Abstract
Description
−特に生物学的媒質中において、蛍光性分子または磁性ナノ粒子の漏出または分解が起こり、組み合わさった蛍光性と磁性が失われ、性能が低下し、ノイズシグナルが出現する;
−蛍光性/磁性活性単位のペイロードが少なく、高用量の使用を余儀なくされる;
−個々の有機蛍光性分子の場合、光退色量が多くなる;
−生物学的周辺に対する蛍光感度が高く、発光消光または色ずれを起こす可能性がある;
−サイズの識別がしにくく、臓器および/または悪性細胞の非特異的標的化が生じる;
−生物学的媒質中での半減期が短い、特に血中循環時間が短いため、静脈内注射後材料が迅速に除去される;
−官能基化により生物学標的化および生物作用の点で強い付加価値がもたらされるが、ナノ粒子表面をさらに官能基化するのが困難である。
−ポリマーとナノ集合体との間の相互作用は、全体としてfluo@mag@ポリマーナノ集合体に対して強い接着を提供しなければならない。そのような目的には、ナノ集合体の正に帯電した表面との静電的相互作用が好ましい。さらに、ポリマーはナノ集合体と相互作用する部位を多数含むのが好ましい。
−ポリマーと磁性ナノ粒子との間の会合定数は、蛍光性コアと磁性ナノ粒子との間の会合定数よりも小さくなければならない、換言すれば、ポリマーは、蛍光性コアの表面に存在する磁性ナノ粒子に対する親和性がナノ集合体をコートするのに十分なものであると同時に、磁性シェルの一部がはがれない程度のものでなければならない。
−製造時および生物学的媒質中での使用時ともに、ポリマーの存在がナノ集合体の凝集を引き起こしてはならない。
−ポリマーは官能化可能なものでなければならず、好ましくは、ポリマーは、多数の官能基化可能パターンを示さなければならない。
−ポリマーは生体適合性を示すものでなければならない。

本発明は蛍光性磁性ナノ集合体(fluo@magナノ集合体)に関する。特に、本発明のナノ集合体は、蛍光性コアであって、その表面に接している磁性ナノ粒子、好ましくは超常磁性ナノ粒子によって取り囲まれた蛍光性コア、好ましくは有機蛍光性コアを含む。
蛍光性有機分子を含む蛍光性コアと、
前記蛍光性コアの表面に接している磁性ナノ粒子と

好ましい実施形態では、本発明のナノ集合体の蛍光性コアは式(Ia)の蛍光性有機分子を含み、磁性ナノ粒子はγ−Fe2O3またはFe3O4のナノ粒子である。特定の実施形態では、蛍光性コアは式(Ia)の蛍光性有機分子を含み、磁性ナノ粒子はγ−Fe2O3のナノ粒子である。別の特定の実施形態では、蛍光性コアは式(Ia)の蛍光性有機分子を含み、磁性ナノ粒子はFe3O4のナノ粒子である。

化学試薬および溶媒はすべて、商業的供給源(Aldrich社、Acros社、SDS社)から購入し、受け取ったものをそのまま使用した。分光計測にはAldrich社から購入した分光グレードの溶媒を使用した。
A.1.合成手順
A.1.1.蛍光性有機分子
Fauconら,J.Mater.Chem.C.,2013,1,3879−3886に記載されている通りに蛍光性分子(Ia)および(Ib)を得た。
硝酸安定化磁性ナノ粒子。Massartら(IEEE Trans.Magn.,1981,17,1247−1248)により記載されている手順に従ってマグヘマイトγ−Fe3O3ナノ粒子を調製した。簡潔に述べれば、Fe(II)塩とFe(III)塩を希塩酸中で混合した。濃アンモニアを添加して媒質を急速にアルカリ化することにより、マグネタイトFe3O4ナノ粒子の共沈殿が可能になり、これを分離した。γ−Fe3O4を硝酸で酸性化した後、硝酸第二鉄で80℃にて化学的に酸化させてγ−Fe3O3ナノ粒子を得た。硝酸イオンはコロイド分散液の正に帯電した表面(ζ=+25mV)の対イオンを安定化させる作用があるため、磁気デカンテーションの後、赤色沈殿を硝酸(pH=1.2)に分散させた。
