Abstract
Description
(GB1からのGB2及びGB3から製造した)ヘパリンの領域選択的脱硫(region−selective desulfation)
領域選択的脱硫されたヘパリン(GB2及びGB3)の合成のために、ヘパリン(MW:14,000、Merck Millipore、生産者番号:375095,Germany、GB1)を先ず脱イオン化超純水の中に溶解させ、Amberlite IR−120H+イオン交換カラム(Sigma−Aldrich,Germany)を用いて、脱塩させた。そのヘパリン溶液に、pH6に達するまでピリジン(Sigma−Aldrich,Germany)を添加することによって、ヘパリン−ピリジン塩を形成させ、それを、B−490ロータリーエバポレーター(Buechi,Germany)中で溶媒蒸発させることによって濃縮し、次いで、−80℃で凍結乾燥させ(GEA Lyovac GT2,Germany)、且つさらなる加工をするまで、−20℃で保存した。
GB1〜GB3の分子量は、Dawn HELEOS II(Wyatt Technology Europe,Germany)上、690nmでの多角度光散乱法により求めた。分子量決定に必要なdn/dcの値は、RI検出器(Optilab T−rEX,Wyatt,Germany)を用い、λ=690nmの波長で測定した。GB1、GB2、及びGB3での、0.1351mL・g−1のRIの増分、dn/dcが、光散乱実験の評価には必要であった。モル質量を求めるための光散乱の結果は、Astraソフトウェア、バージョン6.1(Wyatt Technology,USA)を用いて実施した。
ヒドロゲルを調製するために、ヘパリン(GB1)(Merck−Millipore、生産者番号:375095,Germany)、GB1をベースとする脱硫ヘパリン誘導体を、GB2及びGB3のために、荷電された成分(GB)としてのヒドロゲルのタイプ(表2参照)に従った前述の方法によって、脱イオン化超純水の中に、ボルテックスミキサー(IKA,Germany)を用い、4℃で30秒間、500rpmで溶解させた。それに加えて、4本アームのアミン末端のPEG(MW、10,000、Jenkem Technology,USA)、1−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド(EDC;Sigma−Aldrich,Germany)、及びN−ヒドロキシスルホスクシンイミド(スルホ−NHS、Sigma−Aldrich,Germany)を、同様にして、すべてのタイプのヒドロゲルのために、UGB1として、溶解させた。ヒドロゲル構成単位の完全な溶解を確保するために、これらの溶液をさらに、超音波浴(RK 100H、Bandelin,Germany)を用い、4℃で5分かけて処理した。
4−スチレンスルホン酸:マレイン酸のモル比=3:1のポリ(4−スチレンスルホン酸−コ−マレイン酸)(Sigma−Aldrich、生産者番号:434566,Germany、GB4、表2参照)、及び4−スチレンスルホン酸:マレイン酸のモル比=1:1のもの(Sigma−Aldrich、生産者番号:434558,Germany、GB5、表2参照)、及びUGB1(四価のPEG、アミン末端、MW=10,000、Jenkem Technology,USA)を使用して、以下の方法で、共有結合的に架橋されたヒドロゲルを調製した:GB4又はGB5及びUGB1をそれぞれ、脱イオン化した超純水に、ヒドロゲル混合物の全容積の1/3ずつ、表1に見られるものの、3倍の濃度で溶解させ、さらに、超音波浴(RK 100H、Bandelin,Germany)を用いて、4℃で5分間処理した。その後で、全部の溶液を、次のステップのために4℃でなじませた(tempered):1−エチル−3−(3−ジメチルアミノプロピル)カルボジイミド(EDC、Sigma−Aldrich,Germany)及びN−ヒドロキシスルホスクシンイミド(スルホ−NHS;Sigma−Aldrich,Germany)もやはり、全反応混合物の1/6に溶解し、4℃でなじませた。