Abstract
Description
しかし、上記レジン塗布法においては、有機成分によるアレルギー反応のおそれがあり、また、生体への親和性にも問題がある。
しかし、上記特許文献1の技術の実用化に向けた検討の中で、量産化や、シートの残存性などに、課題があることが分かってきた。
すなわち、特許文献1では、生体親和性セラミックス膜として、結晶化したハイドロキシアパタイト(HAp)膜などが記載されているが、この結晶化には、10時間といった長時間の加熱工程が必要となり、量産化に不向きであることが分かった。
また、非晶質リン酸カルシウムを用いれば、結晶化のための加熱工程は不要であるが、非晶質リン酸カルシウムのシートを象牙質表面に貼付した場合、時間の経過とともにシートが消失する場合があることが判明した。
しかし、フッ素ドープ非晶質リン酸カルシウムシートは象牙質との固着性が乏しい。そこで、非晶質リン酸カルシウム層を、上記フッ素ドープリン酸カルシウムシートに付加することを検討したところ、非晶質リン酸カルシウム層が象牙質との接着層として機能することが分かった。
さらに、このような積層シートであれば、シートの消失の問題も解決されることが分かった。これは、以下の理由によると推測された。
すなわち、まず、シートの消失は、スメア層除去時のエッチング液やシート貼付時の酸性溶液による象牙質表面の脱灰によって、コラーゲン線維が露出し、その露出したコラーゲン線維にシートのCa2+とPO4 3-が吸収されることで生じると推測された。この点、フッ素ドープリン酸カルシウムと非晶質リン酸カルシウム層の複合シートにおいては、接着層の非晶質リン酸カルシウム層が、脱灰部の修復層としても機能し、シートの消失が回避できたものと推測された。
本発明は、以上の如き検討の結果、完成されるに至った。
以下、本発明において、「フッ素ドープ非晶質リン酸カルシウム」を「F−ACP」、「非晶質リン酸カルシウム」を「ACP」と略記することがある。
本発明にかかる歯科治療用複合シートは、フッ素ドープ非晶質リン酸カルシウム層と、非晶質リン酸カルシウム層とを備える。
非晶質リン酸カルシウム(ACP,Ca3(PO4)2・nH2O)は、カルシウムイオンとリン酸イオンを構成要素とする化合物であり、ハイドロキシアパタイト(Ca10(PO4)6(OH)2、以下「HAp」と略記することがある)の前駆体として知られている。
フッ素ドープ非晶質リン酸カルシウムは、上記非晶質リン酸カルシウムにフッ素がドープされた化合物である。
本発明の歯科治療用複合シートは、一般的な薄膜形成方式と薄膜単離技術によって得ることができる。
気相法としては、例えば、レーザーアブレーション法、スパッタリング法、プラズマ法、真空蒸着法、イオンプレーティング法、熱化学気相成長法、プラズマ化学気相成長法などが採用できる。
好ましくはレーザーアブレーション法であるが、このレーザーアブレーション法は、例えば、次のような手順で行なう。
すなわち、まず、基材をレーザーアブレーション装置に入れ、排気する。ACP層形成の際は、水蒸気含有ガス又は炭酸ガス含有ガスを装置内に導入する。F−ACP層形成の際は、組成中のフルオロ基が水酸基で置換されるのを避けるため、水蒸気含有ガスや炭酸ガス含有ガスの導入は行わず、真空状態としておく。
次に、ArFエキシマレーザー発生装置等のレーザー発生装置、ミラー、レンズ等からなるレーザー光源から発生したレーザー光線をターゲットに照射する。これによって、ターゲットが分解して原子、イオン、クラスター等が放出され、基材上に、ターゲットの組成を反映した膜が形成される。
前記溶媒としては、基材上の薄膜を溶解せず、少なくとも基材と薄膜とが接する部分を溶解するものであれば良い。水系溶媒のような極性溶媒、有機溶媒のような非極性溶媒など、特に限定することなく使用できる。水系溶媒の場合には、純水、細胞培養用緩衝液、細胞培養用液体培地等が好ましい。また、有機溶媒の場合は、揮発性で生体セラミックス膜に残らないアセトン、ヘキサン、アルコール等が好ましい。なお、溶解時間や溶媒の温度などは基材の材質や厚さなどに応じて任意に調節すればよい。また、基材の材質に応じて複数の溶媒を組み合わせてもよい。
複数の単一層を重ね合わせる場合には、層間の接着性を高めるために、例えば、脱灰・再石灰化を利用することとしてもよい。
