本発明の一態様において、コンセンサス抗原は、以下を1つ以上有することを含む、転写及び翻訳の改善を提供することが望ましい:転写を増加させる低GC含量リーダー配列;mRNA安定性及びコドン最適化;可能なシス作用配列モチーフ(すなわち、内部TATAボックス)の除去。
本明細書で使用する用語は、特定の実施形態のみを説明するためのものに過ぎず、限定することを意図するものではない。本明細書及び添付の特許請求の範囲で使用するように、単数形「a」、「an」及び「the」は、文脈上他に明確に指示されない限り、複数の指示対象を含む。
本明細書中で使用する「アジュバント」とは、本明細書に記載のDNAプラスミドワクチンに添加し、DNAプラスミドによってコードされる1つ以上のコンセンサスフィロウイルス免疫原の抗原性を増強し、また、以下に記載する核酸配列をコードする任意の分子を意味し得る。
「抗体」とは、クラスIgG、IgM、IgA、IgD、または、IgEの抗体、または、Fab、F(ab’)2、Fdを含む、その断片もしくは誘導体、ならびに、その一本鎖抗体、二特異性抗体、二重特異性抗体、二価抗体及び、誘導体を意味し得る。当該抗体は、哺乳類の血清試料から単離された抗体、ポリクローナル抗体、親和性精製抗体、または、これらの混合物のうち、所望のエピトープ、または、それに由来する配列に対する十分な結合特異性を示すものであり得る。
本明細書中で使用する「コード配列」または「コード核酸」とは、タンパク質をコードするヌクレオチドを含む核酸(RNAまたはDNA分子)を意味し得る。コード配列はまた、RNA配列をコードするDNA配列を含み得る。当該コード配列は、核酸の投与先である個体または哺乳類の細胞での発現を誘導できるプロモーター及びポリアデニル化シグナルを含む調節エレメントと機能的に連結した、開始及び終止シグナルをさらに含むことができる。いくつかの実施形態では、当該コード配列は、必要に応じて、N末端メチオニンまたはIgEもしくはIgGシグナルペプチドなどのシグナルペプチドをコードする開始コドンをさらに含み得る。
本明細書中で使用する「遺伝子構築物」とは、免疫原などのタンパク質をコードするヌクレオチド配列を含むDNA分子またはRNA分子のことを指す。当該遺伝子構築物は、RNA分子から転写されたDNA分子のことも指す場合がある。当該コード配列は、核酸分子の投与先である個体の細胞で発現を誘導できるプロモーター及びポリアデニル化シグナルを含む調節エレメントと作動可能に連結した、開始及び終止シグナルを含む。本明細書中で使用する場合、用語「発現可能な形態」とは、個体の細胞に存在している場合にコード配列が発現するように、タンパク質をコードするコード配列に作動可能に連結した必要な調節エレメントを含んでいる遺伝子構築物のことを指す。
本明細書中で使用する「相補体」または「相補的」とは、ある核酸が、ヌクレオチド間または核酸分子のヌクレオチド類似体間に、ワトソン・クリック(例えば、A-T/U及びC-G)、または、フーグスティーン塩基対を表わすことを意味し得る。
本明細書中で使用する「コンセンサス」または「コンセンサス配列」とは、特定のフィロウイルス抗原の複数のサブタイプのアラインメントの分析に基づいて構築した合成核酸配列、または対応するポリペプチド配列を意味し得、これを用いて、特定のフィロウイルス抗原のサブタイプまたは血清型に対する広範な免疫を誘導することができる。
本明細書中で使用する「定電流」とは、組織、または、当該組織を規定する細胞が、同組織に送達される電気パルスの持続期間中に受容する、または、経験する電流を定義する。当該電気パルスは、本明細書に記載の電気穿孔装置から送達される。この電流は、電気パルスの寿命期間中、当該組織に定電流量で留まるが、それは、本明細書で提供される電気穿孔装置が、好ましくは、瞬間的フィードバックを有しているフィードバック素子を具備しているからである。当該フィードバック素子は、パルスの持続期間中の組織(または、細胞)の抵抗を測定し、及び、電気穿孔装置に自身の電気エネルギー出力を変化させる(例えば、電圧を上げる)ようにすることができるため、同組織での電流は、電気パルスの間ずっと(マイクロ秒のオーダーで)、及び、パルス間において、一定であり続ける。いくつかの実施形態では、フィードバック素子は、制御器を含む。
本明細書中で使用する「電流フィードバック」または「フィードバック」は、互換的に使用されており、及び、提供される電気穿孔装置の活発な応答を意味し得るが、これは、電極間の組織の電流を測定することと、電流を一定レベルで維持するために、EP装置が送達するエネルギー出力を適宜変化させることとを含む。この一定レベルは、パルスシーケンスまたは電気処理を開始する前に、使用者が予め設定する。電気穿孔装置内の電気回路が、電極間の組織の電流を連続的にモニタリングし、そのモニタリングした電流(または、組織内の電流)を予め設定された電流と比較し、連続的にエネルギー出力の調整を行なって、モニタリングした電流を予め設定されたレベルで維持することができるように、フィードバックは、電気穿孔装置の電気穿孔コンポーネント、例えば、制御器によって達成され得る。当該フィードバックループは、瞬時のものであり得るものであり、それは、フィードバックループが、アナログ閉ループフィードバックであるからである。
本明細書中で使用する「分散電流」とは、本明細書に記載の電気穿孔装置の様々な針電極アレイから送達される電流のパターンを意味し得るものであり、これらのパターンは、電気穿孔される組織のあらゆる領域に対する電気穿孔関連の熱ストレスの発生を最小限に抑えるか、または、好ましくは、消失させる。
本明細書中で互換的に使用する「電気穿孔」、「電気透過処理」、または「界面動電増強」(「EP」)は、生体膜に微細経路(細孔)を生じさせるための膜貫通電場パルスの使用を意味し得る。こうした微細経路の存在により、プラスミド、オリゴヌクレオチド、siRNA、薬剤、イオン、及び、水などの生体分子が、細胞膜の片側から他方の側に通過することが可能になる。
