Abstract
Description
ここでの例は、微生物ロドプシンのNanohaloarchaea由来の、まだ特徴づけられていないキセノロドプシンの代表的な包括的な機能研究を示し、それらが内向きプロトンポンプであることを示す。モデル膜システム、大腸菌細胞、ヒト胚性腎細胞、神経芽腫グリオーマ細胞及びラット海馬神経細胞におけるナノサリナ由来のキセノロドプシン(NsXeR)のポンピング活性の厳密な研究により、これら全ての細胞でNsXeRが内向きポンプとして機能することが確認された。また、NsXeRは、代謝回転速度が、400s−1の強力なポンプであり、ラット海馬神経細胞の最大内因性発火頻度まで活動電位を引き出すことが可能であることも実証されている。NsXeRの結晶構造は、既知のロドプシンとは非常に異なるイオン活動経路を明らかにする。プロトンポンプとしての固有の性質により、NsXeRは、イオン条件に完全に依存しないため、このロドプシンは、良く知られているカチオン選択性チャネルとして、ニューロンの光誘起リモートコントロールの興味深い代替品となる。

12.pH7.3に滴定された、129mMグルコン酸カリウム、10mM HEPES、10mM KCl、4mM MgATP及び0.3mM Na3GTPで充填された抵抗3〜8MΩのパッチピペット、及びpH7.3に滴定された、125mM NaCl、2mM KCl、2mM CaCl2、1mM MgCl2、1mM MgCl2、30mMグルコース及び25mM HEPESを含む細胞外溶液を用いた細胞全体の構成において、パッチクランプ測定によりラット海馬ニューロンにおいて測定した場合、前記光駆動内向きプロトンポンプが、3msのλ=532nmのパルス幅及び23mW/mm2の強度で誘発される得る、実施形態1〜11の何れか1つに記載の使用のための光駆動内向きプロトンポンプ。
13.核酸コンストラクトであって、ヌクレオチド配列が、ヒト細胞での発現のためにコドン最適化されており; ここで、前記ヌクレオチド配列が、配列番号16に示される配列を有する、実施形態1〜12の何れか1つで定義される光駆動内向きプロトンポンプをコードするヌクレオチド配列を含む核酸コンストラクト。

23.実施形態17〜19の何れか1つに記載の細胞を含む、非ヒト哺乳動物であって、好ましくはここで、前記細胞が、内因性の細胞である; ただし、人間や動物に対して何れかの動物の苦しみを上回る利益を実質的にもたらさない可能性がある動物は、除外される、非ヒト哺乳動物。

大腸菌懸濁液中でのpH変化
NsXeR(Uniprot ID G0QG75)、HrvXeR(Ghai, R. et al. Sci. Rep. 1,(2011))及びAlkXeR(Vavourakis, C. D. et al. Front. Microbiol. 7,(2016))、のコーディングDNAが、商業的に合成された(Eurofins)。ヌクレオチド配列は、GeneOptimizer(商標)ソフトウェア(Life Technologies、USA)を使用して、大腸菌発現用に最適化された。5’リボソーム結合部位及び追加のLEHHHHHH*タグをコードする3’伸長を有する遺伝子が、XbaI及びBamHI制限酵素部位を介してpET15b発現ベクター(Novagen)に導入された。
タンパク質は、上記のように発現された。しかしながら、誘導の3時間後、細胞は、3,000gで30分の遠心分離により採集された。採集された細胞は、20mM Tris−HCl(pH8.0)、5%グリセロール、0.5% TritonX−100(Sigma−Aldrich、USA)及び50mg/L DNaseI(Sigma−Aldrich、USA)を含む緩衝液中で25,000psiでM−110P Lab Homogenizer(Microfluidics、USA)で破壊された。細胞溶解物の膜画分は、4℃で1時間、90,000gでの超遠心分離によって分離された。ペレットは、50mM NaH2PO4/Na2HPO4(pH8.0)、0.1M NaCl及び1%DDM(Anatrace、Affymetrix、USA)を含む緩衝液に再懸濁され、可溶化のために一晩攪拌された。不溶性画分は、90,000g、4℃で1時間の超遠心分離によって除去された。上清は、Ni−NTAカラム(Qiagen、Germany)に充填され、キセノロドプシンが、50mM NaH2PO4/Na2HPO4(pH7.5)、0.1M NaCl、0.3Mイミダゾール及び0.2%DDMを含む緩衝液で溶出した。溶出液は、100容量の50mMNaH2PO4/Na2HPO4(pH7.5)、0.1M NaCl緩衝液に対して2時間2回透析し、イミダゾール処理を行った。
