Abstract
Description
項1. IFN−I受容体を発現しており、IFN−I誘導性制御領域と、前記制御領域の下流に連結されたレポーター遺伝子とを含むレポーター構築物が導入された細胞を含む、全身性エリトマトーデスの治療薬に対するコンパニオン診断薬。
項2. 前記細胞がヒト胎児腎細胞である、請求項1に記載のコンパニオン診断薬。
項3. BAFF/APRIL受容体又はBAFF受容体を発現しており、BAFF/APRIL誘導性制御領域又はBAFF誘導性制御領域と、前記制御領域の下流に連結されたレポーター遺伝子とを含むレポーター構築物が導入された細胞を含む、全身性エリトマトーデスの治療薬に対するコンパニオン診断薬。
項4. 前記BAFF/APRIL受容体が、BCMA、TACI及びBAFF−Rからなる群より選択される、項3に記載のコンパニオン診断薬。
項5. 前記BAFF受容体が、BAFF−Rの細胞外領域とBCMAの細胞内領域とを含む融合タンパク質である、項3及び4に記載のコンパニオン診断薬。
項6. 前記細胞がヒト胎児腎細胞である、項3〜5のいずれかに記載のコンパニオン診断薬。
項7. 項1又は2に記載のコンパニオン診断薬と、項3〜5のいずれかに記載のコンパニオン診断薬とを含む、全身性エリトマトーデスの治療薬に対するコンパニオン診断キット。
項8. 被験動物から得られた生体試料について、項1又は2に記載のコンパニオン診断薬を用いて測定されるレポーター活性値を、IFN−I活性値として取得する工程1と、
前記生体試料について、項3〜5のいずれかに記載のコンパニオン診断薬を用いて測定されるレポーター活性値を、BAFF活性値又はBAFF及びAPRIL活性値として取得する工程2と、
前記IFN−I活性値並びに前記BAFF活性値又はBAFF及びAPRIL活性値に基づき、前記生体試料中のIFN−IとBAFF又はBAFF及びAPRILとのうち、相対的に多く存在する全身性エリトマトーデス憎悪因子を決定する工程3と、を含む、生体試料の分析方法。
項9. 前記工程3が、
前記IFN−I活性値並びに前記BAFF活性値又はBAFF及びAPRIL活性値を、IFN−I軸とBAFF軸又はBAFF及びAPRIL軸との二次元座標系に展開する工程31と、
展開された座標が、IFN−I活性値が相対的に高くBAFF又はBAFF及びAPRILが相対的に低いグループと、IFN−I活性値が相対的に低くBAFF又はBAFF及びAPRILが相対的に高いグループとのいずれに属するかを決定する工程32とを含む、項8に記載の分析方法。
項10. 前記生体試料が血清である、項8又は9に記載の分析方法。
本発明のコンパニオン診断薬は、全身性エリトマトーデス(SLE)治療薬に対する有効性を予測するコンパニオン診断薬として用いられるものであり、憎悪因子による刺激によってレポーター活性を生じるように設計された細胞(以下において、レポーター細胞とも記載する。)を含む。
本発明のコンパニオン診断薬が有効性を予測するSLE治療薬としては、SLEの疾患憎悪因子又はその受容体に対する抗体が挙げられる。SLEの疾患憎悪因子としては、インターフェロン(IFN−I)、B細胞活性化因子(BAFF)、増殖誘導リガンド(APRIL)が挙げられる。具体的なSLE治療薬としては、IFN−Iをターゲットとする治療薬として、IFN−I受容体の中和抗体(例えば、Anifrolumab)が挙げられ、BAFF又はBAFF及びAPRILをターゲットとする治療薬として、BAFFに対する抗体又はBAFF及びAPRILに対する抗体(例えば、Belimumab)が挙げられる。
本発明のコンパニオン診断薬に含まれるレポーター細胞は、SLE増悪因子に対する受容体を表面に安定発現しており、SLE増悪因子誘導性制御領域と、前記制御領域の下流に連結されたレポーター遺伝子とを含むレポーター構築物が導入された細胞である。このように設計されることで、レポーター細胞は、生体試料中に存在するSLE憎悪因子をレポーター活性により定量するために用いることができる。具体的には、生体試料中に存在するSLE憎悪因子がレポーター細胞表面の受容体に結合することで、SLE憎悪因子のシグナル伝達経路に対してSLE憎悪因子誘導性制御領域が応答又は活性化され、それによりレポーター遺伝子の発現(レポーター分子の産生)を誘導することができ、更に、産生されたレポーター分子に由来するシグナル量(レポーター活性)を定量することで、生体試料中に存在するSLE憎悪因子を定量することができる。
