全てのペプチドは、標準的なFmoc化学を使用して、Syro IIペプチド合成装置を用いて施設内で生成した。引き続いて、HPLCを使用してペプチドを分析し、平均純度は74%であった。UV感光性ペプチドは、2−ニトロフェニルアミノ酸残基を有する感光性構成要素を含有した。ジペプチドGMは、Bachemから調達した。使用前に、ペプチドをDMSO(Sigma、カタログ番号41640)、0.5%TFA(Sigma、カタログ番号T6508)で、所望の使用例に応じて2mg/ml〜10mg/mlの範囲の濃度で溶解した。
組換えHLA−A*02:01野生型(WT−A*02:01、配列番号322)、またはC末端BirAシグナル配列とヒトβ2m軽鎖とを有するジスルフィド修飾HLA−A*02:01重鎖を大腸菌(Escherichia coli)中の封入小体として生成し、既述のように精製した(2)。HLA−A*02:01複合体の再折りたたみ反応は、わずかな修正を加えて既述のように実施した(Saini et al 2013)。手短に述べると、WT−A*02:01またはジスルフィド修飾HLA−A*02:01重鎖、β2m軽鎖、およびペプチドを再折りたたみ緩衝液(100mMトリス・Cl、pH8、0.5Mのアルギニン、2mMのEDTA、0.5mMの酸化グルタチオン、5mMの還元グルタチオン)で希釈し、濃縮前に、撹拌しながら4℃で2〜8日間インキュベートした。濃縮されたタンパク質は、HiLoad 26/600200pgカラム(GE Healthcare)を使用して、AKTAprimeシステム(GE Healthcare)上で、20mMのTris−HCl、pH8/150mMのNaCl中でサイズ排除クロマトグラフィー(SEC)によって精製した。ピーク画分を2000μg/mlに直接濃縮し、分注して−80℃で凍結し、または製造業者の取扱い説明書に従って4℃で一晩BirAビオチン−タンパク質リガーゼ(Avidity)によってビオチン化し、2000μg/mlに最終濃縮する前に第2のゲル濾過に供し、分注して−80℃で保存した。
UV光切断可能ペプチドとのペプチド交換反応は、既述のように実施された。要約すると、所望の九量体ペプチドをビオチン化されたUV感光性pMHC複合体と100対1のモル比で混合し、366nmのUV光(Camag)に少なくとも30分間曝した。
可溶性TCRは、既述のように生成した(20)。要約すると、TCRαおよびTCRβ鎖コンストラクトを大腸菌中の封入小体として別々に発現させ精製した。TCRα鎖はスレオニンをシステインに置き換えることによって48位で変異させ、TCRβ鎖はセリンをシステインに置き換えることによって57位で変異させ、鎖間ジスルフィド結合を形成させる。
bs−868Z11−CD3分子は、scTv 868Z11をヒト化抗CD3抗体のF(ab’)ドメインのC末端に結合することによって生成した(22、23)。この目的を達成するために、scTvのVβドメインをヒトIgG2(APPVAG、配列番号2)に由来する上部CH2領域に直接融合させた。ヒンジ内のシステインノックアウトC226SおよびC229Sは、F(ab) 2分子の形成を防ぐ。記のコンストラクトまたはヒト化抗CD3抗体の軽鎖のどちらかをコード化するHCMV駆動発現ベクターをExpiCHO細胞(Thermo)に一過性に同時形質移入した。12日後、上清をタンデムクロマトグラフィー(プロテインLとそれに続く分取サイズ排除、GE Biosciences)で処理し、高純度のモノマーbsTCRをPBS中で調合した。
異なるpMHC複合体に対するsTCRまたはbsTCR分子の親和性は、動態または定常状態の結合分析を使用して、OctetRED384システム(Pall Fortebio)上で測定した。全ての分析物またはリガンドは、特に指定されていない場合は、動態緩衝液(PBS、0.1%BSA、0.05%ツィーン20)でそれらの最終濃度に希釈した。全てのバイオセンサーは、使用前に動態緩衝液で少なくとも10分間水和させた。負荷および測定は、3mmのセンサーオフセットで、少なくとも40μlの384傾斜ウェルプレート(Pall Fortebio)内で実施した。プレート温度は25℃、振盪機速度は1000rpmに設定した。ステップ間の補正ベースラインを可能にするために、結合期の前と次の解離期を同じウェル内で実施した。必要に応じて、動態緩衝液を適切な濃度でDMSOが添加された解離緩衝液として使用し、分析物の組成を一致させた。
