Abstract
Description
本出願は、米国特許法119(e)に基づき、全体が参照により本明細書に組み込まれる、2016年11月16日に出願された米国特許仮出願第62/422,922号および2017年3月29日に出願された米国仮特許出願第62/478,063号について優先権を主張する。
本発明は、グラントP30 CA023168を通じて国立衛生研究所および国立癌研究所により授与された政府の支援によりなされた。米国政府は本発明において一定の権利を有する。
本明細書中に記載される発明は、連結した治療剤または造影剤を含み得る、リガンドイオノフォアコンジュゲート、およびこのコンジュゲートを含有する医薬組成物に関する。また、記載されたコンジュゲートを、エンドサイトーシスまたは他の同様のプロセスによって内在化される治療剤または造影剤のエンドソーム蓄積および脱出を増加させるために使用する方法も記載される。葉酸受容体を過剰発現する細胞内へのコンジュゲートされたマイクロRNAの送達を媒介する、マイクロRNAの葉酸への直接の結合(FolamiR)による、マイクロRNAの腫瘍組織への送達も記載される。
多くの病気は、薬または生物学的薬剤で治療することができる(生物学的薬剤の例としては、ヌクレオチド、例えばsiRNA、miRNAなど;天然には存在しない合成アミノ酸を含むアミノ酸;酵素、ペプチド、アプタマー、抗原などを含むタンパク質;および抗体、例えば糖タンパク質、免疫グロブリンなど)。これらの薬物または生物製剤は、それらの標的とするリガンド(例えば葉酸受容体結合リガンド)を用いてそれらの標的細胞に送達することができるが、例えば葉酸が介在するエンドサイトーシス後に、その薬物がエンドソームから放出されないことでその有効性が阻害されることがある。したがって、細胞質への閉じ込められた積み荷(cargo)の「エンドソーム放出」のための新しい方法の発見は、標的化された薬物または生物製剤の増大した有効性を達成するために有用であろう。エンドソーム放出は、健康な組織に対して毒性の低い既知のイノフォアを用いて、この積み荷を含むエンドソームを破裂させるための浸透圧を作り出すためのリガンドイオノフォアコンジュゲートの使用によって促進することができることが見出された。理論に縛られることなく、H+イオンの流出とK+イオンの流入が共役するイオノフォアおよびアンチポーターである、ニゲリシンは、細胞内に送達さると、エンドソームの内部でエンドソームの膨潤および/または崩壊をもたらす浸透圧の不均衡とエンドソーム内容物の細胞質内への放出を引き起こすと考えられている。サリノマイシンのようなカリウムイオンを輸送する他のK+イノノフォアもエンドソーム放出に利用できると考えられる。
いくつかの実施形態では、本開示は、共有結合した葉酸とマイクロRNAとのコンジュゲートまたはその模倣体、および薬理学的に許容できる担体、希釈剤、または添加剤を含んでなる標的化されたマイクロRNA送達システムを提供する。

材料
ペプチド合成のためのアミノ酸はAapptec, USAから購入した。N-ヒドロキシベンゾトリアゾール(HOBt)、1-[ビス(ジメチルアミノ)メチレン]-1H-1,2,3-トリアゾロ[4,5-b]ピリジニウム3-オキシドヘキサフルオロホスフェート(HATU)およびベンゾトリアゾール-1-イルオキシトリス(ピロリジノ)ホスホニウムヘキサフルオロホスフェート(PyBOP)はSigma-Aldrichから入手した。固相ペプチド合成(SPPS)は、標準的なペプチド合成装置(Chemglass、Vineland、NJ)を用いて行った。特に記載しない限り、他の化学物質は全てSigma-Aldrichから購入した。葉酸コンジュゲートはいずれも分取逆相(RP)-HPLC(Agilent)によって精製し、LC/MS分析は、UVダイオードアレイ検出器と組み合わせたAgilent質量分析計を使用した。
DMSO中のニゲリシンの葉酸-ピリジルジスルフィドアミド誘導体(0.010 g、0.0069 mmol、1当量)およびNHS-SS-DBCO(0.005 g、0.010 mmol、1.5当量)の攪拌溶液に、DIPEA(0.0013 g、0.010 mmol、1.5当量)を滴加した。室温にて反応混合物の攪拌を続けた。反応の進行をLC-MSでモニターした。葉酸-SS-nigを完全に変換した後、粗製の反応混合物をRP-HPLC(移動相A=10mM酢酸アンモニウム、pH=7;有機相B=アセトニトリル;方法:13ml/分にて35分間で0%Bから50%B)により精製し、葉酸-SS-DBCO-ニゲリシンを65%の収率で得た。LC-MS(A=10mM炭酸水素アンモニウム、pH=7;有機相B=アセトニトリル;方法:12分で0%Bから100%B) RT=6.52分(M/2+H+=945.4)
葉酸-ニゲリシン DBCO miR-34aコンジュゲート:
葉酸-ニゲリシン-SS-DBCO miR-34aコンジュゲート:
葉酸-ニゲリシン DBCO-siLuc2コンジュゲート:
DMSO中の葉酸-Cys(0.010g、0.015mmol、1当量)およびマレイミド-NIR色素(0.019g、mmol、1.1当量))の撹拌溶液に、DIPEA(0.0029g、0.0228mmol、1.5当量)を滴加した。反応混合物を室温で撹拌し続けた。反応の進行をLC-MSによってモニターした。葉酸-Cysを完全に変換した後、粗製の反応混合物をRP-HPLC(移動相A=10mM酢酸アンモニウム、pH=7;有機相B=アセトニトリル;方法:13ml/分にて35分間で0%Bから30%B)により精製し、葉酸-NIRを85%の収率で得た。LC-MS(A=10mM炭酸水素アンモニウム、pH=7;有機相B=アセトニトリル;方法:12分で0%Bから30%B) RT=3.30分(M+H+=1179.0).