ここでは、2つのデータセットの分析結果が報告される: XeRタンパク質は、ナノディスク及びリポソームで再構成される。プロテオナノディスクは、標準プロトコルを用いて再構成された(Ritchie, T. K. et al. in Methods in Enzymology(ed. Duzgunes,N.)464, 211-231(Academic Press, 2009);参照により本明細書に組み込まれる)。1,2−ジミリストイル−sn−グリセロ−3−ホスホコリン、DMPC(Avanti Polar Lipids、USA)が脂質として用いられた。アポリポタンパク質−1の細長いMSP1E3が使用された。アセンブリ中のモル比は、DMPC:MSP1E3:NsXeR=100:2:3であった。リポソームは、上記のように調製された。
NsXeRタンパク質は、実施例1に記載されるように調製及び精製された。最終的に、タンパク質は、結晶化のために70mg/mlに濃縮された。NsXeR結晶は、以前の研究で使用されていたものと同様の(Gordeliy, V. I. et al. Molecular basis of transmembrane signalling by sensory rhodopsin II-transducer complex. Nature 419, 484-487 (2002);参照により本明細書に組み込まれる)メソアプローチ(Landau, E. M. & Rosenbusch, J. P. Lipidic cubic phases: A novel concept for the crystallization of membrane proteins. Proc. Natl. Acad. Sci. 93, 14532-14535 (1996); 及びCaffrey, M. & Cherezov, V. Crystallizing membrane proteins using lipidic mesophases. Nat. Protoc. 4, 706-731 (2009); それぞれ、参照により本明細書に組み込まれる)で成長した。結晶化緩衝液中の可溶化タンパク質は、47℃であらかじめ融解されたモノウンデセノイン(Nu−Chek Prep)と混合して、脂質中間相を形成した。タンパク質−中間相混合物の100nlアリコートが、96ウェルLCPガラスサンドイッチプレート(Marienfeld)にスポットされ、NT8結晶化ロボット(Formulatrix)により600nLの沈殿剤溶液で覆った。20mg/mlのタンパク質濃度及び2.0Mマロン酸ナトリウム、pH8.0(Hampton Research)で最良の結晶が得られた。結晶は、22℃で成長し、1〜4週間で出現した。
レチナールは、13−cis構造である。しかしながら、分解能が不十分であるため、15−syn又は15−anti構造化を区別することができない。NsXeRは、大きなプロトン取り込み空洞があり、これは、タンパク質の細胞外部部分にN末端の非常に短いヘリックスを有するバルクから単離される。空洞は、水分子で満たされていることを提案する。推定のプロトンドナーAsp76は、その空洞から利用できる可能性がある。Asp76のGlu、Ser、Thr及びAsnへの突然変異により、タンパク質が正しく折り畳まれない(突然変異体は、着色されていない。上記の表を参照)。これは、機能的だけでなく、これらのアミノ酸の重要な構造的役割の証拠である。Ser55は、Asp76の近くにあり、この残基を安定化する可能性がある。Ser55のアラニン(BRにおけるAla53)の置換もタンパク質の折り畳みを破壊する。
NsXeRと他の既知の微生物レチナールプロトンポンプの他の大きな違いは、BRのAsp96(NsXeRでは、Ala71である)に相当する位置に荷電アミノ酸が無いことである。しかしながら、残基His48(基底状態のSchif塩基から10Å)及びAsp220(12Å)は、水素結合を介して接続されており、予想されるプロトンポンプ受容体位置の近くに位置する。Asp220をAsnに置き換えると、プロトンポンプが完全に機能しない。
突然変異アミノ酸での構造及び実験は、内向きプロトン輸送の機構への見識を提供する。照射すると、レチナールが異性化し、疎水性環境に囲まれたSchiff塩基が脱プロトン化し、プロトンが脱プロトン化されたHis48−Asp220に移行する。両方の中間体がレチナールSchiff塩基の脱プロトン化状態に対応するため、MI及びMII中間状態で起こる。実際、Asp−His相互作用は、その脱プロトン化状態を安定化させることにより、AspのpKaを大幅に低下させることが知られている。プロトン転位におけるAsp−His対の重要な役割は、突然変異Asp及びHisを用いた上記の実験によって裏付けられる。レチナールの再異性化後、親水性空洞(「プロトン放出空洞」)に接続されたプロトン化されたAsp−His対が、細胞質に直接プロトンを放出することを提案する。レチナールの異性化後、Asp76は、親水性空洞を介してプロトン化する。レチナールの再異性化は、D76からSchiff塩基の再プロトン化をもたらす。