TAP欠損HLA−A*02:01発現細胞株T2をATCC(CRL−1992)から調達し、10%熱不活化FCS(Life Technologies、カタログ番号10270106)および抗生物質ペニシリンおよびストレプトマイシン(Biozym、カタログ番号882082、各100μg/ml)を添加したRPMI培地1640 GlutaMAX(商標)(Thermo Fisher、カタログ番号61870010)中で、必要に応じて継代数16まで培養した。GloResponse(商標)NFAT−luc2 Jurkat細胞株をPromega(カタログ番号CS1764)から継代数6で調達し、10%熱不活化FCS(Life Technologies、カタログ番号10270106)、1%ピルビン酸ナトリウム(C.C.Pro、カタログ番号Z−20M)および抗生物質ハイグロマイシンB(Merck Millipore、カタログ番号10270106)、1%ピルビン酸ナトリウム(C.C.Pro、カタログ番号Z−20M)および抗生物質ハイグロマイシンB(Merck Millipore、カタログ番号400052、200μg/ml)、ペニシリンおよびストレプトマイシン(Biozym、カタログ番号882082、各100μg/ml)を添加したRPMI培地1640 GlutaMAX(商標)(Thermo Fisher、カタログ番号61870010)中で培養し、必要に応じて継代数14まで培養した。
GloResponse(商標)NFAT−luc2 Jurkat細胞およびペプチドを負荷したT2標的細胞を使用したT細胞活性化アッセイは、製造業者の取扱い説明書に従って実施した。要約すると、連続細胞培養からT2細胞を採取し、T2培地中で洗浄して3.3×106細胞/mlの濃度で再懸濁し、96ウェル丸底プレート(Corning costar(登録商標)、カタログ番号3799)に移した。DMSO、0.5%TFA中のペプチドを100nMの最終濃度で添加し、懸濁液を37℃、5%CO2で2〜3時間にわたりインキュベートした。PBS中で調合したbsTCRをT2培養培地中で所望の濃度に希釈し、それぞれの希釈液の25μlを白色96ウェル平底プレート(Brand、カタログ番号781965)に分配した。GloResponse(商標)NFAT−luc2 Jurkat細胞を連続細胞培養から採取して洗浄し、3.0×106細胞ml−1の濃度でT2培地に再懸濁し、細胞懸濁液の25μlをbsTCR希釈液と共に白色96ウェル平底プレートに分配した。
Y84C/A139C HLA−A*02:01−SLLMWITQV複合体と1G4TCRを濃縮して1:1の比率で混合し、結晶化のために7mg/mlの濃度を得た。シッティングドロップ蒸気拡散実験は、0.1M酢酸アンモニウム、0.1Mビストリス(pH5.5)、および17%ポリエチレングリコール(PEG)10,000を含有する母液の存在下で、結晶をもたらした。単結晶は、0.1M酢酸アンモニウム、0.1Mビストリス(pH5.5)、20%(w/v)PEG10,000、および10%グリセロールを含有する凍結防止剤溶液に移した。結晶は、EIGER16M検出器を収容するドイツ電子シンクロトロンで、EMBL P14ビームライン上に載せて、100Kで極低温冷却させた。X線データセットは、2.5Aの分解能で収集した(表2)。
検索モチーフによって許容される潜在的な組み合わせの1つに一致する九量体ペプチドリガンドの検索は、NCBIヒトタンパク質データベースを使用して実施した。このデータベースは、全ての非冗長GenBank CDS翻訳物、ならびにPDB、SwissProt、PIR、およびPRFからの記録をカバーするが、全ゲノムショットガンプロジェクトからの環境サンプルは除外されている。データベースは、NCBIサーバーから直接取得した。
結合モチーフを可視化したSeq2Logosは、それぞれの相互作用のKd値の逆数を取り、108で除して作成した。これらの値は、位置特異的重み行列ファイル形式で組み立て、デンマーク工科大学バイオインフォマティクス部門のSeq2Logoオンラインリソース(27)のPSSM−Logoタイプを用いて処理した。