本明細書中で使用する「フィードバック機構」とは、ソフトウェアまたはハードウェア(または、ファームウェア)のいずれかによって実行されるプロセスであって、(エネルギーパルスの送達前、送達中、及び/または、送達後の)所望の組織のインピーダンスを受容し、それを現在値、好ましくは、電流と比較して、送達されるエネルギーパルスを調整して、予め設定された値を達成するプロセスのことを指し得る。フィードバック機構は、アナログ閉ループ回路により実施され得る。
「断片」は、特定のフィロウイルス抗原を認識することによってフィロウイルスに対する免疫応答を哺乳類において誘起することができるフィロウイルス免疫原のポリペプチド断片を意味し得る。フィロウイルスエンベロープ糖タンパク質免疫原は、場合により、1位にシグナルペプチド及び/またはメチオニン、いずれの場合にも1位にシグナルペプチド及び/またはメチオニンを有するかまたは有さない本明細書に記載のコンセンサス配列に98%以上の相同性を有するタンパク質、本明細書に記載のコンセンサス配列に99%以上の相同性を有するタンパク質、及び本明細書に記載のコンセンサス配列に100%の相同性を有するタンパク質を含む場合がある。断片は、例えば、シグナルペプチド、例えば、免疫グロブリンシグナルペプチド、例えばIgEシグナルペプチドまたはIgGシグナルペプチドに連結したフィロウイルス免疫原の断片を含んでも含まなくてもよい。
本明細書中で、2つ以上の核酸またはポリペプチド配列の関係で使用する「同一の」または「同一性」とは、当該配列が、指定領域に渡って指定の割合の同一残基を有することを意味し得る。この割合を計算するには、2つの配列を好適に整列させ、指定領域に渡って2つの配列を比較し、双方の配列間で同一残基が発生する位置の数を確定して一致位置の数を得て、一致した位置の個数を指定領域の合計位置数で割り、計算結果に100を掛けることで、配列同一性のパーセント値を算出し得る。2つの配列の長さが異なるか、あるいは、アライメントにより1つ以上の付着末端が生じて、指定の比較領域に単一配列のみが含まれる場合には、単一配列の残基を、計算の分母には含めるが、分子には含めない。DNAとRNAを比較する場合には、チミン(T)とウラシル(U)を同等とみなし得る。同一性は、筆算で求めてもよく、あるいは、BLASTやBLAST 2.0等のコンピュータ配列アルゴリズムを使用して計算することもできる。
本明細書中で使用する「インピーダンス」は、フィードバック機構を論ずる際に利用し得るものであり、かつ、オームの法則に従って電流値に変換することができるため、予め設定された電流と比較が可能である。
本明細書中で使用する「免疫応答」とは、1つ以上の提供するDNAプラスミドワクチンを介したフィロウイルスコンセンサス抗原の導入に応答して、宿主の免疫系、例えば、哺乳類の免疫系が活性化することを意味し得る。この免疫応答は、細胞性応答または体液性応答、または、その両方であり得る。
本明細書中で使用する「核酸」または「オリゴヌクレオチド」または「ポリヌクレオチド」は、互いに共有結合している少なくとも2つのヌクレオチドを意味し得る。一本鎖を表現することで、相補鎖の配列も定義される。したがって、核酸は、表現される一本鎖の相補鎖も包含する。ある核酸の多くの変異体を、所定の核酸として同一目的で使用し得る。したがって、核酸は、実質的に同一の核酸、及び、その相補体も含む。一本鎖は、ストリンジェントなハイブリダイゼーション条件下で標的配列とハイブリダイズできるプローブを提供する。したがって、核酸は、ストリンジェントなハイブリダイゼーション条件下でハイブリダイズするプローブも含む。
本明細書中で使用する「作動可能に連結した」は、遺伝子の発現が、遺伝子と空間的に接続しているプロモーターの制御下にあることを意味し得る。プロモーターは、その制御下にある遺伝子の5’(上流)または3’(下流)に位置し得る。当該プロモーターと遺伝子との間の距離は、プロモーターが由来する遺伝子において当該プロモーターを制御する遺伝子とプロモーターとの間の距離とほぼ同一とし得る。当該技術分野で周知の通り、プロモーター機能を喪失させずに、この距離の変化に適応し得る。
本明細書中で使用する「プロモーター」とは、核酸の細胞での発現を、実現し、活性化し、または、増強することのできる合成分子または天然由来分子のことを意味し得る。プロモーターは、発現をさらに増強し、及び/または、空間的発現、及び/または、その一時的発現を改変する目的で、1つ以上の特定の転写調節配列を含み得る。プロモーターは、遠位のエンハンサーエレメントまたは抑制エレメントを含むこともでき、このものは、転写開始部位から数千塩基対も離れた場所に位置することができる。プロモーターは、ウイルス、細菌、真菌、植物、昆虫、及び、動物を含む起源に由来し得る。プロモーターは、遺伝子成分の発現を調節し得るものであり、発現が起こる細胞、組織、もしくは、臓器、あるいは、発現が起こる発生段階については、恒常的もしくは差次的に、または、例えば、生理的ストレス、病原体、金属イオン、あるいは、誘発剤などの外部刺激に応答して同発現を調節し得る。プロモーターの代表例として、バクテリオファージT7プロモーター、バクテリオファージT3プロモーター、SP6プロモーター、lacオペレーター-プロモーター、tacプロモーター、SV40後期プロモーター、SV40初期プロモーター、RSV-LTRプロモーター、CMV IEプロモーター、SV40初期プロモーターまたはSV40後期プロモーター、及び、CMV IEプロモーターなどがある。
