T細胞受容体(TCR:T cell receptor)
国際免疫遺伝学情報システム(IMGT)を使用してTCRを記述することができる。天然αβヘテロ二量体TCRは、α鎖とβ鎖を有する。広義的には、各鎖は、可変領域、連結領域及び定常領域を含み、β鎖は、通常は、可変領域と連結領域との間に短い可変領域を含むが、当該可変領域は、一般的に連結領域の一部として考慮される。独特のIMGTのTRAJとTRBJでTCRの連結領域を決定し、IMGTのTRACとTRBCでTCRの定常領域を決定する。
天然TCRの膜近位領域CαとCβ鎖の間にジスルフィド結合が存在し、本発明では「天然鎖間ジスルフィド結合」と称する。本発明において、人工的に導入されて、天然鎖間ジスルフィド結合の位置と異なる位置を有する鎖間の共有ジスルフィド結合を「人工鎖間ジスルフィド結合」と称する。
用語「腫瘍」とは、病理学的タイプまたは感染の段階に関係なく、すべてのタイプの腫瘍細胞生長または発がん過程を含む転移組織または悪性形質転換細胞、組織、または器官を指す。腫瘍の実施例は、非制限的に、固形腫瘍、軟組織腫瘍、および転移性病変を含む。固形腫瘍の実施例は、肉腫、扁平上皮癌及び癌のような異なる器官系統の悪性腫瘍を含む。例えば、感染した前立腺、肺、乳房、リンパ、腸胃(例えば、結腸)及び尿生殖路(例えば、腎臓、上皮細胞)、咽頭を含む。肺扁平上皮癌は、ほとんどの結腸癌、直腸癌、腎細胞癌、肝臓癌、肺部の非小細胞癌、小腸癌及び食道癌のような悪性腫瘍を含む。前記癌の転移性病変は、同様に、本発明の方法と組成物により治療及び予防することができる。
周知のように、TCRのα鎖可変ドメインとβ鎖可変ドメインは、それぞれ3つのCDRを含み、抗体の相補決定領域と類似している。CDR3は、抗原オリゴペプチドと相互作用し、CDR1及びCDR2は、HLAと相互作用する。従って、TCR分子のCDRが、それと抗原オリゴペプチド-HLA複合体の相互作用を決定した。抗原オリゴペプチドSLLMWITQCとHLA A2複合体(即ち、SLLMWITQC-HLA A2複合体)を結合することができる野生型TCRのα鎖可変ドメインアミノ酸配列とβ鎖可変ドメインアミノ酸配列は、それぞれ配列番号:1と配列番号:2であり、当該配列は、本発明者が初めて発見した。これは、下記のCDR領域を有する。

他の一つの好ましい例において、SLLMWITQC-HLA A2複合体に対する前記TCRの解離平衡定数は、5μM≦KD≦10μMであり、好ましくは、0.1μM≦KD≦1μMであり、さらに好ましくは、1nM≦KD≦100nMであり、
他の一つの好ましい例において、SLLMWITQC-HLA A2複合体に対する前記TCRの解離平衡定数は、100pM≦KD≦1000pMであり、好ましくは、10pM≦KD≦100pMであり、さらに好ましくは、1pM≦KD≦10pMである。
本発明において、Pro60または60Pは、すべて60番目のプロリンを示す。なお、本発明による変異体の具体的な形態の表現方法は、例えば、「T27G」は、27番目のTがGで置換されたことを表し、同様に、「I29A/V」は29番目のIがAで置換され、またはVに置換されたことを表す。他のものもそれに応じて類推する。
(1)前記SLLMWITQC-HLA-A2複合体に対する本発明のTCRの親和力および/または結合半減期は、少なくとも野生型TCRの2倍、好ましくは、少なくとも10倍である。
(2)前記SLLMWITQC-HLA-A2複合体に対する本発明のTCRの親和力および/または結合半減期は、少なくとも野生型TCRの100倍であり、好ましくは、少なくとも1000倍であり、さらに好ましくは、104~107倍に達することができる。
(3)本発明の高親和力TCRを形質導入させる効果細胞は、標的細胞に対して非常に強い殺傷作用がある。