UGB1:EDC:スルホNHSのモル比は、1:8:4である(UGB1のそれぞれのアミノ基では、1:2:1)。次のステップにおいて、GB4又はGB5にEDC及びスルホ−NHSを添加し、ピペッティングによって混合し、且つ30秒間の待ち時間の後に、その反応混合物を、ボルテックスシェーカー(VWR,Germany)で10秒間混合し、次いでまた別の待ち時間20秒をとった。この手順の後で、全活性化時間1分で、ボルテックスシェーカー上のUGB1を、活性化させたGB4又はGB5にピペット移植し、次いでまた別の10秒間、ボルテックスシェーカーを用いて混合した。最終的な反応混合物(ここでは、UGB1及びGB4又はGB5の濃度は、表1に記載の濃度に相当している)は、今や、ピペッティングにより任意の型(forms)の中に注入することが可能であり、ゲル化は、12時間かけて重合させることにより起こさせる。次いで、それらのヒドロゲルを、リン酸緩衝生理食塩液の溶液の中に、数時間の間溶液交換を繰り返すことにより完全に膨潤させ、その後で、そのヒドロゲルを、使用するか、又は特性解析した。
ヒト内皮細胞を培養するために、カバーガラスの上に、1cm2あたり11μLの最終的な反応混合物をピペッティングすることにより、約100μmの最終厚みを有する表面結合された(surface−bound)ゲルを形成させた。そのカバーガラスは、以下の文献の手順に従って、あらかしめ、ポリ(エチレン−old−無水マレイン酸)の薄膜を用いてコーティングして、ヒドロゲルの共有結合を確保しておいた:Pompe et.al.,Biomolecules,2003,4,1072〜1079。RGDペプチドを用いた修飾をするために、EDC/スルホ−NHSを含むリン酸緩衝生理食塩溶液の中で膨潤させた、成形したヒドロゲルを、4℃の、1/15Mのリン酸緩衝生理食塩溶液の中に、50mMのEDC及び25mMのスルホ−NHSの濃度で溶解させ、且つ、GB4又はGB5の残存カルボキシル基を、20分かけて活性化させてから、ホウ酸塩緩衝剤(100mM、pH8.0、4℃)を用いて、洗い流した。次いで、その活性化されたヒドロゲルを、ホウ酸塩緩衝剤(100mM、pH8.0)の中に溶解させた、ペプチド配列
ヒト臍帯静脈内皮細胞(HUVEC、Lonza,Germany)を、フィブロネクチン−コーティングした細胞培養フラスコ上のPromocell C−22010を補充したPromocell C−22010(PromoCell GmbH,Germany)の上に、37℃、及び5%CO2で継代培養し、80%の密集度を達成した。継代2〜6からの細胞を、それに続く実験で使用した。
マレイミド基を用いたポリ(4−スチレンスルホン酸−コ−マレイン酸)の誘導体化:
架橋反応2に従ったヒドロゲルを形成させるためには、最初にGB4又はGB5を、N−(2−アミノエチル)マレイミドトリフルオロアセテート(Sigma,Germany)を用いて官能化させなければならない。この目的のために、GB4又はGB5それぞれの500mg(25μmole)を、2.7mLの脱イオン化超純水の中に溶解させ、氷上で、10分間撹拌する。その試料容器は、25mLのスナップカバーガラス(snap−cover glass)である。次いで、65.14mgのNHS(0.30mmole)を400μLの中に溶解させ、且つ200μLの氷冷した脱イオン化超純水の中に溶解させた115.02mg(0.60mmole)のEDCを、GB4又はGB5の溶液に添加して、5分間待った。次のステップにおいて、200μLのMilliQ水の中に溶解させた、76.25mg(0.30mmole)のN−(−2−アミノエチル)マレイミドトリフルオロアセテートを、GB4又はGB5を活性化させる活性化時間が完了した後で40秒かけて、滴下により添加した。その反応混合物を、氷の上で、さらにまた別の10分間撹拌する。最後に、氷浴を外し、その混合物を、室温で一夜撹拌する。その生成物を、MWCO=5kDaの排除サイズの透析チューブの中に入れる。2日にわたって透析を実施する;第一日には、1モル濃度の塩化ナトリウム溶液2.5リットルを用い、6時間かけて透析を実施する。この場合、その溶液を、2時間ごとに取り替える。6時間経過後は、脱イオン化超純水に対して、透析を一晩がかりで実施する。第二日には、透析を、脱イオン化超純水に対して実施し、水は、2時間ごとに取り替える。最後のステップにおいて、その溶液を凍結乾燥させる。