ヒト胎児腎細胞(HEK)及び神経芽細胞腫神経膠腫細胞(NG)を用いた実験
ヒトコドン最適化NsXeR遺伝子は、商業的に合成された(Eurofins)。遺伝子は、付加的な膜輸送シグナル(Gradinaru, V. et al. Molecular and Cellular Approaches for Diversifying and Extending Optogenetics. Cell 141, 154-165 (2010); 参照により本明細書に組み込まれる)、P2A自己切断ペプチド(Kuzmich, A. I., Vvedenskii, A. V., Kopantzev, E. P. & Vinogradova, T. V. Quantitative comparison of gene co-expression in a bicistronic vector harboring IRES or coding sequence of porcine teschovirus 2A peptide. Russ. J. Bioorganic Chem. 39, 406-416 (2013); 参照により本明細書に組み込まれる)及びC末端のGFPバリアント(Shcherbo, D. et al. Bright far-red fluorescent protein for whole-body imaging. Nat. Methods 4, 741-746 (2007); 参照により本明細書に組み込まれる)を有するpcDNA3.1(−)ベクターにクローニングされた。遺伝子は、CMVプロモーターの下でクローン化された。配列は、シーケンシングによって検証された。
アデノ随伴ウイルスを介した遺伝子導入により、ラット海馬ニューロンでNsXeRが異種発現された。海馬は、出生後のP1Sprague−Dawleyラットから単離され、パパイン(20U ml−1)、37℃で20分間処理された。海馬は、10%ウシ胎児血清を補充したDMEM(Invitrogen/Gibco、高グルコース)で洗浄され、少量のこの溶液で滴定された。24ウェルプレートのポリ−D−リジン/ラミニンコーティングガラスカバースリップに〜96,000個の細胞が播種された。3時間後、プレーティング培地を培養培地(2%B−27サプリメントを含むNeurobasal A、及び2mM Glutamax−I)に置き換えた。
Hernandez達は、げっ歯類の聴覚経路の光遺伝学的刺激の戦略を証明している(Hernandez et al. - 前記光駆動内向きプロトンポンプが、配列番号1(NsXeR)、2(HrvXeR1)、9(HrvXeR)、10(AlkXeR)、11(AlkXeR1)、12(AlkXeR2)、13(AlkXeR3)、14(AlkXeR4)、及び15(AlkXeR5)を含み、好ましくは、からなり;特にここで、前記光駆動内向きプロトンポンプが、配列番号1(NsXeR)のアミノ酸配列を含む、請求項1に記載の使用のための光駆動内向きプロトンポンプ。
- (i)前記光駆動内向きプロトンポンプが、pH6〜pH8の間; 好ましくはpH5〜pH9の間で活動的である; 及び/又は
(ii)前記光駆動内向きプロトンポンプの最大吸収が、560nm〜580nmの間である、
(iii)20℃、pH7.5、532nmの波長で5nsの持続時間のパルス及び3mJ/パルスのエネルギーの供給で、100:2:3の比のDMPC:MSP1E3:光駆動内向きプロトンポンプのモル比を示すプロテオナノディスクで測定した場合、前記光駆動内向きプロトンポンプの光サイクルが、50ms未満、好ましくは45ms未満、より好ましくは40ms未満、より好ましくは35ms未満、さらにより好ましくは、27ms等の30ms未満である; 及び/又は
(iv)pH7.3に滴定された、129mM グルコン酸カリウム、10mM HEPES、10mM KCl、4mM MgATP及び0.3mM Na3GTPで充填された抵抗3〜8MΩのパッチピペット、及びpH7.3に滴定された、125mM NaCl、2mM KCl、2mM CaCl2、1mM MgCl2、30mMグルコース及び25mM HEPESを含む細胞外溶液を用いた細胞全体の構成において、パッチクランプ測定によりラット海馬ニューロンにおいて測定した場合、前記光駆動内向きプロトンポンプが、250s−1超、好ましくは300s−1超、より好ましくは370s−1超、より好ましくは380s−1超、より好ましくは390s−1超、例えば400s−1の代謝回転速度を有する; 及び/又は