次の具体的な実施例では、本発明をさらに説明する。これらの実施例は、単に本発明を説明するためのものであり、本発明の範囲はこれに限定されないことを理解しなければならない。下記の実施例において、具体的な条件を示していない実験方法は、例えば、(SambrookおよびRussell等、分子クローン:実験室ハンドブック(Molecular Cloning-A Laboratory Manual)(第3版)(2001)CSHL出版社)に記載された条件、またはメーカーから提案する条件のような通常の条件に従う。他の説明がなければ、割合と部数は重量に基づいて計算される。
本発明の実施例に使用された実験材料について特別な説明がなければ、すべての市販で入手することができて、ここで、E.coli DH5αはTiangenから購入され、E.coli BL21(DE3)はTiangenから購入され、E.coli Tuner(DE3)はNovagenから購入され、プラスミド(Plasmid)pET28aはNovagenから購入される。
本発明は、部位特定突然変異の方法で、特許文献WO2014/206304による記載に基づいて、1つの柔軟なオリゴペプチド(linker)によってTCRαとβ鎖可変ドメインを連結して構成された安定的な単鎖TCR分子を構築し、そのアミノ酸とDNA配列は、それぞれ配列番号:54と配列番号:55であり、図7a及び図7bに示された通りである。当該単鎖TCR分子を鋳型で高親和性TCR分子をスクリーニングした。当該鋳型鎖のα可変ドメイン(配列番号:3)とβ可変ドメイン(配列番号:4)のアミノ酸配列は、図2a及び図2bに図示された通りであり、これに対応されるDNA配列は、それぞれ配列番号:5と6であり、図3a及び3bに示された通りであり、柔軟なオリゴペプチド(linker)のアミノ酸配列とDNA配列は、それぞれ配列番号:7と8であり、図4a及び4bに示された通りである。
実施例1で製造された組換えプラスミドpET28a-鋳型鎖を含有するBL21(DE 3)ギュンラクをカナマイシンを含有するLB培地に完全に接種し、OD600が0.6~0.8になるまで37℃の温度で培養し、IPTGを入れて最終濃度を0.5mMにして、37℃の温度で継続して4時間培養した。5000rpMで15分間遠心分離して細胞沈殿物を得て、Bugbuster Master Mix(Merck)で細胞沈殿物を分解させ、6000rpMで15分間遠心分離して封入体を回収し、再びBugbuster(Merck)で洗浄し、細胞破壊、水と膜成分を除去し、6000rpMで15分間遠心分離して封入体を収集した。封入体を緩衝液(20mM Tris-HCl pH 8.0、8Mの尿素)に溶解させ、高速遠心分離して不溶性物質を除去し、上清液をBCA法で定量して分割入れて、-80℃の温度下で保管した。
BIAcore分析
BIAcore T200リアルタイム分析システムとしてTCR分子とSLLMWITQC-HLA-A2複合体の結合活性を測定した。抗ストレプトアビジン(streptavidin)の抗体(GenScript)のカップリングバッファ(Coupling buffer)(10mMの酢酸ナトリウム(Sodium acetate)緩衝液、pH4.77)を入れた後、抗体を事前にEDCとNHSによって活性化されたCM5チップに流して、抗体をチップ表面に固定させ、最後にエタノールアミン(Ethanolamine)の塩酸溶液で未反応活性表面を遮断して、カップリング処理を完了しており、カップリングレベルは、約15000RUである。
100mlの重鎖または軽鎖の発現を誘導するE.coli菌液を収集し、4℃の温度下で8000gで10分間遠心分離した後、10mlのPBSで菌体を1回洗浄し、次に5mlのBugBuster Master Mix Extraction Reagents(Merck)で激しく振とうして菌体を再浮遊させ、室温下で20分間スピン培養した後、4℃、6000gの条件下で15分間遠心分離させ、上清を捨て、封入体を収集した。
