ハラミンビニルモノマー(HA)の合成。N-ハラミン前駆体ビニルモノマーであるヒダントインアクリルアミド(HA)[N-(2-メチル-1-(4-メチル-2,5-ジオキソイミダゾリジン-4-イル)プロパン-2-イル)アクリルアミド]を、Bucherer-Bergs反応(Kocer et al., ''N-Halamine Copolymers for Use in Antimicrobial Paints,'' ACS Applied Materials & Interfaces 3(8):3189-3194 (2011)、これはその全体が参照により本明細書に組み入れられる)に基づき以前に報告された方法に従って合成した。簡単に言うと、水/エタノール(体積比1:1)溶媒混合物中の1:2:6モル比のN-(1,1-ジメチル-3-オキソブチル)アクリルアミド(TCI, Japan)、シアン化カリウム、および炭酸アンモニウムを、丸底フラスコ中、室温で4日間撹拌した。エタノールの蒸発後、粗生成物を希HClに曝露し、続いてろ過することにより単離した。生成物を、178℃の融点を有する白色粉末として、収率89%で得た。その構造をNMR、FT-IR、および質量分析により確認した。
フリーラジカル重合法を用いて、N-ハラミン-ドーパミン共重合体を調製した。重合のために、100%MeOH(20mL)を溶媒として用い、HA(7mmol)およびDMA(3mmol)をモノマーとして用い、かつAIBN[2,2'-アゾビス(2-メチルプロピオニトリル)](40mg)を開始剤として用いた。反応混合物に窒素ガスを20分間通気して、酸素を除去した。重合を60℃で3時間行った。室温まで冷却した後、白色の粘稠重合体がフラスコの底に沈澱し、これを回収した。重合体を熱MeOHを用いてさらに洗浄した。湿性重合体生成物を、次いで、80%EtOHに100mg/mLで溶解して保存溶液とし、冷蔵庫に貯蔵した。重合体を1H NMR、FT-IR(フーリエ変換赤外分光法、Bruker Vertex V80V vacuum FTIR system)、およびGPC(ゲル浸透クロマトグラフィ、Waters Ambient Temperature GPC:DMF)で特徴づけた。図1は、本出願のN-ハラミン-ドーパミン共重合体の合成の図を示す。
浸漬コーティング法を用いて、N-ハラミン-ドーパミン共重合体組成物を様々な基体上にコーティングし、塩素化を用いてN-ハラミン部分を活性化した(図2)。N-ハラミン-ドーパミン重合体(poly1、HA-co-DMA、HA:DMA=7:3)の作業溶液を、溶媒として80%EtOHを用いて、保存溶液から2mg/mLで調製した。コーティング接着を試験するために、316Lステンレス鋼、プラスチック(PP、PVC、HDPE)、およびガラスを含む、5つの異なる材料基体を切断して1平方インチの試験片とし、DI水で洗浄した。次に、試験片をコーティング溶液に3時間浸漬した。その後、試験片を80%EtOHで短時間洗浄して、任意の非結合または緩く接着した重合体分子を除去し、ドラフト内で少なくとも2時間乾燥した。poly1コーティングした試験片を10%漂白剤溶液(8.25%次亜塩素酸塩、HClでpHを7.0に調節、利用可能な塩素含有量は約4000ppm)で30分間処理することにより塩素化し、DI水で十分に洗浄して表面上の任意の遊離塩素を除去し、ドラフト内で終夜乾燥した。コーティングを接触角ゴニオメーター(Rame-Hart)およびXPS(X線光電子分光法)で特徴づけた。
N-ハラミン-ドーパミン重合体コーティングした試験片の表面上の固定酸化塩素含有量(N-ハラミン)を、ヨウ素/チオスルフェート滴定法(Qiao et al., ''N-Halamine Modified Thermoplastic Polyurethane with Rechargeable Antimicrobial Function for Food Contact Surface,'' RSC Advances 7(3):1233-1240 (2017)、これはその全体が参照により本明細書に組み入れられる)を用いて判定した。簡単に言うと、2つの試験片(1 in2)をヨウ化カリウム(KI)、水、およびHClを含むフラスコ中に入れ、試験片を室温で10分間撹拌した。次に、得られた溶液に0.5%デンプン溶液を加え、チオ硫酸ナトリウムで滴定した。酸化塩素含有量を、以下の式を用いて算出した:

N-ハラミン-ドーパミンコーティングしたステンレス鋼表面の殺生物効果を、以前に記載された「サンドイッチ」接触致死試験法を用いて判定した(Qiao et al., ''N-Halamine Modified Thermoplastic Polyurethane with Rechargeable Antimicrobial Function for Food Contact Surface,'' RSC Advances 7(3):1233-1240 (2017)、これはその全体が参照により本明細書に組み入れられる)。グラム陰性菌の大腸菌O157:H7およびグラム陽性菌の黄色ブドウ球菌をこの試験で用いた。各細菌の単一のコロニーを15mLの脳心臓浸出物(BHI)ブロス中に移し、37℃で16時間インキュベートした。培養物を遠心分離によってペレット化し、Butterfieldのリン酸緩衝液(BPB)で2回洗浄し、最後にBPB緩衝液に再懸濁した。細菌集団密度をO.D.640nmにより推定し、接種菌を調製した。25μL量の接種菌(約4×107 CFU/mLの細菌)を正方形の試験片の中心に加え、第2の同じ試験片を試料上に置き、サンドイッチを滅菌した重しで圧迫して、確実に接種した菌と完全に接触させる。接触時間10、30、および60分で、試験片を5mLのNa2S2O3溶液(0.05N)中に移し、激しくボルテックス処理して、任意の酸化塩素残渣を失活させ、生存菌を試料から分離した。10倍連続希釈を全ての試料で行い、各希釈液をトリプチケースソイ寒天プレートに加えた。プレートを37℃で48時間インキュベートし、細菌コロニーを数え、殺生物効果試験のために記録した。接種菌の細菌数(CFU/試料)および各接触時間の細菌数を以下に示すとおりに算出した:細菌数(log10CFU/試料)=log10[(寒天プレート上の細菌数×希釈因子×5mL)/(0.025mL)]。
有機負荷下でのコーティング表面の包括的抗菌制御効果を調査するために、細菌生息防止試験を以前に報告された方法に従い、いくつかの変更を加えて実施した(Bastarrachea et al., ''Antimicrobial Food Equipment Coatings: Applications and Challenges,'' Annual Review of Food Science and Technology 6(1):97-118 (2015)、これはその全体が参照により本明細書に組み入れられる)。簡単に言うと、黄色ブドウ球菌の終夜培養物を調製して、LB(Luria-Bertani)培地中の最終細胞密度を106 CFU/mLとした。次に、三つ組試料(1.5×0.5cm2)を細菌溶液に室温で3時間浸漬した。次いで、試料を取り出し、PBS緩衝液で洗浄し、細菌LIVE/DEADキットで染色した。試料を蛍光顕微鏡法(EVOS(商標) FL Cell Imaging System、Thermo Scientific)により観察して、細菌生存度を評価した。Live/dead画像法のための波長は、生細胞に対しては緑色蛍光タンパク質(470/22nm)および死細胞に対しては赤色蛍光タンパク質(531/40nm)である。表面上に接着して生存した細菌だけが、表面上で緑色蛍光(1〜5μmの間の直径を有する明るい点)を発光する。未コーティングのステンレス鋼(SS-Ctrl)、N-ハラミン前駆重合体コーティング(SS-Poly1、非塩素化)およびN-ハラミン重合体コーティング(SS-Poly1-Cl、塩素化)ステンレス鋼の表面上に残っている生菌の密度を比較した。