miR-34a-5pガイド鎖およびmiR-34a-3pパッセンジャー鎖(いずれもIntegrated DNA Technologiesによって調製)として示される2つのRNAオリゴヌクレオチドを使用してmiRNA二本鎖を構築した。miR-34a-3pパッセンジャー鎖は、5’末端にアジドリンカーおよび3’末端に2’-O-メチルRNA塩基で修飾された20nt RNAオリゴ二重鎖(mCmArAmCrCmArGmCrUmArAmGrAmCrAmCrUmGrCC)を含んでなり、miR-34a-5pガイド鎖は、3’上に2’-O-メチルRNA塩基で最小限の修飾を有する22ntのRNAオリゴを含んでなる(rUmGmUrUrGrGrUrCrGrArUrUrCrUrGrUrGrArCrGrGrU/5Phos)。同じ修飾を用いて合成したスクランブルmiRNA(ネガティブコントロール)を用いて対照の二本鎖を形成した。葉酸-DBCO-ニゲリシンまたは葉酸-SS-DBCO-ニゲリシンコンジュゲートとアジド修飾アンチセンスmiR-34a(またはスクランブル)との間でビ-オルトゴナル(bi-orthogonal)なクリック反応を行った。クリック反応は、水中で室温にて8時間、モル比1:10(アジドオリゴ:葉酸DBCOまたは葉酸-SS-DBCO)で行った後、4℃にて4時間冷却した。コンジュゲートしていない葉酸は、Oligo Clean and Concentrator(Zymo Research)を製造業者の説明書に従って使用して反応物から除去した。コンジュゲーションは、15%TAEネイティブPAGEおよびMALDIスペクトル分析を用いて確認した。葉酸-NIR化合物のコンジュゲーションについては、さらにLicor Odyssey CLX(Licor)を用いて確認した。コンジュゲート後、miR-34a-5pガイド鎖を葉酸とNIR-葉酸コンジュゲートにアニーリングした。簡潔には、アニーリング緩衝液:50mMのNaCl(Sigma)、および1mMのEDTA(Sigma)を添加した10mMトリス緩衝液pH7(Sigma)中で、葉酸-miR-34a-3pとmiR-34a-5pとを等モル比(1:1、最終濃度各5μM)で混合し、95℃で5分間インキュベートし、1時間かけて室温まで冷却した後、-80℃で保存した。
二本鎖RNAオリゴヌクレオチドおよびFolamiRコンジュゲートを、37℃の水中の50%ウシ胎児血清(Sigma)中で、表示した時間インキュベートした。RNA試料を回収し、15%TAEポリアクリルアミドゲル電気泳動(PAGE)を用いて分析した。
細胞系
細胞内のFolamiR-34aコンジュゲート活性をモニターするために、miR-34a Renillaセンサー(MB-231センサー)を発現するMDA-MB-231細胞を作製した。最初に、miRNAセンサーの特異性を、MDA-MB-231乳癌細胞中で、miR-34a模倣体またはネガティブコントロール(スクランブルRNA)とともにmiR-34aセンサーまたは変異センサーを一過性に発現させることによってモニターした。その目的のために、1×104の細胞を96ウェルのプレートに播種し、Lipofectamine 2000(Life Technologies)を用いて、25ngのプラスミドおよび6nMのmiRNA模倣体で同時トランスフェクトした。Renilla Gloルシフェラーゼキット(Promega)を用いて、トランスフェクションの48時間後にRenilla活性を測定した。結果は、miR-34aがMB-231細胞において内因的に活性であること、および外因的なmiR-34aの送達がRenillaセンサーのさらなるサイレンシングを促進することを示唆する。これらの結果は、miR-34aセンサーがmiR-34aに特異的であり、MB-231細胞中の内因的レベルのmiR-34aがmiR-34aセンサーを完全に沈黙させるのに十分ではなく外因性のmiR-34aでのノックダウンを増大させる余地があることを示唆している。安定なクローンを作製し、Renilla活性について試験した。15種の安定なクローンのうち、高いRenillaレベルとmiR-34a活性をモニターするその能力から、クローン5をさらなる実験のために選択した。(図1)
MDA-MB-231トリプルネガティブ乳癌細胞(HTB-26、MycoAlert Mycoplasma検出Kit-Lonzaによるマイコプラズマ混入試験によりマイコプラズマ不含を確認済)を、10%胎児ウシ血清(Sigma)、100U/ml-1のペニシリンおよび100μg/mlのストレプトマイシン(Hyclone、GE Healthcare Life Sciences)を添加したRPMI 1640培地(葉酸不含、Life Technologies)で培養し、5%CO2中37℃に維持した。ルシフェラーゼレポーター実験用に、ベクター(psiCHECK、Promega)中のRenillaルシフェラーゼの3’非翻訳領域に、miR-34aに対するアンチセンス配列を挿入することにより、miR-34aセンサープラスミドを作製した。miR-34a特異的サイレンシングは、MDA-MB-231細胞においてmiR-34aセンサーまたは変異miR-34aセンサーを一過性でトランスフェクトすることによって確認した。miR-34aセンサー発現細胞を、Lipofectamine RNAimax(Life Technologies)を使用してmiR-34a模倣体でトランスフェクトし、外因的なmiRNAによって媒介されるサイレンシングを確認した。安定なクローンを作製するために、MDA-MB-231細胞を6ウェルのプレートに1×106細胞/ウェルの密度で播種し、Lipofectamine 2000(Life Technologies)を用いて2μgのmiR-34aセンサープラスミドをトランスフェクトした。選択マーカーとしてハイグロマイシンB(500μg/mL;Hyclone、GE Healthcare Life Sciences)を用いて安定なクローンを選択した。単一のクローンをRenilla発現について評価し、最も高いRenilla発現を有するクローンを選択した。MB-231センサー細胞を、葉酸および血清不含RPMI培地中、葉酸-DBCO-miR-34a、葉酸-ニゲリシン-DBCO-miR-34a、葉酸-ニゲリシン-SS-DBCO-miR-34aおよびFolamiR-NC(陰性対照)を含む(最終濃度50nM)96ウェルのプレートに播種した。対照として、非処理および非コンジュゲート二本鎖miRNAを含めた。Renilla Glo ルシフェラーゼキット(Promega)を製造業者の指示に従って使用し、インキュベーション後12~48時間の間のRenillaルシフェラーゼ値を得た。各時点について、Renillaレベルを FolamiR-NCに対して正規化した。実験は、各条件につき3連で3回行った。
