Abstract
Description
本願は、「VIRAL VECTORS FOR TREATING PARKINSON’S DISEASE」の表題で2016年7月21日に出願された米国仮特許出願第62/365,316号の優先権を主張し、その開示は、参照によって本明細書中に組み込まれる。

本開示の態様では、本開示は、レンチウイルス粒子を発現させるためのレンチウイルスベクター系を提供する。この系は、PARPファミリーメンバーの発現を阻害するためのshRNAを含む、治療用ベクターを含む。多数のPARPファミリーメンバーが存在し、本開示は、いずれか1つの特定のPARPファミリーメンバーに限定されない。しかしながら、実施形態では、レンチウイルスベクター系は、PARP1を特異的に阻害する。
レンチウイルスビリオン(粒子)は、ビリオン(ウイルス粒子)を産生するために必要なウイルスタンパク質をコードするベクター系によって発現される。プロモーターに作動可能に連結した、逆転写および組込みに必要なレンチウイルスpolタンパク質をコードする核酸配列を含有する少なくとも1つのベクターが存在する。別の実施形態では、polタンパク質は、複数のベクターによって発現される。
投薬は、1日1回であってもよく、または1日数回であってもよい。投薬は、投薬間に間隔をあけて行ってもよい。例えば、被験体は、初日に処置を受け、次いで、1日おきに、または2日に1回、または3日に1回、または4日に1回、または5日に1回、または6日に1回、または7日に1回、または2週間に1回、または1カ月に1回など、処置を受けてもよい。しかしながら、投薬は、1年に1回、2回、または数回であってもよく、そのような投薬スケジュールが、年基準で繰り返されてもよい。レンチウイルス粒子は、PDと関連する症状を処置するのに好適な任意の方法によって送達することができる。例えば、投薬は、ガイドされたニードルを使用した、脳幹への直接的な注射によって行われてもよい。これは、脳深部刺激と併せて行われる可能性が高いであろう。

レンチウイルスベクター系を、概して図1に要約されるように、開発した。レンチウイルス粒子は、293T/17 HEK細胞(American Type Culture Collection、Manassas、VAから購入)において、治療用ベクター、エンベローププラスミド、およびヘルパープラスミドのトランスフェクションの後に、産生させた。機能的ウイルス粒子を産生した293T/17 HEK細胞のトランスフェクションには、プラスミドDNAの取込み効率を増加させるために、試薬ポリ(エチレンイミン)(PEI)を利用した。プラスミドおよびDNAは、まず、3:1の比(PEI対DNAの質量比)で、血清不含の培養培地に、別個に添加した。2~3日後に、細胞培地を採取し、レンチウイルス粒子を、高速遠心分離および/または濾過、続いて、アニオン交換クロマトグラフィーによって精製した。レンチウイルス粒子の濃度は、形質導入単位/ml(TU/ml)の単位で表すことができる。TUの決定は、培養液中のHIV
p24のレベルを測定し(p24タンパク質は、レンチウイルス粒子に組み込まれる)、定量的PCR、または細胞に感染して光を使用すること(ベクターがルシフェラーゼもしくは蛍光タンパク質マーカーをコードする場合)により、細胞1個当たりのウイルスDNAコピー数を測定することによって達成した。
材料および方法:
ヘルパープラスミドの構築:ヘルパープラスミドを、まずは、Gag、Pol、およびインテグラーゼ遺伝子を含有するpNL4-3 HIVプラスミド(NIH Aids Reagent Program)に由来するDNA断片のPCR増幅によって、構築した。プライマーは、pCDNA3プラスミド(Invitrogen)において同じ部位に挿入するために使用することができる、EcoRIおよびNotI制限部位を有する断片を増幅するように設計した。フォワードプライマーは、(5’-TAAGCAGAATTC ATGAATTTGCCAGGAAGAT-3’)(配列番号41)であり、リバースプライマーは、(5’-CCATACAATGAATGGACACTAGGCGGCCGCACGAAT-3’)(配列番号42)であった。
