Abstract
Description
本出願は、2017年7月13日出願の米国仮特許出願第62/532,246号の優先権を主張する。
a)iPSCおよび/またはESCを、コーティングされた非組織処理培養プレートに蒔くステップと、
b)1つまたは複数の細胞運命因子をコードするmRNAを個別のmRNAまたは組み合わせたmRNAとしてステップa)の細胞にトランスフェクションするステップと、
c)ステップb)の前記iPSCおよび/またはESCを、細胞がNPCに分化するまで培養するステップと、
d)多数のNPCを必要とする場合、細胞をコーティングされていない超低接着条件に移して懸濁液中に成長させることにより、c)のNPCを増やし得るステップと

を含む。本方法の実施形態では、ステップa)において、6ウェルプレートのウェルには、約0.75×105細胞/ウェル〜約1.5×106細胞/ウェルで播種する。他のサイズの培養容器を使用することもできる。6ウェルプレート以外の培養容器のための播種密度は、約70,000細胞〜約14,000,000細胞を使用し、細胞数を培養容器の成長面積で割ることに基づいて、例えば、表2に示すように決定することができる。別の実施形態では、ステップb)のトランスフェクションに使用するmRNAは、Oligo2、NKX2.2、SOX10およびOligo1のmRNAからなる群のうちの1つまたは複数から個別にまたは組み合わせて選択される細胞運命因子をコードする。別の実施形態では、ステップb)におけるトランスフェクションのための用量は、6ウェル培養プレートを使用する場合、1細胞運命因子あたり約20ng〜約100ngであるか、または他のタイプのプレートもしくは培養フラスコを使用する場合、容器の面積もしくは培養培地量の容量に基づいて、比例的に調整する。別の実施形態では、最初のトランスフェクション後、トランスフェクションは、6ウェル培養プレートを使用する場合、約10ng/細胞運命因子〜約200ng/細胞運命因子のmRNAの用量で少なくともさらに2回反復するか、または他のタイプのプレートもしくは培養フラスコを使用する場合、容器の面積もしくは培養培地量の容量に基づいて、比例的に調整する。別の実施形態では、培養容器プレートは、マトリゲルおよび/またはPLO−ラミニンでコーティングされている。別の実施形態では、ステップa)〜c)に使用する培地は、DMEM/F12、DMEMおよびMEMaから選択される。一部の実施形態では、培地には、KSR、アスコルビン酸、SAG、PDGF、HGF、IGF1、T3、インスリン、RA、B27、CAMPおよび/またはビオチンのうちの1つまたは複数を補充する。本方法の実施形態では、ステップa)〜d)の培養条件は、約2%〜約6%の酸素濃度におけるものである。別の実施形態では、ステップc)の細胞は、OPCの形態特性を有する細胞が観察されるまで約4〜約20日間培養する。別の実施形態では、OPC生成は、双極性形態および/または細胞運命因子のうちの1つの発現に基づいて確認することができる。約4〜約20日後、約100%に近いが約75%よりも多くの細胞が、OPC細胞特有の形態を有する。一部の実施形態では、約40〜60%の細胞が、OPC細胞特有の形態を有する。約40%よりも少ない細胞がOPC細胞特有の形態を有する場合、細胞選別を使用して細胞を単離することができる。懸濁培養システムを使用してさらに培養すると、さらなる選択が可能となる。一部の実施形態では、OPCの確認は、細胞運命因子のうちの1つ、例えば、Oligo2またはSox10の発現により示すことができる。