葉酸-ニゲリシン-DBCO-miR-34aまたは葉酸-ニゲリシン-SS-DBCO-miR-34aで(トランスフェクション試薬非存在下で)処理したMB-231センサー細胞は、処理後48時間で最大80%のRenilla活性の低下を示す(図1)。しかしながら、ニゲリシンを欠く葉酸-DBCO-miR-34aは、48時間までに30%の遺伝子ノックダウン活性しか示さず、このことは、ニゲリシンがエンドソームからのmiR-34aの放出を助けることを裏付ける。データに基づけば、意外にも、放出可能および放出不能の両方の葉酸-ニゲリシン-miR-34aが細胞に効率的に入り、活性を保持しており、このことはコンジュゲートされた葉酸がmiR-34a-5pのアルゴノート(Argonaute)へのローディングを妨げないことを示唆する。
siRNA標的化アッセイとして、MDA-MB-231細胞を1×106細胞/ウェルの密度で6ウェルのプレートに播種し、Lipofectamine 2000(Life Technologies)を用い、2μgのpmiRGloプラスミド(Promega)でトランスフェクトした。24時間後、葉酸および血清不含RPMI培地中、Fol-DBCO-siLuc2、Fol-ニゲリシン-DBCO-siLuc2、Fol-DBCO-NC(ネガティブコントロール)、Fol-ニゲリシン-DBCO-NCを含む(最終濃度50nM)96穴プレートに細胞を再播種した。対照として、非処理および非コンジュゲート二本鎖miRNAを含めた。各時点について、Renillaおよびホタルルシフェラーゼの値を、デュアルルシフェラーゼレポーターキット(Promega)を製造元の指示に従って使用して得た。Firefly/Renillaの比率は、各時点について、Fol-DBCO-NCまたはFol-ナイリシン-DBCO-NCに対して正規化した。実験は、各条件につき3連で3回行った。二元配置分散分析(ANOVA)およびボンフェローニ事後検定を用いて統計的有意性を検定した。
MDA-MB-231細胞を、葉酸とコンジュゲートしたルシフェラーゼ(siLuc2)(Fol-DBCO-siLuc)またはニゲリシンの分子を有する修飾された葉酸リガンドとコンジュゲートしたルシフェラーゼ(siLuc2)(Fol-ニゲリシン-DBCO-siLuc)についてsiRNAとともにインキュベートすることにより、ルシフェラーゼ標的化アッセイを行った。これら細胞ベースの実験は、Fol-ニゲリシン-DBCO-siLucで処理した細胞において、処理後18時間で急速なルシフェラーゼ活性の低下を示し、24時間後で40%に達することを示した(図2)。しかし、Fol-DBCO-siLucでは、32時間でもルシフェラーゼ活性の低下はみられない。興味深いことに、このルシフェラーゼ活性の低下は、Fol-NC(ネガティブコントロール)処理した細胞またはFol-siLuc処理した細胞では観察されず、ニゲリシンの存在に特異的であることを示唆している。
Fol-siLuc2とそのニゲリシン対応物との間でのルシフェラーゼ低下における違いは、エンドソームから細胞質への葉酸-siLuc2コンジュゲートの脱出におけるニゲリシンの促進を実証している。
生細胞イメージング
生細胞イメージングのため、細胞をガラス底の2穴チャンバースライド(Lab-TekTM Chambered Coverglass、Thermo Fisher Scientific、デンマーク)上にプレーティングした。簡潔に言えば、チャンバースライドをポリ-D-リジン(0.1mg/mL;Sigma-Aldrich)で5分間前処理し、PBSで洗浄し、5分間風乾させた。Rab5B-GFPを安定に発現するMDA-MB-231細胞を実験の1日前に3×104細胞/ウェルでプレーティングし、10%ウシ胎児血清(Sigma)、100U/mlペニシリンおよび100μg/mlストレプトマイシン(Hyclone, GE Healthcare Life Sciences)を添加した葉酸不含RPMI 1640培地(Life Technologies)中、5%CO2、37℃で維持した。実験当日、培地を10mMの4-(2-ヒドロキシエチル)-1-ピペラジンエタンスルホン酸(HEPES、Sigma-Aldrich)を添加した培地と交換し、スライドを、共鳴スキャナーと、東海ヒットライブイメージングチャンバー(INU-TIZ-F1;東海ヒット社、日本)を取り付けたピエゾZドライブ(ニコンインスツルメンツ社)とを備えたNikon A1Rsi共焦点顕微鏡(温度は37℃に設定)にセットした。画像取得は単一焦点面で行ない、NIS-elements software 4.5(Nikon Instruments Inc.、日本)を使用し、葉酸-Cy5コンジュゲート(50nM)を添加後に開始した。ImageJ 2.0.0(NIH)を用いて画像をさらに分析した。
最初のエンドソーム脱出実験は、50nMの葉酸-cy5色素コンジュゲートまたは葉酸-ニゲリシン-Cy5色素コンジュゲートを用いて、Rab5B-GFPを安定的に発現するMDA-MB-231細胞において行った。1時間ごとに24時間まで観察を行った。3時間での細胞の代表的な画像を図3および4に示す。葉酸-cy5コンジュゲートの結果の有意な違いが、内在化から3時間以内にみられ、このとき葉酸-ニゲリシン-cy5色素コンジュゲートで処理した細胞において顕著に大きいエンドソームが観察された。3時間後、葉酸-ニゲリシン-cy5で処理したエンドソームは、葉酸-cy5色素コンジュゲートよりも大きくなり、凝集しプルームを形成し始めた。3時間後、膨潤したエンドソームはより大きな構造に凝集したが、葉酸-cy5色素3のみで処理されたものは実験の過程を通して比較的変化しなかった。図3は、葉酸-Cy5で処理したMDA-MB-231細胞が、処理後3時間で主に細胞膜およびエンドソームから点状にCy5蛍光シグナルを生じたことを示す。しかしながら、葉酸-ニゲリシン-Cy5処理細胞では、大きなエンドソームの形成と細胞質中のCy5の蛍光シグナルのぼやけた分散がみられた(図4)。
アプリオリパワー解析を用いて、統計学的有意性0.05、α<0.5、および検出力80%に必要なサンプルサイズを推定した。パワー計算に基づいて、各処置群に含めるべき推奨動物数を6匹とした。次に、小規模のパイロット試験から予想されるエフェクトサイズを決定した。3匹の動物をFolamiR-34aで処置した後に観察されたRenillaレポーター活性が強力で有意な減少であったため(図7A、B)、残り3匹の動物は処置しなかった。この場合、0.5の検出力によって、α<0.5で3匹の動物を使用して有意性が正確に予測される。他の試験はすべて、最初に計算した6匹の動物を使用した。