合成されたオリゴペプチドSLLMWITQC(北京賽百盛遺伝子技術有限公司)を20mg/mlの濃度でDMSOに溶解した。軽鎖と重鎖のボンイプチェを8Mの尿素、20mMのTris pH8.0、10mMのDTTで溶解させ、再生前に3Mの塩酸グアニジン(Guanidine hydrochloride)、10mMの酢酸ナトリウム、10mMのEDTAを入れてさらに変性させた。SLLMWITQCペプチドを25mg/L(最終濃度)で再生緩衝液(0.4M L-アルギニン、100mM Tris pH8.3、2mM EDTA、0.5mM酸化グルタチオン(Glutathione)、5mM還元グルタチオン、0.2mM pMSF、4℃まで冷却)に入れた後、20 mg/Lでの軽鎖と90mg/Lでの重鎖(最終濃度であり、重鎖を3回に分けて入れ、8h/回)を順次入れ、再生は4℃の温度で少なくとも3日間に完成し、SDS-PAGEで正常に再生されたかどうかを測定した。
10体積の20mMのTris pH8.0で透析して再生緩衝液を変えて、緩衝液を少なくとも2回変えて溶液のイオン強度を減らした。透析後、0.45μmのセルロースアセテート(Cellulose acetate)膜タンパク質溶液を濾過した後、HiTrap Q HP(GEゼネラルエレクトリックカンパニー)陰イオン交換カラム(5mlバッドボリューム(Bed Volume))に入れた。Akta精製器(GEゼネラルエレクトリックカンパニー)で、20mMのTris pH8.0で製造された0~400mMのNaClの線形勾配でタンパク質を溶出し、pMHCが約250mMのNaClで溶出し、ピーク成分を収集し、SDS-PAGEで純度を測定した。
Millipore超濾過チューブで精製されたpMHC分子を濃縮するとともに、緩衝液を20mMのTris pH8.0に置換した後、ビオチン化試薬0.05MのBicine pH8.3、10mMのATP、10mMのMgOAc、50μMのD-Biotin、100μg/mlのBirA酵素(GST-BirA)を入れて、室温下で混合液を一晩培養し、SDS-PAGEでビオチン化が完了したかどうかを検出した。
Millipore超濾過チューブでビオチン化標識されたpMHC分子を1mlに濃縮し、ゲル濾過クロマトグラフィーでビオチン化されたpMHCを精製し、Akta精製器(GEゼネラルエレクトリックカンパニー)を使用して、濾過されたPBSでHiPrepTM16/60 S200 HRカラム(GEゼネラルエレクトリックカンパニー)を前平衡させ、1mlの濃縮後のビオチン化pMHC分子を入れた後、PBSで1ml/minの流速で溶出した。ビオチン化されたpMHC分子は、約55mlの単一ピーク溶出であった。タンパク質が含有された成分を併合して、Millipore超濾過チューブで濃縮し、BCA法(Thermo)でタンパク質濃度を測定し、プロテアーゼ(Protease)阻害剤cocktail(Roche)を入れてビオチン化されたpMHC分子を分けて-80℃の温度下で保管した。
実施例4.高親和性単鎖TCRの生成
ファージディスプレイ技術は、高親和力変異体についてスクリーニングするためのTCR高親和力変異体ライブラリーを生成する手段である。Li等((2005)Nature Biotech23(3):349-354)に記載されたTCRファージディスプレイ及びスクリーニング方法を実施例1の単鎖TCR鋳型に応用した。当該鋳型鎖のCDR領域を変異させて高親和性TCRのライブラリーを構築し、選択した。いくつかのラウンドに経て選択されたファージのライブラリーは、すべて対応する抗原に特異的に結合され、その中で、単一のクローンを選択して、配列解析を行った。