蛍光性分子をTHF(50μL)に溶かした0.1重量%ストック溶液を、ボルテックスミキサーを用いて攪拌しているマグヘマイトナノ粒子の0.006重量%溶液(2.5mL)に注入した。磁性ナノ粒子の硝酸溶液を用いた場合、蛍光性分子と酸化鉄ナノ粒子とを含みfluo@magと称されるハイブリッドナノ粒子の形成が即時に起こる。
A.2.1.ナノ集合体の特徴付け
方法。633nmで作動する4mW He−Neレーザーと175°の角度で後方散乱光を収集する光電子増倍検出器とを備えたナノ粒子サイズ分析機であるZetasizer Nano ZS ZEN3600(Malvern Instruments社)を用いて、動的光散乱法(DLS)によりナノ集合体の流体力学的直径およびサイズ分布を決定した。ナノ集合体の水溶液を用いて20℃で測定を実施した。各試料について、コレログラムの自動調整を伴う多重取得モードで強度測定を実施し、3回の取得の測定値の平均値を求めた。各コレログラムをキュムラントアルゴリズムに当てはめることにより、ナノ粒子の平均サイズおよび分布幅を求めた。
方法。Varian Model Cary 5E分光光度計に積分球DRA2500を用いてUV−可視吸収スペクトルを記録した。Jobin−Yvon社の蛍光光度計(Fluorolog 2)を用いて補正発光スペクトルを得た。EtOH中のクマリン540A(φf=0.38)から溶液中での蛍光量子収率φfを求めた。Spectra−Physics社の装置(二次元レーザーダイオードアレイおよび2つのLBO結晶によって励起される2つのYAGレーザー(Millennia社)によって励起されるチタンサファイアTsunamiレーザー)によって生じる450nmのフェムト秒レーザー励起を用いた時間相関単一光子計数法(TCSPC)により、モノクロメータ(Hamamatsu MCP R3809U光電子増倍管)を用いて580nmにおける蛍光強度減衰を測定した。光音響結晶により4MHzの繰り返し率で900nmにおける光パルスを選択した後、450nmで2倍にした。
方法。Quantum Design社のMPMS−5S SQUID磁力計を温度298Kおよび磁場範囲0〜2Tで作動させて、fluo@magナノ粒子のコロイド懸濁液([Fe]約6.5×105mol/L)に関する温度依存性磁化実験の結果を収集した。磁気双極子間相互作用を回避するために十分に試料を希釈した。ZFC試料で298Kにてヒステリシスループを実施した。
fluo@magナノ集合体を適切な高分子電解質(すなわち、ポリマー)でコートすることにより、水性媒質中で凝集も解離も起こらず、in vitroおよびin vivoでの適用に適した安定なナノ集合体のコロイド溶液が得られる。次いで、それを凍結乾燥させると、構造的および機能的特性を全く失わずに様々な化学的性質およびイオン強度の媒質に再分散させることができる長期間保管用の微粉末が得られる。
以下のプロトコルは、ポリマーとしてPAA高分子電解質(Mw=1.8〜2.2kDa)を用いることにより蛍光性磁性fluo@magナノ集合体を安定化させる工程を説明するものである。目的とする様々な媒質中で生物学的に使用する安定化されたfluo@mag@PAAナノ集合体の凍結乾燥を説明する。