上記のとおり培養したFR陽性ヒトMDA-MB-231細胞およびFR陰性ヒトA549細胞をトリプシン処理により剥がし、氷冷リン酸緩衝生理食塩水(PBS;pH7.4)中で2回洗浄し、無血清培地中1×107細胞/密度で再懸濁した。細胞生存率をトリパンブルー排除法により測定し、細胞の生存率が>80%である場合のみ細胞を使用した。次に、標準プロトコルに従ってフローサイトメトリー分析を行った。簡潔に言えば、1×106細胞をPE抗FOLR1抗体(カタログ番号908303、Biolegend)または対照としてマッチアイソタイプ抗体(カタログ番号400213、Biolegend)と共にインキュベートし、LSR Fortessaフローサイトメーター(BD Biosciences, San Jose, CA, USA)を用いてフローサイトメトリー分析により解析した。データは、FlowJoソフトウェアv10(Tree Star、Inc、OR、USA)を用いて解析した。FRの機能性は、第一に、MDA-MB-231およびA549細胞をFolamiR-34a-NIR(50nM)と共にインキュベートした後、上記のようにフローサイトメトリー分析によって確認し、第二に、最終濃度50nMに葉酸-フルオレセインイソチオシアネート(FITC)とインキュベートした細胞の顕微鏡分析によって確認した。1.25倍の対物レンズとOlympus DP80カメラを取り付けたOlympus IX73顕微鏡およびCellSens 1.11を使用して細胞を種々の時点で評価した。
MDA-MB-231トリプルネガティブ乳がん細胞(HTB-26)およびA549非小細胞肺がん細胞(CCL-185)(いずれも、MycoAlert Mycoplasma検出Kit-Lonzaによるマイコプラズマ混入試験によりマイコプラズマ不含を確認済)を、5%CO2、37℃にて、10%ウシ胎児血清(Sigma)、100UmL-1ペニシリンおよび100μgmL-1ストレプトマイシン(Hyclone、GE Healthcare Life Sciences)を添加したRPMI 1640培地(葉酸不含(Life Technologies))中で培養し、維持した。
MDA-MB-231およびA549細胞を500ngのmiR-34a Renillaセンサーでトランスフェクトし、FolamiRと共にインキュベートした。処置の96時間後にRenillaシグナルを測定した。各時点について、データ点をFolamiR-NC(ネガティブコントロールロール:スクランブルmiRNA)に対して正規化した。エラーバーは平均±s.d.を表す。各実験は、各処理につき、少なくとも4回の技術的反復、n=3とし、統計分析は事後ボンフェローニ検定を用いたone-way ANOVAにて行った(**、P<0.01)。
ルシフェラーゼレポーター実験用に、ベクター(psiCHECK, Promega)中のRenillaルシフェラーゼの3’非翻訳領域にmiR-34aに対するアンチセンス配列を挿入することによってmiR-34aセンサープラスミドを作製した。miR-34a特異的サイレンシングは、miR-34aセンサーまたは変異miR-34aセンサーを一過性でトランスフェクトすることによってMDA-MB-231細胞において確認した。Lipofectamine RNAimax(Life Technologies)を使用してmiR-34aセンサー発現細胞をmiR-34a模倣体でトランスフェクトし、外因的なmiRNAによって媒介されるサイレンシングを確認した。安定なクローンを作製するため、MDA-MB-231細胞を1×106細胞/ウェルの密度で6ウェルのプレートに播種し、Lipofectamine 2000(Life Technologies)を使用して2μgのmiR-34aセンサープラスミドでトランスフェクトした。選択マーカーとしてハイグロマイシンB(500μg/mL;Hyclone、GE Healthcare Life Sciences)を用いて安定なクローンを選択した。単一のクローンをRenilla発現について評価し、Renilla発現が最も高いクローンを選択した。
MB-231センサー細胞を、葉酸および無血清RPMI培地中、FolamiR-34a、Fol-SS-34aおよびFolamiR-NC(ネガティブコントロール)を最終濃度50nMで含む96ウェルのプレートに播種した。対照として非処理および非コンジュゲート二本鎖miRNAを含めた。Renilla Glossiferaseキット(Promega)を製造業者の指示に従って使用し、インキュベーションから24時間、48時間、72時間、96時間および120時間後のRenillaルシフェラーゼ値を得た。Renillaレベルを、各時点について、FolamiR-NCに対して正規化した。実験は、各条件につき3連で3回行った。
MDA-MB-231乳癌細胞は、原形質膜上に検出可能なレベルのFRを発現することが報告されており、この細胞株はFolamiR活性を評価するための妥当なモデルである。FRの発現を確認するため、MDA-MB-231細胞とA549細胞(FRネガティブコントロール)を比較するフローサイトメトリー分析を行った。MDA-MB-231細胞は、検出可能なレベルのFRαを発現することが確認された(図5c)。
これらの結果は、フローサイトメトリーを用いてNIR-FolamiR-34aの細胞内取り込みを分析すること(図5d)および蛍光顕微鏡を用いて葉酸フルオレセインイソチオシアネート(Fol-FITC)コンジュゲート取り込みを分析することによって裏付けられた(図5E)。いずれの葉酸コンジュゲートも、FR陽性細胞株MDA-MB-231に取り込まれたが、FR陰性A549細胞株には取り込まれなかった。この観察結果は、miR-34a Renillaセンサーを一過性で発現するMDA-MB-231およびA549細胞株をFolamiRで処理した後、機能的に確認された。このセンサーは、1つのmiR-34a相補的結合部位が後に続くRenilla遺伝子であり、miR-34aによるRenillaの転写後調節のモニタリングを可能にする。両細胞株においてこのセンサーは、トランスフェクトされたmiR-34a模倣体に応答した(図10);但し、このセンサーはFolamiR-34a曝露後のMDA-MB231細胞においてのみ下方制御され(図5F)、FolamiR 標的化はFR発現細胞に依存することを示唆する。これらの結果をまとめると、MDA-MB-231はFR陽性細胞株であることを示唆しており、FR相互作用を介したFolamiRコンジュゲートの特異的取り込みについての予備的証拠となる。
FolamiR-34a活性をモニターするため、MDA-MB-231細胞を作製し、miR-34a Renilla ルシフェラーゼセンサー(MB-231センサー)またはmiR-34aに応答しないこのセンサーの変異体を安定的に発現させた。複数のクローンを作製し、最高レベルのRenillaを有するクローン(図11)を用いてFolamiR-34a活性を評価した。FolamiR-34aまたはFolamiR-SS-34aをMB-231センサー細胞に添加した場合(トランスフェクション試薬の非存在下で)、曝露の72時間後にRenilla活性の低下がみられた(図6a)。Renilla活性は、おそらく細胞内でのFolamiR-34aの複製誘導性の希釈またはFolamiR-34aの減少により、曝露後120時間で回復した。
