標的の発現、精製およびビオチン化
単一ドメイン(FN−B、Uniprot ID P02751、アイソフォーム7、アミノ酸E1265−T1355)としての、またはその周囲のIII型ドメイン(FN−67B89、アミノ酸G1080−E1455)に隣接する、組換えヒトフィブロネクチンEDBは、本試験における標的タンパク質としての役割を果たし、EDBを含まないドメイン6〜9(FN−6789)を対照として使用した。すべての変異体を、c末端ヘキサヒスチジン(H6)タグを付けて大腸菌で発現させ、固定化金属イオンアフィニティクロマトグラフィ(IMAC)およびサイズ排除クロマトグラフィ(SEC)によって精製した。このために、コドン最適化したDNA配列がThermo Fisher Scientificによって合成され、これをpET−21a発現ベクター(Novagen)にクローニングし、大腸菌BL21(DE3)細胞(Agilent)に導入した。タンパク質を、OD600が約0.7に達するまで、30℃、120rpmで、750mLスケールで発現させた。タンパク質産生を誘導するために、750μLの1M IPTGを主培養物に添加し、25℃、120rpmで一晩インキュベートした。細胞を回収し、10mLの平衡化緩衝液(20mM Tris−HCl pH8.0、10%グリセロール、500mM NaCl、10mMイミダゾール)に再懸濁し、ソニフィケーション(Branson Digital Sonifier 250)によって溶解した。上清を、AKTAprime(商標)plus system(GE Healthcare)および20分で10〜500mMイミダゾールの線形勾配を使用して、1mL HisTrapカラム(GE Healthcare)でのIMACによって精製した。続いて、タンパク質をPBS(14mM NaCl、2.7mM KCl、10mM Na2HPO4、1.8mM KH2PO4、pH 7.5)に対して4℃で一晩透析し、HiLoad 26/600 Superdex 200pg(FN−67B89およびFN−6789の場合)または75pg(FN−Bの場合)カラム(GE Healthcare)を使用したサイズ排除クロマトグラフィによってさらに精製した。最終的に精製されたタンパク質を、SDS−PAGE、分析SEC、およびEDB特異的BC−1抗体(ab154210、Abcam)を使用したELISAによって分析した。タンパク質を、−20℃で5%マンニトールおよび5%トレハロースを添加したPBS中でアリコートとして保存した。
発現され、精製されたFN−67B89タンパク質の品質を決定するために、96ウェル形式でタンパク質間相互作用を分析する手法としてELISAを使用した。最初の段階では、50mM Na2CO3、pH 9.4の被覆緩衝液を使用して、アルカリ性条件下での受動吸収によって、10μg/mLの標的タンパク質を100μLの容量でMaxisorb(商標)96ウェルプレートに被覆した。ELISAプレートの被覆を4℃で一晩実施した。被覆緩衝液を除去した後、プレートを300μLのPBS−Tで3回洗浄し、300μLのブロッキング緩衝液(PBS中3%BSA)を添加した。ブロッキングを室温で2時間実施した。一次インキュベーションのために、プレートを300μLのPBS−Tで3回洗浄し、PBS−Tで1:1000に希釈した100μLのBC−1抗体と共に4℃で1時間インキュベートした。PBSで1:5000に希釈したHRP結合抗マウス抗体(554002、BD Pharmingen(商標))を、100μL/ウェルのPBS−Tで3回洗浄した後に添加し、4℃で1時間インキュベートした。次に、プレートをPBS−Tで3回、PBSで3回洗浄した後、HRPを介したTMB基質の変換によって抗原−抗体複合体を検出した。100μLのTMB基質をウェルに添加し、試料中に存在する分析物の量に比例して青色を発現させた。50μLの0.2M HClを添加することによって発色を停止させ、450nmで比吸光度を測定した。
EDB特異的シスチンノットミニタンパク質を、モモルディカ・コキネンシス(oMCoTI−II)からのトリプシン阻害剤IIの開鎖変異体に基づく2つの異なるコンビナトリアルライブラリを使用したファージディスプレイによって選択した。どちらのライブラリもM13ファージミド系に基づくが、適用されるランダム化スキームが異なる。MCopt1.0ライブラリでは、ループ1(6、9および12アミノ酸の長さの相違を含む)および3、ならびにN末端の2つのアミノ酸がランダム化されているが、MCopt2.0ライブラリは、10アミノ酸のみを有するループ1のランダム化によって構築された。さらに、MCopt1.0はメジャーコートタンパク質pVIIIによって提示され、MCopt2.0はpIIIによって提示されるため、ディスプレイの種類においても異なる。