マグヘマイトナノ粒子を用いた合成。蛍光性化合物(Ib)をTHFに溶解した溶液(50μL、0.1重量%)をマグヘマイトナノ粒子の硝酸溶液(2.5mL、0.006重量%、pH=1.2)に激しく攪拌しながら加えた。数秒後、磁性蛍光性ナノ集合体(Ib)−fluo@マグヘマイトが形成された。ポリ(アクリル酸)(2.1kDa、5mg)を粉末で加え、pH=9に達するまで攪拌下で水酸化アンモニウム(1mol/L)を滴加した。得られた半透明の溶液をさらに30分間攪拌し、Spectra Porメンブレン(標準グレード再生セルロース;カットオフ:8〜10kDa)を用いて、fluo@mag@PAAナノ集合体(1)の溶液の最終pHの値が7に達するまで24時間にわたってMillipore水(600mL)に対して透析した。
得られたfluo@mag@PAAナノ集合体を固体状態で長期間にわたって保管するため、様々な体積(1mL〜3mL)のナノ粒子溶液を液体の高さが1cmを超えないようにガラス製バイアルに入れた。液体窒素を用いて溶液を凍結させた。9時間〜12時間にわたって凍結乾燥を実施して暗赤色の粉末を得、これを窒素雰囲気下で−18℃にて保管した。
凍結乾燥試料に十分な溶媒(水、生理的媒質、エタノールなどのアルコール性溶媒)を加えることにより、ナノ集合体の再分散が容易に実施された。ナノ集合体は再分散時に凝集を起こさないため、超音波処理を一切必要としなかった。再分散溶媒を加える量に応じて0.6〜3×10−3mol/Lの範囲、さらには0.6〜7×10−3mol/Lの範囲の鉄濃度を得ることができた。
サイズ、表面電位、組成、光物理的特性および緩和特性に関するfluo@mag@PAAナノ集合体(1)の特徴付けについて記載する。DLS、TEMおよびUV−可視吸収測定(積分球DRA2500を備えたAgilent Model Cary 5E分光光度計を用いてUV−可視吸収スペクトルを記録した)により、時間および溶媒の性質の関数としてコロイド懸濁液の安定性を調べた。質量分析および磁気沈降の両方により組成(コア内の蛍光性分子およびシェルの磁性ナノ粒子の数)を明らかにした。
形成後、凍結乾燥−再分散工程後および超音波処理実施後にfluo@mag@PAAナノ集合体(1)のサイズ特徴付け(DLSおよびTEM測定)を実施した。再分散後、凍結乾燥前の最初の溶液よりも濃度の高い溶液でも、DLSにより凝集は観察されない。さらに、超音波処理後にナノ集合体の解離は全く観察されない。
fluo@mag@PAAナノ集合体(1)の安定性を、時間の関数としてpHおよびイオン強度の異なる様々な溶液で調べた。
有機性コア表面の酸化鉄NPの量を決定する試験を質量分析および磁気沈降それぞれにより実施した。ICP−AES測定(誘導結合プラズマ−原子発光分析)によるナノ集合体の鉄含有量の滴定も同様に実施してもよい。
fluo@mag@PAAナノ集合体(1)の希釈溶液の緩和度測定(1H NMRのT1縦緩和およびT2横緩和時間)を実施した。
様々な媒質に分散させたfluo@mag@PAAナノ集合体(1)の定常状態の吸収および発光測定(図6)、時間分解蛍光測定および広視野蛍光顕微鏡観察を実施して、その発光強度およびスペクトル範囲の特徴を明らかにした。
ナノ集合体の取込みおよびその細胞毒性作用を検討するため、in vitro研究用によく知られたモデル細胞として2種類の異なるヒト細胞株(HEK293およびMDA−MB−468)を用いる。蛍光顕微鏡観察およびTEMイメージングを実施して、細胞内でのナノ集合体の蛍光特性および配置の特徴を明らかにする。
細胞のインキュベーション。細胞を5%CO2雰囲気下、Ibidi社の8ウェルプレート上で少なくとも24時間、37.7℃でインキュベートした。72時間にわたる実験に5000個の細胞をインキュベートした。MDA細胞は、10%のウシ胎仔血清および1%のペニシリン/ストレプトマイシンを含有するRPMI培地で増殖させた。HEK細胞は、10%のウシ胎仔血清および1%のペニシリン/ストレプトマイシンを含有するDMEM培地(高グルコース、グルタミン添加)で増殖させた。