MB-231センサー細胞の増殖は、1回のFolamiR-34a処理後に減少し(図6B)、これはRenilla活性の減少と相関していた。意外にも、放出可能および放出不能のFolamiRはいずれも細胞に効率的に侵入し、データに基づく活性を保持しており、コンジュゲート化した葉酸がmiR-34a-5pのアルゴノートへのローディングを妨げないことを示唆している。
Lipofectamine 2000(Life Technologies)を使用し、FR陽性ヒトMDA-MB-231細胞およびFR陰性ヒトA549細胞を、500ngのmiR-34aセンサープラスミドでトランスフェクトした。24時間後、細胞(4000個/ウェル)を、葉酸および血清不含RPMI培地中、FolamiR-34aおよび FolamiR-NC(ネガティブコントロール)を最終濃度50nMまたは100nMで含有する96ウェルのプレートに播種した。Lipofectamine RNAimax(Life Technologies)でトランスフェクトした細胞を、miR-34a模倣体に対するmiR-34a Renillaセンサー応答をモニターするための対照として使用した。対照として非処理および非コンジュゲート二本鎖miRNAを含めた。Renilla Glossiferaseキット(Promega)を製造業者の指示に従って使用し、インキュベーションの96時間後にRenillaルシフェラーゼ値を得た。FolamiR-34a処理細胞のRenillaレベルを、各時点についてFolamiR-NCに対して正規化し、非コンジュゲート二本鎖miRNA処理細胞を未処理に対して正規化した。実験は、各条件につき3連で3回行った。Renilla活性の用量依存的減少が、FolamiR-34aで処理した細胞においてのみ観察された(図6c)。
in vitro FR結合競合アッセイは、Van Der Heijden, J. W. et al.(“Folate receptor β as a potential delivery route for novel folate antagonists to macrophages in the synovial tissue of rheumatoid arthritis patients,” Arthritis Rheum. 60, 12-21 (2009)) および Gent, Y. Y. et al.(“Evaluation of the novel folate receptor ligand [18F]fluoro-PEG-folate formacrophage targeting in a rat model of arthritis,” Arthritis Res. Ther. 15, R37 (2013))に記載のとおり行った。簡潔に言えば、FR陽性ヒトMDA-MB-231細胞およびFR陰性ヒトA549細胞を前述のように培養し、トリプシン処理によって剥がし、氷冷リン酸緩衝生理食塩水(PBS;pH7.4)で2回洗浄し、血清不含培地中1×107細胞/mLの密度に再懸濁した)。次に、この細胞懸濁液100μLを、(1~100倍モル過剰の葉酸グルコサミンコンジュゲートの存在下または非存在下で)FolamiR-34a-NIRと共に終濃度50nMまでインキュベートした。細胞を4℃で20分間インキュベートし、氷冷PBSで2回洗浄し、LSR Fortessaフローサイトメーター(BD Biosciences、San Jose、CA、USA)を用いるフローサイトメトリー分析によりFolamiR-34a-NIR結合の置換について分析した。FlowJoソフトウェアv10(Tree Star、Inc、Ashland、Ore)を用いてデータを解析した。
機能的競合は、1~100倍モル過剰の葉酸グルコサミンコンジュゲートの存在下で、FolamiR-34aまたはFolamiR-NCと共に終濃度50nMでインキュベートしたMDA-MB-231センサー細胞を用いて検証した。対照として非処理および非コンジュゲート二本鎖miRNAを含めた。Renillaルシフェラーゼ値は、Renilla Glossiferaseキット(Promega)を製造業者の指示に従って使用し、インキュベーションの96時間後に得た。FolamiR-34a処理細胞のRenillaレベルを、各時点についてFolamiR-NCに対して正規化し、非コンジュゲート二本鎖miRNA処理細胞を未処理に対して正規化した。実験は、各条件につき3連で3回行った。葉酸グルコサミンコンジュゲート(葉酸-グルコサミン)の量が増えると、細胞特異的なNIR-FolamiR-34aシグナルは用量依存的に減少し、葉酸-グルコサミンがNIR-FolamiR-34aと競合することが示された(図6D)。さらに重要なことに、葉酸-グルコサミン処理は、用量依存的に、miR-34a Renillaセンサーに対するNIR-FolamiR-34aのサイレンシング効果を無効にした(図6E)。これらの結果は、FolamiRがFRを過剰発現している細胞に機能的miRNAを送達することができ、その送達がFR発現に依存することを裏付けている。
図6D、Eは、漸増濃度の葉酸グルコサミンコンジュゲートの存在下での、FolamiR-34a(50nM、96時間)に対するmiR-34a Renillaセンサー応答を示す。各実験条件について、データポイントをFolamiR-NC(ネガティブコントロールロール:スクランブルmiRNA)に対して正規化した。実験は、各処理につき少なくとも4回反復しておこない(n=3)、統計分析は事後ボンフェローニ検定を用いてone-way ANOVAで行った(* P<0.05)。
スルホローダミンB(SRB、Sigma)アッセイを、96ウェルプレートにおける細胞増殖の代理として使用した。簡潔に言えば、FolamiR処理後、細胞を完全培地中10%トリクロロ酢酸で固定し、1%酢酸中0.4%(wt/vol)のSRBで1時間染色した。未結合の色素を1%酢酸で4回洗浄することにより除去した。最後に、タンパク質が結合した色素を10mmの非緩衝トリス塩基で抽出し、GloMax Multi+分光光度計(Promega)を用いて510nmの吸光度を測定した。吸光度(細胞量に代わるもの)を、各時点について、FolamiR-NCの存在下で培養した細胞の吸光度に対して正規化した。