K)オリゴデンドロサイト細胞が、樹状分枝様突起(tree branch-looking process)を伴って出現する、および/またはミエリン塩基性タンパク質(MBP)を生成するまで、少なくとも約1週間、細胞を培養するステップと
を使用して逐次的または直接的に誘導するための方法である。
的組織細胞を多能性幹細胞から誘導するための最新のプロトコールでは、医学的に適用可能なNPC、OPCまたはオリゴデンドロサイトを生成する、分化カスケードに沿った各ステップにおいて、成長因子、ホルモン、サイトカイン、シグナルペプチドおよび他の細胞間シグナル分子の組合せ(本開示においては便宜上まとめて「成長因子」と呼ぶ)の使用を必要とする。残念ながら、精製したポリペプチドとして供給した場合、成長因子は、一般に高価で不安定であり、バッチ毎に一貫しないため、これらの使用は困難なものとなっている。その上、成長因子が高度に特異的かつ組合せ的方法で機能するため、分化の各段階は、様々な成長因子のセットにより規定され、これら成長因子のセットは、最適化が困難かつコストが高い。本明細書に開示するように、重要な運命変化点において適切な用量および送達条件でmRNAを組み合わせることにより、大量の成長因子を使用することなく、非常に高い効率かつ低いコストで、NPC、OPCおよびオリゴデンドロサイトを実現することができる。

c)ステップb)の前記iPSCおよび/またはESCを、前記細胞がNPCに分化するまで培養するステップと、
d)前記NPCをステップc)から単離するステップと、
e)コーティングされた細胞培養容器にNPCを蒔くステップであって、KSR、ならびにアスコルビン酸、SAG、PDGF、HGF、IGF1、T3、インスリン、RA、B27、CAMPおよびビオチンからなる群から選択される培地サプリメントのうちの1つまたは複数をいずれも有するMEMaまたはDMEM/F12またはDMEMを含む培地中に、前記NPCを、1培養フラスコまたは1培養ウェルプレートあたり約70,000細胞〜約14,000,000細胞を使用して播種するステップと、

g)ステップf)の前記トランスフェクションを少なくともさらに2回反復するステップと、
h)ステップg)の前記細胞を、前記細胞の形態がNPC細胞からOPC細胞の形態に変化するまで培養するステップと、
i)前記OPCをステップh)から単離するステップと、
j)ステップh)から単離したOPCをコーティングされた細胞培養容器に蒔くステップであって、KSR、ならびにアスコルビン酸、N2、B27、ビオチン、Cam、T3およびインスリンからなる群から選択される培地サプリメントのうちの1つまたは複数をいずれも有するMEMaまたはDMEM/F12またはDMEMを含む培地中に、前記OPCを、1培養フラスコまたは1培養ウェルプレートあたり約70,000細胞〜約14,000,000細胞を使用して播種するステップと、
k)オリゴデンドロサイト細胞が、樹状分枝様突起を伴って出現する、および/またはミエリン塩基性タンパク質(MBP)を生成するまで、少なくとも約1週間、前記細胞を培養するステップと
を含む、方法。

- 人工多能性幹細胞(iPSC)および/または胚性幹細胞(ESC)の神経前駆細胞(NPC)への分化を誘導するための方法であって、
a)iPSCおよび/またはESCを、培地を含むコーティングされた非組織処理培養容器に蒔くステップと、
b)1つまたは複数の細胞運命因子をコードするmRNAを個別のmRNAまたは組み合わせたmRNAとしてステップa)の前記細胞にトランスフェクションするステップと、
c)ステップb)の前記iPSCおよび/またはESCを、前記細胞がNPCに分化するまで培養するステップと
を含む、方法。 - ステップb)の前記細胞運命因子が、Ngn2、Pax6、Sox2、Brn2およびFoxOファミリー因子のうちの1つまたは複数からなる群から、個別のmRNAまたは組み合わせたmRNAトランスフェクションとして選択される、請求項1に記載の方法。
- ステップb)の前記細胞運命因子が、Ngn2である、請求項2に記載の方法。
- ステップb)の前記細胞運命因子が、Pax6である、請求項2に記載の方法。
- ステップb)の前記細胞運命因子が、Sox2である、請求項2に記載の方法。
- ステップb)の前記細胞運命因子が、Brn2である、請求項2に記載の方法。
- ステップb)の前記細胞運命因子が、FoxOファミリー因子である、請求項2に記載の方法。
- ステップb)の前記細胞運命因子のうちの1つまたは複数が、トランス活性化ドメインと融合している、請求項2に記載の方法。
- ステップc)の前記細胞を、前記細胞がステップc)の前記細胞運命因子のうちの1つまたは複数を発現するまで培養する、請求項1に記載の方法。

