を含む抗原性フェリチンタンパク質である。

ヒト適合性のグリコシル化は、組換え薬物製品における安全性および効能に関与し得る。規制当局の承認は、極めて重要な品質属性として適切なグリコシル化を証明することを条件とし得る(Zhangら、Drug Discovery Today 21(5):740〜765頁(2016)を参照されたい)。N−グリカンは、アスパラギン側鎖のグリコシル化に起因することができ、ヒトと、細菌および酵母のような他の生物との間で構造が異なり得る。このように、本開示に従うフェリチンにおいて、非ヒトグリコシル化および/またはNグリカン形成を低減または除去することが望ましいであろう。一部の実施形態では、フェリチンのグリコシル化を制御することにより、特にヒトワクチン接種に使用する場合、組成物の効能および/または安全性が改善される。
一部の実施形態では、フェリチンは、本明細書に記載される変異の1つより多くのタイプを含む。一部の実施形態では、フェリチンは、グリコシル化を減少させる変異、内部システインを除去する変異、および表面に露出したシステインを生成する変異から独立して選択される1つまたはそれ以上の変異を含む。一部の実施形態では、フェリチンは、グリコシル化を減少させる変異、内部システインを除去する変異、および表面に露出したシステインを生成する変異を含む。
上記で考察したように、H.ピロリフェリチンに関して記載した変異に対応する変異の位置は、ペアワイズまたは構造アライメントによって同定することができる。構造アライメントは、タンパク質がかなりの配列変動にも関わらず類似の構造を共有し、ファミリーの多くのメンバーが構造的に特徴付けされている、フェリチンのような大きいタンパク質ファミリーにとって適切であり、これもまた、インフルエンザ(例えば、血球凝集素)、ボレリア(例えば、OspA)、RSV(例えば、RSV FまたはG)、およびEBV(例えば、gL、gH、gp220、またはgp42)のような、本明細書に記載される他のポリペプチドの異なる形態における対応する位置を同定するために使用することができる。タンパク質データバンク(PDB)は、その受託番号と共に以下に列挙するフェリチンを含む、多くのフェリチンに関する3D構造を含む。

表面に露出したアミノ酸(例えば、システイン)に結合することができる免疫刺激性部分を、本開示に従ってフェリチンにおいて使用することができる。一部の実施形態では、免疫刺激性部分は、B細胞アゴニストである。

金属フリークリックケミストリーは、タンパク質の起こり得る酸化を回避するためにコンジュゲーション反応に関して使用することができる。金属フリークリックケミストリーは、抗体コンジュゲートを形成するために使用されている(van Geelら、Bioconjugate Chem.2015,26,2233〜2242頁を参照されたい)。
本明細書で使用されるフェリチンは、スルフヒドリルとしても知られるチオールを含むシステインを含むことができ、これはスルフヒドリル反応性化学基との反応に関して利用可能である(または還元を通して利用可能となり得る)。このように、システインは、アジュバントのような免疫刺激性部分をフェリチンに付加するための化学選択的改変を可能にする。塩基性条件下では、システインは、脱プロトン化されて、チオレート求核試薬を生成し、これはマレイミドおよびヨードアセトアミドのような弱い求電子試薬と反応することができる。システインとマレイミドまたはヨードアセトアミドとの反応は、炭素−硫黄結合をもたらす。

EBVは、コア膜融合機構を形成し、ウイルスを細胞内に貫通させる3つの糖タンパク質、すなわち糖タンパク質B(gB)、gH、およびgLを有する。gLおよびgHは、例えば以前に、Matsuuraら、Proc Natl Acad Sci U S A.2010 Dec 28;107(52):22641〜6頁に記載されている。ワクチンとして使用するためのgLおよびgHの単量体および三量体は、例えばCuiら、Vaccine.2016 Jul 25;34(34):4050〜5頁に記載されている。gHおよびgLタンパク質は会合して、ウイルス侵入にとって必要な効率的な膜融合および上皮細胞受容体との結合にとって必要であると考えられるヘテロ二量体複合体を形成する。