MTT試験:5000個の細胞を適切な培地(200μL)で24時間増殖させた。次いで、Millipore水に分散させたfluo@mag@PAAナノ集合体(1)の溶液と様々な時間の間インキュベートした。MTT試験を用いることにより細胞生存率を評価した。MTTの溶液(20μL;5mg/mL)を細胞に添加し、さらに2時間〜2時間30分インキュベートした。上清溶液を除去し、DMSO(200μL)を加えて、着色した酸化産物を溶解させた。570nmでの吸光度を読み取って参照細胞と比較することにより、インキュベートした細胞の生存率がわかる。
反転モードで作動するLSM(Nikon A1R Si、油浸対物レンズPlan Apo、×60、1.4、λexc=488nm)を用いて共焦点モードで、または倒立顕微鏡(Nikon Eclipse Ti、油浸対物レンズPlan Apo、×60、1.4、λexc=482nm)を用いて広視野モードで蛍光顕微鏡観察を実施した。
−45〜50℃の寒天溶液にHEK細胞を加える;
−0.1Mリン酸塩溶液および3%グルタルアルデヒドを用いて寒天ゲルの小片を4℃で2時間固定する;
−リン酸塩緩衝液およびMillipore水で洗浄する;
−四酸化オスミウムのMillipore水溶液(1%)を用いて1時間、後染色し、Millipore水を用いて繰り返し洗浄する;
−エタノール浴を用いて、エタノールの濃度を漸増させながら(30%、50%、70%、85%、95%、100%)1時間脱水する;
−100%エタノール浴を用いて脱水する(2時間および4℃で一晩);
−エタノールを酸化プロピレンに入れ換える;
−EPON樹脂に包埋する;
−55℃で1日および72℃で60時間、重合させる;
−薄切後、酢酸ウラニルで30分間染色し、Millipore水を用いて洗浄する。
一連の5匹のマウスを用いてin vivo磁気共鳴イメージングを実施した。マウスを安楽死させた後、摘出した臓器をスライスし、組織の自家蛍光を最小限に抑えるため、1光子および2光子励起を用いて蛍光顕微鏡観察を実施した。ナノ集合体のイメージングの空間解像度を高めるため、肝臓の超薄スライス(厚さ70nm)を用いて透過型電子顕微鏡測定(TEM)を実施した。
手順。5匹の雌性BALB/Cマウス(5週齢)にMRI分光計BioSpect 4,7T(Bruker社)を用いてT2強調MRI実験を実施した。シーケンス(T2*モード)のエコー時間TEおよび繰返し時間TRをそれぞれ5msおよび300msに設定した。この分析を実施するにあたっては、Fe濃度13μmol/kgマウスを選択した。
組織薄切手順。注射から24時間後、マウスを屠殺した。fluo@mag@PAAナノ集合体(1)の生体内分布を評価するため、ex vivo分析用に5種類の臓器(肝臓、脾臓、腎臓、肺および心臓)を採取した。臓器を、4℃の亜鉛含有固定液(Pharmingen社、フランス)に48時間、次いで4℃の4重量%パラホルムアルデヒド−20重量%スクロース溶液に24時間、連続して浸漬させた。最後に、液体窒素の蒸気を用いてOCT(Tissue−Tek社)の中で凍結させた。亜鉛含有固定液は、固定工程で蛍光性分子が弱まるのを防ぐ。凍結させた臓器は使用するまで−80℃で30日間保管した。クリオトームクリオスタット(Leica Biocut 2030)を用いて、−20℃で厚さ10μmの切片に切断した。切片を直ちにスーパーフロストガラススライドに載せ、2時間乾燥させた後、Prolong(細胞核染色剤のDAPIを含有する、または含有しないもの)で処理し、上部カバースリットで保護して、顕微鏡イメージングを可能にした。切片を観察する前に少なくとも72時間、4℃で放置した。