具体的には、人工唾液や、第一リン酸カルシウム水溶液のようにCa2+とPO4 3-を含む弱酸性の溶液を用いて、各層の界面に一時的な脱灰を起こしたのち、中性若しくはアルカリ性の環境下で過飽和になったCa2+とPO4 3-を歯質に戻して再石灰化を起こし、各層を固着させることで、強固な接着が可能となる。この方法では、無機質しか使用しないので、薬剤や高分子材料などを使用した場合のようなアレルギーの心配が少なく、侵襲を少なくすることができる。
また、歯科治療用複合シートにおける非晶質リン酸カルシウム層の厚みは、特に限定するわけではないが、例えば、歯科治療用複合シートの長期残存性を確保する上では、0.3μm以上が好ましく、2μm以上がより好ましい。
貫通孔の形成は、例えば、特許第4919519号公報に記載されるように、突起物によるパターンを形成した基材を用いることにより行うことができる(特許第4919519号公報の図3(b)〜(d)参照)。
また、孔の空いていない歯科治療用複合シートに対して、レーザーを照射して貫通孔を形成することもできる。
貫通孔としては、特に限定するわけではないが、例えば、直径50〜200μm程度が好ましく、60〜150μm程度がより好ましく、80〜120μm程度がさらに好ましい。また、貫通孔と貫通孔との間隔は、500〜2000μm程度が好ましく、750〜1500μm程度がより好ましく、900〜1200μm程度がさらに好ましい。
そのため、本発明の歯科治療用複合シートは、結晶化ハイドロキシアパタイトを用いる場合と比べると、結晶化に要する時間・コストが削減できる分、量産化に有利である。
本発明の歯科治療用複合シートは、象牙質知覚過敏の治療や、エナメル質、象牙質の再生などのために有効に使用できる。
図1は、本発明の歯科治療用複合シート10の一使用例を模式的に示した図である。21はエナメル質、22は象牙質、23は象牙細管、24は歯髄、25は歯肉である。
図1では、エナメル質21の一部が欠損し、象牙質22が一部露出している。象牙質22に存在する象牙細管23が開口していることで、象牙細管23内の組織液が熱、圧力等の色々な刺激によって動かされ、その動きが象牙質22の近くにある歯髄24の神経を刺激することにより象牙質知覚過敏が生じる状態であると考えられる。
図1に示すように、本発明の歯科治療用複合シート10を、歯質表面、特に、エナメル質21ないし象牙質22の欠損部位に貼付して使用することで、象牙質知覚過敏を治療することができる。
具体的には、本発明の歯科治療用複合シートを、人工唾液や、第一リン酸カルシウム水溶液のようにCa2+とPO4 3-を含む弱酸性の溶液を用いて歯質に付着することによって、歯科治療用複合シートと歯質の界面に一時的な脱灰を起こしたのち、中性若しくはアルカリ性の環境下で過飽和になったCa2+とPO4 3-を歯質に戻して再石灰化を起こし、歯科治療用複合シートと歯質を固着させることで、強固な接着が可能となる。この方法では、無機質しか使用しないので、薬剤や高分子材料などを使用した場合のようなアレルギーの心配が少なく、侵襲を少なくすることができる。
また、上述のように、弱酸性の溶液を用いた接着をする場合においても、本発明の歯科治療用複合シートでは、接着層の非晶質リン酸カルシウム層が脱灰部修復層としても機能し、象牙質の脱灰によって露出したコラーゲン線維にCa2+とPO4 3-を供給するので、複合シートが容易に消失してしまうことはない。
さらに、本発明の歯科治療用複合シートは、透明ないし白色であるため、審美性のための歯科化粧材としても機能させることが可能である。
薄膜作製用基板として、Siウェハ上に単離層としてのレジスト膜をコーティングした基板を使用した。この基板上にパルスレーザーデポジション(PLD)法(レーザーは、KrFエキシマレーザー、波長248nmを使用)により、膜厚が0.3μmのACP修復層を形成し、更にその上に膜厚が2μmのF−ACP薄膜を堆積してF−ACP/ACP複合シートを作製した。
ACP層の形成には、ターゲットとしてハイドロキシアパタイト(「セルヤード」(登録商標)、ペンタックス製)を用い、F−ACP層の形成には、ターゲットとしてフッ化アパタイト(太平化学産業株式会社性)を用いた。
ACP層の成膜条件は、雰囲気ガス:O2+H2O、ガス圧:0.1Pa、基板温度:室温、エネルギー:150mJとした。
F−ACP層の成膜条件は、雰囲気ガス:なし(真空)、ガス圧:1.3×10-4Pa、基板温度:室温、エネルギー:150mJとした。