コーティング厚さをさらに増加させるために、以前に記載されたPEIを架橋剤として用いた(Lee et al., ''Mussel-Inspired Surface Chemistry for Multifunctional Coatings,'' Science 318(5849):426-430 (2007)、これはその全体が参照により本明細書に組み入れられる)。簡単に言うと、2mg/mLの重合体溶液を80%EtOHおよび20%トリス-HCl緩衝液(pH8.5)で調製し、溶液の最終pHは約pH8.0であった。PEI(100mg/mL)溶液をDI水中で調製した。重合体溶液およびPEI溶液をPoly1:PEI=1:5のモル比で混合した。次いで、浸漬コーティング手順のために溶液を用いた:試料を溶液中に10秒間浸漬し、取り出し、ドラフト内で1時間乾燥/硬化させた。その後、試料を80%EtOHで十分に洗浄して、任意の緩く接着した分子を除去した。次いで、試料を前述と同じ塩素化条件下で塩素化した。試料を水接触角、FT-IR、およびXPSにより特徴づけた。加えて、スプレーコーティング法を、PEIを含む、および含まないpoly1溶液に対し、ミスト噴霧器ボトルを用いて実施した(試料を噴霧中は垂直に向けた)。コーティングした試料を乾燥/硬化し、清浄化し、浸漬コーティングと同じ方法で塩素化した。これら全ての浸漬コーティングおよびスプレーコーティングした試料を塩素含有量について滴定した。
Poly1+PEIの最適化された配合を、スプレーコーティングによる実際の食品機器部品のコーティングに用いた。ステンレス鋼(304)パイプを食品加工工場から直接取り、石鹸および水で十分に清浄化した。PEI配合poly1溶液(5mg/mL)を、ミスト噴霧器を用いて外表面に噴霧し、これは5分以内に完全に乾燥した。空気中でさらに1時間硬化した後、コーティングしたパイプ表面およびコーティングしていないパイプ表面の両方に0.5%漂白剤を噴霧し、乾燥させた。塩素化工程を3回繰り返して、十分な塩素化を確実にした。その後、パイプ全体をDI水で十分に洗浄して、表面上のいかなる遊離塩素残渣も除去した。N-ハラミンコーティングしたパイプ上の酸化塩素を可視化するために、ヨウ素滴定のメカニズムに基づく単純な方法を用いた。水、酸、ヨードカリウム、およびデンプン溶液を含む指示薬溶液をインサイチューで調製し、この溶液の一滴を表面に滴下した。十分な酸化塩素が表面に固定されていれば、溶液は5秒以内に無色から青色に変化した。固定された酸化塩素が表面になければ、溶液は少なくとも1分間無色のままである。綿棒を指示薬溶液に浸し、表面を簡単に拭き取るスワビング法も開発した。表面がN-ハラミンコーティングでうまくコーティングされれば、綿は5秒以内に無色から青色に変化した。しかし、コーティングがないか、またはN-ハラミンが活性化されなければ、綿は少なくとも1分間無色のままであった。
N-ハラミン-ドーパミン共重合体p(HA-co-DMA)を、NMRおよびFT-IRによって特徴付けた。NMRスペクトル(図3A)において、HA(δ10.7ppm、N-H)およびDMA(δ6.25〜6.75ppm、カテコール)両方の特徴的なピークが重合体鎖で観察されたが、各モノマーの二重結合(δ5.5〜6.0ppm)は消失し、N-ハラミンおよびドーパミン基を含む純粋な重合体が得られたことが確認された。重合体鎖におけるHAおよびDMA比を、NMRスペクトルのピークを積分することにより7:3と算出し、これはHAおよびDMAモノマーの設計した供給比(7:3)と一致していた。以前の報告によると、20〜30%のDMA含有量は、典型的には、接着効果を提供するのに十分であり(Glass et al., ''Enhanced Reversible Adhesion of Dopamine Methacrylamide-Coated Elastomer Microfibrillar Structures Under Wet Conditions,'' Langmuir 25(12):6607-6612 (2009)、これはその全体が参照により本明細書に組み入れられる)、したがって得られたp(HA-co-DMA)は、分子組成に基づいて十分な接着性を有すると予想された。加えて、GPCの結果は、重合体が分子量Mn=11,957およびMw=18,225を有し、PDI(多分散性)=1.52であることを示した。FT-IRスペクトル(図3B)は、ヒダントイン(1767および1718cm-1)およびドーパミン基(1519cm-1)の特徴的なピークを示した。まとめると、これらのデータは、N-ハラミン-ドーパミン官能基化重合体がうまく合成されたことを示した。最終重合体生成物は、工業用途のために、乾燥が速く、適用が容易で、安全なコーティング溶液を配合するのに便利な溶媒である70〜87%EtOH/H2O溶液に可溶性であった。
Claims (30)
