本発明は、眼充血を治療するための組成物およびそれを用いて眼充血を治療するための方法、より具体的には、眼充血を治療するための抗血管新生剤とαアドレナリン受容体アゴニストとの組み合わせを含む組成物と、それによって眼充血を治療するための方法とに関する。

Claims (16)
- 眼充血の治療に使用するための組成物であって、
前記組成物は、抗血管新生剤およびαアドレナリン受容体アゴニストを含む組成物。 - 前記抗血管新生剤は、血管内皮増殖因子受容体(VEGFR)1,2および3、血小板由来成長因子受容体(PDGFR)α、βおよびLynからなる群から選択される1つ以上を阻害するマルチキナーゼ阻害剤(MKI)である請求項1に記載の組成物。
- 前記MKIは、アファチニブ、アムバチニブ、アキシチニブ、カボザンチニブ、カネルチニブ、セディラニブ、セリチニブ、クレノラニブ、クリゾチニブ、ダブラフェニブ、ダコミチニブ、ダサチニブ、エルロチニブ、フォレチニブ、ゲフィチニブ、ゴルバチニブ、イブルチニブ、イコチニブ、イデラリシブ、イマチニブ、ラパチニブ、レンバチニブ、リニファニブ、モテサニブ、ネラチニブ、ニロチニブ、ニンテダニブ、パルボシクリブ、パゾパニブ、ポナチニブ、キザルチニブ、レゴラフェニブ、ルキソリチニブ、ソラフェニブ、スニチニブ、タンドゥチニブ、チバニチブ、チボザニブ、トラメチニブ、バンデタニブ、ヴァタラニブ、およびベムラフェニブから成る群から選択され、そして、前記MKIは、好ましくはニンテダニブまたはアキシチニブである請求項2に記載の組成物。
- 前記抗血管新生剤は、血管内皮増殖因子(VEGF)に結合してVEGFのシグナル伝達を遮断する生物学的薬剤である請求項1に記載の組成物。
- 前記生物学的薬剤は、ベバシズマブ、ラニビズマブ、ラムシルマブ、アフリベルセプト、およびコンバーセプトからなる群から選択される請求項4に記載の組成物。
- 前記αアドレナリン受容体アゴニストは、ナファゾリン、テトラヒドロゾリン、オキシメタゾリン、メトキサミン、フェニレフリン、キシロメタゾリン、オキセドリン、アプラクロニジン、ミバゼロル、クロニジン、ブリモニジン、アルファメチルドーパ、グアンファシン、デキセメディトミジン、(+)−(S)−4−1−(2,3−ジメチル−フェニル)−エチル−1,3−ジヒドロ−イミダゾール−2−チオン、1−(イミダゾリジン−2−イル)イミノリンダゾール、メトキサミン、フェニレフリン、チザニジン、キシラジン、グアナベンズ、およびアミトラズからなる群から選択される請求項1に記載の組成物。
- 前記αアドレナリン受容体アゴニストは、α2−アドレナリン受容体に対して選択的なα2−アドレナリン受容体アゴニストである請求項1に記載の組成物。
- 前記α2−アドレナリン受容体アゴニストは、ブリモニジン、アプラクロニジン、ミバゼロル、クロニジン、アルファメチルドーパ、グアンファシン、デキセメディトミジン、(+)−(S)−4−1−(2,3−ジメチル−フェニル)−エチル−1,3−ジヒドロ−イミダゾール−2−チオン、1−(イミダゾリジン−2−イル)イミノリンダゾール、メトキサミン、フェニレフリン、チザニジン、キシラジン、グアナベンズ、およびアミトラズからなる群から選択され、そして、前記α2−アドレナリン受容体アゴニストは、好ましくはブリモニジンである請求項7に記載の組成物。
