一部の実施形態では、検出可能な標識は、Forster共鳴エネルギー移動などの、且つ限定されないが、生物発光共鳴エネルギー移動(「BRET」)及び蛍光共鳴エネルギー移動(「FRET」)を含む共鳴エネルギー移動(RET)に基づく。
化学発光は、化学反応の結果として、熱の放出が制限されたエネルギーの放出(発光)である。「化学発光」という用語は、本明細書では、酵素の活性に依存する生物発光を包含するため使用される。非酵素化学発光は、触媒の存在下での有機染料及び酸化剤間の化学反応の結果である。化学発光放出は、化学的に誘起される有機色素の励起状態からのエネルギーが基底状態に崩壊するにつれて発生する。化学発光放出の持続時間及び強度は、反応溶液中に存在する化学試薬の程度にほぼ依存している。

プラスミン、そのチモーゲンプラスミノーゲン、及び細菌プロテアーゼなどのプロテアーゼは、乳製品中でカゼインミセル及び乳脂肪球と結合し得る。これは、これらの試料中に存在するプロテアーゼの量を測定することを困難にし得る。理論に拘束されることを望むものではないが、これらのプロテアーゼに対する天然基質であるカゼインは、プロテアーゼアッセイに使用される基質に対する競合的阻害剤として作用し得ると考えられる。乳製品中のプロテアーゼの量を測定する種々の方法が記載されている(例えば、プラスミンの場合、Saint−Denis et al.(2001)及びRauh et al.(2014)を参照のこと)。これらのアッセイにより測定されたプロテアーゼの量は、試料がアッセイのために調製されている仕方に応じて変動し得る。カゼインミセルからプロテアーゼを解離させ、プロテアーゼアッセイのための試料を調製するために使用することができる改良された方法が必要とされている。
(i)本明細書に記載の方法を使用して、乳製品の試料を処理すること。
(ii)加工された乳製品中のプロテアーゼ活性を測定すること、及び
(iii)プロテアーゼ活性に基づいて乳製品のプロテアーゼ濃度を決定すること、
を含む。一部の実施形態では、乳製品の試料はまた、乳製品の部分的に精製された部分などの抽出物とし得る。一部の実施形態では、未殺菌乳、低脂肪乳、無脂肪乳、低温殺菌乳、UHT乳、ラクトース変性UHT乳、強化UHT乳、フレーバーUHT乳、UHT特殊調製粉乳、チーズ、ヨーグルト、乳清、バターミルク、クリーム、粉乳、乳児用調製粉乳、及びバター、ならびにそれらの組み合わせ、及び/またはカゼインに結合しているプロテアーゼを含むそれらのうちの1つ以上の抽出物からなる群より選択される乳製品。
センサーの構築
BRET2−プラスミンセンサー及びBRET2−細菌プロテアーゼPfluセンサー1、2、及び3をコードするDNA構築物を、GenScript(USA)で合成した。センサーは、化学発光ドナーとしてのRluc2、及びアクセプターとしての緑色蛍光タンパク質(GFP2)の多様体を含んでいた。EcoRI及びXhoI制限部位を使用して、pRSETベクター(BioLabs、オーストラリア)に構築物をクローニングして、プロテアーゼセンサーをコードするpRSET−Pfluベクターを作製した。プロテアーゼセンサーをコードするポリヌクレオチド配列を、配列番号102〜104に示す。プロテアーゼセンサーのポリペプチド配列を、配列番号105〜107に示す。プラスミンセンサーをコードするポリヌクレオチド配列を、配列番号108に示す。プラスミンセンサーのポリペプチド配列を、配列番号109に示す。
材料及び方法
粗細菌プロテアーゼ
OXOID寒天プレート上に、Pseudomonas fluorescens(株65)の分離株をプレーティングし、28℃で2日間インキュベートした。プレートからの3つのコロニーを、滅菌栄養ブロス(NB)100mlに接種し、28℃で48時間インキュベートした。10mLの培養物を、6000×gで10分間遠心分離した。上清を廃棄し、細胞ペレットを滅菌等張生理食塩水(0.85%w/v)1mLに再懸濁し、脱脂UHT乳(COLESオーストラリア脱脂乳、99%脂肪不含)99mLに添加した。培養物を、150rpmで円形撹拌しながら、4℃で8日間インキュベートした。0.85%w/vの生理食塩水1mLを脱脂UHT乳99mLに添加すること、及び同一条件下で8日間インキュベートすることにより、対照培養物を調製した。この後、培養物を、4℃、3,000×gで10分間遠心分離した。上清を収集し、0.22μmフィルタで濾過滅菌し、粗プロテアーゼの供給源として使用した。これを、1mlのアリコートに等分し、−80℃で保存した。