同定工程では、PCRによってシスチンノットミニタンパク質配列を増幅するために、3回目のスクリーニングラウンドに由来する濃縮スクリーニングプールファージミドを調製した。チオレドキシンA融合変異体としてのシスチンノットミニタンパク質の発現を可能にするために、PCRインサートをpET−32a(Novagen)の誘導体である発現ベクターpET−32−LibExにクローニングした。このベクターは、細胞質での効率的なジスルフィド結合形成を可能にする大腸菌チオレドキシンA、迅速な精製のためのH6タグ、抗体による検出のためのsタグ、および融合タグを除去するためのトロンビン切断部位をコードするDNA配列を連続的な順序で担持する。シスチンノットミニタンパク質配列をコードするDNA断片を、トロンビン切断部位の下流で、固有のBam HIおよびKpn I制限部位を介してpET−32−LibExベクターに導入した。ベクターを熱ショックによってE.coli SHuffle(登録商標)T7 Express細胞(New England BioLabs)に導入し、細胞を選択寒天プレートに播種した。単一コロニーを採取し、配列決定し、1mLの自動誘導培地(MagicMedia(商標)、Thermo Fisher Scientific)での小規模発現のために96ウェルプレートに移した。タンパク質生産を30℃、220rpmで一晩行った。細胞を3000xgで15分間の遠心分離によって回収し、凍結融解サイクルと組み合わせて0.1mg/mLリゾチーム(Merck Millipore)および5U/mLベンゾナーゼ(Merck Millipore)を含む緩衝液(20mM Tris、2mM MgCl2、20mM NaCl、pH8)中でインキュベートして溶解し、80℃で10分間加熱した。細胞破片を除去するための最後の遠心分離(3000xg、15分、4℃)の後、E−PAGE(商標)および結合分析のために上清を収集した。
組換えタンパク質の生産を、それぞれのシスチンノットミニタンパク質配列をコードするpET−32−LibExベクターを担持するE.coli SHuffle(登録商標)T7 Express細胞を使用して、30℃、120rpmにて750mLスケールで実施した。培養物が約0.7のOD600に達した後、750μLの1M IPTGを添加し、25℃、120rpmで一晩インキュベートすることによって産生の誘導を達成した。大腸菌細胞を回収し、10mLの平衡化緩衝液に再懸濁し、ソニフィケーションによって溶解し、10分間80℃に加熱した。細胞破片を遠心分離(15.000×g、4℃で30分間)した後、AKTAprime(商標)plusシステムおよび20分で10〜500mMイミダゾールの線形勾配を使用して、1mL HisTrapカラムでのIMACによって上清を精製した。シスチンノットミニタンパク質融合タンパク質含有画分を収集し、トロンビン切断緩衝液(20mM Tris、150mM NaCl、1.5mM CaCl2および5%(w/v)グリセロール、pH8.45)に対して4℃で一晩透析した。
EDB結合に寄与するMC−FN−010内の残基を同定するために、アラニンスキャニング変異誘発を実施した。この方法は、定義された配列位置のアラニンに対するアミノ酸の体系的な置換と、生成された変異体のその後の結合分析を含む。アラニンスキャニングを生成するために、MC−FN−010誘導体の変異をPCRによって導入するか、またはGeneArt(商標)Strings(商標)断片(Thermo Fisher Scientific)の直接合成によってコード配列全体を構築した。それぞれのDNA断片を、固有のBamHIおよびKpnI制限部位を使用してpET−32−LibEx発現ベクターにクローニングし、E.coli SHuffle(登録商標)T7 Expressコンピテント細胞(New England BioLabs)に導入した。すべての変異をDNA塩基配列決定によって検証した。アラニンスキャニング変異誘発変異体を、5mLの選択的自己誘導培地を使用して24ウェル形式で発現させた。生産および融合タンパク質の精製は、96ウェル形式について上述したように実施したが、HisPur(商標)Ni−NTAスピンカラム(Thermo Fisher Scientific)を使用した上清のさらなる精製段階を含んだ。シスチンノット融合タンパク質の標的タンパク質およびオフターゲットタンパク質への結合能力および特異性は、上記のように抗体ベースのELISAアッセイを用いて実施した。