- 前記組成物は、0.001%−1%のニンテダニブと0.01%−1%のブリモニジン、好ましくは、0.01%−0.5%のニンテダニブと0.01%−0.5%のブリモニジン、好ましくは0.1%−0.3%のニンテダニブと0.01%−0.05%のブリモニジン、より好ましくは、0.2%のニンテダニブと0.025%のブリモニジン、を含む請求項1に記載の組成物。
- 前記組成物は、および防腐剤の有無にかかわらず、更に、安定剤、界面活性剤、ポリマーベース担体、ゲル化剤、有機共溶媒、pH活性成分、浸透圧活性成分、から選択される1つ以上の薬学的に許容される賦形剤、を含む請求項1〜9のいずれか一項に記載の組成物。
- 前記組成物は、局所眼用製剤、軟膏、ゲル、徐放性半固形製剤、徐放性固形製剤、または眼用インプラントであり、そして、前記局所眼製剤は、好ましくは、水溶液、懸濁液、または乳濁液である請求項1〜10のいずれか一項に記載の組成物。
- 前記徐放性半固体製剤、前記徐放性固体製剤、または前記眼用インプラントは、ポリ乳酸(PLA)、ポリ乳酸−co−グリコール酸(PLGA)、およびポリ乳酸ポリ乳酸−co−グリコール酸コポリマー、からなる群から選択される生分解性ポリマーを含む請求項11に記載の組成物。
- 患者の眼充血を治療するための方法であって、MKIで患者の複数のキナーゼ受容体を阻害すること、および、患者のαアドレナリン受容体を、前記αアドレナリン受容体のアゴニストで活性化すること、を含み、
ここで、前記キナーゼ受容体は、VEGFR1,2および3、ならびにPDGFRα,βおよびLyn、からなる群から選択され、好ましくはVEGFR1,2または3である方法。 - 前記MKIは、アファチニブ、アムバチニブ、アキシチニブ、カボザンチニブ、カネルチニブ、セディラニブ、セリチニブ、クレノラニブ、クリゾチニブ、ダブラフェニブ、ダコミチニブ、ダサチニブ、エルロチニブ、フォレチニブ、ゲフィチニブ、ゴルバチニブ、イブルチニブ、イコチニブ、イデラリシブ、イマチニブ、ラパチニブ、レンバチニブ、リニファニブ、モテサニブ、ネラチニブ、ニロチニブ、ニンテダニブ、パルボシクリブ、パゾパニブ、ポナチニブ、キザルチニブ、レゴラフェニブ、ルキソリチニブ、ソラフェニブ、スニチニブ、タンドゥチニブ、チバニチブ、チボザニブ、トラメチニブ、バンデタニブ、ヴァタラニブ、およびベムラフェニブから成る群から選択され、そして、前記抗血管新生剤は、好ましくはニンテダニブまたはアキシチニブであり、そして
前記αアドレナリン受容体を活性化する工程は、ナファゾリン、テトラヒドロゾリン、オキシメタゾリン、メトキサミン、フェニレフリン、キシロメタゾリン、オキセドリン、アプラクロニジン、ミバゼロル、クロニジン、ブリモニジン、アルファメチルドーパ、グアンファシン、デキセメディトミジン、(+)−(S)−4−1−(2,3−ジメチル−フェニル)−エチル−1,3−ジヒドロ−イミダゾール−2−チオン、1−(イミダゾリジン−2−イル)イミノリンダゾール、メトキサミン、フェニレフリン、チザニジン、キシラジン、グアナベンズ、およびアミトラズからなる群から選択されるαアドレナリン受容体アゴニストを投与することを含み、そして、前記αアドレナリン受容体アゴニストは、好ましくはブリモニジンである請求項13に記載の方法。 - 前記抗血管新生剤および前記α−アドレナリン受容体アゴニストは、局所眼製剤、軟膏、ゲル、徐放性半固形製剤、徐放性固形製剤又は眼用インプラントの形態で罹患した眼に投与される請求項14に記載の方法。
- 前記徐放性半固体製剤、前記徐放性固体製剤、または前記眼のインプラントは、罹患した眼に注射される請求項15に記載の方法。