単回投与試験のために、200μLのMatrigel(Corning)に懸濁した5×106のMDA-MB-231センサー細胞の皮下注射により、雌性Nu/Nu(NU-Foxn1nu;Charles River)コンジェニックマウス(6週齢、n=5)に皮下腫瘍を誘導した。長期的な研究のために、親のMDA-MB-231細胞を使用した。観察により、げっ歯類は、天然に存在する形態の葉酸である、5-メチル-テトラヒドロ葉酸の高い血漿および組織レベル(ヒトよりも約10倍高い)を示すため、マウスは、腫瘍移植の2週間前と一連の実験期間中、葉酸欠乏食餌(Envigo、TD 95247)で飼育した。葉酸欠乏食餌は、葉酸レベルをヒトで見られる生理的レベルにまで下げることを示した。腫瘍の成長を測定するため、ノギスを用いて個々の腫瘍を測定し、腫瘍体積を次の式によって計算した:腫瘍体積(mm3)=幅×(長さ2)×2-1。処置の時点までに腫瘍が150cm3の体積に達していない場合は動物を除外した。単回投与実験のために、発光および蛍光シグナルの取得後(0日目)、動物に5nmolのFolamiRを静脈内(i.v.)注射した。複数回投与実験では、動物の平均腫瘍サイズの差を最小にすることによって動物を実験群に無作為化した。
腫瘍体積が約200mm3に達したら、指定されたモル濃度のFolamiRを動物に3日ごとに静脈内注射で投与した。すべての実験プロトコルは、Purdue Animal Care and Use Committeeによって承認されており、動物の使用に関するNIHガイドラインに準拠していた。
IVIS Lumina II(Caliper)またはSpectral AMI(Spectral Instruments)を用いてin vivoで腫瘍の生物発光をモニターするため、RediJect Coelenterazine h生物発光基質(PerkinElmer)を製造元のプロトコルに従って投与した。基質の注入後の複数の時点(20分を開始時とした)で発光値を取得し、最大平均放射輝度値のみを報告した。745nm励起でIVIS Lumina II(Caliper)およびICG発光フィルターを使用して赤外線イメージングを実施した。腫瘍のルミネセンスおよび蛍光の非侵襲的な長期的モニタリングは、以下の時点で動物全体の画像化によって実施した:0時間、24時間時間、48時間および72時間(実験群あたりn=3動物)。全体的な臓器画像は、Licor Odyssey CLX(Licor)の800 nmチャンネルを用いて取得した。
マウス特異的サイトカインMulti-Analyte ELISAアレイキット(Qiagen)を製造元の指示に従って用い、複数回投与実験で得た血清試料(最後の注射の24時間後)を用いてIL-6および腫瘍壊死因子(TNFα)濃度を試験した。簡潔に記載すると、血清サンプルを氷上で解凍し、分析前に4℃で10分間1000×gで遠心分離することにより沈殿物を取り除いた。全てのサンプルまたは標準品をアッセイ緩衝液と共に96ウェルプレートに添加した。プレートを穏やかに振盪し、室温で2時間インキュベートした。
上清を除去し、ウェルを洗浄した。検出抗体を添加し、プレートを室温で1時間インキュベートした。プレートをすすぎ、アビジン-HRP溶液と共に室温で30分間インキュベートした。ウェルを洗浄し、GloMaxプレートリーダー(Promega)を用いてデータを取得するため現像液を加えた。450nmおよび570nmで吸光度を測定した。570nmの値を450nmの値から差し引いた。サイトカイン標準曲線を用いて、血清試料中のサイトカイン濃度(pg/ml)を計算した。検出限界は以下の通りであった:IL-6 58.8 pg/mL、およびTNFα 30.5 pg/mL。LPS処置したNu/Nuマウス(NU-Foxn1nu;Charles River)を陽性対照として使用した。マウスに500ng/kgのリポ多糖(LPS、L6529、Sigma)を腹腔内注射し、注射の2時間後に血清を採取した。
Balb/cマウス(8週齢)に33.3nmol、10nmolまたは1nmolのFolamiR-34aを1回静脈内注射した。動物を投与後2週間観察した。マウスの体重の変化および臨床的所見(急速な体重減少、下痢、ボサボサの毛、丸まった姿勢、倦怠、苦しい呼吸、神経学的徴候など)について観察した。マウスには自由に食餌および水を与えた。実験の最後に剖検を行った。さらなる分析のために、全血、血清、および臓器組織を採集した。
各NIR-Folタグ付きmiRNA(NIR-FolamiR)の1回用量(5nmol)を、触診可能なMB-231センサー細胞異種移植片を有する動物に尾静脈注射により投与した。蛍光分布およびルシフェラーゼ活性を、腫瘍細胞標的化の特異性についてモニターするため、ならびに取り込みと細胞内標的の抑制の代理としてそれぞれ測定した。
注射の24時間後、NIR-FolamiRは主に腫瘍組織に保持され、重要なことに、肝臓(図7C、Lv)を含む生物の他の部分からは除去された(図7A、左-NIR)。しかしながら、放出不能なNIR-FolamiR-34aのみがin vivoでRenillaノックダウンを誘導した(図7A、右ルシフェラーゼ、図7Bで定量)。重要なことに、1回の注射のみの後、RenillaレベルはNIR-FolamiR-34a処理後に約50%減少し、これは培養細胞中で観察されたセンサー活性の減少よりもさらに大きかった(図6A、C、E)。NIR-FolamiR-34aで処置した腫瘍には、全RNA 1ナノグラムあたり約3.5×106コピーのmiR-34aが存在した(図7D)。
対照的に、NIR-FolamiR-SS-34aで処置したマウスから採取した腫瘍中のmiR-34aのコピー数は、ネガティブコントロールロールの動物と同様であり、放出可能な葉酸コンジュゲートが循環血において分解するまたは早くに減少し得ることを示唆する。この可能性を検討するため、FolamiRコンジュゲートを50%血清に暴露した。FolamiR-SS-34aは血清の存在下で非常に不安定であり、一方FolamiR-34aは6時間以上無傷のままであった。
FolamiR-34aは、非コンジュゲート型miR-34aよりも安定しているようであり、葉酸が血清ヌクレアーゼからmiRNAを保護することを示唆している。FolamiRがin vivoでFRに特異的に結合するかどうかを決定するため、100倍モル過剰の葉酸-グルコサミンの存在下または非存在下で、5nmolのNIR-FolamiR-34aを、右側の肩にFR陽性ヒトMDA-MB-231センサー細胞を移植し、左側の肩にFR陰性ヒトA549細胞を移植したヌードマウスに静脈内注射した。結果は、FR陽性MDA-MB-231腫瘍がFolamiRコンジュゲートを蓄積する一方、FR陰性A549腫瘍では蓄積せず(図7E、F)、このFR依存性蓄積は過剰の葉酸-グルコサミンによって阻止することができる(図7E、F)ことを示す。