単量体および三量体のシスチンノットミニタンパク質リガンドの標的タンパク質への結合速度論を、PBS−Tをランニング緩衝液として用いるBiacore T−100装置(GE Healthcare Life Science)を使用して決定した。このために、ビオチン化FN−67B89タンパク質(200〜300μg/mL)を、SAセンサチップ(GE Healthcare Life Science)のフローセルに結合することによって捕捉した。単量体リガンドを分析するために、最大750応答単位(RU)の固定化標的密度を適用し、三量体変異体については、最大400のRUを目指した。50〜4000nMの範囲の濃度でマルチサイクル速度論的方法を使用して、単量体リガンドの結合分析を実施した。サイクルを90秒の結合期間で開始し、続いて420秒の解離期間と最後の再生段階を実施した。20μL/分の流量を適用して速度論的測定を実施した。三量体変異体は、同じ結合および解離条件下で分析したが、30μL/mLの一定流量で単一サイクルの速度論的測定モードを使用した。この場合、分析物の濃度は1.25nM〜10nMであった。結合速度論および定常状態分析は、グローバル速度論適合モデル(1:1ラングミュア、Biacore T−100 Evaluation Software、GE Healthcare Life Science)を使用して計算した。
免疫蛍光染色のために、凍結保存された腫瘍または脳の小片を5ミクロンの厚さの切片に切断し、氷冷アセトンで5分間固定し、風乾した。次に、3%BSAを含むPBS中で室温にて5分間、スライドをブロックした。EDBを染色するために、1μgのそれぞれのビオチン化シスチンノットミニタンパク質を、2.9μgのストレプトアビジン−Cy3複合体(Rockland Immunochemicals)と共に室温で30分間インキュベートした。次に、あらかじめ形成した複合体を腫瘍切片に添加し、37℃で30分間インキュベートした。その後、1%BSAを含むPBSでスライドを3回洗浄した。CD31染色を、1%BSAを含むPBSで1:100に希釈したラット抗マウスCD31 IgG抗体(クローン390、eBioscience)を使用して37℃で30分間実施した。PBSで3回洗浄した後、細胞核を、PBSで1:5000に希釈したHochst 33342(Thermo Fisher Scientific)を用いて室温で30分間染色した。スライドを上記のように再度洗浄し、封入剤(Dako)の薄層中で、カバーガラスで覆った。Zeiss Apotome顕微鏡(Carl Zeiss)で画像を捕捉し、ZENソフトウェア(Carl Zeiss)で分析した。
三量体Alexa Fluor 680(AF680)結合リガンドおよびN末端ビオチン化ミニタンパク質をPepscanから購入した。得られたすべてのペプチド構築物は、100μgのアリコートとして−20℃で保存した。実験のために、すべてのペプチドを100μLのDPBS(Gibco)に溶解し、1μg/μLの濃度にした。すべての構築物について、ESI質量分析によって同一性を検証し、分析逆相クロマトグラフィ(Pepscan)によって純度を分析した。さらに、標的結合特性(FN−67B89への結合)および特異性(FN−6789への結合)を特徴付けるために、三量体をSDS−PAGEおよびSPRによって分析した。
ヒト神経膠芽腫U−87 MG(ATCC)細胞株を、無菌条件下に37℃、5%CO2および95%湿度で、10%FCSを添加したEMEM培地(ATCC)で培養した。
三量体構築物の生体内分布および腫瘍標的化の分析のために、所望の腫瘍サイズを担持するマウスを含めた。すべての三量体構築物を、最終容量100μlのPBS緩衝液(3.34nmol/マウス)中で眼球後静脈叢を介して静脈内注射した。マウス(各構築物についてn=3)を、615〜665nmの励起範囲を使用し、695〜770nmで発光シグナルをモニタリングして、IVIS Spectrum System(Perkin Elmer)で画像化した。画像化工程を注射の1時間後、2時間後または6時間後に実施し、安楽死させた後、腫瘍と特定の器官を切除し、画像化し、秤量し、さらなる分析のために凍結保存した。Living Image(登録商標)ソフトウェア(PerkinElmer)を使用して、目的の領域の蛍光強度を定量化した。GraphPad Prismで二元配置分散分析を使用した三重に反復したデータセットに基づいて統計的有意性を計算した。