一部の実施形態では、EBVポリペプチドは、gp220ポリペプチドを含む。gp220−ハイブリッドウシガエル/H.ピロリフェリチンナノ粒子は、Kanekiyo Cell.2015 Aug 27;162(5):1090〜100頁において既に記載されている。このナノ粒子は、本明細書に記載されるある特定のフェリチンからの他の差の中でも、表面に露出したシステインを提供する変異またはシステインを含むリンカーを含まない。
一部の実施形態では、EBVポリペプチドは、gp42ポリペプチドを含む。例示的なgp42配列を、配列番号434として提供する。例えばgLおよびgHポリペプチドとの融合体に含めるために適したさらに例示的なgp42配列を、配列番号239として提供する。gLおよびgHポリペプチドとの融合体に含めるために適したさらに例示的なgp42配列を配列番号240として提供する。

一部の実施形態では、インフルエンザポリペプチドは、完全長または部分長のHAまたはNAを含むHAまたはNAポリペプチドである。任意のHAまたはNAポリペプチドを使用してもよい。HAまたはNAポリペプチドは、天然に存在してもよく、または天然から変化してもよい。一部の実施形態では、HAは、H1〜H18のいずれか1つに由来する。一部の実施形態では、NAは、N1〜N11のいずれか1つに由来する。

N結合グリコシル化は、タンパク質のアスパラギン(Asn;N)残基のアミド窒素に対するグリカンの結合である。結合プロセスは、グリコシル化タンパク質をもたらす。グリコシル化は、タンパク質の翻訳後にグリコシル化酵素により接近可能であり、認識されるタンパク質中の任意のアスパラギン残基で起こり得、NXS/TX部位の一部である接近可能なアスパラギンで最も一般的であり、アスパラギンの後に続く第2のアミノ酸残基は、セリンまたはトレオニンである。非ヒトグリコシル化パターン(例えば、ある特定の非ヒト細胞タイプにおけるグリコシル化部位を含むポリペプチドの発現に起因する)は、抗体を誘発するために使用する場合、ポリペプチドを望ましくないほど反応原性にすることがある。加えて、通常グリコシル化されないポリペプチドのグリコシル化は、その免疫原性を変化させることができる。例えば、グリコシル化は、タンパク質内の重要な免疫原性エピトープを隠すことができる。このように、グリコシル化を低減または除去するために、アスパラギン残基またはセリン/トレオニン残基のいずれかを、例えば別のアミノ酸への置換によって改変することができる。

一部の実施形態では、RSV Fポリペプチドは、完全長または断片の野生型RSV Fポリペプチドである。一部の実施形態では、融合前RSV Fと融合後RSV Fの間で共有されるRSVポリペプチドのエピトープをブロックする。エピトープのブロッキングにより、抗原性RSVポリペプチドを対象に投与した場合に、エピトープに対する抗体の生成が低減または除去される。これは、融合前コンフォメーションのようなFの特定のコンフォメーションに対して特異的なエピトープを標的とする抗体の割合を増加させることができる。Fは、まだ細胞に侵入していないウイルスの融合前コンフォメーションを有することから、融合前Fを標的とする抗体の割合の増加は、より大きい程度の中和を提供することができる(例えば、本明細書に記載されるように中和対結合比として表記される)。ブロッキングは、共有されたエピトープの近位にあるNグリカンのようなかさ高い部分を操作することによって達成することができる。例えば、野生型Fに存在しないNグリコシル化部位を、例えば適切な残基をアスパラギンに変異させることによって付加することができる。一部の実施形態では、ブロックされたエピトープは、RSV Fの抗原性部位1のエピトープである。一部の実施形態では、融合前RSV Fと融合後RSV Fの間で共有される2つまたはそれより多くのエピトープがブロックされている。一部の実施形態では、RSV Fの抗原性部位1の2つまたはそれより多くのエピトープがブロックされている。一部の実施形態では、ブロックされたエピトープと地形学的に重複する1つもしくはそれ以上、または全てのエピトープが、同様にブロックされ、場合によりブロックされたエピトープはRSV Fの抗原性部位1のエピトープである。