ペプチド結合は、300nMの精製された再折りたたみY84C/A139C HLA−A*02:01を用いた蛍光異方性アッセイにおいて評価した。100nMの蛍光標識された高親和性ペプチドNLVPKFITCVATV(Genecast)を折りたたまれたY84C/A139C HLA−A*02:01に添加し、動態測定は、異方性(FITCλex=494nm、λem=517nm)を測定する、Tecan Infinite M1000 PRO(Tecan、独国クライルスハイム)マルチモードプレートリーダーを用いて実施した。Y84C/A139C HLA−A*02:01は、再折りたたみ後に直接使用するか、または測定前に示された時間にわたり保存緩衝液(10%グリセロール、50mMのTris−HCL、pH8.0)中で−80℃で保存した。動態測定は、50mMのHEPESバッファー、pH7.5で室温(22〜24℃)で実施した。データは、GraphPad Prism v7を使用してプロットした。
ストレプトアビジン(Molecular Probes、カタログ番号S888)をPBS中の最終濃度2μg/mlでNunc MAXIsorpプレート(Thermo Fisher、カタログ番号439454)に添加し、密閉プレートを湿った環境下で室温で一晩インキュベートした。翌日、ELx405プレート洗浄機(Biotek)を使用して、プレートを洗浄緩衝液洗浄(PBS、0.05%ツイーン−20)で4回洗浄した。300μlのブロック緩衝液(2%BSA添加PBS)を各ウェルに添加し、密封プレートを37℃で1時間インキュベートした。ブロック緩衝液は、それぞれのUV交換pMHC調製物のブロック緩衝液中の1:100希釈の100μlを添加する前に廃棄した。従来の再折りたたみpMHCモノマーに基づく500ng/ml〜15.6ng/mlの標準系列が、各プレートに含まれていた。エッジ効果を低減するために、縁部ウェルを300μlのブロック緩衝液で充填し、密封プレートを37℃で1時間インキュベートした。プレートを再度4回洗浄した後、100μlの抗β2ミクログロブリンHRP結合二次抗体(Acris、カタログ番号R1065HRP)を最終濃度1μg/mlで各ウェルに添加した。密封プレートを37℃で1時間インキュベートした。プレートを洗浄緩衝液で再度4回洗浄した後、100μlの室温TMB基質(Sigma、カタログ番号T0440)を各ウェルに添加した。プレートを室温で5分間インキュベートした後、各ウェルに50μlの1N H2SO4を添加して、停止させた。Synergy2プレートリーダーを使用して450nmで5秒間にわたり吸光度を読み取ることにより、プレートを即座に分析した。pMHC濃度は、Synergy2ソフトウエアを用いた標準曲線適合(Log(Y)=A*Log(X)+B)に基づいて算出した。データは、GraphPad Prism v7を使用してプロットした。
TAP欠損HLA−A*02:01発現細胞株T2をATCC(CRL−1992)から調達し、10%熱不活化FCS(Life Technologies、カタログ番号10270106)および抗生物質ペニシリンおよびストレプトマイシン(Biozym、カタログ番号882082、各100μg/ml)を添加したRPMI培地1640 GlutaMAX(商標)(Thermo Fisher、カタログ番号61870010)中で、必要に応じて継代数16まで培養した。連続細胞培養からT2細胞を採取し、T2培地中で洗浄して3.3×106細胞/mlの濃度で再懸濁し、96ウェル丸底プレート(Corning costar(登録商標)、カタログ番号3799)に移した。DMSO、0.5%TFA中のペプチドを10μMの最終濃度で添加し、懸濁液を37℃、5%CO2で2時間インキュベートした。プレートをPFEA(PBS、2%FCS、2mMのEDTA、0.01%アジ化ナトリウム)で2回洗浄した後、PFEAで1:250に希釈したウェルあたり50μlのPE標識抗ヒトHLA−A2(Biolegend、カタログ番号343305)を最終濃度0.8μg/mlに添加した。プレートを4℃で30分間インキュベートした後、PFEAで2回洗浄した。最後に、細胞を固定化溶液(PFEA、1%ホルムアルデヒド)に再懸濁して4℃で保存した後、iQue Screener(Intellicyt)を用いて分析した。T2細胞をFSC−A/SSC−Aシグナルに基づいてゲートし、FSC−H/FSC−Aダブレット排除を使用してダブレットを除去した。PEチャネルの正ゲート座標は、未染色対照に基づいていた。データは、GraphPad Prism v7を使用してプロットした。