SLE増悪因子に対する受容体は、所定のSLE増悪因子に対して特異的であり、当該SLE憎悪因子の刺激を細胞内シグナルに変換して細胞の反応を開始させる機能を有するタンパク質である。
本発明で用いられるレポーター細胞において、SLE憎悪因子誘導性の制御領域は、レポーター遺伝子の発現を制御しており、SLE憎悪因子で刺激されることによって、レポーター遺伝子を発現するよう応答又は活性化が誘導される領域である。SLE憎悪因子誘導性の制御領域は、当業者が適宜選択することができる。
レポーター遺伝子は、上記の制御領域の下流に連結されており、制御領域がSLE憎悪因子による刺激で応答又は刺激されることでその発現が誘導(上方制御)される。
レポーター細胞として用いられる細胞としては特に限定されず、原核細胞及び真核細胞を問わない。原核細胞としては、大腸菌、桿菌、酵母等があげられる。真核細胞としては、好ましくは哺乳類細胞、昆虫類細胞等が挙げられ、より具体的には、ヒト胎児腎細胞(HEK293等)、骨髄腫(SP2/0、vNS0等)、肺癌腫(A549等)、結腸癌腫(HCT116等)、子宮頸癌腫(HeLa等)、リンパ球(Jurkat T、Ramos B等)、単球(THP−1等)等のヒト細胞、メラノーマ(B16等)、マクロファージ(J774、RAW 264.7等)等のマウス細胞等が挙げられる。これらの細胞の中でも、好ましくは真核細胞が挙げられ、より好ましくは哺乳類細胞が挙げられ、更に好ましくはヒト細胞が挙げられ、一層好ましくはヒト胎児腎細胞が挙げられる。
レポーター細胞は、必要な遺伝情報を組み込んだ核酸構築物を細胞に導入する公知の方法によって作製することができる。細胞に導入するための核酸構築物は、導入する細胞に応じて、当業者が適宜設計することができる。
本発明のコンパニオン診断薬は、前記レポーター細胞の他に、緩衝液、安定化剤、防腐剤等を含んでいてもよく、また、従来公知の方法に従って製剤化されていてもよい。
本発明のコンパニオン診断キットは、SLE治療薬に対する有効性を予測するコンパニオン診断のために用いられるものであり、上述のコンパニオン診断薬を含む。
本発明の生体試料の分析方法は、上記のコンパニオン診断薬を用いて、生体試料中のSLE憎悪因子を測定する方法であり、後述の工程1、工程2及び工程3を含む。また、工程3は、後述の工程31及び工程32を含むことができる。これによって、生体試料に由来する被験動物の、SLE治療薬の有効性を予測するコンパニオン診断を行うことが可能となる。なお、工程1及び工程2は、順不同であり、工程3は、工程1及び工程2の後に行う。
工程1では、被験動物から得られた生体試料について、上記のIFN−I用レポーター細胞を含むコンパニオン診断薬を用いて測定されるレポーター活性値を、IFN−I活性値として取得する。
工程2では、工程1の対象となった生体試料について、上記のBAFF/APRIL用レポーター細胞又はBAFF用レポーター細胞を含むコンパニオン診断薬を用いて測定されるレポーター活性値を、BAFF活性値又はBAFF及びAPRIL活性値として取得する。
工程3では、工程1で得られたIFN−I活性値、並びに、工程2で得られたBAFF活性値又はBAFF及びAPRIL活性値に基づき、前記生体試料中のIFN−IとBAFF又はBAFF及びAPRILとのうち、相対的に多く存在する全身性エリトマトーデス憎悪因子を決定する。
工程3では、工程31として、前記IFN−I活性値並びに前記BAFF活性値又はBAFF及びAPRIL活性値を、IFN−I軸とBAFF軸又はBAFF及びAPRIL軸との二次元座標系に展開する工程を含むことができる。
工程3では、工程31の後、展開された座標が、IFN−I活性値が相対的に高くBAFF又はBAFF及びAPRILが相対的に低いグループと、IFN−I活性値が相対的に低くBAFF又はBAFF及びAPRILが相対的に高いグループとのいずれに属するかを決定することができる。これによって、相対的に多く存在する全身性エリトマトーデス憎悪因子の決定を視覚的に容易に行うことができる。
(レポーター細胞)