最終体積100μLを含む96ウェルプレート中で、プロテアーゼアッセイを実施した。切断緩衝液(50mMのトリス−HCl、50mMのNaCl、25mMのリシン、pH8)中の、または50μLの全脂肪または脱脂UHT乳(Coles)が追加された様々な量の粗細菌プロテアーゼまたはプラスミン(ヒトもしくはウシ、Sigma)と共に、精製プラスミンセンサー(10nm)を、28℃で10分間インキュベートした。1pM〜10nMのプロテアーゼ濃度をアッセイすることにより、較正グラフを生成した。10分間インキュベーションした後に、BRET2シグナルを測定した。BRET2測定では、10分間のインキュベーション期間の後に、5μLのClz400a基質(100μM)を加えた。
マイクロプレート分光計を使用して、500nm及び470nmで測定された生物発光放出の比としてBRET比を計算した。GFP2強度(平均0〜5秒)をRLuc2強度(平均0〜5秒)で除することにより、BRET比を計算した。Windows(登録商標)用のGraphpad Prism 7の正規化関数を使用して、BRET比を正規化して、Log[プロテアーゼ]値に対してプロットし、Log[アゴニスト]対正規化応答−可変勾配モデルに適合させた。
ウシプラスミン及びヒトプラスミン(図4)または細菌プロテアーゼに対する全脂肪UHT乳中のプラスミンセンサーの検量線を作成した。プラスミンセンサーは、ヒトプラスミンの場合に5.6nMのEC50、ウシプラスミンの場合に108nMのEC50を有していた(表3)。ヒトプラスミン、ウシプラスミン、及び細菌プロテアーゼへのプラスミンセンサーの応答を比較する予備研究(図5)は、細菌プロテアーゼの場合のEC50が全脂肪UHT乳中で約50nMであることを推測する。
本出願のセンサーは、乳製品などの試料中のPseudomonas spp.プロテアーゼを検出するために使用することができる。実施例1に記載されるように、Pfluセンサー1、2、及び3を調製した。実施例2に記載されるように、粗細菌プロテアーゼを調製した。10nMの精製センサーを使用した実施例2に記載されるように、プロテアーゼアッセイを実施した。
細菌プロテアーゼ、ウシプラスミン、及び/またはヒトプラスミンへの全脂肪UHT乳中のPfluセンサー1、2、及び3の検量線を作成した。図7(Pfluセンサー1)、図8(Pfluセンサー1)、図9(Pfluセンサー2)、及び図10(Pfluセンサー3)に検量線を示す。
センサーの標的配列を変えることにより、0.1〜90nMの細菌プロテアーゼの線形範囲の組み合わせを与える細菌プロテアーゼへの細菌プロテアーゼセンサーの感度は、32〜537nMの範囲のウシプラスミンレベル及び0.7〜3680nMの範囲のヒトプラスミンレベルを測定し得る。プラスミンの濃度は、比色測定を使用した未殺菌乳中の1〜5nMの濃度範囲で(Benslimane et al.、2009)、ELISAアッセイを使用して0.6〜22nMの範囲で(Dupont et al.、1997)測定されている。本発明によるセンサーは、細菌プロテアーゼにより切断され、細菌プロテアーゼへの選択性があり、乳汁中のプラスミンの生理学的レベルに対して大部分または比較的感度が鈍いことを、これらのデータが実証する。
材料及び方法
未殺菌乳試料
オーストラリアの乳製品加工業者から、6つの未殺菌乳試料を入手した。これらの試料を得た雌ウシは、貯蔵寿命が長い生乳を一貫して生産することができる高品質の生産者であると認識されていたことを、加工業者は示していた。