Clustal Omegaの多重配列アラインメントを使用して、多配列アラインメントを実施した。(www.ebi.ac.uk/Tools/msa/clustalo/)(Madeira et al.”The EMBL−EBI search and sequence analysis tools APIs in 2019”,Nucleic Acids Research,47:W636−W641,2019,doi:10.1093/nar/gkz268)。
全てのデータは、GraphPad Prismソフトウエアバージョン7を使用してプロットした。xデータセットとyデータセット間の相関は、ピアソン相関係数を算出することによって計算され、GraphPad Prismソフトウェアバージョン7を使用してR2として報告した。バイオレイヤー干渉結合動態測定の曲線適合のためのR2およびX2値は、Octet RED384システムソフトウエアDataAnalysis HTバージョン10.0.3.7を使用して計算した。
空のMHCクラスI分子とペプチド負荷MHCクラスI分子の分子動力学シミュレーションは、前者がペプチドリガンドのC末端を収容するFポケット内での可動性が増加していることを示している(16)。マウスMHCクラスI分子H−2Kbに関する先行研究では、84位のチロシンと139位のアラニンをシステインに変異させることで、Fポケット内の対向する残基間にジスルフィド結合を導入することで、複合体を安定化できた。この変異体は、全長ペプチドなしで再折りたたみができ、遡及的なペプチド結合が可能であった(17、18)。
次に、本発明者らは、ジスルフィド修飾HLA−A*02:01分子が、ペプチド−MHC複合体形成およびTCRリガンド結合できるかどうかを判定した。親和性測定は、可溶性分析物として再折りたたみTCR1G4を用いて、OctetRED 384上でバイオレイヤー干渉法(BLI)によって実施した。このTCRは、がん精巣抗原NY−ESO−1またはその合成変異型SLLMWITQV(ESO 9V、配列番号4)に由来するHLA−A*02:01特異的ペプチドSLLMWITQC(ESO 9C、配列番号3)を認識する(20,21)。ビオチン化されたY84C/A139CHLA−A*02:01は、その空の状態で直接固定化し、またはストレプトアビジン被覆されたバイオセンサー上でペプチドESO 9Vと短時間インキュベートした後に固定化した(図1b)。組立後に親和性測定を開始するのに必要な最短時間である5分間のペプチドインキュベーションと、より長い時間との間に差は検出されなかった。さらなる分析では、高濃度のペプチドを使用した場合、1〜2分以内に完全な交換が実際に達成されたことが示された。動態を複数の1G4濃度にわたって測定し、野生型HLA−A*02:01がESO 9Vで直接再折りたたみされたものを対照として使用した。
未修飾TCRに対するWT−A*02:01と同等のリガンドとしてのY84C/A139C HLA−A*02:01分子の有用性が確立されたことから、本発明者らは、この解析を変異した高親和性TCRおよびより多くのペプチドリガンドに向けて拡張したいと考えた。本発明者らは、HIV p17 Gag由来HLA−A*02:01拘束性ペプチドSLYNTVATL(SL9、配列番号5)を認識するTCRの親和性成熟変異型である成熟一本鎖TCR(scTv)868Z11を使用した(8、22)。

pMHCの迅速かつ柔軟な生成は、多くの異なるpMHCに対する大規模な結合親和性データセットの収集を容易にする。このようなデータセットの一例は、位置スキャンライブラリをスクリーニングしてpMHC−bsTCR結合モチーフを生成することであり、これは、bsTCR安全性スクリーニングアプローチの1つの構成要素の役割を果たし得る。このような測定を実施するためには、pMHCは理想的には可溶性分析物として使用されるべきであり、これは複数の利点を提供するためである。第一に、例えば、bsTCRなどの既知の活性を有する同一リガンドを繰り返し固定化することにより、適合結果、特に報告されたRmax値をより良く解釈することができる。第二に、二重特異性TCRコンストラクトを可逆的に固定化できる多種多様な再生可能なバイオセンサーが存在することから、固定化の代わりに可溶性分析物としてpMHC複合体を使用することは、迅速で費用効果の高い高スループットスクリーニングにのために好ましい。これらのバイオセンサーは通常、抗体で被覆されており、読み取り品質を損なうことなく、動態測定に少なくとも20回使用できる。第三に、bsTCRを固定化することは、bsTCRまたは抗体が複数のpMHC結合部位を有する場合、一価親和性を測定するために利用可能な唯一の方向性であるが、これは、固定化されたpMHCでは、結合力しか測定できないためである。