−1光子蛍光イメージング:励起源であるAr+およびHe−Neレーザーを備えたNikon A1R Si(油浸対物レンズPlan Apo、×60、1.4、λexc=488nmを使用)−IRSUN社/ナントとの共同研究;
−2光子蛍光イメージング:波長可変Ti:サファイアフェムト秒レーザー(Mai Tai、710〜920nm)およびマルチスペクトルイメージングが可能な32チャンネルQUASAR GaAsp検出器を備えたZeiss LSM780。画像はすべて、830nmで励起させ、2種類の検出チャンネル(450(±35)nmの青を中心とするチャンネルと654(±104)nmの赤を中心とするチャンネル)を用いて取得した−KU Leuven/ベルギーとの共同研究。
手順および試薬
カコジル酸ナトリウム緩衝液0.2mol/L、pH=7.4(1年以上保管−500mL)。カコジル酸ナトリウム三水和物(21.4g)、塩化カルシウム二水和物(25mg)および塩化マグネシウム六水和物(50mg)を蒸留水(450mL)に溶解した。磁気攪拌下で0.5mol/LのHCl溶液を加えることにより混合物のpHを調整した。最後に蒸留水を加えて全量を500mLに調整した。
C.1.葉酸による官能基化
fluo@magナノ集合体は、ナノ粒子の細胞内移行の速度を増大させる葉酸(FA)などの生物学的に活性な分子でコートし得る。酸化鉄ナノ粒子の外層に対して錯体を形成するカルボキシル単位があることから葉酸を選択した。
fluo@mag@PAAナノ集合体(1)に抗生物質としての役割を果たすアミノクマリンなどの生物学的に活性な分子を共有結合によりグラフトすることが可能である。アミノクマリンは、アミド結合形成を介してfluo@mag@PAA表面と共有結合させることができアミノ官能基を示す。
fluo@mag@ポリマーのバイオコンジュゲーションには、生物学的実体と効率的かつ特異的に反応することがわかっている反応基の導入が必要である。マレイミド誘導体は、タンパク質および抗体中の存在量がアミノ基に比して少ないチオール基と特異的に反応するため、特に興味深いものである。マレイミド誘導体の導入は、ポリマーシェルとマレイミド単位との間に共有アミド結合を生じさせるペプチドカップリングを介して進行する。マレイミドのグラフトは、蛍光測定で、得られた発光スペクトルの最大発光波長がカップリング前よりもわずかに浅色シフトする(Δλmax≒−10nm)ことにより明らかになった。
ストレプトアビジンと結合した生物学的に関心のある様々な実体と複合体を形成させるのに使用される非常に一般的なビオチンとの共有的コンジュゲーションにより、fluo@mag@ポリマーを官能基化した。ビオチンとストレプトアビジンは安定度定数の極めて高い(Kd=108)アフィニティー複合体を形成することから、以下の結果は、ビオチンで官能基化したfluo@mag@ポリマーを用いて、ストレプトアビジンとコンジュゲートした生物学的実体を容易に認識することが可能であることを示している。

Xが、−COOH、アルキル、アリール、−CO−アルキル、ポリ(エチレングリコール)、−ポリ(エチレングリコール)−OMeを表し、Yが、−(C=O)−O−L1−R8、−(C=O)−S−L1−R8、−(C=O)−NH(−L1−R8)または−(C=S)−NH(−L1−R8)を表し、