実施例4でスクリーニングされた高親和力の単鎖TCRのCDR領域の変異をαβヘテロ二量体TCRの可変ドメインの対応するサイトに導入し、BIAcoreを介しSLLMWITQC-HLA-A2複合体との親和力を測定した。前記CDR領域と親和力突然変異の導入は、当業者に公知された部位の特定の変異体の方法を使用した。前記野生型TCRのα鎖とβ鎖可変ドメインのアミノ酸配列はそれぞれ図1a(配列番号:1)と1b(配列番号:2)に示す通りである。
TCRαとβ鎖の発現ベクターをそれぞれ化学的形質転換法により発現細菌BL21(DE3)に形質転換させ、細菌をLB培養液に生長させ、OD600=0.6時の最終濃度が0.5mMであるIPTGで誘導し、TCRのαおよびβ鎖で発現させた後、形成された封入体をBugBuster Mix(Novagene)で抽出し、BugBuster溶液に何度も繰り返して洗浄し、最終的に封入体を6Mの塩酸グアニジン、10mMのジチオトレイトール(DTT)、10mMのエチレンジアミン四酢酸(EDTA)、20mMのTris(pH8.1)に溶解した。
実施例3による方法を使用して高親和力CDR領域のαβヘテロ二量体TCRとSLLMWITQC-HLA-A2複合体の親和力を測定した。
本発明の高親和性単鎖TCR分子と抗CD3抗体の一本鎖分子(scFv)を融合して融合分子を構築した。オーバーラップ(overlap)PCRの方法を通じて、プライマーを設計して、抗-CD3抗体と高親和性単鎖TCR分子の遺伝子を連結し、中間の連結オリゴペプチド(linker)は、GGGGSに設計され、融合分子の遺伝子断片が制限酵素サイトNcoIとNotIを持つようにした。PCR増幅産物をNcoIとNotI二重酵素で切断し、NcoIとNotI二重酵素で切断されたpET28aベクターによって連結される。連結産物をE.coli DH5αコムピント(competent)細胞に形質転換させ、カナマイシンを含有したLBプレートにコーティングし、37℃の温度の下で一晩反転培養し、陽性クローンをとり、PCRスクリーニングし、陽性組換え体に対してシーケンスし、配列の正確かどうかを確認した後、組換えプラスミドを発現するために抽出して、E.coli BL21(DE3)受容性細胞に形質転換させた。
目的遺伝子を含有した発現プラスミドを大腸菌菌株BL21(DE3)に形質転換させ、LBプレート(カナマイシン50μg/ml)をにコーティングして、37℃の温度の下で、一晩培養した。次の日、クローンを選択して、10mlのLB液体培地(カナマイシン50μg/ml)を2~3時間培養し、1:100の体積比に基づいて、1LのLB培地(カナマイシン50μg/ml)に接種し、継続して0.5~0.8のOD600まで培養した後、0.5mMの最終濃度のIPTGで目的タンパク質の発現を誘導した。4時間誘導した後、6000rpMで10分間遠心分離して細胞を得た。PBS緩衝液で菌体を1回洗浄し、菌体を分けて入れて、200mlの細菌培養物に該当する菌体をとり、5mlのBugBuster Master Mix(Novagen)に細菌を解離し、6000gで15分間遠心分離して封入体を収集した。その後、洗剤で4回洗浄して細胞を破壊物と膜成分を除去した。その後、PBSのような緩衝液で封入体を洗浄して、洗剤と塩を除去した。最終的には、封入体を8Mの尿素を含有したTris緩衝液で溶解させ、封入体の濃度を測定し、これを分けて入れ、-80℃の温度下で冷凍保管した。