- N-ハラミン(HA)およびドーパミン(DMA)の共重合体であって、HAおよびDMAは共重合体中に0.4:9.6〜9.6:0.4(HA:DMA)の比で存在する、共重合体。
- HA:DMAの比が約7:3である、請求項1記載の共重合体。
- モノマー(A)、(B)、(C)、および(D)を含む、請求項1記載の共重合体:

- 液体、粉末、またはエアロゾル形態である、請求項14記載の抗菌コーティング。
- コーティングの共重合体が金属、プラスチック、ガラス、または塗料に共有結合している、請求項14記載の抗菌コーティング。
- コーティングの共重合体が互いに架橋している、請求項14記載の抗菌コーティング。
- 共重合体がポリエーテルイミド架橋剤によって互いに架橋している、請求項17記載の抗菌コーティング。
- 請求項1〜12のいずれか一項記載の共重合体、請求項13記載の組成物、または請求項14〜18のいずれか一項記載の抗菌コーティングでコーティングされている、装置またはシステム。
- 食品貯蔵装置、食品加工装置、または生物医学装置である、請求項19記載の装置またはシステム。
- 抗菌コーティングにより少なくとも10nmの厚さでコーティングされている、請求項19記載の装置またはシステム。
- 請求項1〜12のいずれか一項記載の共重合体を調製するための方法であって、
ラジカル重合可能なN-ハラミン系モノマーを提供する段階;
ラジカル重合可能なドーパミン系モノマーを提供する段階;
N-ハラミン系モノマーおよびドーパミン系モノマーを適切な溶媒に溶解して、N-ハラミン系モノマーおよびドーパミン系モノマーの溶液を生成する段階;ならびに
該溶液を、フリーラジカル開始剤存在下でラジカル重合反応にかけて、N-ハラミン系モノマーおよびドーパミン系モノマーの共重合体を生成する段階
を含む、前記方法。 - 溶媒が、メタノール、エタノール、塩化メチレン、トルエン、ジオキサン、THF、クロロホルム、シクロヘキサン、ジメチルスルホキシド、ジメチルホルムアミド、アセトン、アセトニトリル、n-ブタノール、n-ペンタノール、クロロベンゼン、ジエチルエーテル、tertブタノール、1,2-ジクロロエチレン、ジイソプロピルエーテル、エタノール、酢酸エチル、エチルメチルケトン、ヘプタン、ヘキサン、イソプロピルアルコール、イソアミルアルコール、メタノール、ペンタン、n-プロピルアルコール、ペンタクロロエタン、1,1,2,2,-テトラクロロエタン、1,1,1,-トリクロロエタン、テトラクロロエチレン、テトラクロロメタン、トリクロロエチレン、水、キシレン、ベンゼン、ニトロメタン、グリセロール、およびその混合物からなる群より選択される、請求項22記載の方法。
- フリーラジカル開始剤が、2,2'-アゾビス(2-メチルプロピオニトリル)、過酸化ベンゾイル、1,1'-アゾビス(シクロヘキサンカルボニトリル)、t-ブチルペルオキシド、ジクミルペルオキシド、過硫酸カリウム、アラルキルハライド、アリールハライド 2,2-ジメトキシ-1,2-ジフェニル-エタン-1-オン、および(2,4,6-トリメチルベンゾイル)-ジフェニルホスフィンオキシドからなる群より選択される、請求項22記載の方法。
- ラジカル重合反応が40℃〜120℃の範囲の温度で起こる、請求項22記載の方法。
- 抗菌コーティングした材料を生成する方法であって、
請求項1〜12のいずれか一項記載の1つまたは複数の共重合体を溶媒に溶解して、共重合体溶液を生成する段階;
共重合体溶液の共重合体が材料に結合するのに十分な期間、重合体溶液中に材料を浸漬し、それによりコーティングした材料を生成する段階;
コーティングした材料を共重合体溶液から取り出す段階;および
コーティングした材料をハロゲンを含む第2の溶液で処理する段階
を含む、前記方法。 - 第2の溶液のハロゲンが塩素、臭素またはヨウ素から選択される、請求項26記載の方法。
- 塩素を含む溶液が漂白剤溶液、次亜塩素酸カルシウム溶液、N-クロロスクシンイミド溶液、ジクロロイソシアヌル酸ナトリウム溶液、トリクロロイソシアヌル酸溶液、次亜塩素酸tert-ブチル溶液、N-クロロアセトアミド溶液、またはN-クロラミンの溶液である、請求項26記載の方法。
- 前記浸漬の前に、共重合体溶液に架橋剤を添加する段階をさらに含む、請求項26記載の方法。
- 共重合体溶液中の共重合体に対する架橋剤の比が、1:10〜10:1の範囲である、請求項28記載の方法。