IFN−I用レポーター細胞としては、invivogen社製のHEK−BlueTMIFN−α/β細胞を用いた。このレポーター細胞の模式図を図1に示す。このレポーター細胞は、IFN−Iシグナル経路を構築するヒトSTAT2およびIRF9遺伝子をHEK293T細胞に安定的にトランスフェクションすることにより生成されたものであり、IFN−Iシグナル経路を構築する他の遺伝子(Recetor(IFNAR1、IFNAR2)、JAK1、TyK2、およびSTAT1)も発現している。また、この細胞には、IFN−I誘導性ISG(ISG54プロモーター)及びその制御下にある誘導性SEAP(分泌型アルカリ性ホスファターゼ)レポーター遺伝子が導入されている。つまり、この細胞は、IFNによって細胞が刺激されると、Receptorを介したシグナル経路によりISGが応答し、SEAPを発現するように設計されている。
リコンビナントIFN−a又はIFN−bを1000U/mlで含むサンプルを8段階希釈し、希釈系列サンプルを調製した。希釈系列サンプルそれぞれと、1ウェル当たり25,000細胞のHEK−Blue−IFN−a/b細胞とを混合し、室温(37℃)で24時間培養した。培養後、培養上清のSEAP活性を取得するため、QUANTI−Blue(Invivogen)に対する発色(OD620)を吸光度計にて測定した。
(レポーター細胞の作製)
BAFF/APRIL用レポーター細胞として、以下のように、BCMA強制発現株であるHEK−Blue BCMA細胞を作製した。このレポーター細胞の模式図を図3に示す。このレポーター細胞は、BCMAが強制発現されており、HEK293T細胞にNFκBおよびAP1の結合配列の下流にSEAPを繋いだベクターが導入されたHEK−Blue細胞(invivogen社)に対して、BCMAを強制発現させた細胞である。

BCMAのリガンドであるリコンビナントBAFF及びリコンビナントAPRILを用い、試験例1と同様にして、レポーター細胞の刺激及びそれによるレポーター活性を測定した。
(レポーター細胞の作製)
BAFF/APRIL用レポーター細胞として、以下のように、TACI強制発現株であるHEK−Blue BCMA細胞を作製した。このレポーター細胞の模式図を図5に示す。このレポーター細胞は、TACIが強制発現されており、HEK293T細胞にNFκBおよびAP1の結合配列の下流にSEAPを繋いだベクターが導入されたHEK−Blue細胞(invivogen社)に対して、TACIを強制発現させた細胞である。

TACIのリガンドであるリコンビナントBAFF及びリコンビナントAPRILを用い、試験例1と同様にして、レポーター細胞の刺激及びそれによるレポーター活性を測定した。
HEK−Blue細胞に、BAFFのみと結合するBAFF−Rを受容体として発現させた、HEK−blue−BAFFR細胞を作製した。具体的には、Vector−builder社にてEF1a−BAFFR−T2A−RFPレンチウイルスベクターを作成し(BAFF−Rをコードする配列として用いた配列は配列番号3に示す通りであり、T2Aへの連結のため、BLASTデータベース配列から終結コドンtaaを取り除いて使用した。)、ウイルス化した後、HEK−Blue細胞に強制発現させることによって、HEK−blue−BAFFR細胞を得た。また、FACSにてBAFF−Rの細胞表面への発現を確認した。
ハムスター卵巣由来細胞であるCHO−K1細胞に、AP1遺伝子及びNFκB遺伝子とSEAP遺伝子とを組み込み、BAFFのみと結合するBAFF−Rを受容体として発現させた、CHO−K1−BAFFR細胞を作製した。具体的には、InvivoGen 社より購入したpNiFty3−SEAP vector(AP1−NFkB−mIFN−βpromoter−SEAP)と、EF1a−BAFFR−T2A−RFPレンチウイルスとをレンチウイルス化して、CHO−K1細胞に複数の多重感染度(MOI)にて強制発現させることによって、CHO−K1−BAFFR細胞を得た。また、FACSにてBAFF−Rの細胞表面への発現を確認した。
