγIIIaアフィニティークロマトグラフィーからフロースルーを回収し、ガラクトシダーゼにより処理し、ProAクロマトグラフィーで浄化することにより、対応するDSロットからG0F濃縮トラスツズマブ及びG0F濃縮リツキシマブを作製した(図8、左)。次に、β(1,4)ガラクトシルトランスフェラーゼによるmAbの酵素的リモデリングにより、末端ガラクトースをG0F種に加えた。このような酵素反応のインキュベーション時間を制御することにより、種々のレベルの末端ガラクトースを有する試料を得た。最後に、フコシル化トラスツズマブ及びフコシル化リツキシマブに対する末端ガラクトースの影響を、これらの異なるGal%を有する試料のADCC活性を、それらの対応する機能アッセイを用いて測定することにより評価した。トラスツズマブ(「mAb1」)及びリツキシマブ(「mAb2」)について得られた結果を、それぞれ図12A及び12Bに示す。参照標準と比較したこれらのフコシル化試料の相対ADCC活性は、トラスツズマブ及びリツキシマブともに低かった。最高レベルの末端ガラクトース(約90%)を有する最大ADCC活性は、トラスツズマブで15%未満、リツキシマブで40%未満である。さらに、影響係数(ADCC%/Gal%)が20を超える非フコシル化トラスツズマブ及び非フコシル化リツキシマブに対する末端Galの影響(図9及び図11)とは異なり、フコシル化トラスツズマブ及びフコシル化リツキシマブに対する末端Galの影響は非常に小さく、影響係数はmAb1で約0.1であり、mAb2で約0.2であった(図12A及び図12B)。0〜90%の範囲のガラクトシル化はADCC活性に、それがフコシル化mAbと関連している場合、有意な影響を及ぼす可能性が低いことを、この結果は示した。換言すれば、ADCC活性に対する末端ガラクトースの影響は、コアフコースの非存在によって有意に影響され、フコシル化mAbに対するその影響は無視できるものであった。

Claims (26)
- グリコシル化非フコシル化IgG1抗体組成物の抗体依存性細胞傷害(ADCC)活性を制御する方法であって、
(1)グリコシル化非フコシル化IgG1抗体組成物のADCC活性を決定する工程;及び
(2)前記組成物内のIgG1抗体のコンセンサスグリコシル化部位におけるグリカン種中の末端β−ガラクトースの量を増加又は減少させることによって、前記グリコシル化非フコシル化IgG1抗体組成物のADCC活性を増加又は減少させる工程
を含む方法。 - 参照グリコシル化非フコシル化IgG1抗体組成物の抗体依存性細胞傷害(ADCC)活性にマッチさせる方法であって、
(1)参照グリコシル化非フコシル化IgG1抗体組成物のADCC活性を決定する工程;
(2)前記参照IgG1抗体と同じ抗体配列を有するIgG1抗体を含む第2の抗体組成物のADCC活性を決定する工程;及び
(3)前記第2の抗体組成物内の1つ以上の抗体のコンセンサスグリコシル化部位におけるグリカン種中の末端β−ガラクトースの量を増加又は減少させることによって、前記第2の抗体組成物のADCC活性を変化させる工程
を含み、
末端β−ガラクトースの量を増加又は減少させた後の前記第2の抗体組成物のADCC活性は、前記参照グリコシル化非フコシル化IgG1抗体組成物と同じであるか、又は前記参照グリコシル化非フコシル化IgG1抗体組成物の約10%、約15%、約20%、約25%、約30%、約35%、約40%、約45%若しくは約50%以内であるか、又は前記参照グリコシル化非フコシル化IgG1抗体組成物の約1%〜約50%以内である、方法。 - グリコシル化非フコシル化IgG1抗体組成物の特定の目標抗体依存性細胞傷害(ADCC)活性を操作する方法であって、
(1)グリコシル化非フコシル化IgG1抗体組成物のADCC活性を決定する工程;

末端β−ガラクトースの量を増加又は減少させた後の前記グリコシル化非フコシル化IgG1抗体組成物のADCC活性は、前記目標ADCC活性と同じであるか、又は前記目標ADCC活性の約10%、約15%、約20%、約25%、約30%、約35%、約40%、約45%若しくは約50%以内であるか、又は前記目標ADCC活性の約1%〜約50%以内である、方法。 - 工程1は、工程2及び/又は工程3の前、後又は同時に行われる、或いは工程2は、工程1及び/又は工程3の前、後又は同時に行われる、請求項1〜3のいずれか一項に記載の方法。