排除体積クロマトグラフィー(SEC)は、GB4又はGB51分子あたりのマレイミド基を求めるために使用される。この目的のためには、ペプチドRGD−SP(M=990g/mole)を、各種の過剰状態で、GB4/GB5−マレイミド誘導体にカップリングさせる。検討したRGD−SPの過剰モルは、この場合においては、GB4又はGB5に対して、6.8、10又は12である。最初に試料容器の中で、35μLのそれぞれのRGD−SP濃度と、35μLのリン酸緩衝生理食塩溶液とを混合することにより、キャリブレーションを実施する。
GB4/GB5マレイミド誘導体と4本アームで、チオール末端のPEG(UGB2、Mw=10,000Da、Jenchem,USA、表2)とを、表1に記載の濃度の2倍で、リン酸緩衝生理食塩溶液の中に溶解させる。その後で、両方の成分と等しい容積を、ミクロ反応容器の中にピペット移植し、ボルテックスシェーカー上で10秒間振盪させる。次いで、その反応混合物の所定の容積を抜き出し、Sigma Cote(登録商標)でコーティングした直径9mmのカバーガラスの上に置く。Sigma Cote(登録商標)でコーティングしたまた別のカバーガラスを、その液滴の上に置く。30分後にカバーガラスを取り外し、そのゲルの薄片を、リン酸緩衝生理食塩溶液の中で12時間かけて膨潤させる。高いUGB2濃度のところでは、最適なゲル化を起こさせるために、1MのHClを用いて、UGB2のpHを5〜7のpHに調節することができる。
上述の方法に従い、UGB2に代えて、マトリックスメタロプロテアーゼ(MMP)で切断可能なヒドロゲルを、UGB4(表4、Tsurkan et al.,Adv.Mater.,2013,25(18),2606〜2610により記述された手順に従って合成)を使用して調製した。
間葉幹細胞(MSC、ATCC、脂肪組織から単離、Germany)を、DMEM中、10%のシ胎血清及び1%のペニシリン/ストレプトマイシンを用い、標準培養条件下(5%CO2、37℃)で培養した。実験には、継代2〜4のMSCを使用した。
ヒドロゲルディスク(60μL)を、1mLの50μg/mLゲンタマイシン(Sigma Aldrich,Germany)の中で18時間かけて培養し、次いでリン酸緩衝生理食塩溶液を用いて、2回洗浄した。ゲンタマイシンを導入した(gentamiycin−charged)ヒドロゲルの抗菌活性を、阻止帯アッセイにより測定した。この目的のためには、ルリアブロス(LB)寒天(Sigma Aldrich,Munich,Germany)プレートを、生産者の説明書に従って調節した。エシェリキア・コリ(Escherichia coli)K12 DH5(DSMZ,Germany)及びスタフィロコッカス・エピデルミジス(Staphylococcus epidermidis)PCI 1200(ATCC,USA)の12時間成長培養菌を、0.1にまで希釈して、600nmで光学濃度(OD)を測定した。これらの細菌懸濁液の250μLを、それぞれのプレートの上に塗布した。滅菌した綿布を使用して、細菌を均等に分散させた。それぞれゲンタマイシン導入又は非導入の、ヒドロゲルのUGB1−GB1 01、UGB2−GB4 20、及びUGB2−GB5 23、さらにはUGB2−UGB3 26を、寒天プレートの四隅に塗布し、それらのプレートを、37℃で一夜かけて培養した。その後で、デジタルキャリパーを用いて、阻止帯の測定をした。3種の主題デフィニション(subject definition)で、独立して10回の実験(n=30)で評価したが、すべての条件で、有意な差が見出された(一元配置Anova)。