シスチンノットミニタンパク質は腫瘍イメージングの薬剤として理想的に適することが示されているため(Kimura et al.2009;Moore et al.2013;Miao et al.2009;Soroceanu et al.1998;Veiseh et al.2007,Nielsen et al.2010;Hackel et al.2013;Zhu et al.2014)、標的結合リガンドを選択するためのコンビナトリアルファージライブラリ構築の基礎として、モモルディカ・コキネンシストリプシン阻害剤−II(oMCoTI−II)の開鎖配列を使用した(Hernandez et al.2000)。第1のライブラリ(MCopt 1.0)は、最初のループにランダム化されたアミノ酸、3番目のループに散在する位置、および最初のシステインの前に2つの可変残基を有する配列を含む。シスチンノットミニタンパク質配列を、M13ファージのメジャーコートタンパク質(pVIII)に遺伝的に融合した。さらに、第2のライブラリ(MCopt 2.0)は、配列の最初のループでのランダム化、およびM13ファージのマイナーコートタンパク質(pIII)によるタンパク質の提示を用いて開発した。したがって、ライブラリは、ランダム化されたループ位置と配列の長さ、およびタンパク質提示価で区別され、これはリガンド選択の結果の変動性につながり得る。卵巣または滑膜細胞の間質を除くほとんどの正常組織には存在しないが、様々な腫瘍実体で高度に発現されることが公知である(Carnemolla et al.1989;Castellani et al.1994)フィブロネクチンエクストラドメインB(EDB)に対するシスチンノットミニタンパク質を同定するために、両方のライブラリを並行して適用した。
次に、社内で生産したEDB標的タンパク質(FN−BおよびFN−67B89)、ならびにオフターゲットタンパク質(FN−6789)および様々な対照タンパク質(粉乳、ストレプトアビジンおよびウシ血清アルブミン)を使用して、残りの6つのシスチンノットミニタンパク質の標的結合特異性に焦点を合わせた。エクストラドメインBを欠くフィブロネクチンは多くの異なる細胞型によって発現されるため、FN−6789は完全に対応するオフターゲットタンパク質である(Mao und Schwarzbauer 2005)。すべての候補物は、組換えFN−BおよびFN−67B89標的タンパク質に対して同等に高い妥当なEDB標的結合を示す(図4)。共通のR−I/V−R−(L)モチーフに基づいて選択した変異体(MCopt 1.0−2/−3およびMCopt 2.0−1/−2/−3)は、中程度から低いオフターゲットおよび対照タンパク質シグナルを示した。MCopt 1.0−2およびMCopt 1.0−3はMCopt 1.0ヒット同定で既に同定されたが、MCopt 2.0−1、−2および−3は、それらの共通のモチーフに基づいてMCopt 2.0プールからのみ同定され、したがって以前のFN−B標的結合では検定されなかった。しかしながら、R−I/V−R−(L)モチーフを含まないMCopt 1.0−1は、オフターゲットFN−6789および粉乳を除くすべての対照タンパク質への高い相互作用が観察されたため、特異性評価ができなかった。これらのデータは、観察されたアミノ酸モチーフがEDB結合に関連することを強く示す。
異なる選択されたシスチンノットミニタンパク質におけるアミノ酸モチーフR−I/V−R−(L)についての以前の所見は、既にEDB結合への高い配列寄与を示唆したが、その後、その関連性を実験的により詳細に評価した。MC−FN−010標的配列における単一アラニン置換を取り上げ、合計で14の誘導体構築物を得た(図7Aに示すように、MC−FN−011からMC−FN−0114まで連続的に番号を付している)。すべての構築物を単一ドメインFN−Bに対して試験した。予想されたように、配列の先頭にアラニン交換を有する7つの構築物は、依然として強い標的相互作用を示し、これらの位置がEDB結合にとって重要ではないことを示唆した。対照的に、共通のモチーフ位置に交換を有する4つの構築物は、結合の喪失を明らかにした。5番目のループでのさらなるアラニン置換は、標的相互作用の減少をもたらし、その関連性も示した(図7B)。これらの結果は、図7Cに要約されているように、最初のループ(RIRL)の4つのアミノ酸残基および5番目のループのアルギニン残基が、FN−67B89への結合相互作用に直接影響するか、またはミニタンパク質の立体配座に間接的な影響を及ぼすことを確認する。
