(2R,4R,5R)-4-(ベンジロキシ)-5-((ベンジロキシ)メチル)-3,3-ジフルオロテトラヒドロフラン-2-イル メタンスルホネート
本発明の化合物をもたらす設計原理は、以下のように要約することができる。進歩した従来のフルオロピリミジンであるカペシタビンのチミジンホスホリラーゼ(TP)による活性化は、いくつかの癌がこの酵素を過剰発現するので、改善として一般的に受け入れられている。一般的な見解に反して、TPのための基質活性を設計することはそれが、高TP発現の癌を有する患者のみならず、全ての患者に良好な影響を有する、毒性副作用の大部分の原因であるFUの生成を妨げるので、より有利であり得ると仮定された。最も電気陰性の原子であるフッ素を類似体の2'位に配置すると、隣接する1'-炭素に正電荷密度を有するTP反応の遷移状態が不安定になると考えられた(Schwartz, P.A.等、Journal of American Chemical Society 2010、132、14425参照)。さらに、別のフッ素を添加すると、より大きな効果が得られると考えられた。しかしながら、TPの関与を排除すると、類似体の代謝活性化が妨げられるため、5'位に-OH基を付加し、キナーゼ介在性リン酸化による活性化の代替経路を可能にすることが決定された(図2参照)。
式中、Rは直鎖または分枝鎖アルキル基(C1-7)であり; R'はH、ヒドロキシ保護基であり; R"はH、リン酸エステル、アミノ酸アルキル(C1-7)エステルホスホラミデート、またはホスホロジアミデートである;またはその薬学的に許容される塩を提供する。一例では、R”が一リン酸、二リン酸、三リン酸またはホスホラミデートであっても良い。

本発明のいくつかの細胞内標的は、DNAの生合成および機能に必要とされる必須酵素として同定された。これらの酵素は、本発明の多数の細胞代謝産物による阻害に対して感受性であり、観察された細胞毒性を生じる。酵素阻害活性を担う細胞内代謝物はすべてリン酸化誘導体であり、以下のものであることが見いだされた。2',2',5-トリフルオロデオキシシチジン一リン酸、F3dCMP、デオキシシチジル酸デアミナーゼ(DCTD)の阻害剤;2',2',5-トリフルオロデオキシウリジン一リン酸、F3dUMP、チミジル酸シンターゼ(TS)の阻害剤;2',2',5-トリフルオロデオキシシチジン二リン酸、リボヌクレオチドレダクターゼ(RR)のF3dCDP阻害剤;2',2',5-トリフルオロデオキシチジン三リン酸、F3dCTP、DNAポリメラーゼα(Pol α)の阻害剤;2',2',5-トリフルオロデオキシウリジン三リン酸、F3dUTP、DNAポリメラーゼα(Pol α) の阻害剤、5-フルオロシトシン、DNAに組み込まれたFC、DNA(シトシン-5)-メチルトランスフェラーゼ(MTase) の阻害剤。これらを表1に要約する。
実施例4
この実施例は、N4-ペンチルオキシカルボニル-2'-デオキシ-2',2',5-トリフルオロシチジン(6a)((ペンチル(1-((2R,4R,5R)-3,3-ジフルオロ-4-ヒドロキシ-5-(ヒドロキシメチル)テトラヒドロフラン-2-イル)-5-フルオロ-2-オキソ-1,2-ジヒドロピリミジン-4-イル)カルバメート))の合成の説明を提供する。
この実施例は、N4-ペンチルオキシカルボニル-2'-デオキシ-2',2',5-トリフルオロシチジン(6a)((ペンチル(1-((2R,4R,5R)-3,3-ジフルオロ-4-ヒドロキシ-5-(ヒドロキシメチル)テトラヒドロフラン-2-イル)-5-フルオロ-2-オキソ-1,2-ジヒドロピリミジン-4-イル)カルバメート))の合成の説明を提供する。
Claims (13)
- 前記ヒドロキシ保護基が、アセチルおよびベンゾイルからなるグループより選ばれたものであることを特徴とする請求項1に記載の化合物。
- 前記R"が、一リン酸、二リン酸、三リン酸、またはホスホラミデートであることを特徴とする請求項1に記載の化合物。
- 請求項1の化合物と、薬学的に許容されるキャリアを含むことを特徴とする組成物。
- 請求項5の組成物を個体に投与することを含む、癌に罹った個体を治療するための方法であって、前記組成物の投与が、当該個体における癌の増殖の阻害をもたらすことを特徴とする方法。
- 前記の個体が、ヒトまたは非ヒト哺乳動物であることを特徴とする請求項7に記載の方法。
- 前記の癌が、腎臓腺癌、急性骨髄性白血病、結腸癌、非小細胞肺癌、転移性悪性黒色腫、乳腺癌、膵臓腺癌、及び卵巣癌からなるグループより選ばれたものであることを特徴とする請求項7に記載の方法。
- 癌細胞におけるDNA合成が阻害されることを特徴とする請求項7に記載の方法。
- リボヌクレオチドレダクターゼ、DNAポリメラーゼα、チミジル酸シンターゼ、DNA(シトシン-5)-メチルトランスフェラーゼ、またはそれらの組み合わせが阻害されることを特徴とする請求項7に記載の方法。
- 前記方法が、DNAへの類似体誤取り込みを引き起こし、DNAの構造および機能の変化を生じさせ、癌細胞の死をもたらすことを特徴とする請求項7に記載の方法。