本明細書中で使用する「ストリンジェントなハイブリダイゼーション条件」とは、例えば、核酸の複合混合物において、第1の核酸配列(例えば、プローブ)が、第2の核酸配列(例えば、標的)とハイブリダイズする際の条件のことを意味し得る。ストリンジェントな条件は配列に依存するため、状況に応じて変化する。ストリンジェントな条件は、所定のイオン強度pHにおける特定の配列に対する融点(Tm)より約5~10℃低くなるように選択される。このTmとは、(所定のイオン強度、pH、及び、核酸濃度下で)標的に対して相補的なプローブの50%が、平衡状態で標的配列にハイブリダイズする温度のことである(この標的配列は、過剰に存在するので、Tmにおいて、プローブの50%が平衡状態で占有される)。ストリンジェントな条件は、塩濃度が、pH7.0~8.3において、約1.0M未満のナトリウムイオン、例えば、約0.01~1.0Mのナトリウムイオン濃度(または、その他の塩)であり、及び、温度が、短いプローブ(例えば、約10~50ヌクレオチド)では、低くとも約30℃、長いプローブ(例えば、約50ヌクレオチドを超えるもの)では、低くとも約60℃とし得る。ストリンジェントな条件は、ホルムアミドなどの不安定化剤を添加しても実現し得る。選択的または特異的ハイブリダイゼーションの場合、陽性シグナルは、バックグラウンドハイブリダイゼーションの少なくとも2~10倍であり得る。例示的なストリンジェントなハイブリダイゼーション条件は、以下の条件を含む。50%ホルムアミド、5×SSC、及び、1%SDSを用いて、42℃でのインキュベート、あるいは、5×SSC、1%SDSを用いて、65℃でのインキュベートと、0.2×SSC、及び、0.1%SDSを用いて、65℃での洗浄を含む。
本明細書中で使用する「実質的に相補的」とは、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、100個またはそれ以上の個数のヌクレオチドまたはアミノ酸の領域に渡って、第1の配列が、第2の配列の相補体と、少なくとも60%、65%、70%、75%、80%、85%、90%、95%、97%、98%または99%同一であること、あるいは、2つの配列が、ストリンジェントなハイブリダイゼーション条件下でハイブリダイズすることを意味し得る。
本明細書中で使用する「実質的に同一」とは、8個、9個、10個、11個、12個、13個、14個、15個、16個、17個、18個、19個、20個、21個、22個、23個、24個、25個、30個、35個、40個、45個、50個、55個、60個、65個、70個、75個、80個、85個、90個、95個、100個またはそれ以上の個数のヌクレオチドまたはアミノ酸の領域に渡って、第1の配列と第2の配列が、少なくとも60%、65%、70%、75%、80%、85%、90%、95%、97%、98%または99%同一であること、あるいは、核酸に関して、当該第1の配列が、当該第2の配列の相補体と実質的に相補的であることを意味し得る。
核酸に関して本明細書で使用する「変異体」は、(i)基準ヌクレオチド配列の一部または断片、(ii)基準ヌクレオチド配列またはその一部の相補体、(iii)基準核酸またはその相補体と実質的に同一の核酸、または、(iv)ストリンジェントな条件下で、基準核酸、その相補体、または、それらと実質的に同一の配列とハイブリダイズする核酸のことを意味し得る。
本明細書中で使用する「ベクター」とは、複製起点を含む核酸配列を意味し得る。ベクターは、プラスミド、バクテリオファージ、細菌、ウイルスベクター、人工染色体または酵母人工染色体であってもよい。ベクターは、DNAまたはRNAベクターであり得る。ベクターは、自己複製染色体外ベクターであってもよいし、宿主ゲノムに組み込まれるベクターであってもよい。

本明細書に記載のタンパク質をコードするコード配列は、日常的な方法を用いて生成し得る。コンセンサスザイールエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、第2コンセンサスザイールエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、コンセンサスザイールエボラウイルスギニアエンベロープ糖タンパク質免疫原エンベロープ糖タンパク質免疫原をコードする核酸配列、コンセンサススーダンエボラウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、マールブルグマールブルグウイルスアンゴラ2005エンベロープ糖タンパク質免疫原をコードする核酸配列、第1コンセンサスマールブルグマールブルグウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、第2コンセンサスマールブルグマールブルグウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列、及び第3コンセンサスマールブルグマールブルグウイルスエンベロープ糖タンパク質免疫原をコードする核酸配列を含む組成物は、開示するアミノ酸配列に基づいて生成してもよい。

プラスミドは、フィロタンパク質に対する免疫応答を誘発することができる合成コンセンサス抗原をコードするコード配列を含む、上記に開示される1つ以上の様々な免疫原をコードする核酸配列を含み得る。
一実施形態では、核酸はRNA分子である。したがって、一実施形態では、本発明は、マールブルグマールブルグウイルス(MARV)、スーダンエボラウイルス(SUDV)またはザイールエボラウイルス(ZEBOV)のエンベロープ糖タンパク質(GP)遺伝子の1つ以上をコードするRNA分子を提供する。当該RNAは、プラス鎖であってもよい。したがって、いくつかの実施形態では、当該RNA分子は、逆転写などの介在性複製ステップを必要とせずに、細胞によって翻訳することができる。本発明に有用なRNA分子は、5’キャップ(例えば、7-メチルグアノシン)を有し得る。このキャップは、RNAのin vivoでの翻訳を促すことができる。本発明に有用なRNA分子の5’ヌクレオチドは、5’三リン酸基を有し得る。キャップされたRNAにおいて、これは、5’から5’への架橋を介して7-メチルグアノシンに連結され得る。RNA分子は、3’ポリAテールを有し得る。また、ポリAポリメラーゼ認識配列(例えば、AAUAAA)を、その3’末端の近傍に有し得る。本発明に有用なRNA分子は、一本鎖であってもよい。