a)培地を含むコーティングされた細胞培養容器にNPCを蒔くステップと、
b)1つまたは複数の細胞運命因子をコードするmRNAを個別のmRNAまたは組み合わせたmRNAとしてステップa)の前記細胞にトランスフェクションして、神経前駆細胞をオリゴデンドロサイト前駆細胞に分化させるステップと、
c)ステップc)の前記細胞を、前記NPCの形態がOPCの形態に変化するまで培養するステップと
を含む、方法。 - ステップa)の前記NPCを、1培養フラスコまたは1培養ウェルプレートあたり約70,000細胞〜約14,000,000細胞を使用して播種する、請求項19に記載の方法。
- ステップb)のトランスフェクションに使用する前記mRNAが、Oligo2、NKX2.2、SOX10およびOligo1のmRNAからなる群のうちの1つまたは複数から個別にまたは組み合わせて選択される細胞運命因子をコードする、請求項19に記載の方法。
- 前記mRNAが、Oligo2をコードする、請求項21に記載の方法。
- 前記mRNAが、NKX2.2をコードする、請求項21に記載の方法。
- 前記mRNAが、Sox10をコードする、請求項21に記載の方法。
- 前記mRNAが、Oligo1をコードする、請求項21に記載の方法。
- 6ウェル培養プレートを使用し、ステップb)の前記トランスフェクションのための用量が1細胞運命因子あたり約20ngである、請求項19に記載の方法。

e)コーティングされた細胞培養容器にNPCを蒔くステップであって、KSR、ならびにアスコルビン酸、SAG、PDGF、HGF、IGF1、T3、インスリン、RA、B27、CAMPおよびビオチンからなる群から選択される培地サプリメントのうちの1つまたは複数をいずれも有するMEMaまたはDMEM/F12またはDMEMを含む培地中に、前記NPCを、1培養フラスコまたは1培養ウェルプレートあたり約70,000細胞〜約14,000,000細胞を使用して播種するステップと、
f)Oligo2、NKX2.2、SOX10およびOligo1のmRNAからなる群のうちの1つまたは複数を個別にまたは組み合わせて、トランスフェクション試薬を使用して、ステップe)の前記細胞にトランスフェクションするステップと、
g)ステップe)の前記トランスフェクションを少なくともさらに2回反復するステップと、
h)ステップg)の前記細胞を、前記細胞の形態がNPC細胞からOPC細胞の形態に変化するまで培養するステップと、
i)前記OPCをステップh)から単離するステップと、
j)ステップj)から単離したOPCをコーティングされた細胞培養容器に蒔くステップであって、KSR、ならびにアスコルビン酸、N2、B27、ビオチン、Cam、T3およびインスリンからなる群から選択される培地サプリメントのうちの1つまたは複数をいずれも有するMEMaまたはDMEM/F12またはDMEMを含む培地中に、前記OPCを、1培養フラスコまたは1培養ウェルプレートあたり約70,000細胞〜約14,000,000細胞を使用して播種するステップと、
K)オリゴデンドロサイト細胞が、樹状分枝様突起を伴って出現する、および/またはミエリン塩基性タンパク質(MBP)を生成するまで、少なくとも約1週間、前記細胞を培養するステップと
を含む、方法。 - 前記NPCが、レシピエント対象に由来するものである、請求項1、19、41または42に記載の方法。
- 開始細胞が体液または体組織から採取されたものである、請求項1、19、41または42に記載の方法。
- 開始細胞がレシピエントから採取されたものである、請求項1、19、41または42に記載の方法。