さらに、微小環境に天然のEDBタンパク質を含むU−87 MG腫瘍異種移植片切片を使用して、細胞状況内での親MC−FN−010の特異性を検討した。ヒト神経膠芽腫腫瘍は、血管構造にフィブロネクチンEDBアイソフォームを保有することが公知である(Mariani et al.1997)。アラニンスキャニング変異誘発に基づいて、3つの位置(PMCTQRANRIAACRRDSDCTGACICRGNGYCG)にアラニン置換を有する陰性対照構築物(MC−FN−0115)を生成した。免疫蛍光イメージング実験のために、ビオチン化形式としてのMC−FN−010およびMC−FN−0115をCy3標識ストレプトアビジンで四量体化した。四量体化されたMC−FN−010−bioは、血管マーカと見なされる、内皮細胞の遍在的に発現される表面タンパク質、CD31に対するAlexa Fluor 647結合抗体で確認されたように、血管周囲の領域をほぼ単独で装飾した(図8)。Cy3とAlexa Fluor 647の重ね合わせ画像は、血管に関連する両方の蛍光シグナルの共局在を示す。さらに、周りの血管周囲領域における四量体化MC−FN−010の局在を検出することができた。対照的に、陰性対照構築物MC−FN−0115で染色されたU−87 MG腫瘍切片は、蛍光シグナルを全く示さなかった(図8)。四量体化されたMC−FN−010およびMC−FN−0115の蛍光シグナルは、正常なマウスの脳切片では観察されなかった(図12)。
標的タンパク質を診断ツールとして使用する前に、それらを造影剤と特異的に結合させる必要がある(Spicer und Davis 2014)。リード候補物MC−FN−010は、ループ1にリジンを含むが、これは第一級アミンへの選択的薬剤結合のためには好ましくない。以前の分析に基づいて、このアミノ酸はEDB結合に積極的に寄与しないため、誘導体構築物MC−FN−016を2番目の候補物として選択する。Trx−MC−FN−016のFN−67B89への結合が、親Trx−MC−FN−010と同等のシグナルで用量依存的に観察された(図9)。対照的に、FN−6789への全体的なバックグラウンドシグナルは、継続的に比較的低かった。両方のシスチンノットミニタンパク質候補物は、FN−6789との相互作用を伴わずにFN−67B89を排他的に標的とし、フィブロネクチンは複数の細胞型で広く発現されるため、これは重要である(Pankov und Yamada 2002)。ビオチン化FN−67B89に対するタグなしのMC−FN−010およびMC−FN−016の親和性をSPR分析によって検定した。両方のシスチンノットミニタンパク質の結合速度論は、速いオフ速度で1桁のマイクロモル範囲の低い結合親和性を明らかにした(図10A)。
より強い結合強度を達成するために、リガンドを、潜在的なアビディティ効果を利用するオキシムライゲーションによって化学的に三量体化した。さらに、マウスへの投与後の分布と局在の観察を可能にするために、分子を近赤外蛍光色素であるAlexa Fluor 680でタグ付けした。3つの三量体構築物すべてを化学合成した後、正しいサイズと純度を管理するために様々なアッセイを実施した。SDS−PAGEおよび逆相クロマトグラフィ分析は、すべての構築物で重要な著明点を示さなかった(データは示していない)。特に、オリゴマー化戦略は、両方のEDB結合シスチンノットミニタンパク質(AF680−(MC−FN−010)3およびAF680−(MC−FN−016)3)の親和性を大幅に改善し、3桁のピコモル親和定数および単量体変異体と比較して著しく緩やかなオフ速度をもたらした(図10B)。
凍結保存された腫瘍または脳の小片を6ミクロンの厚さの切片に切断し、氷冷アセトンで5分間固定し、風乾した。次に、3%BSAを含むPBS中で室温にて5分間、スライドをブロックした。次いで、EDBの染色のために、0.1μgのAF680−(MC−FN−010)3および1%BSAを含むPBSで1:100に希釈した抗マウスCD31抗体(RB−10333−P1、Thermo Fisher)を腫瘍切片に添加し、37℃で30分間インキュベートした。その後、1%BSAを含むPBSでスライドを3回洗浄した。一次抗マウスCD31抗体のCD31染色を、37℃で30分間、1%BSAを含むPBSで1:400に希釈した二次抗ウサギIgG−Cy3抗体(111−165−003、Jackson ImmunoResearch)で検出した。PBSで3回洗浄した後、細胞核を、PBSで1:5000に希釈したHochst 33342(Thermo Fisher Scientific)を用いて室温で30分間染色した。スライドを上記のように再度洗浄し、封入剤(Dako)の薄層中で、カバーガラスで覆った。