本明細書で使用される場合、アジュバントは、表面に露出したアミノ酸,例えば、システインを介してフェリチンにコンジュゲートしてもよい。非コンジュゲートアジュバントもまた、本明細書に記載される抗原性フェリチンポリペプチドと共に対象に投与してもよい。一部の実施形態では、抗原性フェリチンポリペプチドと共にアジュバントを投与すると、アジュバントを投与しない非フェリチンポリペプチド単独、または抗原性フェリチンポリペプチド単独の投与と比較して、対象において非フェリチンポリペプチドに対するより高力価の抗体を生じる。アジュバントは、抗原性ポリペプチドに対するより早期の、より強力な、またはより持続性の免疫応答を促進し得る。
様々な実施形態では、本明細書に記載される抗原性フェリチンポリペプチドおよび/または関連する実体を含む医薬組成物を提供する。一部の実施形態では、医薬組成物は、病原体に対して保護的免疫応答のような免疫応答を誘発することが可能な免疫原性組成物(例えば、ワクチン)である。
同様に、本明細書に記載される抗原性ポリペプチドをコードする核酸も提供される。一部の実施形態では、核酸はmRNAである。ポリペプチドをもたらす翻訳を受けることが可能な任意の核酸は、本開示の目的に関してmRNAであると考えられる。
同様に本明細書において、本明細書に記載される1つまたはそれ以上の抗原性ポリペプチド、核酸、抗原性フェリチン粒子、抗原性ルマジンシンターゼ粒子、組成物、または医薬組成物を含むキットも提供される。一部の実施形態では、キットは、溶媒、溶液、緩衝剤、説明書、または乾燥剤の1つまたはそれ以上をさらに含む。
以下の実施例は開示した特定の実施形態を説明するために提供され、本開示の範囲を限定するものとは決して解釈すべきでない。
OspAおよびフェリチンを含む抗原性ポリペプチドを産生した。
OspA−フェリチンナノ粒子の免疫原性を評価するため、Ribiアジュバントの存在下の血清型1OspA−フェリチンナノ粒子、または血清型1で脂質化された完全長の組換えOspAを含むイヌワクチンであるRECOMBITEK(登録商標)Lyme(B.ブルグドルフェリの精製された外表面プロテインA(OspA)の脂質懸濁液)で2回、C3Hマウスを免疫した(図4)。C3H/HeNマウスは、0週および4週で筋肉内ワクチン接種した。2回目の投与の2週後から血清についてELISAを行った。Ribi(Sigma社アジュバントシステムCat#S6322−1vl)をPBS1ml中に再懸濁して1分ボルテックスし、免疫に先立って等体積で抗原に添加した。
このOspA−フェリチンの保護有効性を評価するため、B.ブルグドルフェリを持つダニを免疫マウスまたは対照マウスに感染させるチャレンジモデルを用いた(Rosa,P.A.ら、Nat Rev Microbiol 3(2):129〜43頁(2005)参照)。
クリックケミストリーによってTLRアゴニスト(図6A)またはCpG(配列番号210;ISS−1018、図7A)等の免疫刺激性部分の直接コンジュゲーションを可能にする、フェリチンナノ粒子の表面上のシステインの操作(S111C)によって、自己アジュバント性構築物を産生した。直接コンジュゲーションの手順は下記の通りであった。哺乳動物から生成された材料を50mMのTris、pH8.5中の10mMのTCEP(Amresco社、K831−10G)で1時間還元してシステイン化を解除した。次いでタンパク質を100mMのTris、pH8、50mMのNaCl中に透析してTCEPを除去した。大腸菌から生成した材料は還元する必要はない。DBCO−PEG4−マレイミドリンカー(Sigma−Aldrich社cat#760676、5mg)をDMSO中で5mg/mlに再懸濁した。リンカー2.5mgを体積10mlでタンパク質3mgに加えた(最終DMSO濃度は5%であった)。リンカーを還元タンパク質と室温で30分インキュベートした。Ambicon社の100MWカットオフフィルター濃縮器を用いて緩衝液交換により過剰なリンカーを除去した(Millipore社Cat#UFC910096)。アジド−PEG4−3M−012(社内で合成)およびアジド−CPG(ISS−1018、IDT社によりカスタム合成)を最終のクリックケミストリーステップに用いた。最終のコンジュゲーションステップとしてアジュバント0.5mgをタンパク質0.5mgに加え、37℃で6時間、次いで4℃で終夜インキュベートした。Ambicon社の100MWカットオフフィルター濃縮器を用いて緩衝液交換により過剰なアジュバントを除去した。コンジュゲーションの効率は、3M−012についてはマススペクトロメトリーで、CPGについてはSDS−PAGE解析で確認した。