水疱性口内炎Indianaウイルス糖タンパク質(VSV-G)配列を、隣接するEcoRI制限部位を用いて、MWG Operonによって合成した。次いで、DNA断片を、pCDNA3.1プラスミド(Invitrogen)のEcoRI制限部位に挿入し、正しい配向を、CMV特異的プライマーを使用したシーケンシングによって決定した。DNA配列は、以下の通りであった。
材料および方法:
Revを有さないヘルパープラスミドの構築:
Revを有さないヘルパープラスミドを、RREおよびウサギベータグロビンポリA配列を含有するDNA断片を挿入することによって、構築した。この配列は、隣接するXbaIおよびXmaI制限部位を用いて、MWG Operonによって合成した。RRE/ウサギポリAベータグロビン配列を、次いで、ヘルパープラスミドのXbaIおよびXmaI制限部位に挿入した。DNA配列は、次の通りである。
RSVプロモーターおよびHIV Rev配列を、隣接するMfeIおよびXbaI制限部位を用いて、MWG Operonによって、単一のDNA断片として合成した。DNA断片を、次いで、CMVプロモーターがRSVプロモーターと置き換えられているpCDNA3.1プラスミド(Invitrogen)のMfeIおよびXbaI制限部位に挿入した。DNA配列は、以下の通りであった。
この実施例の目的は、PARP1阻害剤RNAレンチウイルスベクターを開発することであった。
Homo sapiens PARP1に対して設計されたshRNAを、PARP1遺伝子の発現を下方制御するそれらの能力に関して試験した。ヒトPARP1 shRNAを含有するレンチウイルスベクターを、レンチウイルス粒子としてパッケージングした。1~10のMOIのレンチウイルス粒子を、ヒトU251膠芽腫細胞に添加した。48時間後に、細胞を溶解させ、PARP1の発現を、PARP1特異的抗体を用いた免疫ブロット分析によって、測定した。
本明細書において概説されるレンチウイルスベクター系は、マウスニューロンにおける形質導入が可能であることが見出されている。図4を参照すると、野生型マウスに、マウスの脳の黒質領域に挿入したスチール製のニードルを介して、顕微鏡写真の左列では模擬(レンチウイルスなし)を注射し、顕微鏡写真の右列では緑色蛍光タンパク質も発現するLV-shPARP1を注射した。LV-shPARP1-GFPは、およそ1×108形質導入単位を含有する0.1mlで投薬した。14日後に、マウスを殺処分し、黒質領域を脳から切除し、ホルムアルデヒド中に固定し、パラフィンに包埋した。薄切片を、スライドガラスに載せ、蛍光顕微鏡で可視化した。TH+ニューロン(高いレベルのチロシンヒドロキシラーゼを発現する)は、概して黒質領域を特定し、図4において赤色(またはグレースケール写真では白色)に見える。中央のパネルは、模擬(左列)またはLV-shPARP1-GFP(右列における緑色[またはグレースケール写真では白色]の染色を参照されたい)を形質導入した細胞を示す。効率的な形質導入および導入遺伝子の発現を示すGFPによって放出される高い強度の光のため、陽性に形質導入されたニューロンはこの図では黒色に見え、予測した通り、偽対照(左列)には存在していなかった。下のパネルは、レンチウイルス形質導入に由来するTH+ニューロンの染色およびGFP+ニューロンの染色の合体であり、TH+ニューロン内を含め、黒質内の形質導入された細胞の存在を示す。
化学的な神経毒素1-メチル-4-フェニル-1,2,3,6-テトラヒドロピリジン(MPTP)は、マウスにおいて重度かつ不可逆的な運動異常を引き起こし、ヒトPDをモデリングするために広く使用されている。例えば、KopinおよびMarkey、11巻、Annu. Rev. Neurosci、81~96頁(1988年)を参照されたい。MPTPでマウスを処置することにより、線条体ドーパミンおよびその代謝産物のレベルが低下するが、これは、薬物の神経毒性により、黒質におけるドーパミン産生細胞の数が低減するためである。このモデルは、MPTP曝露後のニューロン死を予防する、一酸化窒素を含む化合物の保護効果を試験するために使用されている。