未殺菌乳試料を12分間超音波処理した(Misonix超音波処理装置3000;試料パルサーオン、0.5秒;パルサーオフ、2.5秒;パワー5.5)。未殺菌乳試料を超音波処理して、均質な分散液を作製する。超音波処理後、Rauh et al.(2014)に記載の前処理方法を使用して、乳汁試料を前処理した。要約すると、0.4Mのクエン酸三ナトリウム緩衝液(pH8.9)250μLと、超音波処理した未殺菌乳1mLを混合し、振盪しながら室温で15分間インキュベートして、カゼインミセルを解離させた。8mMのε−アミノカプロン酸(EACA)及び0.4MのNaClを含有する0.1Mのトリス緩衝液(pH8)と、クエン酸処理乳を混合(1:1)し、振盪しながら室温でさらに15分間インキュベートした。
検量線を作成するために、前処理された乳汁試料のアリコートを30秒間90℃に加熱して、任意の内因性プラスミンを不活化した。10μLのプラスミン溶液を50μLの加熱した乳汁試料に添加することにより、加熱した前処理した試料にヒトプラスミン(Sigma)を混ぜた。添加されたプラスミンの最終濃度は、0.001〜500nMに及んでいた。5μLのBRET2プラスミンセンサー(10nM)及び30μLのプラスミン切断緩衝液を混ぜた試料に加え、試料を、ピペッティングすることによりよく混合した。試料を、28℃で10分間インキュベートした。インキュベーション後に、5μLのClz400a基質(100μM)をウェルに添加し、BRET比を上記のように記録した。
未殺菌乳試料中に存在するプラスミンの量を決定するために、50μLの前処理された未殺菌乳試料をマイクロプレートウェル中で5μLのBRET2プラスミンセンサー(10nM)及び40μLのプラスミン切断緩衝液と混合した。混合物を、28℃で10分間インキュベートした。インキュベーション後、5μLのClz400a基質(100μM)をウェルに添加し、BRET比を記録した。検量線から計算した濃度に2.5の希釈係数を掛けた。
比色基質としてD−Val−Leu−Lys−4−ニトロアニリド二塩酸塩(V0882、Sigma Aldrich)を使用したRauh et al.の方法(2014)を使用して、BRET2プラスミンセンサーに基づく測定を検証した。
BRET2プラスミンセンサー
前処理ステップなしで未殺菌乳中のプラスミン濃度を決定する最初の試みは成功しなかった。
Rauh et al.(2014)の前処理プロトコール後に、ヒトプラスミンを混ぜた未殺菌乳試料の場合の、比色基質D−Val−Leu−Lys−4−ニトロアニリド二塩酸塩の検量線を作成した(図12)。元のRauh前処理プロトコール後の未殺菌乳試料中で測定された内因性プラスミン濃度は、2.1nM〜2.9nMの範囲であった(表4)。
材料及び方法
2ステップ前処理プロトコール
クエン酸三ナトリウム緩衝液にEACA及びNaClを添加して、2回の15分間のインキュベーションのステップを組み合わせることにより、実施例4に記載の前処理プロトコールを修正した。要約すると、5mMのEACA及び0.25MのNaClを含有するクエン酸三ナトリウム緩衝液(pH8.9)250μLと、1mLの超音波処理未殺菌乳を混合した。次に、試料を室温で15分間インキュベートした。

BRET2プラスミンセンサー及び比色基質についての検量線を、実施例4に記載のように決定した。
実施例4に記載されるように、未殺菌乳試料中に存在するプラスミンの量を決定した。実施例4に記載されているように、比色基質、D−Val−Leu−Lys−4−ニトロアニリド二塩酸塩を使用して、未殺菌乳試料中に存在するプラスミンの量を検証した。
BRET2プラスミンセンサー
ヒトプラスミンを混ぜた未殺菌乳試料を使用して、2ステップ前処理プロトコール後に、BRET2プラスミンセンサーの検量線を作成した。EC50は、16.04nMである(図13)。未殺菌乳試料中で測定された内因性プラスミン濃度は、5.8nM〜9.0nMの範囲であった(表5)。
2ステップ前処理プロトコールにより処理し、ヒトプラスミンを混ぜた未殺菌乳試料を使用して比色基質の検量線を作成した(図14)。2ステップ前処理プロトコール後の未殺菌乳試料中で測定された内因性プラスミン濃度は、9.7nm〜10.9nMの範囲であった(表5)。