本発明者らはさらに、生成された結合モチーフを用いて、ヒトゲノムから交差反応性ペプチドリガンドを同定し得るかどうか探求したいと考えた。本発明者らは、50nMの例示的なKd閾値を導入することによって、親和性データセットからペプチドリガンド検索モチーフを作成し;その閾値を超えてbs−868Z11−CD3Kdを増加させる全ての単一アミノ酸置換は、モチーフから除外した(表4)。このモチーフに基づいて、本発明者らは、NCBIヒト非冗長タンパク質配列データベースにおいて、モチーフによって許容される組み合わせに一致する九量体配列を検索した。この検索は、ヒトゲノム内で400を超えるヒットを同定し、野生型配列SLYNTVATLとの配列同一性は、1〜6位の同一位置に及んだ。140のペプチドを選択し、より大きなグループの配列同一性分布に相当するようにサンプリングし、合成して親和性測定のために使用した。(表5;配列番号178−317)。本発明者らは、これらのペプチドのうちの91について、1桁のKds以上の結合親和性を検出できた。
pMHC−bsTCRの結合親和性は、この抗スループットスクリーニングプラットフォームを使用して測定され得るが、さらにより有用であるためには、機能性T細胞係合bsTCRとしての生体外活性と一貫性がなくてはならない。一般に、適切な読み取りと連動する標的細胞とエフェクター細胞の生体外同時インキュベーションを使用して、これらのコンストラクトを特性評価する。NFAT応答エレメントによって駆動されるルシフェラーゼレポーター遺伝子を発現する細胞株であるGloResponse(商標)NFAT−luc2 Jurkatエフェクター細細胞、および外因性ペプチド負荷を通じた復元可能なpMHC提示を有するTAP欠損A*02:01細胞株であるペプチド負荷T2標的細胞をbs−868Z11−CD3の存在下でインキュベートし、この文脈での動態スクリーニングの有意性を裏付けた。T2細胞に100nMの濃度で位置スキャニングライブラリからのそれぞれのペプチドを別々に負荷し、引き続いて読み取りの前に、Jurkatsおよび異なるbsTCR濃度と共に18時間にわたり同時インキュベートした。予測通り、本発明者らは、いかなるbsTCR濃度でも検出可能なT細胞活性化がないものから、例えば野生型ペプチド(図5a)のように低濃度から始まる強い応答に至るまで、幅広いスペクトルの結果に遭遇した。選択されたbsTCR濃度範囲の多くの相互作用についてEC50値を判定できなかったので、本発明者らは、3倍を超えるシグナルを誘導することができる最低のbsTCR濃度として定義される、T細胞活性化の開始によって、個々のペプチドを分類した。開始値をそれぞれ、測定されたKDsに対してプロットした(図5b)。
1G4TCRがESO9VY84C/A139CHLA−A*02:01をESO9VWT−A*02:01と区別なく認識することをさらに確認する。野生型ESO9V HLA−A*02:01分子について以前に報告されたように、TCRと、ESO 9Vで再折りたたみされたジスルフィド安定化MHCとを共結晶化し、X線結晶学で分析した(表2)(21)。結晶構造の比較は、双方の複合体間の高度の構造的重複を明らかにした。双方のHLA−A*02:01分子の主鎖は、Cα(T細胞受容体のα鎖の定常部分;図7A)に対して計算された、1.14Aの二乗平均平方根偏差(RMSD)値でほぼ完全に整列していた。同じことは、ジスルフィド結合に近接している場合でさえも、全ての原子にわたって計算された1.27AのRMSD値で、それらの側鎖を含めた双方の結合ペプチドに当てはまった(図7B)。1G4 TCRとの相互作用についても、同様の結論を下し得た。WT−A*02:01 1G4とY84C/A139CHLA−A*02:01 1G4の結晶構造を比較すると、ペプチドおよびMHC主鎖と相互作用する相補性決定領域(CDR)ループ領域は、境界面のわずかな偏差と4.13°の小さなドッキング角の変化を示した。このシフトは、繰り返し結晶化した場合でも、同一複合体の予測偏差の範囲内であった(図7、CおよびD)。総合して、判定された結合親和性および結晶構造は、TCR結合に関して、野生型複合体と比較して、Y84C/A139C HLA−A*02:01 pMHC複合体のペプチド受容性および同様の特性を示す。1G4 Y84C/A139C HLA−A*02:01 ESO 9V複合体の結晶構造は、受入れ番号6Q3SでPDBに寄託されている。