本明細書において、電気穿孔法によって対象に対して効率的に送達され、及び、1つ以上の所望の抗原を発現することができる、線状核酸ワクチンまたは線状発現カセット(「LEC」)も提供する。このLECは、あらゆるリン酸主鎖を欠いた線状DNAであってもよい。DNAは、1つ以上の抗原をコードし得る。LECは、プロモーター、イントロン、終止コドン、ポリアデニル化シグナルを含み得る。抗原の発現は、プロモーターによって制御され得る。このLECは、抗生物質耐性遺伝子、及び/または、リン酸主鎖を含まなくともよい。このLECは、所望の抗原遺伝子発現と無関係の他の核酸配列を含有していなくともよい。
一実施形態では、本明細書において、本発明の核酸を細胞に送達することができるウイルスベクターを提供する。当該発現ベクターは、ウイルスベクターの形態で細胞に提供され得る。ウイルスベクター技術は、当該技術分野で周知であり、例えば、Sambrook et al.(2001)、及び、Ausubel et al.(1997)、及び、その他のウイルス学、及び、分子生物学のマニュアルに記載がされている。ベクターとして有用なウイルスとして、レトロウイルス、アデノウイルス、アデノ随伴ウイルス、ヘルペスウイルス、及び、レンチウイルスなどがあるが、これらに限定されない。一般的に、適切なベクターは、少なくとも1つの生物において機能する複製起点、プロモーター配列、好都合な制限エンドヌクレアーゼ部位、及び、1つ以上の選択マーカーを含む。(例えば、WO01/96584、WO01/29058、及び、米国特許第6,326,193号を参照されたい。)ウイルスベクター、特に、レトロウイルスベクターは、遺伝子を哺乳類、例えば、ヒト細胞に挿入するための最も広く使用されている方法となっている。他のウイルスベクターは、レンチウイルス、ポックスウイルス、単純ヘルペスウイルスI、アデノウイルス、及び、アデノ随伴ウイルスなどから誘導することができる。例えば、米国特許第5,350,674号、及び、第5,585,362号を参照されたい。
核酸分子を含む組成物を提供する。組成物は、単一のプラスミドのような単一の核酸分子の複数のコピー、2つ以上の異なるプラスミドのような2つ以上の異なる核酸分子の複数のコピーを含み得る。例えば、組成物は、複数の2、3、4、5、6、7、8、9または10以上の異なる核酸分子を含み得る。そのような組成物は、複数の2、3、4、5、6、またはそれ以上の異なるプラスミドを含み得る。
本明細書において、フィロウイルス、特にマールブルグウイルス、エボラウイルス・スーダン及び/またはエボラウイルス・ザイールに対する免疫応答を哺乳類において生成可能なワクチンを提供する。ワクチンは、上記のように各プラスミドを含み得る。ワクチンは、複数のプラスミド、またはそれらの組み合わせを含み得る。ワクチンは、治療的または予防的免疫応答を誘導するために提供してもよい。
本明細書において、免疫応答を誘発することができる、フィロウイルス、特にマールブルグウイルス、エボラウイルス・スーダン及び/またはエボラウイルス・ザイールの免疫原に対して、それらを特に効果的なものとするエピトープを含むコンセンサス抗原の遺伝子構築物及びタンパク質を提供するためのワクチンの送達方法を提供する。ワクチンまたはワクチン接種の送達方法は、治療的免疫応答及び予防的免疫応答を誘導するために提供してもよい。ワクチン接種プロセスは、哺乳類において、フィロウイルス、特にマールブルグウイルス、エボラウイルス・スーダン及び/またはエボラウイルス・ザイールに対する免疫応答を生じ得る。ワクチンを個体に送達し、哺乳類の免疫系の活性を調節し、免疫応答を増強してもよい。ワクチンの送達は、細胞内で発現させ、細胞の表面に送達し、そこにおいて免疫系が認識し、細胞性、体液性、または細胞性及び体液性の応答を誘導する核酸分子としてのコンセンサス抗原の形質移入であってもよい。ワクチンの送達は、哺乳類に上記のワクチンを投与することによって、フィロウイルス、特にマールブルグウイルス、エボラウイルス・スーダン及び/またはエボラウイルス・ザイールに対する免疫応答を哺乳類において誘導または誘発するために使用してもよい。

ワクチンのプラスミドの電気穿孔法による当該ワクチンの投与は、細胞膜に可逆的な孔を形成させるのに効果的なエネルギーパルスを哺乳類の所望の組織に送達するように構成することができる電気穿孔装置を用いて達成され得るものであり、及び、好ましくは、当該エネルギーパルスは、使用者が入力したプリセット電流に近い定電流である。当該電気穿孔装置は、電気穿孔コンポーネント、及び電極組立体またはハンドル組立体を含み得る。電気穿孔コンポーネントは、制御器、電流波形発生器、インピーダンステスタ、波形ロガー、入力要素、ステータス報告要素、コミュニケーションポート、メモリーコンポーネント、電源、及び、電源スイッチなどの当該電気穿孔装置の1つ以上の様々な要素を含み、及び、組み込み得る。当該電気穿孔法は、in vivo電気穿孔装置、例えば、CELLECTRA EPシステム(VGX Pharmaceuticals,Blue Bell,PA)、または、Elgenエレクトロポレーター(Genetronics,San Diego,CA)を使用して、プラスミドによる細胞の形質移入を容易にし得る。
本明細書で論じるDNAワクチンを含むDNAプラスミドの調製方法を本明細書において提供する。哺乳類発現プラスミドへの最終サブクローニング工程の後のDNAプラスミドを用いて、当該分野で公知の方法を用いて、大規模発酵タンクに細胞培養物を播種することができる。
方法
プラスミドワクチン構築