次に、FolamiR-34aの有効性を評価するため複数回投与試験を実施した。MB-231異種移植動物を、用量を減らしたFolamiR-NCまたはFolamiR-34a(0.1nmol、0.5nmolおよび1nmol)で3日毎に合計7回投与して処置した。20日間の投与期間中に対照の葉酸-コンジュゲートを投与した動物の腫瘍は約3.5倍に成長したが、FolamiR-34aで処置した動物の腫瘍サイズの成長は緩やか(約1.5倍)であった(図7Gおよび図12)。0.1ナノモル程度の低い用量でも、腫瘍成長の抑制は有意であった。切除した腫瘍組織中のmiR-34aのコピー数は、対照を投与したマウスから切除した腫瘍よりも約1.5倍高かった(図13A)。重要なことに、FolamiR-34aで処置した動物では、臓器全体の毒性の兆候も血清サイトカインIL-6またはTNF-αの上昇も認められなかった(図14A)。これらの結果は、正常免疫能(immunocompetent)マウスにおいて実施された最大耐量(MTD)試験によって裏付けられ、FolamiR-34aを投与したマウスは、試験した最大用量33.3nmolまで毒性の病理学的徴候も有意の体重変化も認められず(図14B)、MTD>33.3nmolを示した。
KrasLSL-G12D/+における腫瘍形成の誘導;Trp53flx/fl(FVB.129バックグラウンド)二重突然変異体マウス(6~10週齢)を用いて、DuPage, et al. (DuPage M, Dooley AL, Jacks T. Conditional mouse lung cancer models using adenoviral or lentiviral delivery of Cre recombinase. Nat. Protoc. 2009; 4: 064-1072)の方法に基づいて行った。簡潔に言えば、肺特異的導入遺伝子活性化を、Creリコンビナーゼをコードするアデノウイルス粒子(106PFU)の気管内送達により行った。実験前の8週間、腫瘍を予備形成させた。
誘導および非誘導動物のMRIスキャン(正常組織のスキャン)は、パーデューMRI施設にて、7.0 Tesla Bruker Biospec 70/30 USRスキャナー(Billerica, MA)および40mmマウスボリュームコイルを用いて得た。動物をO2中2.5%v/vのイソフルランを用いて5分間麻酔した後、温めた動物用ベッドに移し、麻酔を2%に設定した。呼吸数は圧力センサーでモニターした。以下のパラメータを用いて、低解像度マルチプレーンスカウトスキャンを行った:TR=4s、TE=1.5ms、FOV=30×30mm2、スライス厚=1mm、データマトリックス=256×256、7スライス/面(軸方向、冠状および矢状)、マウスあたりのスキャンのおおよその時間=1分。スカウトスキャンを用いてマウスの背骨を整列させ、以下のパラメータを用いて肺の高解像度画像を収集した:TR=4s、TE=1.5ms、FOV=30×30mm2、スライス厚=0.5mm、データマトリックス=256×256、30スライス/面(軸方向および冠状)、マウス当たりのおおよそのスキャン時間=5分。
腫瘍量の定量は、Krupnick et al (“Quantitative monitoring of mouse lung tumors by magnetic resonance imaging, ” Nat. Protoc., 2012, 7, 128-142)に記載されている手動セグメンテーションプロトコルに従って行った。MRIによる腫瘍量のこの分析は、腫瘍組織(明るい)と正常な肺組織(暗い)との間のMR画像強度の大きな差を利用するもので、平均肺画像強度を腫瘍量の代理として用いるものである。簡潔に言えば、肺のMR画像をImageJ 2.0.0を用いて手動でセグメント化し、同じ動物内の肝臓のそれに対して正規化された平均の肺画像の強度を計算する。動物内の腫瘍の進行を決定するために、平均肺画像強度を±s.d.の初日に対して正規化する。さらに、ITK-Snap and Paraview 5.2ソフトウェア(Kitware、NY、USA)を用いた三次元再構成法を用いて、動物あたりの腫瘍と全体の肺の体積を計算した。示された時間での腫瘍/肺全体の比を求め、腫瘍によって占められている肺の体積の割合(%)を示した。
葉酸取り込み試験のために、5nmolのOTL38(On Target Laboratories, LLC, West Lafayette, INにより提供、現在臨床試験中の(臨床試験ID:NCT02769533)蛍光近赤外(NIR)色素に結合した葉酸からなる蛍光イメージングコンジュゲート)を、健常マウスおよび腫瘍保有マウスに尾静脈からシステマティックに送達した(n=3;導入遺伝子活性化の8週間後)。注射から24時間後に、動物を殺して生理食塩水で灌流した。Licor Odyssey CLX(Licor)の800 nmチャンネルを使用して臓器全体の画像を取得した。肺を10%緩衝ホルマリン中で固定し、標準的な手順に従ってパラフィン包埋した。切片をヘマトキシリンおよびエオシン(H&E)で染色し、1.25倍の対物レンズとOlympus DP80カメラを取り付けたOlympus IX73顕微鏡およびCellSens 1.11を用いて評価した。腫瘍量をImageJ 2.0.0を用いて計算し、各動物について3つの独立した切片から得られた肺全体の面積に対する腫瘍面積で表した。染色していない切片をマウントし、Licor Odyssey CLX(Licor)の800 nmチャンネルで、20倍対物レンズ、ICGバンドパスフィルター(Ex:780-800;Ex:810-860;Semrock、 Brightline)、キセノン/水銀光源(Nikon、日本)、Photometrics QuantEM EMCCDカメラ、およびNIS-Elements(Nikon、日本)を取り付けたNikon TiS顕微鏡を用いて評価した。
このモデルはFR発現についてまだ検証されていないため、まず、このモデルの肺腺癌を葉酸受容体発現、ならびに腫瘍に特異的な葉酸コンジュゲートの取り込みおよび保持について評価した。近赤外蛍光(NIR)色素とコンジュゲートさせた葉酸受容体-α(FRα)を標的とするリガンドである、蛍光イメージングリガンドOTL38を、肺腫瘍を有するマウスまたは健常なマウスに静脈内投与した。
臓器全体のレベル(図8A)および組織学的レベル(図8B)で観察されるように、葉酸コンジュゲートは、肺腫瘍において優先的に保持され、正常な健康な組織からは排出された。より高倍率の画像は、近赤外のシグナルが、健康な組織と悪性の組織との間の細胞密度差によるものではないことを示している;OTL-38の受容体介在のエンドサイトーシスに起因して前に観察されたように、明確な点状シグナル伝達がOTL38投与後に腫瘍で観察される(図8Bの挿入図を参照)。肺腺癌におけるOTL38保持がFRとのその相互作用によって媒介されるかを確かめるため、in vivo遮断アッセイを実施した。肺腫瘍を有するマウスにおいて、100倍モル過剰の葉酸-グルコサミンの存在下または非存在下で、OTL38(5 nmol)を静脈内注射した。肺腫瘍におけるOTL38の優先的保持は、過剰の葉酸-グルコサミンによって減少し(図8C、D)、これは腫瘍におけるOTL-38の蓄積がFRに依存することを示唆している。これらのデータによって、KrasLSL-G12D/+;Trp53flx/flx腫瘍がOTL-38を特異的に取り込み保持することが確認され、これはFolamiR-34aが同様に腫瘍組織中に蓄積することを示唆している。