-80℃の超低温冷蔵庫で約10mgの封入体を取り出した後、解凍し、ジチオトレイトール(DTT)を入れて最終濃度を10mMにし、37℃の温度で30分~1時間の間インキュベーションし(Incubate)ジスルフィド結合が完全に解除されることを確保した。その後、封入体サンプル溶液をそれぞれ200mlの4℃の予冷リフォールディング緩衝液(100mM Tris pH8.1、400mM L-アルギニン、2mM EDTA、5M 尿素、6.5mMβ-mercapthoethylamine、1.87mM Cystamine)に滴下して、約4℃の温度下で30分間ゆっくりと攪拌した。再生溶液を8倍体積の予冷されたH2Oで16~20時間の間透析した。再び8倍体積の10mMのTris pH8.0で2回透析し、4℃の温度で継続して約8時間の間透析し、透析後のサンプルをろ過した後、下記のように精製した。
透析されたリポーリング物質(10mM Tris pH8.0に)をPOROS HQ/20陰イオン交換クロマトグラフィー前のロードカラム(Applied Biosystems)を使用して、AKTA精製器(GE Healthcare)で0~600mMのNaClで勾配溶出した。クマシブリリアントブルー(Coomassie Brilliant Blue)で染色されたSDS-PAGEで各成分を分析した後、合併した。
第1のステップの精製合併されたサンプル溶液を濃縮して、このステップの精製に提供し、PBS緩衝液で事前平衡させたSuperdex7510/300GLゲル濾過クロマトグラフィー前のロードカラム(GE Healthcare)を使用して融合タンパク質を精製して、クマシーブリリアントブルーで染色されたSDS-PAGEでのピークの成分を分析した後、合併した。
抗-CD3の一本鎖抗体(scFv)とαβヘテロ二量体TCRを融合して、融合分子を製造した。抗-CD3のscFvとTCRのβ鎖を融合し、前記TCRβ鎖は、任意の前記高親和性αβヘテロ二量体TCRのβ鎖可変ドメインを含むことができ、融合分子のTCRα鎖は、任意の前記高親和性αβヘテロ二量体TCRのα鎖可変ドメインを含むことができる。
1.α鎖発現ベクターの構築
αβヘテロ二量体TCRのα鎖を持った目的遺伝子をNcoIとNotI二重酵素で切断し、NcoIとNotI二重酵素で切断されたpET28aベクターに連結した。連結産物をE.coli DH5αに形質転換させ、カナマイシンを含有したLBプレートにコーティングし、37℃の温度下で一晩反転培養し、陽性クローンをとり、PCRスクリーニングし、陽性組換え体に対してシーケンスで、配列の正確かどうかを確認した後、組換えプラスミドを発現するために抽出して、E.coli BL21(DE3)に形質転換させた。
オーバーラップ(overlap)PCRの方法で、プライマーを設計して抗-CD3 scFvと高親和性ヘテロ二量体TCRβ鎖遺伝子を連結し、中間の連結オリゴペプチド(linker)はGGGGSであり、抗-CD3のscFvと高親和性ヘテロ二量体TCRβ鎖の融合タンパク質の遺伝子断片が制限酵素サイトNcoI(CCATGG)とNotI(GCGGCCGC)を持つようにした。PCR増幅産物をNcoIとNotI二重酵素で切断し、NcoIとNotI二重酵素で切断されたpET28aベクターに連結した。連結産物をE.coliDH5α受容性細胞に形質転換させ、カナマイシンを含有したLBプレートにコーティングし、37℃の温度の下で一晩反転培養し、陽性クローンをとり、PCRスクリーニングし、陽性組換え体に対してシーケンスし、配列の正確かどうかを確認した後、組換えプラスミドを発現するために抽出して、E.coli BL21(DE3)受容性細胞に形質転換させた。