血清型1OspA株B.ブルグドルフェリは米国で疾患を引き起こす一方、B.アフゼリ(血清型2)、B.ガリニ(血清型3、5、6、7)、およびB.ババリエンシス(血清型4)は欧州、アジア、およびその他の地域で疾患を引き起こす。幅広く交差保護する組成物を産生するため、血清型1、2、3、4、5、および7(配列番号1、5、6、7、8、および10)についてOspA−フェリチンナノ粒子を設計した。これらの粒子を発現させ、大腸菌からアニオン交換およびサイズ排除クロマトグラフィーを用いて精製した(図3A)。全てのOspA−フェリチン抗原性ポリペプチドは予想された分子量である47kDaを有し、DLS解析および透過型電子顕微鏡によっても6種のOspAナノ粒子全ての形成が確認された(図3B)。
OspAとフェリチンとの間に可撓性を提供するためにいくつかの異なったリンカーを試験した。構築物は1個から5個の−GGGS−(配列番号443)配列の範囲であった。種々のリンカー構築物を精製し、均一な大きさのナノ粒子を形成した。
もう1つのナノ粒子であるアクイフェックス・エオリクス(Aquifex aeolicus)由来のルマジンシンターゼについて、OspAの抗原呈示を検討した。様々な血清型を含むOspA−ルマジンシンターゼ粒子は、大腸菌細胞からアニオン交換クロマトグラフィーおよびサイズ排除クロマトグラフィーによって容易に精製された。OspA血清型1(配列番号12、図19A〜図19C)、OspA血清型2(配列番号16、図20A〜図20C)、OspA血清型3(配列番号17、図21A〜図21B)、OspA血清型4(配列番号18、図17A〜図17C)、OspA血清型5(配列番号19、図22A〜図22C)、およびOspA血清型7(配列番号21、図23A〜図23C)からなる構築物を産生し特徴解析した。OspA−ルマジンシンターゼ粒子はEMで15.8nmの粒子を形成しており、DLSで大きさは均一であった。
HAのエクトドメイン配列(48個のC末端膜貫通残基を欠除する)をフェリチンのN末端に融合して哺乳動物細胞中で自己集合するナノ粒子を生成させることによって、HAナノ粒子(HA−Np)を産生した。フェリチンは表面に露出したアミノ酸をシステインに置き換える変異(配列番号208のフェリチン配列に対するS26C、A75C、またはS111Cの変異に起因する)を含み、HA−Npへのアジュバントのコンジュゲーションが可能になる。そのようなシステインの表現を図27に提示する。
種々のアジュバントおよびH1/Stem−NpへのTLRアゴニストのコンジュゲーションのH1/Stem−Np組成物の免疫原性に対する影響を評価した。図40Aに、H1/Stem−Np TLR7/8アゴニストコンジュゲートによる免疫で誘起されたH1/ニューカレドニア/1999(NC99)HA三量体に対するIgG抗体力価を示す。H1/Stem−Np 10μgを、アジュバントなしに、等モル量の遊離SM7/8aアジュバント(83.3ng)と混合して、高用量の遊離SM7/8aアジュバント(21.84μg)と混合して、または体積比1:1のPAAもしくはAF03アジュバントと、0週および3週で投与した。H1/Stem−Np−SM7/8aコンジュゲートは、他のアジュバントなしに投与した。2回目の投与の2週後に血清をアッセイした。図40Bに、H1/Stem−Np TLR9アゴニストコンジュゲートによる免疫で誘起されたH1/Stem三量体に対するIgG抗体力価を示す。H1/Stem−Np 10μgを、アジュバントなしに、等モル量の遊離CpGアジュバント(850ng)と混合して、高用量の遊離CpGアジュバント(20μg)と混合して、または体積比1:1のAF03アジュバントと、0週および3週で投与した。H1/Stem−Np−CpGコンジュゲートは、他のアジュバントなしに投与した。2回目の投与の2週後に血清をアッセイした。
上に概要を示したプロセスを用いて、図26Aに提示するインフルエンザポリペプチドを含むHA−Npを発現し、培地中に放出した(HA部分の配列アライメントを図26Aに示し、デンドグラムを図26Bに示す)。図26B中の矢印は候補ポリペプチドを示す。図43Aに列挙するHA−Npはアニオン交換およびサイズ排除クロマトグラフィーによって293Expi細胞培養上清から精製した。ナノ粒子の生成に成功したことを確認するクーマシー染色の結果を図43Bに示す。