本明細書において、本発明者らは、ジスルフィド安定化された機能的に空のHLA−A*02:01分子を提示しており、これは、例えばジペプチドGMなどの典型的に必要とされる高親和性ペプチドを使用することなく、再折りたたみおよび精製され得る。得られたモノマーは、一段階の負荷手順でペプチドを添加した後に、pMHCを形成し得る。ジスルフィド橋はMHC分子の安定性を高めるが、導入は野生型と比較して、ジスルフィド修飾HLA*02:01 pMHC複合体へのTCRの結合を阻害せずまたは有意に変化させない。この技術は、親和性測定に適した大規模なpMHCライブラリを迅速に作成するための優れたツールである。ジスルフィド修飾HLA*02:01で生成されたpMHC複合体ライブラリをバイオレイヤー干渉法に基づく分析と組み合わせることで、高スループットpMHC−bsTCR結合動態スクリーニングが可能なプラットフォームがもたらされる。この設定は、モノクローナル抗体または二重特異性T細胞エンゲージャーなどの二重特異性物質のような、pMHC複合体を標的化するその他の生物製剤の分析にも役立ち得る。このプラットフォームの1つの適用において、本発明者らは、HIV特異的bsTCR bs−868Z11−CD3についてのpMHC−bsTCR結合親和性データセットを迅速に収集できた。それぞれのpMHCに対するbsTCR結合親和性は、提示されたT細胞エンゲージャーと連動して細胞レポーターアッセイで試験した場合の生体外活性の指標であり、これらのデータセットはbsTCRの特性評価のために有益である。広範囲のpMHC−bsTCR親和性にわたる、結合親和性とbsTCR媒介細胞活性化との関係の分析は、このようなデータセットを実行可能に収集するために利用できるツールが限られていることから、これまで困難であった。

Claims (24)
- a)(i)MHC Iの場合、前記安定化されたMHC分子のα1ドメインの1つのアミノ酸とα2ドメインの1つのアミノ酸との間に;および/または
(ii)MHC Iの場合、前記安定化されたMHC分子のα1ドメインの2つのアミノ酸の間に;または
(iii)MHC IIの場合、前記安定化されたMHC分子のα1ドメインまたはβ1ドメインの2つのアミノ酸の間に;および/または
(iv)MHC IIの場合、前記安定化されたMHC分子のα1ドメインの1つのアミノ酸とβ1ドメインの1つのアミノ酸との間に、
少なくとも1つの人為的に導入された共有結合架橋を含んでなる、適切に安定化されたMHC分子を提供するステップと、
b)前記適切に安定化されたMHC分子をその多数のペプチドリガンドに接触させて、ペプチドリガンド/MHC(pMHC)分子複合体を形成するステップと、
c)前記pMHC分子複合体をTCR結合についてスクリーニングするステップと
を含んでなる、TCR結合ペプチドリガンド/MHC分子複合体(pMHC)をスクリーニングする方法。 - 前記MHC分子が、HLA、二量体、三量体、および四量体からなる群から選択される、HLA、MHC IまたはMHC II多量体である、請求項1に記載の方法。
- アミノ酸間の前記人為的に導入された少なくとも1つの共有結合架橋が、組換え的に導入されたジスルフィド橋、架橋されるべき非天然アミノ酸の導入、光架橋アミノ酸の導入、および化学的に導入された架橋から選択される、請求項1または2に記載の方法。
- アミノ酸間の前記人為的に導入された少なくとも1つの共有結合架橋が、
(i)好ましくは、MHC Iの84位のアミノ酸および139位のアミノ酸をシステインに変異させることによって、αらせん間に;
(ii)好ましくは、MHC Iの71位のアミノ酸およびMHC Iの22位のアミノ酸をシステインに変異させることによって、MHC Iのα1ドメインのαらせんとβシートの間に;
(iii)好ましくはMHC Iの51位のアミノ酸およびMHC Iの175位のアミノ酸をシステインに変異させることによって、αらせん間に;または
(iv)好ましくはMHC Iの71位のアミノ酸およびMHC Iの22位のアミノ酸をシステインに変異させることによって、αらせんとβシートの間に;および好ましくはMHC Iの51位のアミノ酸およびMHC Iの175位のアミノ酸をシステインに変異させることによって、αらせん間に、