例えば、Przedborskiら、93巻、Proc. Natl. Acad. Sci. U.S.A.、44565~4571頁(1996年)を参照されたい。このモデルを使用して、PARPのニューロン細胞での発現を低減させるであろうショートヘアピンRNA配列(sh)を発現するように設計された、レンチウイルスベクター(LV-shPARP)で、マウスを事前処置することによって、ドーパミン作動性ニューロンの死滅を予防する能力を測定することができる。このベクターは、形質導入された細胞を特定する緑色蛍光タンパク質マーカーを発現するように、さらに修飾されてもよく(LV-shPARP-GFP)、shPARPを発現しないベクター(LV-GFP)と比較される。
個々のエンベロープ糖タンパク質の特性は、組織の指向性および疾患の部位への治療用遺伝子の送達の効率に影響を及ぼす。PDを処置するために、遺伝子療法の標的は、黒質のTH+細胞である。TH+細胞への標的化を最適化するために、様々なエンベロープ糖タンパク質を、マウス黒質のTH+細胞への形質導入効率を改善するそれらの役割に関して、比較する。実施例1において上述のように、水疱性口内炎ウイルスG糖タンパク質(VSV-G)を含有するエンベローププラスミドを設計し、産生させた。このエンベローププラスミドを、VSV-Gの代わりに、FUG-C(狂犬病ウイルス糖タンパク質のN末端領域)、バキュロウイルスに由来するgp64エンベロープ糖タンパク質、ヒヒ内在性ウイルスに由来するエンベロープ糖タンパク質、またはレンチウイルス粒子をパッケージングするのに好適な他の代替物を含む、設計された他のエンベローププラスミドと比較することができる。それぞれの事例において、エンベローププラスミドのバリアントを使用して、レンチウイルスベクターストックを産生させ、マウスの脳に注射し、マウス黒質のTH+細胞への形質導入の効率性を、試験する。
本明細書において記載される研究には、PARP1、およびその調節をどのようにしてPDを治療的に処置するために使用することができるかに焦点を当てたものが含まれる。しかしながら、PARP1は、同様の機能を有するおよそ16個の緊密に関連するPARP遺伝子のうちの1つに過ぎない。本明細書において記載される、標的の特定、shRNAの産生およびレンチウイルスにより送達されるmiRNAへの変換のための技法を使用して、他のPARP遺伝子を、PDを処置することに有効な治療用ベクターとなり得るそれらの能力に関して、試験することができる。簡単に述べると、他のPARP遺伝子を含有するレンチウイルスベクターを、マウスに注射して、本明細書において記載される方法、技法、および材料を使用して、PDの補正に関して試験することができる。
19~22ヌクレオチドの長さである、標的ショートヘアピン配列を、shRNA設計プログラム、例えば、例として、InvitrogenのBlock-iT RNAi designerまたはBroad InstituteからのRNAi設計プログラム(MIT)から選択する。数個の配列を、特定の遺伝子、例えば、例として、PARPの効率的なノックダウンに関して試験する。標的遺伝子の発現を少なくとも80%減少させるshRNA配列を、次いで、規定のマイクロRNAヘアピン骨格内に挿入する。マイクロRNA(miRNA)ヘアピン構造はmiRBase.orgのウェブサイトから入手することができる。
PDの初期診断から少なくとも5年後に、年齢が35~75歳の12人の患者に、用量漸増研究において、本明細書において記載される(cGMPグレード)LV-shPARP組成物(例えば、図1Bに示されるレンチウイルス構築物に基づく)を、両側定位的被殻内(intraputaminal)注射で受容させた。可能性の高い用量範囲は、5mlの滅菌生理食塩水中、108形質導入単位のLV-shPARPである[1形質導入単位が、平均で、単一の標的細胞の染色体に1コピーの導入遺伝子が組み込まれるのを達成するのに必要なLV-shPARPの量である]。範囲の上限は、およそ1010形質導入単位のLV-shPARPであることが予測される。処置した患者を、少なくとも1年間から最長5年間、自発運動状態の変化に関して追跡する。