(v)pH7.3に滴定された、129mMグルコン酸カリウム、10mM HEPES、10mM KCl、4mM MgATP及び0.3mM Na3GTPで充填された抵抗3〜8MΩのパッチピペット、及びpH7.3に滴定された、125mM NaCl、2mM KCl、2mM CaCl2、1mM MgCl2、1mM MgCl2、30mMグルコース及び25mM HEPESを含む細胞外溶液を用いた細胞全体の構成において、パッチクランプ測定によりラット海馬ニューロンにおいて測定した場合、前記光駆動内向きプロトンポンプが、40Hzを超える周波数、好ましくは50Hzを超える周波数、より好ましくは60Hzを超える周波数、さらにより好ましくは70Hzを超える周波数、最も好ましくは、80Hzを超える周波数で活動電位を誘発することができる; 及び/又は
(vi)pH7.3に滴定された、129mMグルコン酸カリウム、10mM HEPES、10mM KCl、4mM MgATP及び0.3mM Na3GTPで充填された抵抗3〜8MΩのパッチピペット、及びpH7.3に滴定された、125mM NaCl、2mM KCl、2mM CaCl2、1mM MgCl2、1mM MgCl2、30mMグルコース及び25mM HEPESを含む細胞外溶液を用いた細胞全体の構成において、パッチクランプ測定によりラット海馬ニューロンにおいて測定した場合、前記光駆動内向きプロトンポンプが、3msのλ=532nmのパルス幅及び23mW/mm2の強度で誘発され得る、
請求項1〜4の何れか1項に記載の使用のための光駆動内向きプロトンポンプ。 - 請求項1〜5の何れか1項に定義される光駆動内向きプロトンポンプをコードする核酸配列を含む核酸コンストラクトであって、前記核酸配列が、ヒト細胞での発現のためにコドン最適化されている、核酸コンストラクト。
- 請求項1〜5の何れか1項に定義される光駆動内向きプロトンポンプをコードする核酸配列又は請求項6に記載の核酸コンストラクトを含む発現ベクターであって、前記核酸配列が、ヒト細胞での発現のために最適化されており; 特に、前記ベクターは、ウイルスベクターであり; 及び/又は前記光駆動内向きプロトンポンプのコード配列が、神経細胞特異的なヒトプロモーター、好ましくは、ヒトシナプシンプロモーターの制御下にある、発現ベクター。
- 請求項1〜5の何れか1項に定義される光駆動内向きプロトンポンプを発現する哺乳動物細胞であって、ただし、前記哺乳動物細胞が、ヒト胚細胞又はヒトの生殖細胞系列の遺伝的同一性を改変するできる細胞ではない、哺乳動物細胞。
- 請求項6に記載の核酸コンストラクト又は請求項7に記載の発現ベクターを含む哺乳動物細胞。
- 前記細胞が、
(a)海馬細胞、視細胞、杆体細胞; 網膜錐体細胞; 網膜神経節細胞; 双極ニューロン; 神経節細胞; 疑似単極ニューロン; 多極ニューロン; 錐体ニューロン; プルキンエ細胞; 又は顆粒細胞; 又は
(b)神経芽腫細胞腫細胞、特に、NG108−15; HEK293細胞; COS細胞; BHK細胞; CHO細胞; 骨髄腫細胞; 又はMDCK細胞、
である請求項9に記載の哺乳動物細胞。 - 請求項1〜5の何れか1項に定義される光駆動内向きプロトンポンプを含むリポソーム。
- 医薬に使用するための請求項6に記載の核酸コンストラクト、請求項7に記載の発現ベクター、請求項8〜10の何れか1項に記載の哺乳動物細胞、又は請求項11に記載のリポソーム。
- 聴力の回復、視力の回復における使用、又はアルカローシス、神経損傷、脳損傷、脳卒中、又はパーキンソン病及びアルツハイマー病等の変性神経障害の治療又は軽減における使用のための、請求項1〜5の何れか1項に定義される光駆動内向きプロトンポンプ、請求項6に記載の核酸コンストラクト、請求項7に記載の発現ベクター、請求項8〜10の何れか1項に記載の哺乳動物細胞、又は請求項11に記載のリポソーム。
- 請求項8〜10の何れか1項に記載の細胞を含む、非ヒト哺乳動物であって、好ましくは、前記細胞が、内因性の細胞である; ただし、人間や動物に対して何れかの動物の苦しみを上回る利益を実質的にもたらさない可能性がある動物は、除外される、非ヒト哺乳動物。
- (i)電気的に興奮可能な細胞の光刺激のために、
(ii)プロトン濃度勾配に対して膜上でプロトンを輸送するために、
(iii)細胞、細胞画分、小胞、又はリポソームの内部を酸性化又はアルカリ化するために、又は
(iv)又は光遺伝学ツールとしての、
請求項1〜5の何れか1項に定義される光駆動内向きプロトンポンプの非治療的、又はex vivo、又はin vitro使用。