ヒト単球系細胞株THP−1細胞に、AP1遺伝子及びNFκB遺伝子とSEAP遺伝子とを組み込み、BAFFのみと結合するBAFF−Rを受容体として発現させた、THP−1−BAFFR細胞を作製した。具体的には、InvivoGen 社より購入したpNiFty3−SEAP vector(AP1−NFkB−mIFN−βpromoter−SEAP)と、EF1a−BAFFR−T2A−RFPレンチウイルスとをレンチウイルス化して、THP−1細胞に複数の多重感染度(MOI)にて強制発現させることによって、THP−1−BAFFR細胞を得た。また、FACSにてBAFF−Rの細胞表面への発現を確認した。
(レポーター細胞の作製)
BAFF用レポーター細胞として、以下のように、BAFFR/BCMAキメラ受容体発現株であるHEK−Blue BAFFR/BCMA細胞を作製した。このレポーター細胞は、BAFF−Rの細胞外領域とBCMAの細胞内領域とを含む融合タンパク質(キメラ受容体)を、HEK−Blue細胞(invivogen社)に対して強制発現させた細胞であり、図9〜図10に示す手順で作製された。図11に、このレポーター細胞の模式図を示す。

2種のレポーター細胞それぞれについて、キメラ受容体のリガンドであるリコンビナントBAFFと、参考用にリコンビナントAPRILとを用い、試験例1と同様にして、レポーター細胞の刺激及びそれによるレポーター活性を測定した。
試験例1、2、3、4で得られた各レポーター細胞(HEK−BlueTMIFN−α/β細胞、HEK−Blue TACI細胞、及び(1)のDNAデザインによる受容体を発現しているHEK−Blue BAFFR−BCMA細胞)を、96ウェルプレートに2.5x104細胞ずつ播種し、24時間培養した。コンプリート培地100μlに健康人(HC)10名又はSLE患者29名の血清20μlを加えたサンプルを添加し、さらに24時間培養した。培養後、上清を回収し、SEAPの発色基質であるQUANTI−Blueを加えて30分間37℃で反応させ、吸光度計を用いて620nm吸収波長を測定した。得られたSEPA活性を、SLE憎悪因子活性(IFN−I活性、BAFF/APRIL活性、及びBAFF活性)として評価した。
配列番号5は、BAFFRの1−234bp領域と、BCMAの154−162bp領域と、BCMAの163−555bp領域とを結合したキメラ配列である。
配列番号6〜15は、プライマーである。
Claims (10)
- IFN−I受容体を発現しており、IFN−I誘導性制御領域と、前記制御領域の下流に連結されたレポーター遺伝子とを含むレポーター構築物が導入された細胞を含む、全身性エリトマトーデスの治療薬に対するコンパニオン診断薬。
- 前記細胞がヒト胎児腎細胞である、請求項1に記載のコンパニオン診断薬。
- BAFF/APRIL受容体又はBAFF受容体を発現しており、BAFF/APRIL誘導性制御領域又はBAFF誘導性制御領域と、前記制御領域の下流に連結されたレポーター遺伝子とを含むレポーター構築物が導入された細胞を含む、全身性エリトマトーデスの治療薬に対するコンパニオン診断薬。
- 前記BAFF/APRIL受容体が、BCMA、TACI及びBAFF−Rからなる群より選択される、請求項3に記載のコンパニオン診断薬。
- 前記BAFF受容体が、BAFF−Rの細胞外領域とBCMAの細胞内領域とを含む融合タンパク質である、請求項3及び4に記載のコンパニオン診断薬。
- 前記細胞がヒト胎児腎細胞である、請求項3〜5のいずれかに記載のコンパニオン診断薬。
- 請求項1又は2に記載のコンパニオン診断薬と、請求項3〜6のいずれかに記載のコンパニオン診断薬とを含む、全身性エリトマトーデスの治療薬に対するコンパニオン診断キット。
- 被験動物から得られた生体試料について、請求項1又は2に記載のコンパニオン診断薬を用いて測定されるレポーター活性値を、IFN−I活性値として取得する工程1と、
前記生体試料について、請求項3〜6のいずれかに記載のコンパニオン診断薬を用いて測定されるレポーター活性値を、BAFF活性値又はBAFF及びAPRIL活性値として取得する工程2と、
前記IFN−I活性値並びに前記BAFF活性値又はBAFF及びAPRIL活性値に基づき、前記生体試料中のIFN−IとBAFF又はBAFF及びAPRILとのうち、相対的に多く存在する全身性エリトマトーデス憎悪因子を決定する工程3と、を含む、生体試料の分析方法。 - 前記工程3が、