特定のウイルス株からのHA−Npの免疫原性を検討するため、目的の株からのナノ粒子でマウスを2回免疫し、第2の免疫の3週後に血球凝集素阻害アッセイ(HAI)を用いて血清をアッセイした。全ての免疫は動物の取り扱いのための確立されたガイドラインに従った。Balb/cマウス(5匹/群)を0週および3週にHA−フェリチンナノ粒子220ng(HA含量170ng)で、および適用可能な場合には筋肉内注射(後ろ足あたり50μL)の直前に1:1でアジュバントと混合して、免疫した。Ribi(Sigma社アジュバントシステムカタログ#S6322−1vl)またはAF03(Sanofi Pasteur社)を図の凡例に指示するように用いた。二価、三価、および四価の組合せについては、注射の前に各ナノ粒子220ngを事前混合した。ブースト注射の2週および3週後に血清を採取した。
選定したHA−Npの組合せによって誘発されたHAI交差反応性を評価した。個別のナノ粒子を組み合わせることによって作成した二価、三価、または四価の処方で、上述のようにマウスを免疫し、試験した。NC99およびCA09のHA−Npの二価の組合せは、いずれかの一価の組成物と比較して拡張した交差反応性を示した(図46A)。しかし、この二価の組合せは1934年〜1957年および1977年〜1991年の古い分岐した株に対しては検出可能な抗体力価を誘発しなかった。COBRA X6およびCOBRA P1の二価の組合せの免疫原性は同じ傾向に従った(図46B)。この組合せはNC99/CA09の二価組成物と比較して増大した幅を示したが、いくつかの株に対するHAI力価は中程度であった。三価の組合せについては、NC99およびCA09のHA−Npへの第3成分の含有によって、第3成分がFM47 HA−Np(図46C)またはHK77 HA−Np(図46E)の場合には交差反応性が増大したが、MAL54 HA−Np(図46D)は幅を増強しなかった。四価処方における第4成分の添加は、NC99、CA09、およびHK77のHA−Npの三価の組合せと比較して観察可能な交差反応性の付加を生じなかった(図46F〜図46H)。
ある種のHA−Np組合せの有効性を、ヒト疾患に関連する動物モデルであるフェレットで試験した。フェレット(n=12匹/群)を、リン酸緩衝食塩液(図49A)、CA09不活化インフルエンザワクチン(IIV、図49B)、野生型HA−Npの三価の組合せ(NC99+CA09+HK77、図49C)、またはCOBRA P1+COBRA X6+HK77 HA−Npの組合せ(図49D)のいずれかで免疫した。筋肉内注射の前に、これらの組成物をAF03アジュバントと1:1で混合して最終注射量を1mlとした。
EBVに対する抗体を誘発する抗原性ポリペプチドを開発した。EBVのgLおよびgHのポリペプチドを一本鎖として呈示する自己集合性フェリチンナノ粒子を開発し、これらのナノ粒子のマウスにおける免疫原性を評価した。
一本鎖のgL/gHナノ粒子およびgp220ナノ粒子を含む組成物を用いて、二価の免疫を実施した。gp220ナノ粒子(配列番号401)を含ませることは、上述のようにワクチン接種したマウスの血清を用いるELISA結合アッセイによって測定して、一本鎖gL/gHナノ粒子(gL−gH_C5 NP[配列番号419])によって誘発された免疫応答に対して顕著な干渉効果を有しなかった(図56A〜図56B、それぞれ個別の希釈および結合力価における測定を示す)。同様に、一本鎖gL/gHナノ粒子と組み合わせて投与した場合、ELISAによって測定して、gp220ナノ粒子への免疫応答に対する応答において干渉は観察されなかった(図57A〜図57B、それぞれ個別の希釈および結合力価における測定を示す)。
次に、フェリチンナノ粒子へのアジュバントのコンジュゲーションを評価した。図58Aに、その中のフェリチンが表面に露出したアミノ酸をコンジュゲーションのために利用可能なシステインに置き換える変異を含む構築物を図示する。図58Bは、PEG4リンカーおよびマレイミドに連結された例示的な免疫刺激性部分(SM7/8a、TLR−7/8アゴニスト)を示す。このマレイミドはフェリチンの表面に露出したシステインにリンカー(それ自体SM7/8aに結合している)を共有結合でコンジュゲートするために用いることができる。SM7/8aにコンジュゲートされたフェリチンに融合した一本鎖gL/gHポリペプチドを含むポリペプチドを、図58Cの電子顕微鏡写真に示す。