導入される、請求項1〜3のいずれか一項に記載の方法。 - 前記負荷されたHLA/ペプチド分子が、約4℃で、約1日以上、好ましくは、約1週間以上安定である、請求項1〜4のいずれか一項に記載の方法。
- pMHC複合体への結合についてのTCRの親和性スクリーニングの感度レベルが、約Kd1.0×10−9よりも高く、好ましくは約Kd1.0×10−6よりも高く、より好ましくは約Kd1.0×10−3よりも高い、請求項1〜5のいずれか一項に記載の方法。
- 前記TCRまたは前記MHC分子のどちらかが、チップ、バイオセンサー、スライドガラスまたはビーズからなる群から選択される固体表面上に適切に固定化されている、請求項1〜6のいずれか一項に記載の方法。
- 前記方法が、高スループットスクリーニング形式で実施される、請求項1〜7のいずれか一項に記載の方法。
- (i)MHC Iのα1ドメイン内の2つのアミノ酸;および/または
(ii)アミノ酸160〜179位内のMHC Iのα1ドメイン内の1つのアミノ酸とMCH Iのα2ドメイン内の1つのアミノ酸;または
(iii)MHC IIのα1ドメイン内の2つのアミノ酸またはβ1ドメイン内の2つのアミノ酸;および/または
(iv)MHC IIのα1ドメイン内の1つのアミノ酸とβ1ドメイン内の1つのアミノ酸
の間に、少なくとも1つの人為的に導入された共有結合架橋を含んでなる、安定化されたMHC分子またはそのペプチド結合断片を含んでなるまたはそれからなる、ポリペプチド。 - (i)好ましくはアミノ酸12〜32位内、好ましくはアミノ酸17〜27位内、より好ましくはアミノ酸20〜24位内、最も好ましくはアミノ酸22位のβ1ユニット内で、およびアミノ酸61〜81位内、好ましくはアミノ酸66〜76位内、より好ましくはアミノ酸69〜73位内、最も好ましくはアミノ酸71位のα1ユニット内で、1つのアミノ酸がMHC Iのα1ドメインのβ1ユニット内で修飾され、別のアミノ酸がMHC Iのα1ドメインのα1ユニット内で修飾され;および/または
(ii)好ましくはアミノ酸50〜70位内、より好ましくはアミノ酸50〜60位内、より好ましくは50〜54、最も好ましくはアミノ酸51位で、1つのアミノ酸がα1ドメインのα1ユニット内で修飾され、およびα2ドメイン内で修飾された別のアミノ酸は、アミノ酸165〜178位内、好ましくはアミノ酸170〜177位内、より好ましくはアミノ酸173〜176位内、最も好ましくはアミノ酸175位であり;または
(iii)好ましくはアミノ酸10〜40位内、好ましくはアミノ酸13〜35位内、より好ましくはアミノ酸22〜25位内、最も好ましくはアミノ酸22位のβ1ユニット内で、およびアミノ酸45〜78位内、好ましくはアミノ酸50〜70位内、より好ましくはアミノ酸56〜66位内、最も好ましくはアミノ酸59位のα1ユニット内で、1つのアミノ酸がMHC IIのα1ドメインのβ1ユニット内で修飾され、別のアミノ酸がMHC IIのα1ドメインのα1ユニット内で修飾され;および/または
(iv)好ましくはアミノ酸5〜53位内、好ましくはアミノ酸17〜41位内、より好ましくはアミノ酸21〜28位内、最も好ましくはアミノ酸26位のβ3ユニット内で、およびアミノ酸52〜88位内、好ましくはアミノ酸66〜76位内、より好ましくはアミノ酸65〜80位内、最も好ましくはアミノ酸75位のα3ユニット内で、1つのアミノ酸がMHC IIのβ1ドメインのβ3ユニット内で修飾され、別のアミノ酸がMHC IIのβ1ドメインのα3ユニット内で修飾され;および/または
(v)好ましくはアミノ酸70〜95位内、好ましくはアミノ酸74〜94位内、好ましくはアミノ酸83〜93位内、より好ましくはアミノ酸87〜92位内、最も好ましくはアミノ酸89位のα3ユニット内で、およびアミノ酸50〜70位内、より好ましくはアミノ酸50〜60位内、より好ましくは50〜54位内、最も好ましくはアミノ酸51位のα1ユニット内で、1つのアミノ酸がMHC IIのβ1ドメインのα3ユニット内で修飾され、別のアミノ酸がMHC IIのα1ドメインのα1ユニット内で修飾される
請求項9に記載のポリペプチド。 - MHC Iの74〜84位、好ましくは84位のアミノ酸と138〜149位、好ましくは139位のアミノ酸をシステインに変異させることによる、α1ドメインのアミノ酸とα2ドメインのアミノ酸との間の少なくとも1つの人為的的共有結合架橋をさらに含んでなる、請求項9または10に記載のポリペプチド。