各種のタイプのヒドロゲルの物理的性質を測定するために、67μLの未重合のヒドロゲル溶液を、それぞれの場合において、Sigmacote(Sigma−Aldrich,Germany)で処理した2枚の9mmのガラススライド(Menzel Glaeser,Germany)の間で、室温で16時間かけて重合させ、次いでそのようにして得られたゲルの薄片を、ガラススライドから取り外した。それらのゲルの薄片の直径を、タイプFLA−3100(Fujitsu,Japan)のスキャナーを用いて光学的に測定した(非膨潤状態での直径)。その後で、それらのゲルの薄片を、0.9%NaCl緩衝によりpH7.4としたリン酸緩衝生理食塩溶液(PBS)(Sigma−Aldrich,Germany)の中(生理学的条件下)で24時間かけて膨潤させ、FLAタイプ3100(Fujitsu,Japan)のスキャナーを用いて再度測定した(膨潤状態における直径)。測定した直径から、それらのヒドロゲルの膨潤度を、次式に従って求めた:

- スルフェート化及び/又はスルホン化ヒドロゲルの中の異なる物質群A及びBの物質を区分して隔離するため、及びバイオ流体から物質群Aの物質を枯渇させ、同時に、前記スルフェート化及び/又はスルホン化ヒドロゲルから前記バイオ流体の中に物質群A又はBの物質を区分して放出するか、又は前記スルフェート化及び/又はスルホン化ヒドロゲルの中の前記物質群Bの物質の結合性を低下させるための方法であって、前記スルフェート化及び/又はスルホン化ヒドロゲルが、タイプ1、タイプ2、タイプ3又はタイプ4のヒドロゲルの群から選択され、且つ前記ヒドロゲルが、荷電されていない構成単位及び荷電された構成単位から構成され、前記荷電された構成単位が、タイプ1では0.0040〜0.0060mole/g、タイプ2では0.0025〜0.0040mole/g、タイプ3では0.0005〜0.0025mole/g、及びタイプ4では0040〜0.0100mole/gの、繰り返し単位あたりのスルフェート基及び/又はスルホネート基の数を前記繰り返し単位のモル質量で割ったものから計算されたパラメーターを特徴とし、且つ膨潤させたヒドロゲルが、20kPa未満の貯蔵モジュラスを有し、且つ前記膨潤させたヒドロゲルの性質が、タイプ1では0.09〜0.20、タイプ2では0.05〜0.18、タイプ3では0.01〜0.12、及びタイプ4では0.16〜0.8の、mmole/mLの単位での、スルフェート基又はスルホネート基の濃度を有し、且つ、前記バイオ流体中の物質群Aの物質の濃度及び物質群Bの物質の濃度への影響が、前記ヒドロゲルのタイプの選択によって定まることを特徴とする、方法。

- 4−スチレンスルホン酸対マレイン酸のモル比が可変で、6:1から1:6までの範囲、且つモル質量が5,000〜100,000g/moleの範囲であるポリ(4−スチレンスルホン酸−コ−マレイン酸)が前記荷電された構成単位として選択されることを特徴とする、請求項19〜21のいずれか1項に記載の共有結合的に架橋されたヒドロゲル材料。
- 前記ペプチド配列中の反応性アミノ酸としてリシン又はシステインのいずれかを有する、共役結合された酵素的に切断可能なペプチドを含むポリマーが、前記ポリマーネットワークを形成させるための荷電されていない構成単位として使用されることを特徴とする、請求項19〜22のいずれか1項に記載の共有結合的に架橋されたヒドロゲル材料。
- 前記酵素的に切断可能なペプチドが、ヒト又は細菌性プロテアーゼ、特にMMP、カテプシン、エラスターゼ、オーレオリシン及び/又は血液凝固酵素によって切断可能であることを特徴とする、請求項23に記載の共有結合的に架橋されたヒドロゲル材料。