遊離プラスミンの放出を促進するために超音波処理及びインキュベーションステップを組み合わせることにより、実施例5に記載の前処理プロトコールを修正した。要約すると、未殺菌乳1mLに、0.4Mのクエン酸三ナトリウム緩衝液(5mMのEACA及び0.25MのNaClを含有するpH8.9)250μLを混ぜ、試料を12分間超音波処理した(Misonix超音波処理装置3000;試料パルサーオン、0.5秒;パルサーオフ、2.5秒、パワー5.5)。
BRET2プラスミンセンサー及び比色基質についての検量線を、実施例4に記載のように決定した。
実施例4に記載されるように、未殺菌乳試料中に存在するプラスミンの量を決定した。実施例4に記載されているように、比色基質、D−Val−Leu−Lys−4−ニトロアニリド二塩酸塩を使用して、未殺菌乳試料中に存在するプラスミンの量を検証した。
BRET2プラスミンセンサー
1ステップ前処理プロトコール後に、ヒトプラスミンを混ぜた未殺菌乳試料の場合の、BRET2プラスミンセンサーの検量線を作成した。検量線を図15に示す。EC50は、24.2nMであり、線形応答が1.6nM(90%)〜128nM(10%)のプラスミンで観察される。
1ステップ前処理プロトコール後に、ヒトプラスミンを混ぜた未殺菌乳試料の場合の、比色基質、D−Val−Leu−Lys−4−ニトロアニリド二塩酸塩の検量線を作成した。検量線を図16に示す。

低出力超音波処理を含む1ステップ前処理プロトコール
超音波処理装置(Misonix超音波処理装置3000)を、連続的に使用することができる低出力超音波処理装置(Q55超音波処理装置、Daintree Scientific)と交換することにより、実施例6に記載の前処理プロトコールをさらに修正した。要約すると、0.4Mのクエン酸三ナトリウム緩衝液(pH8.9、5mMのEACA及び0.25MのNaClを含有する)250μLと、未殺菌乳1mLを混合した。試料を氷上で2分間連続して超音波処理した(Q55超音波処理装置、振幅を40に設定)。
実施例4に記載されたように、検量線を決定した。
実施例4に記載されるように、未殺菌乳試料中に存在するプラスミンの量を決定した。
低出力超音波処理を含む1ステップ前処理プロトコール後に、ヒトプラスミンを混ぜた未殺菌乳試料の場合の、BRET2プラスミンセンサーの検量線を作成した。検量線を図17に示す。計算されたEC50は、29.3nMであり、応答は、5.5nM(90%)〜107nM(10%)で線形である。
実施例4〜7のデータは、前処理プロトコールへの超音波処理の組み込みが、アッセイの感度及び乳汁試料中で測定された遊離内因性プラスミンの量に大きな好ましい効果を有することを示す(表8)。例えば、1ステップ前処理プロトコールを使用すると、Rauh et al.(2014)の前処理方法と比較して、測定された遊離内因性プラスミンのレベルが5〜10倍増加した。比色基質アッセイを使用して、これを検証した。BRETアッセイ及び比色アッセイの双方について、未殺菌乳中で測定された内因性遊離プラスミンレベルは、2ステップ前処理の代わりに1ステッププロトコールを使用した場合に2〜3倍高かった。低出力超音波処理を組み込むと、同時に、元の方法の場合の50分のアッセイ時間から低出力超音波処理の前処理場合の12分のアッセイ時間に総アッセイ時間を低減しながら、Rauh et al.(2014)の前処理方法と比較して、未殺菌乳中で測定された内因性遊離プラスミンレベルが4〜6倍増加した。データは、EACAの存在下で試料を超音波処理することが、カゼインからの内因性プラスミンの解離の効率を有意に増加させることを示している。

材料及び方法
未殺菌乳中の細菌プロテアーゼを測定するために、Pflu1センサーを本明細書で使用した。試料を4分間超音波処理したことを除いて、実施例7に記載されたように、前処理された未殺菌乳試料を調製した。
検量線を生成するために、前処理された未殺菌乳試料のアリコートを、1時間55℃、続いて、30秒間90℃に加熱して、任意の内因性細菌プロテアーゼ及び内因性プラスミンを不活化した。50μLの加熱された乳汁試料に、10μLの細菌プロテアーゼを添加することにより、加熱前処理された試料に、種々の量の粗製細菌プロテアーゼ画分を混ぜた(調製については実施例2を参照のこと)。添加された細菌プロテアーゼの最終濃度は、1pM〜10nMの範囲であった。5μLのBRET2 Pflu1センサー(10nM)及び30μLのプラスミン切断緩衝液を、混ぜた試料に添加して、試料をピペッティングにより十分に混合した。試料を、28℃で10分間インキュベートした。インキュベーション後に、5μLのClz400a基質(100μM)をウェルに添加し、BRET比を上記のように記録した。