L1は、1〜150個の鎖原子を有するアルキル、アルケン、アリール、アリールアルキル、ポリ(エチレングリコール)またはポリ(プロピレングリコール)連結基から選択されるスペーサーを表し、前記連結基は任意選択で、1つまたは複数の−O−、−S−、−NR9−、−CO−、−NHCO−、−CONH−またはその組合せにより中断または終結されていてよく、R9はHまたはアルキルであり、

L2は、単結合または1〜150個の鎖原子を有するアルキル、アルケン、アリール、アリールアルキル、ポリ(エチレングリコール)もしくはポリ(プロピレングリコール)連結基から選択されるスペーサーを表し、前記連結基は任意選択で、1つまたは複数の−O−、−S−、−NR9−、−CO−、−NHCO−、−CONH−またはその組合せにより中断または終結されていてよく、R9はHまたはアルキルであり;任意選択で、L2とR10とを連結させている反応基の残基をさらに含み、
R10は、アミノ酸、ペプチド、タンパク質、抗体、親和性タンパク質、酵素、多糖、デキストラン、ベンジルグアニン、脂質、脂質集合体、脂肪酸、ヌクレオシド、ヌクレオチド、オリゴヌクレオチド、ハプテン、アプタマー、リガンド、抗生物質、基質、ビオチン、アビジン、ストレプトアビジン、合成ポリマー、ポリ(エチレングリコール)、ポリ(プロピレングリコール)、ポリマー微粒子、ナノ粒子、フルオロフォア、発色団、放射性同位元素、放射性同位元素の大環状複合体およびその組合せから選択される生物活性基を表す、

前記ポリマーがポリ(アクリル酸)である、(1)〜(8)のいずれかに記載のナノ集合体システム。

(1)〜(9)のいずれかに記載のナノ集合体システムを製造する工程であって、攪拌下で磁性ナノ粒子の分散液に蛍光性有機分子の溶液を注入することと、それに続く前記ポリマーの添加段階とを含む、工程。
(11)
生物活性基による前記ポリマーの官能基化のその後の段階をさらに含む、(10)に記載の工程。
(12)
(1)〜(9)のいずれかに記載のナノ集合体システムを、少なくとも1つの薬学的に許容される賦形剤とともに含む、医薬組成物。
(13)
(1)〜(9)のいずれかに記載のナノ集合体システムを含む、薬剤。
(14)
生理的媒質から分子および/または生物学的標的を抽出し検出するための、(1)〜(9)のいずれかに記載のナノ集合体システムの使用。
(15)
(1)〜(9)のいずれかに記載のナノ集合体システムを含む、キット。
Claims (15)
- ナノ集合体システムであって、
蛍光性有機分子を含む有機蛍光性内部コアと
前記蛍光性内部コアの表面に接している磁性ナノ粒子と
を含む、ナノ集合体および
前記ナノ集合体の表面に吸着した少なくとも1つのポリマー
を含み、
但し、シリカ、官能基化シリカ、ドープシリカ、グラフトシリカまたはその組合せを含まない、
ナノ集合体システム。 - 前記蛍光性有機分子が式I
Lが、(i)、(ii)および(iii)
Xが、O、CH2またはNR8を表し、R8は、H、アルキルまたはアルケンを表し、
X’がOまたはNR9を表し、R9は、H、アルキルまたはアルケンを表し、
nが、1、2、3および4から選択される整数を表し、
R1が−CO2Hまたは−P(O)(OH)2を表し、
R2、R3、R4、R5、R6およびR7がそれぞれ独立して、アルキルおよびエステルまたはポリ(エチレングリコール)から選択される任意選択で官能基化された基を表し、好ましくは、R2、R3、R4、R5、R6およびR7が、それぞれメチル基を表す、
請求項1に記載のナノ集合体システム。 - 前記磁性ナノ粒子が、γ−Fe2O3およびFe3O4からなる群より選択される超常磁性ナノ粒子である、請求項1または2に記載のナノ集合体システム。
- 20〜700nmの範囲の流体力学的直径を有する、請求項1〜3のいずれか1項に記載のナノ集合体システム。