pMARV、pEBOS、及びpEBOZプラスミドDNA構築物は、全長GPタンパク質をコードする。pEBOS及びpEBOZに対して、アミノ酸コンセンサス戦略を使用し、一方、pMARVには、2005年のアンゴラ流行株由来の型適合配列(GenBank#VGP_MABVR)を使用した(Towner JS,et al.(2006). Marburgvirus genomics and association with a large hemorrhagic fever outbreak in Angola. J Virol 80:6497-6516)。コンセンサス配列は、優勢なZEBOV及びSUDV GPアミノ酸配列のアラインメント及びそれぞれについてコンセンサスを生成することによって決定した。各ワクチンGP遺伝子を、ヒトにおける発現のために遺伝子最適化し(タンパク質発現を強化するためのコドン及びRNAの最適化を含む(GenScript,Piscataway,NJ))、商業的に合成し(GenScript,Piscataway,NJ)その後、サイトメガロウイルス前初期(CMV)プロモーターの制御下で、改変pVAX1哺乳類発現ベクター(Invitrogen,Carlsbad,CA)にサブクローニングした(GenScript,Piscataway,NJ);修飾は、2A>C、3C>T、4T>G、241C>G、1,942C>T、2,876A>-、3,277C>T、及び3,753G>Cを含む。系統発生解析は、MEGAバージョン5ソフトウェアを使用してClustalWを用いた多重アラインメントによって行った。あるいは、MARV間のGP多様性は、比較してはるかに高い(約70%の同一性)ため、コンセンサス戦略は採択しなかった。MARVの適用範囲については、現在までに最大級の最も致命的なMARV流行の原因となったことから、2005年のアンゴラ(GenBank #VGP_MABVR)でのアウトブレイクに由来するMGPの配列を利用することを選択した。この配列は、Musoke、Popp及びLeiden(10.6%の多様性)、またはウガンダ(01Uga07)、ダーバ(05DRC99及び07DRC99)及びオゾリン(10.3%の多様性)を含む関連株の最も近いクラスターのいずれかから10%より大きく異なっていた。3プラスミド戦略は、新規3価多価-フィロウイルスワクチン戦略の基盤を形成した。
ヒト胎児腎(HEK)293T細胞を培養し、形質移入し、回収した。簡潔に述べると、細胞を、10%FBS、1%Pen-strep、ピルビン酸ナトリウム、及びL-グルタミンを含むDMEM中で増殖させた。細胞を150mm Corningディッシュで培養し、5%CO2を含む37℃インキュベーター中で一晩70%コンフルエンスまで増殖させた。Calphos(商標)哺乳類形質移入キットプロトコール(Clonetech)またはLipofectamine(商標)2000試薬(Invitrogen)のいずれかを使用して製造業者のプロトコールに従って10~25μgのフィロウイルス科pDNAでディッシュを形質移入し、次いで24~48時間インキュベートした。細胞を氷冷PBSで回収し、遠心分離して洗浄し、次いでウェスタンイムノブロットまたはFACS分析のためにペレット化した。標準的なウエスタンブロッティングを使用し、GP1検出のためのGP特異的MAbを生成した。ウエスタンイムノブロッティング実験のデータを図1Bに示す。FACS分析からのデータを図1Cに示す。
成体雌C57BL/6(H-2b)、BALB/cJ(H-2d)、及びBl0.Br(H-2k)マウスをジャクソン研究室から購入し(Bar Harbor,ME)、一方、Hartleyモルモットはチャールズ川(Wilmington,MA)から入手した。すべての動物実験は、UPenn IACUC及びSchool of Medicine Animal Facility、またはPHACのNML Institutional Animal Care Committee及び実験動物の住居及びケアに関するカナダ動物衛生委員会のガイドラインに従って実施し、上記の機関による適切なレビュー及び承認の後にGuide for the Care and Use of Laboratory Animals of NIHの推奨に従って行った。UPenn及びNMLは、動物福祉、動物福祉法、及びその他すべての該当する連邦、州及び地方の法律に関するNIH方針を遵守する。
スクロース精製MARVオゾリンGPまたはZGPのいずれかでコーティングした96ウェルELISAプレートを用いて、または陰性対照スクロース精製ニパGタンパク質を1:2,000の濃度で用いて、抗体(Ab)力価を測定した。簡潔に述べると、プレートを4℃で18時間インキュベートし、PBS及び0.1%Tween-20で洗浄し、100μl/試料の血清を3回(5%スキムミルク及び0.5%Tween-20を含むPBS中、1:100、1:400、1:1,600、及び1:6,400の希釈率で)試験した。湿った容器中、37℃で1時間インキュベートした後、プレートを洗浄し、次いでヤギ抗マウスIgG結合HRP抗体(Cedarlane)100μlを添加し(1:2,000希釈)、さらに湿った容器中、37℃で1時間インキュベートした。洗浄後、ABST(2,2’-アジノ-ビス(3-エチルベンザゾリン-6-スルホン酸)及びペルオキシダーゼ基質(Cedarlane)100μlを添加し、Ab結合を可視化した。プレートを37℃で30分間インキュベートし、その後405nmで読み取った。陰性対照血清から陽性対照を差し引くと>3SDであることをもって、陽性結合結果を特徴付けた。
最終免疫の8~11日後にマウスを殺処分し、脾臓を採取した。簡潔に述べると、10%FBS、1×抗-抗(Invitrogen)、及び1×β-ME(Invitrogen)を補充したRPMI1640培地(Mediatech Inc.,Manassas,VA)に脾臓を入れた。Stomacher machine(Seward Laboratory Systems Inc.,Bohemia,NY)を用いて脾臓の機械的破壊により脾細胞を単離し、得られた産物を、40μm細胞ストレーナー(BD Falcon)を用いてろ過した。次いで、細胞を、RBCの溶解のためにACK溶解緩衝液(Lonza、スイス)で5分間処理し、PBSで洗浄し、次いでELISPOTまたはFACSアッセイで使用するためにRPMI培地に再懸濁した。