未殺菌乳試料中に存在する細菌プロテアーゼの量を決定するために、マイクロプレートウェル中の5μLのPflu1センサー(10nM)及び40μLのプラスミン切断緩衝液と、前処理未殺菌乳試料50μLを混合した。混合物を、28℃で2時間インキュベートした。インキュベーション後、5μLのClz400a基質(100μM)をウェルに添加し、BRET比を記録した。検量線から計算した濃度に2.5の希釈係数を掛けた。
前処理及びPflu1センサー後の未殺菌乳試料の検量線を作成した。検量線を図18に示す。EC50は、0.4nMであり、応答は、48.2pM(90%)〜2.6nM(10%)の線形である。迅速な1ステップ前処理プロトコール後の未殺菌乳試料中で測定された細菌プロテアーゼのレベルは、主に、CYBERTONGUE(登録商標)細菌プロテアーゼセンサーの本発明者らの現在の生成の検出の限界以下のサブピコモル濃度で存在していた(検出限界は現時点では、1pM以上と推定される)。未殺菌乳試料のうち2つを除く全ては、検出可能な細菌プロテアーゼを含まなかった(図19)。内因性細菌プロテアーゼを測定することができた試料では、濃度は、1.4pM〜3.4pMの範囲であった(図19)。
材料及び方法
本明細書では、未殺菌乳試料の任意の前処理なしに、未殺菌乳中の細菌プロテアーゼを測定するために、Pflu1センサーを使用した。
検量線を生成するために、前処理された未殺菌乳試料のアリコートを、55℃に1時間加熱して、任意の内因性細菌プロテアーゼを不活化した。50μLの加熱された乳汁試料に、10μLの細菌プロテアーゼを添加することにより、加熱前処理された試料に、種々の量の粗製細菌プロテアーゼ画分を混ぜた(調製については実施例2を参照のこと)。添加された細菌プロテアーゼの最終濃度は、1pM〜10nMの範囲であった。5μLのBRET2 Pflu1センサー(10nM)及び30μLのプラスミン切断緩衝液を、混ぜた試料に添加して、試料をピペッティングにより十分に混合した。試料を、前処理(低出力超音波処理を含む1ステップ前処理プロトコール)有り及びなしで、28℃で10分間または2時間インキュベートした。インキュベーション後に、5μLのClz400a基質(100μM)をウェルに添加し、BRET比を上記のように記録した。
細菌プロテアーゼセンサーは、2時間のインキュベーション期間の後に(4分間の低出力超音波処理を含む1ステップ前処理プロトコールを使用して)前処理されている未殺菌乳中で検量された同じセンサーと比較して、細菌プロテアーゼを含むセンサーの10分間のインキュベーション後の未殺菌乳のいかなる前処理もなしに、細菌プロテアーゼへの類似の応答及び感度を有することが明らかである(図20〜22、表9)。

Pflu1センサーが複数のP.fluorescens株に応答することができるかどうかを決定するために、本発明者らは、P.fluorescens株117由来の細菌プロテアーゼの粗抽出物を生成した。
実施例2に記載の方法を使用して、P.fluorescens株117を接種した脱脂乳から粗抽出物を調製した。
切断緩衝液及びUHT乳に、株65または117いずれかから産生された1nMの細菌プロテアーゼを混ぜ、応答を、いかなる細菌プロテアーゼも添加しない対照と比較した。実施例2に記載したように、粗細菌プロテアーゼを調製した。10nMの精製センサーを使用して、実施例2に記載されるように、プロテアーゼアッセイを実施した。
P.fluorescens株65及び117から産生された細菌プロテアーゼは、緩衝液中で10分のインキュベーション時間内にPlu1センサーを完全に切断して(図24)、BRET比の95%の減少をもたらした。1nMの細菌プロテアーゼを混ぜたUHT乳中で、アッセイを実施した場合、株117由来の細菌プロテアーゼにおける71.2%の低減と比較して、株65の細菌プロテアーゼにおけるBRET比の80.6%の低減があった。これにより、Pflu1センサーが異なるP.fluorescens株から産生された細菌プロテアーゼに応答することができることが実証される。