FolamiR-34aによる複数回投与実験のため、腫瘍を有する動物(8週齢)を、それらのMRI測定腫瘍量の差を最小にすることによって実験群に無作為化した。動物に1nmolのFolamiRを3日ごとに静脈内注入(合計10回の注入)して処置し、上記のように7.0 Tesla Bruker Biospec 70/30 USR スキャナー(Billerica, MA)を用いて腫瘍の進行を1週間毎に4週間にわたってモニターした。最後の注射から24時間後に、動物を屠殺して生理食塩水で灌流した。肺を摘出し、10%緩衝ホルマリン中に固定し、標準的な手順に従ってパラフィン包埋した。切片をヘマトキシリンおよびエオシン(H&E)で染色し、前述のように評価した。マウスは、腫瘍の誘導から6週目から始めて一連の実験期間中は葉酸欠乏食餌(Envigo、TD 95247)で飼育した。すべての実験プロトコルは、Purdue Animal Care and Use Committeeによって承認されており、動物の使用に関するNIHガイドラインに準拠していた。
雌性Nu/Nu(NU-Foxn1nu;Charles River)コンジェニックマウス(6週齢、n=3)に、200μLのMatrigel(Corning)に懸濁した、5×106FR陽性ヒトMDA-MB-231センサー細胞(右側)およびFR陰性ヒトA549細胞(左側)を注射し、皮下腫瘍を誘導した。腫瘍を形成させ、同様の大きさのA549およびMDA-MB-231腫瘍を有するマウスを実験に含めた。Kras;p53マウスモデルについては、導入遺伝子活性化から8週間腫瘍を形成させた(n=3)。腫瘍形成はMRIを用いてモニターした。5nmol(≧100倍モル過剰)の葉酸グルコサミンコンジュゲートの存在下または非存在下、異種移植モデルについては5nmolのFolamiR-34a-NIRを、Kras;p53マウスモデルについては5nmolのOTL38を、尾静脈を介し同時投与することにより競合試験を行った(n=3)。葉酸グルコサミンコンジュゲートは、葉酸と比較して低pHでの溶解度が高いため、腎臓での析出を防ぐために使用した。in vivoでの動物全体の画像化およびex vivoでの組織分布実験を上記のように実施した。
Nu/Nuマウス(NU-Foxn1nu;Charles River)由来のMDA-MB-231由来腫瘍およびKrasLSL-G12D/+;Trp53flx/flxマウス由来の肺腫瘍をRNA Later(Life Technologies)に回収し、-80℃で保存した。腫瘍組織(50mg)を、700μLのQIAzol溶解試薬(Qiagen)と1.4mmのセラミックビーズが入った2mLのコレクションチューブに入れた。サンプルをビーズミル(Fisher Scientific)を用いて4m s-1にて3分間粉砕した。RNeasy Mini Kit(Qiagen)を製造業者の指示に従って用い全RNAを抽出した。次に、1μgのトータルRNAを用い、miScript II RT Kit(Qiagen)とmiScript HiFlex Bufferを用いてcDNAを作製した。miR-34a標準の生成のため、miR-34a模倣体(Life Technologies)をcDNA合成に使用した。qRT-PCRは、miRNAプライマーアッセイ(Qiagen)を用いて実施した。以下のサイクリング工程の下で、miScript SYBR Green PCR Kit(Qiagen)を使用するQuantStudio 6 Flex リアルタイムPCRマシン(Life Technologies)を用いて反応を行った:95℃で15分;95℃で15秒間、55℃で30秒間、70℃で30秒間を40サイクル;1.6℃/秒で95℃から60℃の融解曲線。各生物学的複製について技術的反復(少なくとも3回)を実施した。1×108コピーから1×103コピーをカバーする標準曲線を用いてmiR-34aコピー数を決定した。
二群間解析のため、両側スチューデントのt検定を用いて群間差を調べた。Prism統計パッケージ(バージョン7、GraphPad Software)を用いた多群間比較のために、事後ボンフェローニ検定によるtwo-wayまたはone-way分散分析(ANOVA)を用いた。図の説明で示したように、エラーバーは、平均±s.d.または平均±s.e.m.のいずれかで表す。統計的に有意なP値は、図および/または凡例において、***、P<0.005;**、P<0.01;*、P<0.05として示す。
KrasLSL-G12D/+;Trp53flx/flx腫瘍がFolamiR-34aに応答性であるかどうかを調べるために、腫瘍を有する動物に3日ごとに1ナノモルのFolamiR-34aを合計10回投与した。実験期間中、腫瘍の成長をMRIによりモニターし、MRIデータから作成して得られた体積の測定値は、対照の葉酸-コンジュゲートを投与した動物と比較してFolamiR-34aが腫瘍の成長を減少させることを示している(図9A、B、C)。腫瘍のサイズは、FolamiR-34aを投与した動物では統計的に変化がなかったが、FolamiR-NCで処置したマウスの腫瘍サイズは投与初日と比較して1.5倍に増大した。試験の終了時に組織学的レベルで同様の応答が観察された(図9D、E)。腫瘍量の定量は統計的有意性を示しており、FolamiR-34aで処置マウスから採取した肺において、FolamiR-NCで処置マウスと比較して、腫瘍体積の1.8倍の減少を示した。
MD-MBA-231異種移植片において観察されたmiR-34aコピー数の増加と同様に、切除された肺腫瘍組織におけるmiR-34aのコピー数は、対照を投与されたマウスから抽出された腫瘍における値より約3倍高かった(図13B)。miR-34aが、miR-34a標的の内在性標的遺伝子の転写レベルを抑制するよう作用していることを検証するために、BCL-2、METおよびMYCを定量した。BCL-2とMYCはいずれも、FolamiR-34aを投与したマウスから採取した腫瘍では統計的に下方制御されており、内在性標的遺伝子に対するmiR-34a活性が確認された(図9G)。

- 治療剤または造影剤が癌に対して標的化されている、請求項16に記載の方法。
- 癌が、卵巣癌、肺癌、乳癌、子宮内膜癌、脳腫瘍、腎臓癌、前立腺癌および大腸癌からなる群から選択される、請求項17に記載の方法。
- 治療剤が炎症部位に標的化されている、請求項16に記載の方法。