標準的なIFNγELISPOTアッセイを行った。簡潔に述べると、96ウェルプレート(Millipore,Billerica,MA)を抗マウスIFN-γ捕捉抗体でコーティングし、4℃で24時間インキュベートした(R&D Systems,Minneapolis,MN)。翌日、プレートをPBSで洗浄し、次いでブロッキング緩衝液(PBS中、1%BSA及び5%スクロース)で2時間ブロッキングした。脾細胞(1~2×105細胞/ウェル)を三連でプレーティングし、37℃、5%CO2で、及びRPMI1640(陰性対照)、ConA(陽性対照)、またはGPペプチドのいずれかの存在下で、個々に(それぞれのGPの長さにまたがって、9アミノ酸をオーバーラップさせた15量体)または全体をプールして一晩刺激した(終濃度2.5μg/ml)。刺激の18~24時間後、プレートをPBSで洗浄し、次いでビオチン化抗マウスIFN-γmAb(R&D Systems,Minneapolis,MN)と共に4℃で24時間インキュベートした。次に、プレートをPBSで再度洗浄し、ストレプトアビジン-アルカリホスファターゼ(MabTech,Sweden)を各ウェルに添加し、室温で2時間インキュベートした。最後に、プレートをPBS中で再び洗浄し、次いでBCIP/NBT Plus基質(MabTech)を各ウェルにスポット現像のために5~30分間添加した。現像プロセスが目視検査で完了するとすぐに、プレートを蒸留水ですすぎ、次に室温で一晩乾燥させた。スポットは、自動ELISPOTリーダー(Cellular Technology Ltd.,Shaker Heights,OH)を用いて計数した。
脾細胞を96ウェルプレートに添加し(1×106細胞/ウェル)、個々のペプチドまたは「最小ペプチドプール」(終濃度2.5μg/ml)のいずれかで5~6時間刺激した。個々のペプチド刺激を、改変ELISPOT(表1~6)によって同定したすべてのペプチドの機能的確認及び表現型の特徴付けに用いた。脾細胞及び形質移入した293Tを、LIVE/DEAD(登録商標)Fixable Violet Dead Cell Stain Kit(Invitrogen)で最初に予め染色した。脾臓については、CD19(V450;クローン1D3)、CD4(PE-Cy7;クローンRM4-5)、CD8α(APC-Cy7;クローン53-6.7)及びCD44(PE-Cy5;クローンIM7)(BD Bioscience, San Jose, CA)について、細胞を表面染色し、PBS+1%FBSで3回洗浄し、BD Cytofix/Cytoperm(商標)キットで透過処理し、次いでIFNγ(APC;クローンXMG1.2)、TNF(FITC;クローンMP6-XT22)、CD3(PE-cy5.5;クローン145-2C11)、及びT-bet(PE;クローン4B10)(eBioscience,San Diego,CA)で細胞内染色した。形質移入した293T細胞におけるGP発現を、形質移入の24時間後に評価した。3回免疫化したH-2bマウス由来の血清を、それらのそれぞれのDNAワクチンまたはpVAX1空ベクター対照を用いてプールすることにより産生した、示したマウス由来GP特異的ポリクローナル血清試薬(1:200希釈)を含有するPBS+1%FBS中、4℃で30分間のインキュベーション後に間接染色を行った。次いで、細胞をFITC結合ヤギ抗マウスIgG(BioLegend,San Diego,CA)で染色し、広範囲に洗浄し、次いでMHCクラスI(HLA-ABC;PE-Cy7;クローンG46-2.6;BD)で染色した。すべての細胞を1%パラホルムアルデヒドで固定した。LSRIIフローサイトメーター(BD)を用いてすべてのデータを収集し、FlowJoソフトウェア(Tree Star,Ashland,OR)を用いて分析した。脾細胞は、CD3+CD44+、CD4+またはCD8+であり、B細胞マーカーCD19及び生存性色素について陰性である活性化IFNγ産生T細胞に関してゲーティングした。
無根系統樹の有意性を最大尤度法によって決定し、ブートストラップ解析によって検証し、MEGAバージョン5ソフトウェアによって有意な支持値(≧80%;1,000回のブートストラップ反復)を決定した。グループの解析は、整合両側独立t検定によって完了し、生存曲線は対数ランク(Mantel-Cox)検定によって解析した。すべての値は平均±SEMであり、統計解析はGraphPad Prism(La Jolla,CA)により実施した。
ワクチンの構築と発現
系統発生解析により、EBOV GP(SUDVでは94.4%、ZEBOVでは92.9%)が相対的に保存され、MARV GP(MGP)はより多様性が高い(約70%の保存)。したがって、優勢なZEBOV及びSUDV GPアミノ酸配列のアラインメントによって決定されるコンセンサス戦略をEBOV GPに採用し、一方、MARVには最大級で最も致命的なMARVアウトブレイクである2005年のアンゴラアウトブレイク配列を用いた型適合戦略を用いた。各GP導入遺伝子を遺伝子最適化し、商業的に合成し、次いで改変pVAX1哺乳類発現ベクターにサブクローニングした。まとめると、3プラスミド戦略が、新規多価-フィロウイルスワクチン戦略の基礎を形成した。
保護の有効性を判定するために、モルモット前臨床チャレンジモデルを採用した。前臨床免疫原性及び有効性試験は、本明細書においてモルモット及びマウスモデルを用いて実施した。モルモット前臨床モデルは、フィロウイルスワクチン開発のためのスクリーニング及び「概念実証」ツールとして広く使用されている。モルモットではMARV及びEBOVの一次単離株は非致死的な病気を引き起こすが、この宿主では少数の継代により、フィロウイルス感染霊長類に認められる病理学的特徴と同様の病理学的特徴を有する致死的疾患を引き起こし得る変異体が選択される。同様に、マウスもまた、フィロウイルスワクチンの開発に広く使用されているが、モルモットモデルとは異なり、免疫及びT細胞応答を評価するための免疫検出試薬が広く入手可能である。マウス適応ZEBOV(mZEBOV)による感染は、EBOVに感染した霊長類に認められるものに類似した、標的器官における高レベルのウイルスによって特徴付けられる疾患ならびに肝臓及び脾臓における病的変化をもたらす。