Pflu1〜3センサーシリーズは、κ−カゼインに見られる切断部位及びポリペプチド配列を組み込む、中央のF−M開裂部位を有するリンカー配列(図1)で隔てられているBRET構成要素を含んでいた。全てのカゼインに見出される全ての切断部位に対するP4〜P4’位に見出される最も主要なアミノ酸を組み込むように、プロテアーゼセンサーの第2のセットを設計した(Pflu−H1〜4)。
新しい細菌プロテアーゼセンサーの構築
GenScript(米国)により、BRET2細菌プロテアーゼPfluセンサーH1、H2、H3、及びH4をコードするDNA構築物を合成した。センサーは、リンカーで隔てられている化学発光ドナーとしてのRluc2及びアクセプターとしての緑色蛍光タンパク質(GFP2)の多様体を含んでいた(表10)。pRSETベクター(BioLabs、オーストラリア)に構築物をクローニングして、プロテアーゼセンサーをコードするpRSET−Pflu−Hベクターを作製した。プロテアーゼセンサーをコードするポリヌクレオチド配列は、配列番号116〜119に示される。プロテアーゼセンサーのポリペプチド配列は、配列番号120〜123に示される。実施例1に記載の方法を使用して、精製センサーを製造した。
検量線の方法及び新規細菌プロテアーゼセンサー(Pflu−H1〜H4)のプロテアーゼアッセイは、実施例2に記載されている。

4つ新しい細菌プロテアーゼセンサー(Pflu−H1〜H4)全ては、緩衝液中の1nMの細菌プロテアーゼに対して有意に応答した(P=0.05)(図26)。センサーの相対感度は、H3>H4>H1>H2であり、応答は、それぞれ89.7%、64.5%、49.5%、及び31.9%であった。これにより、Pflu−H3センサーが、最も感度が高く、Pflu−H4、Pflu−H1、及びPflu−H2センサーのそれぞれの応答と比較して、1nMの細菌プロテアーゼへの応答が39%、82%、及び182%高いことが実証される。

実施例2及び10では、デンシトメトリーを使用して、且つSDS−PAGEの細菌プロテアーゼバンドの相対密度を46kDaバンドのマーカーバンドと比較して、細菌プロテアーゼ濃度を推定した(カラータンパク質標準、Broad Range #P7712S Lot:0021410、BioLaban)。本発明者らは、細菌プロテアーゼの相対密度を既知の濃度のBSAと比較することにより、細菌プロテアーゼ濃度を決定する異なるアプローチを調査した。
実施例2及び10に記載の方法を使用して、P.fluorescens株65及び117を接種した脱脂乳から粗抽出物を調製した。株65(65−1及び65−2)ならびに株117及び異なる濃度のBSAの2つの抽出物のSDS−PAGE分析を実施した(図32)。株65−1は、株65由来の細菌プロテアーゼを使用している本明細書中の全ての実施例に使用される抽出物である。株117は、実施例10に使用される抽出物である。Novex #LC5625 SeeBlue Pre−stained Standardは、使用された市販のマーカーであった。SDS−PAGEでのタンパク質のデンシトメトリー分析を、Image Quant TL(GE Healthcare)及びBSA検量線から推定された細菌プロテアーゼ濃度を使用して実施した(図33)。線形回帰分析を当てはめることにより、GraphPad Prism(バージョン7)を使用して、検量線グラフをプロットした。