一本鎖gL/gH(gL/gH_C5、配列番号419)を含むナノ粒子を投与した3カ月後の免疫原性を評価する検討を実施した。BALB/cマウス(n=5/群)を3週の投与間隔で2回免疫した。裸のフェリチン(すなわち、いずれのポリペプチドまたはアジュバントにもコンジュゲートしていないフェリチン)1μgを一本鎖gL/gHを含むナノ粒子1μgと投与し、ナノ粒子は混合したAF03アジュバントの存在下または非存在下に処方した。13週目にELISA解析のために血液を採取した。AF03アジュバントの投与を受けるマウスについては、体積比1:1でAF03とナノ粒子組成物を混合した。それぞれのマウスは上述のナノ粒子組成物100μLの投与を受けた。数匹のマウスはその中のフェリチンがSM7/8aにコンジュゲートされた一本鎖のgL/gHを含むナノ粒子の投与を受けた(図67〜図68における「7/8a」)。
トリコプルシア・ニイフェリチンならびにgp220および/またはgL/gHポリペプチドを含むナノ粒子も開発した。トリコプルシア・ニイフェリチンナノ粒子は1:1の比で自己集合した重鎖および軽鎖を含む。軽鎖を有する1つの非フェリチンポリペプチドと重鎖上の別の非フェリチンポリペプチドとを組み合わせることにより、個別のナノ粒子の表面上に2つの異なったポリペプチドを提示することができることが見出された。したがって、例えば自己集合したトリコプルシア・ニイフェリチンナノ粒子は、gp220とgL/gHの両方を提示することができた。
フェリチンに融合したgH/gL/gp42の一本鎖構築物の概略図を(配列番号227〜231および241〜242のそれぞれにおいて)図86Aに示す。各タンパク質の間の融合は、可撓性のアミノ酸リンカーまたは硬いアミノ酸リンカーを介している。一本鎖gH/gL/gp42分子は、ナノ粒子上で1:1:1の比のヘテロ三量体を形成する。
他のパラミキソウイルスFタンパク質と同じく、RSV FはN末端シグナルペプチドおよびタンパク質をウイルス表面に固定するC末端膜貫通領域を有する前駆体タンパク質として発現される。RSV Fはプロテアーゼフリンによって細胞内切断を受けて疎水性の融合ペプチド(図87Aの「FP」)を放出する。融合ペプチドの役割は、感染の際に標的細胞に結合することである。融合ペプチドに隣接してヘプタッドリピート領域A(HRA)があり、一方ヘプタッドリピート領域B(HRB)は膜貫通ドメインに隣接している。
動物研究に先立って、DS−CAV1およびRSV Fナノ粒子の濃度を、Octetを用いる結合によって解析した。融合前抗原の融合前特異的抗体D25およびAM14への結合も、ForteBio Octet装置を用いて測定した。全てのアッセイはPBS中、30℃で実施した。抗体をプロテインA(ProA)センサーチップ(forteBio#18−5013)に400秒、負荷して、ほぼ飽和に達するまで捕捉させた。次いでバイオセンサーチップをPBS中で90秒、平衡にし、次いで公知の濃度の抗原とPBS中で300秒会合させ、次いでPBS中で抗原を解離させた。1:1の相互作用を仮定したデータ解析および曲線フィッティングを、公知の濃度の精製したPre−F−NPの結合の外部標準曲線を用いるOctet Data Analysis HT10.0ソフトウェアで行った。CHO馴化培地中のPre−F−NP濃度を決定する例示的なアッセイ結果を図95Bに示す。
マウスにおけるRSV抗原に対するin vivo応答を評価するため、雌BALBcマウスを0週、3週、および6週で指定した用量のRSV抗原で筋肉内免疫した。他に注記しなければ、RSV抗原を(例えばとりわけ図96A〜図96B、および図12A〜図12Bの実験において)ベッドサイド混合戦略によりAF03でアジュバント化した。すなわち、それぞれの後ろ足に50μlを注射する直前に、関連するタンパク質の溶液50μlをSanofi社のアジュバントAF03(スクアレン系エマルジョン、Kluckerら、J Pharm Sci.2012 Dec;101(12):4490〜500頁参照)50μlと混合した。非アジュバント化群については、抗原を上記のように混合したが、AF03を等体積のPBSに置き換えた。