保護DNAワクチン(プラスミドpEBOZ、pEBOS及びpMARV、三価DNAワクチンとも呼ばれる)によって促進される免疫相関をよりよく特徴づけるために、フィロウイルスワクチン開発のためのスクリーニング及び「概念証明」ツールとして広く使用されているマウスモデルを採用した広範囲の免疫検出試薬が利用可能である。最初に、B細胞応答を、2回のワクチン接種のそれぞれ20日後、各一価DNAワクチン40μgの注射の間の3週間、H-2dマウス(n=5/群)で評価した。これらの実験からのデータを図3A~3Cに示す。図3A及び図3Bに示すように、プレブリード対照試料ではGP特異的IgGはほとんど観察されなかったが、ワクチン接種後のすべての動物において有意な増加が検出された。精製SGPは利用できなかったため、精製ZGPを代理として用いた。SUDVワクチン接種したマウスのIgGは、ZGPに結合し、ワクチン誘発性のAb生成能力、及び異種間認識能力を実証した。さらに、1回の免疫化の後にワクチン接種した動物の100%において抗体陽転が起こり、その後、応答は同種のブーストによって有意に増加した;AVE逆エンドポイント希釈力価は、pMARV免疫マウスでは22.1倍、pEBOS及びpEBOZワクチン接種動物ではそれぞれ3.4倍及び8.6倍に増加した。BSL-4施設でZEBOV、SUDV-ボニファス、及びMARV-Angolaの中和について試料を次にアッセイした。中和アッセイの結果を図3Cに示す。すべての動物においてワクチン接種後にNAb力価の有意な増加が検出された。
次に、ZEBOVチャレンジに対するワクチン効力を、前臨床マウスモデルにおいて評価した。強力なNAb誘導及び保護データが観察されたため、マウスには1回のみワクチン接種を行った。マウス(H-2k;n=10/群)を40μgのpEBOZ DNAで免疫化し、28日後にBSL4施設において1,000 LD50のマウス適合ZEBOV(mZEBOV)でチャレンジして保護を評価した。チャレンジ後7日目までにすべての対照動物が感染により死亡したが、図5AはDNAワクチン接種したマウスが完全に保護されたことを示している(P=0.0002)。さらに、図5Bに示すように、対照マウスは、死亡まで体重の進行性の喪失を示した(P<0.0001)。
本発明者らは、前臨床げっ歯類の免疫原性及び有効性試験における多価-フィロウイルスワクチンの開発及び評価を報告する。モルモットでの2回のDNAワクチン接種及びmZEBOVに対するマウスでの「単回投与」DNAワクチン接種後に、gpMARV及びgpZEBOVによるチャレンジに対する完全な保護が観察された。今日まで、モルモットの遺伝子ワクチン接種には、裸のDNAの注射が含まれていた(Sullivan NJ,Sanchez A,Rollin PE,Yang ZY,Nabel GJ(2000). 本明細書において、3つの合成ザイールエボラウイルス(EBOV)糖タンパク質(GP)を発現するDNAワクチン製剤を記載する:GP配列アラインメント(1976-2014)に基づいて設計した2つと、2014のアウトブレイク株に適合する第3のコンストラクト。プラスミドIL-12(pIL-12)もまた、細胞性免疫応答をさらに増強するためのアジュバントとして含めた。多価GP DNAワクチン製剤を、DNA-DNAプライムブースト免疫化レジメンに従ってマカクに投与した。マカク(n=3または4/群)には、筋肉内送達及びその後の電気穿孔法によって多価GP DNA製剤+pIL-12を与えた。免疫原性の差異をアッセイし、異なる用量、レジメン(2、3、4、及び5回の注射)、及びその後の投与の間の異なる間隔での保護をモニタリングした。抗体及びT細胞応答の両方が、第1回注射の2週間後の動物の83%及び第2回注射後の動物の100%で観察された。マカクに、致死量のEBOVギニア・マコーナアウトブレイク株(1000pfu、7-Uウイルス)をチャレンジし、感染後28日間モニタリングした。4週間の間隔で少なくとも3回の注射を受けた動物の100%が致命的なチャレンジを生き延びた。動物は病気の徴候から完全に保護され、血液化学の上昇を示さなかった。興味深いことに、2回の注射を受けた動物の50%が致命的なチャレンジで生き残った。生存動物は病気の徴候を最小限に抑え、さらなる最適化により、2回の注射による完全な保護が潜在的に達成可能であることが示唆された。マウスにおけるさらなる最適化研究では、単回注射が100%保護的であり、ワクチン接種後8か月の長期免疫反応を誘発したことが判明した。
EBOV GP DNAワクチン製剤の開発
現在臨床試験中のワクチンには、rVSVΔG/ZEBOVGP、ChAd3プライム+MVAブースト、及びMVA汎フィロウイルスが含まれる。これらのワクチンは免疫原性であり、非ヒト霊長類(NHP)で保護され、単回投与保護を提供するが、抗ベクター免疫を発達させ、記憶応答の持続時間が不確定であり、ヒト臨床試験において有害反応をもたらし、すべての集団に適してはいない可能性がある。したがって、異種ザイールエボラウイルスに対する強い免疫応答を誘導することができる、よりクリーンな安全特性を有する追加のプラットフォームは、非常に有益であろう(図9)。
DNAワクチンの単回免疫化はマウスにおいて免疫原性である
ワクチン製剤の電気穿孔(EP)に続いて、BALB/cマウスに単一の筋肉内(IM)免疫化を与えた。28日目に、全IgG抗体価及びELISPOT-IFNγを測定した。各ワクチン製剤は、ロバストなIgG応答を生じた。IFNγ、IL2、及びTNFαを分泌する多機能CD4+及びCD8+T細胞は、すべてのワクチン及び製剤について同様であった(図11)。
二価または三価のワクチン製剤で免疫したBALB/cマウスに、1000LD50の異種エボラウイルスで致死的チャレンジを行った。チャレンジ株はマウス適応型Ebola Mayinga1976であった。対照プラスミドpVax1でワクチン接種したマウスは急速に体重を減少させ、7日目には生存していなかった。しかしながら、2価ワクチン製剤接種マウス及び3価ワクチン製剤接種マウスは、最大20日目まで死亡したマウスはなかった(図12)。