BSA較正グラフに最適な直線は、y=107235+79.66x、R2=.9679であると決定された(図32)。抽出物65−1、65−2、及び117のための細菌プロテアーゼ濃度はそれぞれ、それぞれ3.94、2.37、及び3.37μMであると決定された。実施例2及び10において株65−1及び株117の同じバッチの濃度を決定するために使用された以前の方法により、それぞれ、0.34μM及び0.32μMの濃度が推定された。濃度決定のための今日のアプローチを使用すると、細菌プロテアーゼの濃度が、株65(65−1)の場合11.6倍高く、株117の場合10.5倍高くなる。Pflu1センサーの感度に対するこれの効果の一例としては、補正率11.6を使用して、図7をプロットするために使用される全ての細菌プロテアーゼ濃度を補正し、結果として、表3に提示されているものよりも11.6高い8.2nMのEC50及び1.4〜45nMの線形範囲(図34及び表12)になる。細菌プロテアーゼ濃度に関する補正率を、細菌プロテアーゼ測定ために、表3に最初に提示された全ての値に適用した(表12)。
表12.粗プロテアーゼを含む50%の全脂肪乳で計算されたEC50及び線形範囲。10%〜90%のBRET2応答の範囲として定義された線形範囲。細菌プロテアーゼセンサーPflu1、Pflu2、Pflu3、Pflu−H3、及びプラスミンセンサーに対する選択率(細菌プロテアーゼ:プラスミン)。BSA標準との比較により決定した細菌プロテアーゼ。N.D.=測定なし、*は、未殺菌乳試料中で計算された値を示し、1は、株117由来の細菌プロテアーゼへの応答を示し、他の全ての応答は、株65由来の細菌プロテアーゼに対するものである。

Claims (24)
- Pseudomonas spp.プロテアーゼセンサー分子であって、KQ、SF、FM、KK、KN、NQ、NT、EI、QQ、及びそれらの組み合わせからなる群より選択される少なくとも1つのPseudomonas spp.プロテアーゼ切断部位、ならびに以下の特徴:
(i)前記標的配列の長さが、約50アミノ酸未満である、
(ii)検出可能な標識、及び/または
(iii)前記プロテアーゼ切断部位のうちの2つ以上、
のうちの1つ以上を有する標的配列を含み、
前記Pseudomonas spp.プロテアーゼによる標的配列の切断が、検出可能な変化を生じる、前記Pseudomonas spp.プロテアーゼセンサー分子。 - 前記標的配列が、3、4、5、6、または7つのプロテアーゼ切断部位を含む、請求項1に記載のセンサー。
- 前記標的配列が、SFM(配列番号37)、SKMXXPP(式中、Xは、任意のアミノ酸(配列番号40)である)、及び/またはKKNQ(配列番号24)を少なくとも含む、請求項1または請求項2に記載のセンサー。
- 前記標的配列が、
SFMKKNQNTEI(配列番号45)、
SFMNQNTEI(配列番号57)、
LSFMAIP(配列番号70)、
LQGSFMKKNQNTEIGSFE(配列番号72)、
LQGSFMNQNTEIGSFE(配列番号73)、
LQGSLSFMAIPGSFE(配列番号74)、及び

から選択されるアミノ酸配列を含む、請求項1または請求項2に記載のセンサー。 - 前記標的配列が、検出可能な標識に結合されているN末端及び/またはC末端にアミノ酸リンカーを含む、請求項1〜4のいずれか1項に記載のセンサー。
- 前記検出可能な変化が、共鳴エネルギー移動、音響、電気化学ポテンシャル、蛍光、化学発光、燐光、吸光度、抗体結合、BRET比、FRET効率、または質量の変化である、請求項1〜5のいずれか1項に記載のセンサー。
- 前記検出可能な標識が、発色団、ナノ粒子、量子ドット、ビオロゲン、及びそれらの組み合わせからなる群より選択される、請求項1〜6のいずれか1項に記載のセンサー。
- Pseudomonas spp.プロテアーゼによる前記標的配列の切断が、共鳴エネルギー移動(RET)の検出可能な変化をもたらす、請求項1〜7のいずれか1項に記載のセンサー。
- 前記検出可能な標識が、2つの以上の構成要素を含む、請求項1〜8のいずれか1項に記載のセンサー。
- 前記検出可能な標識が、
i)前記標的配列に共有結合されている化学発光ドナードメイン及びアクセプタードメイン、または
ii)蛍光団ドメイン及びアクセプタードメイン、
含む、請求項9に記載のセンサー。 - 1つのドメインが、前記標的配列のN末端に共有結合しており、前記他のドメインが、前記標的配列のC末端に共有結合している、請求項10に記載のセンサー。
- 前記アクセプタードメインが、緑色蛍光タンパク質(GFP)、Venus、mOrange、またはそれらのうちのいずれか1つの生物学的に活性な多様体もしくはフラグメントであり、