発現プラスミドをそれぞれE.coli Tuner(DE3)受容性細胞に形質転換させ、LBプレート(カナマイシン50μg/ mL)にコーティングして、37℃の温度下で一晩培養した。次の日、クローンを選択して、10mlのLB液体培地(カナマイシン50μg/ml)を2~3時間培養し、1:100の体積比に基づいて、1LのLB培地(カナマイシン50μg/ml)に接種し、継続して0.5~0.8のOD600まで培養した後、最終濃度が1mMのIPTGを入れて目的タンパク質の発現を誘導した。4時間誘導した後、6000 rpMで10分間遠心分離して細胞を得た。PBS緩衝液で菌体を1回洗浄し、菌体を分けて入れて、200mlの細菌培養物に該当する菌体をとり、5mlのBugBuster Master Mix(Novagen)に細菌を解離し、6000gで15分間遠心分離して封入体を収集した。その後、洗剤で4回洗浄して細胞を破壊物と膜成分を除去した。その後、PBSのような緩衝液で封入体を洗浄して洗剤と塩を除去した。最終的には、封入体を6Mの塩酸グアニジン、10mMのジチオトレイトール(DTT)、10mMのエチレンジアミン四酢酸(EDTA)、20mMのTris、pH8.1を含有した緩衝液で溶解し、封入体の濃度を測定し、これを分けて入れ、-80℃の温度下で冷凍保管した。
本実施例では、本発明の高親和力TCRが形質感染された効果細胞が標的細胞に対して優れた特異的活性作用があることを検証した。
本実施例は、非放射性細胞毒性実験を通じて、LDHの放出を測定することにより、本発明のTCRを形質導入させた細胞の殺傷機能を検証した。当該試験は、51Cr放出細胞毒性試験の比色代替試験で、細胞分解後放出された乳酸脱水素酵素(LDH)を定量分析した。30分間のカップリングされた酵素反応で培地に放出されたLDHを検出し、酵素反応でLDHはテトラゾリウム塩(INT)を赤色のホルマザン(formazan)に転換させることができる。生成された赤色の産物の量と分解された細胞の数は正比例する。標準96ウェルプレートリーダーで490nmの可視光線データを収集することができる。
本実施例は、本発明の高親和力TCRと抗-CD3抗体の融合タンパク質が効果細胞に対して方向転換することができ、優れた活性作用があることを検証した。
本発明の高親和力TCRで形質感染T細胞をヒト黒色腫異種移植片モデルのマウスに注射して、体内で腫瘍の抑制効果を測定した。
Claims (53)
- T細胞受容体(TCR)であって、
SLLMWITQC-HLA A2複合体に結合する活性を有し、前記T細胞受容体は、TCRα鎖可変ドメイン及びTCRβ鎖可変ドメインを含み、前記TCRα鎖可変ドメインは、3つのCDR領域を含み、前記TCRα鎖可変ドメインの3つのCDR領域の基準配列は、以下の通りであり、
CDR1α:TSINN
CDR2α:IRSNERE
CDR3α:ATDANGKII、また少なくとも1つの下記の突然変異を含み、
および/または、前記TCRβ鎖可変ドメインは、3つのCDR領域を含み、前記TCRβ鎖可変ドメインの3つのCDR領域の基準配列は、以下の通りであり、
CDR1β:SGHDY
CDR2β:FNNNVP
CDR3β:ASSLGSNEQY、また少なくとも1つの下記の突然変異を含む、前記T細胞受容体(TCR)。

好ましくは、突然変異後の前記TCRα鎖可変ドメインは、27G、28W、29Aまたは29V、30Q、51T、52G、53Q、54Qまたは54A、90M、91Y、93Eまたは93Q、94Fまたは94Yまたは94W、95Aまたは95Q、96Wまたは96Rまたは96Tまたは96Sまたは96Mまたは96L、97Wまたは97Vまたは97Yまたは97P、及び98Eまたは98Nまたは98Dまたは98Qまたは98Kまたは98Vまたは98Gまたは98Mまたは98Sから選択される1つまたは複数のアミノ酸残基を含み、アミノ酸残基の番号は、配列番号:1に示される番号を使用し、および/または突然変異後の前記TCRβ鎖可変ドメインは、51H、52G、53A、54V、95Qまたは95M、96Rまたは96Kまたは96M、及び98Aまたは98Gまたは98Pから選択される1つまたは複数のアミノ酸残基を含み、アミノ酸残基の番号は、配列番号:2に示される番号を使用することを特徴とする