SPA09またはアラムと混合した抗原については、AF03をそれぞれ等体積のSPA09またはアラムに置き換えて上記の手順を実施した。いずれの処方についても免疫による有害な影響は観察されなかった。最初の免疫の1日前、およびそれぞれの注射の少なくとも2週後(すなわち、2週、5週、および8週)に血液を採取した。他に特定しなければ、示したデータは3回目の注射の2週後(8週、2wp3とも注記する)のものであった。典型的には、血清は免疫前の動物(ナイーブと注記)、2回目の注射の2週後(ポスト2または2wp2)、または3回目の注射の2週後(ポスト3rdまたは2wp3)から分析した。
RSV Gcc−NPを上述のように調製した。マウスにおけるRSV Gcc−NPに対するin vivo応答を評価するため、雌BALBcマウスを0週、3週、および6週で、高用量(5μg)または低用量(0.5μg)の抗原を用い、指定された用量のRSV抗原で筋肉内免疫した。他に注記しなければ、RSV Gcc−NPをベッドサイド混合戦略によりAF03でアジュバント化した。すなわち、それぞれの後ろ足に50μlを注射する直前に、タンパク質溶液50μlをSanofi社のアジュバントAF03(スクアレン系エマルジョン、Kluckerら、J Pharm Sci.2012 Dec;101(12):4490〜500頁参照)50μlと混合した。免疫による有害な影響は観察されなかった。最初の免疫の1日前、およびそれぞれの注射の少なくとも2週後(すなわち、2週、5週、および8週)に血液を採取した。他に特定しなければ、示したデータは3回目の注射の2週後(8週、2wp3とも注記する)のものである。典型的には、血清は免疫前の動物(ナイーブと注記)、2回目の注射の2週後(ポスト2または2wp2)、または3回目の注射の2週後(ポスト3rdまたは2wp3)から分析した。
Pre−F−NPおよびGcc−NPのヒト細胞における応答を誘発する能力を実証するため、MIMICプラットフォームでの実験を実施した。MIMICプラットフォームはチャレンジに際して先天的なおよび後天的な抗原特異的応答を迅速かつ再現性よく産生することができる自己ヒト免疫細胞のみからなっている。以前の研究により、HBV、破傷風トキソイド、モノクローナル抗体、YF−VAX、およびインフルエンザB細胞応答等の多様な標的に対してin vivoの免疫プロファイルを反復するMIMICシステムの能力が実証されている。Pre−F−NP RF8140(配列番号23)、対、融合後F三量体(配列番号24)による処置によって誘発されるRSV融合前F三量体結合抗体応答をヒトB細胞で比較し、代表的なベースライン応答と比較した。ヒトB細胞において、Pre−F−NP(RF8140、配列番号23)、対、融合後F(配列番号24)による処置によって誘発された融合前F三量体(DS−CAV1、配列番号25)、対、融合後F三量体(配列番号24)に対する測定された結合応答の比を決定した。異なるF抗原による処置によって誘発されたMIMICからの抗体をVERO細胞アッセイを用いて測定した。ヒトB細胞において、Pre−F−NP(RF8140、配列番号23)、対、融合後F三量体(配列番号24)による処理によって誘発された中和力価を非処理群と比較し、RF8140(配列番号23)がヒト細胞において優れた中和性応答を誘発することを示す。Gcc−NPがGccペプチド(配列番号29)単独より優れたG抗体応答を誘発することを実証するため、ヒト細胞をGccペプチド単独(配列番号29)またはヒトB細胞中でナノ粒子にコンジュゲートしたGccペプチド(Gcc−NP)で処置した。Gcc−NPは優れたG結合抗体応答を誘発した。したがって、Pre−F−NPおよびGcc−NPはヒトの免疫において免疫応答を誘発した。
Claims (27)
- 表面に露出したアミノ酸をシステインに置き換える変異を含むフェリチンタンパク質。
- システインを含むNまたはC末端リンカーを含むフェリチンタンパク質。
- 表面に露出したアミノ酸を介してフェリチンタンパク質に連結した1つまたはそれ以上の免疫刺激性部分を含むフェリチンタンパク質。
- 非フェリチンポリペプチドをさらに含む抗原性フェリチンタンパク質である、請求項1〜3のいずれか1項に記載のフェリチンタンパク質。