前記化学発光ドナードメインが、ルシフェラーゼまたは生物学的に活性な多様体もしくはフラグメントであり、
前記標的配列が、SFMKKNQNTEI(配列番号45)、SFMNQNTEI(配列番号57)、またはLSFMAIP(配列番号70)を含む、請求項10または請求項11に記載のセンサー。 - i)Pseudomonas spp.プロテアーゼの場合、0.1〜10nM、もしくは0.1〜100nMのEC50を有し、及び/または
ii)ウシプラスミンと比較して、Pseudomonas spp.プロテアーゼへの感度が、少なくとも20倍、または少なくとも100倍高い、
請求項1〜12のいずれか1項に記載のセンサー。 - 請求項1〜13のいずれか1項に記載のセンサーもしくはその標的配列をコードする単離核酸、または請求項1〜13のいずれか1項に記載のセンサーもしくはその標的配列をコードする核酸を含むベクター。
- i)試料中のPseudomonas spp.プロテアーゼを検出するための、
ii)乳製品の腐敗を検出するための、または
iii)Pseudomonas spp.感染を検出するための、
請求項1〜13のいずれか1項に記載のセンサーの使用。 - 試料中のPseudomonas spp.プロテアーゼを検出する方法であって、前記方法が、
i)請求項1〜13のいずれか1項に記載のセンサーと、試料を接触させること、及び
ii)前記センサーの変化を検出すること、
を含み、前記変化が、前記試料中のPseudomonas spp.プロテアーゼの存在に対応する、前記方法。 - 乳製品の腐敗を検出する方法であって、前記方法が、
i)請求項1〜13のいずれか1項に記載のセンサーと、試料を接触させること、及び
ii)前記センサーの変化を検出すること、
を含み、前記変化が、前記乳製品が腐敗しているか、腐敗し始めているか、または腐敗する可能性があることを示す、前記方法。 - i)少なくとも1pM、もしくは少なくとも10pM、もしくは少なくとも100pmのPseudomonas spp.プロテアーゼを検出し、及び/または
ii)1時間未満、30分未満、または15分未満で実施される、
請求項16または請求項17に記載の方法。 - 請求項1〜13のいずれか1項に記載のセンサー及び1つ以上の許容される担体(複数可)を含む組成物。
- カゼインミセルからプロテアーゼを解離する方法であって、前記方法が、カゼインに結合している前記プロテアーゼと競合する化合物の存在下で、カゼインミセルと結合しているプロテアーゼを含む組成物を超音波処理することを含む、前記方法。
- 以下の特徴:
i)前記組成物が、カゼインに結合しているプロテアーゼを含む乳製品またはその抽出物の試料である、
ii)前記プロテアーゼが、少なくとも部分的には乳汁腐敗を引き起こす、
iii)前記プロテアーゼが、プラスミンである、
iv)前記化合物が、アミノカプロン酸である、
v)前記組成物が、カルシウムキレート剤を含む、
vi)前記超音波処理が、15分未満、10分未満、5分未満、または2分未満実施される、及び
vii)前記超音波処理が、約50℃未満、または40℃未満、または30℃未満の温度で実施される、
のうちの1つ以上が、適用される、請求項20に記載の方法。 - 乳製品中のプロテアーゼの濃度を決定する方法であって、前記方法が、
(i)請求項20または請求項21のいずれか1項に記載の方法を使用して、前記乳製品の試料を処理すること、
(ii)前記加工乳製品中のプロテアーゼ活性を測定すること、及び
(iii)前記プロテアーゼ活性に基づいて前記乳製品のプロテアーゼの濃度を決定すること、
を含む、前記方法。 - ステップ(ii)が、
少なくとも1つのプロテアーゼ切断部位を有する標的配列及び任意に検出可能な標識を含むセンサー分子と、前記加工乳製品を混合すること、
を含み、前記プロテアーゼによる前記標的配列の切断が、検出可能な変化を生じ、さらに前記ステップ(ii)が、
前記検出可能な変化を測定すること、
を含む、請求項22に記載の方法。 - 前記センサー分子が、請求項1〜13のいずれか1項に規定の通りである、請求項23に記載の方法。