さらに、シート貼付時の再石灰化を促進させる目的でF−ACP/ACP積層膜に小孔径150μm、小孔間が1mmの三角配列の構造をもつ金属マスクを取り付け、エキシマレーザー加工法により貫通孔をあけた。
次にSi基板と薄膜の間のレジスト層をアセトンで溶解させ、小孔をもつF−ACP/ACP薄膜を基板から単離し、これを貫通孔のあるF−ACP/ACP複合シートとして回収した。
ACP修復層の厚みを0.5、1、2,4,6μmに変更したこと以外は実施例1と同様にして、実施例2〜6のF−ACP/ACP複合シート(貫通孔あり)を得た。
象牙質表面へのシートの貼付方法としては、象牙質表面を#600の耐水研磨紙で研磨した後、表面をエッチング剤で10秒間洗浄し、水洗いした後、蒸留水中で60秒間超音波洗浄を行い、スメア層の除去と象牙細管の開口を行った。貼付するシートはUVオゾン処理装置で表裏それぞれ300秒ずつ洗浄を行った。pH2.0の第一リン酸カルシウム水溶液を滴下した象牙質表面にシートを貼付し、10分間静置することで象牙質とシートの界面を脱灰させた。その後界面を再石灰化させる目的でpH7.0〜7.2の人工唾液をしみ込ませたメラミンスポンジと50gの分銅を乗せて、37℃の恒温槽で3時間静置した。その後、象牙質上に実施例1〜4(ACP層厚み0.3μm、0.5μm、1μm、2μm)の各F−ACP/ACP複合シートを貼付し、シートが消失するまでの時間を目視で調べた。
さらに、長期残存性に特に優れた実施例4〜6(ACP層厚み2μm、4μm、6μm)については、ブラッシング試験も行うこととし、ブラッシング試験後のシートについて、上記の長期残存性試験をより長期(2週間)で評価することとした。
ブラッシング試験は、以下のようにして行った。
すなわち、シートが貼付された象牙質表面に対し、歯ブラシ(SUNSTAR#211)を用い、荷重200gで、90ストローク/分の速度で、20ストロークブラッシングし、表面状態を分析した。
実施例1〜4(ACP層厚み0.3μm、0.5μm、1μm、2μm)について、脱灰部修復層であるACP層の厚さの違いと、シート消失までの時間を測定した結果を表1に示す。
特に、ACP層の厚みが厚いほどシートの残存性が高くなることが分かる。
なかでも、2μmのACP層の厚さをもつシートは、168時間(7日間)の長時間でもシートが残存することが確認できた。
図2に示す結果から、象牙質上でもシートを長時間残存させるために、2μm以上のACP層を設けることが特に有効であることが分かった。
F−ACP薄膜の特性を評価するため、HAp薄膜,F−HAp薄膜とともに、溶出試験を行った。
直径15mmのTi基板を#320から#1000の耐水研磨紙と直径0.3μmのアルミナ粒子で順次研磨し、鏡面加工をした。
HAp粉末,FAp粉末を共に1g秤量し、油圧式プレス機にて19.6MPaの圧力で加圧形成することで直径16mm、厚さ3mmのバルク体を作製した。その後、電気炉を用いて大気中で750℃−10hで焼結した。
研磨したTi基板にPLD法にて厚さ4μmのACP薄膜、F−ACP薄膜を成膜し、ACP/Ti、F−ACP/Tiを作製した。
電気炉を用いて、大気中、450℃で10時間、ACP/Ti、F−ACP/Tiをポストアニール処理して、HAp/Ti、F−HAp/Tiを作製した。
0.5Mリン酸二水素ナトリウム(NaH2PO4)水溶液と、0.5Mリン酸水素二ナトリウム(Na2HPO4)水溶液を、pHを計測しながら撹拌し、pH5.5になるように混合調整した0.5Mリン酸ナトリウム緩衝液を作製した。その後、オートクレーブ装置にて液体滅菌したものを溶出試験液とした。
上記にて作製した各試料、すなわち、F−ACP/Ti,HAp/Ti,F−HAp/Ti(それぞれ3枚)について、上記溶出試験液(pH5.5の0.5Mリン酸ナトリウム緩衝液)を用いて、ICP−MSによる溶出試験を行った。
具体的には、20mm角のテフロン(登録商標)容器中に作製した溶出試験液を10mL入れ、3種類それぞれの試料を浸漬させたのち、1、3、10日後の溶液をそれぞれ50μLずつ分取する。分取した溶液を硝酸溶液で100倍希釈して試料検液とし、ICS−MS(アジレント・テクノロジー製 Agilent8800型)によりCaイオン濃度を測定した。
図3に示す結果から、F−ACP薄膜が、HAp薄膜と同程度の耐酸性を有することが確認できた。

- 非晶質リン酸カルシウム層の厚みが2μm以上である、請求項1に記載の歯科治療用複合シート。