- アミノ基若しくはカルボキシル基及び/又はセル−インストラクティングペプチドを有する生理活性及び/又は抗接着性分子が、前記ヒドロゲルネットワークに、配列中のリシン又はシステインを介して、前記荷電された構成単位のポリ(4−スチレンスルホン酸−コ−マレイン酸)又はマイケルタイプの付加を可能とする基を有するその誘導体に、共有結合を形成することによって結合されることを特徴とする、請求項23又は24に記載の共有結合的に架橋されたヒドロゲル材料。
- 前記生理活性分子が、抗菌物質、たとえば抗生物質若しくは消毒剤、又は医薬活性成分であることを特徴とする、請求項25に記載の共有結合的に架橋されたヒドロゲル材料。
- 前記抗接着性分子が、ポリエチレングリコール(PEG)又はポリ(2−オキサゾリン)(POX)であることを特徴とする、請求項25又は26に記載の共有結合的に架橋されたヒドロゲル材料。
- 前記セル−インストラクティングペプチドが、細胞外マトリックス、たとえばコラーゲン、ラミニン、テネイシン、フィブロネクチン、及びビトロネクチンの、構造的及び機能的タンパク質から誘導されたペプチドであることを特徴とする、請求項25〜27のいずれか1項に記載の共有結合的に架橋されたヒドロゲル材料。
- 前記生理活性及び/又は抗接着性及び/又は細胞誘導性のペプチドが、前記ヒドロゲルネットワークに、酵素的に切断可能なペプチド配列を介して共有結合的に結合されていることを特徴とする、請求項23〜28のいずれか1項に記載の共有結合的に架橋されたヒドロゲル材料。
- 前記ヒドロゲル材料が、0.2〜22kPaの貯蔵モジュラスを有していることを特徴とする、請求項19〜29のいずれか1項に記載の共有結合的に架橋されたヒドロゲル材料。
- 請求項1〜18のいずれか1項に記載の方法を実施するための物理的に架橋されたヒドロゲル材料であって、ポリ(4−スチレンスルホン酸−コ−マレイン酸)の形態にある荷電された構成単位と、ポリマーの形態にある荷電されていない構成単位との間の物理的相互作用をベースとし、強く正に荷電されたペプチド配列が、前記ポリマーの上に共役結合されている、物理的に架橋されたヒドロゲル材料。
- 前記強く正に荷電されたペプチド配列が、リシン若しくはアルギニンの少なくとも10個の繰り返し、又はリシン及びアラニンを用いるか若しくはアルギニン及びアラニンを用いたジペプチドモチーフの少なくとも5個の繰り返しを含んでいることを特徴とする、請求項31に記載の物理的に架橋されたヒドロゲル材料。
- 前記ヒドロゲルが、インビボで血管形成、免疫疾患、癌、糖尿病、神経変性疾患、クローン病、大腸潰瘍、多発性硬化症、喘息、慢性関節リウマチ、又は皮膚の創傷治癒、及び骨の再生を調節するための因子を取り扱うために使用されることを特徴とする、請求項1〜32のいずれか1項に記載のヒドロゲルの使用。
- 皮膚の創傷治癒のために、タイプ2のヒドロゲルが、炎症誘発性のケモカインの、エオタキシン、GRO−a、IL−8、IP−10、MCP−1、MCP−3、MCD、Rantes、及びSDF−1−アルファの少なくとも1つを前記ヒドロゲルの内側に隔離するために使用され、前記再生誘発因子の、EGF、FGF−2、TGFb1、IL−10、HGF、及びPLGFの少なくとも1つが、前記バイオ流体の中で、影響されないか又はほとんど影響されずに留まることを特徴とする、請求項33に記載の使用。
- 微生物又は真核生物由来の細胞溶解物から、目標とするタンパク質を精製するための、請求項1〜32のいずれか1項に記載のヒドロゲルの使用。
- バイオ流体から、タイプ1〜3の前記ヒドロゲルに弱く結合されている、EGF、FGF−2、TGF−β、IL−10、HGF、及びPLGFのシグナル伝達分子をネガティブに選択及び分離するための、請求項1〜32のいずれか1項に記載のヒドロゲルの使用。
- 胚幹細胞(ES)、人工多能性幹細胞(iPS−)、並びにES及びiPS−を伴わないその他の幹細胞及び前駆細胞、患者から採取した初代細胞、不死化細胞系、さらには心臓、筋肉、腎臓、肝臓、及び神経組織から、インビトロで細胞及び器官培養するための、請求項1〜32のいずれか1項に記載のヒドロゲルの使用。