- 前記蛍光性内部コアが、1×104〜1×107個の範囲の多数の有機蛍光性分子を含む、請求項1〜4のいずれか1項に記載のナノ集合体システム。
- 1×102〜1×106個の範囲の多数の磁性ナノ粒子を含む、請求項1〜5のいずれか1項に記載のナノ集合体システム。
- 前記ポリマーがイオン性ポリマー、好ましくは高分子電解質である、請求項1〜6のいずれか1項に記載のナノ集合体システム。
- 前記ポリマーが式II
mが、20〜150の範囲の正の整数を表し、
x、yおよびzがそれぞれ独立して、0%〜100%の範囲のmの百分率を表し、x+y+zは100%のmに等しく、
Xが、−COOH、アルキル、アリール、−CO−アルキル、ポリ(エチレングリコール)、−ポリ(エチレングリコール)−OMeを表し、Yが、−(C=O)−O−L1−R8、−(C=O)−S−L1−R8、−(C=O)−NH(−L1−R8)または−(C=S)−NH(−L1−R8)を表し、
L1は、1〜150個の鎖原子を有するアルキル、アルケン、アリール、アリールアルキル、ポリ(エチレングリコール)またはポリ(プロピレングリコール)連結基から選択されるスペーサーを表し、前記連結基は任意選択で、1つまたは複数の−O−、−S−、−NR9−、−CO−、−NHCO−、−CONH−またはその組合せにより中断または終結されていてよく、R9はHまたはアルキルであり、

Zが、−(C=O)−O−L2−R10、−(C=O)−S−L2−R10、−(C=O)−NH(−L2−R10)または−(C=S)−NH(−L2−R10)を表し、
L2は、単結合または1〜150個の鎖原子を有するアルキル、アルケン、アリール、アリールアルキル、ポリ(エチレングリコール)もしくはポリ(プロピレングリコール)連結基から選択されるスペーサーを表し、前記連結基は任意選択で、1つまたは複数の−O−、−S−、−NR9−、−CO−、−NHCO−、−CONH−またはその組合せにより中断または終結されていてよく、R9はHまたはアルキルであり;任意選択で、L2とR10とを連結させている反応基の残基をさらに含み、
R10は、アミノ酸、ペプチド、タンパク質、抗体、親和性タンパク質、酵素、多糖、デキストラン、ベンジルグアニン、脂質、脂質集合体、脂肪酸、ヌクレオシド、ヌクレオチド、オリゴヌクレオチド、ハプテン、アプタマー、リガンド、抗生物質、基質、ビオチン、アビジン、ストレプトアビジン、合成ポリマー、ポリ(エチレングリコール)、ポリ(プロピレングリコール)、ポリマー微粒子、ナノ粒子、フルオロフォア、発色団、放射性同位元素、放射性同位元素の大環状複合体およびその組合せから選択される生物活性基を表す、
請求項1〜7のいずれか1項に記載のナノ集合体システム。 - 前記ポリマーがポリ(アクリル酸)である、請求項1〜8のいずれか1項に記載のナノ集合体システム。
- 請求項1〜9のいずれか1項に記載のナノ集合体システムを製造する工程であって、攪拌下で磁性ナノ粒子の分散液に蛍光性有機分子の溶液を注入することと、それに続く前記ポリマーの添加段階とを含む、工程。
- 生物活性基による前記ポリマーの官能基化のその後の段階をさらに含む、請求項10に記載の工程。
- 請求項1〜9のいずれか1項に記載のナノ集合体システムを、少なくとも1つの薬学的に許容される賦形剤とともに含む、医薬組成物。
- 請求項1〜9のいずれか1項に記載のナノ集合体システムを含む、薬剤。
- 生理的媒質から分子および/または生物学的標的を抽出し検出するための、請求項1〜9のいずれか1項に記載のナノ集合体システムの使用。
- 1〜9のいずれか1項に記載のナノ集合体システムを含む、キット。