- a)事前選択されたTCRを含んでなる、請求項1〜8のいずれか一項に記載の方法を実施するステップと、
(b)TCR結合が検出された前記ペプチドリガンド/MHC分子複合体中のそれらのペプチドリガンドのアミノ酸配列を判定および比較し、それによって前記事前選択されたTCRの特定のアミノ酸結合モチーフを同定する追加的なステップと
を含んでなる、TCRの特定のアミノ酸結合モチーフを検出または生成するための方法。 - 前記ペプチドリガンド/MHC分子複合体が、異なる濃度を有する並行アッセイ反応で使用され、好ましくは、前記方法ステップが、同定された前記事前選択されたTCRのための修飾アミノ酸結合モチーフからなるペプチドのプールを含んでなるように繰り返される、請求12項に記載の方法。
- a)請求項12または13に記載の方法を実施するステップと、
b)TCR結合が検出された前記ペプチドリガンド/MHC分子複合体中のそれらのペプチドリガンドのアミノ酸配列を判定および比較し、それによって前記TCRの交差反応性を同定する追加的なステップと
を含んでなる、TCRの交差反応性を検出または判定するための方法。 - a)事前選択されたTCRを含んでなる、請求項1〜8のいずれか一項に記載の方法を実施するステップと
b)TCR結合が検出された前記ペプチドリガンド/MHC分子複合体中のそれらのペプチドリガンドのアミノ酸配列を判定および比較し、それによって前記TCRの交差反応性を同定する追加的なステップと
を含んでなる、TCRの交差反応性を検出または判定するための方法。 - TCRと、前記TCRに結合するTCR結合ペプチドリガンド/MHC分子複合体とを含んでなる、T細胞活性化を測定するステップをさらに含んでなる、請求項1〜8または12〜15のいずれか一項に記載の方法。
- 少なくとも
(i)MHC Iのα1ドメイン内の2つのアミノ酸;および/または
(ii)アミノ酸160〜179位内のMHC Iのα1ドメイン内の1つのアミノ酸とMCH Iのα2ドメイン内の1つのアミノ酸;または
(iii)MHC IIのα1ドメイン内の2つのアミノ酸またはβ1ドメイン内の2つのアミノ酸;および/または
(iv)MHC IIのα1ドメイン内の1つのアミノ酸と、β1ドメイン内の1つのアミノ酸
を含んでなり、ビーズ、フィラメント、ナノ粒子、またはその他の適切な担体に結合されており、任意選択的にペプチドリガンドが負荷されている、安定化されたMHC分子またはそのペプチド結合断片を含むかまたはそれらからなるポリペプチドを含んでなる、医薬組成物。 - i)好ましくはアミノ酸12〜32位内、好ましくはアミノ酸17〜27位内、より好ましくはアミノ酸20〜24位内、最も好ましくはアミノ酸22位のβ1ユニット内で、およびアミノ酸61〜81位内、好ましくはアミノ酸66〜76位内、より好ましくはアミノ酸69〜73位内、最も好ましくはアミノ酸71位のα1ユニット内で、1つのアミノ酸がMHC Iのα1ドメインのβ1ユニット内で修飾され、別のアミノ酸がMHC Iのα1ドメインのα1ユニット内で修飾され;および/または
(ii)好ましくはアミノ酸50〜70位内、より好ましくはアミノ酸50〜60位内、より好ましくは50〜54、最も好ましくはアミノ酸51位で、1つのアミノ酸がα1ドメインのα1ユニット内で修飾され、およびα2ドメイン内で修飾された別のアミノ酸は、アミノ酸165〜178位内、好ましくはアミノ酸170〜177位内、より好ましくはアミノ酸173〜176位内、最も好ましくはアミノ酸175位であり;または

(v)好ましくはアミノ酸70〜95位内、好ましくはアミノ酸74〜94位内、好ましくはアミノ酸83〜93位内、より好ましくはアミノ酸87〜92位内、最も好ましくはアミノ酸89位のα3ユニット内で、およびアミノ酸50〜70位内、より好ましくはアミノ酸50〜60位内、より好ましくは50〜54位内、最も好ましくはアミノ酸51位のα1ユニット内で、1つのアミノ酸がMHC IIのβ1ドメインのα3ユニット内で修飾され、別のアミノ酸がMHC IIのα1ドメインのα1ユニット内で修飾される、
請求項17に記載のポリペプチドを含んでなる医薬組成物。 - 例えば、抗CD28抗体または抗41BB抗体などの共刺激分子および/またはこれらの共刺激分子の時系列配列の1つまたは組み合わせをさらに含んでなる、請求項17〜18に記載の医薬組成物。
