培養後、1%FBS含有DMEM培地で、所定濃度になるように溶解した被験試料を各wellに200μLずつ添加し、24時間培養した。被験試料を添加し1%FBS含有DMEM培地を添加したサンプルをコントロールとした。培養終了後、各wellから培地を抜き、400μLのPBS(−)にて洗浄後、150μLのM−PER(R)(Thermo Fisher SCIENTIFIC社)を用いて細胞を溶解した。このうちの100μLを用いて総グルタチオンの定量を行った。すなわち、96wellプレートに溶解した細胞抽出液100μL、0.1mol/Lリン酸緩衝液50μL、2mol/LのNADPH25μLおよびグルタチオンレダクターゼ25μL(終濃度17.5unit/mL)を加え、37℃で10分間加温した後、10mmol/L 5,5’−dithiobis(2−nitrobenzonic acid)25μLを加え、5分後までの波長412nmにおける吸光度を測定し、ΔOD(光学濃度)/minを求めた。総グルタチオン濃度は酸化型グルタチオンを用いて作成した検量線をもとに算出した。得られた値は総タンパク量当たりのグルタチオン量に補正した後、下記式に従い、グルタチオン産生促進率を算出した。試験は、各試料n=6で行った。
(Test method)
Normal human skin fibroblasts (NB1RGB) were cultured in α-MEM medium containing 10% FBS (fetal bovine serum), and then the cells were recovered by trypsin treatment. The collected cells were diluted with α-MEM medium containing 10% FBS to a concentration of 2.0 × 10 5 cells / mL, 200 μL per well was seeded on a 48-well plate, and the cells were cultured overnight.
After culturing, 200 μL of the test sample dissolved in DMEM medium containing 1% FBS to a predetermined concentration was added to each well, and the cells were cultured for 24 hours. The sample to which the test sample was added and the DMEM medium containing 1% FBS was added was used as a control. After completion of the culture, the medium was removed from each well, washed with 400 μL of PBS (−), and the cells were lysed with 150 μL of M-PER (R) (Thermo Fisher SCIENTIFIC). Total glutathione was quantified using 100 μL of this. That is, 100 μL of cell extract dissolved in a 96-well plate, 50 μL of 0.1 mol / L phosphate buffer, 25 μL of NADPH of 2 mol / L and 25 μL of glutathione reductase (final concentration 17.5 units / mL) were added, and the mixture was added at 37 ° C. for 10 minutes. After warming, 25 μL of 10 mmol / L 5,5'-diziobis (2-nitrobenzonic acid) was added, and the absorbance at a wavelength of 412 nm up to 5 minutes later was measured to determine ΔOD (optical concentration) / min. The total glutathione concentration was calculated based on the calibration curve prepared using oxidized glutathione. The obtained value was corrected to the amount of glutathione per total protein amount, and then the glutathione production promotion rate was calculated according to the following formula. The test was performed on each sample n = 6.
グルタチオン産生促進率は、以下の式より算出した。平均値を表1に示す。

プトレシン、スペルミジン、スペルミン、アセチルスペルミジン、ジアセチルスペルミジン、アセチルスペルミンにグルタチオン産生促進作用が確認できた。