- 前記抗体組成物の末端β−ガラクトースの量は、約10%、約15%、約20%、約25%、約30%、約35%、約40%、約45%、約50%、約55%、約60%、約65%、約70%、約75%、約80%、約85%、約90%、約95%、約100%、約125%、約150%、約175%若しくは約200%増加若しくは減少するか、又は総量で約0.5%、約1%、約2%、約3%、約5%、約7%、約10%、約15%、約20%、約25%、約30%、約35%、約40%、約45%、約50%、約55%、約60%、約65%、約70%、約75%、約80%、約85%、約90%、約95%、約96%、約97%若しくは約98%まで増加若しくは減少するか、又は総量で約0%〜100%まで増加若しくは減少する、請求項1〜13のいずれか一項に記載の方法。
- 前記ADCC活性は、細胞ベースのアッセイ又は結合アッセイを用いて測定又は決定される、請求項1〜14のいずれか一項に記載の方法。
- 前記細胞ベースのアッセイは、NK92細胞又はPMBC細胞を含む、請求項15に記載の方法。
- 前記結合アッセイは、FcγRIIIaを含む、請求項15に記載の方法。
- 前記抗体組成物中の末端β−ガラクトースの量は、抗体を発現する細胞を、前記抗体の前記グリカン種中の末端β−ガラクトースの量を調節する細胞培養培地中で培養することによって増加又は減少させる、請求項1〜17のいずれか一項に記載の方法。
- 前記末端β−ガラクトースの量は、化学薬品又は酵素を用いて増加又は減少させる、請求項1〜18のいずれか一項に記載の方法。
- 前記酵素は、Endo−S2、β−(1−4)−ガラクトシダーゼ、Endo−H、β−1,4−ガラクトシルトランスフェラーゼ、及びPNGアーゼ−Fからなる群から選択される、請求項18に記載の方法。
- 請求項1〜20のいずれか一項に記載の方法によって生成されるIgG1抗体組成物であって、前記抗体組成物は、参照IgG1抗体組成物と比較して、増加又は減少したADCC活性を有し、前記参照IgG1抗体組成物は、前記増加又は減少したADCC活性を有するIgG1抗体と同じ抗体配列を有するIgG1抗体を含む、IgG1抗体組成物。
- 前記抗体組成物は、前記参照抗体組成物と比較して、約30%、約35%、約40%、約45%、約50%、約55%、約60%、約65%、約70%、約75%、約80%、約85%、約90%、約95%、約100%、約125%、約150%、約175%若しくは約200%、又は約1倍、約2倍、約3倍、約4倍、約5倍、約6倍、約7倍、約8倍、約9倍若しくは約10倍増加したADCC活性、或いは前記参照抗体組成物と比較して、約0.5倍〜約8倍増加したADCC活性を有する、請求項21に記載のIgG1抗体組成物。
- 前記抗体組成物は、前記参照抗体組成物と比較して、約30%、約35%、約40%、約45%、約50%、約55%、約60%、約65%、約70%、約75%、約80%、約85%、約90%、約95%、約100%、約125%、約150%、約175%若しくは約200%、又は約1倍、約2倍、約3倍、約4倍、約5倍、約6倍、約7倍、約8倍、約9倍若しくは約10倍減少したADCC活性、又は前記参照抗体組成物と比較して、約0.5倍〜約8倍減少したADCC活性を有する、請求項21に記載のIgG1抗体組成物。
- 前記IgG1抗体組成物は、トラスツズマブ、リツキシマブ又はインフリキシマブを含む、請求項21〜23のいずれか一項に記載の抗体組成物。
- 前記トラスツズマブ抗体は、
a.(i)配列番号1で示されるアミノ酸配列を含む軽鎖CDR1配列、(ii)配列番号2で示されるアミノ酸配列を含む軽鎖CDR2配列、及び(iii)配列番号3で示されるアミノ酸配列を含む軽鎖CDR3配列を含む軽鎖可変ドメイン、及び
b.(i)配列番号4で示されるアミノ酸配列を含む重鎖CDR1配列、(ii)配列番号5で示されるアミノ酸配列を含む重鎖CDR2配列、及び(iii)配列番号6で示されるアミノ酸配列を含む重鎖CDR3配列を含む重鎖可変ドメイン
を含むか、若しくは
c.配列番号7を含む軽鎖可変ドメイン、及び
d.配列番号8を含む重鎖可変ドメイン
を含む;
又は、前記リツキシマブ抗体は、
a.(i)配列番号11で示されるアミノ酸配列を含む軽鎖CDR1配列、(ii)配列番号12で示されるアミノ酸配列を含む軽鎖CDR2配列、及び(iii)配列番号13で示されるアミノ酸配列を含む軽鎖CDR3配列を含む軽鎖可変ドメイン、及び
b.(i)配列番号14で示されるアミノ酸配列を含む重鎖CDR1配列、(ii)配列番号15で示されるアミノ酸配列を含む重鎖CDR2配列、及び(iii)配列番号16で示されるアミノ酸配列を含む重鎖CDR3配列を含む重鎖可変ドメイン
を含むか、若しくは
c.配列番号17を含む軽鎖可変ドメイン、及び
d.配列番号18を含む重鎖可変ドメイン
を含む;
又は、前記インフリキシマブ抗体は、

c.配列番号31を含む軽鎖可変ドメイン、及び