Zeiss Apotome顕微鏡(Carl Zeiss)で画像を捕捉し、ZENソフトウェア(Carl Zeiss)で分析した。図13は、ヒト神経膠芽腫異種移植腫瘍および正常な脳組織試料での三量体構築物(AF680−(MC−FN−010)3および対照AF680−(MC−FN−0115)3)によるさらなる免疫蛍光染色を示す。AF680−(MC−FN−010)3は、血管マーカCD31で局在化されたU−87 MG切片の腫瘍血管周辺の領域を染色したが、対照AF680−(MC−FN−0115)3は全く染色を示さなかった。正常なマウス脳切片ではAF680−(MC−FN−010)3による染色は観察されず、腫瘍血管系の特異性を示した。

- 前記EDB結合ペプチドまたはEDB結合剤が、固体支持体上に解除可能または解除不能に固定化されている、請求項37に記載のアッセイ装置。
- 試料中のフィブロネクチンエクストラドメインB(EDB)の存在および/または量を検定するための方法であって、請求項1〜22、25、および30〜32のいずれか一項に記載のEDB結合ペプチドまたは請求項23〜32のいずれか一項に記載のEDB結合剤を使用することを含む方法。
- 患者における癌の診断、検出またはモニタリングのための方法であって、請求項1〜22、25、および30〜32のいずれか一項に記載のEDB結合ペプチド、または請求項23〜32のいずれか一項に記載のEDB結合剤を使用して、前記患者におけるフィブロネクチンエクストラドメインB(EDB)の存在および/または量を検定することを含む方法。
- 前記検定が、前記患者から単離された生物学的試料に対して実施される、請求項40に記載の方法。
- EDBの存在、または癌のない参照と比較してより高いEDBの量が、前記患者が癌を有することを示す、請求項40または41に記載の方法。
- EDBの存在および/または量を検定することが、
(i)試料を前記EDB結合ペプチドまたは前記EDB結合剤と接触させること、ならびに
(ii)前記EDB結合ペプチドまたは前記EDB結合剤とEDBとの間の複合体の形成を検出することおよび/または複合体の量を決定すること
を含む、請求項39〜42のいずれか一項に記載の方法。 - 前記EDB結合ペプチドまたはEDB結合剤が、少なくとも1つの検出可能な標識またはレポータを含むか、またはそれに結合されている、請求項39〜43のいずれか一項に記載の方法。
- 前記EDB結合ペプチドまたはEDB結合剤が、固体支持体上に解除可能または解除不能に固定化されている、請求項39〜44のいずれか一項に記載の方法。
- 請求項1〜22、25、および30〜32のいずれか一項に記載のEDB結合ペプチド、請求項23〜32のいずれか一項に記載のEDB結合剤、請求項33に記載の組換え核酸、または請求項34に記載の宿主細胞を含む医薬組成物。
- 治療法に使用するための、特に患者における癌を治療または予防するのに使用するための、請求項1〜22、25、および30〜32のいずれか一項に記載のEDB結合ペプチド、請求項23〜32のいずれか一項に記載のEDB結合剤、請求項33に記載の組換え核酸、請求項34に記載の宿主細胞、または請求項46に記載の医薬組成物。
- 患者における癌を標的化するのに使用するための、請求項1〜22、25、および30〜32のいずれか一項に記載のEDB結合ペプチド、または請求項23〜32のいずれか一項に記載のEDB結合剤。
- 請求項1〜22、25、および30〜32のいずれか一項に記載のEDB結合ペプチド、請求項23〜32のいずれか一項に記載のEDB結合剤、請求項33に記載の組換え核酸、請求項34に記載の宿主細胞、または請求項46に記載の医薬組成物を患者に投与することを含む、前記患者を治療する方法であって、好ましくは、前記患者が癌を有するかまたは癌を発症するリスクがある、方法。
- 前記EDB結合ペプチドまたはEDB結合剤が、少なくとも1つの治療エフェクタ部分を含むかまたはそれに結合されている、請求項46に記載の医薬組成物、請求項47に記載のEDB結合ペプチド、EDB結合剤、組換え核酸、宿主細胞もしくは医薬組成物、請求項48に記載のEDB結合ペプチドもしくはEDB結合剤、または請求項49に記載の方法。
- 前記癌がEDB陽性である、および/またはEDBを発現する細胞が関与する、請求項40〜45のいずれか一項に記載の方法、請求項47もしくは50に記載のEDB結合ペプチド、EDB結合剤、組換え核酸、宿主細胞もしくは医薬組成物、請求項48もしくは50に記載のEDB結合ペプチドもしくはEDB結合剤、または請求項49もしくは50に記載の方法。