- 抗原性フェリチンタンパク質であって、(i)表面に露出したアミノ酸をシステインに置き換える変異およびシステインに連結した免疫刺激性部分;ならびに(ii)非フェリチンポリペプチドを含む前記抗原性フェリチンタンパク質。
- 抗原性フェリチンタンパク質であって、(i)表面に露出したシステイン、(ii)該フェリチンタンパク質のN末端側のペプチドリンカー、および(iii)該ペプチドリンカーのN末端側の非フェリチンポリペプチドを含む前記抗原性フェリチンタンパク質。
- 表面に露出したアスパラギンを非アスパラギンアミノ酸に置き換える変異をさらに含む、請求項1〜6のいずれか1項に記載のフェリチンタンパク質。
- 内部システインを非システインアミノ酸に置き換える変異をさらに含む、請求項1〜7のいずれか1項に記載のフェリチンタンパク質。
- 内部システインは、H.ピロリフェリチンの31位、またはペアワイズもしくは構造アライメントによって決定される、H.ピロリフェリチンの31位に対応する位置にある、請求項8に記載のフェリチンタンパク質。
- 抗原性フェリチンタンパク質であって:
a.表面に露出したアミノ酸をシステインに置き換える変異およびシステインに連結した免疫刺激性部分;
b.H.ピロリフェリチンの31位の内部システイン、またはペアワイズもしくは構造アライメントによって決定される、非H.ピロリフェリチンの31位と類似の位置の内部システインを、非システインアミノ酸に置き換える変異
c.表面に露出したアスパラギンを非アスパラギンアミノ酸に置き換える変異;ならびに
d.非フェリチンポリペプチド
を含む前記抗原性フェリチンタンパク質。 - 非システインアミノ酸はセリンである、請求項8〜10のいずれか1項に記載のフェリチンタンパク質。
- アスパラギンはH.ピロリフェリチンの19位、またはペアワイズもしくは構造アライメントによって決定される、非H.ピロリフェリチンの類似の位置にある、請求項7〜11のいずれか1項に記載のフェリチンタンパク質。
- フェリチンは、H.ピロリフェリチンのE12C、S26C、S72C、A75C、K79C、S100C、およびS111C変異の1つもしくはそれ以上、またはペアワイズもしくは構造アライメントによって決定される、非H.ピロリフェリチンにおける1つまたはそれ以上の対応する変異を含む、請求項1〜12のいずれか1項に記載のフェリチンタンパク質。
- 非フェリチンポリペプチドは、インフルエンザ、エプスタイン−バーウイルス、呼吸器合胞体ウイルス、またはボレリア由来のポリペプチドである、請求項4〜13のいずれか1項に記載のフェリチンタンパク質。
- 非フェリチンポリペプチドはRSV Gポリペプチドを含み、場合により該RSV Gポリペプチドは、Gポリペプチド中央保存領域を含む、請求項14に記載のフェリチンタンパク質。
- フェリチンと非フェリチンポリペプチドとの間にペプチドリンカーをさらに含む、請求項4〜5または7〜15のいずれか1項に記載のフェリチンタンパク質。
- システインに連結され、水素結合またはイオン結合することが可能な部分を含む免疫刺激性部分を含む、請求項1〜16のいずれか1項に記載のフェリチンタンパク質。
- TLR2、TLR7/8、TLR9、またはSTINGのアゴニストである免疫刺激性部分を含む、請求項1〜17のいずれか1項に記載のフェリチンタンパク質。
- 配列番号201〜207または211〜215のいずれか1つと80%、85%、90%、95%、98%、または99%の同一性を有するアミノ酸配列を含む、請求項1〜18のいずれか1項に記載のフェリチンタンパク質。
- 請求項1〜19のいずれか1項に記載のフェリチンタンパク質を含むフェリチン粒子。
- 請求項1〜20のいずれか1項に記載のフェリチンタンパク質またはフェリチン粒子、および薬学的に許容される担体を含む組成物。
- 第1のフェリチンタンパク質および第2のフェリチンタンパク質を含む組成物であって、該第1のフェリチンタンパク質はフェリチン重鎖および第1の非フェリチンポリペプチドを含み、該第2のフェリチンタンパク質はフェリチン軽鎖および第2の非フェリチンポリペプチドを含み、該第1および第2の非フェリチンポリペプチドは異なり、場合によりフェリチン粒子が該第1のフェリチンタンパク質および該第2のフェリチンタンパク質を含む、前記組成物。
- アジュバントをさらに含む、請求項21または22に記載の組成物。