BALB/cマウスに、一価ワクチンの3xIM 40μg免疫化、続いて0、28及び84日の電気穿孔を与えた。IgG抗体価、INFγ、CD4+CD44+記憶T細胞、及びCD8+CD44+記憶T細胞を測定した。ロバストな免疫応答は、最後の注射後数か月で検出可能であった(図13)。
カニクイザルは、エボラワクチンの有効性と致死的チャレンジのモデルであるため、これをNHPとして使用した。カニクイザルには、Rhesus pIL12アジュバントとともに二価製剤、またはRhesus pIL12アジュバントI.M.とともに三価製剤のいずれかを投与し、続いてEPを投与した。免疫原性を理解するための異なる注射レジメン。グループ1には、4週間間隔で2価製剤の2回のIM-EP注射を与えた。グループ2には、4週間間隔で3価製剤の2回のIM-EP注射を与えた。グループ3には3価製剤の3回のIM-EP注射を与えた。免疫原性試験のための試料は、毎月採取し、最後の投与後1か月間採取した。(図6)。各DNAワクチン製剤は、ロバストな抗エボラGP抗体応答(GMT>103)及び抗GP T細胞応答を誘導した。各注射後に免疫応答を増強した(図14)。
グループ1、2及び3由来のカニクイザルに、最終的なDNA免疫化から投与28日後の1000TCID50ギニア-マコーナ2014C07ウイルス(7-U参照株)をチャレンジした。チャレンジ後28日間、動物をモニタリングした。チャレンジ後10日目に、対照動物が生存していなかったのに対し、チャレンジ後28日目にグループ3動物の4/4が生存し、チャレンジ後28日目にグループ2動物の4/8が生存し、チャレンジ後28日目にグループ1動物の3/4が生存した。(図15)。生存動物には重大な疾患の徴候はなかった。それらはまた、正常なCBC及び酵素レベルを維持していた。IM-EPによって送達される二価及び三価のDNAワクチンは、最終的なDNA注射後3か月以上検出可能な長期抗体及びT細胞応答を誘発する(図43)。
INO-4212の非盲検試験(INO-9012の存在下または非存在下)を実施した。INO-4212をIMまたはIDで投与した後、健康なボランティアで電気穿孔を行った。安全性及び免疫学的評価をモニタリングした。皮内送達及び筋肉内送達を比較した。被験者は合計69人であった。免疫化の前、ならびに2、6、及び14週において、ELISA分析を行った(ベースライン)。血清学的検査は、エボラ・ザイール糖タンパク質に対する陽性IgG抗体応答として定義される。
実施例7に記載の3つのEBOV DNA構築物に対する免疫応答データを記載する:ZEBOV(1976-1996)(ConEBOVGP#1またはINO-4201)、ZEBOV(2002-2008)(ConEBOVGP#2)のコンセンサス配列、及び2014年のギニアアウトブレイクからのマッチZEBOV配列(ギニアGP)。5つのワクチン、単一のDNA構築物のみを含む一価ワクチン、いずれかのConEBOVGP#1とギニアGPを含む二価ワクチン製剤、ならびにConEBOVGP#1、ConEBOVGP#2、及びギニアGPの3つすべてを含む三価製剤を開発した。コホート1~5の69名の被験者を、ELISAによる免疫応答の分析に含め、コホート1~5の75名の被験者を、ELISpotによる免疫応答の分析に含めた(図21、25)。
抗EBOVRの力価を、第2、6及び14週で各コホートについて測定した。第6週までに、各コホートは、0日目を上回る抗体力価の増加が認められた(図22~23)。各コホートの初回投与の反応性はほとんどまたは全くなかった。用量2は抗体陽転を開始し、コホート3及び5は最大の頻度を示す。3投与は4/5コホートを>90%抗体陽転させる。コホート3及び5は、この時点で100%の抗体陽転を示す(図24)。
コホート応答をペプチドプールによって分析した(図26~33)。ELISpot異常値を分析するために、各プールの第0日の値及び総EBOV応答を使用して異常値の閾値(平均日0値+(0日の3×STDEV))を作成した。この閾値は、正規分布した母集団の99%を包含すべきである。異常値の閾値より大きいベースライン値を表示した被験者はすべて除去し、残りの被験者とともに応答基準を生成した。
全コホートからの47人の被験者を分析に含めたが、コホート5は不十分であった(図35)。ベースライン及び14週目にICS分析を行った。プール1~4からなる単一のEBOVペプチドプールを刺激に使用する。CD4及びCD8区画の両方からのIFNgまたはTNFa産生の形態でのT細胞活性の分析は、コホート3のみでのTNFα及び/またはIFNgの上昇と同様にCD4及びCD8区画の両方におけるTNFαの有意な上昇を示唆する(Wilcoxon整合ペア分析、両側)。
コホート3(ID)患者の100%が、2回の投与後に抗体陽転した。コホート5(IM+IL12)患者の92%が2回投与後に抗体陽転し、3回投与後に100%抗体陽転した。他のコホートは、2回の投与後には最高で67%を示し、3回の投与後には93%の高さに及んだ。
本明細書において、清潔な安全性特性を有し、かつ血清に依存せず、可能な反復ベクター投与を可能にする新規エボラウイルス疾患(EVD)DNAワクチンを提示する。3つの新規合成ザイールエボラウイルス(EBOV)GP DNAワクチンを設計し、2価または3価製剤を開発した。両方のEBOV-GP DNAワクチンは、カニクイザルにおける致死的EBOVマコーナC07チャレンジに対して高度に保護的(75~100%)であった。異なるレジメンの動物(n=4~5/群)を追跡し、DNA免疫後の長期間の免疫原性をモニタリングした。すべてのNHPは迅速に抗体陽転した。NHPは、IFNγ、IL2、及びTNFαを発現する多機能性CD4及びCD8 T細胞及び記憶サブセット集団(図47~59)における応答を含むEBOV GP抗原に対する耐性全IgG抗体力価及びT細胞応答を有する。同時に、データは強力な記憶免疫応答の生成と保護のためのEBOV-GP DNAワクチンの供給を強く支持している。
